The international community’s implementation of the Responsibility to Protect (R2P) Principle: a Comparative Study of Sudan and Libya

  • Description: In the history of international relations, the adoption of the R2P principle in 2005 was widely understood to be a break-through in the international community’s efforts to respond to massive human rights violations and atrocities. Despite this, the international community’s implementation of the R2P has been widely contested, including in relation to its application in Sudan and Libya. There is agreement in the literature that, in both cases, the implementation of the R2P failed to achieve its intended purpose of protecting civilian populations. This thesis addresses the question of why the R2P failed in Sudan and Libya. Most explanations concerning the failure of the R2P in Sudan and Libya tend to focus on the politics of big power countries. The study argues that the failure to implement the R2P is related to the interactions between regional organisations and the international community (global organisations like UN) in attempting to solve large-scale human rights violent conflict. The study is firmly embedded in scholarly debates about humanitarian intervention so as to demonstrate the fact that this study is part of efforts to advance knowledge of scholarly debates on humanitarian intervention in this contemporary world order. The study combines a case study approach and qualitative research approach to achieve a nuanced understanding of the reason why the R2P failed in Sudan and Libya. The study relied on the use of documentary reviews and in-depth interviews of experts to collect data which was analysed using critical discourse analysis. The thesis demonstrated that the implementation of the R2P was characterised by a breakdown of the relationship between regional actors like the AU and global institutions like the UN. This breakdown contributed to the failure of the R2P in Sudan and Libya. The lack of financial and operational capacity for rapid deployment, the internal divisions with the AU, and the lack of political will within the AU members states were also cited as obstacles to the realisation of the R2P efforts in Sudan. To establish in-depth knowledge the study revealed other factors related to the politics of big power countries and how they played a part in the failure of the R2P in Sudan and Libya. Among these factors include implementation inconsistencies, the ICC factor, the absence of UNSC consensus in the response over Darfur and Libya, and the veto power factor and its implications in the efforts of the international community in both cases. This study also showed that the implementation of the R2P may not work effectively if left only to international actors to implement. Regional and sub-regional actors are important stakeholders of the R2P, and their interactions with the international community in actualising the implmementation of the R2P on the ground are essential. The manner in which the R2P was applied in Sudan and Libya is an indication its implementation requires improved interaction between regional actors and the international community at all level of collaboration including at global, regional and sub-regional level. This can build a strong foundation to drive the effective implementation of the R2P in future interventions. The findings of this thesis will significantly improve the available literature on the reason why the R2P failed in Sudan and Libya, particularly by paying much attention to the role of regional and sub-regional actors.

  • Description: Uncertainty as to the future of the liberal international order and the position of the United States within an international system in which it is not the hegemon remains a topic of debate amongst scholars of International Relations (Acharya: 2017; Duncombe and Dunne: 2018; Ikenberry: 2009, 2011, 2014; Nye: 2012 and Monteiro: 2011/2012). Fukuyama’s (1989: 4) “end of history” has not happened and a resurgence of populist leaders within established liberal democratic countries has contributed to a rapid decline of moral and ethical leadership and has further compromised the future of the liberal international order (Duncombe and Dunne, 2018: 27). As the relative power of the United States declines and the future of the liberal international order becomes increasingly uncertain, support for its future must be sought from outside the West (Duncombe and Dunne, 2018: 25 and Ikenberry: 2009). This thesis seeks to locate where potential non-Western support for the future liberal international order may be found. It does so through an analysis of how Islamic states who are part of the Organisation of Islamic Cooperation vote on the United Nations Human Rights Council (UNHRC). The OIC has a significant presence at the UNHRC and can influence the direction of the liberal international human rights regime. The evidence examined in this research project suggests that the future liberal international order and human rights regime can indeed expect some form of cooperation from the OIC. However, the OIC, and by extension Islamic states, would likely offer more support at least for human rights, if a more common understanding were to be found.

