Isolation and molecular characterization of Bacillus cereus from cow’s raw milk

- Description: Bacillus cereus is a group of ubiquitous facultative anaerobic spore forming Gram-positive rods commonly found in soil. It has been detected and implicated in several contaminated food products and raw milk in dairy farms and it causes foodborne gastroenteritis by producing several toxins. This study is aimed at characterizing virulence determinants of B. cereus from cow‟s raw milk. A total of 400 raw milk samples were collected in Fort Hare Dairy Trust and Middledrift Dairy Farm; and cultured on Polymyxin pyruvate Egg-Yolk Mannitol Bromothymol Agar (PEMBA) for 48 hours at 37°C. DNA was isolated from the isolates and 16S rDNA was amplified and sent to Central Analytical Laboratory for sequencing. The gyrB gene of B. cereus was also used to confirm the identity of the isolates. Antibiotic susceptibility profiles of the isolates together with virulence genes were investigated. Multilocus Sequence typing was used to investigate the genetic relatedness of some selected isolates. Furthermore, spores of the isolates were produced, harvested and their concentrations determined. All (100%) of the isolates were identified as having a 96-99% similarity to B. cereus, B. thuringiensis and B. anthracis using sequencing; while gyrB gene was observed in all (100%) of the isolates. Three virulence genes nheA, nheB, nheC encoding for non haemolysin enterotoxin were amplified in all (100%) the isolates. All (100%) of the isolates were susceptible to doxycycline, gentamycin, tetracycline, ciprofloxacin, chloramphenicol and streptomycin. Resistance to rifampicin and penicillin G was predominant with equal rate of 100%, while susceptibility to erythromycin, clindamycin and doxycycline ranged from 60% to 100%. The selected isolates were related and are descendants of the same ancestor. All (100%) the isolates produced spores. The B. cereus isolates contain virulence genes, has multiple antibiotic drug resistance and produce spores, which poses a health risk to the public and cannot be used as probiotics.

- Description: My aim was to resolve the taxonomy of the South African forms of Cercopithecus albogularis by exploring morphological and genetic variation in the two samango subspecies described for the region: C. a. erythrarchus and C. a. labiatus. In addition, I estimated their geographic distributions and habitat requirements from the provenance data I collected during my study of museum specimens. My analysis has shown clear morphological differences between C. a. labiatus and C. a. erythrarchus. The two subspecies differ in pelage coloration, but also in cranial shape and tail lengths. Furthermore, C. a. labiatus is slightly smaller, shorter-tailed and stocky compared with the long-tailed slender northern forms of C. albogularis, which may be adaptations to cold environments like montane forest. The northern C. albogularis subspecies are distinguishable from the southern taxa with a high degree of reliability (98.5%), suggesting that C. albogularis consists of more than one species. The most appropriate name for the southern species is C. labiatus. My genetic study did not detect variation among the 10 animals sampled in Hogsback; while it clustered the different taxa, it could not resolve relationships between them, with the exception of the outgroup. The lack of resolution of the deeper nodes could be a result of the fact that our sequence was very short (274 bp). The mt 12S rRNA gene was not an ideal gene for this study, which should have involved a less conserved section of the mtDNA molecule, like the rapidly evolving D-loop. More genetic work is clearly needed to resolve the phylogenetic relationships within the C. mitis supergroup. However, preliminary genetic data indicate that the southern samangos are distinct from the C. mitis of West Africa, while my morphometric study suggests they may also be distinct from C. albogularis in East Africa. Molecular and karyological studies comparing the genomes of Hogsback “C. a. labiatus” with the neighbouring “C. a. erythrarchus” and Zanzibar C. albogularis would be extremely enlightening.

- Description: This study analyses the economic impact between foreign debt and economic growth in South Africa. By fitting a production function model to annual data for the period 1980-2011, the study examines the dynamic effect of debt service, capital stock and labour force on the economic growth of the country. By following Cunningham (1993), it has identified the long-run and short-run causal relationships among the included variables. The results indicate that the debt servicing burden has a negative effect on the productivity of labour and capital, and thereby affect economic growth adversely. The results also illustrate that the debt service ratio tends to negatively affect GDP and the rate of economic growth in the long-run, which, in turn, reduces the ability of the country to service its debt. Similarly, the estimated error correction term shows the existence of a significant long-run causal relationship among the specified variables. Overall, the results suggest the existence of short-run and long-run causal relationships running from debt service to GDP.

