Tuple space is accessed through a small set of primitive operations and is conceptually easy to understand and manipulate. The physical implementation of a Linda tuple space may of course be completely different from the conceptual model. Rhodes has implemented versions of Linda on a ring of RS-232 joined PC's and on a cluster of T800 transputers with a single copy of tuple space on one transputer. Current research targets the implementation of a distributed Linda server on a network of heterogeneous processors. This work describes the design and implementation of a distributed Linda server. Emphasis is placed on aspects of the design which enhance portability and efficiency.

- Description: Iron-formations are ferruginous sedimentary rocks which have their source from fumarolic activity associated with submarine volcanism, with deposition of iron as oxides, hydroxides, and hydrous oxide-silicate minerals in shallow and/or deep marine sedimentary systems. The Precambrian ironformations of southern Africa have a wide age range, but are more prominently developed before 1.SGa. These iron formations occur in greenstone belts of the Kaapvaal and Zimbabwean cratons, in the Limpopo mobile belt, in cratonic basins and in the Damara mobile belt. The Archaean-Proterozoic sedimentary basins and greenstone belts host iron ore deposits in iron-formation. Iron formations have a lengthy geological history. Most were subjected to intense, and on occasions repeated, tectonic and metamorphic episodes which also included metasomatic processes at times to produce supergene/hypogene high grade iron ores. Iron-formations may be enriched by diagenetic, and metamorphic processes to produce concentrating-grade ironformations. Uplift, weathering and denudation, have influenced the mineral association and composition of the ores, within which magnetite, haematite and goethite constitute the major ore minerals. The iron resources of the southern Africa region include the Sishen deposits, hosting to about 1200 Mt of high grade direct shipping ore, at >63% Fe. Deposits of Zimbabwe have more than 33 000 Mt of beneficiable iron-formation. The evaluation of an iron ore prospect involves many factors which must be individually assessed in order to arrive at an estimate of the probable profitability of the deposit. Many of these are geological and are inherent in the deposit itself. Other factors are inherent aspects of the environment in which the ore is formed. Although the geological character of the ore does not change, technological advances in the processing techniques may have a great effect on the cost of putting the ore into marketable form. Geochemical, geophysical and remote sensing methods would be used for regional exploration. Chip sampling and drilling are useful for detailed exploration. Purely geological exploration techniques are applicable on a prospect scale in the exploration of iron ore deposits. Regional exploration targeting should choose late Archaean greenstone belts containing oxide facies iron-formation or Early Proterozoic basins located at craton margins as they are both known to host high-grade haematite orebodies formed by supergene/hypogene enrichment. Most types of iron ore deposits in southern Africa are described and classified. An attempt is made to emphasize the major controls on mineralisation, in the hope that these may be applicable to exploration both in the southern African region and within analogous settings around the world.

- Description: This thesis presents a detailed chemical and spectroscopic determination of the capsular, polysaccharide K-antigen isolated from the Klebsiella bacterium, serotype K43 (culture #2482). The repeating unit of the capsular polysaccharide was found to be of the "3 + 2" repeating unit type. A uronic acid was found as part of a disaccharide side chain and the main chain of the polysaccharide was found to be composed of a neutral trisaccharide of mannose and galactose. The work forms part of an ongoing research interest in bacterial polysaccharides of this laboratory and now completes the structural elucidation of all the Klebsiella K-antigens, bar three antigens which were originally assigned to other laboratories. These data together with the respective serological characteristics of each serotype are available to the molecular biologist, and may result in the production of: vaccine(s) against Klebsiella infections, diagnostic products and novel carrier molecules enabling targeted drug delivery.

Algorithmic skeletons as a method of parallel programming

- Description: A new style of abstraction for program development, based on the concept of algorithmic skeletons, has been proposed in the literature. The programmer is offered a variety of independent algorithmic skeletons each of which describe the structure of a particular style of algorithm. The appropriate skeleton is used by the system to mould the solution. Parallel programs are particularly appropriate for this technique because of their complexity. This thesis investigates algorithmic skeletons as a method of hiding the complexities of parallel programming from the user, and for guiding them towards efficient solutions. To explore this approach, this thesis describes the implementation and benchmarking of the divide and conquer and task queue paradigms as skeletons. All but one category of problem, as implemented in this thesis, scale well over eight processors. The rate of speed up tails off when there are significant communication requirements. The results show that, with some user knowledge, efficient parallel programs can be developed using this method. The evaluation explores methods for fine tuning some skeleton programs to achieve increased efficiency.

