- Description: In the present study, silver nanoparticles (AgNPs) were synthesized using aqueous leaf extracts of three Congolese plant species, namely Brillantaisia patula (BR-PA), Crossopteryx febrifuga (CR-FE) and Senna siamea (SE-SI). The obtained AgNPs were studied for their optical, structural, surface morphological and antibacterial properties. The prepared AgNPs were characterized by using UV-Visible spectra, Transmission Electron Microscopy (TEM), Fourier Transform Infrared Spectroscopy (FTIR), X-ray spectroscopy (EDX) and X-ray diffractometer (XRD). The synthesized nanoparticles were spherical shaped and well-dispersed with average sizes ranging from 45 to 110 nm. The AgNPs derived from BR-PA, CR-FE and SE-SI exhibited higher antibacterial activity against three bacterial pathogens of the human skin compared to their respective crude extracts and AgNO3. This indicated that the biomolecules covering the nanoparticles may enhance the biological activity of metal nanoparticles. Hence, our results support that biogenic synthesis of AgNPs from Congolese plants constitutes a potential area of interest for the therapeutic management of microbial diseases such as infectious skin diseases.

- Description: Pereskia aculeata Miller (Cactaceae) is an invasive alien cactus which has negative impacts on indigenous plant biodiversity in South Africa. Catorhintha schaffneri Barilovsky and Garcia (Coreidae), the pereskia stem-wilter, was collected at coastal sites in the subtropical region of Santa Catarina state in southern Brazil and released as a biological control agent to control P. aculeata, in South Africa, in 2014. The introduced distribution of P. aculeata covers a wide variety of climatic conditions which was expected to influence the establishment success of the new agent. The thermal tolerance of C. schaffneri was investigated by developing a degree-day model and calculating the thermal limits of the species. The influence of humidity on egg hatchability and adult survival was also investigated. These data were then used to determine where the agent was likely to establish in South Africa using both weather station and microclimate temperature data. To ground-truth these predictions, sixteen release sites were selected covering a wide range of climatic conditions in the introduced distribution. Three releases of 30 adult C. schaffneri were conducted at each site and population establishment was recorded. Field establishment was recorded at only two of the sixteen experimental release sites. Low winter temperatures were predicted to prevent establishment at seven of the sixteen sites, but nine sites were considered climatically suitable according to the agent’s thermal physiology. Low rainfall and humidity could explain why the agent did not survive at some sites with suitable thermal climates. Many of the thermally suitable sites were affected by a severe drought over the course of the experiment, so further releases during periods with average rainfall and humidity are warranted and could confirm whether the drought was a significant factor influencing the failure of the agent to establish. Although climatic-matching and thermal physiology studies are valuable for tentative predictions of establishment success, there are numerous variables involved that require ground-truthing. Releasing the agent over a wide range of climatic zones believed to be within the thermal limits of the agent, and following releases with detailed post-release evaluations may be the best method of determining where future releases should be focused.

- Description: Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is native to South America but has invaded the Afro-Eurasian supercontinent where it is currently the most devastating invasive arthropod pest of tomato. As a part of the first classical biological control programme against T. absoluta in Africa, a larval parasitoid, Dolichogenidea gelechiidivoris Marsh. (Syn.: Apanteles gelechiidivoris Marsh) (Hymenoptera: Braconidae), of T. absoluta was imported from Peru into the quarantine facility of the International Centre of Insect Physiology and Ecology, Kenya. We report on the host larval preference of D. gelechiidivoris and the host suitability, and the parasitoid’s reproductive strategy, including lifetime fecundity and egg maturation dynamics. Dolichogenidea gelechiidivoris females preferentially oviposited in early (1st and 2nd) larval instars of T. absoluta but parasitized and completed development in all four instars of the host. Host instar did not affect D. gelechiidivoris sex-ratio but females reared on the first instar had significantly fewer eggs than when reared in late larval instars (3rd and 4th). Females of the parasitoid emerged with a high mature egg load which peaked 2 d post eclosion. The females of D. gelechiidivoris survived 8.51 ± 0.65 d and produced 103 ± 8 offspring per female at 26 ± 4 °C (range: 24 to 29 °C) and 50–70% relative humidity (RH) with males present and fed honey-water (80% honey). Increasing maternal age decreased the proportion of female offspring. Under the aforementioned laboratory conditions, the Gross and Net reproductive rates were 72 and 39.5 respectively, while the mean generation time was 20 d. The potential intrinsic rate of natural increase was 0.18. This study shows that D. gelechiidivoris is a potential biological control agent of T. absoluta and should be considered for release in Kenya and across Africa following host specificity testing and risk assessments.