Modelling and mapping the suitability of land for crop production using a combination of GIS and remote sensing in the Eastern Cape: a case study of Mbashe and Mquma local municipalities-South Africa

Crop-land suitability analysis is a prerequisite to achieve optimum utilization of the available land resources for sustainable agriculture rural production (T.R. Nisar Ahamed et al., 2000). It is indeed of paramount importance to identify suitable land for cropping while causing minimum impact to the environment. The Food and Agriculture Organization (FAO, 1976), recommended an approach of land suitability evaluation for crops in terms of suitable land based on climatic and terrain data and soil properties. In this study, an attempt was made to identify suitable areas for massive crop production using remote sensing and GIS methodologies and the knowledge from extension officers. The primary method aims at generating land cover data using SPOT 5 satellite imagery and modeling with the existing land capability. The research purpose was to map the map the number hectares suitable areas for crop production number hectares suitable areas for crop production number hectares suitable areas for crop production number hectares suitable areas for crop production. Spatial modeling techniques were utilized to model land suitability model in an effective and efficiently way. The spatial modeling extension from ESRI product was used to model the crop suitability areas. The model run on ArcGIS platform and due to the fact that modeling only uses raster formats, all the data sets were projected and converted to raster format. The weighted overlay model was used to create land suitability map. The model results revealed that 4046251.79 hectares were suitable for cropping in the study area. The final outputs of suitable areas were calculated and each ward was given a value of suitable area as well as unsuitable area. The validation of the final maps compliments the 500 000 hectares that were mapped by Dept of Agriculture EC using 8% slope as the best potential areas. The method provides a cheap, effective and efficient way to map suitable areas over a large area and it also uses remote sensing data. It is hoped that decision makers will make use of the information produced in this paper as the whole world is in crisis of food security.

Possible factors that affect export diversification considered as independent variables in this study include gross capital formation, human capital, real effective exchange rate and trade openness. Results of the study reveal that export diversification and trade openness are positively related to economic growth while real effective exchange rate, capital formation and human capital have negative long run relationships with economic growth. The study recommended the continual implementation of trade liberalisation by the South African government. The South African government is also encouraged to promote the production of a diversified export basket through subsidisation, promotion of innovation and production of new products.

A web based help desk system is developed and implemented to allow knowledge sharing and collaboration amongst users and experts as a way of ensuring the technical sustainability of the Siyakhula Living Lab, a remote rural based Information and Communications Technology (ICT4D) intervention. The research describes the use of Transactional and Innovation & Creative knowledge management strategies and their associated applications such as case-based reasoning (CBR) methods and collaborative knowledge approaches to develop a help desk support system for a geographically distributed multipurpose ICT centre in Dwesa, a rural remote area in the Eastern Cape Province, South Africa. The system implemented in this study has been validated for functional adequacy and usability within the Siyakhula Living Lab. The results of the evaluation are presented in this thesis to provide the initial validation of the effectiveness of the help desk system. - Description: In recent years, immense effort has been channelled towards the Information and Technological development of rural areas. To support this development, telecommunication networks have been deployed. The availability of these telecommunication networks is expected to improve the way people share ideas and communicate locally and globally, reducing limiting factors like distance through the use of the Internet. The major problem for these networks is that very few of them have managed to stay in operation over long periods of time. One of the major causes of this failure is the lack of proper monitoring and management as, in some cases, administrators are located far away from the network site. Other factors that contribute to the frequent failure of these networks are lack of proper infrastructure, lack of a constant power supply and other environmental issues. A telecommunication network was deployed for the people of Dwesa by the Siyakhula Living Lab project. During this research project, frequent visits were made to the site and network users were informally interviewed in order to gain insight into the network challenges. Based on the challenges, different network monitoring systems and other solutions were deployed on the network. This thesis analyses the problems encountered and presents possible and affordable solutions that were implemented on the network. This was done to improve the network‟s reliability, availability and manageability whilst exploring possible and practical ways in which the connectivity of the deployed telecommunication network can be maintained. - Description: In recent years, immense effort has been channelled towards the Information and Technological development of rural areas. To support this development, telecommunication networks have been deployed. The availability of these telecommunication networks is expected to improve the way people share ideas and communicate locally and globally, reducing limiting factors like distance through the use of the Internet. The major problem for these networks is that very few of them have managed to stay in operation over long periods of time. One of the major causes of this failure is the lack of proper monitoring and management as, in some cases, administrators are located far away from the network site. Other factors that contribute to the frequent failure of these networks are lack of proper infrastructure, lack of a constant power supply and other environmental issues. A telecommunication network was deployed for the people of Dwesa by the Siyakhula Living Lab project. During this research project, frequent visits were made to the site and network users were informally interviewed in order to gain insight into the network challenges. Based on the challenges, different network monitoring systems and other solutions were deployed on the network. This thesis analyses the problems encountered and presents possible and affordable solutions that were implemented on the network. This was done to improve the network‟s reliability, availability and manageability whilst exploring possible and practical ways in which the connectivity of the deployed telecommunication network can be maintained. Multivariate co-integration, Granger causality, vector error correction model, impulse response function analysis and variance decomposition analysis are employed to determine the semi-strong form efficiency in South African equity market. Monthly data of Johannesburg Stock Exchange index, money supply (M1 & M2), short term interest rate, inflation, rand/dollar exchange rate, London Stock Exchange index (FSTE100) and GDP from 2000-2009 are the variables of interest.Weak form efficiency is examined using unit root tests. The results of this study show evidence of weak form efficiency of the JSE using the Augmented-Dickey Fuller and Philip-Perron unit root tests. The results reject the hypothesis that the JSE is semi-strong and have important implications for government policy, regulatory authorities and participants in the South African stock market.
