- Description: Mixed Reality is a relatively new field in computer science which uses technology as a medium to provide modified or enhanced views of reality or to virtually generate a new reality. Augmented Reality is a branch of Mixed Reality which blends the real-world as viewed through a computer interface with virtual objects generated by a computer. The 21st century commodification of mobile devices with multi-core Central Processing Units, Graphics Processing Units, high definition displays and multiple sensors controlled by capable Operating Systems such as Android and iOS means that Mobile Augmented Reality applications have become increasingly feasible. Mobile Augmented Reality is a multi-disciplinary field requiring a synthesis of many technologies such as computer graphics, computer vision, machine learning and mobile device programming while also requiring theoretical knowledge of diverse fields such as Linear Algebra, Projective and Differential Geometry, Probability and Optimisation. This multi-disciplinary nature has led to a fragmentation of knowledge into various specialisations, making it difficult to integrate different solution components into a coherent architecture. Software design patterns provide a solution space of tried and tested best practices for a specified problem within a given context. The solution space is non-prescriptive and is described in terms of relationships between roles that can be assigned to software components. Architectural patterns are used to specify high level designs of complete systems, as opposed to domain or tactical level patterns that address specific lower level problem areas. Pattern Languages comprise multiple software patterns combining in multiple possible sequences to form a language with the individual patterns forming the language vocabulary while the valid sequences through the patterns define the grammar. Pattern Languages provide flexible generalised solutions within a particular domain that can be customised to solve problems of differing characteristics and levels of iii complexity within the domain. The specification of one or more Pattern Languages tailored to the Mobile Augmented Reality domain can therefore provide a generalised guide for the design and architecture of Mobile Augmented Reality applications from an architectural level down to the ”nuts-and-bolts” implementation level. While there is a large body of research into the technical specialisations pertaining to Mobile Augmented Reality, there is a dearth of up-to-date literature covering Mobile Augmented Reality design. This thesis fills this vacuum by: 1. Providing architectural patterns that provide the spine on which the design of Mobile Augmented Reality artefacts can be based; 2. Documenting existing patterns within the context of Mobile Augmented Reality; 3. Identifying new patterns specific to Mobile Augmented Reality; and 4. Combining the patterns into Pattern Languages for Detection & Tracking, Rendering & Interaction and Data Access for Mobile Augmented Reality. The resulting Pattern Languages support design at multiple levels of complexity from an object-oriented framework down to specific one-off Augmented Reality applications. The practical contribution of this thesis is the specification of architectural patterns and Pattern Language that provide a unified design approach for both the overall architecture and the detailed design of Mobile Augmented Reality artefacts. The theoretical contribution is a design theory for Mobile Augmented Reality gleaned from the extraction of patterns and creation of a pattern language or languages.

The effects of selected proline-based cyclic dipeptides on growth and induction of apoptosis in cancer cells

- Description: An increasing number of cyclic dipeptides (CDPs) have been shown to exhibit important biological activity including antifungal, antibacterial, anticonvulsant and immunomodulatory activity. Furthermore, some CDP derivatives have been shown to exhibit antitumour activity in vitro and in vivo. Several proline-based CDPs that exhibit biological activity have been detected in various processed foods and beverages. In the present study, the potential of seven proline-based CDPs to inhibit cancer cell growth was investigated in HT-29 (colon), HeLa (cervical), MCF-7 (breast) and WHCO3 (oesophageal) cancer cell lines. The CDPs used in this study were cyclo(Phe-Pro), cyclo(Tyr-Pro), cyclo(Gly-Pro), cyclo(Pro- Pro), cyclo(His-Pro), cyclo(Leu-Pro) and cyclo(Thr-Pro). The sulforhodamine B (SRB) cell growth assay was used in an initial screening phase to investigate the effects of the CDPs in HT-29, HeLa and MCF-7 cells. After exposing the cells to 10mM of the respective CDPs for 48 hours, the SRB assay results showed that only cyclo(Phe-Pro) exhibited more than 50% growth inhibition (p<0.01) in the three cell lines. The other CDPs showed comparatively marginal growth-inhibitory effects, except for cyclo(Tyr-Pro), which exhibited a pronounced effect in MCF-7 cells compared to HT-29 and HeLa cells. The MTT assay was used to confirm the SRB assay results for cyclo(Phe-Pro) and cyclo(Tyr-Pro), extending the investigation to the use of the fourth cell line WHCO3 and using a longer exposure time of 72 hours. The MTT assay demonstrated a dosedependent (0.008-10 mM) growth inhibition by cyclo(Phe-Pro) with an IC50 value of 4.04 ± 1.15 mM for HT-29 cells. Cyclo(Phe-Pro) was subsequently used to investigate whether the growth-inhibitory effects of this CDP were related to the induction of apoptosis in HT-29 cells. Hoechst 33342 staining showed that 5mM cyclo(Phe-Pro) induced characteristic chromatin condensation and nuclear fragmentation in 18.3 ± 2.8% (p<0.01) of HT-29 cells after 72 hours. Furthermore, annexin V binding revealed that HT-29 cells treated with 5 mM cyclo(Phe-Pro) displayed phosphatidylserine externalization after 48 hours. In addition, it was shown that 10 mM cyclo(Phe-Pro) induced poly(ADP-ribose)polymerase PARP cleavage, one of the hallmark events of apoptosis. The use of the broad-range caspase inhibitor Z-VAD-FMK, showed that this PARP cleavage was caspase-dependent, which in turn was confirmed by demonstrating an increase in caspase-3 activity (p<0.01) in cyclo(Phe- Pro)-treated HT-29 cells. In conclusion, these findings demonstrate that cyclo(Phe-Pro) inhibited the growth of HT- 29, MCF-7, HeLa and WHCO3 cells, and induced apoptosis in HT-29 colon cancer cells, suggesting the potential antitumour activity of cyclo(Phe-Pro)-related CDPs.

