- Description: Mammalian predators are keystone species and can have disproportionately large impacts, relative to their abundance, on the communities and ecosystems they inhabit. In addition, predator-prey interactions (including bottom-up and top-down effects) are the fundamental linkage among species in food webs. Thus, understanding the food requirements of predators is imperative. There have been no comprehensive studies on the diet of black-backed jackals (Canis mesomelas) across a full seasonal cycle in the Albany Thicket Biome of the Eastern Cape Province. This biome is rich in plant diversity, is highly nutritious for herbivores and supports a large number of mammalian species, yet it is poorly conserved. The aim of the present study was to establish the diet of black-backed jackals on two conservation areas where thicket vegetation dominates (Great Fish River Reserve and Kwandwe Private Game Reserve). The diet was assessed using scats collected monthly from June 2009 to May 2010 and a sample of 25 stomachs over the same period. At both sites and across all seasons, mammalian hair was encountered most often (93% of all scats) and a total of 23 mammal species were consumed. Of the mammals recorded in the scats, hair from several ungulate species was encountered most frequently and these were mostly hider species (offspring hidden in tall vegetation just after birth). On a seasonal basis, the proportion of mammalian hair was lowest in autumn. However, the contribution of fruits (Carissa haematocarpa, Diospyros lycioides and Grewia robusta) to the diet was significantly higher during this season. This suggests that black-backed jackals may employ diet switching in the Eastern Cape as a mechanism to capitalise on seasonally abundant food sources. Based on the stomach content analysis, invertebrates were consumed as frequently as mammalian prey throughout the year but mammalian prey was recorded in greater quantity (i.e. volume and mass). Thus, scat analysis may have overestimated mammalian prey and fruits and underestimated the importance of invertebrates, particularly highly digestible species (e.g. termites). In spite of these differences, scat analysis demonstrated similar general and seasonal trends as the stomach content analysis. Thus, scat analysis can provide an efficient and relatively accurate source of information on the diet of blackbacked jackals in the Eastern Cape. Both stomach content and scat collection methods depicted the black-backed jackal as a generalist and opportunistic predator, mainly relying on mammals and invertebrates. Although the results of my study suggest that black-backed jackal may be important predators of certain ungulate species, further study on their prey preferences, actual biomass ingested and the effects on the population viability of prey species is required in order to develop a robust management plan for the species in the protected areas of the Eastern Cape.

Current research trends are geared towards utilizing food crops as substrate for bioethanol fermentation; however, this is the source of much controversy. Utilizing food crops for fuel purposes is anticipated to cause massive food shortages worldwide. Cellulose is the most abundant renewable resource on earth and is subject to a wide array of scientific study in order to utilize the glucose contained within it. Waste paper has a high degree of cellulose associated with it, which makes it an ideal target for cellulose biotechnology with the ultimate end goal of bioethanol production. This study focussed on producing the necessary enzymes to hydrolyse the cellulose found in waste paper and using the sugars produced to produce ethanol. The effects of various printing inks had on the production of sugars and the total envirorunental impact of the effluents produced during the production line were also examined. It was found that the fungus Trichoderma longibrachiatum DSM 769 grown in Mandel's medium with waste newspaper as the sole carbon source at 28 °C for 6 days produced extracellular cellulase enzymes with an activity of 0.203 ± 0.009 FPU.ml⁻¹, significantly higher activity as compared to other paper sources. This extracellular cellulase was used to hydrolyse waste newspaper and office paper, with office paper yielding the highest degree of sugar production with an end concentration of 5.80 ± 0.19 g/1 at 40 °C. Analysis by HPLC showed that although glucose was the major product at 4.35 ± 0.12 g/1, cellobiose was also produced in appreciable amounts (1.97 ± 0.71 g/1). The sugar solution was used as a substrate for Saccharomyces cerevisiae DSM 1333 and ethanol was produced at a level of 1.79 ± 0.26 g/1, the presence of which was confirmed by a 600 MHz NMR spectrum. It was found that cellobiose was not fermented by this strain of S. cerevisiae. Certain components of inks (the PAHs phenanthrene and naphthalene) were found to have a slight inhibitory effect (approximately 15% decrease) on the cellulase enzymes at very high concentrations (approximately 600 μg/1 in aqueous medium), while anthracene had no effect. Whole newsprint ink was shown not to sorb glucose. The environmental analysis of the effluents produced showed that in order for the effluents to be discharged into an aqueous ecosystem they would have to be diluted up to 200 times. They were also shown to have the potential to cause severe machinery damage if reused without proper treatment.

