- Description: A new Schiff base probe (QT) consisting of 8-aminoquinoline (Q) and thiophene-2-carboxaldehyde (T) moieties has been synthesized. QT undergoes chelation-enhanced fluorescence quenching when exposed to Hg2+ due to coordination by the sulfur and nitrogen atoms of QT thus forming a facile “turn-off” sensor. The formation of the chelation complex was confirmed by UV–visible absorption and emission spectral measurements, 1H NMR titration and density functional theory calculations. These studies revealed that the probe exhibits high selectivity and sensitivity towards Hg2+ in the presence of other common metal ions. A low detection limit of 23.4 nM was determined and a Job plot confirmed a 2:1 stoichiometry between QT and Hg2+. The potential utility of QT as a sensor for Hg2+ ions in human HeLa cells was determined by confocal fluorescence microscopy, and its suitability for use in the field with environmental samples was tested with Whatman filter paper strips.

- Description: Two readily synthesized Sn(IV) porphyrins (SnP, SnPH) have been prepared with and without the cationic triphenylphosphonium moiety (TPP+), which have high singlet oxygen quantum yields (ca. 0.72) and long triplet state lifetimes. The Sn(IV) porphyrin with a TPP+ moiety (SnPH) exhibits favorable photodynamic activity against MCF-7 cells with IC50 values of 2.9 μM. SnPH exhibits higher cellular uptake than SnP in MCF-7 cells. A Rhodamine 123 (Rh123) assay showed that SnPH targets mitochondria and induces apoptosis by generating reactive oxygen species. The results demonstrate that this structural modification strategy merits further in-depth study.

- Description: In the present work, the performance of aptamer and antibody bioreceptors for the detection of the human epidermal growth factor receptor (HER2) cancer biomarker on a glassy carbon electrode is reported. The carboxylic acid group rich graphene quantum dots (GQDs) modified with gold nanoparticles and a porphyrin binuclear framework (CoP-BNF) were used to modify the glassy carbon electrode. The aptamer and antibody were both amine functionalized and attached to GQDs and CoP-BNF through an amide bond. The designed immunosensors and aptasensors in this work were characterized using electrochemical impedance spectroscopy. The aptasensors, compared to the immunosensors gave better limit of detection values. The aptasensor outperforms the immunosensor in terms of its reusability and storability, while the immunosensor could not be regenerated for subsequent experiments. The potential applicability of all sensors in this work was also investigated, by detection of HER2 in spiked human serum with acceptable results.

- Description: In this study, two cobalt phthalocyanines (asymmetric complex 1) and symmetric (complex 2) were synthesized and conjugated to nitrogen-doped graphene quantum dots (NGQDs) through covalent and non-covalent means to create Co phthalocyanine (CoPc)-NGQDs hybrid systems. The CoPcs and conjugates were applied as electrode modifiers on a glassy carbon electrode in the electrochemical sensing of nitrite. Of the CoPcs alone, complex 1 performed better than complex 2 regarding the limits of detection (LoD) recorded (5.74 μM for 1 and 15.1 μM for 2). Regarding the conjugates/nanocomposites, the π-π stacked conjugate derived from complex 2 (2πNGQDs) demonstrated highly favourable electrochemical potential with an LoD value of 0.70 μM. The nanocomposites fashioned from complex 1 were marred by a reduced loading which rendered the conjugates poor electrochemical sensors. These observations however do not disqualify GQDs as complementary nanomaterials to phthalocyanines but rather shed light on seeking alternative routes to increasing the Pc loading in conjugates (more so in π-π stacked conjugates).

