Evidence of Late Holocene extreme marine wave event (tsunami) deposits along the South African coast and implications for coastal zone management

- Description: Although extreme marine wave events such as tsunamis and storm surges have always been part of human existence, their impact on ever-growing coastal population of the world’s coast was highlighted by the recent catastrophic events over the past three decades. Although in situ observations and eyewitness accounts have improved our ability to record and interpret wave inundation by modern extreme marine events, records documenting inundation by pre-historic and historical events are relatively sparse to provide reasonable information on their distribution, magnitude and frequency intervals in most parts of the world. Owing to consequences these natural coastal hazards have on coastal population and infrastructure of the world’s coast, onshore sedimentary deposits preserved in the geological records have been extensively utilized by scientists worldwide to reconstruct the record of pre-historic and historical events and establish the recurrence interval of future events. Although studies investigating inundation of past tsunami has increased worldwide over the past two decades, research on extreme marine inundation has received little attention along the South African coast, despite known inundation by recent events and potential source mechanisms. Therefore, the aim of this study is to investigate sedimentary evidence of past tsunamis and related extreme marine wave events (i.e., storm surges) preserved in the recent geological archive of several coastal environments along the South African coast. The Swartkops, Kabeljous, Kromme and Keurbooms estuaries located in log-spiral bays along the south-east coast and sheltered coastal systems of the Berg Estuary and Verlorenvlei as well as coastal lowlands of the Dwarskersbos on the west coast were selected as the study sites. The sediment cores were collected from these coastal environments and identification of extreme marine wave inundation was based on the recognition of distinct anomalous sand layers. To characterise sediment transport and reconstruct provenance source origin of the deposits, a combination of sedimentological (i.e., grain size distribution), exoscopy (i.e., quartz grain surface microtextures) and micropaleontological (i.e., foraminiferal assemblages) proxies were utilized. Furthermore, age estimation of the deposits was determined using a combination of radiocarbon (14C) and radiometric (137Cs and 210Pb) dating methods.Evidence of coastal flooding by high-energy marine wave events is found preserved in the stratigraphic records of Swartkops and Kabeljous estuaries along the south-east coast and lowlands of Dwarskersbos on the west coast in the form of anomalous sand deposits (layers). The anomalous deposits of Swartkops and Kabeljous estuaries generally consist of gravel-sized shell fragments often with a sharp erosive contact suggesting deposition by a highly energetic marine wave event. Although the preservation varies between cores, a maximum of three anomalous sand depositional layers are preserved in the Swartkops whereas only a single layer in the Kabeljous Estuary with a basal layer of calcrete deposits. 210Pb and 14C dating, constrain the ages of the younger deposits (i.e., the uppermost layer) at Swartkops between 1770-1804 AD and 339-3 cal. BP (1611-1947 cal. AD), respectively. Although these ages span over a wide period, they are interpreted to represent evidence of one of the historical tsunamis generated in the subduction zones of the Indian Ocean e.g., 1762 AD and 1797 AD events generated by 8.8 and 8.0 Mw earthquakes in the Arakan and Sunda Subduction Zones, respectively. Furthermore, the deposits could also represent geological evidence of the 1833 Mentawai events whereas evidence of 1883 Krakatau tsunami is not disregarded, since this event reportedly inundated Algoa Bay. On the other hand, the 14C dating constrain the ages of the second anomalous deposits between 1526-1253 cal. BP (424-697 cal. AD), whereas the older anomalous deposits preserved in the deeper sections of the Swartkops cores as well as the backwater channel of the Kabeljous Estuary are constrained to have been deposited around 4000 cal. BP. Although there is no known locally documented extreme marine wave event during this period, these deposits are interpreted to represent late and mid-Holocene tsunamis associated with mega-thrust subduction zones of the Indian Ocean whereas local submarine landslide generated event are also not disregarded entirely. On the west coast, three anomalous deposits representing extreme marine wave inundation are preserved in the coastal lowlands of the Dwarskersbos. Although the general sedimentary features and thickness vary between the deposits, the first anomalous deposits in the uppermost section of the cores consist of medium sand with scattered shell fragments whereas, the third anomalous deposits in the deeper section of the cores are generally coarser with gravel-sized shell fragments and sharp erosive basal contacts. By contrast, the second anomalous deposits consist of three successive sub-layers of yellow sand with variable sedimentary features and these layers represent successive wave inundation events. Although the sedimentary characteristics of these sub-layers varies, it was noted that the lowermost layers consist of coarser and less well sorted sediments often with basal erosive contact, suggesting that the first wave was more energetic than the two-succeeding waves. Furthermore, calibrated 14C dated shell fragments from the first and second anomalous deposits yielded ‘post-bomb’ ages suggesting deposition after 1950 AD. Given their position within the stratigraphic records, the two deposits are interpreted to represent first geological evidence of the 2008 west coast and 1969 Dwarskersbos events, respectively. On the other hand, the 14C dated shell fragments constrain the deposition of the third anomalous deposits between 315-0 cal. BP (1635-11950 cal. AD) and 358-0 cal. BP (1592-1950 cal. AD), respectively. The third anomalous deposits are interpreted to the 07 April 1620 event, which was described as ‘Two startling thunderclaps’ near Robben Island. Although initially associated to earthquakes, there are no records of an earthquake during this period, therefore, we postulate that the 1620 event could have been a similar event to that of 