Exploring the linkages between biodiversity conservation and ecotourism in protected areas

- Description: The prospect of using ecotourism to add value to conservation initiatives has frequently been stated. However, there is a general lack of information on the costs and benefits of biodiversity and its management, and how this can be influenced by the ecotourism industry. This study seeks to explore the value of biodiversity from the perspective of ecotourism in a hierarchical series of studies that move from the biome-level perspective down to the value of juveniles in ecotourism. Ecotourism plays a significant role in the establishment and management of Private Protected Areas (PPAs) and hence the opportunity to be added to the conservation estate. The financial opportunity to be found in ‘big five’ areas had led to an uneven biome coverage of PPAs which has resulted in the over-representation of certain areas and a skewed contribution towards national conservation targets. The landscape-use of PPAs have indicated that ecotourism operators are largely driven to achieve successful game-viewing sightings which has influenced management and affected the landscape use of PPAs, the stocking rate of species, the introduction of extralimital species, and management of flagship species. These management decisions are based on anecdotal sources and have been made at the expense of biodiversity and therefore apparent conflict exists between ecotourism and conservation objectives. This is based on lack of information: ecotourism operators, despite the large area they protect, have not realised their potential to contribute to conservation. It is therefore important to develop policy and incentives to motivate the establishment of PPAs in areas of high conservation significance in order to contribute to national conservation targets. Ecotourism should be used as a solution to provide important insights into tourist preferences which contribute to the larger debate of how to establish a balance between managing animal populations to ensure tourist satisfaction while supporting conservation. To ensure sustainability of PPAs, it is strongly recommended to manage PPAs more intensively with regards to PPA size, species stocking rates and landscape use. The presence of juveniles at an animal sighting significantly contributes to tourist satisfaction which suggests that the loss of juveniles may reduce the attraction. This is used to provide recommendations that using contraception to control elephant population numbers should be applied with caution as a management tool as it may be counterproductive by negatively impacting ecotourism. The findings from this study not only contribute to the conservation of biodiversity through the use of ecotourism, but will also ensure sustainability of the ecotourism industry and build capacity for sustainable living.

- Description: Mobile phones are increasingly being equipped with a wide range of sensors which enable a variety of interaction techniques. Sensor-based interaction techniques are particularly promising for domains such as map-based applications, where the user is required to interact with a large information space on the small screen of a mobile phone. Traditional interaction techniques have several shortcomings for interacting with mobile map-based applications. Keypad interaction offers limited control over panning speed and direction. Touch-screen interaction is often a two-handed form of interaction and results in the display being occluded during interaction. Sensor-based interaction provides the potential to address many of these shortcomings, but currently suffers from several limitations. The aim of this research was to propose enhancements to address the shortcomings of sensor-based interaction, with a particular focus on tilt interaction. A comparative study between tilt and keypad interaction was conducted using a prototype mobile map-based application. This user study was conducted in order to identify shortcomings and opportunities for improving tilt interaction techniques in this domain. Several shortcomings, including controllability, mental demand and practicality concerns were highlighted. Several enhanced tilt interaction techniques were proposed to address these shortcomings. These techniques were the use of visual and vibrotactile feedback, attractors, gesture zooming, sensitivity adaptation and dwell-time selection. The results of a comparative user study showed that the proposed techniques achieved several improvements in terms of the problem areas identified earlier. The use of sensor fusion for tilt interaction was compared to an accelerometer-only approach which has been widely applied in existing research. This evaluation was motivated by advances in mobile sensor technology which have led to the widespread adoption of digital compass and gyroscope sensors. The results of a comparative user study between sensor fusion and accelerometer-only implementations of tilt interaction showed several advantages for the use of sensor fusion, particularly in a walking context of use. Modifications to sensitivity adaptation and the use of tilt to perform zooming were also investigated. These modifications were designed to address controllability shortcomings identified in earlier experimental work. The results of a comparison between tilt zooming and Summary gesture zooming indicated that tilt zooming offered better results, both in terms of performance and subjective user ratings. Modifications to the original sensitivity adaptation algorithm were only partly successful. Greater accuracy improvements were achieved for walking tasks, but the use of dynamic dampening factors was found to be confusing. The results of this research were used to propose a framework for mobile tilt interaction. This framework provides an overview of the tilt interaction process and highlights how the enhanced techniques proposed in this research can be integrated into the design of tilt interaction techniques. The framework also proposes an application architecture which was implemented as an Application Programming Interface (API). This API was successfully used in the development of two prototype mobile applications incorporating tilt interaction.

- Description: Semiconductor materials used for making photovoltaic (PV) cells have defects and impurities due to constraints of keeping production costs low. Electrically active defects that are distributed over the bulk material lead to reduced overall performance of a photovoltaic (PV) cell. In this study an adaptable Light Beam Induced Current (LBIC) system was designed in order to characterize the local performance of PV cells. The system uses a laser source and objective lens mounted vertically above the sample on the X-Y stage. Two current pre-amplifiers are used for converting the photo-generated current from the PV cell and the signal from the reflection detector cell into a voltage that can be measured by the data acquisition board. Two configurations for measuring the photo-current maps of either bare cells or encapsulated PV mini-modules can be used. To add flexibility it was envisaged that the system would be built in such a way that it allows easy integration of carrier lifetime mapping capabilities while keeping costs to a minimum. The carrier lifetime measurement technique integrated into the LBIC system is based on the optical Open-Circuit Voltage-Decay (OCVD) method. In a single-crystalline silicon PV cell that was tested, photo-current and opencircuit voltage LBIC scans revealed shunting behaviour due to scratch marks on the front surface. The marks are believed to have been caused by poor handling during manufacturing process. Reduced photo-current due to edge shunting was observed towards the edges of the PV cell. In another sample of single crystalline silicon cell an edge shunt resulted in a 30 percent drop in photo-current measured. LBIC measurements performed on multi-crystalline cells revealed nonuniformities such as enhanced photo-current on one side of grain boundaries. These asymmetric enhancements of local photo-current are attributed to the incline of the grain boundary into the bulk of the material. LBIC results obtained from mini modules showed a high degree of mismatching from cell to cell. It is well known that mismatching can degrade the performance of a PV module with series connected cells. The LBIC measurements presented also illustrated the negative effects of delamination on the photo-current of PV modules. The LBIC measurements performed on mini PV modules highlight some of the benefits of using an LBIC technique as a tool for investigating PV cell’s local photo-current response. The effective lifetime maps obtained highlight the importance of optical OCVD method as a tool that can be used in conjunction with conventional LBIC technique. Effective minority carrier lifetimes around 40 ms were measured on a single crystalline silicon cell of 2.5 x 2.5 cm2. Local features such as cracks and lifetime degrading defects were revealed by LBIC maps and were also confirmed on effective lifetime maps. The results presented demonstrate the importance of using effective carrier lifetime maps to complement photocurrent maps of PV cells and identification of areas where defects are located.

