- Description: A research was conducted on cotton to test different plant extracts with potential insecticidal properties against bollworms (Noctuidae) and leafhoppers (Cicadellidae) for the use by small-scale, cotton farmers. The study was carried out during the 2006/07 and 2007/08 seasons at ARC Institute for Industrial Crops, Rustenburg (25°39.0 S, 27°14.4 E) in the North West Province of South Africa. Four organic insecticides, tobacco (Nicotiana tabacium), khaki weed (Tagetes minuta), thorn apple (Datura stramonium) and garlic (Allium sativum) were compared with two chemical treatments, Mospilan® (acetamiprid) and Decis® (deltamethrin), and an untreated control. The cultivar, DeltaOPAL, was planted and the trial conducted using standard practices. Overall, Decis® and tobacco treatments exhibited significantly less bollworm larvae than the other treatments. Tobacco was the most promising biological pesticide against American bollworm (Helicoverpa armigera) and spiny bollworm (Earias insulana) larvae. All the treatments were significantly effective against the red bollworm (Diparopsis castanea) larvae, but Decis® and Mospilan® provided the best control. Although khaki weed, garlic and thorn apple were promising biological pesticides against the bollworm complex, tobacco was the most effective organic treatment. Mospilan® seemed to be more effective on the control of leafhoppers (Jacobiella fascialis), followed by Decis® and tobacco. Among the plant extracts, tobacco and garlic gave higher seed cotton yields compared to the khaki weed and thorn apple during both study seasons. In 2006/07, none of the treatments reached the corresponding economic threshold levels (ETLs). In contrast, in 2007/08, only rarely were the treatments (mostly Decis® and Mospilan®) below the corresponding ETLs. These differences were attributed to the higher rainfall recorded during the 2007/08 season, which reduced the effectiveness of the treatments by partly washing away the pesticidal applications. In addition, a high weed infestation also created competition for nutrients in the soil, thus resulting in lower (0.50-1.25 ton/ha in 2007/08 vs 2.50-5.00 ton/ha in 2006/07), but acceptable cotton seed yields. In conclusion, it is suggested that some plant extracts (particularly from tobacco and garlic) can be used as a cheaper and more environment-friendly alternative to chemical insecticides for the control of bollworms and leafhoppers, although it has been demonstrated that their efficacy do not reach the one of chemical treatments. Further research in the near future is recommended.

The formation of thin films of these complexes using different modification methods was also studied. Hexylthio functionalized metallophthalocyanine complexes were immobilized onto gold electrode surfaces using the self-assembly techniques. Surface modifications using cobalt and manganese tetraamino phthalocyanine as polymers, monolayers (onto electrografted surfaces) and as carbon nanotube – metallophthalocyanine conjugates was also carried out. The new method of modifying gold electrodes with metal tetraamino phthalocyanine complexes was investigated. The modified electrode surfaces were studied for their electrocatalytic properties and as potential electrochemical sensors for the detection of hydrogen peroxide (H₂O₂). The limits of detection for the H₂O₂ were of the orders of ~10⁻⁷ M for all the modified electrodes. The modified electrodes gave very good analytical parameters; such as good sensitivity, linearity at studied concentration range and well-defined analytical peaks with increased current densities. The modification methods were reproducible, highly conducting thin films were formed and the modified electrodes were very stable. The design of electrochemical immunosensors for the detection of measles-specific antibodies was also carried out. The modified surface with measles-antigen as sensing element was accomplished using covalent immobilization for an intimate connection of the measles-antigen as a sensing layer onto an electrode surface. Two methods of detecting measles-specific antibodies were investigated and these methods were based on electrochemical impedance, i.e. label-free detection, and voltammetric method using horse-radish peroxidase (HRP) labeled antibody as a reporter. The detection of measles-specific antibodies was accomplished using both these methods. The potential applications of the designed immunosensor were evaluated in real samples (human and newborn calf serum) and the electrodes could detect the antibodies in the complex sample matrix with ease.

The photophysical and photochemical properties and photocatalytic oxidation of cyclohexene properties of the newly synthesised in the presence of gold nanoparticles were investigated. The compounds were stable, well within the stability range for phthalocyanines. The singlet oxygen quantum yield values increased drastically in the presence of gold nanoparticles. The photocatalytic products obtained from the reaction were cyclohexene oxide, 2-cyclohexen-1-ol, 2-cyclohexene-1-one and 1,4-cyclohexanediol. The percentage conversion values, yields and selectivity values improved significantly in the presence of AuNPs. Singlet oxygen was determined to be the main agent involved in the photocatalytic oxidation of cyclohexene. The electrocatalytic oxidation of bisphenol A and p-nitrophenol was carried out using nickel tetraamino phthalocyanine and all the newly synthesised metallophthalocyanine in the presence of gold nanoparticles and single walled carbon nanotubes. The charge transfer behaviour of AuNPs was enhanced in the presence of TaPc, TiPc and VPc complexes. The presence of single walled carbon nanotubes further improved electron transfer and minimised electrode passivation.

