Compound 1 exhibited moderated antibacterial activity towards Staphylococcus aureus with MIC value of 62.5 μg/mL. The crude MeOH extract (TMr) highly reduced Plasmodium falciparum growth with an IC50 value of 10.11 μg/mL, while hexane fraction (F1) highly reduced Trypanosoma brucei brucei growth with an IC50 value of 5.60 µg/mL. All tested samples presented little or no in vitro cytotoxicity on HeLa cell line. The present work confirms that T. mantaly is medicinally important and may be used effectively as an antimicrobial, an antiplasmodial and an antitrypanosomial with promising therapeutic index.

The prevalence of use and value of wild edible herbs in South Africa

- Description: The prevalence of use and commercial value of wild edible herbs in South Africa is examined from four recent quantitative studies at eight different sites. The use of wild edible herbs was widespread in rural communities, with over 90% of households using them in all but one sample site. Mean frequency of use in season was generally between two and five times per week, with a mean of four times across all sites. But many households consume them daily. Use in winter was less than in summer. The mass of wild edible herbs consumed ranged from 12 kg to over 130 kg per household per year. Local (farm-gate) prices ranged from R2.65 to R72 per kilogram, but were generally between R30 and R40 per kg. Direct-use value to consuming households ranged from R85 to almost R5000 across the eight sites, with a mean of R1020 per user household per year. Although harvesting of wild herbs takes time, the high gross direct-use value represents a considerable saving on having to purchase commercial alternatives. Key species differ from place to place both in availability and use, and include both indigenous and exotic species. Commonly used genera include Amaranthus, Bidens, Chenopodium, Cleome, Corchorus, and Momordica. The use, value and trade in wild edible herbs currently receives no recognition in land and agrarian reform policies. It is imperative that this be addressed, and the relationships between rural livelihoods, use of wild edible herbs, food security, and land and resource tenure be clarified, and debated within the policy forums around different models for, and delivery of, agrarian reform and rural development.

The use of wild edible herbs was widespread in rural communities, with over 90% of households using them in all but one sample site. Mean frequency of use in season was generally between two and five times per week, with a mean of four times across all sites. But many households consume them daily. Use in winter was less than in summer. The mass of wild edible herbs consumed ranged from 12 kg to over 130 kg per household per year. Local (farm-gate) prices ranged from R2.65 to R72 per kilogram, but were generally between R30 and R40 per kg. Direct-use value to consuming households ranged from R85 to almost R5000 across the eight sites, with a mean of R1020 per user household per year. Although harvesting of wild herbs takes time, the high gross direct-use value represents a considerable saving on having to purchase commercial alternatives. Key species differ from place to place both in availability and use, and include both indigenous and exotic species. Commonly used genera include Amaranthus, Bidens, Chenopodium, Cleome, Corchorus, and Momordica. The use, value and trade in wild edible herbs currently receives no recognition in land and agrarian reform policies. It is imperative that this be addressed, and the relationships between rural livelihoods, use of wild edible herbs, food security, and land and resource tenure be clarified, and debated within the policy forums around different models for, and delivery of, agrarian reform and rural development.

This is because, the African continent was since ancient times characterized by certain traditions and cultures which mainly have contributed to the plight of the women. This goes to emphasise that women in most cases are the victims of misdeeds such as early marriage, forced marriage, prejudices, violence, marginalisation, exploitation, discrimination; in short, vices that lead to devalorisation of African women in Africa. Over time, some contemporary French-speaking African writers have responded to the devaluation and valuation of women in their novels using 21st century objective view. With regard to the theme that we decided to address in this research, it is worth knowing that the preponderant task is based on the socio-critical theory, postulated by Claude Duchet. Meanwhile, recourse is also made to the comparative method by Rens, Bod et al. However, since the author’s message could be understood by the reader through the use of some expressions or terms that speak directly to the themes of the research, and that a reader’s role in reading experience cannot be overemphasised, we have been able to also lean on the Reader-response theory of Louise Michelle Rosenblatt. The three methods were used concurrently especially that the analysis of each text is based on how characters are being depicted leading us to realizing that our findings on how the African woman is devalued in the first part of the research, while in the second part of the thesis, our findings demonstrate that a lot of tributes are showered on the African woman due to her numerous outstanding qualities. In effect, our findings also reinforce a significant shift in the narrative that concerns the contemporary view on womanhood. Hence, moving from a dogmatic overconcentration on her lot to the new era woman whose lot is now redefined via the new wave feminist perspective on the negation of the African woman. This could be said to be in line with Amadiume Ifi’s contributions and pioneering work in feminist discourse towards new ways of thinking about sex and gender, the question of power, and women’s place in history and culture. , Pour commencer, il est nécessaire de se souvenir qu’à l’époque actuelle, comme juste après les indépendances de la plupart des pays africains, bon nombre des écrivains venant du continent africain décident de se préoccuper des méfaits sociaux tels que le mariage précoce et/ou forcé, les préjugés, la violence, la marginalisation, l’exploitation, la discrimination, entre autres. Evaluation of incidence of Mycobacterium tuberculosis complex associated with soil, hayfeed and water in three agricultural facilities in Amathole District Municipality in the Eastern Cape Province, South Africa