  • Description: Research on African women and HIV, particularly research that investigates their experiences of living with the virus, has been relatively peripheral. As a response to the apparent knowledge and research gaps, this project is a qualitative study involving women living with human immunodeficiency virus (WLHIV) and attending an anti-retroviral treatment (ART) clinic at Zomba Central Hospital in Southern Malawi. The study utilised an interpretative phenomenological analysis (IPA) approach, which informed all aspects of the data-collection and data-analysis processes. Using purposive sampling, 12 women were recruited and interviewed on three occasions over a period of six months. The interviews were conducted in Chichewa and audio recorded and later transcribed into English. Fourteen superordinate themes emerged from the analysis representing the women’s prominent life experiences, how they received their diagnosis, and how they contained the trauma of their diagnosis. The analysis also developed themes regarding how they coped with the challenges of living with the human immunodeficiency virus (HIV) and made sense of their experiences. The results showed that the women struggled with multiple challenges experienced through their various identities as WLHIV. The findings also pointed to interlocking disadvantages that put the women at risk of infection, and which were present from childhood. This has implications for more structural and multidisciplinary interventions for WLHIV.

  • Description: First-generation African students contend with psychosocial, structural, educational background and financial struggles to gain access to university. The first-generation students exercised their available resources and power, agency and acquired skills to negotiate their journey and entrance to university. They had to figure out for themselves how to navigate a daunting and complex path to university without relying on the knowledge and informative engagement with previous older familial generations who had the experience of attending university. They are a testimony of a generation of young people who have the resilience and grit to compensate for the structural deficits they have and experienced through their disadvantaged educational systems and their financially disadvantaged backgrounds. Although one would expect that teachers would automatically be the first encounter a high school student has of acquiring information on how to gain access to university, many of the previously disadvantaged school teachers are simply too inundated with work to be able to facilitate this process. First-generation students found themselves mostly having to look beyond the parameters of the classroom to acquire these resources. Following a qualitative approach of narrative enquiry, five African students with life experience of being first-generation Rhodes University students whose mothers were or are domestic workers, narrated their personal stories. A first-generation student’s agency, negotiation and navigation through obstacles, struggles and setbacks in the backdrop of having mothers who were/are domestic workers who socialised their children in a specific ideology to value and pursue education are the foundations for this narrative enquiry. This research provides a framework for investigating the concept of adequate and appropriate university preparedness to address the inadequate resources that previously disadvantaged schools have in terms of preparing their students to compete (in often times with more advantaged students) to gain access to university. These first-generation students did not come from educational environments where career counselling or aptitude tests are done to better equip them for entrance into university and appropriate subject and degree choice. Several common traits emerged that give perspective to the narrative of the journey that first-generation students’ had to endure and overcome to gain access to university.

  • Description: Reintroduced lion (Panthera leo) populations pose several ecological and management challenges in small (< 1,000 km2), fenced wildlife reserves. Changes in the natural socialecological conditions of reintroduced lion populations may lead to rapid population growth and a breakdown of natural predator-prey relationships. Reduced competition with other lions also likely reduces the potential for reintroduced lions to naturally form groups. My study used a combination of questionnaire surveys with tourists, existing lion demographic data from 16 wildlife reserves across South Africa and a controlled lion social experiment to address these ecological and management issues. Tourism was the primary reason for lion reintroductions. Tourists scored lions highly in terms of preference for viewing on safari, in particular, lions in larger, natural groups and adult males. Viewing lions also enhanced a tourists’ overall safari experience. The breakdown of natural social behaviour may likely therefore reduce tourist satisfaction related to lions. The number of resident prides and male coalitions in a reserve affected lion vital rates. Lion population growth rate was highest in reserves that contained a single resident pride, and the presence of unknown adult males significantly reduced cub survival and lioness birth intervals. The ratio of male cubs born also increased in reserves with a higher density of unknown adult males. Fertility control measures (deslorelin implants and unilateral hysterectomy) were effective at limiting lion population growth. Deslorelin treatment increased the age of first reproduction or the birth interval and decreased the subsequent litter size of treated lionesses to closer reflect natural vital rates in larger (> 10, 000 km2) systems. However, there was variability in infertility response between lionesses including adverse reactions in a small proportion of treated individuals. The number of resident prides and male coalitions in a reserve affected lion social behaviour. Lionesses formed larger groups in reserves with a higher density of unknown adult female neighbours, likely driven by territory defence. Lion prides with resident cubs were generally more fragmented, likely in response to reduced competition from unknown adult males. However, in areas with a high density of unknown adult female neighbours, prides with cubs formed larger groups likely in response to heightened territory defence. Therefore, with smaller foraging group sizes, predation rate was increased in reserves with reduced competition from unknown lions. My study supports a metapopulation approach for the management of lions in small, fenced reserves, and the standardisation of lion management procedures and database management. Endorsed by the Biodiversity Management Plan for lions in South Africa, this will enhance the long-term conservation potential of isolated populations.