Electrospun nanofibers : an alternative sorbent material for solid phase extraction

Three miniaturised electrospun nanofiber based solid phase extraction devices were fabricated. For the first two, 10 mg of electrospun polystyrene fibers were used as a sorbent bed for a micro column SPE device (8 mm bed height in a 200 μl pipette tip) and a disk (I) SPE device (5 mm 1 mm sorbent bed in a 1000 μl SPE barrel). While for the third, 4.6 mg of electrospun nylon nanofibers were used as a sorbent bed for a disk (II) SPE device, (sorbent bed consisting of 5 5 mm 350 μm stacked disks in a 500 μl SPE barrel). Corticosteroids were employed as model analytes for performance evaluation of the fabricated SPE devices. Quantitative recoveries (45.5-124.29 percent) were achieved for all SPE devices at a loading volume of 100 μl and analyte concentration of 500 ng ml-1. Three mathematical models; the Boltzmann, Weibull five parameter and the Sigmoid three parameter were employed to describe the break through profiles of each of the sorbent beds. The micro column SPE device exhibited a breakthrough volume of 1400 μl, and theoretical plates (7.98-9.1) while disk (I) SPE device exhibited 400-500 μl and 1.39-2.82 respectively. Disk (II) SPE device exhibited a breakthrough volume of 200 μl and theoretical plates 0.38-1.15. It was proposed that the formats of future electrospun nanofiber sorbent based SPE devices will be guided by mechanical strength of the polymer. The study classified electrospun polymer fibers into two as polystyrene type (relatively low mechanical strength) and nylon type (relatively high mechanical strength).

There is considerable knowledge and literature available on the process of internationalisation, globalisation and expansion within the services sector and as this information is important to understand the internationalisation process itself, the decision to expand (cross border) presents a number of opportunities and challenges to the hotel industry. Hotels are comprised of a number of divisions and departments, which collectively offer unique services and facilities to various markets. Within this context, careful consideration is required with respect to an organisation‟s overall strategic objectives, the internal structure and how government and cultural influences and differences affect global expansion. Organisational matters such as strategic objectives as well as operational functions such as financial, marketing and sales, human resources and organisational design must also be considered as hotels require synergy within each of these various components in order to function optimally. When international or globalisation opportunities present themselves or are proactively sought, organisations need to ensure that various factors are clearly understood, analysed and identified. These include the most appropriate strategy required, the organisations own internal capacity levels, governmental influences and cultural dynamics. Exploring this relationship this thesis investigates whether the actions of grassroots movements can bring local government in line with their developmental role as accorded to them by the Constitution. The assumption embedded in this question is that the current balance of power at the local level exists outside of the service of the historically disadvantaged. Following on from that, the thesis explores, through different modes of analysis, theoretical and historical, the policy and constitutional framework for local government, and then it unravels the context set by the political economy of South Africa. The aim is to make a significant attempt at understanding the possible implications of the interventions grassroots movements make in the public space. The thesis does this also by looking at the strengths and weaknesses of the strategies of the UDF to makes an assessment of the possible endurance of post-apartheid grassroots movements. Since civil society ‘suffers’ from nationalist politics, with its own corporatist institutions, the thesis searches deep within or arguably 'outside of civil society', subjecting AbM and UPM to academic critique, to see how movements embedded among the poor and carrying the political instrument of anger and marginalisation, can dislodge the power of capital. More importantly, the thesis situates the post-apartheid moment within postcolonial politics; navigating through the legacy of Colonialism of a Special Type (CSP), the thesis explores the limits and opportunities at the disposal of grassroots movements. From a different perspective, the thesis is an examination of the organisation and movement of power and the spaces within which power and ideas are contested. Drawing on the political and economic engagements, dubbed the Dar Es Salaam debates, in the 1970’s and 1980’s spurred on by Issa Shivji, the late Prof Dani Wadada Nabudere and Mahmood Mandani, the conclusions of this thesis develops these engagements, essentially making a case for the continued centrality of the post-apartheid state. However, the thesis also asserts the indubitable role that both grassroots movements and civil society need to play, not necessarily in the democratisation of the state, though that goes without saying, but in taking the post-colonial state on its own terms. Consequently, the thesis puts forward the idea that issue-based mobilisation does exactly this, and in the manner that acknowledges the state’s centrality and makes paramount the self-organisation (popular assemblies) of ordinary people in public affairs. The thesis categorically concludes that the centrality of the post-apartheid state and its progressive outlook (constitutional values) is contingent on organs of popular assemblies which need to take the state on its (progressive) terms.

