Agricultural disturbance affects taxonomic and functional diversity of Afrotropical macroinvertebrate composition in a South African river system

- Description: Developing species-level biomonitoring tools to monitor riverine systems threatened by anthropogenic pollution, including local agricultural activities in the Afrotropical region, remain a critical challenge. Here we explored the utility of taxonomic-based (diversity, richness, and composition) as well as functional-based (functional diversity) indices to examine the effects of agricultural disturbance on macroinvertebrate communities in the Kat River, Eastern Cape Province of South Africa. We collected physicochemical parameters and macroinvertebrates from eight sites delineated into four land-use categories (highly impacted, HIC; impacted category, IC; moderately impacted, MIC and least impacted, LIC) using agricultural land cover. We recorded 70 macroinvertebrate taxa belonging to 49 families and 48 genera in the Kat River. Redundancy analysis (RDA) and Pearson correlation analysis revealed that species of Lymnaeidae, Belostomatidae, Planorbidae and Libellulidae families and class Oligochaeta were tolerant to agricultural disturbance, as they were dominant in the highly impacted sites and were significantly associated with high salinity, temperature, total dissolved solids (TDS), flow velocity and nutrients. Conversely, species of Baetidae, Caenidae and Potamonautidae were negatively associated with the highly impacted sites and high salinity, temperature, and nutrients. On the other hand, taxonomic indices showed more sensitivity to indicators of agricultural pollution than functional indices, with taxon richness, Shannon index, Simpson's index and Margalef's index declining significantly in the highly disturbed sites (p less than 0.05). They were negatively associated with high electrical conductivity, large river width, and high nitrite and nitrate concentrations; hence they were identified as indicator metrics sensitive to agricultural pollution. Overall, our study revealed that agricultural disturbance could differentially affect the structure and function of macroinvertebrates, and indicator taxonomic and functional indices were identified for long-term monitoring of rivers that drain agricultural landscapes.

- Description: Internet security is an ever-expanding field. Cyber-attacks occur very frequently, and so detecting them is an important aspect of preserving services. Machine learning offers a helpful tool with which to detect cyber attacks. However, it is impossible to deploy a machine-learning algorithm to detect attacks in a non-centralized network. Software Defined Networks (SDNs) offer a centralized view of a network, allowing machine learning algorithms to detect malicious activity within a network. The InSDN dataset is a recently-released dataset that contains a set of sniffed packets within a virtual SDN. These sniffed packets correspond to various attacks, including DDoS attacks, Probing and Password-Guessing, among others. This study aims to evaluate various machine learning models against this new dataset. Specifically, we aim to evaluate their classification ability and runtimes when trained on fewer features. The machine learning models tested include a Neural Network, Support Vector Machine, Random Forest, Multilayer Perceptron, Logistic Regression, and K-Nearest Neighbours. Cluster-based algorithms such as the K-Nearest Neighbour and Random Forest proved to be the best performers. Linear-based algorithms such as the Multilayer Perceptron performed the worst. This suggests a good level of clustering in the top few features with little space for linear separability. The reduction of features significantly reduced training time, particularly in the better-performing models.

- Description: In this work, cobalt tetra phenoxy acetic acid phthalocyanine (CoTAPc) is investigated as an electron mediator, immobilization platform for an HB5 aptamer and to enhance the electrochemical signal for the detection of human epidermal growth factor receptor 2 (HER2). Furthermore, the CoTAPc was combined individually with sulphur/nitrogen doped graphene quantum dots (SNGQDs), gold nanoparticles (AuNPs) and cerium oxide nanoparticles (CeO2NPs), on a glassy carbon electrode (GCE) via sequential adsorption. The CoTAPc and SNGQDs were also π-π stacked, used for electrode modification similarly to the rest of the other surfaces and applied towards the electrochemical detection of HER2. The designed sensors were characterized using electrochemical impedance spectroscopy (EIS). The designed aptasensors showed detection limits as low as 6.0 pg/mL. The real life applicability of the designed aptasensors was tested in human serum samples. The aptasensors showed great storage stability, sensitivity and specificity towards HER2, implying great potential for applications in early diagnosis of breast cancer.