2008 that inundated the west coast region and/or an unknown event. While it is acknowledged that the absence of reliable source mechanisms poses a great challenge in the reconstruction of extreme marine wave events inundation along the south-east coast, the findings of exoscopy and micropaleontological analyses suggest that the deposits were transported to their depositional environments, although resultant microtextural features and occurrence of individual foraminiferal species and their taphonomical conditions is dependent on coastal setting. The south-east coast deposits consist of highly diverse foraminiferal assemblages, which are dominated by marine and marginal-marine taxa with relatively low abundances of estuarine-brackish species and variable taphonomical conditions. Furthermore, the mechanical-induced features such as percussion marks, fresh surfaces with sharp edges were still preserved on quartz grain surface of the Swartkops deposits, despite dominance by chemical-induced features such as dissolution. By contrast, grain surface microtextural signatures of the Dwarskersbos deposits are dominated by mechanical features such as fresh surfaces and v-percussion marks. Furthermore, the foraminiferal assemblages of the Dwarskersbos deposits consist of exclusively marine and marginal-marine species, suggesting that the sediments were sourced from shallow water environments as well as the adjacent beach. An efficient approach for the synthesis of dolutegravir and its analogue exploiting flow chemistry

- Description: Africa has the highest Human Immunodeficiency Virus (HIV) prevalence in the world and has invested heavily in running its HIV programmes and conducting research of medical significance. Despite efforts in combating this disease the production, distribution, cost, and availability of antiretroviral (ARV) generics remains a major problem as they are imported from China and India. Exploring the recently emerged “enabling technique”, namely flow chemistry in the production of APIs has gained a lot of attention. This study was devoted on exploring the application of flow chemistry on the synthesis of a newly approved anti-HIV drug dolutegravir (DTG); an integrase inhibitor with a high genetic barrier to resistance with a daily dosing scheduled compared to other ARVs and its third-generation inhibitor analogue, cabotegravir. Chapter one covers a comprehensive background and literature review of the HIV epidemic, an introduction of antiretroviral therapy as well as detailed dolutegravir and cabotegravir synthesis. A brief introduction of continuous flow technology with its advantages and disadvantages is discussed in this chapter. The efficient seven-step continuous flow procedure afforded dolutegravir and cabotegravir in improved reaction times and yields compared to the traditional batch procedure was demonstrated. The significant advantage of this flow process includes the reduction of the overall reaction time from step one to step seven, from prolonged 68 hours in batch to 34 minutes. The overall yield of each reaction step improved dramatically upon flow optimization. The yields of the second step (selective ester hydrolysis), fourth step (cyclization) and fifth step (amidation) increased from 64 %, 40 % and 33 % in batch to 98 %, 71 % and 100 % yield by HPLC respectively. Other than improved yields and residence times which was aided advantages of microreactor technology including intensive mixing, the flow process is also much better because it is cost effective. An evaluation of the effectiveness of area-based conservation interventions in avoiding biodiversity loss in South Africa

- Description: Counterfactual impact evaluation studies form an important evidence base for the effectiveness of conservation projects, programs, and policies (collectively referred to as conservation interventions). In South Africa, counterfactual impact evaluation methods have rarely been applied to local conservation interventions, and therefore evidence for the effectiveness of key strategic national conservation approaches is lacking. This study evaluated three area-based interventions that together aim to avoid the loss of areas most important for the persistence of biodiversity in the terrestrial realm as evidence towards the effectiveness of South Africa’s landscape approach to biodiversity conservation. The first intervention, South Africa’s National Protected Area Expansion Strategy (NPAES), set ambitious targets to double the extent of South Africa’s protected area network while ensuring that the expansion preferentially occurs in areas of under-represented biodiversity. The strategy was evaluated in terms of its effectiveness in guiding protected area expansion towards more equitable representation of South Africa’s biodiversity through an assessment of changes in indicators of protected area expansion decision-making before and after the implementation of the strategy. The second intervention is the use of maps of biodiversity priorities to guide land use change decisions outside protected areas. Impact was evaluated as avoided loss of Critical Biodiversity Areas (CBAs), which need to remain in a natural condition to meet in situ conservation targets for species, ecosystems, and ecological processes. Avoided loss in CBAs was benchmarked against avoided loss in protected areas, to contextualize the effectiveness of land use planning as a conservation intervention. Lastly, the effectiveness of stricter land use regulations for threatened ecosystems to reduce land conversion pressure on these ecosystems was evaluated. An exploration of the Participatory Guarantee System (PGS) facilitating the growth and certification of small-scale organic farmers: Giyani, Garden Route and Overberg case studies