- Description: The degradation of rifampicin (RIF) in an acidic medium to form 3-formyl rifamycin SV, a poorly absorbed compound, is accelerated in the presence of isoniazid, contributing to the poor bioavailability of rifampicin. This manuscript presents a novel approach in which isoniazid is formulated into gastric-resistant sustained-release microspheres and RIF into microporous floating sustained-release microspheres to reduce the potential for interaction between RIF and isoniazid (INH) in an acidic environment. Hydroxypropyl methylcellulose acetate succinate and Eudragit® L100 polymers were used for the manufacture of isoniazid-loaded gastric-resistant sustained-release microspheres using an o/o solvent emulsification evaporation approach. Microporous floating sustained-release microspheres for the delivery of rifampicin in the stomach were manufactured using emulsification and a diffusion/evaporation process. The design of experiments was used to evaluate the impact of input variables on predefined responses or quality attributes of the microspheres. The percent degradation of rifampicin following 12 h dissolution testing in 0.1 M HCl pH 1.2 in the presence of isoniazid gastric-resistant sustained-release microspheres was only 4.44%. These results indicate that the degradation of rifampicin in the presence of isoniazid in acidic media can be reduced by encapsulation of both active pharmaceutical ingredients to ensure release in different segments of the gastrointestinal tract, potentially improving the bioavailability of rifampicin.

- Description: As part of an ongoing study of natural products from local medicinal plants, the methanol extract of stem bark of Rauvolfia caffra Sond was investigated for biological activity. Column chromatography and preparative thin-layer chromatography were used to isolate lupeol (1), raucaffricine (2), N-methylsarpagine (3), and spegatrine (4). The crude extract, fractions and isolated compounds were tested for anti-oxidant, antitrypanosomal and anti-proliferation activities. Two fractions displayed high DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging activity and reducing power with IC50 (The half maximal inhibitory concentration) and IC0.5 values of 0.022 ± 0.003 mg/mL and 0.036 ± 0.007 mg/mL, and 0.518 ± 0.044 mg/mL and 1.076 ± 0.136 mg/mL, respectively. Spegatrine (4) was identified as the main antioxidant compound in R. caffra with IC50 and IC0.5 values of 0.119 ± 0.067 mg/mL and 0.712 ± 0 mg/mL, respectively. One fraction displayed high antitrypanosomal activity with an IC50 value of 18.50 μg/mL. However, the major constituent of this fraction, raucaffricine (2), was not active. The crude extract, fractions and pure compounds did not display any cytotoxic effect at a concentration of 50 μg/mL against HeLa cells. This study shows directions for further in vitro studies on the antioxidant and antitrypanosomal activities of Rauvolfia caffra Sond.

