Synthesis, characterization & application of visible light responsive nitrogen doped Tio2 and copolymer-grafted asymmetric membranes with ozonolysis for water treatment

- Description: The use of titanium dioxide for the photo-catalytic removal of organic, inorganic, and microbial pollutants from natural water and wastewater has been considered a very promising technique. The aim of this study was to prepare nitrogen doped titanium dioxide, immobilize it on asymmetric polymeric membranes of poly (methacrylic acid) grafted onto poly (vinylidene difluoride) (PVDF) blended with poly (acrylonitrile) (PAN), and evaluate the photo-catalytic, antimicrobial, and antifouling properties of the membranes. Nitrogen doped titanium dioxide (N-TiO2) nano-particles were prepared by a low temperature sol gel synthesis technique. The modification of TiO2 with nitrogen allows photo-sensitization of the photo-catalyst towards visible light utilization. The N-TiO2 nano-particles were characterized by fourier transform infrared spectroscopy (FT-IR), scanning x-ray photoelectron spectroscopy (SXPS), X-ray diffraction analysis (XRD), diffuse reflectance spectroscopy (DRS), Brunauer Emmett Teller (BET) surface area analysis, and transmission electron microscopy (TEM). The characterizations revealed the presence of the expected functional groups and confirmed successful doping and that the product was visible light responsive. Novel poly (methacrylic acid) grafted onto poly (vinylidene difluoride)/ poly (acrylonitrile) (PMAA-g-PVDF/ PAN) asymmetric membranes were prepared by the dry-wet phase inversion technique. The poly (methacrylic acid) (PMAA) side chains where grafted onto an activated PVDF backbone by reversible addition fragmentation chain transfer (RAFT) polymerization. The photo-catalytic membranes were generated by blending N-TiO2 with the polymer solution before casting the membranes. The membranes were characterized by FT-IR, nuclear magnetic resonance spectroscopy (NMR), scanning electron microscopy (SEM), and thermo-gravimetric analysis (TGA). FT-IR and NMR analyses confirmed successful grafting of MAA chains onto PVDF while SEM confirmed the successful preparation of membranes with asymmetric structure. The efficacy of the photo-catalytic asymmetric membranes was evaluated on the removal of herbicides from synthetic water. Bentazon was easily degraded while atrazine and paraquat were recalcitrant and proved difficult to degrade. The best results were observed with 3 % N-TiO2-PMAA-g-PVDF/ PAN asymmetric membranes on the photo-degradation of bentazon, atrazine and paraquat in water. Significant enhancement in the photo-degradation of the three herbicides was observed when photo-catalytic degradation was coupled with ozonation. Liquid chromatography-mass spectrometry (LC-MS) analysis confirmed the presence of a degradation by-product during the photo-catalytic degradation of bentazon. The photo-catalytic membranes were also evaluated on the photo-catalytic reduction of heavy metals Pb2+ and Fe3+ in water, and the best results were obtained using 1 % N-TiO2-PMAA-g-PVDF/ PAN and 1 % N-TiO2-PAN asymmetric membranes. All prepared photo-catalytic membranes where capable of completely inactivating E. coli ATCC 8739 within 120 minutes of exposure and inactivation rate increased with increasing N-TiO2 photo-catalyst loading. However, there was an indication from the results obtained that N-TiO2 supported on PMAA-g-PVDF/ PAN showed a higher inactivation rate of E. coli ATCC 8739 compared to N-TiO2-PAN and N-TiO2-PVDF membranes. The 1 % N-TiO2-PMAA-g-PVDF/ PAN membranes gave the highest pure water flux (421.83 L/m2h). This increase (PVDF = 30.50 L/m2h, PAN = 73.85 L/m2h) in pure water flux is owedb to PMAA grafting as well as addition of N-TiO2. These modifications resulted in an increased membrane surface hydrophilicity, which promoted permeation of pure water through the membrane structure. A high bovine serum albumin (BSA) rejection (76.5 %) was noted and can be attributed to steric hindrance brought about by PMAA side chains which prevented the bulky BSA molecules from attaching to the membrane surface for PMAA-g-PVDF/ PAN membranes. However, the supporting porous sub-layer of an asymmetric membrane seemed to play a very important role in the overall permeability of a membrane. PVDF membranes are highly hydrophobic hence they gave a very low pure water flux.

The antifungal activity of an aqueous Tulbaghia violacea plant extract against Aspergillus flavus

