Change in narrative therapy : a pragmatic hermeneutic case study

- Description: The client of this case study was a twenty two year old female in her first year at university. The client had come into therapy because she had felt depressed, lonely and riddled with selfdoubt. The author used a Narrative Therapy approach with the client and was focussed on helping the client generate new meanings and stories that were more useful and empowering for the client. In this case study, the author was interested in exploring the process of change that the client underwent during the therapy process and he would rely on identifying innovative moments to track these changes. This interest informed the research question; what is the process of change in narrative therapy as tracked through the therapeutic dialogue? How does the change process in this case study track with the heuristic model of change put forward by Gonçalves and his colleagues? The author chose to use a pragmatic hermeneutic case study method in order to analyse the data and the results were organised into a coherent narrative. The data was collected from twenty two therapy sessions and these were grouped together into themes, namely a quick start, the beginning of change, thickening the innovative moments and lighting the fire. The results of this study reveal that despite being considered a good outcome case by the author, the process of change differed somewhat to that proposed by the heuristic model of change.

The self-assembled films are stable and showed blocking characteristics towards the following Faradaic processes; gold surface oxidation, under potential deposition of copper and solution redox chemistry of [Fe(H2O)6 3+ ]/[Fe(H2O)6 2+ ]. The solution chemistry of [Fe(CN)6]−3/[Fe(CN)6]−4 redox process was used to study the orientation of the CoPcs-SAMs and this revealed a possible deviation from flat orientation of the complexes on the gold electrodes when the SAM formation times were greater than 24 h. For SAM formation time at 24 and 48 h, CoTDMPc-SAM showed more inhibition of the [Fe(CN)6]−3/[Fe(CN)6]−4 redox process than the CoTBMPc-SAM counterpart, indicating more blocking characteristics of the dodecylmercapto ring substituents compared to the benzylmercapto counterpart. To avoid SAM desorption, the potential application should be limited to −0.2 to +0.8 V vs. Ag∣AgCl (NaCl, sat’d) in acidic and neutral pH.

- Description: Tetra-carboxy acid chloride phthalocyanine complexes of cobalt, iron and manganese were synthesized and characterized using spectroscopic and electrochemical techniques. These complexes were fabricated as thin films on gold electrode following a covalent immobilization and self-assembling methods. Surface electrochemical and spectroscopic characterization showed that these complexes are surface-confined species. The characterization using spectroscopic and electrochemical methods confirmed the formation of thiol and MPc SAMs on gold electrode. The electrocatalytic behaviour of the SAM modified gold electrodes was studied for the detection of L-cysteine and hydrogen peroxide. The limits of detection (LoD) for Lcysteine were of the orders of 10[superscript -7] mol.L[superscript -1] for all the MPc complexes studied and the LoD for hydrogen peroxide at cobalt phthalocyanine modified gold electrode was of the orders of 10[superscript -7]mol.L[superscript -1] for both electrocatalytic oxidation and reduction. The modification process for gold electrodes was reproducible and showed good stability, if stored in pH 4 phosphate buffer solutions and can be used over a long period of time. The cobalt phthalocyanine modified gold electrode was also investigated for the fabrication of glucose oxidase (GOx)-based biosensor and as an electron mediator between the enzyme and gold electrode. The behaviour of the enzyme modified gold electrode towards the detection of glucose was studied and the results gave a limit of detection of the orders of 10[superscript -6] mol.L[superscript -1] with low binding constant (4.8 mM) of enzyme (GOx) to substrate (glucose) referred to as Michaelis-Menten constant. The practical applications, i.e. the real sample analysis and interference studies, for the enzyme modified gold electrodes were investigated. These studies showed that the enzyme electrode is valuable and can be used for glucose detection.

The gold disk electrode gave better catalytic performance in terms of stability and reduction of overpotential.