- Description: In this study, I evaluated the Participatory Guarantee System (PGS) as a framework to support the growth and certification of small-scale organic farmers. I used the interpretivism and constructivism lenses to understand the PGS from the perspective of members of three PGS groups. The research was qualitative, treating the three PGS groups as case studies. I collected data using focus groups, semi-structured interviews, questionnaires, field notes and observations. The theoretical frameworks that guided the research and data analysis were Socio-ecological Systems (SES) based principles and the Multi-Level-Perspective (MLP) framework. The data were analysed using Atlas.ti content analysis software. The key results were grouped into four themes: capacity; market and logistics; learning process; and disturbance, shock and stress. The theme of ‘capacity’ dealt with the functioning of a PGS, where three categories were identified: group composition and capacity; resource limitations and institutional support; marketing and consumer education. The more diversity in age, education and culture of PGS stakeholders, the better performance and chances of continuity and propagation. PGS has limited capacity partly due to its voluntary nature and infancy. It needs institutional support to grow and be useful for members. Department of Agriculture extension officers knew little about organic farming, thus limiting their engagement and assistance to such farmers. The theme of ‘market and logistics’ looked at the various factors that influence the farmers’ market reach. The perception from the members was that there was minimal consumer awareness about organic agriculture and the PGS. The marketing of organic produce relied largely on the individual farmer’s efforts and ability to search for and secure markets. The theme of the ‘learning process’ was categorised into learning and knowledge exchange. Members of the PGS groups highly regarded the invaluable knowledge shared through farm visits and online platforms, such as their WhatsApp groups. This knowledge improved and informed their farming techniques and led to the growth of v their skill base and experimentation. They viewed the way knowledge was exchanged as non-threatening and highly useful. The fourth theme of ‘disturbance, shock and stress’ dealt with the effects of COVID19. The three groups were all affected by lockdown restrictions (at the time of data collection) and were initially cut off from their usual markets. This led to a loss of income, loss of farm staff and a loss of the desired growth of the farms. - Description: In this study, I evaluated the Participatory Guarantee System (PGS) as a framework to support the growth and certification of small-scale organic farmers. I used the interpretivism and constructivism lenses to understand the PGS from the perspective of members of three PGS groups. The research was qualitative, treating the three PGS groups as case studies. I collected data using focus groups, semi-structured interviews, questionnaires, field notes and observations. The theoretical frameworks that guided the research and data analysis were Socio-ecological Systems (SES) based principles and the Multi-Level-Perspective (MLP) framework. The data were analysed using Atlas.ti content analysis software. The key results were grouped into four themes: capacity; market and logistics; learning process; and disturbance, shock and stress. The theme of ‘capacity’ dealt with the functioning of a PGS, where three categories were identified: group composition and capacity; resource limitations and institutional support; marketing and consumer education. The more diversity in age, education and culture of PGS stakeholders, the better performance and chances of continuity and propagation. PGS has limited capacity partly due to its voluntary nature and infancy. It needs institutional support to grow and be useful for members. Department of Agriculture extension officers knew little about organic farming, thus limiting their engagement and assistance to such farmers. The theme of ‘market and logistics’ looked at the various factors that influence the farmers’ market reach. The perception from the members was that there was minimal consumer awareness about organic agriculture and the PGS. The marketing of organic produce relied largely on the individual farmer’s efforts and ability to search for and secure markets. The theme of the ‘learning process’ was categorised into learning and knowledge exchange. Members of the PGS groups highly regarded the invaluable knowledge shared through farm visits and online platforms, such as their WhatsApp groups. This knowledge improved and informed their farming techniques and led to the growth of v their skill base and experimentation. They viewed the way knowledge was exchanged as non-threatening and highly useful. The fourth theme of ‘disturbance, shock and stress’ dealt with the effects of COVID19. The three groups were all affected by lockdown restrictions (at the time of data collection) and were initially cut off from their usual markets. This led to a loss of income, loss of farm staff and a loss of the desired growth of the farms. Solanum incanum is among the plants utilised to control cabbage aphids in Mkoba village, Zimbabwe. Solanum species are known for their steroidal compounds which comprise glycoalkaloids and saponins. This study evaluated the knowledge, opinions, and attitudes of the vegetable peasant farming community in Gweru regarding their use of the indigenous plant (S. incanum) as a pesticide. The study also reported the phytochemical profiling, structural characterisation of the isolated compounds, and biological and pesticidal activity evaluation of phytochemicals isolated from S. incanum. A descriptive survey was carried out to assess the knowledge, attitude, and practices of a conveniently sampled group of vegetable farmers in Mkoba village who use S. incanum as a pesticide. Forty-nine respondents comprised of 19 males and 30 females of ages ranging from 15 to above 60 years took part in the study by answering an open and closed-ended questionnaire. The survey revealed that parents and neighbours were instrumental in disseminating pesticidal information in the community. Brassica napus were the most grown.vegetable and vulnerable to cabbage aphids. Mixed opinions amongst the respondents varied regarding the health and environmental impact of S. incanum as a pesticide. Seventy-five percent (75%) of the respondents supported the use of S. incanum as a pesticide whilst 25% claimed that the use of S. incanum was the source of the health problems experienced in the community. The survey demonstrated that (45)91% of the farmers displayed poor practices regarding the disposal of empty pesticide containers and the use of personal protective clothing. - Description: Creep-resistant 1CrMoV steels are used for high-pressure turbine rotors in fossil fuel power stations operating at steam temperatures up to 565 °C in the creep regime. The upper bainitic microstructure of 1CrMoV rotors changes when exposed to high temperature service over long periods. The aim of this study was to relate these microstructural changes for service exposed 1CrMoV rotor steels to the remaining creep life as determined using existing methods. Analytical electron microscopy was used to quantitatively study the microstructural evolution of service-exposed 1CrMoV steel rotors. 