CD23 is believed to play an important role in regulating allergic responses and in inflammation, amongst others. β2 integrins are important in a variety of cell-adhesion reactions during immune-inflammatory mechanisms and the binding of their natural ligands generates outside-in cellular signalling, leading to cell activation. Although the binding of CD23 to β2 integrins contributes to this signalling in monocytes, the interaction site for CD23 is unknown. This study focused on the interaction of three soluble CD23 proteins with the β2 integrins CD11b/CD18 and CD11c/CD18. Differentiated HL60, THP1 and U937 monocytic cells were used to demonstrate the binding of three recombinant CD23 constructs (corresponding to 16, 25 and 33 kDa human soluble CD23) to upregulated CD11b/CD18 and CD11c/CD18. This binding was partially blocked by an antibody specific for the CD11b/CD18 αI domain, demonstrating that αI domains are involved in binding to CD23. Recombinant αI domain proteins of CD11b and CD11c were demonstrated to bind CD23 using ELISA and in surface plasmon resonance spectroscopy. The dissociation constants for CD23-CD11b/CD18 and CD23-CD11c/CD18 are comparable to other integrin ligands. This study has shown that CD23 interacts directly with the αI domains of β2 integrins and that the interaction surface likely spans the lectin domain as well as either the stalk and/or C-terminal tail of CD23. This study also looked at the effect that soluble CD23 proteins had on monocyte biology. It appears that iv sCD23 proteins have little effect on the phagocytic or chemotactic ability of monocytes, while an increase in oxidative burst was shown with the 16 kDa and 25 kDa CD23 proteins. The antifungal activity of an aqueous Tulbaghia violacea plant extract against Aspergillus flavus

- Description: Phytochemical analysis of both HEA1 and the crude plant extract showed the presence of phenolics, tannins and saponins. Saponins were the predominant secondary metabolites and were mostly abundant in the plant extract and to a lesser extent in the active compound. Steroidal saponins, tannins and phenolics were also detected in the plant extract, but only the phenolics were detected in the active compound. The results of the phytochemical analysis showed that those compounds that were not present in the active compound could be removed from the crude extract during the TLC purification process. Investigation on the mechanism of action of the crude plant extract on the sterol production by A. flavus showed that the plant extract affected ergosterol biosynthesis by causing an accumulation of oxidosqualene in the ergosterol biosynthetic pathway resulting in a decline in ergosterol production. An oscillatory response in lanosterol production was observed in the presence of the plant extract, which may be an adaptation mechanism of A. flavus to unfavourable conditions and compensation for the loss of enzyme activity which may have occurred as a result of the accumulation of oxidosqualene. The antifungal activity of the plant extract on ergosterol production by A. flavus may also be due to saponins which target the cell membrane and ergosterol production in fungi. The effect of the plant extract on the fungal cell wall of A. flavus also showed that the plant extract caused a decline in β-(1, 3) glucan production by inhibiting β-glucan synthase. The plant extract also affected the chitin synthesis pathway of A. flavus, by causing a decline in chitin production, which was due to the inhibition of chitin synthase. Investigation of chitinase production using 4MU substrates showed that the plant extract caused an accumulation of chitobioses, by activating chitobiosidases and endochitinases. A decline in N-acetylglucosaminidase activity in the presence of the plant extract was observed and this prevented the formation of N-acetylglucosamine. The accumulation of chitobiosidase and endochitinase may be as a result of autolysis that may be triggered by A. flavus as a survival mechanism in the presence of the plant extract and as a compensatory mechanism for the loss of β-glucans and chitin. The antifungal effect of the plant extract on various components of the cell wall of A. flavus, makes T. violacea aqueous plant extract an ideal chemotherapeutic agent against both human and plant pathogens of Aspergillus. The broad spectrum of antifungal activity of T. violacea against A. flavus also eliminates any chances of the fungus developing resistance towards it and would make it a candidate for use as a potential antifungal agent. Further identification and possible chemical synthesis is needed to shed light on the safety and efficacy of the active compound for further development as a chemotherapeutic agent.

- Description: Penguins spend a large part of their life cycle at sea and are amongst the most threatened seabirds as they suffer high mortality and have low reproductive success. Across the world, many populations are currently declining despite the constant development of conservation actions. The lack of information at the individual level reduces the understanding of individual variability and its potential role in the population dynamics of these vulnerable species. For example, long-lived species are characterised by substantial individual heterogeneity in their contribution to the overall population dynamic that should be understood when considering the implemention of conservation policies. This thesis explored some of the primary mechanisms behind inter-individual differences in behaviour and breeding success in the endangered African penguin Spheniscus demersus. On Bird island (Algoa Bay), nest defence behaviours were investigated on adults brooding chicks 1-3 weeks-old and chicks were measured at regular intervals (every 5 days) to assess their growth rate, a proxy for breeding success in this species. All adults were sexed and implanted with a passive integrated transponder which was used for individual identification. In this species, males are usually larger than females and have a higher survival rates both as juveniles and as adults. A subsample of them was equipped with tracking devices (GPSs and Time Depth Recorders), sometimes over consecutive foraging trips and across years. Finally, mate and nest fidelity were investigated to understand the drivers and consequences of this trait in that population. Breeding success was linked to nest defence behaviours, with bolder birds generally showing lower breeding success. This relation could not, however, be explained by differences in foraging strategies. Males were generally bolder than females when defending the nests, but their foraging behaviour did not vary with their personality. They generally had lower foraging effort than females. By contrast, bolder females performed more sinuous path, more wiggles and travelled larger vertical distances than shyer ones. Overall, females were more flexible than males over consecutive trips, suggesting that they probably adjust their behaviours to the offspring needs in this species. However, no sex difference in behavioural flexibility between sexes was visible across years, indicating that environmental variability may equally impact females and males’ behaviour over such time-scale. Generally, consistency was time-scale dependent as penguins increased their foraging flexibility across years, potentially adjusting to the variability of the environment. Interestingly, foraging consistency over consecutive trips increased during years of poor environmental conditions, and individuals with consistent foraging strategies were more successful than more flexible individuals when resources were extremely scarce. Finally, while nest and mate fidelity were high in this species, individuals that changed nest/partners, improved their breeding outcome. Particularly, after low breeding success females were more likely to change nest site and thereby partner to possibly improve their fitness. The potential biased adult sex ratio towards males in this population may facilitate changes in females which may have more partners available to re-mate than males. The present thesis reveals the existence of individual differences in behaviour and breeding success which relate to personality in penguins. These findings highlight the importance to integrate individual variability to predict future population dynamics in the context of global changes to understand the resilience and vulnerability of the species.