- Description: The work is based on the synthesis and photodynamic antimicrobial chemotherapy (PACT) activities of neutral (1 and 2) and cationic (1Q and 2Q) morpholine substituted phthalocyanines. For applicability, these complexes were covalently linked to modified polyacrylonitrile (PAN) to form 1-PAN, 2-PAN, 1Q-PAN, and 2Q-PAN, respectively. Chitosan was conjugated to PAN (to form PAN-CS) which was then linked to the Pcs to form PAN-CS-1, PAN-CS-2, PAN-CS-1Q, and PAN-CS-2Q, respectively. Singlet oxygen quantum yields improved following the inclusion of chitosan. The PACT activities of the complexes alone and when anchored to both PAN and PAN-CS was evaluated against bacteria: S. aureus, E. coli and fungi C. albicans. Cationic phthalocyanine showed high efficacy values of >7 log reduction value for all microorganisms. These results translated into excellent bacterial colony reduction of >90% against both S. aureus and C. albicans after 1 h of photoirradiation on PAN-CS support.

- Description: A method is presented for the electrochemical detection of the breast cancer biomarker human epidermal growth factor receptor 2 (HER2). A glassy carbon electrode was modified using two techniques known as sequential adsorption and electro-polymerization, and the results are compared. The highly conductive polypyrrole (PPy) is used, in the presence of sulfur/nitrogen doped graphene quantum dots (SNGQDs) and a known cobalt phthalocyanine (CoPc). The different nanomaterials were used as an immobilization platform for the HER2 specific HB5 aptamer via amide linkage. The nanomaterials were arranged in various ways on the glassy carbon electrode, to investigate the effect of the electrode interface on the operational characteristics of a biosensor. The immobilized aptamer selectively recognizes HER2 on the electrode interface, and this leads to an increased charge transfer resistance (Rct) of the electrode when using ferricyanide as the electrochemical probe. The developed immunosensors showed high sensitivity with the best detection limit of 0.00141 ng/mL. The results showed that the method is simple and sensitive enough for the determination of HER2 in serum samples with good reproducibility and accuracy.

- Description: A novel asymmetrical zinc (II) phthalocyanine (Pc) 4 bearing three dimethoxy groups and one carboxyl group was linked to glutathione capped graphene quantum dots (GQDs) by the reaction of carboxylic acid substituent on Pc 4 with the amino group on the GQDs. On the other side, the symmetrical Pc analog 3 was linked to the same nanoparticles through π-π interactions. The as-formed nano-photosensitizers were fully characterized by spectroscopic methods and their photophysicochemical properties were investigated as well. Photodynamic antimicrobial chemotherapy was performed on the planktonic cells of S. aureus strain. And the results show that these nano assemblies were able to completely inhibit the metabolic activity of the resistant bacteria strain S. aureus with a 10.26 log reduction in the viable count. Again, asymmetrical Pc showed higher photocatalytic activity compared to the symmetrical complex with higher kobs and fast initial rates for the former. The photocatalysis obeyed the Langmuir-Hinshelwood kinetic model. The target conjugates showed all the advantages of two different groups existing on a single entity.

- Description: In this study, the photodynamic antimicrobial activities of a series of new tetra-substituted indium phthalocyanine (InPc) complexes are assessed. An aldehyde substituted complex (2) was initially prepared, which was converted through a condensatioreaction to an imine-pyrrolidine substituted complex (3), which in turn was quaternized to form a tetracationic species (4). Favorable photophysicochemical properties were obtained by incorporating a heavy In(III) ion into the central cavity. Aggregation studies revealed that 2–4 remain non-aggregated in DMSO at concentration below 25 µM. The photodeactivation of S.aureus and E.coli was studied. Log reduction values > 9.0 were obtained for cationic InPc 4 after 30 min of incubation and exposure to light for 75 min.

A comparison of molecular modelling with the conventional time dependent—density functional theory (TD-DFT) approach and its simplified sTD-DFT approximation provides further evidence that the latter method accurately reproduces the key trends in the spectral properties, providing colossal savings in computer time for quite large molecules. This demonstrates that it is a valuable tool for guiding the rational design of new phthalocyanines for practical applications.