1CrMoV rotor steel samples were characterised in the initial state, and three long-term (272 h to 300 kh) service exposed states. Detailed microstructural investigations were conducted to analyse the cavities; bainitic laths and packets; dislocations; carbide phases and their size, population, composition. The creep life fractions were estimated using conventional life assessment methods based on creep cavitation and hardness. The creep life fraction consumed of the rotors, estimated based on the creep cavities and hardness data, ranged from ~0.5 to 1. Electron backscattered diffraction (EBSD) was used to analyse the bainitic packets, laths, misorientations, and dislocations, which showed that the bainitic lath surface area to volume (SV) correlated with service time. The precipitates were extracted from the Fe-matrix using extraction replication and were further investigated using transmission Kikuchi diffraction (TKD), scanning transmission electron microscopy (STEM), selected area electron diffraction (SAED), and energy dispersive spectroscopy (EDS). STEM-EDS and TKD identified M3C, MC, M2C, M7C3, and M23C6 (M = Fe, Cr, Mo, V, Mn) in the 1CrMoV steels. STEM-EDS measurements revealed that Fe/Cr ratios in M3C decreased from 4.5 (initial state) to 2.5 (>272 kh). The Fe/Cr and Fe/Mo ratios in overall carbides decreased from 5 and 10 (initial state) to 1 and 2 (>272 kh), respectively. The TKD analysis indicated that the relative phase proportion of M2C and M7C3 carbides, relative to the total extracted carbides, is positively correlated with service exposure. The effect of Cannabis extract on the morphological and metabolic characteristics of various fat depots in diet-induced Obese and STZ-induced male wistar rats

- Description: To investigate the potential anti-diabetic/obesity properties of oral cannabis administration in an obese and streptozotocin (STZ)-induced diabetic rat model, as well as an obese rat model, and to determine the mechanism of action, with a focus on the peritoneal and intramuscular fat depots. Experimental Design: Obese and STZ-induced diabetic rats were allocated a high fat diet (HFD) and intraperitoneally injected with STZ to mimic an obese and diabetic state. The rats were then orally administered cannabis extract (CE) of 1.25, 2.5 and 5.0 mg/kg body weight (relative to tetrahydrocannabinol (THC) content) or metformin as a positive control. For the obese rat model, the rats were allocated either a high carbohydrate diet (HCD) or high fat diet (HFD) and orally administered with cannabis extract of 1.25 mg/kg body weight (relative to THC content). Weight, blood and insulin-resistant parameters of the rats were monitored. The mitochondrial to genomic DNA ratio (MT:18S DNA), average adipocyte area of the various adipose tissues, citrate synthase and carnitine palmitoyltransferase 1 (CPT1) enzyme activities of the peritoneal and intramuscular fat were measured. Gene expression levels of uncoupling protein 1 (UCP1), cell-death inducing DNA fragmentation factor alpha like effector-a (Cidea), perilipin, hormone-sensitive lipase (HSL) and mitochondrial transcription factor A (TFAM) were measured in peritoneal fat, intramuscular fat and brown adipose tissue (BAT). Main Findings: Obese and STZ-induced diabetic rat model: Due to the biphasic nature of cannabinoids, cannabis dosage plays an important role in the observed effects. CE1.25 was the only cannabis treatment effective in improving the insulinresistant parameters of the rats unlike the other higher cannabis concentrations (CE2.5 and CE5.0). In the peritoneal fat, CE1.25 increased MT:18S DNA, increased citrate synthase activity, and decreased the average adipocyte area when compared to the STZ group. CE1.25 also induced fat beigeing by upregulating gene expression levels of UCP1 and Cidea. XIX Furthermore, an increase in gene expression levels of perilipin, HSL, and TFAM showed increased fat mobilization and metabolic activity. In the intramuscular fat, CE1.25 also reduced the average adipocytes area. Use of flow reactors for an improved synthesis of Tenofovir derivatives

- Description: Tenofovir disoproxil fumarate (TDF) is a prescription drug used to treat and prevent human immunodeficiency virus (HIV) infections. It aids in the reduction of HIV in the body, allowing the immune system to function more effectively. This reduces the risk of HIV complications, such as infections, whilst improving the quality of life. TDF belongs to the nucleotide reverse transcriptase inhibitors category of drugs. These drugs limit the ability of the reverse transcriptase, an enzyme essential for each virus to replicate itself. A reduction in the cost of HIV treatment could improve supply security and make it more accessible to patients in need. Thus, this project aims to use flow technology to achieve a guaranteed supply chain and access. In this thesis, a method for the synthesis of TDF was developed using microreactors starting from the readily available adenine. The method was adapted from the traditional batch synthesis as currently there is no literature on the synthesis of TDF in flow chemistry. The batch methods used for the synthesis of the working standards were successfully translated to flow systems. For the best results to be obtained, optimisation of the reaction temperature, reagent molar equivalents and concentration was critical. From the optimisation studies, an improvement in conversion is evident throughout all stages. The flow synthesis of the first intermediate resulted in a conversion of 100 % in N,N-dimethylformamide in 6 min residence time. This study was conducted at 200 °C, which is higher than the boiling point of the solvent with the aid of a back pressure regulator. A key reagent in the synthesis of tenofovir, namely ((tosyloxy)methyl)phosphonic acid, had to be synthesised and it was successfully produced in 96 % yield. The second intermediate was synthesised through a successful coupling reaction between (R)-9-(2-hydroxypropyl)adenine and ((tosyloxy)methyl)- phosphonic acid, which afforded a 100 % conversion of tenofovir in 20 min total residence time. The multistep synthesis of tenofovir was a success as it afforded a 65 % overall