Ammonia removal from water by ion exchange using South African and Zambian zeolite samples

- Description: One problem of intensive fish culture systems is the progressive build-up of toxic wastes such as ammonia. The possibility of improving aquaculture water quality using two kinds of zeolite is discussed. Zeolites are alumino-silicates whose framework allows them to exchange cations. Ion exchange has been demonstrated to be competitive with other methods of ammonia removal due to the high selectivity for ammonia exhibited by zeolite materials. In this study an unknown Zambian zeolite (identified as laumontite by X-ray diffraction techniques) and Pratley clinoptilolite (a South African zeolite) were tested under laboratory conditions and in a fresh water recirculating system. Ammonia cation exchange capacities (CEC) and suitable application rates for efficient water treatment were determined using the batch and column ion exchange procedures. Estimated ammonia uptake, the most important criterion used to assess performance of zeolite filters was strongly influenced by zeolite type, particle size, pre-treatment, regeneration and ion exchange method used. Statistical analysis showed significant differences in average ammonia CEC values between clinoptilolite (14.94 mg g⁻¹) and laumontite (2.77 mg g⁻¹), with the former displaying a higher Na⁺ ® NH₄⁺ exchange rate especially in the early reaction stages. This difference accords with the higher purity of clinoptilolite, 47% as opposed to 4.7% for laumontite, which makes it a better zeolite for ammonium removal. CEC increased linearly as particle size of the clinoptilolite was reduced resulting in a linear regression model (y = 18.29 – 3.704 x; r² = 74%). Pre-treatment of clinoptilolite using 1N NaCl significantly improved the ammonia CEC of clinoptilolite. Overall performance of both the batch and column methods achieved after regeneration (18.3 mg g⁻¹) was 25% higher than the estimated CEC values (13.0 mg g⁻¹) for the unregenerated samples of clinoptilolite. Comparison of CEC estimates using Pratley clinoptilolite, showed that average batch CEC estimates were significantly lower than the column method estimates. The average ammonia CEC values estimated in a fresh water recirculating system (5.80 mg g⁻¹ and 4.12 mg g⁻¹ for the 0.7-1.0 and 1.0-1.4 mm particle sizes, respectively) were significantly lower than the column and batch estimates for the same particle sizes (P < 0.05). Some nitrite (NO₂) and nitrate (NO3) build up was experienced probably due to the growth of autotrophs in the filters. Mass balance of nitrogen (N) for the three treatments of the fish trial (0.7-1.0 mm, 1.0-1.4 mm and the control treatment that had no zeolite in the filter) indicated that less that 10% of the N was retained for growth. It was found that 60% of the NH₄-N present associated with the soluble N was available for absorption by the zeolite filter or biological nitrification and that a total of approximately 22% of NH₄-N available was absorbed by clinoptilolite. The results indicate that the rate of nitrification can be deductively estimated by allowing a zeolite filter to become a biological filter. It is concluded that water treatment by ion exchange using natural zeolites, provides a reliable and efficient method for ammonia removal and appears to be a viable supplementary water treatment method for fresh water systems.

- Description: This research will appraise the production and utility of digital atlases in an African context, using personal experience gained through the production of a digital atlas for Swaziland, the opinions obtained from other producers of African digital atlases and relevant literature. Since the 1950s, decision makers and researchers have viewed information as a powerful contributor to national development. Over the past thirty years, developed countries have increased the efficient collation and dissemination of geographic information through the utilization of the digital environment. The development of Geographic Information Systems (GIS) has increased the range of applications attainable in digital mapping projects. Applications have ranged from resource inventories to the monitoring of environmental degradation, crime patterns and service provision. The patchy history of data collection, analysis and mapping in Africa, has had a limiting effect on the ability of countries to identify, plan and control their resources efficiently. Despite the desire to automate the mapping process and reap some of the planning benefits evident in the developed world, Africa has not succeeded in mobilising full technological potential. Political instability, poor infrastructure, the absence of national policy guidelines, and a lack of skilled manpower are some of the issues which have limited utilization. Despite many of the hurdles faced by African countries, automated mapping and analysis technologies are still being pursued. The role of computers in the utilisation of data has become apparent through several means, one of which is the digital atlas. Analytical functions in many of these products allow situation modelling and provide superior graphic displays in comparison to their paper counterparts. Several African countries have embarked on the development of national digital databases and in some cases have produced digital national atlases. The potential to improve resource utilization, service provision and land use planning using these atlases, does exist. The production and utility of these atlases in an African context, however, needs closer assessment. "Computers don't clothe, don't cure, don't feed. Their power begins and ends with information. Their usefulness is therefore strictly linked to the ffectiveness of the information" (Gardner,1993:16).

- Description: Three artificial stream designs, termed Large Artificial Stream Units (LASUs), Raceways, and Channels, at two major scales (1700 L, 12.5 L and 20 L recirculated volume) were developed at the Institute for Water Research, Rhodes University, Grahamstown, in order to explore the possibilities of using indigenous rheophilic macroinvertebrates in routine toxicity tests. This study compared these systems, using 96h-EC50 values from sodium sulphate toxicity tests as the experimental response. Two local Ephemeroptera (Leptophlebiidae: Adenophlebia auriculata Eaton, and Baetidae: Afroptilum sudafricanum Lestage) were evaluated for their suitability in routine toxicity tests; and the possible effects of elevated salinity levels in South African rivers on the test species were assessed. Two sets of experiments with each mayfly species were conducted, following an unreplicated regression design. Dechlorinated tap water was used as the water source. Experiments in the Channels were repeated to determine experimental variability. Results were compared statistically by testing for overlap of 95% confidence limits (95%Cls) of EC50 values. The differences between A. auriculata EC50 values in the different systems were statistically significant (no overlap of 95%CLs), but they were not more variable than has been considered normal for biological systems (Coefficient of variation 20.1 %; ratio of greatest EC50 / smallest EC50 1.63). The differences were not related to the scale or the average current velocity characteristic of each stream design (average current velocity LASUs - Raceways - Channels 0.090 - 0.083 - 0.038 m/s). The Channels proved to be most efficient with regard to practical performance as they are portable and easily transportable, user-friendly, reliable, splash-free, cost effective to construct, and can easily be adapted to specific requirements. These systems are therefore recommended for regular use. The suitability of the two mayfly species for routine toxicity testing was evaluated. A. auriculata EC50 values showed a significant negative correlation with the corresponding average body-size (range 1476 - 1610 μm, mean 1555 μm). The different average body-sizes probably reflected the abundance of a certain size range present in the Palmiet River at the time of collection. Both species reacted similarly to Na₂S0₄ (similar slopes of the toxicity curves), identifying this salt as a slow acting toxicant. A. sudafricanum populations were more sensitive to Na₂S0₄ (EC50 3.404 g/L) than A. auriculata (EC50 8.090 g/L), probably because of its smaller body-size (mean 709 μm) and a lack of extremely tolerant individuals. In comparison to other freshwater macro invertebrates, including the standard toxicity test organism Daphnia spp., both mayfly species seemed to be moderately tolerant of Na₂S0₄; therefore there was no particular advantage to using these indigenous taxa rather than Daphnia spp. An assessment of the effects of elevated salinity/TDS levels on the test taxa yielded preliminary insights. An investigation into the influence of the environment on spawning aggregations and jig catches of chokka squid Loligo Vulgaris reynaudii off the south coast of South Africa