- Description: The synthesis, photophysicochemical and photodynamic therapy (PDT) activities of benzothiazole substituted zinc phthalocyanine (Pc): 1 (asymmetrically substituted and composed of no charges), 2 (asymmetrically substituted and composed of three positive charges), and 3 (symmetrically substituted and composed of four positive charges), are presented. The triplet and singlet oxygen quantum yields were highest for complex 2 showing the importance of asymmetry and charge. The complexes are covalently and non-covalently linked to B doped detonation nanodiamonds (B@DNDs) to yield nanohybrids (B@DNDs-1, B@DNDs-2, B@DNDs-3). The presence of B@DNDs, asymmetry and positive charge resulted in improved PDT with the lowest cell viability being observed for B@DNDs-2 at 5%. The cell viability ranged from 5% to 7% for the nanohybrids compared to 19–26% for Pcs alone.

- Description: Photodynamic therapy (PDT) is a minimally invasive therapeutic procedure for cancer treatment. This study focuses on the synthesis, photophysicochemical properties, and PDT activity of Sn (IV) porphyrin (2), when linked to biotin decorated nitrogen doped graphene quantum dots (B-NGQDs). The porphyrin complex 2 was conjugated through an ester bond to B-NGQDs to form 2-B-NGQDs. Singlet oxygen quantum yield increased for 2 when linked to B-NGQDs to form 2-B-NQGDs. The dark toxicity and photodynamic therapy studies were conducted for 2, NGQDs and their conjugates using MCF-7 breast cancer cells. The cell viability for dark toxicity of all the compounds was above 90%, and 2-B-NGQDs showed high PDT activity at a concentration of 40 µg/mL with cell viability of 22%.

- Description: A series of A3 type RhIIItriarylcorroles 2a-c with different meso-substituents with differing electron-donating and withdrawing properties and two A2B type RhIIItriarylcorroles 4a-b with meso-methylthiophenyls at the B position have been prepared and characterized. An analysis of structure-property relationships of 2a-c and 4a-b has been carried out by comparing the optical spectroscopy and electrochemistry of the dyes to trends predicted in DFT and TD-DFT calculations. Rational structural modification strategies that enhance the suitability of 2a-c for use as highly efficient catalysts for hydrogen evolution reactions on glassy carbon electrodes coated with 2a-c/reduced graphene oxide composites and of 4a-b in surface-modified Au electrodes are explored.

- Description: Heterogeneous photocatalysis is a promising approach for environmental remediation from contaminants including microorganisms and organic pollutants. In this work, hematite nanofibers are fabricated and modified with a novel monosubstituted Pc (4) as well as an asymmetrical tetrasubstituted Pc (5) with the aim of creating hybrid photocatalysts. The photocatalytic activities of the unmodified and phthalocyanine modified hematite nanofibers were compared based on their efficiencies in the photoinactivation of S. aureus and photooxidation of methyl orange. For both applications, the hybrid nanofibers were found to be more efficient photocatalysts than the unmodified hematite nanofibers. Comparison of the modified nanofibers (4-Fe2O3 and 5-Fe2O3) showed that they have comparable antibacterial activity while the 5-Fe2O3 nanofibers are the best for the photooxidation of methyl orange. The singlet oxygen generation efficiency, high activity, versatility, regenerability and thus reusability of the fabricated hybrid nanofibers makes them ideal candidates for real life water treatment studies.

- Description: Prostate specific antigen (PSA) aptasensors are fabricated using a novel asymmetrically substituted Co phthalocyanine (CoPc), gold nanoparticles (AuNPs) and PSA-specific antigen. The fabricated aptasensors are: GCE-AuNPs-Aptamer, GCE@CoPc-Aptamer and GCE-AuNPs@CoPc-Aptamer (GCE = glassy carbon electrode). The fabricated sensors are characterized at each modification step to monitor the changes occurring at the sensor surface. Concentration studies were carried out using differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) to determine detection limits. All the fabricated aptasensors were found to be highly specific and selective but the GCE-AuNPs@CoPc-Aptamer nanoconjugate performed the best. The aptasensors were also tested in spiked serum samples and detection limits, as well as % recoveries were determined. The results obtained showed that the GCE-AuNPs@CoPc-Aptamer has the potential to be used for clinical studies as the results agree with those obtained for detection of PSA in buffer.