- Description: The primary aim of this thesis was to explore the use of ambient seismic noise as a tool to map the subsurface of the Cape-Karoo and Karoo region of South Africa. The Karoo is an ideal laboratory to use ambient seismic signal to map the shallow subsurface, as it is a quiet and pristine environment with a relatively well known geology. Ambient seismic signals were continuously recorded at three independent networks (ArrayA, ArrayB and ArrayC). ArrayA and ArrayB comprised 17 temporary stand-alone seismic stations each and recorded ambient noise wavefields for a ten week period between August and October 2015. ArrayC comprised 19 temporary stand-alone seismic stations, recording ambient seismic noise for a period of six weeks between June and July 2016. ArrayA and ArrayB were installed in the south-eastern Cape-Karoo region, near the town of Jansenville and ArrayC was installed near the Cradock-Tarkastad region of South Africa. This thesis is made up of two separate studies. Firstly, the retrieval and coherency of Rayleigh surface waves extracted from the vertical component recordings. For the first time in the south-eastern Cape-Karoo and Karoo area, estimates of Green’s function from cross-correlating ambient noise data between stations pairs were reconstructed and shown, which can be successfully used to image the subsurface. The stacked cross-correlations between all station pairs show clear arrivals of the Rayleigh surface waves. The group velocities of the Rayleigh waves in the 2 to 7 seconds period range were picked and inverted to compute the 2-D group velocity maps. For ArrayA and ArrayB, the resulting 2-D group velocity maps at different periods resulted in a group velocity model from approximately 2 to 7 km depth, which generally show a high velocity anomaly in the north of the study area, most likely imaging the denser, thick sedimentary basin of the Karoo (Carboniferous-Permian). To the south, the low velocity anomaly likely corresponds to the overlying Jurassic- Cretaceous sequences of the younger Algoa Basin (Uitenhage Group). For ArrayC, the group velocity maps showed high velocity regions, which is consistent with the dolerite sill intrusions in the Karoo and the low velocity structures, which was interpreted as the Karoo sediments. Secondly, the study comprised of characterizing the ambient seismic noise source. The first order analysis of the symmetry of the cross-correlation function showed that, although the ambient noise sources are relatively homogeneously distributed in the study area, most (energetic) of the ambient seismic noise propagates from the coast of South Africa. This was verified by analyzing the azimuthal distribution of the ambient seismic noise.

- Description: The primary aim of this thesis was to explore the use of ambient seismic noise as a tool to map the subsurface of the Cape-Karoo and Karoo region of South Africa. The Karoo is an ideal laboratory to use ambient seismic signal to map the shallow subsurface, as it is a quiet and pristine environment with a relatively well known geology. Ambient seismic signals were continuously recorded at three independent networks (ArrayA, ArrayB and ArrayC). ArrayA and ArrayB comprised 17 temporary stand-alone seismic stations each and recorded ambient noise wavefields for a ten week period between August and October 2015. ArrayC comprised 19 temporary stand-alone seismic stations, recording ambient seismic noise for a period of six weeks between June and July 2016. ArrayA and ArrayB were installed in the south-eastern Cape-Karoo region, near the town of Jansenville and ArrayC was installed near the Cradock-Tarkastad region of South Africa. This thesis is made up of two separate studies. Firstly, the retrieval and coherency of Rayleigh surface waves extracted from the vertical component recordings. Novel cytotoxic agents developed with unique mechanisms of action have not been therapeutically suitable, since many of these compounds lack tumour selectivity. The adverse effects and the inefficacies of most chemotherapeutic therapies have motivated extensive investigations of alternatives. Numerous studies have demonstrated the advantages of using combination therapy with naturally-derived agents as an alternative, due to the higher therapeutic efficacy; with the added benefit of lower drug usage and reduction in drug-resistance development. An excellent example of a naturally derived and common dietary agent is curcumin, the active constituent of turmeric. Curcumin is known to modulate several signalling pathways in addition to displaying a diverse range of anti-tumour activities against a number of cancer cells. Several reports point to curcumin being beneficial if used in addition to chemotherapeutic drugs. In this study, synthetic aminonaphthoquinone derivatives (coded Rau 008, Rau 010, Rau 015 and Rau 018) were tested individually and in combination with curcumin for the potential as anti-breast cancer agents in different tumour cell lines. Notably, the aminonaphthoquinone class of compounds have shown potential as anti-cancer agents in various tumour cell lines. This study was thus aimed at screening the Rau compounds (and selected combinations with curcumin) for anti-cancer activity using a range of in vitro biological assays, and was not mechanistic in nature. The IC50 values of the individual and combined drugs were determined from dose-response curves using non-linear regression analyses. Synergistic, additive and antagonistic drug interactions were assessed using combination index and isobologram approaches. Based on these interactions, selected drug combinations were then further analysed for their potential anti-estrogenic, cytotoxic, anti-angiogenic and anti-metastatic effects against the ER(-) MDA-MB-231 and ER(+) MCF-7 breast cancer cell lines. The potential anti-cancer effects of the selected drug combinations on other estrogen-responsive models, such as the osteosarcoma (MG-63) and endometrial cancer (HEC-1A) cell lines, were also investigated. Overall, the findings of this study indicated that the sensitivity of the tumour cells to the various test compounds was significantly enhanced when combined with curcumin. For instance, in the presence of curcumin, Rau 008 or Rau 010 reduced the metastatic capability of ER(-) breast cancer cells significantly. The anti-proliferative effect of the Rau 015+curcumin combination was enhanced in ER(+) and ER(-) breast cancer, including oteosarcoma cells, while a combination of Rau 015 and curcumin induced a significant cytotoxic effect in ER(-) breast cancer and endometrial cancer-derived cells. A combination of Rau 018 and curcumin inhibited the proliferation of ER(+) breast cancer, and the combined effect was significantly more enhanced compared to individual treatment. A similar effect was noted in osteosarcoma-derived cells. Further, the Rau 018+curcumin combination exerted a marked cytotoxic and anti-angiogenic effect in ER(-) breast cancer, osteosarcoma and endometrial cancer in addition to inhibiting the attachment of ER(-) breast cancer cells to collagen. Notably, curcumin enhanced the anti-proliferative effect of Tamoxifen in breast cancer (MCF-7 and MDA-MB-231), osteosarcoma and endometrial cancer, and also induced a significant cytotoxic effect against ER(+) breast cancer and osteosarcoma, whilst also reducing the invasive potential of ER(-) breast cancer. A combination of 17β-estradiol and curcumin induced a marked anti-proliferative effect in ER(+) and ER(-) breast cancer, reduced the adhesion of ER(-) breast cancer to laminin, and elicited a significant cytotoxic and anti-angiogenic effect against endometrial cancer. Notably all the selected combinations (except Rau 018 and curcumin) inhibited bone mineralization. These findings point to the vital influence of curcumin in the responsiveness of ER(+) and ER(-) tumours towards the relative test agents. Further, among the Rau+curcumin combinations tested in this study, the anti-cancer activity of Rau 015+curcumin and Rau 018+curcumin appeared to be the most effective in the different cell lines tested. Nonetheless, it appears that a combination of 30 μM Rau 018 with 100 μM curcumin may be more promising as an anti-breast cancer agent against ER(+) and perhaps triple negative breast cancer, and warrants further investigation.