- Description: Erratic and highly variable catches in the South African chokka squid Loligo vulgaris reynaudii fishery, cause socio-economic hardship for the industry and uncertainty for resource managers. Catch forecasting can reduce this problem as it is believed that catch variability is strongly influenced by environmental factors. In this study, data were collected at varying temporal and spatial scales. Data for the hourly time-scale study were collected from 1996-1998, aboard commercial vessels, whilst for the longer time-scales, data were extracted for Kromme Bay (a single fishing area) from existing databases (1991-1998) that were comprised of compulsory catch returns and oceanographic data. The environment-catch relationship for chokka squid on the inshore spawning grounds was then investigated using multiple correlation and regression analysis, analysis of variance, contingency table analysis and cross-correlation statistical techniques. This simple, direct, 'black box' statistical approach was relatively successful in developing a predictive capability. On a short time-scale (hourly), the regression model accounted for 32% of the variability in catch, with turbidity the main determinant (13%). On a daily monthly time-scale, the best prediction model was on a monthly scale, accounting for 40% of the variability in catch. The principal determinant, bottom temperature anomaly (11 %), was found to lag one month forward. Seasonal and diel catch variations induced changes in the relative importance of turbidity, water temperature and wind direction on catches. A strong, positive relationship was found between easterly winds (which cause upwelling) and catch, particularly in summer. Catch rates, however, decreased with an increase in turbidity. The correlation between temperature and catch was generally negative, however, higher catches were associated with a temperature range of 13-18°C. Highest catch rates were associated with easterly winds, zero turbidity conditions and sea surface temperatures from 15.0-16.9°C. Selected case studies (in situ observations) suggested that upwelling and turbidity events act as environmental triggers for the initiation or termination of the spawning process, respectively. A holistic approach is required to improve predictive capability of chokka squid abundance. Although short-term predictability remains essential (i.e. hourly-scale), future research should concentrate on long-term prediction models (e.g., monthly time-scales) involving greater spatial variation, which are the most important for management.

The objective of the analysis is to determine the viability and the economic benefits of using these technologies when developing systems that facilitate music distribution over the Internet. A prototype of a distributed digital music library and purchasing system named WAPS (for Web-based Audio Purchasing System) was developed and implemented in the Java programming language. In this thesis both the physical and the logical component elements of WAPS are explored in depth so as to provide an insight into the inherent problems of creating such a system, as well as the overriding benefits derived from the creation of such a system.

- Description: Rhabdomys pumilio, in the Eastern Cape of South Africa, does not use short day length as an inhibitory cue for reproductive activity, and, despite previous records indicating that these mice are strictly seasonal in their reproduction, litters have been found during the winters of some years,both in the Eastern Cape and in the Western Cape. This led me to believe that the reproductive activity of Rhabdomys pumilio is more opportunistic and that the cue or cues used to control reproduction are less predictable and, or more variable than the photoperiod cue used by many seasonally reproducing rodents. Two experiments were conducted, investigating the influence of low ambient temperature (15⁰C)and reduced food availability on the reproductive activity of both male and female four striped field mice. Mice were maintained in one of four conditions (food restricted at 15⁰C, food restricted at 26⁰C, ad lib. food at 15⁰C, and ad lib. food at 26⁰C) for 4 (males) and 8 weeks (females)(photoperiod 12L:12D, humidity 40%). Results indicated that the males reduced their reproductive activity slightly when exposed to either low temperature or low food availability and that maximum inhibition of reproduction occurred when mice were exposed to both low temperature and low food availability. However, female reproductive activity was inhibited when exposed to low food availability, irrespective of the temperature. Both sexes of mice showed varying abilities to resist fat loss and, in the males, the size of the fat store had a significant effect on reproduction. This varying ability to resist fat loss could be related to levels of activity and in the females (where activity was quantified), high activity scores were significantly associated with reproductive inhibition. These results support the hypothesis that reproduction in Rhabdomys pumilio is opportunistic and controlled by the availability of energy. I propose that the females will be more sensitive to reproductive inhibition due to their far greater post-fertilization responsibilities, where the reproductive activity of the females is rapidly inhibited by a reduction in food availability, while the males are less readily inhibited by low food availability or low temperature, unless the change in the controlling factors is severe enough, or prolonged, at which stage their reproductive activity will cease. The significance of opportunistic reproduction in the seasonal but unpredictable climate of the study area is discussed.