Empirical modelling of the solar wind influence on Pc3 pulsation activity

- Description: Geomagnetic pulsations are ultra-low frequency (ULF) oscillations of the geomagnetic field that have been observed in the magnetosphere and on the Earth since the 1800’s. In the 1960’s in situ observations of the solar wind suggested that the source of pulsation activity must lie beyond the magnetosphere. In this work the influence of several solar wind plasma and interplanetary magnetic field (IMF) parameters on Pc3 pulsations are studied. Pc3 pulsations are a class of geomagnetic pulsations with frequency ranging between 22 and 100 mHz. A large dataset of solar wind and pulsation measurements is employed to develop two empirical models capable of predicting the Pc3 index (an indication of Pc3 intensity) at one hour and five minute time resolution, respectively. The models are based on artificial neural networks, due to their ability to model highly non-linear interactions between dependent and independent variables. A robust, iterative process is followed to find and rank the set of solar wind input parameters that optimally predict Pc3 activity. According to the parameter selection process the input parameters to the low resolution model (1 hour data) are, in order of importance, solar wind speed, a pair of time-based parameters, dynamic solar wind pressure, and the IMF orientation with respect to the Sun-Earth line (i.e. the cone angle). Input parameters to the high resolution model (5 minute data) are solar wind speed, cone angle, solar wind density and a pair of time-based parameters. Both models accurately predict Pc3 intensity from unseen solar wind data. It is observed that Pc3 activity ceases when the density in the solar wind is very low, even while other conditions are favourable for the generation and propagation of ULF waves. The influence that solar wind density has on Pc3 activity is studied by analysing six years of solar wind and Pc3 measurements at one minute resolution. It is suggested that the pause in Pc3 activity occurs due to two reasons: Firstly, the ULF waves that are generated in the region upstream of the bow shock does not grow efficiently if the solar wind density is very low; and secondly, waves that are generated cannot be convected into the magnetosphere because of the low Mach number of the solar wind plasma due to the decreased density.