- Description: Phytochemical analysis of both HEA1 and the crude plant extract showed the presence of phenolics, tannins and saponins. Saponins were the predominant secondary metabolites and were mostly abundant in the plant extract and to a lesser extent in the active compound. Steroidal saponins, tannins and phenolics were also detected in the plant extract, but only the phenolics were detected in the active compound. The results of the phytochemical analysis showed that those compounds that were not present in the active compound could be removed from the crude extract during the TLC purification process. Investigation on the mechanism of action of the crude plant extract on the sterol production by A. flavus showed that the plant extract affected ergosterol biosynthesis by causing an accumulation of oxidosqualene in the ergosterol biosynthetic pathway resulting in a decline in ergosterol production. An oscillatory response in lanosterol production was observed in the presence of the plant extract, which may be an adaptation mechanism of A. flavus to unfavourable conditions and compensation for the loss of enzyme activity which may have occurred as a result of the accumulation of oxidosqualene. The antifungal activity of the plant extract on ergosterol production by A. flavus may also be due to saponins which target the cell membrane and ergosterol production in fungi. The effect of the plant extract on the fungal cell wall of A. flavus also showed that the plant extract caused a decline in β-(1, 3) glucan production by inhibiting β-glucan synthase. The plant extract also affected the chitin synthesis pathway of A. flavus, by causing a decline in chitin production, which was due to the inhibition of chitin synthase. Investigation of chitinase production using 4MU substrates showed that the plant extract caused an accumulation of chitobioses, by activating chitobiosidases and endochitinases. A decline in N-acetylglucosaminidase activity in the presence of the plant extract was observed and this prevented the formation of N-acetylglucosamine. The accumulation of chitobiosidase and endochitinase may be as a result of autolysis that may be triggered by A. flavus as a survival mechanism in the presence of the plant extract and as a compensatory mechanism for the loss of β-glucans and chitin. The antifungal effect of the plant extract on various components of the cell wall of A. flavus, makes T. violacea aqueous plant extract an ideal chemotherapeutic agent against both human and plant pathogens of Aspergillus. The broad spectrum of antifungal activity of T. violacea against A. flavus also eliminates any chances of the fungus developing resistance towards it and would make it a candidate for use as a potential antifungal agent. Further identification and possible chemical synthesis is needed to shed light on the safety and efficacy of the active compound for further development as a chemotherapeutic agent.

The associative behaviour of silky sharks, Carcharhinus falciformis, with floating objects in the open ocean

- Description: The silky shark Carcharhinus falciformis forms the primary elasmobranch bycatch in tuna purse seine fisheries using fish aggregating devices (FADs) in all of the world’s tropical oceans. Its life-history traits of slow growth, late maturation and low fecundity make it vulnerable to over exploitation, as is apparent from historical bycatch trends. Very little is known about the associative behaviour of this species with floating objects, information which is essential in formulating effective mitigation and management measures. This study aims to address this knowledge gap through the use of various electronic tagging techniques in conjunction with dietary analysis. Dietary data were collected from 323 silky sharks incidentally caught at FADs. Approximately 40 percent of the diet consisted of prey associated with FADs while the remaining 60 percent were non-associated species of crustaceans, cephalopods and fishes. These results suggest that the associative behaviour is not primarily driven by trophic enhancement, but is likely a combination of predator avoidance, social interactions and feeding. Fine-scale behavioural data from silky sharks associated with drifting FADs were collected through the use of acoustic telemetry techniques. Acoustic tags were implanted into 38 silky sharks (69- 116 cm TL) at eight FADs. FADs were equipped with satellite linked acoustic receivers and abandoned to drift freely. Presence/absence and swimming depth data were telemetered via the Iridium satellite system. A total of 300 d of behavioural data were collected from 20 tagged individuals. Individuals remain associated with the same FAD for extended periods (min = 2.84 d, max = 30.60 d, mean = 15.69 d). Strong diel patterns were observed in both association and swimming depth. Typically individuals moved away from FADs after sunset and return later during the night, then remain closely associated until the following evening. Vertical behaviour also changed around sunset with sharks using fairly constant depths, within the upper 25 m, during the day and switching to rapid vertical movements during the night, with dives in excess of 250 m recoded. Broader scale movement behaviour was investigated using pop-up archival satellite tags (PSATs). Tags were deployed on 46 silky sharks (86-224.5 cm TL) for a total of 1495 d. Light data were used to calculate geolocation estimates and reconstruct the sharks’ trajectories. Movement patterns differed between animals and according to deployment duration. Several extensive horizontal movements were observed, with an average track length of 3240 km during an average tag deployment of 44.02 d. Horizontal movement patterns were found to correlate very closely with drift patterns of FADs. Consequently, it appears that the movement behaviour of juvenile silky sharks is strongly influenced by the movement of drifting FADs in this region. Ghost fishing of silky sharks through entanglement in FADs was also investigated using data derived from PSATs as well as underwater visual censuses. Thirteen per cent of the tagged sharks became entangled in FADs and entangled sharks were observed in 35 percent of the 51 FADs surveyed. Using this information in conjunction with estimated time that sharks remained entangled in the FAD (from depth data from PSATs), and scaling up according to estimates of FAD numbers, it was found that between 480 000 and 960 000 silky sharks are killed annually in this manner in the Indian Ocean. Subsequent management measures in this region prevent the deployment of FADs with netting that could lead to entanglement. Overall, floating objects appear to play a significant role in the juvenile life stages of silky sharks in this region. While their association with floating objects is clearly advantageous in an evolutionary sense, under current tuna fishery trends, this behaviour is certainly detrimental for the population.