- Description: Helcion pruinosus, the rayed limpet, is found in the midshore region of the intertidal zone, (i.e. the Balanoid zone) of rocky shores, from Saldanha Bay to Sodwana Bay along the coast of South Africa. At Gonubie (32°57'S/25°01'E) this species of limpet was found under boulders, (when not feeding), and could reach average densities of 85/ m². The limpets prefer smooth boulders, avoiding those encrusted by coralline algae or boulders with a rough texture. There was no habitat segregation between adult and juvenile H. pruinosus, i.e. there was no sign of up-shore migration by larger individuals. Males out-numbered females by, on average 1.4 : 1, which was a similar sex ratio to that of the congeneric species, H. pectunculus (Gray, 996). Unlike H. pectunculus, there were no differences in shell lengths between males and females (p = 0.946 at Gonubie, south-east coast and p = 0.961 at Kommetjie, south-west coast; t-test). Shell height could also not be used as a criterion to differentiate between the sexes. The average maximum shell length of H. pruinosus at Gonubie and Kommetjie was 25 mm, but the maximum shell lengths found were 30 mm, (one individual in each case), on both the southeast coast and west coast. Growth occurred most rapidly in the first year of life, with individuals reaching 15 mm after 1 year. In the second year limpet growth slowed to only 7 mm. Sexual maturity was reached at a shell length of between 11 to 14 mm (at an age of 8 months to 1 year). The life-span of H. pruinosus was determined at 2.55 years on the south-east coast and 2.9 years on the south-west coast. Micro-growth bands were produced tidally. Growth occurred allometrically, i.e. shell length increased at a faster rate than shell height. The limpets foraged during low tide, at night, presumably feeding on epilithic algae on the boulders. A second smaller peak of limpet activity was often recorded during the day, when low tide coincided with dawn). More limpets (up to ten times more) were active at spring tides compared to neap tides. Limpet activity was greatest during the spring tides of autumn. During all seasons limpet activity peaked 30 minutes before low water, after which numbers decreased rapidly. All limpets had retreated beneath the boulders before being covered by the flooding tide. It was concluded that the limpets were responding to both endogenous and exogenous cues, but the actual stimulus for retreat could not be determined. The limpets did not return to a fixed scar nor did they return rigidly to a home site. H. pruinosus was found to be gonochoristic. Sexes could be separated, on dissection, according to the colour of their gonads (males had white gonads, whilst that of the females was olive-green). Gonads were present throughout the year in most individuals, i.e. totally spent animals were never observed. Reproductive seasonality was similar in west coast and south-east coast animals. The gonad indices of the west coast limpets were higher, (maximum of 35%), compared to that on the southeast coast, (25%). The animals were probably trickle spawners, with some individuals liberating gametes throughout the year. There was however synchrony in gametogenesis between male and female limpets on both the west coast and south-east coast lines. Gonad indices peaked in the summer months (October to December) with a second minor peak in April/May. These peaks occurred before the release of the gametes in spawning events. The gonad index was lowest in winter, (July). The ovaries were full of mature eggs (oocytes having a diameter of 200 to 250 ~) prior to spawning. The acinal wall thickness had been decreasing prior to spawning. After spawning there was an increase in previtellogenic eggs « 100 μm) in females, and an increase in the acinal wall thickness to indicate spermatogenesis. The two populations are subjected to different environmental conditions and the actual cue for stimulating the release of gametes could not be determined. Finally, a table of differences between H pruinosus and its sister species, H pectunculus was created to show the life-style of H pruinosus.

Asymmetric α-alkylation reactions

- Description: A novel camphor-derived hydroxy ketal 138 has been developed as a crural auxiliary, and used to prepare a series of six carboxylic esters of increasing steric bulk. The α-benzylation of this series of esters was achieved with diastereoselectivities of 59 - 83% d. e. and in 39 - 48% material yield. These results compared very favourably with those obtained in earlier studies using a regioisomeric analogue as the chiral auxiliary. Computer.modelling studies of the putative enolate intermediate has provided some insight into the possible mode of electrophilic attack at the α-carbon and the roles of the ketal protecting group and the lithium cation in these asymmetric transformations. In a related investigation, based on earlier work, a camphor-derived imino lactone has provided convenient access to α-alkyl α-amino acids, the imino lactone serving as a masked glycine equivalent. Using straight chain primary alkyl iodides [RI; R = Me, Et, Pr, Bu, CH₃(CH₂)₄ and CH₃(CH₄)₅], alkylation of the potassium enolate of the camphor-derived imino lactone was effected with 54 - 89% d.e. and in 54 - 87% material yield. Four novel alkylated derivatives were synthesised using isopropyl iodide, sec-butyl iodide and allyl iodide, the latter reagent resulting in both the monoallylated and diallylated products. While very good diastereoselectivities were achieved (83 - 88% d. e.) in these reactions, the material yields from reaction with the secondary alkyl iodides were low (31- 35%) due, presumably, to their decreased electrophilicity. Computer modelling studies of the enolate were carried out and support the hypothesis of endo attack by the electrophile on the enolate intermediate. These studies also indicate the possibility of coordination of the postassium cation to the endocyclic ester oxygen, thus effectively anchoring the bulky cation away from the reaction site.

Characterization of amide bond hydrolysis in novel hydantoinase-producing bacteria

- Description: This thesis describes a series of investigations into the amide bond-hydrolyzing activity of bacterial strains RU-KM1, RU-KM3L, RU-KM3S, and RU-OR, which were previously isolated for their ability to hydrolyze hydantoins to amino acids. The main aim of the study was to develop biotransformations with potential application in the production of enantiomerically pure amino acids and related compounds. Several compounds may be used as substrates by biocatalysts for the production of amino acids, such as hydantoins, amino nitriles, and amides. These compounds are not only important for amino acid production, but they may be used for production of other industrially important compounds, such as 2- arylpropionic acids, which are non-steroidal anti-inflammatory drugs. Thus, the ability of the above-mentioned strains to hydrolyze these substrates was investigated, with the view to utilizing the maximum potential of these biocatalysts. The compounds used as substrates in the investigation are all essentially amides. Thus, the ability of the strains to hydrolyze imides, hydantoins, and amides, was investigated. In particular, imides have a structure which is very similar to that of hydantoins, and thus it was an objective of the study to determine whether these strains could hydrolyze imides. Imidehydrolyzing activity has only recently been discovered in microorganisms. Hydantoin conversion involves a two-step hydrolysis reaction which yields, initially, an Ncarbamylamino acid intermediate, and subsequently, an "-amino acid. The hydantoinhydrolyzing enzymes of a Pseudomonas putida strain, RU-KM3S, were characterized in a crude extract preparation and reaction conditions for its biocatalytic application were optimized. The optimum conditions for conversion of 5-methylhydantoin were found to be 3 hours at 40°C, with conversion yields greater than 50% achieved. The enzymes of RU-KM3S demonstrated considerable stability, retaining 80% of their activity after incubation at 40°C for 3 hours. The activities of the enzymes were increased by the addition of a detergent to the extraction medium, suggesting that the enzymes might be membrane-bound. The results of the determination of the metal-dependence of the hydantoinase and N-carbamylase of RUKM3S suggested that these enzymes required metal ions for activity, with metal ions such as Mg²⁺, Mn²⁺, Zn²⁺, and Co²⁺ resulting in activation of the enzymes. However, Cu²⁺ and Fe²⁺ caused inactivation of these enzymes. The stereoselectivity of the enzymes was investigated, and the results suggested that the hydantoinase was non-selective, whereas the N-carbamylase was L-selective. The hydantoin substrate selectivity of RU-KM3S was compared to that of three other hydantoinase-producing bacteria, RU-KM1, RU-KM3L, and RU-OR. The four strains were able to hydrolyze all of the seven substrates tested. However, there was a difference in activity levels between crude extract preparations and whole cells, with crude extracts generally showing higher activity than whole cells, except in the case of RU-KM1. Some difference was also observed in the order of preference of substrates between whole cells and crude extracts. The preferred substrate for RU-KM1 whole cells was isopropylhydantoin, whereas the crude extract preparation preferentially hydrolyzed p-hydroxyphenylhydantoin. RU-KM3L whole cells achieved a higher conversion yield with isobutylhydantoin, whereas the crude extract achieved a higher yield with 5-t-butylhydantoin. RU-KM3S whole cells and crude extract preferentially hydrolyzed 5-n-butylhydantoin, although the yield was greater with the crude extract. The highest conversion yields were observed with RU-KM3S crude extract, with conversion yields of 71.6% and 100% for n-butylhydantoin and phydroxyphenylhydantoin, respectively.The ability of RU-KM1, RU-KM3L, and RU-KM3S to hydrolyze nitriles, initially to amides and subsequently to carboxylic acids, was investigated. These strains were demonstrated to be unable to utilize acrylonitrile, propionitrile and benzonitrile as nitrogen sources, but were able to hydrolyze acrylonitrile, propionitrile and acetonitrile, in resting cell reactions. Nitrile hydrolysis was demonstrated to be inducible in all three strains, and the enzyme system responsible for nitrile hydrolysis was proposed to be a nitrile hydratase-amidase system. Amidase activity in the four bacterial strains was investigated. The ability of RU-KM1, RUKM3L, RU-KM3S, and RU-OR to utilize amides as a nitrogen source was investigated, and the results showed that propionamide was a good nitrogen source for all four of the strains. Amide-hydrolyzing activity, by resting cells, was shown to be inducible by propionamide in all four strains. RU-KM3S demonstrated superior amide-hydrolyzing ability in that it hydrolyzed propionamide, acetamide, and acrylamide to a greater extent than the other strains. Resting cells of RU-KM1 and RU-OR were demonstrated to have the ability to hydrolyze the imide substrate, succinimide, and this imidase activity was found to be inducible. These strains were also able to utilize this imide as the sole source of nitrogen for growth, which is a novel finding, as to date, bacteria have only be reported to utilize imides as a carbon source. The identity of the enzyme system responsible for succinimide hydrolysis is not yet clear. In conclusion, the hydantoin-hydrolyzing enzymes of RU-KM3S have been shown to be possibly membrane associated, which is a novel finding that has also been proposed in three other hydantoinase-producing strains in our laboratory. This study has shown that the Ncarbamylase of RU-KM3S is L-stereoselective, which, to our knowledge, is the first report of an L-stereospecific N-carbamylase in a Pseudomonas putida. Publication of these findings is already in progress. This is the first report on the study of imide hydrolysis in either an Agrobacterium tumefaciens or a Pseudomonas sp., and publications reporting these results are in preparation.