An historical study of channel change in the Bell river, north eastern Cape

- Description: Channel instability has occurred in the Bell river, north eastern Cape, in the form of meander cutoffs, incipient meander cutoffs, channel straightening and general channel instability. Recent cutoffs occurred in 1974 and 1988. The study examines the spatial and temporal controls of channel form and pattern in the Bell river in order to assess the causes of channel instability. From the 17 km surveyed stretch, it was found that the main spatial controls of channel form were riparian vegetation density and channel bed material. Discharge as estimated in the field was not the main controlling variable of channel form. Two distinct groups of stream beds were identified from the survey; an upper gravel-bed stream and a lower sand-bed stream. These sites displayed distinct form ratios, channel gradients and bed material characteristics. The incidences of major channel instability were identified as being the transitional zone between the two reaches. Examination of temporal controls of channel form included climatic trend analysis and catchment sediment production analysis. Rainfall analysis indicated that no long term progressive trends in the annual or seasonal data existed. Distinct wet and dry cycles occur with peaks every 16 to 19 years. Wet cycles are the result of an increase in the frequency of daily events rather than in the magnitude of events. Flow record analysis demonstrated the relationship between regional discharge and upper catchment rainfall. Coincidence of peak flows and channel straightening were also noted. Soil erosion surveys showed that erosion had increased in the catchment and that accelerated erosion were probably the result of overstocking and poor veld management. It was concluded that channel changes in the Bell river are possibly the result of anthropogenic influence in catchment and channel processes. Increased sediment production to the channel resulted in channel aggradation with attendant instability. The plantation of riparian vegetation led to perimeter stability in the short term at flows less than bankfull discharge, but served to reduce cross-sectional area in the long term, thereby increasing the potential for flooding, meander cutoffs and channel change

An investigation into cholinergic interactions in the rat pineal gland

- Description: The mammalian pineal gland is mainly innervated by the sympathetic nervous system which modulates the activity of indole pathway enzymes and the secretion of pineal hormones. Recently researchers have demonstrated and characterized the presence of muscarinic cholinergic receptors in the pineal gland. However the role of these receptors remains unclear. In an attempt to investigate the role of cholinergic receptors in the pineal gland, a number of studies were carried out on the various steps in the indole metabolic pathway, using various agents which act on the cholinergic system. Investigations using pineal organ cultures showed that stimulation of these muscarinic cholinergic receptor sites with a parasympathomimetic agent, a rise in levels of aHT occurred without a concomitant increase in aMT levels. Further organ culture experiments using the cholinergic agonist acetylcholine and anticholinesterase agent physostigmine, produced a similar rise in aHT without altering aMT levels. This acetylcholine-induced rise in aHT levels were not altered by the ganglion blocking agent hexamethonium whilst the antimuscarinic agent atropine prevented the acetylcholine-induced rise in aHT levels. Thesefindings suggest that cholinergic agents may play a role in regulating indoleamine synthesis in the pineal gland. Cyclic-AMP assay studies showed that acetylcholine increases pineal cAMP levels significantly and does not influence the isoproterenol-induced cAMP rise in the pineal gland. The cAMP regulator cAMP-phosphodiesterase (cAMP-PDE) was found to increase significantly in the presence of the anticholinesterase agent physostigmine. NAT enzyme studies revealed that physostigmine does not affect NAT enzyme levels significantly and HIOMT studies showed that this agent does not inhibit HIOMT activity. The mechanism by which acetylcholine and physostigmine are able to cause a increase in aHT and not aMT levels needs to be researched further. Acetylcholinesterase enzyme assay studies revealed that the AChE enzyme undergoes a diurnal rhythm in the pineal gland with activity being higher during the day and lower at night. Investigations using the drug reserpine showed that this rhythm is not under the control of the sympathetic nervous system. Further research needs to be done however, in determining whether or not this enzyme is present in the pineal gland to regulate the levels of acetylcholine interacting with muscarinic receptors in the gland, or for some other reason. Choline acetyltransferase studies demonstrate the presence of the enzyme in the rat brain cerebral cortex as well as showing that melatonin increases ChAT enzyme activity in this tissue. This suggests that melatonin plays a role in cholinergic transmission there. ChAT activity could not be measured in the pineal gland however. Muscarinic receptor binding studies also carried out on rat brain cerebral cortex show that melatonin enhances cholinergic receptor affinity and receptor number in this tissue. An investigation into the potential immunogenicity of various extracts of the South African bont tick Amblyomma hebraeum