- Description: The perception that someone has of an entrepreneurial career ultimately influences his/her decision to pursue such a career path. By investigating employees' perceptions of owning a business, this study provides insights regarding the question of why so few South Africans embark on this path. Entrepreneurship is regarded by many as the solution to South Africa’s employment and economic problems. As such, an understanding of the reason why some people become entrepreneurs and others do not could provide solutions on how to stimulate entrepreneurship among all South Africans, and hopefully increase the levels of entrepreneurship in the country. The purpose of the study was to contribute to the field of entrepreneurship by investigating the perceptions and attitudes that employees have regarding an entrepreneurial career, that is, to ask employees what they think having their own business might be like. By investigating these perceptions, the researcher looked for reasons why many individuals remain in the employment of others rather than embarking on an entrepreneurial career, as well as establishing whether these employees had intentions of starting their own businesses, Therefore the primary objective of this study was twofold, namely to establish the perceptions that employees have regarding self-employment, running their own business or following an entrepreneurial career, and to establish the influence of these perceptions on their entrepreneurial intentions. After conducting a comprehensive literature study and examining the various models of intentions, attitude towards the behaviour was identified as having the strongest influence on entrepreneurial intentions. Attitude towards the behaviour is the extent to which an individual makes a favourable or unfavourable evaluation of the behaviour in question, and additionally is a function of beliefs applicable to the behaviour. Attitude towards the behaviour is closely related to perceived desirability, and “desirability” is a form of value. More specifically, work values have the propensity to significantly predict career choice. For the purpose of this study, the 14 work values identified by Farrington et al. (2011) served as the factors to be investigated in establishing the perceptions of an entrepreneurial career. The independent (work values) and dependent (entrepreneurial intentions) variables investigated in this study were defined and operationalised. Reliable and valid items were sourced from existing studies. Respondents were identified by means of convenience and snowball sampling, and a structured questionnaire was made available to the respondents. The data gathered from the 184 usable questionnaires was subjected to various statistical analyses. The validity and reliability of the measuring instrument were confirmed by means of an exploratory factor analysis and calculating Cronbach-alpha coefficients. Descriptive statistics were calculated to summarise the sample data distribution. Pearson’s moment product correlations were calculated to establish the correlations between the various work values and entrepreneurial intentions. Furthermore, a multiple regression analysis was undertaken to investigate whether relationships existed between the perceptions of an entrepreneurial career in terms of several work values and entrepreneurial intentions. The following work values were identified as influencing the entrepreneurial intentions of employees working in small businesses: - Stimulating; - Stability and advancement; - Flexibility; - Autonomy. The more the work values above were perceived to be experienced when following an entrepreneurial career, the more likely it was that the respondents in this study reported intentions of following such a career. In order to investigate the influence of the various demographic variables on the independent and dependent variables, an Analysis of Variance was performed. In addition, the post-hoc Bonferroni test was done to identify significant differences between the means scores of the various categories within each demographic variable. The practical significance of these differences was assessed by means of calculating Cohen’s d. The results showed a significant positive relationship between the demographic variable Population group and the dependent variable Entrepreneurial v intentions. In addition, relationships were identified between Population group and the intrinsic work values Challenging and Stimulating, the extrinsic-related work values Financial benefit and security and Stability and advancement, and the social-related work value Serving the community. A significant positive relationship was also reported between the demographic variable Entrepreneurial parents and the intrinsic-related work value Stimulating, the extrinsic-related work value Future prospects and the social-related work value Serving the community. The demographic variable Age showed a significant positive relationship with the extrinsic-related work value Financial benefit and security. - Description: Schools are traditionally established as the main meeting platforms for awareness, education, training and capacity building in the local communities, and as such, are highly regarded and respected. They are situated and integrated in nearly all local communities. They are thus perfectly suited to reach, draw on and provide capacity building to the communities. Zambia recently introduced a new curriculum aspect named localised curriculum, which makes up 20% of the curriculum. This curriculum area encourages schools to address issues of sustainable development that are locally relevant through this curriculum component, but it is not clear what kind of learning emerges from this curriculum aspect. The study looked at the shift in pedagogical discourse since the introduction of the localised curriculum. Using case study methodology and observation of a number of classroom activities in the Keemba District near Monze in Zambia, I observed social learning interactions taking place in the implementation of the localised curriculum. In particular, I was interested in how the different aspects of the localised curriculum enabled social learning, at different levels. What is important in this research is that it focused not so much on what people should know, or be able to do, but rather, drawing on some of the questions raised by Wals (2007), I considered questions of how people learn, and how social learning emerges in school-community interactions in the localised curriculum. The main research question was to investigate: How does localised curriculum enable social learning in response to environmental issues in Zambian schools? This research drew on a number of associated research projects that were carried out in southern Africa and beyond. In his research, Namafe (2008) states that he saw a great need to institutionalise increased participation of Zambian Basic Schools and their local communities in sustainable development of the country in order to alleviate poverty and achieve equitable development. Hogan (2008, p. 122 ) researching in a different context, argues that contextualisation of the curriculum allowed for indigenous knowledge to come in to the classroom, stating that ‘Weakening of the frames provided the opportunity of closer relationships to give space for knowledge other than teacher selected knowledge or book knowledge to enter the classroom.’ In this study I found that: - Teaching localised curriculum allowed for contextualisation and integration of environment and natural resource management issues - Localising the curriculum contributed to curriculum relevancy: teaching localised curriculum helped learners, teachers and community to develop concern for quality of life and contributed to an understanding of sustainable development - Localised curriculum allowed for frame awareness, frame deconstruction and reframing to take place: this allowed teachers to use methods which promoted active pupil participation and interaction, encouraging learners to be reflexive thinkers rather than just using rote memory - Participation of teachers, community and learners in the implementation of the localised curriculum: The involvement of community members to teach topics of the localised curriculum was useful and articulated processes of inter–epistemological dialogue between traditional or every day and scientific or institutional knowledge - The localised curriculum implementation involved various roles and was influenced by various factors. Of key interest to this study was the fact that neither parents, learners nor teachers saw the localised curriculum as an opportunity for problem solving, but rather emphasised the learning of practical life skills relevant to the community. This differed from the curriculum manuals, and reduced the environmental education potential and also social learning opportunities.