The hydantoin substrate selectivity of RU-KM3S was compared to that of three other hydantoinase-producing bacteria, RU-KM1, RU-KM3L, and RU-OR. The four strains were able to hydrolyze all of the seven substrates tested. However, there was a difference in activity levels between crude extract preparations and whole cells, with crude extracts generally showing higher activity than whole cells, except in the case of RU-KM1. Some difference was also observed in the order of preference of substrates between whole cells and crude extracts. The preferred substrate for RU-KM1 whole cells was isopropylhydantoin, whereas the crude extract preparation preferentially hydrolyzed p-hydroxyphenylhydantoin. RU-KM3L whole cells achieved a higher conversion yield with isobutylhydantoin, whereas the crude extract achieved a higher yield with 5-t-butylhydantoin. RU-KM3S whole cells and crude extract preferentially hydrolyzed 5-n-butylhydantoin, although the yield was greater with the crude extract. The highest conversion yields were observed with RU-KM3S crude extract, with conversion yields of 71.6% and 100% for n-butylhydantoin and phydroxyphenylhydantoin, respectively.The ability of RU-KM1, RU-KM3L, and RU-KM3S to hydrolyze nitriles, initially to amides and subsequently to carboxylic acids, was investigated. These strains were demonstrated to be unable to utilize acrylonitrile, propionitrile and benzonitrile as nitrogen sources, but were able to hydrolyze acrylonitrile, propionitrile and acetonitrile, in resting cell reactions. Nitrile hydrolysis was demonstrated to be inducible in all three strains, and the enzyme system responsible for nitrile hydrolysis was proposed to be a nitrile hydratase-amidase system. Amidase activity in the four bacterial strains was investigated. The ability of RU-KM1, RUKM3L, RU-KM3S, and RU-OR to utilize amides as a nitrogen source was investigated, and the results showed that propionamide was a good nitrogen source for all four of the strains. Amide-hydrolyzing activity, by resting cells, was shown to be inducible by propionamide in all four strains. RU-KM3S demonstrated superior amide-hydrolyzing ability in that it hydrolyzed propionamide, acetamide, and acrylamide to a greater extent than the other strains. Resting cells of RU-KM1 and RU-OR were demonstrated to have the ability to hydrolyze the imide substrate, succinimide, and this imidase activity was found to be inducible. These strains were also able to utilize this imide as the sole source of nitrogen for growth, which is a novel finding, as to date, bacteria have only be reported to utilize imides as a carbon source. The identity of the enzyme system responsible for succinimide hydrolysis is not yet clear. In conclusion, the hydantoin-hydrolyzing enzymes of RU-KM3S have been shown to be possibly membrane associated, which is a novel finding that has also been proposed in three other hydantoinase-producing strains in our laboratory. This study has shown that the Ncarbamylase of RU-KM3S is L-stereoselective, which, to our knowledge, is the first report of an L-stereospecific N-carbamylase in a Pseudomonas putida. Publication of these findings is already in progress. This is the first report on the study of imide hydrolysis in either an Agrobacterium tumefaciens or a Pseudomonas sp., and publications reporting these results are in preparation.

