- Description: Comparison of poliovirus strains was carried out to determine the origin of the virus in two isolates obtained during the 1982 outbreak of poliomyelitis in Gazankulu. Comparisons of the outbreak isolates with vaccine and wild-type strains of the same poliovirus type were carried out using four biochemical techniques. SDS-polyacrylamide gel electrophoresis (SDS-PAGE), two-dimensional thin-layer chromatography (TLC) and reversed-phase high-performance liquid-chromatography (RP-HPLC) were used for comparing viral capsid proteins. Comparison of poliovirus strains at a genetic level was carried out using two-dimensional oligonucleotide mapping of viral RNA. Results showed the type 1 poliovirus isolate, 5061, to be a novel wild-type poliovirus. The type 2 isolate, 5068, was closely related to the poliovirus type 2 Sabin vaccine strain, P712. It was concluded that the intrinsic variability of poliovirus strains was responsible for the appearance of isolate 5068

It is an integral membrane protein composed of four different subunits, tightly associated with other functional, but non-specific proteins. Purification of the nicotinic cholinergic receptor by chromatofocusing demonstrates an improved method over that of affinity and ion-exchange chromatography. Gel chromatography and SDS-polyacrylamide gel electrophoresis show evidence of four subunits; a(40-44 kDa), 6(53 kDa ),'Y(63 kDa) and 6(66 kDa) despite some degradation of receptor molecules by intracellular proteases. Spectrophotometric and fluorimetric studies of receptor-ligand interactions, show the functional and chemical integrity of the receptor to remain intact after solubilisation. The effect of cholinergic ligands on purified receptor preparations indicate quenching of the intrinsic fluorescence of the receptor. Agonists, like acetylcholine, bind and cause local conformational transitions, changing the active region from a hydrophobic to a hydrophilic environment. This phenomenon is illustrated by the 10-fold increase in fluorescence when the receptor is in a desensitised state. Antagonists, such as d-Tubocurarine, block this conformational transition. In vitro rectus abdominis muscle preparations . show the nitrosamines, dimethylnitrosamine and diphenylnitrosamine, to be true agonists of the nAChR. However their low affinity and specificity for the receptor precludes them as photoaffmity labelling agents. Photoactivation of dimethylnitrosamine occurs when associated with an acidic hydrogen at the active site of the receptor, suggesting energy-transfer labelling to be more facile than photoaffmity labelling. The membrane-bound receptor, in the presence of these nitrosamines, undergoes conformational transitions regulating the opening and closing of the ion-channel. Desensitisation and receptor activation are shown to involve one and the same molecular transition.

First the study sought to establish to what degree was enmeshment between the patient and his mother responsible for his maladaptive behaviour. The second aspect was to establish the extent to which the patient responded to psychotherapy. The patient was a 20 year old male university student. He was experiencing a number of problems including difficulty in interpersonal relations, deteriorating academic performance, depression, anxiety and uncertainty about career choice and about the future. The history revealed that the patient had very close and enmeshed relations with his mother. The mother was a very domineering person and had great influence on her son. On the other hand his father was weak and emotionally unavailable. The patient had never been away from home until after he graduated from High School and decided to join the army. It was then his problem began to manifest. He left the army and registered at university. However, the problems continued and even increased in intensity. It was hypothesized that as a result of enmeshment with his mother and the fact that his mother was a domineering personality, he never had the opportunity to learn social skills that would have enabled him to emancipate himself from his mother . Thus when he moved out of home to be on his own, first in the army and later at university, he could not cope. He lost self-confidence, became passive, indecisive and dependent. Psychotherapy was successful in helping him gain insight into the genesis of his problem. This insight resulted in the patient's efforts towards individuation.

