- Description: Driver fatigue is a complex phenomenon that has a range of causal factors including sleeprelated and task-related factors. These manifest as different safety and performance outcomes. Extensive research has been applied to linking these factors to performance impairment. However, little research focuses on the mechanisms by which this link exists. This research project therefore focuses on the processes underlying how driving performance is controlled and maintained during the development on non-sleep-related driver fatigue. The main aim was to establish whether progressive impairment of driving control over a prolonged drive could be attributed to a depletion of attentional resources, as proposed by Resource Theory, or to a withdrawal of effort, as proposed by Effort-Regulation Theory. As a multicomponent skill, driving requires perception, cognition and motor output. The secondary aim of this research was therefore to assess whether a prolonged drive impairs stage-specific information processing. Participants (n=24) in three experimental groups performed a 90-minute simulated drive wherein they were expected to keep the bonnet of a car on a lane (tracking task). The three groups differed in terms of lane width: small, medium and large, corresponding to low, medium, and high task-demand, respectively. To assess the impacts of this task on stagespecific information processing, participants performed a set of resource specific tests before and after the prolonged drive. Each task had two difficulty variations to ensure that performance decrement was due not only to the task-characteristic, but specifically to resource depletion. The tests probing information processing were: a modified Fitts' tapping task for motor programming, a digit recall task for perception, and an object recognition reading task for cognition. Performance was measured as lateral deviation of the car. Physiological measures included heart rate frequency (HR) and various time- and frequencydomain heart rate variability (HRV) parameters, eye blink frequency and duration. The Borg CR-10 scale was used to evaluate subjective effort and fatigue during the task. Driving control declined over time and was supplemented by HR, HRV, blink frequency and duration, indicating an increase in parasympathetic activity (or a reduction in arousal). An increase in blink frequency was considered as a sign of withdrawal of attentional resources over time. Driving control declined to a greater extent in the large road width group and reflected a lower parasympathetic activity, whereas the inverse was observed for the small road width group. Resource tests reveal a non-specific impairment of information processing following the prolonged drive. However, this was accompanied by an increase in parasympathetic activity. Overall, results indicate that Effort-Regulation Theory better accounts for the impairment of driving control in prolonged driving than does Resource Theory. This suggests that the impact of fatigue is guided more by task goals and intrinsic motivation than by the manner in which the fatigue state developed. Moreover, performance impairment by effort-regulation is dependant more on time on task than on task-demand

The research was carried out on secondary school learners at boarding schools in Grahamstown. The participating learners were in Grades 8 to 12. The goals of the research included determining the actual levels of awareness, the difference between these and self-perceived levels of the participants, the assessment of risk in terms of online behaviour, and the determination of any gender differences in the answers provided by the respondents. A review of relevant literature and similar studies was carried out, and data was collected from the participating schools via an online questionnaire. This data was analysed and discussed within the frameworks of awareness of threats, online privacy social media, sexting, cyberbullying and password habits. The concepts of information security and online privacy are present throughout these discussion chapters, providing the themes for linking the discussion points together. The results of this research show that the respondents have a high level of risk. This is due to the gaps identified in actual awareness and perception, as well as the exhibition of online behaviour patterns that are considered high risk. A strong need for the construction and adoption of threat awareness programmes by these and other schools is identified, as are areas of particular need for inclusion in such programmes. Some gender differences are present, but not to the extent that, there is as significant difference between male and female respondents in terms of overall awareness, knowledge and behaviour.