MTBC has members that may contaminate an extensive range of hosts, including wildlife. Diverse wild species are known to cause disease in domestic livestock and are acknowledged as TB reservoirs. It has been a main study worldwide to deliberate on bTB risk factors as a result some studies focused on particular parts of risk factors such as wildlife and herd management. The objectives of this study were to design questionnaires from commercial farms and smallholding farms; isolate and identify MTBC from collected samples using culture and PCR assays recovered from Fort Hare, Middledrift and Seven star dairy farms; and assessing genotypic drug resistance through detection of mutations conferring resistance to INH and RMP associated with first line treatment for MTBC infection. Questionnaires were administered to thirty (30) smallholding farm owners in the two villages (kwaMasele and Qungqwala) and three (3) three commercial farms (Fort Hare dairy farm, Middledrift dairy farm and Seven-star dairy farm). Detection of M. tuberculosis complex was achieved by Polymerase Chain Reaction using primers for IS6110; whereas a genotypic drug resistance mutation was detected using Genotype MTBDRplus assays. Nine percent (9 percent) of respondents had more than 40 cows in their herd, while 60 percent reported between 10 and 20 cows in their herd. Relationship between farm size and vaccination for TB differed from forty-one percent (41 percent) being the highest to the least five percent (5 percent). The highest number of respondents who knew about relationship between TB cases and cattle location was ninety-one percent (91 percent). Approximately fifty-one percent (51 percent) of respondents had knowledge about wild life access to the farms. Relationship between import of cattle and farm size ranged from nine percent (9 percent) to thirty-five percent (35 percent). Cattle sickness in relation to farm size differed from forty-three (43 percent) being the highest to the least three percent (3 percent); while thirty-three percent (33 percent) of respondents had knowledge about health management. Respondents with knowledge about the occurrence of TB infections in farms were forty-eight percent (48 percent). The frequency of DNA isolation from samples ranged from the highest forty-five percent (45 percent) from water to the least twenty-two percent (22 percent) from soil. Fort Hare dairy farm had the highest number of positive samples forty-four percent (44 percent) from water samples; whereas Middledrift dairy farm had the lowest positive from water, seventeen percent (17 percent). Twelve (22 percent) out of 55 isolates showed resistance to INH and RMP that is, multi-drug resistance (MDR) and nine percent (9 percent) were sensitive to either INH or RMP. The mutations at rpoB gene differed from 58 percent being the highest to the least (23 percent). Fifty-seven percent (57 percent) of samples showed a S315T1 mutation while only 14 percent possessed a S531L in the katG gene. The highest inhA mutations were detected in T8A (80 percent) eighty percent and the least was observed in A16G (17 percent). The results of this study reveals that risk factors for bTB in cattle and dairy farm workers is a serious issue abound in the Eastern Cape of South Africa; with the possibility of widespread dissemination of multidrug resistant determinants in MTBC from the environment.

- Description: Sharptooth catfish Clarias gariepinus was introduced into the Eastern Cape Province, South Africa, in 1976 and there are concerns about its possible negative impacts on native biota. This study investigated its trophic impact by examining its feeding habits. Stomach content and stable isotope analyses were compared from three localities—the Great Fish River, Sundays River and Glen Melville Dam. Stomach content analysis indicated a catholic diet dominated by fish particularly in all localities.Spatially, however, the diets revealed differences based on the dominance of macrophytes that were only present in the rivers, and aquatic invertebrates that appeared more diverse within the Great Fish River compared to other localities. By contrast, stable isotopes revealed a more generalised feeding pattern with no clear dominance of particular prey. Stable isotopes further showed that the catfish was a complex predator, with large catfish being top predators whereas smaller size groups appeared to feed lower in the food chain. An ontogenetic shift in diet was evident, with small fish predominantly consuming aquatic invertebrates and shifting towards fish with increasing size. High dietary overlap suggests the potential risk associated catfish feeding, especially the potential of piscivory by small catfish that are more likely to persist in shallow and marginal where endangered indigenous minnows occur. The alteration of environmental conditions, especially flow by inter basin water transfer (IBWT) schemes, was inferred to have had a probable influence its invasion success. Occurrence of other invaders, which was facilitated by the IBWT together with the catfish, posits the risk of invasion meltdown within the study systems.

