- Description: The main aim of this research is to examine the hip hop scene in Grahamstown, Eastern Cape and whether hip hop can serve as a significant platform to discuss women’s lived experiences. This study also places focus on how female rappers construct their rap personas, as well as how they are perceived by their male counterparts. In order to understand the multifaceted viewpoints, it was necessary to interview both male and female hip hop artists. Eight interviews were conducted with eight young black rappers who are actively involved in the Grahamstown hip hop scene. It was found that hip hop, like any other art form, is a significant platform for women to express themselves, however gender constraints limit who is willing to listen to and promote their music. Male rappers advocate for women to talk about their stories, yet are more unlikely to listen because it does not relate to their struggles. In addition, there seem to be four specific tropes that female rappers choose to construct their identities from. Female rappers tend to create their personas around: Queen Mother, Fly Sista, Bitch with Attitude, and Lesbian. However, these categories are fluid and it was found that women navigate these categories depending on their audience and message they want to convey at a particular moment. Lastly, there is a split between Grahamstown West (Rhodes University) and Grahamstown East (township). University students are unlikely to perform in the township, and township residents rarely perform at organised events in Grahamstown West. In addition, Rhodes University students are more likely to feature on the university run radio station, rather than Radio Grahamstown, the local community radio.

- Description: The main objective of this thesis is to investigate how conscious hip hop culture is used to reconstruct and influence urban youth identities in South Africa, in the case of Fingo Village, Makhanda. In addition, it aims to understand how the South African conscious hip hop scene, appropriated from the West, can be used to shape the identities of the urban youth, instead of the materialistic commercial/mainstream gangsta rap that is currently dominating popular culture and media today. Kellner’s (1995: 10) multicultural critical approach is appropriate for this thesis, as it “provides a critical multiculturalist politics and media pedagogy that aims to make people sensitive to the relations of power and domination which are encoded in cultural texts, such as those of television and film, or new technologies and media such as Internet, and social networking”. Moreover, in the words of Cvetkovich and Kellner (cited in Dolby, 2010:11), this dissertation “investigates the increasing influence of global popular culture and its possibility to equip the urban youth with new sources of identification”, as well as opportunities for social change. In Makhanda, rappers, community activists, B-Boy crews, graffiti artists, spoken-word poets, photographers, and journalists all assembled in Fingo Village for the social event Around Hip Hop, at the multipurpose Fingo square, between 2011 and 2019. Today, Around Hip Hop is an arts based organization that produces hip hop mixtapes, short documentaries, and educational events aimed at creating a cross-cultural exchange and increasing awareness of South African politics. Around Hip Hop has hosted various events, dialogues, and performances where hip hop is used as a cultural expression, which continuously creates spaces for the urban youth to rethink and reconstruct their identities; not only as artists, but as audience members as well. Events, such as The Return of the Cypher, Intyatyamo Elityeni, and Business beyond Fingo Festival all form part of Around Hip Hop. As such, music has been closely related to youth culture, especially as it pertains to youth rehabilitation and identity formation. Studies (e.g., Duffy and Fuller, 2000; Miranda et al, 2013; Strayhorn, 2011; Tomasello, 2016) highlight the value of utilising music in youth rehabilitation, and acknowledge the intrinsic value of music to facilitate psychological and social change for individuals who would not otherwise benefit from psycho-education, or other didactic modes of youth rehabilitation. That said, Tyson (2005) developed the RAP music scale to psychometrically validate the underlying constructs of youth’ attitudes and perception of rap music in the United States of America, in order to use the instrument for music-based youth rehabilitation programs. The development of the RAP music scale informed the creation and validation of the Kwaito-music Attitude and Perception (KMP) Scale, the subject of my Masters thesis. Following suggestions from Morrison (2014), my study sought to psychometrically validate the properties of the Kwaito-music Attitude and Perception (KMP) Scale. The KMP was found to be a reliable measure of kwaito music attitudes and perception amongst a sample of South African youth who listen to this genre of music. The internal consistency of the questionnaire revealed a Cronbach’s α (Alpha) value of 0.820. Limitations of the project including reduced factor extraction and sample size constrains are discussed. It is envisioned that results from this study will find resonance with youth-community programs, and rehabilitation projects utilising music as a medium of healing amongst South African youth.