Currently, Virtual Reality is used mainly in the field of visualisation where 3D graphics allow users to more easily view complex sets of data or structures. The field of interaction in Virtual Reality has been largely neglected due mainly to problems with input devices and equipment costs. Recent research has aimed to overcome these interaction problems, thereby creating a usable interaction platform for Virtual Reality. This thesis presents a background into the field of interaction in Virtual Reality. It goes on to propose a generic framework for the implementation of common interaction techniques into a homogeneous application development environment. This framework adds a new layer to the standard Virtual Reality toolkit – the interaction abstraction layer, or interactor layer. This separation is in line with current HCI practices. The interactor layer is further divided into specific sections – input component, interaction component, system component, intermediaries, entities and widgets. Each of these performs a specific function, with clearly defined interfaces between the different components to promote easy objectoriented implementation of the framework. The validity of the framework is shown in comparison with accepted taxonomies in the area of Virtual Reality interaction. Thus demonstrating that the framework covers all the relevant factors involved in the field. Furthermore, the thesis describes an implementation of this framework. The implementation was completed using the Rhodes University CoRgi Virtual Reality toolkit. Several postgraduate students in the Rhodes University Computer Science Department utilised the framework implementation to develop a set of case studies. These case studies demonstrate the practical use of the framework to create useful Virtual Reality applications, as well as demonstrating the generic nature of the framework and its extensibility to be able to handle new interaction techniques. Finally, the generic nature of the framework is further demonstrated by moving it from the standard CoRgi Virtual Reality toolkit, to a distributed version of this toolkit. The distributed implementation of the framework utilises the Common Object Request Broker Architecture (CORBA) to implement the distribution of the objects in the system. Using this distributed implementation, we are able to ascertain that CORBA is useful in the field of distributed real-time Virtual Reality, even taking into account the extra overhead introduced by the additional abstraction layer. We conclude from this thesis that it is important to abstract the interaction layer from the other layers of a Virtual Reality toolkit in order to provide a consistent interface to developers. We have shown that our framework is implementable and useful in the field, making it easier for developers to include interaction in their Virtual Reality applications. Our framework is able to handle all the current aspects of interaction in Virtual Reality, as well as being general enough to implement future interaction techniques. The framework is also applicable to different Virtual Reality toolkits and development platforms, making it ideal for developing general, cross-platform interactive Virtual Reality applications.

- Description: This thesis investigates the development of a low cost, component based facial virtual conferencing system. The design is decomposed into an encoding phase and a decoding phase, which communicate with each other via a network connection. The encoding phase is composed of three components: model acquisition (which handles avatar generation), pose estimation and expression analysis. Audio is not considered part of the encoding and decoding process, and as such is not evaluated. The model acquisition component is implemented using a visual hull reconstruction algorithm that is able to reconstruct real-world objects using only sets of images of the object as input. The object to be reconstructed is assumed to lie in a bounding volume of voxels. The reconstruction process involves the following stages: - Space carving for basic shape extraction; - Isosurface extraction to remove voxels not part of the surface of the reconstruction; - Mesh connection to generate a closed, connected polyhedral mesh; - Texture generation. Texturing is achieved by Gouraud shading the reconstruction with a vertex colour map; - Mesh decimation to simplify the object. The original algorithm has complexity O(n), but suffers from an inability to reconstruct concave surfaces that do not form part of the visual hull of the object. A novel extension to this algorithm based on Normalised Cross Correlation (NCC) is proposed to overcome this problem. An extension to speed up traditional NCC evaluations is proposed which reduces the NCC search space from a 2D search problem down to a single evaluation. Pose estimation and expression analysis are performed by tracking six fiducial points on the face of a subject. A tracking algorithm is developed that uses Normalised Cross Correlation to facilitate robust tracking that is invariant to changing lighting conditions, rotations and scaling. Pose estimation involves the recovery of the head position and orientation through the tracking of the triangle formed by the subject's eyebrows and nose tip. A rule-based evaluation of points that are tracked around the subject's mouth forms the basis of the expression analysis. A user assisted feedback loop and caching mechanism is used to overcome tracking errors due to fast motion or occlusions. The NCC tracker is shown to achieve a tracking performance of 10 fps when tracking the six fiducial points. The decoding phase is divided into 3 tasks, namely: avatar movement, expression generation and expression management. Avatar movement is implemented using the base VR system. Expression generation is facilitated using a Vertex Interpolation Deformation method. A weighting system is proposed for expression management. Its function is to gradually transform from one expression to the next. The use of the vertex interpolation method allows real-time deformations of the avatar representation, achieving 16 fps when applied to a model consisting of 7500 vertices. An Expression Parameter Lookup Table (EPLT) facilitates an independent mapping between the two phases. It defines a list of generic expressions that are known to the system and associates an Expression ID with each one. For each generic expression, it relates the expression analysis rules for any subject with the expression generation parameters for any avatar model. The result is that facial expression replication between any subject and avatar combination can be performed by transferring only the Expression ID from the encoder application to the decoder application. The ideas developed in the thesis are demonstrated in an implementation using the CoRgi Virtual Reality system. We investigate user sign-on and two of the main authentication technologies that can be used to control a resource through authentication and providing additional security services. The problems with the existing sign-on scenario are that users have too much credential information to manage and are prompted for this information too often. Single Sign-On (SSO) is a viable solution to this problem if physical procedures are introduced to minimise the risks associated with its use. The Generic Security Services API (GSS-API) provides security services in a manner in- dependent of the environment in which these security services are used, encapsulating security functionality and insulating users from changes in security technology. The un- derlying security functionality is provided by GSS-API mechanisms. We developed the Secure Remote Password GSS-API Mechanism (SRPGM) to provide a mechanism that has low infrastructure requirements, is password-based and does not require the use of long-term asymmetric keys. We provide implementations of the Java GSS-API bindings and the LIPKEY and SRPGM GSS-API mechanisms. The Secure Authentication and Security Layer (SASL) provides security to connection- based Internet protocols. After finding deficiencies in existing SASL mechanisms we de- veloped the Secure Remote Password SASL mechanism (SRP-SASL) that provides strong password-based authentication and countermeasures against known attacks, while still be- ing simple and easy to implement. We provide implementations of the Java SASL binding and several SASL mechanisms, including SRP-SASL.