There are approximately 400 different species of microbiota which may be beneficial, harmful or both, but each play an important role in the regulation and modulation of the hosts’ bowel processes (McOrist et al. 2008; Dethlefsen et al. 2008). Many of these colon microbiota allow for saccharolytic fermentation of non-digestible dietary fibres and carbohydrates into by-products and intermediates, followed by a subsequent conversion into short chain fatty acids (SCFAs) (mainly n-butyric acid, propionic acid and acetic acid) each of which play an important role in maintaining colon homeostasis (Topping & Clifton 2001). A balance of ‘good’ microbiota (e.g., Bacteroides spp./ Bifidobacteria spp.) and ‘bad’ microbiota (e.g., Veilonellae) and the optimal production of various SCFAs within the gut could possibly allow for proper functioning of the large intestine and assist in decreasing the onset of various colonic disorders such as Irritable Bowel Syndrome (IBS). The sample group for the study consists of male and female patients, with an average age of 40 to 50 years old, whom of which have been diagnosed with either constipation IBS (C-IBS) or diarrhoea IBS (D-IBS) via the Rome III criteria system for IBS diagnosis. DNA and SCFA extractions were optimised for human stool, colonic fluid and tissue biopsy sample obtained from the aforementioned patients. Optimization steps allowed for starting material with high analysis integrity. Different methods of microbiota analysis, such as ARISA, were investigated; however, real-time qPCR was selected as the best method to identify and quantify specific microbiota. Extracted SCFAs were separated via gas chromatography and identified and quantified via Mass Spectrometry. Significant changes in microbial content and SCFA profiles were found to be associated with healthy and IBS patients. Results obtained would however be influenced by external factors typical of clinical studies of this nature. This study allows for opportunities for future research into understanding IBS.

Pelagic fish dominate the mid trophic level and can exert both a top-down control of zooplankton and a bottom-up control on predatory fish and marine top predators. They also make up a substantial contribution of the world catches in seas around the world where they co-exist in upwelling areas. The fluctuations of sardine and anchovy are largely influenced by recruitment from the larval stage and by the physical environmental forces that determine the planktonic compositions upon which they feed. In southern Africa, the biology and behaviour of two threatened seabird species, African penguins (Spheniscus demersus) and Cape gannets (Morus capensis), are directly influenced by the availability of sardine (Sardinops sagax) and anchovy (Engraulis encrasicolus). This study aimed to provide further insight into the fluctuations of sardine and anchovy in relation to the variability of environmental factors such as wind, sea surface temperature, rainfall and plankton abundance, focusing specifically on Algoa Bay in the Eastern Cape, South Africa. Algoa Bay is particularly important as it is home to the world’s largest African penguin colony and the world’s largest gannetry. Therefore, to further investigate the populations of these two seabird species, the fluctuations of their food source were studied.

Using this approach one has at one's disposal a variety of algorithms that make the solution process of many differential equations algorithmic. Vital properties of a given differential equation can often be inferred from the symmetries admitted by the equation. However, Lie group analysis has not enjoyed wide-spread application to systems of first-order ordinary differential equations. This is because such systems typically admit an infinite number of Lie point symmetries, and there is no systematic way to find even a single nontrivial one-dimensional Lie symmetry algebra. In the few applications available, the approach has been to circumvent the problem by transforming a given system of first-order ordinary differential equations into one in which at least one of the equations is of order two or greater. It is therefore fair to say that the full power of Lie group analysis has not been sufficiently harnessed in the solution of systems of first-order ordinary differential equations. In this dissertation we review some applications of Lie group analysis to systems of first order ordinary differential equations. We shed light on the integration procedure for first-order systems of ordinary differential equations admitting a solvable Lie algebra. We do this via instructive examples drawn from mathematical epidemiology models. In particular we revisit the work of Nucci and Torrisi [54] and improve the exposition of the Lie-symmetry-inspired solution of a mathematical model which describes a HIV transmission. To aid implementation of the integration strategy for systems of ordinary differential equations, we have developed ad-hoc routines for finding particular types of admitted symmetries and checking if a given symmetry is indeed admitted by a system of ordinary differential equations.

- Description: Livelihood stressors in southern Africa, such as HIV/Aids and climate change, do not act in isolation but rather interact concurrently in complex socio-ecological systems with diverse, interrelated and compounded affects. Households experience differential vulnerability to such stressors based on contextual factors such as geographical location, income level and the gender and age of its members. Households’ differential experiences of vulnerability are further defined by the households’ use of their capital stocks: the human, social, natural, financial and physical capital available to the household to form livelihoods and resist the detrimental effects of a stressor. The capital stocks of 340 households were measured in two sites in the Eastern Cape, South Africa, using a household survey. These data were analysed to determine differences between the sites, households with heads of different gender and households of different income levels. Further data relating to the drivers and interactions of stressors over temporal and spatial scales, as well as the perceived value of various forms of capital by different social groups in the two sites, were collected via Participatory Learning and Action (PLA) methods including timelines, mental modelling and pair-wise ranking. Although the two sites have similar levels of income and fall within the same province, many significant differences emerged. The two sites showed different distributions of household head genders and different stressors and perceptions of vulnerability, perhaps owing to differences in their capital stocks, acting alongside the influence of culture and access on a shifting rural-urban continuum. These discrepancies further transpired to reflect crucial differential experiences along gender lines and income levels in each site. Vulnerability was often context specific, not only because of unique drivers of stress in different areas, but also because socio-economic groups and localities often had characteristics that could potentially exacerbate vulnerability, as well as characteristics that can potentially facilitate adaptive capacity. Stressors were found to have depleted multiple forms of capital over time, while new stressors were emerging, raising concerns over the most appropriate means of social protection within these contexts.