The investigation focuses specifically on the work of two visual artists: Zimbabwean born, Johannesburg-based Kudzanai Chiurai, and Cape Town born, Berlin-based Robin Rhode. The introduction familiarises the reader with the two artists and briefly outlines their histories and methods, as well as giving a short history of the development of hip-hop as a subculture from its beginnings in 1970s New York. The first chapter follows this brief introduction to outline some of the parallels, especially concerning race relations, between 1970s America and post-apartheid contemporary South Africa. This comparison aims to highlight similarities that gave rise to the hip-hop phenomenon and which also place South Africa in a prime position to welcome such influences. The second half of the chapter explores how migration theory and issues of diaspora have not only influenced the development of hip-hop, but have also become points of focus for both artists, who are in fact disporans themselves. The second chapter explores ‘ground level’ concerns of everyday life in the city. Development of a low-cost bioprinting system for the fabrication of cell-laden sodium alginate hydrogels

- Description: Bioprinting is a rapidly expanding technology with the ability to fabricate in vitro 3D tissues in a layer-by-layer manner to ultimately produce a living tissue which physiologically resembles native in vivo tissue functionality. Unfortunately, large costs associated with commercially available bioprinters severely limit the amount of people/research groups with access to the technology. Here, we investigated the potential for modifying a commercially available RepRap Prusa iteration 3 (i3) three-dimensional (3D) printer, by replacing the traditional plastic-based print-head with various open-source syringe-housed microextrusion print-head units, such that deposition of composite bioinks consisting of cells, biopolymer scaffolds and/or biomolecules may be achieved at a relatively low cost. Using adipose-derived human mesenchymal stromal stem cells (ad-HMSC) induced for adipogenic differentiation, as well as human umbilical vein endothelial cells (HUVEC), the potential for fabricating vascularised adipose tissue was investigated. The non-toxic, inexpensive algal polysaccharide, sodium alginate, was used to test the printability of the system, as well as for investigating the functionality unmodified sodium alginate has for use as a potential bioink in adipose tissue engineering. Cell viability assays, namely WST-1 and fluorescein diacetate (FDA)/propidium iodide (PI) live/dead cell staining, revealed that ad-HMSC were viable after 7 days of culture. However, viability of HUVEC encapsulated hydrogels revealed significantly lower cell viability. Live/dead cell staining revealed that the modified printing system was able to print ad-HMSC/HUVEC co-cocultures with a large degree of cell viability after 1 day of culture. However, after 7 days of culture, the majority of cells were revealed to be dead. Furthermore, due to the lack of mechanical integrity possessed by alginate in a liquid-like state, printing sodium alginate hydrogels in air consistently resulted in deformation of printed constructs. The newly developed 3D printing technique termed freeform reversible embedding of suspended hydrogels (FRESH) was therefore investigated as a means for achieving 3D spatial control of printed hydrogels using the modified system. Printing cell-free sodium alginate hydrogels within gelatin sacrificial support baths allowed for fabricating constructs in a spatially defined manner. However, overprinting and swelling of alginate hydrogels negatively affected the overall printing accuracy. The present study aimed to pave the way for further system modifications and refinements, such that the ultimate goal of low-cost bioprinting may be achieved. Further optimisation of printing parameters, hydrogel characteristics and sterilisation techniques may allow for fabricating viable, physiologically relevant tissues using the modified system developed.