- Description: Rabbits and goats were inoculated with crude, membrane-associated and soluble components extracted from unengorged adult females and nymphs of the bont tick Amblyomma hebraeum. Inoculation provided some protection against nymphal infestation, however it had little effect on adult feeding. Histological examination of adults fed on inoculated hosts showed evidence of gut damage. Skin provocation testing with tick extracts elicited a Type I immediate hypersensitivity which was influenced by antihistamine. A delayed skin reaction was also evident. Whether this was attributable to Type III Arthus reaction or Type IV cell-mediated hypersensitivity was not determined. A comparative histological study of sites of tick extract injection, on inoculated and naive hosts, demonstrated the role of eosinophils in the hosts response to tick feeding. Serological examination revealed elevated anti-A hebraeum lgG titres following inoculation. - Description: Rabbits and goats were inoculated with crude, membrane-associated and soluble components extracted from unengorged adult females and nymphs of the bont tick Amblyomma hebraeum. Inoculation provided some protection against nymphal infestation, however it had little effect on adult feeding. Histological examination of adults fed on inoculated hosts showed evidence of gut damage. Skin provocation testing with tick extracts elicited a Type I immediate hypersensitivity which was influenced by antihistamine. A delayed skin reaction was also evident. Whether this was attributable to Type III Arthus reaction or Type IV cell-mediated hypersensitivity was not determined. A comparative histological study of sites of tick extract injection, on inoculated and naive hosts, demonstrated the role of eosinophils in the hosts response to tick feeding. Serological examination revealed elevated anti-A hebraeum lgG titres following inoculation. Aspects of the biology and ecology of the South African abalone Haliotis midae Linnaeus, 1758 (Mollusca Gastropoda) along the eastern Cape and Ciskei coast

- Description: The South African abalone Haliotis midae Linnaeus, 1758, is an important commercial, recreational and aquaculture mollusc species. It is the largest of the six haliotid species found in South African waters and has the second largest distributional range aside from Haliotis spadicea which is widely exploited by rock and surf anglers as bait. Analysis of population structure at Great Fish Point revealed that H. midae exhibited a high degree of microhabitat specificity, and that while dietary habits played a role in habitat selection, it was ultimately the activities of predators which confined size classes to particular niches and restricted all animals to nocturnal activities. Large (> 100 mm SL) exposed animals relied on shell thickness and adhesion to combat predators, while small (> 45 mm SL) sub-boulder animals and medium sized (50 - 95 mm SL) animals relied on their cryptic microhabitats and the protective spine canopies of co-resident urchins (Parechinus angulosus) for daytime protection. Populations of H. midae were discontinuously distributed along the coast, occupying small isolated reefs which offered a suitable array of microhabitats and a good food supply. They mostly inhabited shallow intertidal and subtidal reefs, but were occasionally encountered on deeper subtidal reefs at 4 - 5 meters. Mean length- and width-at-age were determined from growth rings composed of alternate conchiolin (dark) and aragonite (white) bands in the internal nacreous shell layer. Growth was described by the Special Von Bertalanffy growth equation: Lt(mm) = 176.998918 (1 - e⁻°·²⁴²⁴¹⁹⁽t ⁺ °·⁴⁹⁵⁴⁹⁴⁾) Wt(mm) = 159.705689 (1 - e⁻°·¹⁹⁵⁴³⁹⁽t ⁺ °·²¹¹⁶⁾) The ageing technique used was validated for animals from Great Fish Point and Mgwalana using independent tag-return data. The same data provided evidence that growth rates varied between animals from Great Fish Point and Bird Island. The growth data also showed that H. midae exhibited a high degree of individual variation in growth rate. Males and females exhibited similar growth rates. Exposed large animals showed a preference for red seaweeds, in particular Plocamium corallorhiza and Hypnea spicifera, while small sub-boulder cryptic animals included larger proportions of brown (Ralfsia expansa) and green (VIva spp.) algae in their diets. Exposed individuals also exhibited a higher degree of selectivity towards prey items, but in general, stomach contents reflected the most abundant seaweed types. Both drift and attached algal species were utilized by H. midae which was a nocturnal feeder. Pigments from red algae were incorporated into the shell layers giving the shells a pink or brick red colour. Haliotis midae is a dioecious broadcast spawner. Gonad Bulk Indices in combination with detailed histological examination of gonads showed that individuals were iteroparous, asynchronous spawners and that the breeding season extended from March through to October, although the peak spawning activity was between April and June. Males and females can spawn partially, totally or not at all, with atresia of residual gametes occurring after spawning. There is no resting stage, and gametogenesis is initiated directly after spawning. The structure of the ovary and testis and the process of gametogenesis is typical of haliotid species. AI: 1 sex ratio was observed from all populations studied. Sexual maturity was first attained in the 40 - 59 mm SL size class, although evidence for the smallest size at first spawning was recorded at 54.6 mm SL for females and 69 mm SL for males. Sizes at 50% sexual maturity were 72.5 mm SL (52.8 mm SW) at Great Fish Point, 72.5 mm SL (57.4 mm SW) at Mgwalana, 73.7 mm SL (51.2 mm SW) at Cape Recife, and 73.5 mm SL (53.8 mm SW) at Kelly's Beach. Haliotis midae was typically highly fecund, although a high degree of variation resulting in poor relationships between fecundity/shell length and gonad weight/shell length. The relationship between fecundity and gonad weight was linear. In the Eastern Cape, H. midae possessed a faster growth rate, smaller size at sexual maturity, smaller maximum size and lower longevity when compared to con specifics in Western Cape waters. A smaller minimum legal size of 93 mm SW is proposed for Eastern Cape animals and it is suggested that the closed season be moved to the peak spawning period between April and June. The benefit of a closed season during the spawning period is questioned, and the feasibility of closed areas as a management option for H. midae in the Eastern Cape is discussed.