- Description: There is consensus that high CO2 results in enhanced growth and yield for most crop plants. However, most of these studies were carried out in the presence of adequate nutrients, which is also the case in agricultural systems (managed ecosystems). About 20% of the earth’s land mass have sufficiently low levels of nutrients to cause some kind of stress to plants. On the other hand, elevated [CO2] decreases foliar nutrient elements in plants and as a result partitioning of certain nutrient elements in plants is altered. Little data is available on the partitioning of most nutrient elements in plants, and this will definitely impact on growth and yield. To investigate this, wheat (Triticum aestivum L. c.v. Gamtoos) was grown in controlled environment cabinets at 360 and 700 µmol mol -1 CO2. The full Long-Ashton nutrient solution comprising of three-nitrogen concentrations ([N]) viz. (4,6 and 12 mM) was used to water plants everyday. The measurement of net assimilation rate (NAR), stomatal conductance (gs), transpiration rate (E), water use efficiency (WUE), foliar [N], nitrogen use efficiency (NUE) and growth parameters (total plant biomass (TPB), total plant height (TPH), leaf area (LA), shoot and root dry weight) were made 7 days after germination (7 DAG) till the onset of flowering. The increase in nitrogen supply in the order of 4, 6 and 12mM resulted in an increase in NAR, g_s_ , WUE and a decline in E under elevated [CO2]. Under elevated [CO2] NAR was observed to increase during the first two weeks reaching its maximum at 14 DAG, thereafter followed by a decline reaching its maximum at 28 DAG. This was later followed by an increase at 35 DAG onwards. Under elevated [CO2], NAR was increased significantly between the nitrogen regimes during the first (7-14 DAG) and the last two (35-42 DAG) weeks. The response of assimilation as a function of internal [CO2] (Ci), showed a decrease with age at ages 14, 28 and 35 DAG. This negatively affected the initial slope and the CO2 saturated photosynthetic rates under all treatments. This suggest that acclimation may have been as a result of both stomatal and biochemical limitations. All the photosynthetic pigment levels (chl_a_, chl_b_, chl_(a+b)_, and C_(x+c)_ ) increased with an increase in nitrogen supply from 4 to 6mM [N]. A 12mM [N] resulted in a significant decline in the photosynthetic pigment levels compared to a 6mM [N]. Chla remained higher than chlb under all treatments. Also, NAR was seen to increase and decrease concomitantly with the photosynthetic pigment levels. Foliar [N] was seen to decrease with an increase in nitrogen supply from 4 to 6 mM [N] under elevated [CO2] and the effects were adverse under the 4mM [N]. Under the 6mM N regime foliar [N] was positively correlated to NAR for elevated [CO2] grown plants. Similarly, E was positively correlated to foliar [N] under the same conditions. Elevated CO2 and increase in nitrogen supply had a pronounced effect on total plant height (TPH), total plant biomass (TPB), leaf area (LA), shoot and root dry weight and nitrogen use efficiency (NUE). The effects were more pronounced under a 6mM [N] as a result of high NUE. However, under 12mM [N] growth was not as expected as a result of lower NUE. Under all treatments shoot dry weight (SDW) was positively correlated to NUE. Anatomical studies revealed that total leaf and midrib thickness was significantly increased with an increase in nitrogen supply under elevated CO2 to support the larger leaf areas. There were no significant changes in the chloroplast ultrastructure as a result of the increase in nitrogen supply and CO2 enrichment. Starch grain surface area was seen to decline with an increase in nitrogen under both ambient and elevated CO2. Elevated CO2 and increase in nitrogen supply significantly increased total grain dry weight per plant by 47 and 46% respectively under 6 and 12mM [N]. In contrast, the increase was by about 21, 61 and 67% respectively under 4, 6 and 12mM [N] between the CO2 regimes.

Under elevated [CO2], NAR was increased significantly between the nitrogen regimes during the first (7-14 DAG) and the last two (35-42 DAG) weeks. The response of assimilation as a function of internal [CO2] (Ci), showed a decrease with age at ages 14, 28 and 35 DAG. This negatively affected the initial slope and the CO2 saturated photosynthetic rates under all treatments. This suggest that acclimation may have been as a result of both stomatal and biochemical limitations. All the photosynthetic pigment levels (chl_a_, chl_b_, chl_(a+b)_, and C_(x+c)_ ) increased with an increase in nitrogen supply from 4 to 6mM [N]. A 12mM [N] resulted in a significant decline in the photosynthetic pigment levels compared to a 6mM [N]. Chla remained higher than chlb under all treatments. Also, NAR was seen to increase and decrease concomitantly with the photosynthetic pigment levels. Foliar [N] was seen to decrease with an increase in nitrogen supply from 4 to 6 mM [N] under elevated [CO2] and the effects were adverse under the 4mM [N]. Under the 6mM N regime foliar [N] was positively correlated to NAR for elevated [CO2] grown plants. Similarly, E was positively correlated to foliar [N] under the same conditions. Elevated CO2 and increase in nitrogen supply had a pronounced effect on total plant height (TPH), total plant biomass (TPB), leaf area (LA), shoot and root dry weight and nitrogen use efficiency (NUE). The effects were more pronounced under a 6mM [N] as a result of high NUE. However, under 12mM [N] growth was not as expected as a result of lower NUE. Under all treatments shoot dry weight (SDW) was positively correlated to NUE. Anatomical studies revealed that total leaf and midrib thickness was significantly increased with an increase in nitrogen supply under elevated CO2 to support the larger leaf areas. There were no significant changes in the chloroplast ultrastructure as a result of the increase in nitrogen supply and CO2 enrichment. Starch grain surface area was seen to decline with an increase in nitrogen under both ambient and elevated CO2. Elevated CO2 and increase in nitrogen supply significantly increased total grain dry weight per plant by 47 and 46% respectively under 6 and 12mM [N]. In contrast, the increase was by about 21, 61 and 67% respectively under 4, 6 and 12mM [N] between the CO2 regimes.