- Description: Iris pseudacorus is both a prized ornamental and an invasive aquatic plant that tends to grow dense monospecific stands, displacing the local vegetation and altering the hydrology of freshwater ecosystems. Originally from Europe, this species has historically invaded North America, China and Japan, and more recently spread through Argentina, South Africa and Australasia, where it is now a target for biological control. Field surveys within its native range have led to the selection of three candidate biocontrol agents. Prioritizing the best candidates for different regions constitutes a critical step, which could save significant time and resources before further cost-intensive experimental studies are conducted. Climate change is seldom taken into consideration in the prioritization process. In this regard, climatic suitability can be used to model the potential distributions of weeds and their candidate agents, both in space and time, thus allowing to identify areas at risk of invasion and predict where agents will be able to establish long-term. Accordingly, the objectives of this work were (i) to predict I. pseudacorus invasions and range shifts in the context of climate change; (ii) to identify wetland areas most at risk of invasion under present and future climatic conditions; and (iii) to prioritize the best suite of candidate biocontrol agents for different invaded ranges, worldwide. To do so, we modelled the present and future (2040–2060) climatic suitability of I. pseudacorus and its candidate agents using the software MaxEnt. Our results highlight a clear distinction between predictions for the Northern and Southern Hemispheres. In North America and eastern Asia, the area climatically suitable for I. pseudacorus is expected to increase and shift northwards. As for its biocontrol agents, very low suitability is predicted across these regions, further decreasing under future climatic conditions. On the other hand, climatically suitable areas for the plant in South America, southern Africa and Australasia are predicted, on average, to reduce in response to climate change. A decrease in climatic suitability is also expected for its candidate biocontrol agents which, however, would still maintain a significant range overlap with their host. These results can be used to prioritize areas most at risk of invasion and identify which combination of candidates could potentially provide the best level of control across different invaded ranges.

- Description: The network security sector has observed a rise in severe attacks emanating from the darknet or encrypted networks in recent years. Network intrusion detection systems (NIDS) capable of detecting darknet or encrypted traffic must be developed to increase system security. Machine learning algorithms can effectively detect darknet activities when trained on encrypted and conventional network data. However, the performance of the system may be influenced, among other things, by the choice of machine learning models, data preparation techniques, and feature selection methodologies. The histogram-based gradient boosting strategy known as categorical boosting (CatBoost) was tested to see how well it could find darknet traffic. The performance of the model was examined using feature selection strategies such as correlation coefficient, variance threshold, SelectKBest, and recursive feature removal (RFE). Following the categorization of traffic as “darknet” or “regular”, a multi-class classification was used to determine the software application associated with the traffic. Further study was carried out on well-known machine learning methods such as random forests (RF), decision trees (DT), linear support vector classifier (SVC Linear), and long-short term memory (LST) (LSTM). The proposed model achieved good results with 98.51% binary classification accuracy and 88% multi-class classification accuracy.

- Description: Tuberculosis, a predominantly pulmonary pathology, is currently the deadliest infection worldwide. Its treatment is based on combination therapy involving selected antimicrobials including Isoniazid. However, physicochemical properties of isoniazid negatively affect the clinical performance of current tuberculosis regimens, causing drug resistance development and increasing mortality rates. Liposomal encapsulation improves antituberculosis drug delivery; however, nano-formulation of isoniazid remains challenging due to its small molecular size and high hydrophilicity. Therefore, this study aimed to derivatize isoniazid and formulate a controlled delivery system using the concept of drug-in-cyclodextrins-in-liposomes to enhance drug biopharmaceutical properties. A prodrug of isoniazid was synthesized and screened for its ability to form stable complexes with α, β, and γ cyclodextrins. A selected inclusion complex with β-cyclodextrin was encapsulated in liposomes and assessed for controlled release of isoniazid. Successful formation of a 1:1 complex was established and characterized, followed by molecular modeling studies to demonstrate strength of the interactions within the complex and predicted complex structure. The inclusion complex was successfully encapsulated in liposomes using the thin film hydration method and the ethanol injection ultrasonic dispersion, with the latter giving the best results. These findings demonstrate the potential.