- Description: This thesis is an investigation of the call centre labour process focusing on both the way work is organised and experienced by the operators and the management of employee relations inside a local government workplace – The City of Johannesburg (COJ) Call Centre. The City of Johannesburg is used to understand labour process dynamics in local government. Johannesburg was recently named the ‘World’s Second Most Inspiring City’ (by Global City Index, 2014) possessing economic and political influence within and around South Africa. Johannesburg is the economic hub of the country, contributing nine per cent to the South African GDP. But it needs to be emphasised that the focus of this thesis is on the labour process rather than the City of Johannesburg. The nature of work in the COJ local government call centre involves answering calls about water, electricity, refuse removal, accounts and other general municipal services. The government’s objective was to have a more customer centred service delivery plan in compliance with the Batho Pele (People first) framework (RSA, 1997, White Paper on Local Government). This resulted in a shift in the government ‘mantra’ and philosophy from viewing Johannesburg residents as customers rather than citizens. This shift from public administration to New Public Management (NPM) has meant the introduction of private sector business principles into public services. This seeks to improve organisational and individual performance to enhance service delivery within South African municipalities. This is also meant to promote responsive and performance management culture. The study uses Labour Process Theory set out in Labour and Monopoly Capital (LMC-1974) by Harry Braverman to argue for the need to contextualise the labour process debate within the socio-political understanding of work in South Africa. In developing the argument about the narrow analysis of public sector call centres based on economic lines I have reviewed literature on call centres which is influenced by Michel Foucault and Harry Braverman to understand the public sector call centre labour process. Inspired by Adesina (2002) and Mafeje (1981) this study uses a ‘complementarity’ approach to knowledge. This means the local context tends to complement the global trends though not without contradictions within the local context. The case study approach was used for in-depth analysis of the local context through different methods (semi-structured interviews, focus group discussions, observations and survey questionnaires) to collect data from different stakeholders within the City of Johannesburg between 2010 and 2013. The concept of a ‘(dis)connecting city’ is fitting for the COJ as this city connects the rest of Sub-Saharan Africa and South Africa to the world through its commerce and political influence. Based on the challenges experienced in this local government call centre I attempt to show the the politicised nature of the public sector call centre labour process. I use the differential power relations in the frontline between customer and Joburg connect workers to illustrate this point. The lack of emphasis on the politicised nature of public sector call centres within literature is informed by the de-contextualised nature of these call centres. In the end, I argue that public sector call centre labour process is multilayered, contradictory and complicated because of these workers’ role as citizens, customers, community members and service providers from a politicised community environment.

- Description: This thesis is an investigation of the call centre labour process focusing on both the way work is organised and experienced by the operators and the management of employee relations inside a local government workplace – The City of Johannesburg (COJ) Call Centre. The City of Johannesburg is used to understand labour process dynamics in local government. Johannesburg was recently named the ‘World’s Second Most Inspiring City’ (by Global City Index, 2014) possessing economic and political influence within and around South Africa. Johannesburg is the economic hub of the country, contributing nine per cent to the South African GDP. But it needs to be emphasised that the focus of this thesis is on the labour process rather than the City of Johannesburg. The nature of work in the COJ local government call centre involves answering calls about water, electricity, refuse removal, accounts and other general municipal services. The government’s objective was to have a more customer centred service delivery plan in compliance with the Batho Pele (People first) framework (RSA, 1997, White Paper on Local Government). This resulted in a shift in the government ‘mantra’ and philosophy from viewing Johannesburg residents as customers rather than citizens. This shift from public administration to New Public Management (NPM) has meant the introduction of private sector business principles into public services. This seeks to improve organisational and individual performance to enhance service delivery within South African municipalities. This is also meant to promote responsive and performance management culture. The study uses Labour Process Theory set out in Labour and Monopoly Capital (LMC-1974) by Harry Braverman to argue for the need to contextualise the labour process debate within the socio-political understanding of work in South Africa. In developing the argument about the narrow analysis of public sector call centres based on economic lines I have reviewed literature on call centres which is influenced by Michel Foucault and Harry Braverman to understand the public sector call centre labour process. Inspired by Adesina (2002) and Mafeje (1981) this study uses a ‘complementarity’ approach to knowledge. This means the local context tends to complement the global trends though not without contradictions within the local context. The case study approach was used for in-depth analysis of the local context through different methods (semi-structured interviews, focus group discussions, observations and survey questionnaires) to collect data from different stakeholders within the City of Johannesburg between 2010 and 2013. The concept of a ‘(dis)connecting city’ is fitting for the COJ as this city connects the rest of Sub-Saharan Africa and South Africa to the world through its commerce and political influence. Based on the challenges experienced in this local government call centre I attempt to show the the politicised nature of the public sector call centre labour process. I use the differential power relations in the frontline between customer and Joburg connect workers to illustrate this point. The lack of emphasis on the politicised nature of public sector call centres within literature is informed by the de-contextualised nature of these call centres. In the end, I argue that public sector call centre labour process is multilayered, contradictory and complicated because of these workers’ role as citizens, customers, community members and service providers from a politicised community environment.

However, this study also found that many of the guest houses in the area are not aware of government incentives available to support them and very few of them have made use of these services. This is something that needs to be addressed if these guest houses are to continue to strengthen the local economy and provide employment opportunities in a sustainable manner. The study revealed that many of the guest houses are operating in an environmentally friendly manner and this will ensure their future sustainability. The broader situation and the contribution of the accommodation sector as critical assets in local and national tourism economies has been thoroughly researched in tourism research around the world. Existing work on the accommodation sector in the South African tourism economy is mainly urban-focused and indicates that its local development impacts can be positive albeit not always maximised through local linkages, however, only a limited amount of academic investigations examines the contribution of tourism sub-sectors to economic growth and employment. - Description: The Republic of South Africa’s energy economy is under immense pressure and the main energy utility, Eskom, cannot reliably generate sufficient electrical energy to meet its customer demand. Water security in South Africa is also under threat as the country is currently amidst a water crisis. The South African manufacturing industry is the sector consuming the largest portion of the total energy consumption and second largest portion of the total water consumption (excluding urban use) nationally. Water and energy efficiency improvement is becoming imperative for organisational success as well as national economic sustainability. This is due to the significant increase in the cost of both electrical energy and water in recent years, and the drop in the reserve energy margin below the minimum level required for sustainable operation of energy utilities. The research objective therefore was to develop a Lean-based water and energy efficiency improvement framework for the manufacturing industry to encourage and realise a reduced demand for energy and water in it. A literature review was conducted on the water and energy landscape in South Africa, Lean manufacturing principles and their positive natural influence on water and energy efficiency, and the Value Stream Mapping tool. The survey of water and energy wastes in the manufacturing industry yielded the creation of standardised water and energy waste categories for use in conjunction with the conventional Lean wastes, tools and techniques. The literature survey indicated that the implementation of Lean manufacturing techniques naturally leads to the improvement of energy and water intensity. The researcher hypothesised that there is even greater potential for water and energy savings if a Lean-based tool is developed to specifically focus on the improvement of energy and water efficiency. A framework for this tool was developed through the literature survey. The tool is based on the Value Stream Mapping tool. A practical experiment was then conducted to establish the effectiveness of the tool in the South African manufacturing industry. The experiment design included an outline of the forecasting and statistical validation techniques to ensure the integrity of data utilised for approving or disproving the set hypotheses. The results of the practical experiment yielded positive results, confirming the hypothesis that the implementation of the developed framework will lead to improved energy and water efficiency in the manufacturing industry. After an explanation of the experimental results, the study concluded with recommendations and conclusions based on the findings.