Khellin) and research in this field has eventually led to the discovery of chromoglycic acid, which is widely used as a sodium salt in asthma therapy. Since biological activity may be related to acidity, a range of chromone-2-carboxylic acids have been prepared via Claisen acylation of substituted o- hydroxyacetophenones and their acid dissociation constants determined potentiometrically to explore substituent effects. From this study it has been found that introduction of certain groups does have a marked effect on acidity. A variety of acrylamide derivatives have been prepared via the dimethylamine-mediated ring opening of chromone-2-carboxamides which, in turn, were prepared from the chromone-2- carboxylic acids via the corresponding acid chlorides. Variable temperature NMR spectroscopy was employed to examine the effect of substituents on the rotational barriers and it has been found that for the acrylamides examined, ring substituents have little effect on the rotational barriers. A combination of low resolution, high resolution and meta-stable peak analysis has been used to study mass fragmentation patterns for a series of acrylamide derivatives. The proposed fragmentation pathways for selected peaks have been found to be common to all the spectra examined when differences in the atomic masses of substituents were taken into account.

Much work has been undertaken on developing the technology for the commercial culture of the species inclusive of determining the nutritional requirements for cost effective feed formulation. However, digestible energy (DE) values for specific feed ingredients were unknown. As DE values are not interchangeable between species, this study set out to determine the DE values for conventional and unconventional feed-ingredients for the formulation of an optimal feed. The ultimate goal in feed formulation is to produce a diet that supports the maximum production at the least cost. The indirect method was found to be most suitable in obtaining DE values for the catfish. Feed samples were marked by adding 2% chromic oxide as an inert indicator. The fish were then force-fed and faecal samples were obtained from the hindgut by stripping the fish 10 hours after feeding as this was found to be when the hind-gut was at its fullest and the feed maximally digested. A linear based computer programme was used to formulate experimental diets using the established DE values as an additional variable. The formulated feed was tested and compared to a commercial catfish diet and a standard reference diet (H-440). All of the newly formulated feeds were found to be cheaper to manufacture (by between 5 and 10%). Statistically (ANOVA), all of the experimental diets resulted in higher specific growth rates (SGR) than the standard reference diet. The commercial diet also resulted in a lower SGR than those experimental diets with similar protein levels.