- Description: The tuberculosis epidemic is escalating in South Africa as well as globally. This escalation is exacerbated by the increasing prevalence of multidrug-resistant tuberculosis (MDRTB), which is defined by the World Health Organisation (WHO) as resistance of Mycobacteria to at least isoniazid and rifampicin. Multi-drug resistant tuberculosis is estimated to occur in 1-2% of newly diagnosed tuberculosis (TB) patients and in 4-8% of previously treated patients. MDRTB is both difficult and expensive to treat, costing up to 126 times that of drug-sensitive TB. Resource constrained countries such as South Africa often lack both the money and the infrastructure to treat this disease. The aim of this project was to determine whether the performance of a systematic review with subsequent economic modelling could influence the decision making process for policy makers. Data was gathered and an economic evaluation of MDRTB treatment was performed from the perspective of the South African Department of Health. Three treatment alternatives were identified: a protocol regimen of second line anti-tuberculosis agents, as recommended in the South African guidelines for MDRTB, an appropriate regimen designed for each patient according to the results of culture and drug susceptibility tests, and non-drug management. A decision-analysis model using DATA 3.0 by Treeage® was developed to estimate the costs of each alternative. Outcomes were measured in terms of cost alone as well as the ‘number of cases cured’ and the number of ‘years of life saved’ for patients dying, being cured or failing treatment. Drug, hospital and laboratory costs incurred using each alternative were included in the analysis. A sensitivity analysis was performed on all variables in order to identify threshold values that would change the outcome of the evaluation. Results of the decision analysis indicate that the individualised regimen was both the cheaper and more cost-effective regimen of the two active treatment options, and was estimated to cost R50 661 per case cured and R2 070 per year of life saved. The protocol regimen was estimated to cost R73 609 per case cured and R2 741 per year of life saved. The outcome of the decision analysis was sensitive to changes in some of the variables used to model the disease, particularly the daily cost of drugs, the length of time spent in hospital and the length of treatment received by those patients dying or failing treatment. This modelling exercise highlighted significant deficiencies in the quality of evidence on MDRTB management available to policy makers. Pragmatic choices based on operational and other logistic concerns may need to be reviewed when further information becomes available. A case can be made for the establishment of a national database of costing and efficacy information to guide future policy revisions of the South African MDRTB treatment programme, which is resource intensive and of only moderate efficacy. However, due to the widely disparate range of studies on which this evaluation was based, the outcome of the study may not be credible. In this case, the use of a systematic review with subsequent economic modelling could not validly influence policy-makers to change the decision that they made on the basis of drug availability.

An investigation into the possible neuroprotective role of antidepressant drugs

- Description: Antidepressants are widely used in the treatment of depressive illnesses associated with neurodegenerative disorders such as Alzheimer’s disease, Parkinson’s disease and Huntington’s disease. Neuroprotection in such disorders is of vital importance in order to delay the progression of the primary disorder. The pathology of neurodegeneration is not fully understood. It is however widely accepted that oxidative stress and excitotoxicity play a major role. Brain tissue is rich in phospholipids, which are especially prone to oxidation due to the high level of oxygen utilization in the brain. In addition, the brain lacks defence mechanisms to protect it against the wrath of free radicals. Presently, there is a wide variety of antidepressant drugs available. These range from the original tricyclic antidepressants to the newer selective serotonin reuptake inhibitors. It is not known whether antidepressant drugs, old or new, offer neuroprotection or how the existing state and/or the progression of neurodegeneration, is influenced by these agents. The present study was undertaken to determine how nortriptyline, trimipramine and fluoxetine affect neurodegeneration. Initial in vitro and in vivo studies show that all three of the antidepressants (0-1mM) studied provide neuroprotection from quinolinic acid induced lipid peroxidation. A histological investigation supported these findings by showing that a marginal degree of neuroprotection is apparent when treating animals with antidepressants (10mg/kg) before and following quinolinic acid intrastriatal injection. Further studies were undertaken in an attempt to determine the mode of neuroprotective action of the agents studied. An in vitro study of superoxide anion induced lipid peroxidation indicates that these agents do not act as antioxidants. The influence of the antidepressants on tryptophan 2,3-dioxygenase activity was assessed, based on the understanding that inhibition of this enzyme results in increased levels of the known antioxidant indoleamine, melatonin. Nortriptyline hydrochloride is seen to inhibit tryptophan 2,3-dioxygenase activity and as such it is possible that this antidepressant can indirectly provide neuroprotection by increasing available melatonin. Electrochemical and UV/visible studies show that trimipramine maleate interacts with free iron (II) and iron (III) ions. Free metal ions can catalyse the formation of damaging free radicals. Through interaction with trimipramine maleate, these ions will be unavailable to the system and thus cannot contribute to oxidative stress. The findings of this study indicate that antidepressants may be able to provide neuroprotection to neuronal cells. The mode of such neuroprotective actions need to be further examined so that patients suffering from depression coexisting with neurodegenerative diseases can be safely and effectively treated.