  • Description: Hip-hop is frequently linked to misogyny and other forms of violence. This link, in many instances, is often presented outside of a critical understanding of complex underlying societal and gender dynamics. South Africa’s high rates of violence against women make it necessary to understand how hip-hop videos interact with society, as hip-hop, in its commercial form, has become a growing music genre in South Africa. Rhodes University, which has a notable student following of hip-hop, has experienced concerning levels of gender-based tensions as evidenced by the rise of the “fallist” movement’s #RapeCultureMustFall, #RUReferenceList and the suicide of Khensani Maseko, at the instigation of an alleged incident of rape, perpetrated by her boyfriend and fellow student. It is therefore of interest to investigate how a select group of Rhodes University student-fans of hip-hop make meaning out of selected South African commercial hip-hop music videos. The hip-hop music videos chosen for the study, hence Pitbull Terrier (Die Antwoord), Pearl Thusi (Emtee), Dlala ka yona (L’Tido), All eyes on me (AKA featuring Burna Boy, Da L.E.S and JR) and Ragga Ragga (Gemini Major featuring Casper Nyovest, Riky Rick and Nadia Nakai), were selected on the strength of their popularity and uniquely gendered scenarios. This study draws on qualitative research methods, thus qualitative thematic content analysis, focus group and in-depth interviews. The study establishes that despite gender being a contentious issue at Rhodes University, students make meaning out of the gendered portrayals based on their own experiences, socialisation, cultural values and level of submission to the discourse of hip-hop. This study seeks to understand how selected student-fans of hip-hop read the gendered portrayals of the music videos, based on their own experiences, socialisation, cultural values, and level of submission to the discourse of hip-hop.

The novobiocin-induced turnover of fibronectin via low density lipoprotein receptor-related protein 1 alters matrix morphology with physiological consequences on cell growth and migration