- Description: Lycium ferocissimum Miers (Solanaceae) is an indigenous shrub in South Africa but has become invasive in several countries including Australia, where chemical and mechanical control methods have proved costly and unsustainable. In Australia, biological control is being considered as a management option, but the herbivorous insects associated with the plant in its native range are not well known. The aim of this study was to survey the phytophagous insects associated with L. ferocissimum in South Africa and prioritise promising biological control agents. In South Africa, the plant occurs in two geographically distinct areas, the Eastern and Western Cape provinces. Surveys for phytophagous insects on L. ferocissimum were carried out repeatedly over a two-year period in these two regions. The number of insect species found in the Eastern Cape Province (55) was higher than that in the WesternCapeProvince (41), but insect diversity based on Shannon indices was highest in the Western Cape Province. Indicator species analysis revealed eight insect herbivore species driving the differences in the herbivore communities between the two provinces. Based on insect distribution, abundance, feeding preference and available literature, three species were prioritised as potential biological control agents. These include the leaf-chewing beetles Cassida distinguenda Spaeth (Chrysomelidae) and Cleta eckloni Mulsant (Coccinellidae) and the leaf-mining weevil Neoplatygaster serietuberculata Gyllenhal (Curculionidae).

- Description: At a recent academic conference in the South, nine of us grappled for four days with these “old” questions and their presence within the “new” discourse of “post” environmental education research. We struggled for an additional six-month period of email exchange to see and feel these questions in our own research. That slow, rich, and deeper academic exchange between us culminated in a research agenda, partially (re)presented in a collectively constructed Mindmap (Figure 1), for critiquing the post/new whose framing is described in the remainder of this Introduction to the politics of knowledge production. That politic of slowly and judiciously engaging a collective form of criticism culminates in identifying the research problem, and questions, of this Special Issue (SI) about the role and place of allegedly new theory in the global discourse of allegedly post environmental education research. This specially assembled issue of The Journal of Environmental Education (JEE) is our best effort to (partially) represent a considerable amount of thought about the challenge presented by “post” and “new” Western thought. In translating our collective thought processes to a SI, we anticipate the reflexivity of the field will be critically advanced through engaging a number of emerging debates (Robottom and Hart, 1993) identified in the following pages of this Introduction, and in three “sample” articles specially written by Isabel Carvalho, Carlos Steil and Francisco Abraão Gonzaga, Louise Sund and Karen Pashby, and Phillip Payne, and an “in process” Conclusion written by Cae Rodrigues. There remains much work to do. This SI is only a start of reengaging overdue debates about the post (Hart, 2005).

The complexes were characterized by a number of techniques including UV–Vis, mass, and infrared spectra, as well as elemental analysis. The glassy carbon electrodes were first azide functionalized then clicked to low symmetry phthalocyanines. The click reaction was confirmed using X-ray photoelectron spectra. The constructed electrodes showed excellent electrocatalytic activity towards hydrazine oxidation. Oxidation peaks with low potentials of 0.21 V and 0.26 V, for complexes 3 and 5, respectively were obtained. Complex-5 gave a better detection limit of 0.94 μM and electrocatalytic rate constant of 5.6 × 106 M−1 s−1.

- Description: In the geological record, fossil phosphatic stromatolites date back to the Great Oxidation Event in the Paleoproterozoic, but living phosphatic stromatolites have not been described previously. Here, we report on cyanobacterial stromatolites in a supratidal freshwater environment at Cape Recife, South African southern coast, precipitating Ca carbonate alternating with episodes of Ca phosphate deposition. In their structure and composition, the living stromatolites from Cape Recife closely resemble their fossilized analogues, showing phosphatic zonation, microbial casts, tunnel structures and phosphatic crusts of biogenic origin. The microbial communities appear to be also similar to those proposed to have formed fossil phosphatic stromatolites. Phosphatic domains in the material from Cape Recife are spatially and texturally associated with carbonate precipitates, but form distinct entities separated by sharp boundaries. Electron Probe Micro-Analysis shows that Ca/P ratios and the overall chemical compositions of phosphatic precipitates are in the range of octacalcium phosphate, amorphous tricalcium phosphate and apatite. The coincidence in time of the emergence of phosphatic stromatolites in the fossil record with a major episode of atmospheric oxidation led to the assumption that at times of increased oxygen release the underlying increased biological production may have been linked to elevated phosphorus availability. The stromatolites at Cape Recife, however, form in an environment where ambient phosphorus concentrations do not exceed 0.28μM, one to two orders of magnitude below the previously predicted minimum thresh-old of >5 μM for biogenic phosphate precipitation in paleo-systems. Accordingly, we contest the previously proposed suitability of phosphatic stromatolites as a proxy for high ambient phosphate concentrations in supratidal to shallow ocean settings in earth history.