Here, previously determined lipid normalisation equations and trophic fractionation values were re-evaluated using freshwater fish species from three rivers in the Upper Zambezian floodplain ecoregion in southern Africa. The parameters commonly used in lipid normalisation equations were not correct for the 18 model species (new D and I parameters were estimated as D = 4.46‰ [95% CI: 2.62, 4.85] and constant I = 0 [95% CI: 0, 0.17]). We suggest that future isotopic analyses on freshwater fishes use our new values if the species under consideration do not have a high lipid content in their white muscle tissue. Nitrogen fractionation values varied between species and river basin; however, the average value closely matched that calculated in previous studies on other species (δ15N fractionation factor of 3.37 ± 1.30 ‰). Here we have highlighted the need to treat stable isotope data correctly in food web studies to avoid misinterpretation of the data.

Investigating the role of Hsp90 and LRP1 in FN matrix dynamics

- Description: Fibronectin (FN), a matrix protein responsible for regulating processes including migration and differentiation, is secreted as a soluble dimer which is assembled into an insoluble extracellular matrix. The dynamics of FN matrix assembly and degradation play a large role in cell migration and invasion contributing to the metastatic potential of cancer cells. Previous studies from our group have shown the direct binding of Hsp90 and FN in vitro and that inhibition of Hsp90 with novobiocin (NOV) caused internalisation of the FN matrix. However, the receptor mediating this internalisation is currently unknown. Low density lipoprotein 1 (LRP1) is a likely candidate as it is a ubiquitous receptor responsible for regulating internalisation of diverse ligands and is known to bind both Hsp90 and FN. We used wild type and knockout LRP1 cell lines to study the endocytosis of FN via this receptor. Here, we demonstrate that LRP1-deficient cells accumulated greatly increased levels of FN and were found to be less sensitive to pharmacological inhibition of Hsp90 by NOV. LRP1-expressing MEF-1 and Hs578T breast cancer cells experienced an increase in total FN in response to NOV, at concentrations below the EC50 value, followed by a dose-dependent loss of FN. We attributed greater FN levels to a loss of extracellular FN matrix coupled with increased internalisation of FN. Cell-surface biotinylation and DOC assays showed that loss of extracellular FN was specific to LRP1-expressing MEF-1 cells. Furthermore, we demonstrate that the loss of extracellular FN is not affected by changes in FN mRNA levels as determined by qRT-PCR, and that treatment with NOV resulted in the accelerated degradation of FN in the presence of cycloheximide. Immunoprecipitation studies reveal a putative complex exists between FN, Hsp90 and LRP1 in both cancer and non-cancer cells which is not perturbed by NOV. Western analyses revealed increased proteolytic processing of LRP1 in response to NOV which we proposed, based on literature, to modulate signalling pathways as a potential mechanism for regulating FN turnover. Moreover, using wound healing assays we identified increased migration to be one of the consequences associated with loss of extracellular FN by Hsp90 inhibition but only in cells containing LRP1. In summary, this study provides new insights into the Hsp90-LRP1 mediated loss of FN matrix and also reveals for the first time the functional consequence related to FN turnover by NOV was an increase in migration in LRP1-expressing cells.

- Description: Rural communities have limited access to health information which is made available on the internet. This is due to poor infrastructure (i.e., lack of clinics or Internet access) and that gives them problems in accessing information within the domain of health. The availability of Information and Communication Technologies (ICTs) in a rural community can provide the community with a number of beneficial solutions to their problems as they maximize the potential of knowledge sharing and delivery. This research seeks to make use of ICTs deployed in the community of Dwesa, in order to contribute to improving the health standards of the community. It seeks to accomplish this by carrying out an investigation and literature review with the aim of understanding health knowledge sharing dynamics in the context of marginalized communities. The knowledge acquired will then be used in the development and implementation of a semantic web-based e-Health portal as part of the Siyakhula Living Lab (SLL) project. This portal will share and deliver western medical knowledge, traditional knowledge and indigenous knowledge. This research seeks to make use of a combination of Free and/or Open Sources Software in developing the portal to make it affordable to the community.