- Description: South Africa has the highest species diversity of ericaceous plants belonging to the Erica genus. There are over 850 identified species in the Cape Floral Region. The Albany Centre of Endemism (ACOE) is located within this region and is a hotspot of diversity consisting of various plant genera. The success of Erica plants is ubiquitously attributed to mycorrhizal relationships they engage in with a diverse group of fungi. This symbiosis is known as the ericoid mycorrhizal (ERM) association. The overall aim of this study was to establish the diversity of root fungi associated with Erica plants using morphological, molecular and 454 pyrosequencing techniques. Six Erica species were identified using leaf and flower morphology according to taxonomic keys. The identified plants were Erica cerinthoides, Erica demissa, Erica chamissonis, Erica glumiflora, Erica caffra and Erica nemorosa. Roots from sampled plants were stained and examined microscopically to determine their mycorrhizal status. Ericoid mycorrhizal associations together with dark septate endophyte (DSE) structures and hyphae that did not form any specific structure were observed in all the roots. In addition arbuscular mycorrhizal (AM) structures in the form of vesicles were detected in E. glumiflora and E. cerinthoides. In order to identify the culturable fungi associated with the respective hosts, sterilised roots were placed on various culture media for cultivation. Thereafter isolated fungi were morphologically classified into 67 morphotypes. These were mostly sterile and darkly pigmented. Non-sporulating mycelia of variable colouration such as white, cream-yellowish, beige, green and brown were also observed. Further identification was carried out using molecular techniques. DNA was extracted separately from pure cultures and amplified using ITS1 and ITS4 primers in a polymerase chain reaction (PCR). Thereafter sequencing and Basic Local Alignment Search Tool (BLAST) were used to identify the isolates to generic level. The fungi were taxonomically classified into 54 operational taxonomic units and 94 percent were Ascomycetes and Helotiales was the dominant order. Unclassified Helotiales with affinities to fungi currently identified as Epacrid root fungus was common in all hosts. Other isolates that were identified included Oidiodendron, Meliniomyces, Phialocephala, Cadophora, Lachnum, Leohumicola Cryptosporiopsis, Chaetomium, Acremonium and Epicoccum species. Basidiomycetes were represented by two OTUs belonging to the genus Mycena. Four OTUs comprised fungi that had no significant alignments in the reference databases. Direct root DNA extraction together with 454 pyrosequencing was used to detect the diversity of culturable and unculturable fungi associated with the identified hosts. The ITS2 region was targeted for sequencing. Although Ascomycetes remained the dominant phyla, Basidiomycetes were also detected in all host plants. Glomeromycota was present in E. caffra and E. cerinthoides. Helotiales was dominant in all Erica plants with the exception of E. cerinthoides and E. chamissonis which were dominated by the order Chaetothyriales. The OTUs identified to genus level included Epacris pulchella root fungus, Oidiodendron cf. maius, Acremonium implicatum, Leohumicola, Lachnum, Capronia and Mycena species. Culture-based techniques and pyrosequencing detected similar fungal composition comprising Ascomycetes, while, pyrosequencing was able to detect Glomeromycetes and Basidiomycetes.

The ITS2 region was targeted for sequencing. Although Ascomycetes remained the dominant phyla, Basidiomycetes were also detected in all host plants. Glomeromycota was present in E. caffra and E. cerinthoides. Helotiales was dominant in all Erica plants with the exception of E. cerinthoides and E. chamissonis which were dominated by the order Chaetothyriales. The OTUs identified to genus level included Epacris pulchella root fungus, Oidiodendron cf. maius, Acremonium implicatum, Leohumicola, Lachnum, Capronia and Mycena species. Culture-based techniques and pyrosequencing detected similar fungal composition comprising Ascomycetes, while, pyrosequencing was able to detect Glomeromycetes and Basidiomycetes.