- Description: Sharptooth catfish Clarias gariepinus was introduced into the Eastern Cape Province, South Africa, in 1976 and there are concerns about its possible negative impacts on native biota. This study investigated its trophic impact by examining its feeding habits. Stomach content and stable isotope analyses were compared from three localities—the Great Fish River, Sundays River and Glen Melville Dam. Stomach content analysis indicated a catholic diet dominated by fish particularly in all localities.Spatially, however, the diets revealed differences based on the dominance of macrophytes that were only present in the rivers, and aquatic invertebrates that appeared more diverse within the Great Fish River compared to other localities. By contrast, stable isotopes revealed a more generalised feeding pattern with no clear dominance of particular prey. The combination of quinoline scaffolds with other antimalarial compounds and metals-based drugs have been reported to be a potential approach to overcome drug resistance common in the currently used antimalarials. 4-Aminoquinoline was hybridized with selected organic molecules and metal-based compounds to form a class of hybrid compounds containing either an amide bond or ester bond as a linker between the parent molecules. 4-Aminoquinoline derivatives are known compounds and they were prepared via known synthetic routes and characterized. The hybrid compounds were characterized and the FTIR results confirmed the successful linkage of 4-aminoquinoline derivatives to selected organic scaffolds to form hybrid compounds. NMR results confirmed the successful formation of hybrid compounds. MS showed signals of the hybrid molecules confirming the successful isolation of the hybrid compounds. In vitro antiplasmodial assay was performed against asexual parasite and chloroquine was used as a reference drug. The percentage inhibition effects of the hybrid compounds were in a range of 96-102% at 5 µM and 36-96% at 1 µM suggesting that the percentage inhibition effect of the hybrid compounds was influenced by the drug concentration. Hybridization of either 4-aminosalicylic scaffold or ferrocene butanoic acid with 4- aminoquinoline derivatives is a potential synthetic route that can result in potent antimalarials. However, more research is needed to fully understand the structure-activity relationship of these hybrid compounds.

Diet of the yellow mongoose (Cynictis penicillata) in the Albany Thicket Biome of South Africa

Diet was determined by identifying undigested food remains in the scats. Hair samples were examined under the compound microscope to identify which mammals were ingested. In the field, both arthropod and small mammal abundance were determined in order to evaluate whether yellow mongooses are opportunistic or specialist feeders. Furthermore, inter-annual (2006–2008) and inter-group/inter-site variations in the diet were assessed. Based on faecal analysis, arthropods were the principal prey, while small mammals acted as supplementary food item. Other food categories like vegetation, reptiles and birds were only ingested or preyed upon infrequently. Intermediate dietary diversity, and low standardised diet breadth indices were obtained for all seasons, as well as for the whole sampling period. The diet overlaps calculated for pairs of seasons were very high and did not differ substantially. However, the diet of the yellow mongoose varied seasonally, yearly and between groups (sites), mostly due to variations in arthropod and small mammal consumption. Food availability displayed an increase in biomass of both small mammals and arthropods in summer and autumn. Results obtained in this study could suggest that the yellow mongoose at the GFRR is insectivorous by preferring to feed on insects (and arthropods in general) when both insects and rodents are abundant. However the fact that its diet varies both seasonally, yearly and locally, as well as the presence of remains of other food categories in the scats, indicates that this species is rather an opportunistic, generalist feeder than an insect specialist.

- Description: Plasmodium falciparum is the pathogen that is responsible for the most virulent, severe and dangerous form of human malaria infection, accounting for nearly a million deaths every year. To survive and develop in the unusual environment of the red blood cells, the parasite causes structural remodelling of the host cell and biochemical changes through the export of virulence factors. Among the exportome are the molecular chaperones of the heat shock protein family, of which Hsp40s and Hsp70s are prominent. PF A0660w, a type II P. falciparum Hsp40, has been shown to be exported in complex with PfHsp70-x into the infected erythrocyte, suggesting possible functional interactions. However, the chaperone properties of PF A0660w and its interactions with proteins of parasite and human origin are yet to be investigated. Using a codon optimised coding region, PF A0660w was successfully expressed in E. coli M 15 [pREP4] cells. However, the expressed protein was largely deposited as insoluble pellet, and analysis of the pellets revealed a high percentage of PF A0660w, characteristic of inclusion body formation. PF A0660w was purified from inclusion bodies using additive enhanced solubilisation and refolding buffers followed by nickel affinity chromatography. SDS-PAGE and western analysis revealed that the purified protein was of high purity. Size exclusion chromatography showed that the protein existed as a monomer in solution and the secondary structure analysis using Fourier transformed infrared spectroscopy (FTIR) confirmed the success of the refolding approach. Its monomeric state suggests that PF A0660w may be functionally different from other Hsp40 that form dimers and that for PF A0660w, dimer formation may not be needed to maintain the stability of the protein in solution, but may occur in response to functional necessities during its interaction with partner Hsp70. PFA0660w was able to significantly stimulate the ATPase activity ofPfl-Isp70-x but not Pfl-Isp70-1 or human Hsp70 (HsHsp70), suggesting a specific functional interaction. Also, PF A0660w produced a dose dependent suppression of rhodanese aggregation and cooperated with Pfl-Isp70-1, PfHsp70-x and HsHsp70 to cause enhanced aggregation suppression. Its ability to independently suppress aggregation may help to maintain substrates in an unfolded conformation for eventual transfer to partner Hsp70s during refolding processes. There is no convenient manual to guide the would-be local historian of the Cape. There are few models that provide an insight into the main problems encountered by the local historian of a Cape community. In such local histories as exist, attention has been focussed predominantly on the foundation and physical growth of towns, the naming of streets, the establishment of schools and hospitals. Many of these accounts were written for publicity purposes or to commemorate the founding of towns.