- Description: Livelihood stressors in southern Africa, such as HIV/Aids and climate change, do not act in isolation but rather interact concurrently in complex socio-ecological systems with diverse, interrelated and compounded affects. Households experience differential vulnerability to such stressors based on contextual factors such as geographical location, income level and the gender and age of its members. Households’ differential experiences of vulnerability are further defined by the households’ use of their capital stocks: the human, social, natural, financial and physical capital available to the household to form livelihoods and resist the detrimental effects of a stressor. The capital stocks of 340 households were measured in two sites in the Eastern Cape, South Africa, using a household survey. These data were analysed to determine differences between the sites, households with heads of different gender and households of different income levels. Further data relating to the drivers and interactions of stressors over temporal and spatial scales, as well as the perceived value of various forms of capital by different social groups in the two sites, were collected via Participatory Learning and Action (PLA) methods including timelines, mental modelling and pair-wise ranking. Although the two sites have similar levels of income and fall within the same province, many significant differences emerged. The two sites showed different distributions of household head genders and different stressors and perceptions of vulnerability, perhaps owing to differences in their capital stocks, acting alongside the influence of culture and access on a shifting rural-urban continuum. These discrepancies further transpired to reflect crucial differential experiences along gender lines and income levels in each site. Vulnerability was often context specific, not only because of unique drivers of stress in different areas, but also because socio-economic groups and localities often had characteristics that could potentially exacerbate vulnerability, as well as characteristics that can potentially facilitate adaptive capacity. Stressors were found to have depleted multiple forms of capital over time, while new stressors were emerging, raising concerns over the most appropriate means of social protection within these contexts.

- Description: Globally largemouth bass Micropterus salmoides is one of the most widely introduced game fish species which has now become invasive in many countries. Well researched in its native North America, there are few studies on this species in its introduced range. This study aims to improve the understanding of the biology and ecology of M. salmoides in temperate southern Africa, where it threatens native biota and supports a popular recreational fishery. This aim was addressed by assessing the age, growth, maturity, reproductive seasonality, feeding, mortality and movement, of two M. salmoides populations from Wriggleswade (1000 ha, 723 m amsl, 32º35'S; 27º33'E) and Mankazana (35 ha, 66 m amsl, 33º09'S; 26º57'E) Dams in the Eastern Cape of South Africa. The most suitable structure for ageing M. salmoides was determined by comparing the accuracy and precision of otoliths and scales. Scales tended to underestimate the age of fish older than five years. Sectioned sagittal otoliths were the more precise structures for ageing (otoliths CV = 15.8 vs. scales CV = 21.9). The periodicity of growth zone formation was validated as annual for otoliths using both edge analysis (EA), and mark recapture of chemically tagged fish (MRCT). EA indicated one annual peak in the frequency of opaque margins between September and January using a periodic logistic regression and a binomial model linked with a von Mises distribution for circular data. MRCT showed the linear relationship between time at liberty and number of growth zones distal to the fluorescent band was not significantly different from one (slope = 0.89). Reproduction was similar to that in native and non-native populations with fish reaching age at 50% maturity at ca. two years, and length at 50% maturity at 259-290 mm FL depending on growth rate. Spawning season occurred in the spring months of August to October. The Wriggleswade Dam population reached similar ages (14 yrs) to populations in temperate North America. Growth was described using the von Bertalanffy growth equations of (Lt = 420(1-ᵉ⁻°·³³⁽ᵗ⁺°·²¹⁾) mm FL in Wriggleswade and (Lt = 641(1-ᵉ⁻°·²²⁽ᵗ⁺°·⁷⁶⁾) mm FL in Mankazana. Growth performance of both populations, described using the phi prime index, seemed to be correlated with temperature and the Wriggleswade Dam population (Φ’ = 2.8) grew similarly to those native temperate North American populations and the non-native populations of Japan and Italy. The Mankazana population had a higher growth performance (Φ’ = 2.9) compared to Wriggleswade and growth was comparable to other nonnative African populations. Both populations utilised all available food resources, with the Mankazana population being in better condition than the Wriggleswade population as a result of a more diverse diet, highly abundant food sources throughout the year, and potentially the adoption of a more conservative ambush feeding strategy. The Wriggleswade population, which was utilised by competitive anglers, showed low mortality rates (Z = 0.43 yr⁻¹), and translocated fish dispersed from the common release site, travelling an average distance of 4km over the study time of 494 days. These results indicated that M. salmoides are successful in these environments, growing at expected temperature dependant rates, maturing at two years, using all available food groups, and suffering from low mortality. It is therefore likely that they will persist in South Africa and establish where introduced.