- Description: The main objective of the current study was to determine the effect of distance travelled (DT), lairage duration (LDhr) and animal-related factors (animal class, breed, sex) on pre-slaughter stress indicators, carcass characteristics, nanostructure and technological properties of beef. The study was conducted in an Eastern Cape abattoir under the Buffalo City District Municipality. Six genotypes of cattle (Beefmaster, Bonsmara, Brahman, Holstein-Friesian, Non-descript and Nguni) brought for slaughter at the abattoir on different days were used in the study. Slaughter weight (SWT), warm and cold carcass weight (WCW and CCW), fatness and conformation of each animal were measured and recorded. Representative samples of blood plasma and meat quality from the Muscularis thoracis et lumborum (LTL) were collected at slaughter and 24 hours after slaughter, respectively. Heat shock proteins 70kDa (HSPA1A), glucose (GLU), cortisol (CORT), creatine kinase (CK) and lactate dehydrogenase (LDH) were determined from the blood plasma. The representative LTL muscle was used to determine ultimate pH (pHu), lightness (L*), redness (a*), yellowness (b*), meat temperature (Tm), Warner Bratzler Shear Force (WBSF) and histological structure of beef. There were significant effects of distance on pHu, Tm, a*, b*, TL%, CK and LDH. Pearson correlations were observed between distance travelled (DT), creatine kinase (CK), lactate dehydrogenase (LDH), ultimate pH (pHu), colour coordinates (L*, a* and b*) and Warner Bratzler Shear Force (WBSF). Principal component analysis (PCA) revealed that the 1st (pHu), 2nd (Tm) and 3rd (L*) principal components were the critical meat quality attributes. Positive relationships between carcass weights and beef quality characteristics for bovine cattle where the cumulative percentage of the first six principal component`s (PC`s) (SWT, WCW, CCW, DP, pHu and Tm contributed to 93.4 % of the total variance. Significant correlation coefficients between DT (r = 0.15), LDhr (r = 0.29), Ta (r = -0.52), Tm (r = 0.26), L* (r = 0.33), a* (r = 0.32) and WBSF (r = -0.49) were observed. Meat from Brahman (BR), Non-descript (ND) and Nguni (NG) cows had visible skeletal muscle fibres which were thin and long indicating improved tenderness of beef. Exposing animals to longer hours of transportation did not only reduce glucose levels but also increased the expression of HSPA1A and levels of cortisol. The CK and LDH were not related to beef quality. Therefore, CK and LDH can be used as indicators of animal welfare in slaughter cattle but cannot be used to predict the quality of meat. Bulls were the heaviest, although heifers had the highest dressing percentage than cows. Furthermore, the nanostructure of beef indicated that heifers had the best muscle fibres, sarcomere length and visible intercalated discs. The observed nanostructures of Nguni genotype are an indication of tender meat. Beef from heifers of Nguni had visible and tender meat with traces of intramuscular fat while males had shorter sarcomere length and tougher meat.

A total of 85 profit-making firms in Nairobi with a sample of 165 management employees were selected for this study. The empirical findings obtained relate to four outsourcing theories. Transaction Cost Analysis (TCA) Theory focuses on the cost savings that result from outsourcing. Agency Outsourcing Theory centres on outsourcing firms hiring agents to achieve productivity. Hiring agents may result in permanent staff being retrenched and additional outsourcing personnel being contracted and job creation and/or job loss results. Expectation Confirmation Theory (ECT) emphasises the importance of an outsourcing provider conforming to quality management principles. Resource Based Theory (RBT) proposes that organisations need a collection of resources and capabilities to execute outsourcing successfully. Findings further suggested that outsourcing can yield positive and/or negative outcomes depending on risks encountered, the business environment, company policies, function/s to be outsourced, and the competence and commitment of an outsourcing vendor. To further enhance the positive impact of outsourcing three improvements need to be executed: formulation of standard policies, price regulations, and commitment of outsourcing firms in adhering to set contract deadlines. It is suggested that the following would allow organisations to gain more from outsourcing in the future: the adoption of international/offshore outsourcing practices, more commitment by outsourcing consultants, the standardisation of charges for outsourcing contracts, and the use of new technology that would improve how outsourcing is conducted. It is concluded that the positive impacts of outsourcing would foster development to some extent while the possible negative impact of outsourcing would impede development.

Data mining can create a competitive advantage for corporations by discovering business relevant information, such as patterns, relationships, and rules. The role of the human user within the data mining process is crucial, which is why the research area of domain knowledge becomes increasingly important. This thesis investigates the impact of domain knowledge-driven variable derivation on classifier performance for corporate data mining. Domain knowledge is defined as methodological, data and business know-how. The thesis investigates the topic from a new perspective by shifting the focus from a one-sided approach, namely a purely analytic or purely theoretical approach towards a target group-oriented (researcher and practitioner) approach which puts the methodological aspect by means of a scientific guideline in the centre of the research. In order to ensure feasibility and practical relevance of the guideline, it is adapted and applied to the requirements of a practical business case. Thus, the thesis examines the topic from both perspectives, a theoretical and practical perspective. Therewith, it overcomes the limitation of a one-sided approach which mostly lacks practical relevance or generalisability of the results. The primary objective of this thesis is to provide a scientific guideline which should enable both practitioners and researchers to move forward the domain knowledge-driven research for variable derivation on a corporate basis. In the theoretical part, a broad overview of the main aspects which are necessary to undertake the research are given, such as the concept of domain knowledge, the data mining task of classification, variable derivation as a subtask of data preparation, and evaluation techniques. This part of the thesis refers to the methodological aspect of domain knowledge. In the practical part, a research design is developed for testing six hypotheses related to domain knowledge-driven variable derivation. The major contribution of the empirical study is concerned with testing the impact of domain knowledge on a real business data set compared to the impact of a standard and randomly derived data set. The business application of the research is a binary classification problem in the domain of an insurance business, which deals with the prediction of damages in legal expenses insurances. Domain knowledge is expressed through deriving the corporate variables by means of the business and data-driven constructive induction strategy. Six variable derivation steps are investigated: normalisation, instance relation, discretisation, categorical encoding, ratio, and multivariate mathematical function. The impact of the domain knowledge is examined by pairwise (with and without derived variables) performance comparisons for five classification techniques (decision trees, naive Bayes, logistic regression, artificial neural networks, k-nearest neighbours). The impact is measured by two classifier performance criteria: sensitivity and area under the ROC-curve (AUC). The McNemar significance test is used to verify the results. Based on the results, two hypotheses are clearly verified and accepted, three hypotheses are partly verified, and one hypothesis had to be rejected on the basis of the case study results. The thesis reveals a significant positive impact of domain knowledge-driven variable derivation on classifier performance for options of all six tested steps. Furthermore, the findings indicate that the classification technique influences the impact of the variable derivation steps, and the bundling of steps has a significant higher performance impact if the variables are derived by using domain knowledge (compared to a non-knowledge application). Finally, the research turns out that an empirical examination of the domain knowledge impact is very complex due to a high level of interaction between the selected research parameters (variable derivation step, classification technique, and performance criteria).