Enzymology of activated sewage sludge during anaerobic treatment of wastewaters : identification, characterisation, isolation and partial purification of proteases

- Description: During anaerobic digestion bacteria inside the digester require a carbon source for their growth and metabolism, sewage sludge was used as a carbon source in this study. The COD content was used to measure the disappearance of the substrate. COD content was reduced by 48.3% and 49% in the methanogenic and sulphidogenic bioreactors, respectively, while sulphate concentration was reduced by 40%, producing 70mg/L of hydrogen sulphide as the end product over the first 5-7 days. Sulphate (which is used as a terminal electron acceptor of sulphur reducing bacteria) has little or no effect on the sulphidogenic and methanogenic proteases. Sulphite and sulphide (the intermediate and end product of sulphate reduction) increased protease activity by 20% and 40%-80%, respectively. Maximum protease activity occurred on day 21 in the methanogenic reactor and on day 9 in the sulphidogenic reactor. The absorbance, which indicates the level of amino acid increased to 2 and 9 for methanogenic and sulphidogenic bioreactors, respectively. Proteases that were active during anaerobic digestion were associated with the pellet (organic particulate matter) of the sewage. These enzymes have an optimum activity at pH 10 and at temperature of 50°C. The proteases that were active at pH 5 and 7, had optimum temperatures at 30°C and 60°C, respectively. Due to their association with organic particulate matter, these enzymes were stable at their optimum temperatures for at least five hours at their respective pH. Inhibition by PMSF, TPCK and 1.10-phenanthroline suggested that proteases inside the anaerobic digester are a mixture of cysteine, serine and metalloproteases. At pH 5, however, EDTA appeared to enhance protease activity by 368% (three-fold). Acetic acid decreased protease activity by 21%, while both propionic and butyric acid at 200 mg/L cause total inhibition of protease activity while these acids at higher pH (where they exist as their corresponding salts) exerted little effect. Copper, iron and zinc inhibited protease activity by 85% at pH 5 with concentrations ranging between 200 and 600 mg/L. On the other hand, nickel, showed an increase in protease activity of nearly 250%. At pH 7 and 10, copper had no effect on protease activity while iron, nickel and zinc inhibited these enzymes by 20-40%. Proteases at pH 7 were extracted from the pellet by sonication, releasing 50% of the total enzymes into the solution. The enzymes were precipitated by ammonium sulphate precipitation, and further purified by ion exchange chromatography and gel filtration. Ion exchange chromatography revealed that most of the enzymes that hydrolyse proteins are negatively charged while gel filtration showed that their molecular weight is approximately 500 kDa.

