Exploring the relationship between intergroup contact and anti-gay prejudice amongst heterosexual South African students

- Description: Throughout history gay men have experienced oppression in the form of harassment, violence and discrimination and in South Africa it is no different (Brouard et al., 2016; OUT, 2016). According to the contact hypothesis, intergroup contact has shown to be one of the most successful ways to reduce prejudice and improve attitudes towards an outgroup (i.e. a group that you do not identify yourself with; Allport, 1954; Pettigrew & Tropp, 2006). Therefore, this study investigated whether positive intergroup contact plays a role in improving attitudes, and lessening prejudice towards gay men in South Africa by exploring the relationship between intergroup contact with gay men amongst heterosexual South African students and anti-gay prejudice. Furthermore, this study aimed to get a better understanding of how this process operates by exploring whether two affective mediators, namely intergroup anxiety and empathy, mediates the contact-prejudice relationship. A quantitative, cross-sectional research design was used where participants completed an online questionnaire (N = 125). The findings from the present study indicate that contact with gay men amongst heterosexual students at a South African University positively and significantly predicted more positive attitudes towards gay men in general. Moreover, the results show that decreased intergroup anxiety and increased intergroup empathy fully mediates the relationship between intergroup contact and anti-gay prejudice. This research contributes to the body of contact literature in South Africa and could offer practical means for interventions that aim to reduce prejudice and improve attitudes towards gay men.

- Description: Narcissism, often linked to a sense of entitlement and inflated sense of self, is a complex condition that has been studied for approximately a century. It is typically associated with individualistic cultures, which place emphasis on the self. Although much is known about narcissism, there is far less knowledge on narcissism in collectivist cultures. The Xhosa culture is commonly seen as a collectivist culture. No research to date has been conducted on this construct with Xhosa-speaking South Africans. The aim of the study was to explore and describe therapist perceptions of narcissism in traditional cultural contexts. The objectives of the study included an investigation into how narcissism presents in the Xhosa culture, and an exploration of the narcissistic elements that manifest more prominently. A qualitative, exploratory descriptive research design was employed, and snowball sampling was used to identify psychologists from the Nelson Mandela Metropole for inclusion in the study. Semi-structured interviews were used to gather data, and data was analysed by thematic analysis. The findings of the thematic analysis consisted of six themes, which are thoughts on the Xhosa culture, culture and personality, contemporary Western theory lacking, traits, parenting, and interpersonal and personal difficulties. These themes reflect the participants’ experience, thoughts, and opinions on narcissism in individuals from the Xhosa culture.

- Description: This study aimed to provide public relations professions with insight into the use of Social Customer Relationship Marketing as a marketing 2.0 tool which delivers the potential to elicit the opportunity for organisations to monitor, engage and manage conversations and relationships through Facebook. Drawing on literature from the fields such as new media, public relations, social customer relationship marketing, and public relations, a qualitative content analysis of case studies of selected organisations was conducted including 10 South African organisations that have incorporated Facebook into their Social Customer Relationship Marketing strategies. The content analysis was conducted to determine the amount of communications taking place on each fan page in order to support the data and provide additional insight into the research question, primary data was collected through two survey questionnaires of, firstly, social media users who subscribe to an organisation‘s Facebook page and, secondly, a survey conducted among the social organisations. Surveys were conducted to firstly determine the social organisations interactions with Facebook as secondly, to determine the social media users‘ interactions with the Facebook. The study found that the majority of the organisations under study in the content analysis successfully elicited user generated conversations about the brand, through content that was advocated by the organisations Facebook administrator. It is also evident as seen in the minority of organisations that these platforms are only likely to elicit consumer- driven dialogue on Facebook if the platform is managed effectively through initiated daily conversations about the brand. The continuous involvement by the administrator in encouraging conversations was necessary, as well as response to all wall posts by the social customer, participation in all conversations by providing accurate information to customers and potential customers about the brand. Conclusions from the survey questionnaires among the social identified the social customers need to be communicated with pro-actively by an organisations who can provide, through collaborative conversations, information pertaining to the brand in real time conversation, while the survey among the social organisation concluded the need for an organisation to not only create a social media footprint, but to allow for a space in which through conversation with the social customer, conversations can turn into relationships.