  • Description: Fibronectin (FN), an extracellular matrix protein, is secreted as a soluble dimer which is assembled into an insoluble extracellular matrix. The dynamics of FN matrix assembly and degradation play a large role in cell migration and invasion thereby contributing to the metastatic potential of cancer cells. Previous studies have shown the direct binding of Heat Shock Protein 90 kDa (Hsp90) and FN in vitro, and that inhibition of Hsp90 with novobiocin (NOV) caused internalisation of the FN matrix. Low density lipoprotein receptor-related protein 1 (LRP1) is a ubiquitous receptor known to bind both Hsp90 and FN. Using an LRP1 expressing Hs578T breast cancer cell line and an isogenic mouse embryonic fibroblast (MEF) model system of differential LRP1 expression we demonstrate that LRP1 is involved in turnover of FN in response to C-terminal Hsp90 inhibition. The first objective of this study was to identify the mechanism of NOV-induced LRP1-mediated FN turnover. Our data show that NOV-mediated FN turnover via LRP1 did not require the activity of matrix metalloproteinases (MMPs), which play an important role in processing and degradation of the extracellular matrix and FN. In addition, the levels of the main FN receptor responsible for its extracellular assembly, β1-integrin, did not change in response to NOV. LRP1 is known to undergo regulated intramembrane proteolysis (RIP) which generates smaller fragments that may translocate to the nucleus and modulate gene transcription. Using inhibitors of LRP1 cleavage and nuclear fractionation we determined that LRP1 processing was not required for the NOV-induced FN response suggesting that a mechanism unrelated to LRP1 RIP is involved. A possible mechanism may be in altered Hsp90-LRP1 cell signalling as we observed disruption of the FN-Hsp90-LRP1 complex at the cell surface in NOV treated cells. How this affects downstream eHsp90-LRP1 signalling is still to be determined but may be related to a significant increase in phospho-AKT and loss of phospho-ERK upon NOV-treatment; two key signalling proteins involved in FN matrix regulation and which are downstream of LRP1 signalling. The second objective of this study was to determine the physiological consequences associated with FN turnover in response to NOV treatment. Using migration assays we demonstrated that levels of insoluble matrix-associated FN and FN concentration are not solely responsible for migratory capacity of cells on decellularized extracellular matrices, but rather that structural composition and integrity of the matrix plays a bigger role. Using confocal and scanning electron microscopy, we identified NOV treated matrices to be flatter, less mature and contain thicker, rope-like FN fibrils to which cells adhered better but were generally less proliferative. Comparatively, cells adhered less to the more mature and 3-dimensional untreated matrices but exhibited increased spreading and cell growth, which may in part be due to the thinner fibrils and web-like matrix. In summary, this study substantiates the role of LRP1 in NOV-mediated FN turnover, and provides new insights into the possible mechanisms of the Hsp90-LRP1 mediated loss of FN matrix. The past meets the present: exploring the biogeography of extant plecoptera of South Africa with reference to ancient middle permian fossil forms from the Onder Karoo locality near Sutherland

  • Description: The Onder Karoo fossil locality near Sutherland, Northern Cape, recently yielded an unprecedented diversity of middle Permian insects, which were preserved in deposits of an aquatic lake margin system, in the Lowermost Abrahamskraal Formation. A large number of Plecoptera (stonefly) specimens were found, and this study represents an in-depth analysis of these species in the context of plecopteran evolution. A significant contribution to current knowledge of the Plecoptera from the middle Permian to the present has been made, utilizing a combination of fossil and phylogenetic evidence to better understand the origins, evolution, diversity and biogeography of the stoneflies. Three new plecopteran species have been described from the Onder Karoo locality, which is recognized here as the first Lagerstätte of its kind in Gondwana. Possible ecological links and relationships with the plecopterans described here have been suggested, using extant Plecoptera as analogues. An in-depth, fossilcalibrated, dated phylogeny of the Plecoptera was completed, using representatives from all three dominant lineages, the Antarctoperlaria, Systellognatha and the Euholognatha. This provides strong support for the monophyly of the Notonemouridae and the Antarctoperlaria, however the monophyly of Systellognatha and Euholognatha was not supported. The Notonemouridae were found to represent an earlier divergence than previously believed, forming a sister group to the remaining Plecoptera. Through a combination of fossil and molecular evidence, strong support was found for the current distribution of the Plecoptera worldwide being attributable to vicariance caused by the rifting of Pangea, and subsequently Gondwana and Laurasia, and long range dispersal. The focussed effort to include Southern Hemisphere stoneflies throughout this study has been a valuable step in reducing the Northern Hemisphere bias which currently dominates plecopteran research and has assisted in opening the way for future research into this important group on a global scale.