Changes in muscle recruitment, functional strength and ratings of perceived effort during an 8-over bowling spell: impact on performance

- Description: Background: The musculoskeletal demands placed on the lower limb musculature of fast bowlers over time have not received much attention. In particular, measures of muscle recruitment changes have, to the author’s knowledge, not been considered. Objective: The present study, therefore sought to establish any associations between an eight over, simulated fast bowling spell, and muscle activation patterns, power output, perceptual demands, and changes in performance. This will enable improvements in the development of training programmes. Methods: Players’ were required to attend two sessions in total. The purpose of the initial session was to collect specific demographic, anthropometric and physiological data and injury history information from each player. In addition, this first session allowed for habituation with the treadmill, the jump meter and all other equipment involved in experimentation. The second testing session involved electrode attachment sites being identified on player’s dominant leg. The areas were then shaved, wiped with an alcohol swab and left to dry, to ensure good connectivity. Pre- and post- measures of muscle activity and functional strength of the lower limbs were recorded in the Department of Human Kinetics and Ergonomics. The protocol took place at the Kingswood High Performance Centre, which is in close proximity to the initial testing site. The protocol involved players bowling eight overs (48 balls). During the protocol, accuracy, ball release speed and perceptual measures were recorded at the end of each over. After the protocol, players were driven back to the Human Kinetics and Ergonomics Department where post-testing measures were completed. The dependable variables of interest were muscle activation, functional strength of the lower limbs, ‘local’ ratings of perceived exertion (RPE), body discomfort, accuracy, and ball release speed. Results: For all muscles it was shown that, as the speed increased so did the muscle activity in players’ lower limbs. There were no significant changes in muscle activity preversus post-protocol. There was however, a general trend of decreasing muscle activity post protocol at higher testing speeds. There were significant (p<0.05) decreases in peak power following the simulated eight over bowling spell. ‘Local’ RPE displayed a significant (P<0.05) increase with each additional over and were observed to reach the ‘heavy’ category. The players’ highest discomfort area was in the lower back, with 13 players perceiving discomfort in this region following the eight over spell. The shoulder and chest were another two areas player’s indicated discomfort with eight players selecting the dominant shoulder. Seven players complained of the dominant side pectoral muscle, leading foot and dominant latissimus dorsi muscle being uncomfortable. Interestingly, the dominant pectoral showed the highest body discomfort ratings amongst players. There were no significant changes in accuracy between overs although there were large interindividual differences in accuracy points between players. The decrease in ball release speed observed during over seven was shown to be significantly (p<0.05) lower than overs one to four. Conclusion: The power output and perceived strain results of the players, appears to indicate the presence of fatigue in players. However, the results are not conclusive, as the fatigue was not shown in muscle recruitment patterns, as well as the body discomfort ratings. There was a non-significant trend observed in the lower limb muscle activation decreasing at higher speeds. Players were able to maintain accuracy. However, the significantly lower ball release speed observed during over seven showed players performance decreasing.