- Description: The descriptive phrase that comprises my title can refer both to a pervasive comic trope and a mode of self-expression. There is a tension here, insofar as the comic trope leads us not to take cross-dressing, or drag, seriously. The first half of the comic film Some Like It Hot (1959), with its cross-gender plot and its (sophisticated but) straightforward use of the comic trope of men-in-women’s clothes, appears to fall foul of this tension and to be susceptible to criticism in this regard. However, the film rectifies itself, portraying the cross-dressing relationship which develops through the second half of the film as a potentially meaningful one for both partners. In this article, I interpret the film as inviting its viewers to adopt a (particular kind of) skeptical ironic (that is, Pyrrhonian) attitude toward gender-presentation practices. While the film in no way attempts to discourage us from participating in such practices, it does invite us—through our partiality toward the characters Osgood and Jerry/Daphne, as we follow their budding, transgressive relationship—to acknowledge that a violation of gender-presentation practices can be a meaningful feature of sincere relationships.

Metabarcoding Analysis of Bacterial Communities Associated with Media Grow Bed Zones in an Aquaponic System

- Description: The development of environmentally sustainable plant and fish production in aquaponic systems requires a complete understanding of the systems’ biological components. In order to better understand the role of microorganisms in this association, we studied the bacterial communities in the dry, root, and mineralized zones of a flood-and-drain media bed aquaponic system. Bacterial communities were characterized using metabarcoding of the V3-V4 16S rRNA regions obtained from paired-end Illumina MiSeq reads. Proteobacteria, Actinobacteria, and Bacteroidetes accounted for more than 90% of the total community in the dry zone and the effluent water. These phyla also accounted for more than 68% of the total community in the root and mineralized zones. The genera Massilia, Mucilaginibacter, Mizugakiibacter, and Rhodoluna were most dominant in the dry, root, and mineralized zones and in the effluent water, respectively. The number of shared operational taxonomic units (OTUs) for the three zones was 241, representing 7.15% of the total observed OTUs. The number of unique OTUs in samples from dry zone, root zone, mineralized zone, and effluent water was 485, 638, 445, and 383, respectively. The samples from the root zone harbored more diverse communities than either the dry or mineralized zones. This study is the first to report on the bacterial community within the zones of a flood-and-drain media bed. Thus, this information will potentially accelerate studies on other microbial communities involved in the bioconversion of nitrogen compounds and mineralization within these types of aquaponic systems.

- Description: Microbial mats were the dominant habitat type in shallow marine environments between the Palaeoarchean and Phanerozoic. Many of these (termed ‘microbialites’) calcified as they grew but such lithified mats are rare along modern coasts for reasons such as unsuitable water chemistry, destructive metazoan influences and competition with other reef-builders such as corals or macroalgae. Nonetheless, extant microbialites occur in unique coastal ecosystems such as the Exuma Cays, Bahamas or Lake Clifton and Hamelin Pool, Australia, where limitations such as calcium carbonate availability or destructive bioturbation are diminished. Along the coast of South Africa, extensive distributions of living microbialites (including layered stromatolites) have been discovered and described since the early 2000s. Unlike the Bahamian and Australian ecosystems, the South African microbialites form exclusively in the supratidal coastal zone at the convergence of emergent groundwater seepage. Similar systems were documented subsequently in southwestern Australia, Northern Ireland and the Scottish Hebrides, as recently as 2018, revealing that supratidal microbialites have a global distribution. This review uses the best-studied formations to contextualise formative drivers and processes of these supratidal ecosystems and highlight their geological, ecological and societal relevance.