Its surface properties were investigated, relative to that of 2-SAM film, by cyclic voltammetry and scanning electron microscopy (SEM). Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry, using [Fe(CN)6]3− as redox probe, were used to investigate the dynamics of electron transport in the SWCNT-2-SAM film. Lower charge transfer resistance, RCT, of the SWCNT-2-SAM modified Au electrode, relative to that of the 2-SAM modified Au electrode, was an indication of enhanced electron transport in the presence of SWCNT. The SAM film of the nano-composite showed better electrocatalytic behaviour, relative to that of 2-SAM film, towards the oxidation of the insecticide, carbofuran. The electrode (SWCNT-2-SAM) showed good selectivity for carbofuran, in the presence of diazinon (a non-electroactive interferent) and at lower concentration of the electroactive interferents (chlorpyrifos and dichlorfos).

Also reductively, the sensor exhibited a very fast amperometric response time (~1 s), linearity up to 5 μM with a coefficient R² = 0.986 and a detection limit of 0.2 μM. The cobalt tetracarboxylic acid phthalocyanine self-assembled monolayer was then evaluated as a mediator for glucose oxidase (GOx)-based biosensor. The GOx (enzyme) was immobilized covalently onto Au-ME-CoTCAPc SAM using coupling agents: N-ethyl-N(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxy succinimide (NHS), and the results demonstrated a good catalytic behavior. Kinetic parameters associated with the enzymatic and mediator reactions were estimated using electrochemical versions of Lineweaver–Burk and Hanes equation, and the stability of the sensor was tested. The biosensor (Au-ME-CoTCAPc-GOx SAM) electrode showed good sensitivity (7.5 nA/mM) with a good detection limit of 8.4 μM at 3σ, smaller Michaelis–Menten constant (4.8 mM from Hanes plot) and very fast response time of approximately 5 s.

The GOx (enzyme) was immobilized covalently onto Au-ME-CoTCAPc SAM using coupling agents: N-ethyl-N(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxy succinimide (NHS), and the results demonstrated a good catalytic behavior. Kinetic parameters associated with the enzymatic and mediator reactions were estimated using electrochemical versions of Lineweaver–Burk and Hanes equation, and the stability of the sensor was tested. The biosensor (Au-ME-CoTCAPc-GOx SAM) electrode showed good sensitivity (7.5 nA/mM) with a good detection limit of 8.4 μM at 3σ, smaller Michaelis–Menten constant (4.8 mM from Hanes plot) and very fast response time of approximately 5 s.

The complexes synthesised were characterised using spectroscopic and electrochemical means. The complexes displayed interesting spectroscopic properties with absorption of the Q band being observed in the near infrared region. These complexes have unusual colours for MPc complexes, with purple or red colour being observed in solution. Interesting electrochemical properties were observed, with rare observation of the MnIV/MnIII redox couple. There was observation of oxidation peaks for the pentylthio derivatised Pc with titanium as the central metal; this is unusual since for reported thio derivatives based on TiPc, no oxidation was observed. The vanadium based Pc showed an interesting spectrum for the first ring based reduction. The absorption spectrum obtained for the 1st reduction of the vanadium complex using spectroelectrochemistry would normally indicate a metal based process but comparing with literature it was concluded that it is a ring based reduction process. Generally all the MPc complexes formed a well ordered stable monolayer on the gold electrode. Electrocatalytic studies using L-cysteine revealed that the SAM based on manganese (III) octapentylthio phthalocyanine (AcOMnOPTPc) was the most effective since it catalyses L-cysteine at much lower oxidation potentials and it is also much more stable.

The catalytic current was found to be linear with sulphite ion concentration in the concentration range employed in this work (0.1–1 mM) and sensitivities ranging from 1.68 to 2.96 μA/mM were observed for all the modified electrodes. Mechanisms of the catalytic oxidation of sulphite ions are proposed.