- Description: Heat Shock 40 kDa proteins (Hsp40s) partner with heat shock 70 kDa proteins (Hsp70s) in facilitating, among other chaperone activities; correct protein transport, productive protein folding and assembly within the cells; under both normal and stressful conditions. Hsp40 proteins regulate the ATPase activity of Hsp70 through interaction with the J-domain. Plasmodium falciparum Hsp70s (PfHsp70s) do not contain a Plasmodium export element (PEXEL) sequence although PfHsp70-1 and PfHsp70-3 have been located outside of the parasitophorous vacuole. Studies reveal that a type I P. falciparum (PfHsp40) chaperone (PF14_0359) stimulates the rate of ATP hydrolysis of the cytosolic PfHsp70 (PfHsp70-1) and that of human Hsp70A1A. PFE0055c is a PEXEL-bearing type II Hsp40 that is exported into the cytosol of P. falciparum-infected erythrocytes; where it potentially interacts with human Hsp70. Studies reveal that PFE0055c associates with structures found in the erythrocyte cytosol termed “J-dots” which are believed to be involved in trafficking parasite-encoded proteins through the erythrocyte cytosol. If P. falciparum exports PFE0055c into the host cytosol, it may be proposed that it interacts with human Hsp70, making it a possible drug target. The effect of PFE0055c on the ATPase activity of human Hsp70A1A has not been previously characterised. Central to this study was bioinformatic analysis and biochemical characterisation PFE0055c using an in vitro (ATPase assay) approach. Structural domains that classify PFE0055c as a type II Hsp40 were identified with similarity to two other exported type II PfHsp40s. Plasmids encoding the hexahistidine-tagged versions of PFE0055c and human Hsp70A1A were used for the expression and purification of these proteins from Escherichia coli. Purification was achieved using nickel affinity chromatography. The urea-denaturing method was used to obtain the purified PFE0055c whilst human Hsp70A1A was purified using the native method. PFE0055c could stimulate the ATPase activity of alfalfa Hsp70, although such was not the case for human Hsp70A1A in vitro.

- Description: Heat Shock 40 kDa proteins (Hsp40s) partner with heat shock 70 kDa proteins (Hsp70s) in facilitating, among other chaperone activities; correct protein transport, productive protein folding and assembly within the cells; under both normal and stressful conditions. Hsp40 proteins regulate the ATPase activity of Hsp70 through interaction with the J-domain. Plasmodium falciparum Hsp70s (PfHsp70s) do not contain a Plasmodium export element (PEXEL) sequence although PfHsp70-1 and PfHsp70-3 have been located outside of the parasitophorous vacuole. Studies reveal that a type I P. falciparum (PfHsp40) chaperone (PF14_0359) stimulates the rate of ATP hydrolysis of the cytosolic PfHsp70 (PfHsp70-1) and that of human Hsp70A1A. PFE0055c is a PEXEL-bearing type II Hsp40 that is exported into the cytosol of P. falciparum-infected erythrocytes; where it potentially interacts with human Hsp70. Studies reveal that PFE0055c associates with structures found in the erythrocyte cytosol termed “J-dots” which are believed to be involved in trafficking parasite-encoded proteins through the erythrocyte cytosol. If P. falciparum exports PFE0055c into the host cytosol, it may be proposed that it interacts with human Hsp70, making it a possible drug target. The effect of PFE0055c on the ATPase activity of human Hsp70A1A has not been previously characterised. Central to this study was bioinformatic analysis and biochemical characterisation PFE0055c using an in vitro (ATPase assay) approach. Structural domains that classify PFE0055c as a type II Hsp40 were identified with similarity to two other exported type II PfHsp40s. Plasmids encoding the hexahistidine-tagged versions of PFE0055c and human Hsp70A1A were used for the expression and purification of these proteins from Escherichia coli. Purification was achieved using nickel affinity chromatography. The urea-denaturing method was used to obtain the purified PFE0055c whilst human Hsp70A1A was purified using the native method. PFE0055c could stimulate the ATPase activity of alfalfa Hsp70, although such was not the case for human Hsp70A1A in vitro.