  • Description: This study redefines the political in dance by drawing on the scholarly concept of the “choreopolitical” (André Lepecki) and extending it into analysing related concepts such as the “postdramatic” (Hans-Thies Lehmann), performance and/as research, among others from Performance Studies scholarship as well as from First Physical Theatre Company’s pioneering legacy of production, pedagogy and research in making Phyical Theatre performance. Following from the notion that performance is both a site and a method of study/knowing, the research invites a rethinking of the relationship between art (performance), epistemology and the political, in the sense that performance becomes a way, not of simply re-presenting the political but, as its own way of knowing, actively questioning the very categories on which the political is premised. The argument for Physical Theatre as having nascent potential to invoke what I call “the power of the small” is analysed as a choreopolitical method and community of practice that has a generative capacity to produce the “intimate revolts” (Julia Kristeva) or body of questions that can perform the imaginative curiosities/forms required to create provocative, subversive, ethical, reflexive and charged performance. My argument is supported by critical commentary, insight, choreological analysis and reflection on the dramaturgical strategies and choreopolitics of selected commissioned choreographers and dance forms that extended FPTC’s manifesto and production between 1993 and 2015. My project has the following three goals: (i) to contextualise, conceptualise and identify key issues in the identity, pedagogy and performance ethos of Physical Theatre as a performance philosophy and form; (ii) to engage critically with the praxis of Physical Theatre within the contextual, cultural, historical and political relationships between Physical Theatre and other performance practices in South Africa; and (iii) to document, analyse and interpret selected claims, works and performance processes from the archive of FPTC’s repertory and training manifesto from 1993 to 2015. The research evaluates the political significance and consequence of FPTC’s heritage and legacy problematising constraints, possibilities, tensions, failures and proposing the hope of imaginative entanglements with practising freedoms.

  • Description: On 4 August 1983, a thirty-three year-old army captain seized power in Burkina Faso and embarked on what can be described a revolutionary journey. Over the next four years, until his assassination in 1987 the government, led by Captain Thomas Sankara, attempted to redeem Burkina Faso from the clutches of neo-colonialism. Through popular mobilisation and organisation, infrastructure (schools, hospitals, bridges) was built, millions of children were vaccinated and diseases such as river blindness were eliminated. Women, long-subjugated by patriarchal systems took up space and led their own initiatives in freedom, including holding senior roles in the public service. On the international stage, practical solidarity was extended to countries either fighting or threatened by neo-colonialism despite the fact that Burkina Faso was poor and was itself threatened by France and her lackeys. What Sankara inherited in August 1983, twenty-three years after Burkina Faso’s independence, was a fragile neo-colonial state which was not allowed by dominant imperialist interests to set an example of what true independence means. So, in just four years, it was all over. Sankara was assassinated by his comrades and the revolutionary project he had led came to a halt. The tragedy of Sankara was the tragedy of all those attempts at revolution which occur before mass movements have had the opportunity to develop and organise themselves independently of the state. Despite this, it is apparent today that Sankara has been influential on current political movements and parties in Africa, from Burkina Faso to South Africa. One of these political movements is the Economic Freedom Fighters (EFF) in South Africa. This half-thesis is an investigation of Sankara’s political thought.   • Description: The study explored the potential of Geography fieldwork projects to develop learners’ action competence in response to social-ecological problems in two schools in the Omusati and Oshana Education Regions. The study investigates ways in which learners’ activities create an opportunity for them to act for the environment. That is, to investigate ways in which learners can be shaped into environmentally literate citizens who act for the environment. A total of 43 grade 10 learners and two grade 10 Geography teachers participated in this qualitative, interpretive case study. Data was generated through document analysis, focus group interview with learners, semi-structured interviews with educators, and participant observations. Such data were analysed into three phases, first inductively and then abductively using Jensen’s (2004) Investigations, Visions, Actions and Change (IVAC) model to produce analytic statements that form the basis of the research findings. The study found that Geography fieldwork projects foster understandings of environmental problems in a local area. Through fieldwork projects, schools initiate indirect environmental actions, however, some environmental problems are too large or complex to be resolved by the actions initiated by learners. Based on these findings, the study identifies Geography fieldwork as an opportunity to create a community of practice in effort to address socioecological concerns. The study further recommends that learners should be allowed to engage with both social investigative actions and technical, scientific investigative actions when doing project work.