conversion. The prodrug TDF was successfully synthesised at 120 °C with a 30 min residence time, yielding a conversion of 100 %. ii | P a g e This thesis consists of four chapters. Chapter one describes the background on HIV and a literature review on HIV drugs. - Description: Globally social media has shown unprecedented levels of adoption and Social Media Analytics (SMA) is a rapidly growing topic. For governments, SMA holds the promise of providing tools and frameworks to collect, monitor, analyse and visualise social media data, usually driven by specific requirements from a target application. However, social media data is noisy and unstructured, and organisations struggle to extract knowledge from this data, and convert it into actual intelligence. This study argues that SMA can support intelligent decision-making for Citizen Relationship Management (CzRM). CzRM is a growing effort of governments around the world to strive to respond rapidly to their citizens by fostering a closer relationship thereby creating more effective and efficient service delivery. However, there is a little evidence in literature on empirical studies of any existing decision-making framework for CzRM and SMA adoption. In particular, there is a gap with regards incorporating SMA into decision-making for CzRM of governments, particularly in developing countries like South Africa. The aim of this study was to develop a framework that provides guidelines, including methods and tools, incorporating SMA into decision-making for CzRM in the Gauteng Provincial Government (GPG) and the Free State Provincial Government (FSPG) of South Africa. A Systematic Literature Review (SLR) and conceptual analysis method was conducted to design the Social Media Analytics Framework for Decision-making in the context of CzRM (the SMAF). The findings from the literature review revealed several benefits and challenges with SMA, in particular the shortage of skills, guidelines, methods and tools for SMA. These challenges were used to draft guidelines that were included in the framework, which consists of five components that can be used to derive intelligent information from SMA. The pragmatic philosophy and a case study design was used to generate an in-depth, multifaceted understanding of the underlying problems in the case of the GPG and the FSPG. The German North-West Metropolitan region was used as a third case study to provide a more global perspective and a case of a developed country in terms of Gross Domestic Product. The scope of the study was limited to social media posts by provincial citizens related to CzRM and service delivery. Both formative and summative evaluations of the proposed theoretical framework were conducted. - Description: Globally social media has shown unprecedented levels of adoption and Social Media Analytics (SMA) is a rapidly growing topic. For governments, SMA holds the promise of providing tools and frameworks to collect, monitor, analyse and visualise social media data, usually driven by specific requirements from a target application. However, social media data is noisy and unstructured, and organisations struggle to extract knowledge from this data, and convert it into actual intelligence. This study argues that SMA can support intelligent decision-making for Citizen Relationship Management (CzRM). CzRM is a growing effort of governments around the world to strive to respond rapidly to their citizens by fostering a closer relationship thereby creating more effective and efficient service delivery. However, there is a little evidence in literature on empirical studies of any existing decision-making framework for CzRM and SMA adoption. In particular, there is a gap with regards incorporating SMA into decision-making for CzRM of governments, particularly in developing countries like South Africa. Moult and energetics in three species from the Euplectes genus representing a gradient of elaborate plumage ornamentation

The evolution and persistence of elaborate and costly ornamentation seemed to be in opposition to the theory of natural selection. However, over the years research has revealed the role and functional significance of elaborate plumage ornamentation in sexual selection. Recent studies have shown that the energetic costs associated with moulting into new plumages are higher than maintenance costs, but these costs have not been quantified for species that exhibit elaborate breeding plumage ornamentation for sexual selection. In addition, for these sexually selected ornamental traits (signals) to have evolved and be maintained, there must be some aspect of individual quality that they convey to the receiver. Despite a large body of literature on sexual selection, honest signalling and ornamental plumage in birds, the physiological correlates of honest signalling during sexual selection and the messages these signals convey to the receivers are poorly understood. In this study, I investigated four aspects related to the honesty of the sexual selection signal. These were (i) seasonal variation in standardised resting metabolic rates of breeding and non-breeding birds, (ii) moult phenology, (iii) physiological parameters linked to honest signalling, and (iv) the energetic costs of production and maintenance of elaborate plumage ornamentation using three species from the Euplectes genus as model species. The three species selected, the red-collared widowbird (Euplectes ardens), fan-tailed widowbird (Euplectes axillaris) and the southern red bishop (Euplectes orix) represent a gradient of plumage elaboration and although the receivers of the sexual selection signals are known for these three species, the energetic cost of the signals and the underlying message the signals convey remain unknown. I measured seasonal variation in resting metabolic rate (RMR), body mass (Mb), thermoneutral zone (TNZ) and body temperature (Tb) over the course of two years in the three selected Euplectes species (E. ardens, E. axillaris and E. orix; chapter 2). Spatial and temporal analysis of the critical zone in the Western rift valley corridor: towards earth stewardship science in East Africa