Because it has emerged relatively recently as a field of study, a number of scholars have called for ongoing research in the area, see Swarbrooke (2003), Pomfret (2006), Buckley (2010), Rogerson (2007), and Williams & Soutar (2005). Much of this research, to date, has focused on marketing, injury caused by AT participation, environmental impacts, and the broad notion of “risk” associated with AT. The problem is that little of this work has focused on the significance of the environment in adventure tourism – an important element inherent in its definition. Thus, this thesis aims to move beyond a general risk-based analysis of AT, and focus on the role of the environment. This is done through a human-environment interaction approach undertaken in Tsitsikamma Village where five adventure tourist operations are based. The first step in operationalizing this approach was to develop a conceptual framework that integrated adventure tourism into a general framework on humanenvironment interaction. The second step was to use a research instrument which enabled one to examine the influence of the environment on human motivations and adventure tourism experiences, and not only the influence of humans on the environment. The former was accomplished through questionnaires based on external, environmental elements that attract adventure tourists as well as inherent personality traits impel such tourists to participate in AT. The latter, based on interviews and participant perceptions, sought to establish the significance of impacts on the environment as well as initiatives taken by AT companies to minimize environmental damage. The findings show that although risk/thrill does play a role, AT motivations and experiences are complex and multidimensional. The environment does significantly influence both motivations and even more so AT experiences. Furthermore, there are distinct differences between the motivations and experiences of participants in different types of AT activities, indicating that generalizing the industry as whole neglects these significant differences. In terms of influences on the environment, the results show that most AT operators in the Tsitsikamma have some consideration for environmental protection and take some initiatives to minimize environmental damage, though the degree to which this occurs differs significantly. Furthermore, activity participants do not believe that the activities have a strong negative impact on the environment, though the majority acknowledge small impacts. The types of impacts observed by participants vary significantly between companies. This thesis demonstrates the manner in which the two-way relationship between humans and the environment manifests itself in the commercial AT industry. The result is a well-rounded examination of the influence of human-environment interaction on AT participants.

- Description: Sodwana Bay, situated within the iSimangaliso Wetland Park, is ecologically important as it contains high-latitude corals and the most southerly known population of coelacanths. This thesis utilised stable isotope and lipid analyses to investigate the trophic ecology of the area, in particular, understanding the relative contribution of inshore and offshore primary production to consumers inhabiting intertidal and shallow subtidal, coral reef, deep reef, canyon head and pelagic habitats. Seaweeds, excluding certain species of red seaweeds with highly depleted carbon signatures, and phytoplankton, such as diatoms, were found to be the principal sources of primary production for all consumers. Offshore production was typified by dinoflagellates. Particulate organic matter (POM) was spatio-temporally variable. Three distinct productivity periods related to nutrient cycling were noted with enriched carbon signatures and higher organic matter loads associated with warmer water. Inshore primary production was an important source of carbon to consumers in all habitats with the exception of zooplankton that were more reliant on pelagic primary production. Benthic invertebrates reflected a gradient in the utilisation of inshore production, due to the reduced availability of this source further offshore. Consumers at the furthest sites offshore were found to include a substantial quantity of inshore-derived production in their diets. Fishes, which are more mobile, were found to incorporate a similar proportion of inshore production into their diets regardless of where they were collected from.

Cretaceous dyke swarms and brittle deformation structures in the upper continental crust flanking the Atlantic and Indian margins of Southern Africa, and their relationship to Gondwana break-up

- Description: Permanent brittle deformation of rocks of the upper crust is often manifested in the growth of fractures, or sliding along fractures, which may subsequently be intruded by magma and other fluids. The brittle deformation structures described here include faults, joints and dykes. Brittle deformation structures along passive continental margins result from continental fragmentation and related uplift, as is seen around the southern African margins in response to Gondwana break-up. In many cases the fragmentation is accompanied by significant magmatic events, for example the Cretaceous mafic dyke swarms that form major components of the South Atlantic Large Igneous Province (LIP) and originated during the break-up of West Gondwana (Africa and South America). The magmatic events accompanying the break-up of Gondwana resulted in crustal extension and the formation of joint systems and dyke swarms that exhibit distinct geometric features that appear to display fractal patterns. This work analyses the relationship between the Henties Bay-Outjo Dyke Swarm (HOD) on the west coast of Namibia, and the Ponta Grossa Dyke Swarm (PG) on the coast of Brazil, both of which formed ca. ~130 Ma, to test for their co-linearity and fractal geometry before and during West Gondwana break-up. This was achieved by reconstructing Gondwana‘s plates that contained the PG and HOD swarms, using ArcGIS and Gplates software. The dyke analyses was complemented with a comparative study of joints of the Table Mountain Group quartzites (TMG, ca. 400 Ma) in the Western Cape Province and Golden Valley Sill (GVS, ca. 180 Ma) in the Eastern Cape Province, to compare their fractal patterns and possible relationship. Mapping of joints was carried out in the field with the use of a compass and GPS. The HOD trend is positioned largely NNE > NE, but a NW dyke trend is also common. The dominant joints in the TMG trend NNW > WSW and the GVS joints trend WNW > NNE and others. The GVS and HOD orientations appear strongly correlated, while TMG shows no simple orientation correlation with GVS and HOD. The lack of correlation is attributed to the TMG‘s formation in different host-rocks with variable anisotropy and/or the presence of different mechanical processes acting at a different time in geological history. All mapped dykes and joints were analysed to test for fractal geometry. The fractal dimension results of about 18605 HOD dykes from microscopic to mega scale (0.1 mm – 100 km) shows fractal patterns that range between Df = 1.1 to 1.9; and the fractal dimension of about 1716 joints in the TMG and about 1026 joints in the GVS at all scales range between ca. Df = 1.6 to 1.9. The similarity of the fractal patterns indicates that joints and dykes may have formed in response to similar tectonic stress events; and similar orientations may indicate that joints pre-dated the dyke intrusions. Deploying DNSSEC in islands of security