- Description: We report on the facile synthesis of graphene quantum dots (GQDs), nitrogen-doped quantum dots (NGQDs) and graphitic carbon nitride quantum dots (gCNQDs) by the bottom-up hydrothermal synthetic process. The time is varied to study its effects on the structural, hydrodynamic and optical properties of these nanostructures. The techniques used to characterize these nanostructures were transmission electron microscopy, X-ray diffractometry, dynamic light scattering, and zetametry (zeta potential), and by energy dispersive X-ray, UV/vis, fluorescence, Fourier transform infrared, Raman and X-ray photoelectron spectroscopies. For gCNQDs and NGQDs, Raman spectroscopy showed an increase in disorder with synthesis time, indicating introduction of more triazine groups for the former and increase in doping with the N atoms for the latter, hence higher temperatures are recommended. For GQDs, Raman spectra showed an increase in the spatial order of the π-conjugated structure with synthesis time. Considering all the techniques employed in this work, the synthesis times of 6h and 8 h are recommended for GQDs and NGQDs.

- Description: The synthesis and characterization of a novel 1,3-diethyl-2-thiobarbituric-acid-substituted N-confused porphyrin (NCP-TB) is reported, along with a study of its photodynamic activity against MCF-7 cells using 530 (110 mW cm−2) and 660 nm (280 mW cm−2) Thorlabs light-emitting diodes for 30 min. The singlet oxygen quantum yield for NCP-TB is 0.38 compared to 0.23 for the parent unsubstituted N-confused porphyrin (NCP) due to the presence of a sulfur atom. NCP-TB exhibits enhanced PDT activity compared to NCP at both wavelengths. A significantly lower IC50 value of 5.2 μM was obtained at 530 nm (14.7 μM at 660 nm) despite a smaller light dose, due to a large red shift of the intense B band into the green region of the spectrum. 2′,7′-Dichlorofluorescein diacetate (DCFDA) assays demonstrate that there is intracellular generation of reactive oxygen species upon exposure to light.

This work compares the photophysicochemical and PACT activities of InCl 5-p-carboxyphenyl-10-15-20-(tris-4-methylpyridinium)-triiodide porphyrin (1), InCl 5-p-carboxyphenyl-10-15-20-(tris-4-hexylpyridinium)-triiodide porphyrin (2), In (para-aminophenyl) 5,10,15,20-tetrakis(4-methylpyridinium)-tetraiodide porphyrin (3), and In (para-aminophenyl) 5,10,15,20-tetrakis(4-hexylpyridinium)-tetraiodide porphyrin (4). The comparisons were based on the structure and number of charges. Hydrophilic and hydrophobic characters of the amphiphilic porphyrins were studied by comparing the effect of the different alkyl halides (iodomethane and iodohexane) quaternizing agents. The porphyrin complexes were further linked to magnetic nanoparticles to facilitate antimicrobial synergy and recovery of the complexes after use. High Log reduction values ranging from 7.19 to 9.58 were obtained for the quaternized complexes.

- Description: Herein, we present syntheses of three asymmetrical zinc(II) phthalocyanine (ZnPc) complexes and hydrothermal synthesis of cobalt tungstate nanoparticles. The ZnPc derivatives were covalently linked to cobalt tungstate nanoparticles. Photophysical and photochemical behaviour of Pcs and nanoconjugates were investigated. The nanoconjugates generated higher triplet quantum yield and singlet oxygen quantum yield than the ZnPcs alone. Furthermore, in this paper photocatalytic efficiencies of ZnPc complexes and nanoconjugates were investigated for the degradation of methylene blue and higher photocatalytic behaviour was observed for the conjugates. The kinetic model was used to study photocatalytic behaviour of all the prepared photocatalysts.