- Description: Over the coming decades, Africa is expected to experience disturbing effects due to climate change and increased land cover change due to human influences presenting a significant concern for the future well-being of human and biological systems, the latter being the foundation of ecosystem services supplied to humanity. Therefore, unprecedented transdisciplinary cooperation, coordination, and integration amongst researchers, government, and civil society are necessary to increase the resiliency of these systems. This study aims to provide an outline of the Africa Alive Corridors (AAC) as an essential model for the encouragement of sustainable development through Earth Stewardship science. These aims are accompanied by the quantification and forward modelling for land cover change of the Critical Zone over 10 Great Lake Basins across one of the AAC, the Western Rift Valley Corridor (WRVC), in East Africa between the years 2018 and 2060. This approach provides the foundation for implementing improved regional governance, better encouragement of sustainable development beyond the 2030 United Nations Sustainable Development Goals, and education programs, such as the AAC, that promote socio-ecological resilience through Earth Stewardship Science. The AAC archives a portion of East Africa as the WRVC, a corridor along the western branch of the East African Rift System that highlights twenty heritage nodes, primarily great lakes, mountain ranges, national parks, and biological hotspots. These heritage entities are associated with ca. 12-million-years of evolution and transformation of the East African topography and related African Great Lake (AGL) systems. The thesis defines the study area by delineating AGL basins intersected by the WRVC. Across these basins, land cover change analysis provides a platform for an integrated assessment of the projected health of the corridor region. Existing land cover datasets provide the initial conditions of the study area for 2008 and 2013. Land cover between 2008 and 2013 is cross-tabulated using the Land Cover Module in the Terrset software, followed by the iii delineation of sub-models and driver variable identification. The Multi-Layer Perceptron algorithm provides the transition potentials between tree cover, urban area, cropland, wetland, and open area classes. Change quantification and prediction using Markov Chain analysis are then established for 2018, 2030, and 2060. The model successfully simulated future land cover change and concluded that: (1) proximity to existing human activity, proximity to existing tree cover, and population are the primary drivers of change; (2) the dominant land cover of the ten lake basins for 2018 was cropland at ca. 48%, followed by tree cover at ca. 33%; (3) total anthropogenic change over the coming four decades equates to over ca. 52 000 km2 (5 200 000 ha), and particularly (4) an urban area is expected to increase by >130%. This assessment ultimately provides a platform for regional governance development at the basin scale and Earth Stewardship science in East Africa. These changes require transdisciplinary action from researchers to civil society. At the same time, the fifth generation (5G) of mobile communication systems are also being researched and developed. Throughout history, mobile communication networks are known to affect the activities of radio astronomy. If not carefully managed, radio frequencies from mobile communication devices can severely affect radio astronomy observations. To that end, many techniques have been proposed to protect the radio astronomer from RFIs coming from radio communication networks. Some of the proposed techniques such as RFI quite zones and spectrum assignment by regulatory authorities will not be convenient during the implementation of 5G mobile networks. This is because 5G radio communication systems are expected to support spectrum-hungry application such as video-on-demand, augmented realities, high-definition television and so on. To realize this, the 5G networks will be forced to have access to protected radio spectrum, including those at which radio astronomy activities are being researched. To facilitate this, the 5G radio communication networks should have the intelligence to coexist within such protected spectrums without the consequences of radio frequency interferences (RFI) to the primary user. In this thesis, we present novel 5G networks with the intelligence that allow them to coexist within radio astronomy areas without introducing RFIs to the primary user. We proposed a photonic solution, keeping in mind the characteristic requirements for future 5G radio communication networks. The thesis begins by reviewing the current trend of radio astronomy research in Africa. It was found that radio astronomy research in Africa is growing rapidly. Many African countries such as South Africa and Ghana are at advanced stages when it comes to radio astronomy research. Therefore, the finding and proposal of this thesis will be valuable to such countries. In order to develop a radio access network (RAN) that can coexist within radio astronomy areas, the thesis reviewed past and present state-of-the-art RANs. Each access network was analyses for its feasibility to be implemented within radio astronomy areas to realize mobile communication without the consequences of RFIs to the astronomer. It was motivated that the current centralized radio access network (C-RAN) the best solution to be developed for radio communication within radio astronomy areas. This is because the C-RAN architecture is centralized by pooling network resources to a common point. From such pool, network resources can be controlled and shared among 5G network user, including radio astronomers and the surrounding communities. However, the phytochemistry, safety and efficacy of these medicinal plants remain less investigated. Based on available traditional medicinal knowledge (through traditional healers, university library resources, ethnobotanical and science journals and other internet resources), traditional uses and availability, three plant species, namely Bulbine latifolia L. Wild, Dicerothamnus rhinocerotis Koek. and Olea europaea subsp. africana (Mill.) P.S. Green were selected and investigated for their hytoconstituents and biological effects. Phytochemical screening of the three medicinal plants was carried out quantitatively and qualitatively using spectrophotometric methods. Chemical profiling of the crude methanol extracts of all the plant species was successfully done by high-performance thin layer chromatography (HPTLC) and tentative identification of the secondary metabolites was achieved by liquid chromatography–mass spectrometry (LCMS). Cytotoxicity effect investigations on the crude extracts of