- Description: The Domain Name System (DNS), a name resolution protocol is one of the vulnerable network protocols that has been subjected to many security attacks such as cache poisoning, denial of service and the 'Kaminsky' spoofing attack. When DNS was designed, security was not incorporated into its design. The DNS Security Extensions (DNSSEC) provides security to the name resolution process by using public key cryptosystems. Although DNSSEC has backward compatibility with unsecured zones, it only offers security to clients when communicating with security aware zones. Widespread deployment of DNSSEC is therefore necessary to secure the name resolution process and provide security to the Internet. Only a few Top Level Domains (TLD's) have deployed DNSSEC, this inherently makes it difficult for their sub-domains to implement the security extensions to the DNS. This study analyses mechanisms that can be used by domains in islands of security to deploy DNSSEC so that the name resolution process can be secured in two specific cases where either the TLD is not signed or the domain registrar is not able to support signed domains. The DNS client side mechanisms evaluated in this study include web browser plug-ins, local validating resolvers and domain look-aside validation. The results of the study show that web browser plug-ins cannot work on their own without local validating resolvers. - Description: The Domain Name System (DNS), a name resolution protocol is one of the vulnerable network protocols that has been subjected to many security attacks such as cache poisoning, denial of service and the 'Kaminsky' spoofing attack. When DNS was designed, security was not incorporated into its design. The DNS Security Extensions (DNSSEC) provides security to the name resolution process by using public key cryptosystems. Although DNSSEC has backward compatibility with unsecured zones, it only offers security to clients when communicating with security aware zones. Widespread deployment of DNSSEC is therefore necessary to secure the name resolution process and provide security to the Internet. Only a few Top Level Domains (TLD's) have deployed DNSSEC, this inherently makes it difficult for their sub-domains to implement the security extensions to the DNS. This study analyses mechanisms that can be used by domains in islands of security to deploy DNSSEC so that the name resolution process can be secured in two specific cases where either the TLD is not signed or the domain registrar is not able to support signed domains. The DNS client side mechanisms evaluated in this study include web browser plug-ins, local validating resolvers and domain look-aside validation. The results of the study show that web browser plug-ins cannot work on their own without local validating resolvers. The tabletop environment also enables users to sit in a natural environment and synchronously communicate without bulky desktops or laptops. Multi-touch tabletops provide the hardware necessary to support co-located GIM. Existing multi-touch interaction techniques were analysed and proved insufficient to support the advanced functional requirements of GIM. The goal of this research was therefore to support co-located GIM by designing new multi-touch tabletop interaction techniques. An architecture was proposed to support co-located GIM with new multi-touch interaction techniques. A software prototype was developed based on the proposed architecture to facilitate the main activities of GIM and to collaboratively compile documents. The prototype was named CollaGIM (Colla – collaborative, GIM – group information management). CollaGIM supports the main activities of GIM using natural gesture interaction on a multi-touch tabletop. - Description: Malaria, caused by an intracellular Plasmodium parasite, remains a devastating disease, having claimed approximately 655 000 lives worldwide in 2010. The Medicines for Malaria Venture suggests a "single-dose radical cure" as the ideal malaria treatment since rapid clearance of blood-stage parasites and symptom relief improves patient compliance and limits drug resistance. Thus, novel antimalarials should be rapid-acting and assessing their rate of activity is critical to drug discovery. Traditional evaluation of this rate by morphological assessments is flawed by highly subjective, operator-specific interpretations, mainly due to heterogeneous parasite morphology under routine culture conditions. This study aimed to develop an alternative, quantitative assay. Energy is vital for the growth and maintenance of all living organisms. Commercially available kits allow rapid quantification of the cell's energy currency, ATP. Therefore, quantification of parasite ATP shows potential for diagnosing abnormal parasite metabolism and the kinetics of drug action. In this study, a rapid protocol for detecting ATP in Plasmodium falciparum parasites using a luminescence-based kit was developed and optimised. Furthermore, luciferase-expressing transgenic parasites, in which luciferase activity is detected using a similar kit, were acquired. The utility of both methods for evaluating the rate of drug-induced stress was explored using antimalarials with varying modes of action and, presumably, rates of activity. Results showed that parasite ATP remained unchanged, increased or decreased during drug exposure. Morphological examinations by light microscopy and a Recovery assay, aided interpretation of the drug-induced changes in parasite ATP. These investigations suggested that unchanged parasite ATP levels reflect poor drug action, increased ATP levels indicate a stress response and partially compromised viability, while significantly reduced ATP reflects severely compromised viability. Concerning the Luciferase assay, parasite luciferase activity decreased during drug exposure, even in the presence of proteasome inhibitors. Changes in parasite ATP and luciferase activity occurred at rates which suggested that chloroquine is slow-acting, mefloquine has a moderate rate of activity and artemisinin is rapid-acting. These findings are compatible with the expected rates of activity of these established antimalarials. Hence, measurement of parasite ATP and/or luciferase activity may support assessments of parasite health and the kinetics of antimalarial action during drug discovery