- Description: This research is based on reflections of various practitioners and their views on how public participation processes may or may not contribute to effective EIA decisions. It is therefore grounded in what is described as practitioner-based research. To study comparative development activities effectively, one must draw on many disciplines and construct a balanced understanding of historical and contemporary development processes. No single conventional disciplinary area of research is able to integrate the issues of public participation, EIA and decision-making in the context of the current development debate. The emergence of Trans-disciplinary areas of research allows for such integration. The Africa Earth Observatory Network (AEON) institute creates the space for this study to achieve an integrated response to the question of the effectiveness of public participation in EIA and decision-making. This further creates the opportunity to contribute and expand the growing body of knowledge and literature of public participation in the earth stewardship science discipline. Making use of triangulation, this study fulfils four major tasks: firstly, an assessment is made on the historical and theoretical importance, process and outcome of both EIA and public participation internationally and in South Africa. Secondly, three prominent case studies (i) the Coega IDZ, (ii) the Pondoland N2 Toll Road, and (iii) the anticipated Hydraulic Fracturing in the Karoo each focusses on concerns of an environmental, socio-economic, and political nature to assess if the public participation process has had influence, if any, on the final decisions for these projects to go-ahead. Thirdly, a survey was conducted to establish the views and perceptions of practitioners in the EIA and public participation domain on the effectiveness of public participation in EIA processes. Lastly, face-to-face interviews were conducted with various ‘pracademics’, i.e. consultants, practitioners, government, and non-governmental officials to establish their views on how public participation may or may not influence EIA decisions. Utilising the International Association for Public Participation’s (IAP2) participation spectrum as an evaluation tool, this research explores thirteen key criteria normally attributed to effective public participation. This provides a scale (inform, involve, consult, collaborate) to assess whether public participation in EIA in South Africa is least effective (inform) or most effective (collaborate). My research confirms that in South Africa an enabling environment to address impacts on our environment is emerging. One of the main challenges remains putting in place robust, clear and effective regulations, models or approaches that provide for effective public participation and decision-making in EIAs. My research also suggests that the legislation on which EIA is based cannot by itself guarantee the effectiveness of public participation processes. In practice, EIA is an institutional process of power division between different actors, and the practitioner or ‘pracademic’ has to play a more fundamental role to ensure effectiveness and fairness in the public participation process. Academic commentators should therefore call for new approaches that emphasise collaborative interaction between decision-makers and the public as well as deliberation amongst participants.

Ptdach2 2+Ptox2 2-). In another novel synthetic method for the same compound the ligand exchange reactions at various temperatures could be investigated and kinetic curves obtained served to illuminate the chemistry involved, indicating the role of small amounts of water in the essentially non-aqueous solvent systems dmf and isoamyl alcohol respectively. These allowed ligand exchange without resulting in hydrolyses even up to 85°C. The ionisation rate of divalent platinum halide complexes was determined for various amine ligands as well as N-S chelate ligands. A comparison of these could suggest why N-S complexes have poor anticancer action. Ionisation was not only studied for neutral molecular species but also for monocationic ones. Relationships could be found with stereochemical aspects of the chelates used. By investigating results of EV-CAD studies thermodynamic data could be obtained which indicated that bond strength decreases from chloro to iodo analogues although extent of ionisation in aqueous solution, i.e kinetic stability, is the reverse. Products formed by the reaction of NO2 gas with Platinum(II) compounds could be identified and separated which greatly contributed to the understanding of the chemistry involved in the formation of mononitro platinum(IV) complexes. Some of these proved to have exceptional anticancer properties. Studies of the interaction of thiol containing biomolecules were performed as a function of time. The results contributed to the understanding of the action of the anticancer agents.