- Description: Internalised stigma is highly prevalent among people with mental illness. This is concerning because internalised stigma is often associated with negative consequences affecting individuals’ personal, familial, social, and overall wellbeing, employment opportunities and recovery. Currently, there is no psychometrically validated instrument to measure internalised stigma among Xhosa people in their home language. Our study aimed to translate the Internalised Stigma of Mental Illness (ISMI) scale into isiXhosa. Following WHO guidelines, the ISMI scale was translated using a five-stage translation design which included (i) forward-translation, (ii) back-translation, (iii) committee approach, (iv) quantitative piloting, and (v) qualitative piloting using cognitive interviews. The ISMI isiXhosa version (ISMI-X) underwent psychometric testing to establish utility, within-scale validity, convergent, divergent, and content validity (assessed using frequency of endorsements and cognitive interviewing) with n = 65 Xhosa people with schizophrenia. The resultant ISMI-X scale demonstrated good psychometric utility, internal consistency for the overall scale (α = .90) and most subscales (α > .70, except the Stigma Resistance subscale where α = .57), convergent validity between the ISMI Discrimination Experiences subscale and the Discrimination and Stigma (DISC) scale's Treated Unfairly subscale (r = .34, p = .03) and divergent validity between the ISMI Stigma Resistance and DISC Treated Unfairly subscales (r = .13, p = .49). But more importantly the study provides valuable insights into strengths and limitations of the present translation design. Specifically, validation methods such as assessing frequency of endorsements of scale items and using cognitive interviewing to establish conceptual clarity and relevance of items may be useful in small piloting sample sizes.

- Description: Despite that counselling psychologists represent a substantial group of registered psychologists in South Africa, literature specifically on the sub-speciality within the country is limited. The aim of this scoping review was to provide a comprehensive summary of the literature available on counselling psychology in South Africa and examine the extent to which literature is available from a lifespan or career-stage perspective. Three electronic databases (EBSCOhost, Sabinet®, and PubMed) were searched for articles published between 2000 and 2021. Titles and abstracts were reviewed, and data extracted and synthesised thematically. Of 507 citations identified, 10 met the inclusion criteria. Findings indicate that literature on counselling psychology in South Africa is scarce, subject to methodological limitations, and dominated by a small number of authors conducting multiple analyses on the same sets of data. Furthermore, literature on counselling psychologists at key career stages across the professional lifespan is largely missing from the professional discourse. Emphasis is instead placed on counselling psychology as embedded in the sociopolitical history of South Africa, professional identity, the contemporary status of the profession, professional threats and challenges, and the profession’s future promise. Our review highlights the need for more empirically informed studies making use of different methodologies, involving multiple authors with diverse backgrounds, tracking employment trends, and soliciting first-person accounts of counselling psychologist’s experiences at key career stages. Without doing so, ideas about how best to support and utilise this particular group of practitioners may be misguided, in turn compromising the successful provision of mental health care within the country.

- Description: In the present work, the performance of aptamer and antibody bioreceptors for the detection of the human epidermal growth factor receptor (HER2) cancer biomarker on a glassy carbon electrode is reported. The carboxylic acid group rich graphene quantum dots (GQDs) modified with gold nanoparticles and a porphyrin binuclear framework (CoP-BNF) were used to modify the glassy carbon electrode. The aptamer and antibody were both amine functionalized and attached to GQDs and CoP-BNF through an amide bond. The designed immunosensors and aptasensors in this work were characterized using electrochemical impedance spectroscopy. The aptasensors, compared to the immunosensors gave better limit of detection values. The aptasensor outperforms the immunosensor in terms of its reusability and storability, while the immunosensor could not be regenerated for subsequent experiments. The potential applicability of all sensors in this work was also investigated, by detection of HER2 in spiked human serum with acceptable results.

- Description: Emerging insects transfer valuable lipids originating in aquatic food sources to terrestrial consumers. The objective of this study was to determine how the export of physiologically important fatty acids from a river to adjacent land via insect emergence relates to the type and quality of the aquatic food consumed, and which emerging insects were primarily responsible for these fluxes. We ran mixing models and hypervolumes incorporating stable carbon isotope ratios of basal resources and emergent invertebrates to determine the major contributors to insect larvae diets. Our data revealed that aquatic food sources (epiphyton, epilithon and suspended particulate matter) were the major organic matter sources (more than 50 %) supporting consumers (Diptera, Ephemeroptera, Odonata and Trichoptera), with allochthonous food sources (C3 plants) being of importance at upstream sites. We calculated fluxes of highly unsaturated fatty acids (HUFAs) via emerging insects and found that these corresponded with the spatial and temporal patterns in the nutritional quality of these same basal resources in the river (quality measured as concentrations of HUFAs). These patterns provide evidence of a direct coupling between food quality and trophic subsidy fluxes from water to land, particularly by emergent dipterans and ephemeropterans.