The availability of Information and Communication Technologies (ICTs) in a rural community can provide the community with a number of beneficial solutions to their problems as they maximize the potential of knowledge sharing and delivery. This research seeks to make use of ICTs deployed in the community of Dwesa, in order to contribute to improving the health standards of the community. It seeks to accomplish this by carrying out an investigation and literature review with the aim of understanding health knowledge sharing dynamics in the context of marginalized communities. The knowledge acquired will then be used in the development and implementation of a semantic web-based e-Health portal as part of the Siyakhula Living Lab (SLL) project. This portal will share and deliver western medical knowledge, traditional knowledge and indigenous knowledge. This research seeks to make use of a combination of Free and/or Open Sources Software in developing the portal to make it affordable to the community.

In the modified binders EVA offers plastomer properties whereas the crumb rubber confers elasticity to the bitumen. 13.75% crumb rubber and 2.5% EVA loadings based on aging indices gave the best optimized mixture. The thermorheological behaviour of the best optimized mixture was compared to industrial grade EVA (AP-1) and crumb rubber (AR-1) modified bitumen. Improving aging behaviour of the bitumen was evaluated using four antioxidants: carbon black, hydrated lime, Irganox 1010 and Irgafos 168. The least aging indices at 58˚C and 64˚C were obtained from a 1:1:1 Irganox 1010/hydrated lime/carbon black mixture. SARA (saturates, aromatics, resins and asphaltene) analysis gave the chemical background for application of antioxidants to reduce the propensity of short term aging. To further improve the EVA/crumb rubber optimized mixture for workability FT wax (Sasobit®) was assayed at loadings from 0-2.5%. This was done to match the EVA content and maintain the original maximum loading of 2.5% in the optimized mixture. The phase change abilities of FT wax owing to the high latent heat enabled co-crystallization with the EVA with the elastomeric backbone of crumb rubber acting as support material. The energy-sustainability nexus was found to have worked at less than 1% loading of FT wax in the EVA/crumb rubber modified bitumen. Response surface methodology approach to all the sections of work was used to come up with the optimized mixtures based on rheological parameters complex shear modulus, phase angle and rutting parameter at test temperature 64˚C. The interaction chemistry of bitumen, crumb rubber, EVA and FT wax was found to be first and second order in all cases involving individual contributions and co-interaction amongst the factors. Project costing for each section of work (optimizing section, antioxidant section and FT wax section) was conducted involving the raw materials used, equipment used, labour involved and other costs incurred for all the project work.

Analysis of the interaction of Hsp90 with the extracellular matrix protein fibronectin (FN)

- Description: Mounting evidence suggests that Hsp90 is present and functionally active in the extracellular space. The biological function of extracellular Hsp90 (eHsp90) remains relatively uncharacterized compared to that of intracellular Hsp90. eHsp90 has been shown to interact with a finite number of extracellular proteins, however, despite the identification of eHsp90 interacting proteins, the function of eHsp90 in these complexes is unknown. Several reports suggest a role for eHsp90α in cell migration and invasion. Reported targets for eHsp90 stimulated cell migration include MMPs, LRP-1, tyrosine kinase receptors and possible others unidentified. Limited studies report a role for eHsp90β. Recently, Hsp90α and Hsp90β were isolated in a complex containing fibronectin (FN) on the surface of MDA-MB-231 breast cancer cells. Herein, we report direct binding of Hsp90α and Hsp90β to FN using a solid phase binding assay and surface plasmon resonance (SPR) spectroscopy. SPR spectroscopy showed that Hsp90β bound the 70 kDa amino-terminal fragment of FN (FN70), but that binding of FN to Hsp90β was not limited to FN70. Confocal microscopy showed regions of colocalization of Hsp90 with extracellular FN matrix fibrils in Hs578T breast cancer cell lines. Treatment of Hs578T breast cancer cells with novobiocin (an Hsp90 inhibitor) and an LRP-1 blocking antibody resulted in a loss of FN matrix and FN endocytosis (novobiocin treated). Addition of exogenous Hsp90β was able to recover such effect after both treatments. FN was shown to colocalize with intracellular LRP-1 in novobiocin treated Hs578T cells. Immunoprecipitation of an LRP-1 containing complex showed the presence of Hsp90 and 70 and 120+ kDa FN fragments. Treatment of Hs578T cells with novobiocin increased the level of FN120+ bound in LRP-1 immunoprecipitate. Exogenous Hsp90β decreased the level of low and high molecular weight FN fragments in a complex with LRP-1, despite the fact that higher levels of lower molecular weight FN fragments were detected in this cell lysate compared to the other treatments. We report FN as a novel interacting protein of eHsp90. Taken together, we provide evidence for a direct role of eHsp90β in FN matrix remodeling. We suggest that Hsp90 plays a direct role in FN matrix dynamics through interaction with FN and LRP-1. The identification of FN as a novel interacting protein of eHsp90 suggests a role for Hsp90 in FN matrix remodeling, which is important for a number of fundamental cellular processes including cell migration and metastasis.