- Description: This study explores the theoretical consequences of Freud's renunciation of the seduction theory. The dissertation defends the thesis that the seduction theory was shaped as much by Freud's adherence to the nomonological model as by the empirical evidence of child sexual abuse. A renunciation of the seduction theory was inevitable, not because the accounts of the daughters were lies, but because the methodology was inappropriate. The nomonological model obscured the emotional structure of the nuclear family in which the structure itself, through which sexuality emerged, directed the girl's entrance into womanhood and caused the woman's dis-ease. Freud's methodology forced him to isolate an event as cause of an illness and to attribute the event to an agent. The universal perversity of the Victorian father thus became the central theme around which an explanation of a female disease was built. When this theme became theoretically untenable, Freud renounced the seduction theory and, still using the nomonological model, built up the construct of the Oedipus complex in which the father was vindicated. In order to exonerate the father, the transactions through which the child's libido developed were represented as originating in inherent tendencies. As a result, the hierarchical nature of the interaction between parent and child was distorted, and this led to the formulation of the distinction between real events and fantasies as a basic premise on which the difference between the pleasure principle and the reality principle rests. This formulation gave rise to the sharp duality between fantasy and reality which eventually compelled him to separate psychic reality and social reality. The theoretical structure built on this duality could not but fuse hysteria, masochism and "normal" femininity into an explanation of the female state, and obscure the essential social relations between men and women which were structured in terms of dominance and submission. The thesis traces the journey from the perverted father as cause of a female disease, hysteria, to the theoretical conjunction of masochism and hysteria. It comes to the conclusion that Freud's model is unable to explain the self-mutilation of the hysteric; nor is it capable of explaining the hysteric's refusal to participate in the circuit of symbolic exchanges which constituted Victorian society. The study further attempts to understand hysteria in terms of the complex interlacing of fact and fantasy and tries to show that fantasy was rooted in the facts of Victorian culture.

Operating in turbulent times: an investigation into the characteristics of strategic flexibility exhibited by South African export companies

- Description: Broadly stated, the major objective of this study was to investigate the extent to which South African export companies exhibited characteristics of strategic flexibility, which is seen to be the most viable strategic option for a firm operating within a turbulent environment. This entailed:- (1) Determining the degree of environmental turbulence as perceived by South African export companies and identifying the most important elements of the external environment as perceived by these companies. (2) Identifying the nature and strength of the various components of strategic flexibility possessed by South African export companies facing environments of either low, medium or high turbulence. (3) Identifying the nature and strength of the various components of strategic flexibility possessed by South African export companies of different size. In order to elicit this information, 1361 questionnaires were posted to the managing directors of export firms who were members of the South African Foreign Trade Organization. The major findings of this study indicate that:- (1) Political elements of the remote environment contribute most to environmental turbulence. (2) Respondents identified economic elements as being the most important in terms of their potential impact on the long term profitability of the firm. (3) Although at present the majority of the firms surveyed were classified as operatIng in medium levels of turbulence, in the future they perceive the external environment as becoming very turbulent. (4) Generally speaking, the firms surveyed are ill-equipped to operate in a turbulent environment as they displayed signs of inflexibility in certain key aspects of their business. In this regard, particular mention must be made of their decision-making processes, the extent to which they had specialist planners, the techniques used to identify future trends in the environment, the extent to which computer technology was utilised in the firm and their structure. (5) Finally, the results do not generally support the view that a relationship exists between the perceived degree of turbulence in the environment and the extent to which strategic flexibility exists in the firm. Often, however, the size of tne firm was more of a determining factor on the components of strategic flexibility than the degree of turbulence in the environment. In this regard, particular mention must be made of the impact of size on the planning processes of the firm, the techniques used to monitor trends in the environment and the emphasis placed on management development and training.