The secondary metabolome of these organisms consists of structurally diverse molecules possessing unprecedented carbon skeletons, the biosynthesis of which occurs via complex metabolomic pathways driven by specialist enzymes. This structural novelty is highly influential on the favourable bioactivity these compounds display. A prominent example of such a compound is trabectedin marketed as Yondelis®. Registered for the treatment of soft tissue sarcomas, this marine drug was developed from extracts of the tunicate Ecteinascidia turbinata. South Africa is renowned for possessing a highly diverse marine biota including several endemic species of marine red algae belonging to the Laurencia sensu stricto genus, which falls within the Laurencia complex. Despite having a good reputation for fascinating secondary metabolites, the taxonomy of Laurencia natural products is proving challenging for reasons including the presence of cryptic species, as well as individual species displaying morphological variability. The aim of this study was thus to isolate secondary metabolites from various South African Laurencia spp. and subsequently assess their chemotaxonomic significance by analysis of a parallel plastid rbcL phylogeny study of Laurencia spp. This study reports the first phycochemical investigation into Laurencia natalensis Kylin, Laurencia cf. corymbosa J.Agardh, Laurencia complanata (Suhr) Kützing, Laurencia sodwaniensis Francis, Bolton, Mattio and Anderson submitted, Laurencia multiclavata Francis, Bolton, Mattio and Anderson submitted, and a South African specimen of Laurenciella marilzae Gil-Rodríguez, Sentíes, Díaz-Larrea, Cassano and M.T. Fujii (basionym: Laurencia marilzae) originally described from Spain. Additionally, the chemical profiles of previously explored species Laurencia flexuosa Kützing and Laurencia glomerata Kützing were re-investigated. The organic extracts of the above species afforded 31 compounds belonging to a wide array of structural classes including halo-chamigranes, linear C15 acetogenins, indole alkaloids, cuparanes and cyclic bromo-ethers. A new tri-cyclic keto-cuparane (4.4) was isolated from L.cf. corymbosa alongside the new cuparanes 4.1 and 4.7. Algoane (5.9), a unique marker compound isolated from L. natalensis, was previously only reported from a sea-hare. Such marker compounds which are exclusive to an individual algal species increase the ease of their subsequent identification. The feasibility of chemotaxonomy as an additional tool to classify Laurencia spp. Was established as broad predictions of a specimen’s phylogeny, based on representatives of its secondary metabolome, proved viable. The study specimens were shown to possess similar chemical profiles to their sister species e.g. L. complanata, L. sodwaniensis and L. multiclavata produced similar metabolites to their sister species as inferred by an rbcL phylogeny tree. Finally, a 1H NMR profiling study on the crude organic extracts of various Laurencia spp. generated distinctive, reproducible spectra, exposing the value of NMR spectroscopy as a rudimentary species discernment tool.

The potential of dance education to promote social cohesion in a post-conflict society: perspectives of South African pre-service student teachers

Theoretical connections between culture, dance education and social cohesion are explored. The empirical investigation is designed as a qualitative case study interrogating pre-service student teachers’ experiences and perceptions of a particular dance education course in a culturally and politically diverse university classroom in post-apartheid South Africa. Open-ended questionnaires, reflective journals and focus group interviews were employed to generate data. Findings indicate that involvement in creative movement and ethno-cultural dances raised awareness of the Self and the Other, engendering perspective and personal transformation, important requisites for social transformation and subsequently social cohesion in a formerly divided society, such as South Africa. In addition, these dance education experiences provided participants with unique encounters with the Other’s culture. These occurred through embodied experiences of the culture of the Other, as well as through bodily negotiations with the Other. It argues further that the biopolitical philosophy upon which South African citizenship is anchored is itself a hindrance to the realisation of efforts aimed at naturalising refugees and their descendants. The research methodology used in this study is non-empirical. This is so because the study is based on available data, information already available in print or on the internet. The study attempts to accomplish the above by undertaking an in-depth analysis of the history of refugees, the current position of naturalisation under international law, and identifies the inherent challenges. In the South African context, the study makes use of extensive statutory, constitutional and case law materials to justify that the current treatment of refugees in their quest for naturalisation is indefensible within the context of a human rights-based approach and the dictates of the Constitution. This study concludes by making recommendations that would help close the legal and policy gaps that obtain presently. These include amendments to the Refugees, Immigration and Citizenship Acts and strengthening policy implementation at the DHA. It is hoped that the recommendations will strengthen and evolve a human rights culture and bring refugee, immigration and citizenship laws in line with the Constitution. It will also pave the way for a more just and peaceful South Africa as she strives to meet her obligations under regional and international law.