  • Description: This psychosocial study investigates the maternal subjectivities of mothers returning to work after maternity leave, who are living in scarcely-resourced Cape Town communities in South Africa. Engaging with interview texts and listening to mothers’ talk, I explore how and why maternal subjectivity is constructed discursively and defensively in our talk. This thesis claims that these particular mothers predominately employ instrumental mothering discourse. The traditional subject position of the intensive mother – which is typically assumed to be the ‘good mother’ – is not a position available to these mothers due to their social circumstances and working role. Subsequently, material provision, the baby’s thriving and surviving body, finding substitute carers and maternal preoccupation are constructed as qualities of ‘good mothering’ in their talk. This ‘good mother’ position, however, is a precarious position that both these mothers and I invest in to defend against feelings towards their babies and themselves as well as to deny (maternal) ambivalence in a problematic social system. In a combined analysis drawing on discursive theory and psychoanalysis, more specifically contemporary attachment theory and intersubjectivity theory, I illustrate how both these mothers and I – as emotional, social and political subjects – co-construct maternal subjectivity. Based on the findings, recommendations for parent-infant interventions are discussed. Arguing that a purely psychoanalytic reading of investment perpetuates notions of individual blame and pathology, I advocate for a psychosocial reading that does not neglect failing social systems but rather pursues an open and reflective, yet critical, mindfulness when listening to talk.

  • Description: The use of the yield curve in forecasting economic recessions is well established in the literature. A new avenue of use for the yield curve has emerged in the form of using it to forecast bull and bear stock markets. This has the potential to change how investors manage portfolios. A dynamic market-timing strategy would allow investors to shift out of or in to stock markets based on the probability of bear stock market in the future. The relationship between the yield curve and the stock market is tested using an adapted probit model. This has proven positive with encouraging results for the US, India and Spain. This is tested for South Africa using the adapted probit model and the SA yield spread. Bear stock markets are identified on the JSE and forms part of the probit modelling process. Bear markets are identified using a six- and four-month criteria. As South Africa is a small, open and developing economy, the probit is also modelled using the US yield spread. The three probit models do not appear to track bear markets well. This is substantiated through the Henriksson-Merton parametric model test which tests for market timing ability. The results for the SA yield spread using both bear market criteria do not show market timing ability, however, the SA and US yield spread model does show potential market timing ability.

  • Description: The main objective of this study was to question and investigate the primacy of illicit financial flows (IFFs) in developing countries, specifically focused on two countries namely China and South Africa. Africa is estimated to have lost approximately $1 trillion to IFFs over the last 50 years, which exceeds the financial assistance that these nations needed over the same period. For years. Africa has been the feeding ground for exploitation and resource plunder, and the narrative has always been Africa is underdeveloped because of this crime. Although this statement holds true in most African countries, what this paper seeks to do is to question whether capital flight, IFFs and more specifically tax evasion and tax haven activity are the reason for the deterioration of African economies or are IFFs perpetuated by economies with unsustainable growth paths. IFFs are an important factor when it comes to obstacles of economic growth. But are they the cause or effect? A very strong case can be made that they are the latter however, it is beyond the scope of this article to resolve this question. Its purpose is merely to assert that the question is a valid one and that presuming the answer could divert attention from the real question of economic development. This study contextualized the way in which IFFs are currently viewed in the world economic system according to the two approaches to development finance, and discussed modern monetary theory as an extension off these theories. Due to the nature of the study, the methodology employed is a case study approach between China and South Africa by means of extensive numerical and document analysis. Upon conducting this analysis on the primacy of illicit financial flows in developing countries there was difficulty in measuring IFFs. The reason for this is because IFFs have a range of estimates so it was very difficult to produce precise and accurate results. The key findings of this paper were that there seems to be some kind of parallel between developing countries with large volumes of illicit financial outflows, and a dependency these countries have on external debt. This means it seems that weak economies, that are highly dependent on external debt and have large amounts of this debt, seem to have the largest volumes of illicit financial outflows. Weak regulation, high levels of debt and liberalised trade markets seem to be contributing factors to the degree to which companies evade taxes and partake in tax haven activity in these regions. Another key finding was that in 2012, despite China being ranked number one in the the countries which have the largest amounts of outflows on average, it still managed to achieve large amounts growth in the last 20 years. Indicating that there is some form of indication that IFFs could be viewed as symptomatic of weak financial systems and weak economies, instead of IFFs being the core of the problem.