The results demonstrate the effectiveness of one of these methods in the production of a flat base level. The filamentary structure of the North Polar Spur is enhanced and results are obtained which support current theories of the origin of this object. The HII region surrounding Ophiuchi is examined in detail. A number of parameters are derived for the HII region. The spur associated with the HII region S54 is also examined. - Description: The purpose of this study was to examine possible differences between the field and laboratory ratings of perceived exertion (RPE) when the performance and physiological measures for the two conditions were equated. Furthermore, the interactive effects of local, central and overall RPE were examined. Finally, the question of the potential effects of attitudes on RPE was addressed. Eleven well-conditioned adult male marathon runners voluntarily participated in the study. After a period of treadmill habituation, biographical and anthropometric data were collected. This was followed by a VO₂ max test and a speed-matching session at 70% of VO₂ max to determine overground running speed at 3.8% and 7. 5% grade. Subjects then completed an attitudinal questionnaire and ran 4km overground. Finally, the above test was repeated on the treadmill, with the gradient and running speed of the overground condition being replicated. Physiological measures and differentiated RPE were obtained during the final two sessions. There were no physical environmental, task characteristic or performance differences between the overground and treadmill conditions. No heart rate or VO₂ differences were observed between the two conditions, but VE was significantly elevated in the laboratory. Local and overall RPE were significantly higher in the laboratory than in the field, but there was no difference for central RPE. Attitudes were more favourable towards the field than towards the laboratory work task. The results suggest that neither heart rate nor VO₂ are major factors directly influencing the perception of exertion. VE however appears to be a potent central signal mediating RPE. The results also indicate that local factors play a more important role in the perception of exertion than was previously thought. Attitudes towards a work task could possibly mediate the self-reports of exertion. The findings of this study also suggest that RPE are influenced by cognition to a large degree. Finally, environmental cues, or the ambience of a particular working environment, can exert a substantial influence on RPE. Direct perceptual translations from laboratory to field situations may therefore be invalid.

- Description: Since in our educational system a great deal of learning is supposedly dependant upon a child's ability to read and assimilate information from textbooks, this study investigated what reading skills are required by a second language reader of English to read textbooks with comprehension and understanding in relation to the reading skills of a competent reader and how Ciskeian Standard 6 pupils perform in relation to a Geography text prescribed at that level. A test, designed to measure eight reading comprehension skills, was given to a sample of 250 children from four schools in Zwelitsha, Ciskei, to establish whether the subjects are able to: a) give the literal meaning of words; b) derive the appropriate meaning of an ambiguous word from the context in which it appears; c) find answers to questions by making direct reference to the text; d) identify the major points and details in a text; e) use the information in the text to predict what the writer is going to talk about next; f) find the referent for anaphoric terms; g) use discourse markers to predict information/meaning to come, and see the relationships between what they have just read and what they are about to read; h) activate and use the background knowledge and schemata that they have to understand the text topic.The results of this study indicate that these children are: a) unfamiliar with the structure of expository texts; b) linguistically bound to a text and that they fail to use linguistic and contextual clues even when they are explicit in the text. The study also shows that the ability to make inferences and predictions is determined to a large extent by the prior knowledge and background experience that a pupil brings with him to the text and by his ability to activate that background knowledge. The findings suggest that in the English classroom, in an English as a second language (L2) medium situation, the L2 teacher has a responsibility to prepare the child for the study, which includes reading, writing, listening and speaking, of all subjects across the curriculum through the second language, which is the medium of instruction

This idea has spread to various countries and today teaching and learning by correspondence takes place in most countries of the world. It is different to conventional classroom education and thus has certain unique characteristics. The definition, characteristics, background and methodology are important if the problems involved are to be understood. These aspects indicate how effective and popular this form of study has become. Particular attention is given to the teaching of in-service teachers who are upgrading their qualifications. The existing and proposed College history syllabi are examined in as far as their appropriateness and relevance are concerned. Emphasis is given to history at Graaff- Reinet College because the author is a history lecturer at this institution. Syllabus differentiation in mathematics in Ciskei : the reality as against intention