- Description: Schools are traditionally established as the main meeting platforms for awareness, education, training and capacity building in the local communities, and as such, are highly regarded and respected. They are situated and integrated in nearly all local communities. They are thus perfectly suited to reach, draw on and provide capacity building to the communities. Zambia recently introduced a new curriculum aspect named localised curriculum, which makes up 20% of the curriculum. This curriculum area encourages schools to address issues of sustainable development that are locally relevant through this curriculum component, but it is not clear what kind of learning emerges from this curriculum aspect. The study looked at the shift in pedagogical discourse since the introduction of the localised curriculum. Using case study methodology and observation of a number of classroom activities in the Keemba District near Monze in Zambia, I observed social learning interactions taking place in the implementation of the localised curriculum. In particular, I was interested in how the different aspects of the localised curriculum enabled social learning, at different levels. What is important in this research is that it focused not so much on what people should know, or be able to do, but rather, drawing on some of the questions raised by Wals (2007), I considered questions of how people learn, and how social learning emerges in school-community interactions in the localised curriculum. The main research question was to investigate: How does localised curriculum enable social learning in response to environmental issues in Zambian schools? This research drew on a number of associated research projects that were carried out in southern Africa and beyond. In his research, Namafe (2008) states that he saw a great need to institutionalise increased participation of Zambian Basic Schools and their local communities in sustainable development of the country in order to alleviate poverty and achieve equitable development. Traditional interaction techniques have several shortcomings for interacting with mobile map-based applications. Keypad interaction offers limited control over panning speed and direction. Touch-screen interaction is often a two-handed form of interaction and results in the display being occluded during interaction. Sensor-based interaction provides the potential to address many of these shortcomings, but currently suffers from several limitations. The aim of this research was to propose enhancements to address the shortcomings of sensor-based interaction, with a particular focus on tilt interaction. A comparative study between tilt and keypad interaction was conducted using a prototype mobile map-based application. This user study was conducted in order to identify shortcomings and opportunities for improving tilt interaction techniques in this domain. Several shortcomings, including controllability, mental demand and practicality concerns were highlighted. Several enhanced tilt interaction techniques were proposed to address these shortcomings. These techniques were the use of visual and vibrotactile feedback, attractors, gesture zooming, sensitivity adaptation and dwell-time selection. The results of a comparative user study showed that the proposed techniques achieved several improvements in terms of the problem areas identified earlier. The use of sensor fusion for tilt interaction was compared to an accelerometer-only approach which has been widely applied in existing research. This evaluation was motivated by advances in mobile sensor technology which have led to the widespread adoption of digital compass and gyroscope sensors. The results of a comparative user study between sensor fusion and accelerometer-only implementations of tilt interaction showed several advantages for the use of sensor fusion, particularly in a walking context of use. Modifications to sensitivity adaptation and the use of tilt to perform zooming were also investigated. These modifications were designed to address controllability shortcomings identified in earlier experimental work. The results of a comparison between tilt zooming and Summary gesture zooming indicated that tilt zooming offered better results, both in terms of performance and subjective user ratings. Modifications to the original sensitivity adaptation algorithm were only partly successful. Greater accuracy improvements were achieved for walking tasks, but the use of dynamic dampening factors was found to be confusing. The results of this research were used to propose a framework for mobile tilt interaction. This framework provides an overview of the tilt interaction process and highlights how the enhanced techniques proposed in this research can be integrated into the design of tilt interaction techniques. The framework also proposes an application architecture which was implemented as an Application Programming Interface (API). This API was successfully used in the development of two prototype mobile applications incorporating tilt interaction.

I undertook this research to gain a basic knowledge about critical reading as well as to form a knowledge base that would enable me to teach it effectively. First, I gave students a diagnostic assessment activity, analyzed the result and established the students’ ability to read a text critically. After I had established the students’ strengths, weaknesses, and the requirements to move students from their existing critical reading competency to an improved one, I reviewed the literature to provide me with a theory of teaching critical reading. The lessons were then planned and taught. The lessons; focus group interviews; researcher’s diary; students’ reflections and non-participant observer comments formed the data of the study. The data were analyzed using the framework of “what went well”, “what did not go well” and “what needs to be improved if the lessons were to be repeated”. The main purpose was to address the goals of the research, which were to find effective ways of teaching critical reading, appropriate critical reading materials, and to identify any pedagogic shortcomings. The 30 students who took part in this research were third-year students studying English Second language and Mother tongue pedagogies to become specialists in the teaching of both at Junior Secondary level (grades 8 – 10). The study was guided by constructivist theory, which underpins learner-centred education, which continues to inform and shape the development of curricula in Namibia.

Reassurance must be given to the effect that incorporating a social clause in the WTO legal framework would not be used as a trade restricting mechanism which might have direct, negative effects on the development of countries worldwide. Such an argument may not be won easily from a legal and economic perspective. There is an omnipresent conflict at the international level between the basic values underlying multilateral trade agreements and principles governing the protection of core labour standards. On the one hand, supporting the stance of free trade is the fundamental force of profit maximization while on the other hand, and in direct opposition to this market driven value system, are human rights-based calls for recognising core labour standards in employment matters related to trade. Increased international trade is a powerful tool for tackling poverty and social misery worldwide. It could thus be important to adopt a legal framework in the multilateral trade system to harness potential opportunities a trade-labour linkage could provide. In that case, the legal questions of whether or not there should be a tradeoff between the right to trade and compliance with core labour standards and whether a social clause in the WTO would achieve this purpose had to be addressed. However, without a compatible underpinning legal framework of universally accepted trade-labour standards, incorporating a social clause in the WTO would be a misplaced legal objective which is unachievable as it could lead to a conflict between the WTO and the International Labour Organisation (ILO) and at most could create a legal fiction whose results may not be positively measurable. Setting two international legal norms at conflict with each other is systematically studied as a conflict in which the values of the global market economy are in a supposed confrontation with those protecting core labour standards as human rights. Therefore, this study undertook a contemporary legal analysis of the possibilities and challenges of incorporating a social clause in the WTO for purposes of entrenching the protection of core labour standards. It put forward arguments and tentative proposals for a trade-labour linkage legal framework which could dispel calls for excluding a social clause in the WTO.