- Description: Diarrhoeal disease is a major cause of mortality in children in developing countries. It also remains a serious problem among all age groups throughout the world. Whereas studies to determine the epidemiological and aetiological factors of diarrhoeal disease have been reported for other parts of South Africa and the world, as yet no information is available for the Eastern Cape. Therefore this study was undertaken to determine the factors for this area. Enteropathogens were compared for the different ages in the various population groups and, where possible, seasonal and geographical differences were emphasised. A total of 7 278 faecal samples were examined by six laboratories in the Eastern Cape during the period November 1988 to October 1990. Data was recorded noting the age, sex and population group of the patients. The towns selected were Port Elizabeth, Uitenhage, Cradock, Grahamstown and their surrounding areas. The isolation rates for the pathogens studied in the various population groups were compared to those reported in similar studies in other countries. The seasonal incidences of the various selected pathogens were compared with those reported from elsewhere in South Africa. It was thought that the higher temperature of summer may influence the finding in the White population group, while rainfall would play a greater role for the Coloured and Black populations. The geographical distribution of the pathogens emphasised the difference in living conditions between the different population groups. For example a generally higher infestation rate of Helminths occurred in rural areas and in the groups living under poorer conditions. The low isolation rates for certain bacteria and the large percentage of samples from which no pathogens were isolated indicate the need for further research. However, the finding should be valuable for determining Public Health priorities and in the management of outbreaks of diarrhoeal disease.

- Description: Diarrhoeal disease is a major cause of mortality in children in developing countries. It also remains a serious problem among all age groups throughout the world. Whereas studies to determine the epidemiological and aetiological factors of diarrhoeal disease have been reported for other parts of South Africa and the world, as yet no information is available for the Eastern Cape. Therefore this study was undertaken to determine the factors for this area. Enteropathogens were compared for the different ages in the various population groups and, where possible, seasonal and geographical differences were emphasised. A total of 7 278 faecal samples were examined by six laboratories in the Eastern Cape during the period November 1988 to October 1990. Data was recorded noting the age, sex and population group of the patients. The towns selected were Port Elizabeth, Uitenhage, Cradock, Grahamstown and their surrounding areas. The isolation rates for the pathogens studied in the various population groups were compared to those reported in similar studies in other countries. The seasonal incidences of the various selected pathogens were compared with those reported from elsewhere in South Africa. It was thought that the higher temperature of summer may influence the finding in the White population group, while rainfall would play a greater role for the Coloured and Black populations. The geographical distribution of the pathogens emphasised the difference in living conditions between the different population groups. For example a generally higher infestation rate of Helminths occurred in rural areas and in the groups living under poorer conditions. The low isolation rates for certain bacteria and the large percentage of samples from which no pathogens were isolated indicate the need for further research. However, the finding should be valuable for determining Public Health priorities and in the management of outbreaks of diarrhoeal disease.

Lateritisation and secondary gold distribution with particular reference to Western Australia

- Description: Lateritisation is associated with tropical climates and geomorphic conditions of peneplanation where hydromorphic processes of weathering predominate. Laterites are products of relative (residual) and absolute(chemical) accumulation after leaching of mobile constituents. Their major element chemistry is controlled by the aluminous character of bedrock and drainage. Bauxitisation is characterised by residual gibbsite neoformation and lateritisation, by both residual accumulation and hydromorphic precipitation of goethite controlled by the redox front at the water table. The laterite forms part of a weathering profile that is underlain by saprock, saprolite, the mottled zone and overlain by a soil horizon. The secondary gold in laterites has its source invariably with mineralised bedrock. The distribution of secondary gold is controlled by mechanical eluviation and hydromorphic processes governed by organic, thiosulphate and chloride complexing. The precipitation of secondary gold is controlled by pH conditions, stability of the complexing agent and ferrolysis. Gold-bearing laterites are Cainozoic in age and are best developed on stable Archean and Proterozoic cratons that have suffered epeirogenesis since lateritisation. Mechanical eluviation increases in influence at the expense of hydromorphic processes as a positive function of topographic slope and degradation rate. Gradients greater than 10⁰ are not conducive for lateritisation, with latosols forming instead. High vertical degradation rates may lead to the development of stone lines. In the Western Australian case, post-laterite aridification has controlled the redistribution of secondary gold at levels marked by stabilisation of the receding palaeowater table. Mineable reserves of lateritic ore are located at Boddington, Westonia and Gibson toward the south-west of the Yilgarn Block. A significant controlling variable appears to be the concentration of chloride in the regolith. Based on the Boddington model, the laterite concentrates the following elements from bedrock gold lodes: i) Mo, Sb, W, Hg, Bi and Au as mobile constituents. ii) As and Pb as immobile constituents. Geochemical sampling of ferruginous lag after bedrock and laterite has provided dispersed anomalies that are easily identifiable. "Chalcophile corridors" up to 150 km in length are defined broadly by As and Sb but contain more discrete anomalies of Bi, Mo, Ag, Sn, W, Se or Au, in the Yilgarn Block. The nature of the weathered bedrock, the tabular distribution of secondary gold ore deposition and the infrastructural environment lends the lateritic regolith to low cost, open-cut mining. The western Australian lateritic-gold model perhaps can be adapted and modified for use elsewhere in the world.