- Description: The topic of this half-thesis was conceived after the author had noted with interest, that an alarmingly high number of pupils in the Mdantsane Central District of the Ciskei were registering for Higher Grade (HG) mathematics for their final (Standard 10) year. What mattered most was that the failure rate was abnormally high. As a teacher of the subject for 14 years as at the end of 1988, the author had seen very weak pupils passing through his hands who had nevertheless insisted on registering for the examination in HG mathematics. A look through the Human Sciences Research Council (HSRC) Report of 1971 which recommended the syllabus differentiation, revealed that the intention of the relevant HSRC Committee was to offer a subject at a level in accordance with a pupil's interest, aptitude and ability. In a survey undertaken by the author, samples of teachers, pupils and parents indeed confirmed the existence of a very high number of HG candidates, as suspected. The survey also revealed an aversion to the standard grade (SG) course by some pupils, teachers and even principals. A desire for university education was shown to be in the minds of most pupil respondents. Well sourced registration statistics clearly showed that the Department of Education and Training (DET) in the Republic of South Africa and the Department of Education in the Ciskei had by far the greatest proportion of pupils on the HG in mathematics in comparison with the other race groups.Standard 10 (Std 10) results for two years taken separately confirmed the author's suspicion that aptitude and/or ability were not taken into consideration by most pupils, in the choice of grade. The results were not good at all. In most cases the uninformed pupil was acting alone in taking the important decision, perhaps with only peer group pressure to contend with. Three independent opinions suggested that there was probably a general feeling that Black education was already inferior, even without the syllabus differentiation hence the reluctance to register for the SG option. Furthermore, the investigation indicated that there was no adequate provision for technical education for Blacks in either the RSA or Ciskei which would provide the alternative to a university career for a pupiL Amongst other things, a mild form of pupil screening and a well structured careers guidance programme are suggested in the concluding chapters

The effects of a South African Black youth jive on selected biophysical physiological and psycho-social parameters

- Description: This study investigated the effects of a South African Black youth jive on selected Biophysical, Physiological and Psycho-social parameters, using 31 Black youths, males and females (mean age 19.29 yrs) as subjects. All subjects participated in the pre- and post-programme testing protocols (acting as their own control) and in a 7-week jive programme. While the female subjects were significantly (p<0.05) heavier with a greater percentage body fat than their male counterparts, a two factor analysis of variance revealed no significant changes in body composition (p<0.05) of either sex group. However, significant improvements did occur in the cardio-respiratory . parameters of working and recovery heart rates, predicted V0₂ max, and the anaerobic capacity. Here, the males exhibited superior cardio-respiratory qualities and performed better in all motor fitness parameters except flexibility, where no significant sex difference occurred. Also, there were significant improvements in all motor fitness tests with the exception of power (as tested in the 18-Item Illinois test). No significant differences occurred between male and female psycho-social responses with no changes occurring after the 7- week programme. - Description: This study investigated the effects of a South African Black youth jive on selected Biophysical, Physiological and Psycho-social parameters, using 31 Black youths, males and females (mean age 19.29 yrs) as subjects. All subjects participated in the pre- and post-programme testing protocols (acting as their own control) and in a 7-week jive programme. While the female subjects were significantly (p<0.05) heavier with a greater percentage body fat than their male counterparts, a two factor analysis of variance revealed no significant changes in body composition (p<0.05) of either sex group. However, significant improvements did occur in the cardio-respiratory . parameters of working and recovery heart rates, predicted V0₂ max, and the anaerobic capacity. Here, the males exhibited superior cardio-respiratory qualities and performed better in all motor fitness parameters except flexibility, where no significant sex difference occurred. Also, there were significant improvements in all motor fitness tests with the exception of power (as tested in the 18-Item Illinois test). No significant differences occurred between male and female psycho-social responses with no changes occurring after the 7- week programme. The survey of students' opinions included students from two universities. The major findings showed that students, regardless of whether or not they had studied accounting at secondary level, believed those who had to be advantaged in the first year financial accounting course. The majority of respondents indicated that high school accounting was, in their opinion, a desirable preparation for the university course. For the purposes of this study, the above-mentioned stage was defined as that period in the family when the youngest child matriculates. Thirty-five empty-nest couples were interviewed during 1984, in the city of Port Elizabeth, South Africa. The mean age of the subjects was 49.5, and the standard deviation was 4.9. The couples were asked about their attitudes towards the empty-nest, using a structured questionnaire, the Family Attitude Survey (FAS). This survey consisted of nine-point attitude statements, which focused on theoretical issues pertinent to this stage, viz. children are on-time or off-time with regard to major life events, impact of children leaving home on the parents, degree of parental involvement with children, parent-child relationships, ageing, sexuality, menopause, work-career, and attitudes towards the past, future and death. The general purpose of the present study was to determine the extent to which the empty-nest stage of life constitutes a negative crisis period, or a positive period of stability and growth for the empty-nest parents. Overall, it was concluded that the empirical evidence depicting the empty- nest stage of life as a positive period of stability and growth rather than a negative crisis period, is persuasive for some of the empty-nest parents in the present study, in view of the empirical findings regarding certain of the above-mentioned theoretical issues examined in the present thesis. Notwithstanding this, it was deemed essential to qualify the above conclusion, given the fact that the same and other respondents experienced difficulty with the following issues, viz. children being off-time with regard to major life events, the departure of children from the home, overinvolvement with children, problematic relationships with them, perceptions of themselves as failures as parents, inability to accept their own ageing, problems with changing sexuality, diminishing enjoyment in their occupations, and lack of prospects for future career advancement, negative preoccupation with the past and future, anxiety about death, and an impoverished marital relationship. It was also demonstrated empirically that wives experience particular psychological problems at this time, viz., firstly, they are more adversely affected by their children's departure from the home than their husbands, secondly, they undergo a rehearsal for widowhood more frequently than them, and thirdly, a minority of them are unable to come to terms with the menopause. Finally, the finding that the majority of wives experienced relief with the onset of the menopause when viewed from the perspectives of general emotional impact, children, and the spousal relationship, conflicts with existing theories in this area. However, it is supported by and large by the majority of empirical studies.