This project set out to improve the potential economic value of Sorghum bicolor as a crop. The task was undertaken by screening for selected enzymes in the plant that would have a potential market for use in industrial applications and in biotransformations, specifically proteases, polyphenol oxidases and peroxidases. Asurveywas conducted using standard enzyme assays and crude plant extracts, to determine whether the selected enzymes were present. Grain tissue did not appear to have significant protease or polyphenoloxidase activity, but high levels of peroxidases were detected, withthe young grain extracts showing more activity(4.63U/mL)thanripegrain extracts (0.62 U/mL). Leaf tissue extracts contained low levels of protease activity, a considerable amount of polyphenol oxidase (0.127 U/mL), and peroxidase (4.7 U/mL) activities comparable with that found in grain tissue. Root tissue extract was found to contain the highest levels of peroxidase activity (7.8 U/mL) compared to the other extracts. Therefore, sorghum peroxidase from the root was isolated, purified, characterized and applied to biotransformation reactions. Different sorghum strains,withvaryinggraincolour, (Zimbabwe - bronze, Seredo - brown and Epurpur - cream/white) were investigated for the presence of polyphenol oxidase and peroxidase activities. Results of spectrophotometric analysis showed that the enzymes did not appear to be strain specific. However, gel electrophoresis analysis revealed differences in band patterns among the strains. Partial purification of sorghum root peroxidase was achieved after centrifugation, extraction with polyvinylpolypyrrolidone (PVPP), ultrafiltration, and hydrophobic chromatography with phenyl Sepharose, followed by polyacrylamidegelelectrophoresis (PAGE). The specific activity of the 5-fold purified enzyme was found to be 122.3 U/mg. After PAGE analysis, two bands with molecular weights of approximately 30 000 and 40 000 were detected, which compares well with horse radish peroxidase (HRP) which has a molecular weight of approximately 44 000. The colour intensity of the bands in the activity gels indicated that sorghum root peroxidase had apparently higher levels of peroxidase activity than commercial horseradish peroxidase (HRP). Characterizationexperiments revealed that sorghumroot peroxidase is active over a broad temperature range and remains active at temperatures up to 100°C. It also has a broad substrate range. The optimum pH of the enzyme was found to be pH 5 - 6. Under standardized assay conditions, the optimal substrate concentration, using o-dianisidine as substrate, was 50 mM, and the optimal H2O2 concentration under these conditions was found to be 100 mM. Sorghum root peroxidase was applied in a preliminary investigation into the oxidative biotransformationof a number of aromatic compounds. The products obtained were comparable withthose whenthe compounds are reacted with HRP which is the most commonly used commercial peroxidase and has been extensively studied. - Description: The motivation for this study was to examine the exploitation patterns of the M. capensis and M. paradoxus hake fisheries on the Southeast Coast, and determine the size and species of hake caught in each of the hand-line, long-line, and trawl methods. The handline hake fishery has increased substantially over the last several years and concerns are beginning to emerge about the impact this will have on the inshore resource collected on the South Coast between August 1998 and July 1999 was used to describe the hand-line method and estimate annual landings. Data on the size and species in longline catches of hake caught during 1997 were already available for this study. Size distributions in trawl catches were determined from commercial category landing data reported by catch weight and depth. The species composition in these catches determined by comparison using RV Afrikana survey data collected in the same depth regions. Location plays a significant role in determining the sizes and species of hake caught by each gear. Hand-lines catch smaller sizes on average than do long-lines, inshore trawls target mainly M. capensis while offshore trawls catch both hake species. A substantial amount of the hand-line hake caught on the South Coast is not reported. Examination of the exploitation patterns reveal that intense trawling pressure is directed at the smaller sized M. paradoxus inhabiting the depth region between 160-400-meters. Inshore trawls discard a large amount of small sized M. capensis within the 100-meter isobath. A preliminary stock assessment on the status of each hake species found that M. paradoxus appears to be over-exploited while M. capensis was in better shape. However, length-based pseudo-cohort analysis, used in this assessment, is critically reliant on having length frequency data from a steady state population in equilibrium. This limits the application of this model for management purposes and this finding is purely theoretical at this stage. Results suggest that each hake species is under a different pattern and level of exploitation and the multi-species nature of hake stocks on the South Coast should be considered in developing optimum management policies. Future work should focus on developing appropriate age/length keys so that an age-based VPA, which is more powerful than the length-based approach, can be applied towards stock assessments on the South Coast. Alternatively, length-data covering a longer period should be compiled and the equilibrium assumption further investigated so that the results from length-based models can be used with more confidence.

Feeding biology of three euphausiid species in the vicinity of the Prince Edward Archipelago (Southern Ocean)

Data on the abundance and biomass of these species, estimated from bongo net tows, were investigated. Trophic position was assessed using gut contents and stable nitrogen isotope measurements. Feeding rate and daily carbon ration were estimated using the gut fluorescence and the gut fullness techniques. Vertical migrations into the surface waters at night were found to be strong for Euphausia vallentini adults and juveniles. Associated with these migrations were clear diel feeding patterns. Insufficient data during daylight hours for E. longirostris made it impossible to determine diel feeding patterns, but high feeding activity did occur during dark hours. Nematoscelis megalops did not show any distinct diel feeding pattern, but slightly higher gut fullness indices in the late afternoon suggested that feeding activity may have been highest during this period. For both Euphausia spp. high gut pigment levels were recorded in 1999, which corresponded to higher ambient chlorophyll a concentrations for that year. Highest initial gut pigment levels and highest ingestion rates were found for Euphausia longirostris in both years and lowest values were observed for N. megalops. High phytoplankton and low metazoan contributions to the diet of Euphausia vallentini juveniles, as shown in the gut content analysis, and low stable nitrogen isotope ratios (ð¹⁵N = 1.39±0.31), both indicated that this group was principally herbivorous. The results of gut content analysis of the adults of E. vallentini were similar to those of the juveniles, however, stable nitrogen isotope results showed that there was a higher degree of omnivory (ð¹⁵N = 3.81±0.66). Daily ration estimates from the gut fluorescence and fullness techniques showed that between 3.3 and 25.7 % of E. vallentini adults total daily carbon ration was derived from autotrophic sources. Although the contribution of carnivory to the diet was difficult to determine, the adults of this species may be considered omnivorous. Irrespective of the degree of carnivory, a dietry shift with an increase in size was evident for this species. Gut content analysis for Euphausia longirostris showed that this species consumed large amounts of both phytoplankton and metazoan prey and this was reflected in the stable nitrogen isotope results (ð¹⁵N = 6.88±0.60). These findings were supported by the results of the daily carbon ration estimates which showed that autotrophic carbon contributed between 6.9 and 20.3 % of the daily carbon consumption. The gut content analysis suggested that N. megalops was omnivorous, and the stable nitrogen isotope results place it in a trophic position equivalent to that of E. longirostris (ð¹⁵N = 6.83±0.78). - Description: Mesozooplankton community structure in the vicinity of the Prince Edward Islands (PEIs) was investigated during six surveys conducted in late austral summer (April/May), 1996 to 1999. Each year zooplankton samples were collected with a bongo net (300 µm mesh) at stations upstream (west), between and downstream (east) of the islands. Chlorophyll a concentrations were determined fluorometrically, corresponding with each net tow. The positions of the Subantarctic Front (SAF) and the Antarctic Polar Front (APF), in relation to the islands in the upstream region, were determined by a line of CTD and/or XBT stations. Both the SAF and the APF were characterised by a high degree of meridional variation in position. Changes in position of the fronts were shown to occur very rapidly. In 1999 the APF moved southwards by ~40nm and the SAF northwards by ~60nm in a period of two weeks, while in 1996 the SAF appeared to move ~120nm northwards in a two week period. The positions of the SAF and APF appeared to have a significant impact on phytoplankton biomass in the vicinity of the PEIs, through the alteration of local flow dynamics. Water retention in 1996, associated with the location of the SAF and APF far to the north and south of the PEIs respectively, corresponded with high chlorophyll a concentrations in the inter-island region (reaching 1.54 mg.m^(-3)). When the fronts were close to the islands, in 1997 and 1999, and a flow through environment existed, chlorophyll a concentrations in the inter-island region were comparatively low. Although biomass enhancement was only observed at the SAF in 1996, phytoplankton size structure in 1999 indicated that, when close to the PEIs, frontal production may be transported to the island system. This is potentially an important source of allochthonous input into the island system. Zooplankton assemblages were a mix of sub-Antarctic and Antarctic communities, with a weak presence of sub-tropical species. Cluster analysis showed that during each survey the region in the vicinity of the PEIs was divided into different, spatially separated zooplankton communities, associated with water masses of different origins. These communities were identified by variations in the abundance and biomass of species rather than variation in species composition and, in general, there was a relatively high degree of similarity both within and between surveys. Inter-annual community analysis revealed that, in many cases, there was greater similarity between communities from different years than communities within years, indicating that short-term variability exceeded inter-annual variability. Multiple regression analysis showed that the major correlate with zooplankton community structure during all surveys was sea temperature, accounting for as much as 77% of the variation in community structure. Temperature was indicative of the relative contribution of sub-Antarctic and Antarctic communities, with low temperatures being characterised by an increased predominance of Antarctic communities and vice versa. The differentiation between sub-Antarctic and Antarctic waters, and their respective communities, was particularly pronounced when the SAF and APF were in close proximity to the islands. Surface salinity and sounding had limited effect on community structure. The affect of sounding was intrinsically related to zooplankton interaction with the island ecosystem. Predation by the islands' land based predators and benthic fish appears to decrease zooplankton biomass over the island shelf, particularly the macrozooplankton size fraction. However, this may only be an important factor under conditions of water retention when replenishment of zooplankton stocks is low. The low densities of many macrozooplankton species, and other deep migrators, on the island shelf may be due to their limited advection onto shallow topography. By contrast, there are indications that zooplankton species occurring at shallower depths may be concentrated in the inter-island region by mesoscale flow patterns. Analysis of the population structure of the copepod Calanus simillimus showed that this species occurred at different stages in its life cycle during different years, even though sampling took place in the same calendar months, indicating that there was inter-annual variation in the timing of the biological season. Differences in the population structure of species, and consequently their contribution to abundance and biomass, may therefore have been an important contributor to inter-annual variation in community structure. Evidence is provided for a long-term trend of southward movement of the SAF. This may have a significant affect on the PEI ecosystem, increasing the proportion of allochthonous input and altering the tropho-dynamics of the island ecosystem