Database and guide for Lesotho wool and mohair production and quality

- Description: Lesotho produces significant quantities of Merino apparel wool and mohair, both of a quality that allows them to compete on the global market and to make a significant contribution to the economy of the country. Nevertheless, very little production and quality data and trends of these fibres have been documented. This is a serious disadvantage in terms of international trading and benchmarking as well as attempts to improve the production and quality of Lesotho wool and mohair. In the light of the aforementioned, the available production and quality data of the past 10 years have been captured and analysed for trends and also benchmarking, where considered applicable. The main focus is on fibre diameter (fineness), staple length, and yield since they largely determine fibre quality, application and price. Some quality related tests were undertaken to fill certain important gaps in the available data. In addition, prickle and medullation were evaluated on representative wool and mohair samples, respectively, because they represent important quality measures for apparel wool and mohair, respectively. It was found that Lesotho wool and mohair are of a fairly good and internationally competitive quality, with the wool having an average fibre diameter (MFD) of ≈20μm, an average staple length of about 64mm, an average VM level of about 4%, an average yield of about 57%, and the annual production being about 4 million kilograms greasy. The average staple length of the mohair was about 140mm, average MFD about 29μm and the average medullation, which unless otherwise specified, refers to the objectionable medullated fibres (kemp type) including the flat medullated fibre, relatively high at about 5.7%. The latter is certainly an area of concern which needs attention and improvement. It was found that the prickle level (Comfort Factor), of some of the wools tested was of such a level as to make the wool suitable for wearing against the skin. With respect to mohair, there is considerable scope to substantially reduce the level of objectionable medullated (kemp style) fibre level through the appropriate breeding interventions. Production of both wool and mohair has increased slightly over the ten years covered by this study. It also became apparent that Lesotho wool and mohair quality and production are greatly influenced by the farming practices and climatic conditions. Greater adoption of the merino sheep breed, sheds/barns and sheep coats are suggested as ways to reduce mortality rate (due to extremely cold temperatures), improve quality and increase yield and production. Some farming practices such as the lack of barns, supplementary feeding and veterinary care present constraints in terms of production of both wool and mohair. The districts in the Highlands region had the highest production of mostly wool, this being ascribed to better pastures, climatic and other conditions conducive to wool and mohair production. Both wool and mohair are considered to have potential for improved quality and production, which could be affected by appropriate interventions by the Small Agricultural and Development Project (SADP) and others through National Wool and Mohair Growers Association (NWMGA). Nevertheless, since the wool and mohair growers (farmers) do not form part of these and other interventions, they do not readily adopt the various strategies and decisions and do not receive the associated benefits immediately. It is therefore advised that local farmers, relevant educators and researchers be represented in policy and other decision making forums. In this way, educational campaigns will be demand driven with greater chance of adoption and success.

- Description: Lesotho produces significant quantities of Merino apparel wool and mohair, both of a quality that allows them to compete on the global market and to make a significant contribution to the economy of the country. Nevertheless, very little production and quality data and trends of these fibres have been documented. This is a serious disadvantage in terms of international trading and benchmarking as well as attempts to improve the production and quality of Lesotho wool and mohair. In the light of the aforementioned, the available production and quality data of the past 10 years have been captured and analysed for trends and also benchmarking, where considered applicable. The main focus is on fibre diameter (fineness), staple length, and yield since they largely determine fibre quality, application and price. Some quality related tests were undertaken to fill certain important gaps in the available data. In addition, prickle and medullation were evaluated on representative wool and mohair samples, respectively, because they represent important quality measures for apparel wool and mohair, respectively. It was found that Lesotho wool and mohair are of a fairly good and internationally competitive quality, with the wool having an average fibre diameter (MFD) of ≈20μm, an average staple length of about 64mm, an average VM level of about 4%, an average yield of about 57%, and the annual production being about 4 million kilograms greasy. The average staple length of the mohair was about 140mm, average MFD about 29μm and the average medullation, which unless otherwise specified, refers to the objectionable medullated fibres (kemp type) including the flat medullated fibre, relatively high at about 5.7%. The latter is certainly an area of concern which needs attention and improvement. It was found that the prickle level (Comfort Factor), of some of the wools tested was of such a level as to make the wool suitable for wearing against the skin. With respect to mohair, there is considerable scope to substantially reduce the level of objectionable medullated (kemp style) fibre level through the appropriate breeding interventions. There is, however, a need for a plant dimension in road ecology, especially in those areas where plant biodiversity, and threats to this biodiversity, are concentrated. The research presented in this thesis aimed to further our understanding of road effects on the plant component of fynbos ecosystems in the megadiverse Cape Floristic Region (CFR). The exceptional plant diversity of the Cape and the presence of an extensive road network in the region present an ideal system for studying the interactions of roads and plants. Road verges have been emphasised as valuable habitats for plants. However, to assess the viability of road verges as habitats for fynbos plants, we first need to understand how these communities are influenced by the disturbances that exist in these environments. To this end, I investigated fynbos plant communities along roads and showed that road-mediated changes in the soil environment can bring about subtle changes in their composition while key components of the communities persist in road verges. Even though fynbos community patterns remain intact in road verges, the ecological processes that facilitate the persistence of plant species may be disrupted in these environments. As such, I studied the pollination of an ornithophilous plant in road verges and fynbos fragments of a highly transformed landscape. Here, landscape context was an important determining factor in the visitation rate of birds to plants in verges, with those occurring next to transformed land experiencing reduced visitation rates. Furthermore, I studied the pollination and fecundity of a highly threatened entomophilous plant and showed that, while there were no impacts on reproductive output of plants, road effects bolstered the reproductive growth of plants near the road. ii Finally, I implement the road-effect distances identified in the above studies to estimate the proportion of extant fynbos vegetation that may be impacted by roads. While the proportion of vegetation affected by roads is small, a substantial proportion is in close proximity to roads, potentially rendering large areas vulnerable to road-mediated human impacts. I further highlight the fact that several plant species of conservation concern in the Cape are affected by roads, and that many of these constitute a significant proportion of South Africa’s threatened flora. A case study from the N2 national road in the south-eastern Cape, where vegetation in road verges is in a better condition than that of adjacent, demonstrates the potential conservation value of road verges for fynbos vegetation in the region.

A technical and economic evaluation of a passive underground mine-water purification system (PUMPS): a geothermally powered geo-engineering system designed for in-situ bio-remediation of acid mine water