- Description: During anaerobic digestion bacteria inside the digester require a carbon source for their growth and metabolism, sewage sludge was used as a carbon source in this study. The COD content was used to measure the disappearance of the substrate. COD content was reduced by 48.3% and 49% in the methanogenic and sulphidogenic bioreactors, respectively, while sulphate concentration was reduced by 40%, producing 70mg/L of hydrogen sulphide as the end product over the first 5-7 days. Sulphate (which is used as a terminal electron acceptor of sulphur reducing bacteria) has little or no effect on the sulphidogenic and methanogenic proteases. Sulphite and sulphide (the intermediate and end product of sulphate reduction) increased protease activity by 20% and 40%-80%, respectively. Maximum protease activity occurred on day 21 in the methanogenic reactor and on day 9 in the sulphidogenic reactor. The absorbance, which indicates the level of amino acid increased to 2 and 9 for methanogenic and sulphidogenic bioreactors, respectively. Proteases that were active during anaerobic digestion were associated with the pellet (organic particulate matter) of the sewage. These enzymes have an optimum activity at pH 10 and at temperature of 50°C. The proteases that were active at pH 5 and 7, had optimum temperatures at 30°C and 60°C, respectively. Due to their association with organic particulate matter, these enzymes were stable at their optimum temperatures for at least five hours at their respective pH. Inhibition by PMSF, TPCK and 1.10-phenanthroline suggested that proteases inside the anaerobic digester are a mixture of cysteine, serine and metalloproteases. At pH 5, however, EDTA appeared to enhance protease activity by 368% (three-fold). Acetic acid decreased protease activity by 21%, while both propionic and butyric acid at 200 mg/L cause total inhibition of protease activity while these acids at higher pH (where they exist as their corresponding salts) exerted little effect. Copper, iron and zinc inhibited protease activity by 85% at pH 5 with concentrations ranging between 200 and 600 mg/L. On the other hand, nickel, showed an increase in protease activity of nearly 250%. The Makganyene Formation has diamictite as the main rock type, but siltstone, sandstone, shale, and iron-formations are also present. A glacial origin has been proposed in the past due to the presence of dropstones, faceted and striated pebbles. Typically, the Makganyene Formation contains banded iron-formations interbedded with clastic rocks (shale, siltstone, sandstone and diamictites) at the contact with the underlying iron-formations. This transitional zone is generally overlain by massive or layered diamictites which contain poorly sorted clasts (mainly chert) within a shaly matrix. Striated pebbles have been found during field work, and dropstones have been observed in diamictites and banded iron-formations during the study. The top of the Makganyene Formation contains graded cycles interbedded with diamictites and thin layers of andesitic lavas from the Ongeluk Formation. The basal contact of the Makganyene Formation with the underlying Koegas Subgroup was described as unconformable by previous workers. However field work localised in the Rooinekke area shows a broadly conformable and interbedded contact with the underlying Koegas Subgroup. As described above, banded iron-formations are interbedded with the clastic rocks of the Makganyene Formation. Moreover, boreholes from the Sishen area display the same interbedding at the base of the Makganyene Formation. This suggests that no significant time gap is present in the whole succession between the Ghaap and Postmasburg Group. The Transvaal Supergroup in the Northern Cape displays the following succession : carbonates-BIFs-diamictites/ lava-BIFs-carbonates. The Makganyene Formation is thus at the centre of a symmetrical lithologic succession. Bulk rock compositions show that the diamictites have a similar composition to banded iron-formation with regard to their major element contents. Banded iron-formations acted as a source for the diamictites with carbonates and igneous rocks representing minor components. Differences in bulk composition between the Sishen and Matsap areas emphasize that the source of the diamictite was very localised. The Chemical Index of Alteration (CIA) has been calculated, but since the source dominant rock was iron-formation, this index cannot be usefully applied to the diamictites. ACN, A-CN-K, and A-CNK-FM diagrams confer a major importance in sorting processes due to the separation between the fine and coarse diamictites. The interbedded iron-formations display little clastic contamination indicating deposition in clear water conditions. However, dropstones are present in one borehole from the Matsap area, indicating that iron-formation took place under ice cover, or at least under icebergs. Stable isotope studies show that the iron-formations, interbedded towards the base of the Makganyene Formation, have similar values to the iron-formations of the Koegas Subgroup. As a result of the above observations, new correlations are proposed in this study, relating the different Transvaal Supergroup basins located on the Kaapvaal Craton. The Pretoria Group of the Transvaal Basin has no correlative in the Griqualand West Basin, and the Postmasburg Group of the Northern Cape Basin has no lateral equivalent in the Transvaal Basin. These changes have been made to overcome problems present in the current correlations between those two basins. The Makganyene Formation correlates with the Huronian glaciations which occurred between 2.4 and 2.2 Ga ago in North America. Another Precambrian glaciation is the worldwide and well-studied Neoproterozoic glaciation (640 Ma). At each of these glaciations, major banded iron-formation deposition took place with associated deposition of sedimentary manganese in post-glacial positions. The central position of the Makganyene Formation within the Transvaal Supergroup in the Northern Cape emphasizes this glacial climatic dependence of paleoproterozoic banded iron-formation and manganese deposition. However these two Precambrian glaciations are interpreted in paleomagnetic studies as having occurred near to the equator. The controversial theory of the Snowball Earth has been proposed which proposes that the Earth was entirely frozen from pole to pole. Results from field work, sedimentology, petrography and geochemistry were integrated in a proposed depositional model of the Makganyene Formation occurring at the symmetrical centre of the lithologic succession of the Transvaal Supergroup. At the beginning of the Makganyene glaciation, a regression occurred and glacial advance took place. The diamictites are mostly interpreted as being deposited from wet-based glaciers, probably tidewater glaciers, where significant slumping and debris flows occurred. Any transgression would cause a glacial retreat by rapid calving, re-establishing the chemical sedimentation of banded iron-formations. These sea-level variations are responsible for the interbedding of these different types of rocks (clastic and chemical). The end of the Makganyene glacial event is characterised by subaerial eruptions of andesitic lava of the Ongeluk Formation bringing ashes into the basin. Banded iron-formation and associated manganese accumulations are climate-dependant. Glacial events are responsible for the build up of metallic ions such as iron and manganese in solution in deep waters. A warmer climate would induce a transgression and precipitation of these metallic ions when Eh conditions are favourable. In the Transvaal Supergroup, the climatic variations from warm to cold, and cold to warm are expressed by the lithologic succession. The warm climates are represented by carbonates. Cold climates are represented by banded iron-formations and the peak in cold climate represented by the diamictites of the Makganyene Formation. These changes in climate are gradual, which contradict the dramatic Snowball Earth event: a rapid spread of glaciated areas over low-latitudes freezing the Earth from pole-to-pole. Therefore, to explain low-latitude glaciations at sea-level, a high obliquity of the ecliptic is most likely to have occurred. This high obliquity of the ecliptic was acquired at 4.5 Ga when a giant impactor collided into the Earth to form the Moon. Above the critical value of 54° of the obliquity of the ecliptic, normal climatic zonation reverts, and glaciations will take place preferentially at low-latitudes only when favourable conditions are gathered (relative position ofthe continents and PC02 in the atmosphere).