- Description: Intrapreneurship expresses organisational activities aimed at increasing quality of products, reducing production cost, capture or creates new product markets, and/or improving processes and services. This research study covers the investigation of potential influences on the intrapreneurial activity within an established organisation, and the possible outcome on financial performance. In the rapidly changing business environment of today, it has become necessary for the organisations to move from boundary-oriented thinking to continuous improvement in order to provide the disruptive competitive advantages necessary to survive and thrive in an environment where the ‘rules of the game’ change quickly in almost all companies and industries (Voelpel, Leibold and Tekie, 2005). Moreover, the automotive industry is no exception to this rule. In general, established companies deal with two significant challenges. First, they have to adapt to the external challenges of constantly changing and developing markets to keep pace with rapid technological evolution, globalisation, and progressively sophisticated competitors (Kemelgor, 2002; Kuratko, Hornsby and Goldsby, 2004). Second, they must deal with the internal challenges of modernising bureaucratic structures and processes, which can lead to slow development, decision-making and an inability to adapt easily to new situations (Hammer and Champy, 1994). In today's context of increasing market globalisation, companies wishing to maintain their competitiveness must innovate constantly (Carrier, 2001). Recognising the importance of meeting these challenges, organisational leaders must create high performance organisations in order to compete in a global mega-economic world. The old ways of doing business as usual and overwhelmed policies and procedures must be rooted out in order for the organisation to compete by identifying and sustaining diversified employees within a global economy (Kennedy, 2010). Companies generally engage in innovation for achieving an increase in quality of products, a reduction in production cost, capture or create new product markets, and reduce the firm’s reliance upon unreliable factors of production (Webster, 2004). There is a growing consensus that established companies must nurture intrapreneurial activity throughout their operations to continue to compete successfully (Sathe, 2003). Numerous authors have suggested intrapreneurship as a method of stimulating innovation and using the creative energy of employees by giving them the resources and independence they need to innovate within the firm (Carrier, 2001; Amo and Kolvereid, 2005). However, there is a certain amount of ambiguity around the concept of intrapreneurship, and this may lead to questions about the difference between intrapreneurship and intrapreneurship (Carrier, 2001). It is therefore important, before introducing the object of this research, to look more closely at the concept on which intrapreneurship is based, and to examine the trends in the research on intrapreneurship. The research conducted by Eesley and Longenecker (2006) suggest that intrapreneurship is a practice of creating new business products and opportunities in an organisation through proactive empowerment and risk-taking. This is considered a key component to organisational success; especially in organisations that operate in rapidly changing industries (Eesley and Longenecker, 2006). Intrapreneurship can manifest itself at every level of the company and regardless of the nature of the position held. Hence, we could have intrapreneurs in technical or non-technical functions; senior, middle or junior management levels; line or staff functions, and manufacturing or service related roles. Beyond this wide perspective, other authors have suggested that intrapreneurship requires a culture built around emotional commitment, autonomy, empowerment, earned respect, and a strong work ethic (Axtell, Holman, Unsworth, Wall, Waterson and Harrington, 2000). They believed intrapreneurship is inseparably connected with leadership, since it involves mobilising teams of people towards a cause much greater than the individuals involved often in the face of significant resistance from status quo preserving forces within and outside the organisation (Seshadri and Tripathy, 2006). Therefore, the failure of organisations to take members inputs on organisational improvement; sanction, promote and encourage risk-taking, empowerment, and improvement actions; give clear organisational direction, priorities, and objectives; and lack of top management support in risk-taking and improvement initiatives, could stifle intrapreneurship (Eesley and Longenecker, 2006). In light of the scope and the group target, intrapreneurship can be considered beneficial for the revitalisation and performance of companies, both large corporations and small and medium-sized enterprises. Previous research conceptualised intrapreneurship in terms of four dimensions that were somewhat distinct in terms of their activities and orientations (Antoncic and Hisrich, 2001): New business venturing; Innovativeness; Self-renewal; and, Proactiveness. The intended contribution of this study is to make use of a self-constructed measuring instrument to demonstrate that intrapreneurship has had a positive influence on corporate performance within an established organisation, in the automotive industry of South Africa. - Description: Intrapreneurship expresses organisational activities aimed at increasing quality of products, reducing production cost, capture or creates new product markets, and/or improving processes and services. This research study covers the investigation of potential influences on the intrapreneurial activity within an established organisation, and the possible outcome on financial performance. In the rapidly changing business environment of today, it has become necessary for the organisations to move from boundary-oriented thinking to continuous improvement in order to provide the disruptive competitive advantages necessary to survive and thrive in an environment where the ‘rules of the game’ change quickly in almost all companies and industries (Voelpel, Leibold and Tekie, 2005). Moreover, the automotive industry is no exception to this rule. In general, established companies deal with two significant challenges. First, they have to adapt to the external challenges of constantly changing and developing markets to keep pace with rapid technological evolution, globalisation, and progressively sophisticated competitors (Kemelgor, 2002; Kuratko, Hornsby and Goldsby, 2004). Second, they must deal with the internal challenges of modernising bureaucratic structures and processes, which can lead to slow development, decision-making and an inability to adapt easily to new situations (Hammer and Champy, 1994). In today's context of increasing market globalisation, companies wishing to maintain their competitiveness must innovate constantly (Carrier, 2001). Recognising the importance of meeting these challenges, organisational leaders must create high performance organisations in order to compete in a global mega-economic world. The old ways of doing business as usual and overwhelmed policies and procedures must be rooted out in order for the organisation to compete by identifying and sustaining diversified employees within a global economy (Kennedy, 2010). Companies generally engage in innovation for achieving an increase in quality of products, a reduction in production cost, capture or create new product markets, and reduce the firm’s reliance upon unreliable factors of production (Webster, 2004). There is a growing consensus that established companies must nurture intrapreneurial activity throughout their operations to continue to compete successfully (Sathe, 2003). Numerous authors have suggested intrapreneurship as a method of stimulating innovation and using the creative energy of employees by giving them the resources and independence they need to innovate within the firm (Carrier, 2001; Amo and Kolvereid, 2005). However, there is a certain amount of ambiguity around the concept of intrapreneurship, and this may lead to questions about the difference between intrapreneurship and intrapreneurship (Carrier, 2001). It is therefore important, before introducing the object of this research, to look more closely at the concept on which intrapreneurship is based, and to examine the trends in the research on intrapreneurship. The research conducted by Eesley and Longenecker (2006) suggest that intrapreneurship is a practice of creating new business products and opportunities in an organisation through proactive empowerment and risk-taking. This is considered a key component to organisational success; especially in organisations that operate in rapidly changing industries (Eesley and Longenecker, 2006). Intrapreneurship can manifest itself at every level of the company and regardless of the nature of the position held. Hence, we could have intrapreneurs in technical or non-technical functions; senior, middle or junior management levels; line or staff functions, and manufacturing or service related roles. Beyond this wide perspective, other authors have suggested that intrapreneurship requires a culture built around emotional commitment, autonomy, empowerment, earned respect, and a strong work ethic (Axtell, Holman, Unsworth, Wall, Waterson and Harrington, 2000). They believed intrapreneurship is inseparably connected with leadership, since it involves mobilising teams of people towards a cause much greater than the individuals involved often in the face of significant resistance from status quo preserving forces within and outside the organisation (Seshadri and Tripathy, 2006). Therefore, the failure of organisations to take members inputs on organisational improvement; sanction, promote and encourage risk-taking, empowerment, and improvement actions; give clear organisational direction, priorities, and objectives; and lack of top management support in risk-taking and improvement initiatives, could stifle intrapreneurship (Eesley and Longenecker, 2006). In light of the scope and the group target, intrapreneurship can be considered beneficial for the revitalisation and performance of companies, both large corporations and small and medium-sized enterprises. Previous research conceptualised intrapreneurship in terms of four dimensions that were somewhat distinct in terms of their activities and orientations (Antoncic and Hisrich, 2001): New business venturing; Innovativeness; Self-renewal; and, Proactiveness. The intended contribution of this study is to make use of a self-constructed measuring instrument to demonstrate that intrapreneurship has had a positive influence on corporate performance within an established organisation, in the automotive industry of South Africa. Enumeration of insect viruses using microscopic and molecular analyses: South African isolate of cryotophlebia leucotreta granulovirus as a case study