- Description: Plasmonic gold nanoparticles significantly improved the efficiency of a TiO2 and Eosin Y based dye-sensitized solar cell from 2.4 to 6.43%. The gold nanoparticles’ sizes that were tested were 14 nm, 30 nm and 40 nm synthesized via the systematic reduction of citrate concentration using the Turkevich method. Prestine TiO2 without plasmonic gold nanoparticles yielded an efficiency of 2.4%. However, the loading of 40 nm gold nanoparticles into the TiO2 matrix yielded the highest DSSC efficiency of 6.43% compared to 30 nm (5.91%) and 14 nm (2.6%). The relatively high efficiency demonstrated by plasmonic gold nanoparticles is ascribed to light absorption/scattering, hot electron injection and plasmon-induced resonance energy transfer (PIRET), influenced by the size of the gold nanoparticles.

- Description: The information on a licence plate is used for traffic law enforcement, access control, surveillance and parking lot management. Existing li-cence plate recognition systems work with clear images taken under controlled conditions. In real-world licence plate recognition scenarios, images are not as straightforward as the ‘toy’ datasets used to bench-mark existing systems. Real-world data is often noisy as it may contain occlusion and poor lighting, obscuring the information on a licence plate. Cleaning input data before using it for licence plate recognition is a complex problem, and existing literature addressing the issue is still limited. This paper uses two deep learning techniques to improve li-cence plate visibility towards more accurate licence plate recognition. A one-stage object detector popularly known as YOLO is implemented for locating licence plates under challenging situations. Super-resolution generative adversarial networks are considered for image upscaling and reconstruction to improve the clarity of low-quality input. The main focus involves training these systems on datasets that include difficult to detect licence plates, enabling better performance in unfavourable conditions and environments.

- Description: The socio-economic and ecological impacts of invasive alien aquatic plant (IAAP) species have been well studied globally. However less is known about ecosystem recovery following the management of IAAP species. This study employed a before-after study design to investigate ecological recovery following the management of Salvinia molesta D.S. Mitchell, at four field sites in South Africa. We hypothesized that the presence of S. molesta would have a negative impact on the ecosystem food web structure, and that following S. molesta control, the systems would show positive ecosystem recovery. Aquatic macroinvertebrate and macrophyte samples collected before and after mechanical or biological control of S. molesta, were analysed for δ13C and δ15N stable isotopes. Salvinia molesta infestations negatively impacted the food web structure, indicated by reduced food chain length, trophic diversity and basal resources. This represented an altered aquatic food web structure, that in some cases, led to the collapse of the aquatic community. In contrast, after either mechanical or biological control, there were increases in food chain length, trophic diversity and abundance of energy resources accessed by consumers, indicating improved food web structure. Although the study showed positive ecosystem recovery following control, we noted that each control method followed a different recovery trajectory. We conclude that S. molesta invasions reduce aquatic biodiversity and alter ecosystem trophic dynamics and related ecosystem processes, necessitating control.

- Description: Evidence-informed decision-making is in increasing demand given growing pressures on marine environments. A way to facilitate this is by knowledge exchange among marine scientists and decision-makers. While many barriers are reported in the literature, there are also examples whereby research has successfully informed marine decision-making (i.e., ‘bright-spots’). Here, we identify and analyze 25 bright-spots from a wide range of marine fields, contexts, and locations to provide insights into how to improve knowledge exchange at the interface of marine science and policy. Through qualitative surveys we investigate what initiated the bright-spots, their goals, and approaches to knowledge exchange. We also seek to identify what outcomes/impacts have been achieved, the enablers of success, and what lessons can be learnt to guide future knowledge exchange efforts. Results show that a diversity of approaches were used for knowledge exchange, from consultative engagement to genuine knowledge co-production. We show that diverse successes at the interface of marine science and policy are achievable and include impacts on policy, people, and governance. Such successes were enabled by factors related to the actors, processes, support, context, and timing. For example, the importance of involving diverse actors and managing positive relationships is a key lesson for success. However, enabling routine success will require: 1) transforming the ways in which we train scientists to include a greater focus on interpersonal skills, 2) institutionalizing and supporting knowledge exchange activities in organizational agendas, 3) conceptualizing and implementing broader research impact metrics, and 4) transforming funding mechanisms to focus on need-based interventions, impact planning, and an acknowledgement of the required time and effort that underpin knowledge exchange activities.