the three species was done using the 3-[4,5- dimethylthiazole-2-yl]-2,5- hiphenyltetrazolium bromide (MTT) assay against HeLa cervical cancer cells. Isolation and purification of single compounds was done using chromatographic techniques (column chromatography (CC), and preparative thin-layer chromatography (PTLC)). Characterisation of single compounds was achieved using spectroscopic techniques such as Nuclear Magnetic Resonance (NMR), High Resolution Mass Spectrometry (HRMS), Ultra Violet Spectroscopy (UV-vis), Fourier-Transformed Infra-Red Spectroscopy (FTIR) and Single-Crystal X-ray Diffraction (XRD). For the first time from the methanol crude extract of the tubers of B. latifolia, the crystal structure of the phenylanthraquinone, knipholone (3.12) (1-(3-acetyl-2,6- dihydroxy-4- methoxyphenyl)-4,5-dihydroxy-2-methylanthraquinone), was isolated and elucidated along with its derivatives, isoknipholone (3.14), knipholone-6’-methyl (3.27). - Description: Anthropogenically-derived eutrophication and Harmful Algal Blooms (HABs) are an increasing threat to estuarine ecosystems globally. The rapid population growth in coastal areas and the mismanagement of water resources have led to the rapid increase in eutrophication in estuaries. Consequently, HABs have become frequent phenomena with severe impacts on the ecological functioning of these systems. A paucity exists in understanding the specific mechanisms of how HABs impact estuarine communities globally. This has not been studied in South Africa, despite the observed increase in the occurrence of HABs in the past few decades. Therefore, this study investigated the impact of HABs on the ecological functioning of the anthropogenically altered Sundays Estuary, South Africa, which is affected by predictable and recurrent HABs of the raphidophyte, Heterosigma akashiwo. This study found that a high density (> 1000 cell m L-1 ) of H. akashiwo was present during all but the winter months in the poly- to mesohaline zone, yet it peaked in warmer spring/summer months with a density of ca. 70 000 cell m L-1 corresponding to 800 µg Chl-a L -1 . This species significantly altered the estuary's water quality, causing supersaturated surface- and hypoxic bottom-water during severe HABs or referred to as hypereutrophic bloom phases (> 60 µg Chl-a L -1 ). Despite these changes, the common copepod, Pseudodiaptomus hessei, occurred at similar densities among HAB phases and responded to similar triggers to the HAB. However, a shift in predator-prey interactions within the zooplankton community was evident, with scavenger or opportunist species such as the isopod, Cirolana fluviatilis, and the polychaete, Perinereis falsovariegata, occurring at higher densities during hypereutrophic bloom phases. The overall abundance of juvenile and small adult fishes were high during HABs, but the diversity decreased during HABs. Some species, notably the Mugilidae occurred at lower densities during HABs. Mugilidae also had pronounced histological gill alterations and a lowered proportion of secondary lamellae available for gas exchange during an in situ cage experiment where fishes were isolated inside and outside the HAB. Deformities such as tumorous growths were also found for the first time in a South African Estuary. This indicated a stressed population of Mugilidae in the Sundays Estuary. The ubiquitous mugilid, Mugil cephalus, actively avoided HABs during an acoustic telemetry study, despite having a home range that overlapped the HAB area. Tagged individuals mostly occupied areas directly above the HAB in the upper mesohaline to lower oligohaline zones and only iv briefly transitioned through the HAB area to utilise the lower reaches of the estuary. Since the formation of HABs is reliant on various factors, including specific species responses, environmental variables, nutrient enrichment, and flow characteristics, the impacts of HABs on estuarine communities may vary between estuarine systems or affected species. However, the chemistry and bioactivity associated with these plants' antidiabetic activity are still lacking. Cissampelos capensis L.f. and Strychnos henningsii Gilg are among the plants utilised to manage diabetes in the Eastern Cape Province, South Africa. C. capensis and S. henningsii are known for their isoquinoline – and indole alkaloids, respectively. Other phytochemical groups associated with antidiabetic activity are the phenols, flavonoids, and terpenes. Thus, this study aimed to evaluate in-depth the phytochemical profiling, alkaloidal identification, and structural characterisation of phytochemicals isolated from these plants. Also, subsequent in vitro antidiabetic screening of the crude extracts and isolated compounds from both plants was done. The phytochemical profiling was performed by employing a qualitative and quantitative approach through high-performance thin layer chromatography (HPTLC) and UV-spectrometry. Further analysis for the identification of alkaloids was done by using LCMS. Fractionation and purification of crude extracts were achieved through crystallisation and various chromatographic techniques. NMR, HRMS, UV/Vis, FTIR, and XRD spectroscopic techniques were used to characterise isolated compounds. In vitro antidiabetic activity screening involved the investigation of cytotoxicity, antioxidant activity, alpha-amylase – and/or alpha-glucosidase inhibition (particular to type II diabetes). C. capensis and S. henningsii extracts tested positive for alkaloids, phenolics, flavonoids, and terpenes through HPTLC screening. Quantitative estimation by UV spectrometry of these phytochemical groups detected the highest concentration of alkaloids, followed by flavonoids, phenolics, and terpenes content. The in vitro antidiabetic biological screening of the various crude extracts suggested that the presence of alkaloids, flavonoids, phenolics, and terpenes fostered favourable biological activity. The LCMS of the known alkaloidal constituents from C. capensis detected the presence of ten alkaloids, including glaziovine, lauroscholtzine, pronuciferine and vi salutardine. Seven known alkaloids and one triterpene were detected from S. henningsii, including holtsiine, 23-hdroxyspermostrychnine, henningsiine, and the triterpene, friedelin. These findings suggest that C. capensis and S. henningsii have similar phytochemical constituents compared to the species reported in literature. C. capensis and S. henningsii extracts tested positive for