Morphological variation and its taxonomic implications for insular populations of Pseudocrenilabrus philander (Pisces: Cichlidae)

- Description: The cichlid fish Pseudocrenilabrus philander is widely distributed in southern Africa. Many of the populations occur in small, insular, geographically isolated water bodies, some of which are in arid areas. These small allopatric populations have been isolated for thousands of years and gene flow between them is non-existent or severely restricted. Populations were found to be different in terms of coloration, size of individuals, sexual dimorphism and behaviour. This thesis involved the determination of the taxonomic status of these isolated populations, from a morphological point of view. This has been part of a larger project, involving genetic and behavioural studies, to determine whether the different populations are geographic races of a single species, or whether they are species. Heritability of the observed differences was tested by breeding through three generations under uniform laboratory conditions. The populations from which wild-caught individuals were drawn could be identified on the basis of colour. There were some differences in anatomy between populations, but none of these were entirely distinctive for a particular population. When bred under laboratory conditions, populations did not show a tendency towards uniformity, as would be expected if morphological differences were environmentally induced. However, there were slight changes in the oral and pharyngeal bone form which may be diet related. Although there are morphological differences between populations and between wild-caught and laboratory-bred populations, it is suggested that the populations of P. philander are not fully differentiated and thus in the stages of incipient speciation. other studies on P. philander have introduced an element of uncertainty in that they show different results. Behavioural work suggests that different populations would behave as different species if they were to become sympatric. Karyological and mitochondrial studies showed that there were no differences between populations. Protein electrophoresis showed that populations were genetically unique. Since the various species and subspecies of Pseudocrenilabrus have been based on morphological criteria, the approach and conclusions in this study are based entirely on morphological criteria. These studies have practical implications for conservation, as some of the small populations are threatened with extinction. If the differences between the populations were ecophenotypic (i.e. related to their environment), then threats to some populations would not affect the conservation status of the species as a whole. If, however, such differences were genetic (i.e. the populations have evolved separately), then the extinction of small, isolated populations could mean the loss of actual species. This study strongly suggests that populations are incipient species and thus should be conserved.

Much controversy exists regarding the efficacy of the drug as an anorectic agent, the related adverse reactions caused by the relatively high doses required for appetite suppression and the potential of this drug for abuse. Whilst numerous studies have been carried out to assess the central and cardiovascular safety of PPA and many investigations have been performed to evaluate efficacy in terms of weight loss in humans, there is a relative paucity of information regarding the effects of PPA on appetite and food intake. A pilot trial was conducted to determine the feasiblility of a multidimensional approach to evaluate the safety and efficacy of PPA as ananorectic agent in humans. Eight normotensive caucasian women who were overweight participated in a randomised double-blind cross-over comparison of PPA (75 mg) and placebo and were dosed to steady-state on a 12-hour fixed-dose schedule for a period of eleven weeks. Aspects of efficacy evaluated included the effects of PPA on hunger, appetite and satiety,salivation, macro-nutrient food intake and body weight. Standardised scales were used to quantitatively assess the possible subjective mood and behavioural reinforcing effects of PPA. Supine systolic and diastolic blood pressures were monitored continually throughout the trial. In addition, peak and trough blood samp1es were taken to monitor serum concentrations of PPA reached at steady-state and patient compliance with the dosing schedule. An adaptation of a published reverse-phase high performance liquid chromatographic (HPLC) assay for PPA in serum using U.V. detection at 210 nm is presented. A significant decrease in body weight, salivation, total food intake and carbohydrate consumption was demonstrated following PPA administration. Phenylpropanolamine produced significant decrements in subjective reports of hunger and appetite, whilst apparently having little effect on satiety. No significant changes were observed for blood pressures and PPA did not produce significant mood alterations or behavioural reinforcing effects. The study demonstrates the feasibility of using this muti-faceted approach, with certain design modifications, to evaluate the overall safety and efficacy of PPA as an appetite suppressant.