- Description: Defining leadership effectiveness (LE) remains a controversial subject with scholars and researchers disagreeing on a common definition. Most studies term leadership effectiveness as the leaders’ ability to successfully exercise personal influence and abilities in order to accomplish set standards. Such leadership effectiveness was measured using three elements, namely influence, follow commitment and versatility. To be effective, leaders must possess certain competencies and perform specific tasks (independent variables). In this study, three leadership competencies, namely strategic thinking, emotional effectiveness and transformational leadership were examined. Similarly, seven leadership tasks (set organisational direction, develop human capital, build core competencies, create organisational alignment, sustain appropriate organisational culture, manage change and establish balanced organisational control) were identified as essential for the attainment of leadership effectiveness (intervening variable) which, in turn, leads to organisational effectiveness (dependent variable). Given the importance of leadership effectiveness to organisational effectiveness, the purpose of this study was to identify, investigate and empirically test the possible relationship between leadership effectiveness and organisational effectiveness within the Kenyan indigenous banks. To achieve the aim of the study, a survey was undertaken using a structured self-administered questionnaire. The respondents were identified using both probability and non-probability techniques with the survey yielding 257 usable questionnaires that were statistically analysed. The proposed hypothetical relationship between leadership effectiveness and organisational effectiveness was assessed using Structural Equation Modelling (SEM), a multivariate statistical technique. The SEM included Cronbach’s alpha and confirmatory factor analyses to assess the dicriminant reliability and validity of the measuring instrument, and Goodness-of-fit indices. The study findings demonstrated the existence of a strong relationship between leadership competencies, tasks performed and leadership effectiveness which in turn impacts on organisational effectiveness, in this case the overall effectiveness of the Kenyan indigenous banks.

The study showed that the role of AMMs in the planning and implementation of curriculum change in PHEIs was too complex and demanding because they spent most of their time on daily administrative routines instead of on core academic activities such as planning and implementing curriculum change in their departments. The AMMs in the PHEIs under study operated more like managers in academic departments than academics in management. As a result the study showed that AMMs faced more challenges than opportunities in their planning and implementation of curriculum change in PHEIs. The major challenges AMMs faced in the planning and implementation of curriculum change were a highly controlled and strict work environment, role conflict, lack of autonomy, role strain and heavy workloads which limited the time AMMs spent on the core business of managing curriculum change in their departments. The study also highlighted some of the strategies albeit a few, which, despite the numerous challenges AMMs faced, are used to try and make the planning and implementation of curriculum change by AMMs was to some extent successful. The study provided insight on the influence of AMMs biographical characteristics as well as the influence of AMM job requirements (such as having a detailed job description and having authority over curriculum matters) on how AMMs enacted their role in curriculum change. Based on the results of the study, a model to assist AMMs in the effective planning and implementation of curriculum change was proposed.

Added to this, the South African mining industry is facing an unparalleled number of challenges, including an uncertain regulatory environment, infrastructure constraints, frequent industrial actions, rising costs and shortages of skills. The dynamism of discontinuous change has increased considerably and the South African mining industry is today facing an uncertain future with a blurred outlook. The results of the detailed analysis of future studies theory and practice in this research study support the argument that there is a strong need to fundamentally change the ways of planning for the future of the South African mining industry. The practice of developing new insight through the application of futures studies is central to this process. Today, collective decisions and strategies are progressively more founded on and informed by futures studies. The research study sought to develop insight regarding the future of the South African mining industry through the construction of four scenarios towards 2055: Divided We Fall, where a confident industry is threatened by social divisions as industry transformation is disregarded; Rock Bottom, where weak global economic conditions coincide with lacklustre industry innovation; Rising from Ashes, with similar economic conditions, but the industry responding positively through accelerated industry innovation; and Renaissance, set against positive global economic conditions with the South African mining industry adopting a collaborative, innovative approach to industry transformation. The research study further strived to uncover the preferred future for the South African mining industry as basis for the South African Mine of the Future Vision towards 2055. Throughout the research study, Inayatullah’s pillars of futures studies were applied as a guideline in mapping the present and future, deepening the future, broadening the future through the development of scenarios, and transforming the future by narrowing it down to the preferred. The study provides valuable insight into the driving forces relevant to the South African mining landscape. In addition, it provides insight on how to anticipate the changes these driving forces may bring about for the industry over the next 40 years from a decision-maker’s point of view. It is up to the mining industry to select the road to follow in terms of progress and sustainable development. Through an innovative approach, the creation of an environment of trust, the sharing of values, purposes and benefits, the South African Mine of the Future Vision is attainable. The South African mining industry must commit itself to working in collaborative partnerships with local communities, government, society and labour; stepping boldly into a world of social, environmental, technological and commercial innovation.