  • Description: This dissertation explored the views of academics who teach and research in the area of Industrial/Organisational Psychology in South Africa about the utility of the field in engaging with the post-1994 South African workplace, thereby remaining relevant. When it emerged as a field of study and practice, Industrial/Organisational Psychology research’s aim was to inform workplace practice and contribute to the betterment of society. It appears as if today this research is deemed irrelevant to the workplace and society, with many practitioners relying on their own knowledge and irrelevant repetitive one size-fit-all Euro-American developed theoretical framework and research evidence to solve the challenges of the post-1994 South African workplace, and to serve its society. The latter propelled the researcher to ask broadly the question of relevancy of the discipline in meeting the demands of the post-1994 South African workplace. Face-to-face, semi-structured interviews were employed to collect data on 8 senior and younger generations of academics in 3 different universities. The collected data was analysed using Braun and Clarke’s 6 steps of thematic analysis. The data and study as a whole was approached with a ‘post’-colonial lens and a Contexualist paradigm in order to contextualise in the present time the past nuances that arose in our country during the colonial and apartheid eras. Themes discussed seemed to indicate a lack of research focus by academics and their students due to limited time and stringent bureaucratic publication structures present both within their universities and outside. When it came to the discipline as seen through the lens of the data and what this suggested in terms of speaking to post-1994 workplace organisational psychological problems, the study found that the findings arising out of the research in I/O psychology appear to be of little relevance to whom they are currently aimed, leading to the idea of these studies acting as a change agent in the workplace and society to fall to the wayside.

  • Description: Environmental sustainability and agriculture are key development and transformational concerns in South Africa while rural development and youth unemployment are key national issues pertinent in the Eastern Cape which ranks in the top three provinces for both concerns. As a formative interventionist researcher, working in a singular case study with youth in Lenye village located in the Eastern Cape Province of South Africa, I sought to investigate the following: Can, and if so, how can expansive learning facilitated by a change laboratory intervention contribute to the development of youth as potential value creators for rural development? This was investigated through the following sub questions: 1) What value is created at each point of the expansive learning process of the change laboratory process and for who? 2) What value can still be created at each point of the expansive learning process of the change laboratory? 3) How is that value created via an expansive learning process? I collected data as I participated as a youth member through: extended contextual profiling via a focus group interview, individual interviews, note taking and document analysis. Furthermore, I attended village meetings, youth meetings and emergent youth development/youth in agriculture/agriculture opportunities. Additionally, change laboratory workshops were conducted and I used audio recordings and notes to capture data. The data collected was then analysed through second generation Cultural Historical Activity Theory using the concepts of immediate, potential, applied, realised and reframing value. In conclusion, the expansive learning process may contribute to the development of rural youth as value creators for sustainable development through youth development, agriculture and community development. All forms of value were created along the expansive learning process for the formative interventionist researcher, the Lenye youth and the greater Lenye youth community. All forms of value may still be created for the Lenye youth, the formative interventionist researcher and the greater Lenye youth community.