- Description: Mesozooplankton community structure in the vicinity of the Prince Edward Islands (PEIs) was investigated during six surveys conducted in late austral summer (April/May), 1996 to 1999. Each year zooplankton samples were collected with a bongo net (300 µm mesh) at stations upstream (west), between and downstream (east) of the islands. Chlorophyll a concentrations were determined fluorometrically, corresponding with each net tow. The positions of the Subantarctic Front (SAF) and the Antarctic Polar Front (APF), in relation to the islands in the upstream region, were determined by a line of CTD and/or XBT stations. Both the SAF and the APF were characterised by a high degree of meridional variation in position. Changes in position of the fronts were shown to occur very rapidly. In 1999 the APF moved southwards by ~40nm and the SAF northwards by ~60nm in a period of two weeks, while in 1996 the SAF appeared to move ~120nm northwards in a two week period. The positions of the SAF and APF appeared to have a significant impact on phytoplankton biomass in the vicinity of the PEIs, through the alteration of local flow dynamics. Microemulsions : a new perspective in the treatment of paediatric and geriatric tuberculosis patients

- Description: Tuberculosis(TB) was declared to be a global emergency in 1993, with South Africa declaring it to be the country’s top health priority in 1996, but ineffective treatment strategies have led to fewer than half of all treated patients in South Africa being cured. At present,paediatric treatment remains a problem, as the antitubercular preparations of rifampicin, isoniazid and pyrazinamide, that are currently available, were not initially designed for the treatment of paediatric TB patients, providing a motivation for this project. The aim of this project is thus the development of a microemulsion dosage form for the oral delivery of RIF(Rifampicin), INH(Isoniazid) and PZA(Pyrazinamide) in combination. RIF, INH and PZA were adequately characterised with reference to the monograph standards referenced and were found to be sufficiently pure to be used in subsequent work. A chromatographic system and conditions were selected and validated as being optimal for HPLC analysis of RIF, INH and PZA in combination, with a drug partitioning method for miglyol 812 developed and validated. Ternary and pseudo-ternary phase diagrams were constructed and reported, all employing miglyol 812 as the lipid. It was undoubtedly the imwitor 308 and crillet 3 combination o/w microemulsion system that proved most successful, maintaining homogeneity on dilution. The microemulsion used in formulation comprised imwitor 308 (27.63%), crillet 3 (27.63%), miglyol 812 23.68%) and water (21.06%). The stability of RIF, INH and PZA was investigated in aqueous solution, miglyol 812, corn oil, 10%m/v cremophor RH, 5%m/v imwitor 308, 10%m/v crillet 3 and 70%m/v sorbitol solution. Trends in the stability assessments conducted on RIF, INH and PZA were noted, with slight variation depending on the formulation component being evaluated. RIF invariably demonstrated temperature and oxidation dependent degradation in all vehicles, with a definite distinction possible between samples stored at 25, 40 and 600C over a 7 day trial period. A definite advantage of storing RIF solutions under nitrogen was observed, with these solutions showing less degradation over the course of the trial, than those stored under air. INH produced a pronounced increase in the degree of degradation of RIF, whereas PZA had a negligible effect on it’s stability. INH proved to be most stable in the 70%m/v sorbitol solution with no significant oxidation or temperature dependent degradation indicated. Temperature dependent degradation was only noticable when INH was in combination with RIF, most significant in crillet 3 solution. PZA was the most stable of the three drugs, remaining relatively unaffected by temperature and the presence of air, independent of the vehicle employed, although the drug remaining did decrease slightly in the presence of RIF.Due to drug dose specifications and solubility limitations, the final formulation assessed, only contained RIF and INH, despite INH and PZA having no significant effect on the stability of each other. The solubility of PZA in the lipid and aqueous components of the microemulsion was not great enough to achieve the required 500 mg/10ml dose, while RIF and INH could achieve the respective 150mg/10ml and 100mg/10ml dose. RIF stability was improved, as anticipated, with the incorporation of RIF into the internal phase decreasing contact with INH which has been shown to affect it’s stability. RIF behaved as predicted, possessing greater stability than shown in the individual formulation components, however, INH did not, being less stable in formulation in the absence of antioxidant, than in it’s presence. A novel microemulsion formulation capable of delivering the incompatible RIF and INH in combination, with numerous microemulsion systems mapped,with the ability of being used for the delivery of other lipophilic drugs and drug combinations, was produced.The final formulation provided valuable information into possible future improvements of the microemulsion to improve drug stability.