The adrenal glands play a central role in maintaining homeostasis. The major neuroendocrine consequence of long-term stress is elevated circulating glucocorticoid levels. In this study, the effect of chronic, oral hydrocortisone treatment on pineal biochemistry was investigated in male Wi star rats of the albino strain. The results show that seven days of oral hydrocortisone treatment endows the pineal gland with the ability to increase melatonin synthesis in organ culture. The increase is accompanied by a rise in NAT activity, cyclic AMP levels and enhanced specific binding to the pineal B-adrenergic receptors. It appears that hydrocortisone sensitizes the pineal gland to stimulation by B-adrenergic agonists. thus rendering the pineal more responsive to B-adrenergic agonists. Further studies were directed at demonstrating an anti-stressogenic function for the pineal gland by investigating whether the principal pineal indole, melatonin. could protect against the deleterious effects of elevated. circulating drocortisone levels. The results show that chronic, oral hydrocortisone treatment significantly increases liver tryptophan pyrrolase activity. The catabolism of tryptophan by tryptophan pyrrolase is an important determinant of tryptophan availability to the brain, and therefore, brain serotonin levels. The findings show that melatonin inhibits basal and hydrocortisone-stimulated liver tryptophan pyrrolase apoenzyme activity in a dose-dependent manner. This inhibition suggests that melatonin may protect against excessive loss of tryptophan from circulation and against deficiencies in the cerebral serotinergic system which are associated with mood and behavioural disorders. The results show that 10-4 M glutamate significantly inhibits the isoprenaline-stimulated synthesis of N-acetylserotonin and melatonin in organ culture when the pineal glands were pre-incubated with glutamate for 4 hours prior to stimulation with isoprenalin and when glutamate and isoprenaline were administered together in vitro. GABA, a glutamate metabolite could not mimic the decrease in isoprenalinestimulated melatonin, and it is likely that the observed effects were directly attributed to glutamate. Incubation of the pineal gland with 10-4 M glutamate in organ culture did not affect HIOMT activity in pineal homogenates, but significantly elevated both basal and isoprenaline-stimulated NAT activity. It was concluded that glutamate only inhibits melatonin synthesis in intact pineal glands and not in pineal homogenates. The present study has provided further support for an interaction between the pineal and the adrenal glands. There is an ever increasing likelihood that melatonin is an anti-stressogenic hormone and that the pineal gland may have a protective role to play in the pathology of stress-related diseases.

In view of the neurotoxic effect of glutamate in the eNS, the functional significance of recently described glutamate receptors in the pineal gland was investigated. The results show that 10-4 M glutamate significantly inhibits the isoprenaline-stimulated synthesis of N-acetylserotonin and melatonin in organ culture when the pineal glands were pre-incubated with glutamate for 4 hours prior to stimulation with isoprenalin and when glutamate and isoprenaline were administered together in vitro. GABA, a glutamate metabolite could not mimic the decrease in isoprenalinestimulated melatonin, and it is likely that the observed effects were directly attributed to glutamate. Incubation of the pineal gland with 10-4 M glutamate in organ culture did not affect HIOMT activity in pineal homogenates, but significantly elevated both basal and isoprenaline-stimulated NAT activity. It was concluded that glutamate only inhibits melatonin synthesis in intact pineal glands and not in pineal homogenates. The present study has provided further support for an interaction between the pineal and the adrenal glands. There is an ever increasing likelihood that melatonin is an anti-stressogenic hormone and that the pineal gland may have a protective role to play in the pathology of stress-related diseases.