A problematisation of Afrikaner identity subject positions as found in the coverage of contemporary Afrikaans music in HUISGENOOT magazine discourse

- Description: Through the lens of Foucauldian genealogy, this thesis problematises the subject positions towards Afrikaner identity reflected in contemporary Afrikaans songs covered by Huisgenoot magazine. Such a genealogical exploration thematises the games of truth underpinning the formation of Huisgenoot, highlights its role in solidifying white Afrikaner Nationalist discourse of apartheid, observes its more populist, profit-oriented approach from the 1970s onwards and considers the residual echoing of its former discursive orientation. The song analysis of selected Afrikaans songs/music videos covered in Huisgenoot is guided by the semiotics of Nicholas Cook, such that it frames the analytical question of how such music is driven by conformance, contest and complementation between multi-media elements in ways that make meaning possible. Accordingly, this methodology resonates with the non-essentialising focus of Foucauldian problematisation, premised on critical resistance that makes new responses simultaneously possible. The self-reflexive autoethnographical approach employed, entailing the author’s dialogical engagement with her own ‘Afrikaner self’, similarly facilitates the dynamic interplay between multiplicities. Correlatively, on the one hand, more ‘closed’ and more ‘open’ Afrikaner identity positions reflected within the songs are analysed, as well as seemingly ‘reconciliatory’ / collaborative’ musical manifestations which ultimately revert back to more ‘closed’ identity constructs. But on the other hand, observation of adherence to a ‘white-black’ dichotomy within songs that manifest more ‘closed’ identity positions also leads to consideration of the parallels between the goals of white Afrikaner Nationalism and the current Decolonisation narrative, which is informed by Black Consciousness thought and generated by the ‘Colonisation of a Special Kind’ ANC policy. Both such Decolonisation rhetoric and the countering rhetoric of the Neo-Afrikaner Protest Movement (NAP) are thus identified as metanarratives, underpinned by a ‘white-black’ dichotomy. In this regard, while Huisgenoot’s predisposition towards songs manifesting more ‘closed’ Afrikaner identity positions is considered in terms of its aforementioned profit-oriented endeavour to appeal to its ostensible primary readership, an encouraging increase in coverage of coloured artists is both noted and occasionally seen to overlap with its coverage of songs manifesting more ‘open’ Afrikaner identity positions. Among the latter one may – again in Foucauldian terms – differentiate between songs focussed on an engagement in cross-cultural relations of power, and songs intent on shattering states of domination of power blocks, often through shocking juxtaposition of disparate elements. These more violently dismantling endeavours, it is argued, comprise examples of Foucauldian transversal struggles, although their coverage in Huisgenoot is also attributed to their shock value and/or record sales, rather than their transformational potential. Furthermore, such Huisgenoot coverage is also seen to place more transformationally oriented artists at risk of media ‘domestication’, with its concomitant inhibiting of creativity.