MobiSAM, a system which aims to enhance the Social Accountability Monitoring (SAM) methodology at local government level, has been designed and implemented. The research presented in this thesis examines tools and techniques for the development of cross-platform client applications, allowing access to the MobiSAM service, across heterogeneous mobile platforms, handsets and interaction styles. Particular attention is paid to providing an easily navigated user interface (UI), as well as offering clear and concise visualisation capabilities. Depending on the host device, interactivity is also included within these visualisations, potentially helping provide further insight into the visualised data. Guided by the results obtained from a comprehensive baseline study of the Grahamstown area, steps are taken in an attempt to lower the barrier of entry to using the MobiSAM service, potentially maximising its market reach. These include extending client application support to all identified mobile platforms (including feature phones); providing multi-language UIs (in English, isiXhosa and Afrikaans); as well as ensuring client application data usage is kept to a minimum. The particular strengths of a given device are also leveraged, such as its camera capabilities and built-in Global Positioning System (GPS) module, potentially allowing for more effective engagement with local municipalities. Additionally, a Short Message Service (SMS) gateway is developed, allowing all Global System for Mobile Communications (GSM) compatible handsets access to the MobiSAM service via traditional SMS. Following an iterative, user-centred design process, a thorough evaluation of the client application is also performed, in an attempt to gather feedback relating to the navigation and visualisation capabilities. The results of which are used to further refine its design. A comparative usability evaluation using two different versions of the cross-platform client application is also undertaken, highlighting the perceived memorability, learnabilitv and satisfaction of each. Results from the evaluation reveals which version of the client application is to be deployed during future pilot studies.

Guided by the results obtained from a comprehensive baseline study of the Grahamstown area, steps are taken in an attempt to lower the barrier of entry to using the MobiSAM service, potentially maximising its market reach. These include extending client application support to all identified mobile platforms (including feature phones); providing multi-language UIs (in English, isiXhosa and Afrikaans); as well as ensuring client application data usage is kept to a minimum. The particular strengths of a given device are also leveraged, such as its camera capabilities and built-in Global Positioning System (GPS) module, potentially allowing for more effective engagement with local municipalities. Additionally, a Short Message Service (SMS) gateway is developed, allowing all Global System for Mobile Communications (GSM) compatible handsets access to the MobiSAM service via traditional SMS. Following an iterative, user-centred design process, a thorough evaluation of the client application is also performed, in an attempt to gather feedback relating to the navigation and visualisation capabilities. The results of which are used to further refine its design. A comparative usability evaluation using two different versions of the cross-platform client application is also undertaken, highlighting the perceived memorability, learnabilitv and satisfaction of each. Results from the evaluation reveals which version of the client application is to be deployed during future pilot studies.

- Description: Zinc, cobalt and iron phenylthio substituted phthalocyanines have been synthesized and characterized. Cyclic and square wave voltammetry in dimethylformamide containing tetrabutylammonium perchlorate revealed five and six redox processes respectively for the cobalt and iron phenylthio substituted phthalocyanines. These complexes are easier to reduce compared to the corresponding unsubstituted MPc and to butylthio substituted derivatives. Spectroelectrochemistry (in dimethylformamide containing tetrabutylammonium perchlorate) was employed to assign the cyclic voltammetry peaks, and gave spectra characteristic of Fe(I)Pc for reduction of iron phenylthio substituted phthalocyanine and Co(I)Pc for the reduction of cobalt phenylthio substituted phthalocyanine. The spectrum of the former is particularly of importance since such species have not received much attention in literature. Cobalt and iron phenylthio substituted phthalocyanines have been deposited on Au electrode surfaces through the self assembled monolayer (SAM) technique. The so formed layers were studied using voltammetric techniques. These SAMs blocked a number of Faradic processes and electrocatalyzed the oxidation of L-cysteine. Amine substituted cobalt phthalocyanine (CoTAPc) was deposited on gold surfaces by using an interconnecting SAM of mercaptopropionic acid or dithiobis(N-succinimidyl propionate) through the creation of an amide. Reductive and oxidative desorption of the SAMs limit the useful potential window. The SAM-CoTAPc layers show electrocatalytic activities towards oxygen reduction through the Co(I) central metal ion. Both SAMs were highly stable and hence will be interesting tools for further research in surface modification and sensor development.