- Description: The environment in which higher education institutions operate is changing, and these changes are impacting on all aspects of higher education, including postgraduate levels. Changes wrought by globalisation, heralded by rapid advances in technology have inaugurated a new era in which there are long term consequences for higher education. The shift towards more quantitative and measurable "outputs" signifies a fundamental change in the educational ethos in institutions. Effectiveness is now judged primarily on numbers of graduates and publications rather than on other aspects. The drive is to produce a highly educated population, especially through increasing postgraduates who can drive national innovation and improve national economies. This affects academics in a range of ways, not least in the ways in which they engage in teaching, what they are willing to do and how they do it. Such changes influence the kinds of research done, the structures and funding which support research, and thus naturally shapes the kinds of postgraduate programmes and teaching that occurs. This study, situated in the field of Higher Education Studies, adopting a critical realist stance and drawing on the social theory of Margaret Archer and the concepts of expert and novice, explores the experiences of postgraduate supervisors from one South African institution across a range of disciplines. Individual experiences at the level of the Empirical and embodied in practice at the level of the Actual allow for the identification of possible mechanisms at the level of the Real which structure the sector. The research design then allows for an exploration across mezzo, macro and micro levels. Individuals outline their own particular situations, identifying a number of elements which enabled or constrained them and how, in exercising their agency, they develop their strategies for supervision drawing on a range of different resources that they identify and that may be available to them. Student characteristics, discipline status and placement, funding, and the emergent policy environment are all identified as influencing their practice. In some instances supervisors recognise the broader influences on the system that involve them in their undertaking, noting the international trends. Through their narratives and the discourses they engage a number of contradictions that have developed in the system with growing neo-liberal trends and vocationalism highlighting tensions between academic freedom and autonomy, and demands for productivity, efficiency and compliance, and between an educational focus and a training bias in particular along with others. Especially notable is how this contributes to the current ideologies surrounding knowledge and knowledge production. Their individual interests and concerns, and emergent academic identities as they take shape over time, also modifies the process and how individual supervisors influence their own environments in agentic moves becomes apparent. Whilst often individuals highlight the lack of support especially in the early phases of supervision, the emergent policy-constrained environment is also seen as curtailing possibilities and especially in limiting the possibilities for the exercise of agency. Whilst the study has some limitations in the range and number of respondents nevertheless the data provided rich evidence of how individual supervisors are affected, and how they respond in varied conditions. What is highlighted through these experiences are ways pressures are increasing for both supervisors and students and changing how they engage. Concerns in particular are raised about the growing functional and instrumental nature of the process with an emphasis on the effects on the kinds of researchers being developed and the knowledge that is therefore being produced. As costs increase for academics through the environments developed and with the varied roles they take on so they become more selective and reluctant to expand the role. This research has provided insights into ideas, beliefs and values relating to the postgraduate sector and to the process of postgraduate supervision and how it occurs. This includes the structures and cultural conditions that enable or constrain practitioners as they develop in the role in this particular institution. It has explored some of the ways that mechanisms at international, national and institutional levels shape the role and practices of supervisors. The effects of mechanisms are in no way a given or simply understood. In this way the research may contribute to more emancipatory knowledge which could be used in planning and deciding on emergent policies and practices which might create a more supportive and creative postgraduate environment.

- Description: The environment in which higher education institutions operate is changing, and these changes are impacting on all aspects of higher education, including postgraduate levels. Changes wrought by globalisation, heralded by rapid advances in technology have inaugurated a new era in which there are long term consequences for higher education. The shift towards more quantitative and measurable "outputs" signifies a fundamental change in the educational ethos in institutions. Effectiveness is now judged primarily on numbers of graduates and publications rather than on other aspects. The drive is to produce a highly educated population, especially through increasing postgraduates who can drive national innovation and improve national economies. This affects academics in a range of ways, not least in the ways in which they engage in teaching, what they are willing to do and how they do it. Such changes influence the kinds of research done, the structures and funding which support research, and thus naturally shapes the kinds of postgraduate programmes and teaching that occurs. This study, situated in the field of Higher Education Studies, adopting a critical realist stance and drawing on the social theory of Margaret Archer and the concepts of expert and novice, explores the experiences of postgraduate supervisors from one South African institution across a range of disciplines. Individual experiences at the level of the Empirical and embodied in practice at the level of the Actual allow for the identification of possible mechanisms at the level of the Real which structure the sector. The research design then allows for an exploration across mezzo, macro and micro levels. Individuals outline their own particular situations, identifying a number of elements which enabled or constrained them and how, in exercising their agency, they develop their strategies for supervision drawing on a range of different resources that they identify and that may be available to them. Student characteristics, discipline status and placement, funding, and the emergent policy environment are all identified as influencing their practice. In some instances supervisors recognise the broader influences on the system that involve them in their undertaking, noting the international trends. Through their narratives and the discourses they engage a number of contradictions that have developed in the system with growing neo-liberal trends and vocationalism highlighting tensions between academic freedom and autonomy, and demands for productivity, efficiency and compliance, and between an educational focus and a training bias in particular along with others. Especially notable is how this contributes to the current ideologies surrounding knowledge and knowledge production. Their individual interests and concerns, and emergent academic identities as they take shape over time, also modifies the process and how individual supervisors influence their own environments in agentic moves becomes apparent. Whilst often individuals highlight the lack of support especially in the early phases of supervision, the emergent policy-constrained environment is also seen as curtailing possibilities and especially in limiting the possibilities for the exercise of agency. Whilst the study has some limitations in the range and number of respondents nevertheless the data provided rich evidence of how individual supervisors are affected, and how they respond in varied conditions. What is highlighted through these experiences are ways pressures are increasing for both supervisors and students and changing how they engage. Concerns in particular are raised about the growing functional and instrumental nature of the process with an emphasis on the effects on the kinds of researchers being developed and the knowledge that is therefore being produced. As costs increase for academics through the environments developed and with the varied roles they take on so they become more selective and reluctant to expand the role. This research has provided insights into ideas, beliefs and values relating to the postgraduate sector and to the process of postgraduate supervision and how it occurs. This includes the structures and cultural conditions that enable or constrain practitioners as they develop in the role in this particular institution. It has explored some of the ways that mechanisms at international, national and institutional levels shape the role and practices of supervisors. The effects of mechanisms are in no way a given or simply understood. In this way the research may contribute to more emancipatory knowledge which could be used in planning and deciding on emergent policies and practices which might create a more supportive and creative postgraduate environment.