- Description: PUMPS mimics natural geothermal vents as a conceptual model designed for the remediation of acid mine water (AMW) in voids of abandoned gold mines of the Witwatersrand Basin in South Africa. In this system, a reaction chamber containing Desulfotomaculum kuznetsovii sulfate reducing bacteria will be set at the bottom of a 3-4 km deep mine that will be flooded. A geothermal system with at least one (1) doublet will be drilled from the bottom of the mine to the depth of 8km, where the temperatures are sufficient for geothermal energy harvesting. AMW, used as a geothermal fluid, will be pumped down the injection well and circulate through hot rock. The hot water is then used to generate electricity and then channelled into the reaction chamber to undergo bio-remediation. Following treatment, the water flows back into the mine voids where it will improve the quality of untreated AMW through dilution. Eventually, the mine will be flooded with clean water that can be stored underground and/or pumped up to surface for social and ecosystem services. Following an introduction and proof of concept for the PUMPS, the research builds further on the technical and economic evaluation of the PUMPS in order to assert its viability and sustainability. The technical viability includes testing the ability for Desulfotomaculum kuznetsovii to survive in high pressure condition; quantifying the amount of energy that can be drawn from the geothermal reservoir; determining the placement and scheme of the geothermal wells; and, finally, developing a robust economic model of the system. Experiments show that Desulfotomaculum Kuznetsovii can tolerate high pressure conditions in of at least 100bar at their ideal sulfate reducing temperature of 63°C. Geochemical modelling shows that AMW can be used effectively as a geothermal fluid for PUMPS. To achieve highest efficiency and minimal fluid loss, the geothermal wells should be placed along the SSE-NNW direction, based on the known stress field across the Witwatersrand Basin. With a flow rate of 30l/s the energy drawn from the geothermal reservoir is sufficient to drive PUMPS and the surplus energy is determined by the volume of AMW treated per day. All results indicate that the PUMPS is technically and economically viable. The economic model shows that the value and viability of the PUMPS is best reflected with a comprehensive inclusion of potential revenue (for example from chemical solution mining of deep seated gold) and financial/environmental incentives.

Comprehensive characterization of the antidiabetic potential of selected plants and macrofungi from Africa using an in vitro target-directed screening platform and cellomics

- Description: Several synthetic antidiabetic drugs have been developed to date, however, most are accompanied by adverse side-effects while remaining expensive and largely inaccessible to the vast majority of those who need it. To provide enough scientific evidence to support the inclusion of more affordable African antidiabetic medicinal plants and macrofungi into healthcare programs, this study sought out to develop a comprehensive in vitro antidiabetic target-directed screening platform incorporating high content screening and analysis/ cellomics. To test the success of this model, the potential antidiabetic mechanisms of five plants (Aspalathus linearis, Brachylaena discolor, Carpobrotus deliciosus, Sutherlandia frutescens and Tarchonanthus camphoratus) and two macrofungal species (Ganoderma lucidum and Hericium erinaceus) were explored. The screening model consisted of approximately 22 assays exploring the antidiabetic effects of selected aqueous and ethanolic extracts in five well-characterised antidiabetic targets: the intestine, liver, skeletal muscle, adipose tissue/ obesity and pancreatic β-cells. These targets were further categorised and scored under three mechanistic classes/ therapeutic targets (postprandial hyperglycaemia; insulin resistance and inflammation; pancreatic β-cell function) to elucidate their potential mechanisms of action and select appropriate animal models for future studies. Almost any normal or diabetic rodent model would be suitable to explore the antidiabetic potential of extracts such as A. linearis, B. discolor ethanol, C. deliciosus ethanol or T. camphoratus which obtained high cumulative scores under postprandial hyperglycaemia while high fat diet and genetic models of obesity appear more suited towards extracts such as H. erinaceus aqueous that obtained their highest cumulative score under insulin resistance. In general, a combination of rodent models ranging from non-obese models to models of obesity and β-cell destruction presenting symptoms from all three mechanistic classes should be considered due to the pleiotropic nature of the tested extracts, however, establishing appropriate experimental designs is crucial. To demonstrate the versatility of the screening platform and emphasise the importance of in vitro screening pertaining to diabetic complications, a more detailed biochemical investigation into the potential therapeutic benefits of A. linearis in the treatment of diabetic wounds was conducted. Several properties supporting the therapeutic potential of rooibos were highlighted with the green and fermented extracts presenting distinctly different characteristics. The pro-inflammatory nature of fermented rooibos may have therapeutic value for wounds characterised with a delayed initial inflammatory phase, such as early diabetic wounds while the green extract appears more suited to wounds burdened with excessive inflammation as it attenuated COX-2 levels and effectively protected fibroblasts against oxidative stress. To date, this appears to be the most comprehensive antidiabetic screening platform in existence and consequently provides the only feasible solution that will enable natural product antidiabetic research to progress to the point where natural products can be commercialised and incorporated into meaningful healthcare programs. Future research should be focussed towards further expanding this model by incorporating additional targets, more sophisticated cell culturing techniques, multiplexed high content screening assays and carrying out combination treatments that explore the antidiabetic effects of two or more crude extracts to establish whether they are capable of acting in a synergistic manner.

An assessment of the social ecological system of Mabira forest and environs: implications for vulnerability to climate variability

- Description: Tropical forests are attracting attention in climate change debates considering the fundamental role they play as carbon sinks. Mabira Central Forest Reserve (CFR) found in the Lake Victoria basin region and of significance in achieving the Sustainable Development Goals. It has however undergone drastic degradation since the 1970s and its social economic ecological and environmental functions are at risk. The present study assessed the Social Ecological System (SES) of Mabira CFR and environs as well as the implications for vulnerability to climate variability. It focused on; i) assessment of the spatial temporal changes of Mabira CFR; to which Landsat images of 1975, 1986, 2000 and 2015 were used in combination with ground truthing for a successful classification of land use/cover types. ii) Determine the implications of land use/cover types and forest management options for above ground biomass (AGB) and soil organic carbon (SOC); here Velle, (1995) allometric equation was adopted for the AGB while the Walkley and Black (1934) was used to determine soil organic carbon iii) Determine the implications of land use cover change for livelihood and vulnerability to climate change impacts; to this the Social Ecological systems framework by Ostrom (2009) was adopted to understand the vulnerability implications for land use cover change (LUCC) and Climate variability in a forest based community. The study reveals that heavy deforestation and degradation continue to pose threats to Mabira CFR and environs. Major drivers of LUCC are: population expansion, loss of soil fertility, poor agricultural practices, establishment of roadside markets, industrialization and unclear forestry boundary. Findings indicate that although forest comprised the biggest coverage between 1975 and 2000, it has faced massive reduction between 2000 and 2015. Major forest conversions have been to subsistence farming (5.94%). The implications of LUCC are reflected in; the exposure of soils to degradation as seen from deforestation; soil erosion; carbon sequestration and climate variability constraining livelihoods. AGB and carbon was found highest in the strict nature reserve where on average the parameters noted above were found to be bigger. The recreation buffer had less carbon compared to the strict nature reserve; however the production zone, which had scattered trees with smaller parameters, registered the lowest average total AGB and carbon. Results indicate a perfectly strong relationship between the AGB and carbon variables. Results also indicate that approximately 6,771092.344 tonnes are sunk in the strict nature reserve. SOC analyzed based on the soil carbon percentage content. In terms of soil depth, the 0-10cm and 10-20cm soil layers had relatively similar variations of least square means for carbon than at 20-30cm soil layer. The highest SOC was observed in the soil surface of 0-10 cm depth, with the highest mean of 2.78 % across all the management zones. The amount of soil organic matter also varied with land use /cover types. Study findings reveal that LUCC and land management have a direct influence on soil organic carbon accumulation. Results indicate that LUCC implications for AGB and SOC as well as climate variability lead to livelihood vulnerability. Climate hazards experienced have affected SES, temperature variation range between 1oC -2oC. These variations are likely to increase vulnerability. This study recommends mapping of LUCC to identify priority areas for intervention. Priority should be given to reducing deforestation and restore degraded areas. Future research should focus on building adaptive capacity for forest based communities to mitigate climate change, improving forest tenure as a strategy for climate change impacts, integrating climate change into forest policy reviews.