- Description: It is the aim of this study to elucidate the influence of progesterone on body weight regulation in intact female rats. A study of the literature includes a description of the body weight regulation and the effects of ovarian hormones on it. The controlled-system approach tries to link behavioral and physiological factors altering energy balance. The experimental study is subdivided into food-intake - and food-selection studies, a locomotor activity study, a study eliciting a possible role of thermogenesis, and finally rat liver studies which consist of a gas chromatography analysis of hepatic fatty acids and an electron microscopy study examining the ultrastructure of hepatocytes. It can be concluded that the effect of progesterone treatment on the body weight of intact female rats depends on the route of administration. There is a significant increase in body weight after subcutaneous progesterone injections without changes in total caloric intake and nutrient selection habits, indicating the importance of energy expenditure. But changes in spontaneous activity make no contribution in the progesterone-induced energy storage. It is also concluded that peripherally located brown adipose tissue thermogenesis is not changed, without ruling out the effect of more centrally located thermogenic organs as the liver. In this organ, small but significant changes in the fatty acid profile occur during the subcutaneous progesterone treatment.

- Description: The stratigraphy at the Gamigab Sn prospect consists of two mainly schistose units separated by a thick marble unit which have been assigned to the Orusewa, the Karibib and the Kuiseb Formations respectively. Four phases of folding affected the lithologies with the south-south-west trending F2 folds defining the main structures in the region. The area underwent low grades of metamorphism. Temperatures were in the range 420° to 500°C and pressures less than 2 kbars. The effects of contact metamorphism are seen in the south-east and south-west. Regional metamorphism outlasted the deformation and contact metamorphism started late during deformation. Two Karoo-age intrusions penetrated the metasediments north of the mineralisation. One is an altered porphyry plug and the other is a weathered dolerite plug, the latter containing xenoliths of undeformed Karoo sediments. Cassiterite is hosted within east-west trending quartz veins that cross-cut previously altered schistose country rocks. The alteration types include sericitisation, tourmalinisation, carbonatisation and ferruginisation. Preliminary Rb/Sr dating on muscovite from the alteration zone suggests an age of 509 ± 11 Ma. Breccias of probable hydrothermal origin are spatially associated with the mineralisation. These hydraulic breccias occur in antiformal structures within the marble and developed in response to a sudden pressure release due to a build up of fluids at the contact between the schistose Orusewa and carbonate Karibib Formations.