- Description: Baculoviruses have been used as biocontrol agents to control insect pests in agriculture since the 1970s. Out of the fifteen virus families known to infect insects, baculoviruses offer the greatest potential as insect biopesticides, due to their high host specificity which makes them extremely safe to humans, other vertebrates, plants and non-target microorganisms. They comprise of two genera: nucleopolyhedroviruses (NPVs) and granuloviruses (GVs). The South African isolate of Cryptophlebia leucotreta granulovirus (CrleGV-SA) which is infectious for the false codling moth (FCM), Thaumatotibia leucotreta, (Meyrick) (Lepidoptera: Tortricidae), has been successfully developed into two commercial biopesticides; Cryptogran® and Cryptex®, for the control of FCM in citrus crops. The current method of enumeration used for CrleGV-SA virus particles in routine experiments during the production of the GV as biopesticides, is dark field microscopy. However, due to the small size of GVs (300-500 nm in length), the technique is not easy to perform on these viruses, and no systemic comparison has been made of potential alternative methods. Therefore, the main objective of this study was to develop a quantitative enumeration method for CrleGV-SA occlusion bodies (OBs) which is accurate, reliable, and feasible, and compare the developed methods of enumeration to the current method. Purified and semi-purified CrleGV-SA viral stocks were prepared for enumeration studies using spectrophotometry, dark field microscopy, scanning electron microscopy (SEM) and real time qPCR. Spectrophotometry was found to be an unreliable method for enumeration of GVs in the production, standardisation, and quality control of biopesticides. Dark field microscopy and SEM were found to be accurate, and statistically comparable (p = 0.064) enumeration techniques. qPCR is currently being optimised for the enumeration of GVs. This technique was demonstrated to generate accurate standard curves for absolute quantification of virus particles for pure and semi-pure virus preparations. qPCR offers the greatest potential as an accurate enumeration method because it is not affected by contamination with non-biological contaminating debris, nor by other biological material due to the specificity of PCR primers. Further work is required to fully develop qPCR as an enumeration method for GVs. However, dark field microscopy has been successfully validated as an enumeration method. SEM, which has a high resolution compared to light microscopy, has an added advantage over dark field microscopy, which is to distinguish virus particles in semi-pure viral stock preparations during counting. Therefore, SEM currently provides the most unambiguous and feasible enumeration method for GVs in both purified and semi-purified virus samples.