Diet and foraging distribution of gentoo penguins pygoscelis papua at Cow Bay, Falkland Islands

Furthermore, this information is required for effective management and conservation of these species. For gentoo penguins at the Falkland Islands a previous overlap in dietary items with commercial fisheries was recognised. Despite this the at-sea distribution has been poorly studied and only at a few localities. Therefore, this thesis looks to broaden the knowledge of foraging ecology for gentoo penguins at another site in the archipelago, Cow Bay. The aim was firstly, through stomach content analysis, to determine if diet changed over a large temporal scale (2002/03/04 – 2011/12/13), and between the chick guard and crèche periods of chick rearing. Secondly, through GPS loggers, to assess distribution at sea during the incubation and chick guard periods (2012/13). Additionally, foraging ecology between genders was investigated. Gentoo penguins at Cow Bay foraged primarily on fish (66-99 percent) throughout the entire study period. The principle prey item by mass was Patagonotothen spp., considered previously as discard by fisheries but now an important commercial resource, for all periods (47-78 percent) except that of the 2012/13 crèche period (19 percent). During this period Sprattus fugensis made up the bulk of the diet (52 percent). Cephalopods, particularly Loligo gahi (1-24 percent), were also prominent in the diet and crustaceans contributed negligibly. The diet indicated that gentoo penguins are capable of both a benthic and pelagic feeding strategy which likely reflects changes in local prey abundance. Birds typically conducted foraging activities during daylight hours with greater trip duration, path length, maximum distance and area utilised during the incubation period than the guard. All trips were confined to the continental shelf and within the boundaries of a marine management zone, the Falkland Islands Interim Conservation Zone (FICZ). Males and females fed on the same size and composition of prey items. Trip duration, path length and maximum distance from the colony were not significantly different, although males foraged in a slightly larger area than females marginally to the north. As at-sea distribution was confined within the FICZ this management area provides protection to food resources of gentoo penguins breeding at Cow Bay. At this site, the population is increasing, likely associated with increased prey availability as evidenced by record catches of Patagonotothen spp. in recent years at the Falkland Islands. Continued long term monitoring of gentoo penguin diet and fishery yields to allow detection of possible competition in the future is recommended. As marine sentinels, on-going monitoring of gentoo penguin populations at the Falkland Islands is likely to provide early warning signs of reduced prey availability.