- Description: The emergence of antimicrobial resistance has made the development of photodynamic therapy (PDT) related applications essential, since microorganisms can not form resistance to this method. Porphyrins are well-known photosensitizers for PDT related applications. Thus, the present study outlines the synthesis, characterization and evaluation of the utility of palladium porphyrins and their chitosan inclusion complexes as photosensitizer dye in photodynamic antimicrobial therapy (PACT). Before in vitro cell studies, the photophysical-chemical studies of all obtained structures were performed in solution. It was observed that the immobilization of the porphyrins into the chitosan influenced the photophysical-chemical and PACT activity properties. The determined fluorescence quantum yield was very low, in the range of 0.007–0.028 for all samples indicating the efficient triplet state population to cause high singlet oxygen quantum yield (ΦΔ). The measured ΦΔ values were in the range of 0.51–0.61 for the porphyrins and 0.53–0.66 for porphyrin chitosan immobilization complexes. Our results demonstrate that the PACT activity of cationic porphyrin (P3) and its chitosan immobilization form (P3-Ct) were more efficient in decreasing the number of viable cells up to 100% in vitro.

- Description: Synthetic molecularly imprinted polymers (MIPs) based functional materials become the best alternatives to alleviate the stability and cost issues related to biological receptors commonly used in biochemical sensors. To address this demand, we report the development of a highly selective and sensitive MIPs-based electrochemical sensor for the detection of 17β-estradiol (E2). The sensor was prepared based on the MIP-graphene oxide (GO)-silver nanoparticle (AgNP) nanocomposite functional materials electrodeposited on the surface of the glassy carbon electrode (GCE). At first, AgNp formation was facilitated by using ascorbic acid to reduce and stabilize it. A very stable MIP-GO-AgNP sensing layer with multifunctional units were formed using imidazole as a functional monomer (p-type-electron acceptor), GO (n-type-electron donor), and AgNP by using the electrodeposition method. The role of GO in the system is providing additional functional units to bind the template and improve materials morphology while that of AgNPs is acting as a catalyst and charge carrier. The characterization of the sensing materials was done by using Fourier transform infrared, scanning electron microscopy, energy dispersive X-ray spectroscopy, and cyclic voltammetry. After optimization of the essential parameters, the sensor was successfully applied to detect the target analyte using the square wave voltammetric technique. The prepared sensor exhibited a wide linear range of 10 fM-250 nM with the limit of detection and limit of quantification of 3.01 fM and 10.03 fM, respectively. The high percentage recoveries, sensitivity, repeatability, and easy fabrication of the MIP-GO-AgNP materials made the proposed sensor promising for environmental monitoring applications in the future.

- Description: Objectives: To summarise SNP associations identified by genome-wide association studies (GWASs) of anxiety disorders and neuroticism; to appraise the quality of individual studies, and to assess the ancestral diversity of study participants. Methods: We searched PubMed, Scopus, PsychInfo and PubPsych for GWASs of anxiety disorders, non-diagnostic traits (such as anxiety sensitivity), and neuroticism, and extracted all SNPs that surpassed genome-wide significance. We graded study quality using Q-genie scores and reviewed the ancestral diversity of included participants. Results: 32 studies met our inclusion criteria. A total of 563 independent significant variants were identified, of which 29 were replicated nominally in independent samples, and 3 were replicated significantly. The studies had good global quality, but many smaller studies were underpowered. Phenotypic heterogeneity for anxiety (and less so for neuroticism) seemed to reflect the complexity of capturing this trait. Ancestral diversity was poor, with 70% of studies including only populations of European ancestry. Conclusion: The functionality of genes identified by GWASs of anxiety and neuroticism deserves further investigation. Future GWASs should have larger sample sizes, more rigorous phenotyping and include more ancestrally diverse population groups.