alkaloids, phenolics, flavonoids, and terpenes through HPTLC screening. Quantitative estimation by UV spectrometry of these phytochemical groups detected the highest concentration of alkaloids, followed by flavonoids, phenolics, and terpenes content. The in vitro antidiabetic biological screening of the various crude extracts suggested that the presence of alkaloids, flavonoids, phenolics, and terpenes fostered favourable biological activity. The LCMS of the known alkaloidal constituents from C. capensis detected the presence of ten alkaloids, including glaziovine, lauroscholtzine, pronuciferine and vi salutardine. Seven known alkaloids and one triterpene were detected from S. henningsii, including holtsiine, 23-hdroxyspermostrychnine, henningsiine, and the triterpene, friedelin. These findings suggest that C. capensis and S. henningsii have similar phytochemical constituents compared to the species reported in literature. Smart catchment management and application in the Swartkops River and Estuary

- Description: The influence of urbanization and industrialization on natural resources results in complex water resource management. In South Africa, there has been escalating rural-urban migration, a growing population in urban areas, and the mushrooming of informal settlements in cities over the last five decades. In the pursuit of protecting water resources, these contextual realities create a significant challenge to policymakers, planners, and implementers. Due to rapid population growth, climate change, recent droughts, and growing competition among agricultural, industrial, commercial, environmental, and domestic sectors, water resources are presently under severe stress in South Africa. This situation necessitates the effective management of water resources. This study completed a situational assessment for the Swartkops Catchment to examine the extent of pollution, mapped sources of pollution, investigated water quality governance and identified hot spot areas to provide the foundation for effective and efficient catchment management. To determine the current water quality status of the Swartkops Catchment, this current study investigated spatial and temporal variation in physico-chemical parameters, nutrients, and faecal bacteria. This study applied a mixed-method research design using semi-structured interviews and focus groups discussions as data collection methods to investigate the water quality governance of the Swartkops Catchment. Following the situational assessment of the Swartkops Catchment, the effectiveness of applying innovative smart catchment practices to improve water quality was investigated. The word smart in this context refers to innovative mechanisms that will ensure effective and efficient water resources management. The PHP scripting language (Hypertext Preprocessor), Android studio, Wampserver, and JavaScript were used to develop the Water Use Screening System (WUSS) and Pollution Incident Reporting System (PRS). In South Africa, stormwater infrastructure suffers from unsustainable utilization. Stormwater systems discharge into many of the country‘s rivers and estuaries, including Swartkops, leading to water quality degradation. To improve the Recommended Ecological Category (REC) of the Swartkops Estuary to Category C, drivers that contribute to poor water quality must be understood to inform appropriate management interventions. Results from sampling in Markman Canal were compared with historical water quality data. The present study showed that the Markman Canal contributed faecal bacteria, nutrients, and trace metals to the Swartkops Estuary. The Markman Canal was the greatest source of trace metals to the Swartkops Estuary compared to the other point sources entering the estuary. The results show that arsenic and mercury levels in the Markman Canal and the Swartkops Estuary were higher than that previously measured. - Description: The continuous growth of mankind has not been considerate to the environment. The release of millions of tonnes of toxic heavy metal cations and anionic species through industrial, mining, agricultural, and electronic dumping has led to disease and, in many instances, death. This is usually suffered by low-income informal populations residing in third world countries. Moreover, many unnecessary deaths of children are becoming more prevalent because of consumption and contact with contaminated water, agricultural, and animal sources. Bioaccumulation of these toxic species in fish, plants, and animals, inevitably make their way back to the unaware general population. As growth by mining, agriculture, and electronics are indeed vital aspects of human development, the negative side effects of these activities usually continue unregulated. Therefore, as these processes are set to continue until more stringent regulatory processes are put into legislature; low-cost, sensitive, selective organic based sensors are a step in the right direction towards highlighting the need for environmental restoration and remediation; whilst also aiming to preventing unnecessary disease and death in the process. Herein, coumarin derived small-molecule fluorescent and colorimetric sensors for the quantitative and qualitative assessment of cationic and anionic species in aqueous and organic media are described. Ten fluorescent sensors supporting 1,4-disubstituted triazolyl moieties were synthesized according to Cu(I)-catalyzed azide-alkyne cycloaddition “click” reactions. These sensors were screened for their cationic and anionic affinities in a variety of solvent systems. Majority of the sensors responded well towards Fe3+, characterized by a strong fluorescent quenching response with a good degree of sensitivity and selectivity. Selected sensors were further investigated for their affinities towards anionic species; however, they did not display the same degree of selectivity or sensitivity towards these chosen anions. Titration studies of selected sensors with Fe3+ were able to be used towards determining the modes of fluorescent quenching; the photophysical mechanisms by which quenching occurs; stoichiometric binding ratios, association constants, and the number of coordination sites present between the sensors and Fe3+. Reversibility studies of the sensor-metal complex was investigated with EDTA. Partial reversibility was achieved for the chosen sensors with Fe3+. Hydrogen potential studies further described the application of these sensors over a good pH range.