  • Description: This research was a Case Study on the role of Flexible Working in achieving a Work – Life Balance, based on the Information Technology Consultants of Kestrel Business Solutions – a company based in London, the United Kingdom. The study set out to determine what aspects of flexible working can reduce work - related stress; home - related stress; improve personal wellbeing; identify the pitfalls of flexible working, and to recommend mitigating actions. The literature review identified five different, but complimentary theories that included: The Spill over Theory; Balanced Theory; Resource Based Theory; Compensation Theory and the Wellness Theory to unpack and explore the concept of Work - Life Balance. The study followed a predominantly qualitative methodology approach and used thematic analysis to identify key ideas from the data. Interviews and questionnaires were used to obtain data from the respondents, in addition to being a data triangulation exercise. From a population of 10 subjects - purposeful sampling was used to select a sample of 10 respondents - made up of 3 males and 7 females. All respondents were employees of Kestrel Business Solutions. 6 respondents were interviewed and given questionnaires to fill out. 6 face to face interviews were conducted, and 6 questionnaires completed until saturation was achieved - indicating a survey response rate of 100%. The results from the data collection generally showed how aspects of flexible working like telecommuting, flexitime and homeworking ultimately helped the employees to achieve a Work-Life Balance - by primarily influencing how they balanced their work, home, and personal commitments. Importantly, the study also identified pitfalls of flexible working; like employees feeling isolated and the pressure to perform. Potential mitigating efforts identified in the study included, providing more company support and education for flexible working initiatives. The study concluded by acknowledging the limitations of the study, which included time constraints and the small population, and sample size of the study. In its conclusion, the study identified recommendations for future studies like: increasing the population and sample size, and devoting more time to the study, for more generalized and conclusive findings.

The Role of HSP70/HSP90 Organizing Protein (Hop) in the Heat Shock Factor 1 (HSF1)-mediated Stress Response

  • Description: Molecular chaperones regulate cellular proteostasis. They control protein conformation and prevent misfolding and aggregation under both normal and stressful environments, ultimately resulting in cell survival. The project aimed to understand the role of the HSP70 – HSP90 Organizing Protein (Hop/STIP1) in the survival of stressed cells and the function of the stress-responsive transcription factor, Heat Shock Factor 1 (HSF1). HSF1 protein levels were significantly reduced in Hop-depleted HEK293T cells compared to controls by ELISA, western blot, and mass spectrometry. HSF1 transcriptional activity at the HSP70 promoter, and binding of a biotinylated HSE oligonucleotide under basal conditions were significantly reduced, consistent with the reduced levels of HSF1. In response to heat shock, HSF1 levels in Hop-depleted cells increased to that of controls, but there was still significantly lowerHSF1 transcriptional activity and HSE binding. Hop-depleted HEK293T cells were more sensitive than controls to the HSF1 inhibitor KRIBB11 and showed reduced short-term and long-term proliferation. Unlike the HSP90 inhibitor 17-DMAG, which had no effect, the HSP70 inhibitor JG98, further decreased the levels of HSF1 in Hop-depleted cells, suggesting a role for HSP70 in the Hop-mediated effects. There was punctate nuclear staining for HSF1 in Hop-depleted cells under both basal and heat shock conditions, as well as reduced nuclear localization and increased cytoplasmic accumulation of HSF1 in response to heat shock. Hop and HSF1 colocalized in cells, and HSF1 could be isolated in complex with Hop and HSP70. Loss of Hop reduced HSF1 in HSP70complexes but did not affect HSF1 abundance in HSP90 complexes. Hop-depleted cells showed reduced short-term and long-term survival compared to controls, an effect that was potentiated by the JG98 HSP70 inhibitor. Taken together, these data suggest that Hop regulation of HSF1activity is via a mechanism involving reductions in HSP70 interaction, as well as reduced nuclear localization, and DNA binding, and is consistent with reduced cellular fitness under basal and stress conditions. , Thesis (PhD) -- Faculty of Science, Biochemistry and Microbiology, 2020