- Description: This thesis deals with the history of the amaHleke people and Pirie Mission, which have become so closely associated that they cannot be separated. It covers the period from the time of Chief Hleke to 1967, the year in which the amaHleke cheiftainship was resuscitated. The first chapter relates the origin of the amaHleke, from the time of Hleke himself (17th century) to Jwarha (about 1820). It explains the relationship between the different branches of the Hleke royal line, and it covers the Hleke settlement at the Mgqakhwebe river. The second chapter deals with the establishment of Pirie Mission by the Presbyterian missionaries John and Bryce Ross. It discusses the various aspects of the mission operation, and explains why and how the amaHleke opposed it. But the situation changed as a result of the 1850-3 Frontier War. Whereas the other Xhosa were expelled from their lands, the Hleke connection with Pirie Mission enabled them to stay on. The Hleke were therefore united with the mission, whether they liked it or not. The remainder of the chapter describes the educational and cultural changes which the mission imposed on them. The third chapter covers economic change at Pirie. Like other mission stations, it was converted from communal to individual land tenure. This was opposed by Chief Jwarha as a blow to his authority, but it did not result in the growth of a peasant class. The chapter concludes with the implementation of betterment in 1963. The fourth chapter explains what happened to the mission after the death of Bryce Ross. The Ross missionaries had frustrated black aspirations in teh church. This was especially frustrating to Burnet and Ntsikana Gaba, the great-grandsons of the prophet Ntsikana. Burnet broke away under the banner of the "Wee Free" branch of the Church of Scotland. This church also could not accommodate Burnet's aspirations. The remainder of the chapter deals with educational developments, with an emphasis on the introduction of Bantu Education. The last chapter deals with the political history of Pirie after the death of Chief Jwarha. The Cape government tried to replace chieftainship by a headman and a Village Management Board. But the Board did not function satisfactorily, and it was scrapped in 1921. Pirie continued to be administered by headmen. Applications for the revival of chieftainship were turned down, partly because there was no agreement on Jwarha's heir. However, this was finally resolved in 1967 with the appointment of Chief Pani Busoshe.