Adaptive interfaces for mobile preference-based searching

- Description: Today's mobile computing devices provide a convenient means to search for points-of-interest (POIs) such as restaurants and accommodation. Mobile Preference-Based Search Tools (PBSTs) allow users to identify POIs such as restaurants or accommodation most suited to their needs and constraints using a mobile device. These devices however, have several design constraints including limited screen space and hardware capabilities. Adaptive User Interfaces (AUIs) have been proposed to address these issues but have not been extensively applied to mobile PBSTs such as mobile tourist guides. In addition, AUIs possess several benefits and advantages over static (traditional) interfaces, which do not take a user's preferences, skill set and experience into account. Little research, however, has been conducted into identifying the potential benefits of AUIs for mobile preference-based searching (PBS). The aim of this research was to determine the extent to which an AUI could improve the effectiveness and user satisfaction of mobile PBS. A literature study was conducted to determine the benefits and limitations of existing mobile PBSTs and determine how these could be improved. The potential benefits of AUIs for mobile PBSTs and a mobile map-based visualisation system were identified. A suitable model for incorporating an AUI into a mobile PBST was identified. The requirements for a mobile PBST were combined with the potentially adaptable objects of a Mobile Map-based Visualisation (MMV) system to provide adaptation suggestions for POInter, an existing mobile tourist guide. A field study using POInter was conducted in order to measure the extent to which participants agreed with suggestions provided for adapting the information, interaction and visualisation aspects of the system. These results were used to derive adaptation requirements for A-POInter, an adaptive version of POInter. Using a model-based design approach, an AUI was designed and implemented for A-POInter. An extensive field study was then conducted to evaluate the usability of the adaptations provided by A-POInter. The quantitative and qualitative data collected from the evaluations allowed the usability of A-POInter to be determined. The results of the field study showed that the participants were highly satisfied with the usability and the usefulness of the adaptations provided by A-POInter. Conclusions and recommendations for future work based on the results of the research were then outlined to conclude the dissertation.

Production of biologically active recombinant HIV-1 protease and intehrase for the purpose of screening medicianl plant extracts

- Description: Human immunodeficiency virus (HIV) and its gradual weakening of the immune system is an ever growing threat. Acquired immune deficiency syndrome (AIDS), the final stage of HIV, renders a person vulnerable to various opportunistic infections, which in the end lead to death. Apart from intensive vaccine studies, treatment research mainly focuses on preventing the individual HIV enzymes (reverse transcriptase, integrase and protease) from performing their functions. Entry inhibitors, however, block viral entry into the cell, while antisense drugs lock onto the viral genome to keep it from functioning. In this study production of active recombinant HIV-1 protease and integrase was attempted for future drug screening programs. HIV-1 protease was cloned into a pET28b(+) vector and expressed in ROSETTA(DE3)pLysS cells. The protein was purified using a nickel-affinity column utilizing the hexa-histidine tag encoded by the vector. Gel filtration chromatography was attempted after refolding of the protease, but protease yield seemed to decrease with the additional purification step. Partially purified protease was characterized with kinetic studies. Kinetic parameters of HIV-1 protease were determined to be Km = 592 μM, Vmax = 0.59 μM/min and kcat = 31 s-1. HIV-1 integrase, which was cloned into a pET15b vector, was expressed in E. coli BL21(DE3) cells. The coding sequence had been mutated to introduce the amino acid substitutions F185K and C280S, increasing solubility of the protein. The first step in purification of this protein was nickel-affinity chromatography, after which cation exchange chromatography was attempted. HIV-1 integrase concentration was low throughout experiments and no clear elution from the cation exchange column could be observed. A non-radioactive enzyme linked HIV-1 integrase assay failed to detect integrase activity. Modifications to future studies of the integrase are suggested in the chapter involved.