Environmental cues and sensory preferences directing the nesting process in loggerhead turtles, caretta caretta, nesting in Maputaland, South Africa

- Description: Animals use their senses for everything on an immediate, and day to day fashion – detecting danger, finding food, finding mates among other activities. In sea turtles and other migratory species senses are used for long-distance migrations. Senses such as smell, vision and hearing, have been studied experimentally under laboratory conditions but seldom have been investigated in the field. This thesis takes a combination of field and laboratory experiments and investigates some of the hypotheses involved in natal homing and nest site selection. The loggerhead’s nesting distribution in Maputaland is distinct with characteristic high and low density nesting areas which are consistent from year to year. Investigations by earlier researchers on these beaches suggested that beach characteristics, such as beach morphodynamic type and beach width, do not appear to influence the beaches at which loggerheads emerge to nest. The high density nesting area (with approximately 440 loggerhead emergences/km) have similar beach characteristics as the low density nesting area (with <50 loggerhead emergences/km). It is therefore suggested that there is another cue that drives nest site selection. It can either be related to a physical characteristic not yet realised, or is a non-physical (but chemical or biological) cue. This dissertation aims to identify the sensory inputs received during the nest selection process, as well as sea-finding ability after nesting. To investigate the mechanism causing the high-density as opposed to the low-density nesting area, three potential drivers were investigated namely: chemical imprinting (as a natal beach cue), ambient and artificial light (as deterrents) and social facilitation (as a learned behaviour). It was also attempted to identify the strength of the most common senses – vision, hearing and smell. As animal ethics restricts interfering with emerging or nesting turtles, the strengths of these senses were tested during sea-finding by adult loggerheads. The results indicate that sulfide concentrations appear to be used as chemical cues for nesting as these concentrations are elevated (>150 percent) in the high density nesting beaches compared to the low density nesting beaches within and among seasons however further investigations are required. Artificial light (range: 0.045–0.5 lux) is an active deterrent of female emergences while ambient light, even under extreme conditions such as lightning during electric storms (up to 8.2 lux), appears to have no observable influence on the spatial or temporal distributions of emergences. Social facilitation appears unlikely as a primary nest site selection factor for loggerheads. It may however, play a minor secondary role to preferred areas or hotspots. Sea-finding in post-nesting female loggerheads appears to be driven exclusively by visual cues such as the light horizon, with minimal to no influence from other cues (the sound of the breakers, slope or smell of the ocean) which solidify the visual system’s use in sea-finding. This research on the nest site selection of loggerheads and the sensory systems involved in this process has added valuable information to the limited pool of knowledge already present and has created a solid framework on which further investigations can be based. Future work in this field should focus on integrating a suite of sensory stimuli and cues to receive a greater understanding of the sensory systems used in nest site selection.