- Description: Background: As science and modern technology have brought many advances, we have also come to overshoot planetary boundaries, while still falling short of development goals to eradicate poverty and inequality. A growing recognition of the complexity of development problems and contexts calls for new framings, including a new approach to monitoring and evaluation (M and E) as one of the mechanisms by which modern societies aim to steer towards a more sustainable future. New approaches to M and E mean new skills for the M and E practitioner. Objectives: This article proposed a framing for M and E skills, comprising of technical, relational and transformational (T-R-T) competences. Method: Adapted from the literature, this competence framework was tested in a broader learning needs assessment and then applied retrospectively to author’s experience in developmental evaluations in complex social–ecological contexts in southern Africa. Results: The emerging insights were that not only technical competence is needed, but also relational competence that goes beyond interpersonal skills, to enable the production and uptake of evaluation findings. In addition, the limitations of mainstream M and E methods in the face of complexity seemed to create a need for ‘transformational’ competence, which included evaluators’ ability to develop credible M and E alternatives. Conclusion: The T-R-T framework helped to advance the notions of ‘hard’ and ‘soft’ skills and expanded on existing M and E competence frameworks. Recommendations included a call for innovative educational and professional development approaches to develop relational and transformational competencies, in addition to training for technical competence.

- Description: In this article we contribute to critical risk approaches to studying pregnancy and childbirth in the global South. Following Sarah Rudrum’s work, our approach focusses on sociocultural inequalities amid the regulation of individuals. We draw on data from our Foucauldian-inspired ethnography of two antenatal clinics in an under-resourced area of South Africa to illustrate how multi-layered risk management operates in these spaces. These data were collected over a period of six months in the form of semi-structured interviews, observations of consultations and waiting room interactions, documents used in the clinic, and posters appearing on the clinic walls. Our findings show how a scientific-bureaucratic approach to pregnancy risk management, as encoded in international, national and institutional guidelines, is well known, highly visible, and practised through surveillance and reporting mechanisms in clinics. This approach incites healthcare practitioners to achieve particular performance standards and to monitor their professional agency. Managing pregnancy risk thus entails regulating the healthcare practitioners themselves. In implementing approved pregnancy risk management strategies in an over-subscribed and under-resourced public healthcare setting, however, healthcare practitioners face potential risk to their professional reputation and integrity. In managing this risk, they resist the scientific-bureaucratic approach through: depicting themselves as victims of unfair institutional arrangements or unreasonable patients; instituting street-level bureaucracy to control access to the clinics; and controlling patients’ actions in authoritarian ways. Our research shows that without engagement with the on-the-ground realities of the antenatal clinic in resource-poor environments, a scientific-bureaucratic approach to pregnancy risk management is inevitably limited in its effectiveness.

- Description: The potato (Solanum tuberosum L.) is an important tuber crop with high dietary value that could potentially help to alleviate malnutrition and hunger in Africa. However, production is expensive, with high fertilizer and pesticide demands that lead to environmental pollution, and tillage practices that negatively affect soil structure. Microorganisms of different types have increasingly been found to be useful as biofertilizers, and arbuscular mycorrhizal (AM) fungi are an important crop symbiont. AM fungi have been shown to increase tolerance of crop plants to drought, salinity and disease by facilitating water and nutrient acquisition and by improving overall soil structure. However, the establishment and maintenance of the symbioses are greatly affected by agricultural practices. Here, we review the benefits that AM fungi confer in potato production, discuss the role and importance of mycorrhiza helper bacteria, and focus on how AM fungal diversity and abundance can be affected by conventional agricultural practices, such as those used in potato production. We suggest approaches for maintaining AM fungal abundance in potato production by highlighting the potential of conservation tillage practices augmented with cover crops and crop rotations. An approach that balances weed control, nutrient provision, and AM fungal helper bacterial populations, whilst promoting functional AM fungal populations for varying potato genotypes, will stimulate efficient mycorrhizal interventions.