The present study assessed the Social Ecological System (SES) of Mabira CFR and environs as well as the implications for vulnerability to climate variability. It focused on; i) assessment of the spatial temporal changes of Mabira CFR; to which Landsat images of 1975, 1986, 2000 and 2015 were used in combination with ground truthing for a successful classification of land use/cover types. ii) Determine the implications of land use/cover types and forest management options for above ground biomass (AGB) and soil organic carbon (SOC); here Velle, (1995) allometric equation was adopted for the AGB while the Walkley and Black (1934) was used to determine soil organic carbon iii) Determine the implications of land use cover change for livelihood and vulnerability to climate change impacts; to this the Social Ecological systems framework by Ostrom (2009) was adopted to understand the vulnerability implications for land use cover change (LUCC) and Climate variability in a forest based community. The study reveals that heavy deforestation and degradation continue to pose threats to Mabira CFR and environs. Major drivers of LUCC are: population expansion, loss of soil fertility, poor agricultural practices, establishment of roadside markets, industrialization and unclear forestry boundary. Findings indicate that although forest comprised the biggest coverage between 1975 and 2000, it has faced massive reduction between 2000 and 2015. Major forest conversions have been to subsistence farming (5.94%). The implications of LUCC are reflected in; the exposure of soils to degradation as seen from deforestation; soil erosion; carbon sequestration and climate variability constraining livelihoods. AGB and carbon was found highest in the strict nature reserve where on average the parameters noted above were found to be bigger. The recreation buffer had less carbon compared to the strict nature reserve; however the production zone, which had scattered trees with smaller parameters, registered the lowest average total AGB and carbon. Results indicate a perfectly strong relationship between the AGB and carbon variables. Results also indicate that approximately 6,771092.344 tonnes are sunk in the strict nature reserve. SOC analyzed based on the soil carbon percentage content. In terms of soil depth, the 0-10cm and 10-20cm soil layers had relatively similar variations of least square means for carbon than at 20-30cm soil layer. The highest SOC was observed in the soil surface of 0-10 cm depth, with the highest mean of 2.78 % across all the management zones. The amount of soil organic matter also varied with land use /cover types. Study findings reveal that LUCC and land management have a direct influence on soil organic carbon accumulation. Results indicate that LUCC implications for AGB and SOC as well as climate variability lead to livelihood vulnerability. Climate hazards experienced have affected SES, temperature variation range between 1oC -2oC. These variations are likely to increase vulnerability. This study recommends mapping of LUCC to identify priority areas for intervention. Priority should be given to reducing deforestation and restore degraded areas. Future research should focus on building adaptive capacity for forest based communities to mitigate climate change, improving forest tenure as a strategy for climate change impacts, integrating climate change into forest policy reviews.

Creating Citizen-Science for Groundwater Monitoring prior to potential Shale Gas Development in Cradock (South-Eastern Karoo, South Africa)

- Description: Citizen Science is an integral tool for community engagement in scientific project design, implementation, data interpretation and reporting, in the quest to promote local capacity development as well as scientific knowledge. Based on the relevance of public engagement in aspects of the natural environment and associated scientific issues associated with the risks and opportunities of potential Shale Gas Development (SGD), a Citizen-Science (CS) study was designed in the Eastern Karoo region of South Africa linked to a Shale Gas Baseline Study initiated by the Africa Earth Observatory Network (AEON). As the pilot area of the baseline study, the town of Cradock was chosen to be the study area for this research; and CS was used to facilitate the identification of the Cradock community’s abilities to monitor the effects of potential SGD in this region. The development of new knowledge, skills and support, as well as a deeper understanding of the community’s role in Citizen Science studies, was also facilitated by this process. This study demonstrates the feasibility of integrating Citizen Science into existing community structures in Africa, thereby encouraging community engagement in the developmental programmes through participatory methods. Using a collaborative Action-Research Approach, consistent community roundtable meetings and key informant interviews served as invaluable platforms for the establishment of a 7-member community working group, that played the liaison role between the researcher and the Cradock community in the CS implementation process. Coupled with this was the recruitment and the training of eight young citizens in conducting a hydro-census and groundwater sampling for six boreholes within the identified Cradock commonage farms, as well as two boreholes on the farm of an identified emergent farmer. The engagements between the researcher, AEON scientists, the community working group and the community at large, enabled the successful implementation of CS training in two groundwater monitoring aspects and the testing of eleven water quality parameters. The training process was combined with the design and the development of a customised ‘Xoras’ Online Application, which was used to capture and share the hydro-census data recorded. Experiential learning in hydro-census and groundwater sampling resulted subsequently in an increased understanding and awareness of these aspects (Figure A). Even if SGD does not materialise in the South-Eastern Karoo, CS training will enable communities in the Shale-Gas Development precincts to participate in local decision-making forums on ground water, health, or on any related regional development projects. It is anticipated that the adoption of CS will ix promote future community engagements, especially about water across this water-scarce region, allowing for greater community-voice representation in resource-policy decisions related to potential Shale-Gas and related natural resource industries in the Karoo.