It is therefore necessary to consider all these components when assessing the rehabilitation, although in the initial stages the successful revegetation of the disturbed areas is the most important criterion. Richards Bay Minerals, on whose mining site this project was carried out, is dredge mining heavy minerals on the north coast of Natal, where they have rehabilitated mined areas since 1978. This project has been carried out to establish: 1) The success of their dune forest rehabilitation using quantitative techniques. 2) The available seed bank in their rehabilitation stands. 3) The similarities in the succession taking place in rehabilitation stands compared to the revegetated stands in the vicinity of Richards Bay. 4) The best methods for creating alternative vegetation communities, especially grasslands, with a high species diversity on the mined tailings. This study reviews only the success of rehabilitation of the natural vegetation but other studies focusing on the insect, reptile, mammal and bird populations are also being undertaken by other researchers. No particular method of determining the success of vegetation rehabilitation has been chosen by restoration ecologists. Therefore in this study a broad range of quantitative techniques were used to show whether successional changes are occurring in the vegetation and physical environment. The results obtained from sampling the rehabilitated vegetation have shown that both the species richness and diversity are increasing as the returned vegetation matures. Levels of soil properties such as Sodium, Phosphate, Calcium and percentage organic matter have also risen with increasing stand age. Community complexity is also increasing with stand age, and TWINSPAN and DECORANA plots have separated out the differently aged stands based on their differences. A "pilot" study was done on the seed bank present in the rehabilitation stands. This has shown the presence of large amounts of early successional, mostly herbaceous species. Seeds of later successional and woody species were scarce which may be a result of the sampling intensity used. However seeds of late successional ground cover species were found in the older stands. Comparisons between the natural revegetation of disturbed areas in the vicinity of Richards Bay and the rehabilitation stands revealed similarities in both species composition and complexity. Species richness and diversity values are comparatively similar for the younger revegetated and older rehabilitation stands, and lWlNSPAN and DECORANA analysis techniques clustered the samples recorded from these areas in close proximity on their relative plots. The oldest revegetated sites contain a number of species found in the rehabilitated vegetation but as Acacia karroo has thinned-out in these stands many of these other species are now mature individuals. Attempts at rehabilitating an area of grassland at Richards Bay Minerals has not produced satisfactory species diversity and experimental manipulations were used to try and increase the diversity of the existing Eragrostis curvula dominated community. Of the several treatments used for the manipulation, a combination of burning and further topsoiling was the most successful in reducing Eragrostis importance and in increasing the species richness. Grassland topsoil spread directly onto the bare tailings produced an extensive vegetation covering over a short period but species richness was not significantly greater than for the existing Eragrostis dominated grassland, and further treatments and management needs to continue if this technique is to be employed. Only a limited amount of alien infestation of the rehabilitated areas was evident from the sampling undertaken during this research. As the removal of alien plants is an ongoing process and the rehabilitation stands are continuously monitored to identify any new invaders, this is not expected to become a problem. From the results of work done overseas and the rehabilitation carried out in South Africa it appears that it is possible to return natural vegetation communities on mined areas. That this is a lengthy process is to be expected but by manipulating the vegetation and continuously monitoring the process it may be possible to speed up development. Areas in need of further research have been identified based on the findings of this project. This will help to reinforce the undertaking of management proposals that will enhance the vegetation recovery and the success of the rehabilitation programme.

The biological control of Hakea sericea Schrader by the Hakea seed-moth, Carposina autologa Meyrick, in South Africa

- Description: Hakea sericea Schrader was introduced to South Africa from Australia and has become a major problem in nearly all the coastal mountain ranges of the Cape Province. The hakea seed-moth, Carposina autologa Meyrick was released in South Africa for the biological control of H. sericea. The impact of the moth on the canopy-stored seeds of H. sericea was evaluated at two study sites in the south-western Cape over three years. The moth has reduced the accumulated seeds at the two study sites by 59.4% and 42.6%, respectively. The moth has shown a surprising ability to disperse and establish new colonies at low population levels. Factors contributing to the slow colonization of C. autologa in South Africa was investigated. The moths appear to be unable to distinguish between healthy and previously attacked fruits; 42.5% of the eggs were laid on attacked fruits. Only 13.1% of the healthy fruits with eggs yielded mature larvae. The high pre-penetration mortality found in the present study is similar to that found in Australia. The effect of the indigenous fungus, Colletotrichum gloeosporioides (Penz.) Sacc., on both H. sericea and C. autologa was investigated. H. sericea trees and branches that die as a result of fungus cause the accumulated fruits on the affected trees or branches to dehisce. This seed loss occurs at a crucial stage during C. autologa larval development. Only 42.1% and 33.0% of the trees were found to be healthy at the two study sites, respectively. One seed crop will always be available for regeneration, since recruitment is linked to fires, and wild-fires occur at a stage when the latest seed crop has escaped attack by c. autologa. C. autologa was released at six sites in the south-western Cape by attaching egg-bearing follicles to healthy fruits in the field. Three release sites were evaluated the year following release to determine whether the moth established or not. The role of C. autologa in the H. sericea biological control programme is discussed. Although seed destruction by C. autologa is not severe, it is expected to contribute to the control of H. sericea.