- Description: The current COVID-19 pandemic has tested the resolve of the global community with more than 35 million infections worldwide and numbers increasing with no cure or vaccine available to date. Nanomedicines have an advantage of providing enhanced permeability and retention and have been extensively studied as targeted drug delivery strategies for the treatment of different disease. The role of monocytes, erythrocytes, thrombocytes, and macrophages in diseases, including infectious and inflammatory diseases, cancer, and atherosclerosis, are better understood and have resulted in improved strategies for targeting and in some instances mimicking these cell types to improve therapeutic outcomes. Consequently, these primary cell types can be exploited for the purposes of serving as a "Trojan horse" for targeted delivery to identified organs and sites of inflammation. State of the art and potential utilization of nanocarriers such as nanospheres/nanocapsules, nanocrystals, liposomes, solid lipid nanoparticles/nano-structured lipid carriers, dendrimers, and nanosponges for biomimicry and/or targeted delivery of bioactives to cells are reported herein and their potential use in the treatment of COVID-19 infections discussed. Physicochemical properties, viz., hydrophilicity, particle shape, surface charge, composition, concentration, the use of different target-specific ligands on the surface of carriers, and the impact on carrier efficacy and specificity are also discussed.

- Description: The synthesis of new Schiff bases and their dinuclear boron complexes is described, along with their characterization by 1H and 13C NMR, FT-IR, and UV–visible absorption spectroscopy, mass spectrometry, and EDX for elemental analysis. The optical and photophysical properties were examined in terms of their absorption and emission behavior, fluorescence quantum yields and fluorescence lifetimes. The flexible dinuclear boron complexes that are linked by a flexible carbon chain exhibited large Stokes shifts in the range from 92 nm to 115 nm in contrast to BODIPY dyes. Those properties make these complexes precious for applications in fluorescence materials. And also theoretical calculations were obtained by using Density Functional Theory (DFT) methods.

- Description: Two near-infrared (NIR) absorbing di(thien-2-nyl)-di(dimethylanilino)azaBODIPY dyes 2a and 2b were synthesized and characterized that differ depending on whether the dimethylaniline substituents are introduced at the 3,5- or 1,7-positions of the azaBODIPY core. The main spectral bands lie at 824 and 790 nm, respectively, in CH2Cl2. The effect of substituent position on the photophysical and pH sensing properties was analyzed through a comparison of the optical properties with the results of time-dependent density functional theory (TD-DFT) calculations. Protonation of the dimethylamino nitrogen atoms eliminates the intramolecular charge transfer properties of these compounds, and this results in a marked blue-shift of the main absorption bands to 696 and 730 nm, respectively, in CH2Cl2, and a fluorescence “turn-on” effect in the NIR region. The pH dependence studies reveal that the pKa values of the non-protonated 2a and 2b molecules are ca. 6.9 (±0.05) and 7.3 (±0.05), respectively, while that of the monoprotonated species for both dyes is ca. 1.4 (±0.05) making them potentially suitable for use as colorimetric pH indicators under highly acidic conditions.

- Description: In this article, we report on the linking of cobalt tetrakis (4-pentyn-oxy) phthalocyanine (CoTPPc) to ZnO nanoparticles via click chemistry. Subsequently, electrocatalytic activity and nonlinear optical properties were investigated (the latter using an open Z-scan technique at 532 nm). The linking of CoTPPc with ZnO resulted in the lowest limiting intensity value of 0.27 J.Cm−2, the βeff (cmW−1) values were found to be 1.51 × 10-8 and 7.10 × 104 for ZnO and CoTPPc-ZnO respectively. The catalytic rate constants (M−1s−1) (and limits of detection) were 4.1 × 104 (12.87 µM), 5.7 × 104 (8.62 µM) and 7.36 × 104 (4.35 µM) for ZnO, CoTPPc and CoTPPc-ZnO. Hence linking ZnO nanoparticles to CoTPPc result in the enhancement of both nonlinear optical behaviour and catalytic activity.
