Distribution and habitat preferences of marine megafauna in Nosy Be, Madagascar

- Description: Marine megafauna have important ecological roles including the top-down regulation of lower trophic levels and the transport of nutrients. They are also charismatic species of socioeconomic importance, due to their public appeal and focus for ecotourism. However, these taxa face numerous anthropogenic threats including bycatch, habitat-loss, noise disturbance, prey reductions, pollution, and vessel traffic. Northwest Madagascar has been identified as an important marine mammal area and whale shark aggregation site, yet limited information on megafauna within Madagascar exists. In this thesis, I investigated the community structure, distribution trends, related environmental factors (Chapter 2) and predicted habitat suitability (Chapter 3) using a species distribution modelling approach for marine megafauna around Nosy Be, North-west Madagascar. Data collection consisted of opportunistic boatbased surveys conducted while looking for whale sharks, mobulid rays and cetaceans. These activities were performed by dedicated ‘swim with whale shark’ tourism operators, Les Baleines Rand'eau and Safari Baleine during tourism activities directed by the Madagascar Whale Shark Project over four years (2016-2019). A total of 1792 sightings were reported with 13 identified species, including elasmobranchs: whale shark, Rhincodon typus; spinetail mobula ray, Mobula japonica; giant oceanic manta ray, Manta birostris; shortfin devil ray, Mobula kuhlii; balaenids: humpback whale, Megaptera novaeangliae; Omura's whale, Balaenoptera omurai, delphinids: Indo-Pacific bottlenose dolphin, Tursiops aduncus; spinner dolphin, Stenella longirostris; pantropical spotted dolphin, Stenella attenuata; Indian ocean humpback dolphin, Sousa plumbea; false killer whale, Pseudorca crassidens; melonheaded whale, Peponocephala electra and one cheloniid species: leatherback sea turtle, Dermochelys coriacea. Sightings were spatially analysed at a community level according to feeding guilds (i.e., filter feeders vs. predators), and distribution of the megafauna groups v were compared to selected remotely sensed physico-chemical data (sea surface temperature and chlorophyll-a concentration) and bathymetry charts (depth and slope) using generalized linear models (Chapter 2). All four variables had a significant effect on filter feeder distribution. Spatial distribution of filter feeder sightings peaked at an area of steep underwater topography (18-50m; x slope=0.5%) located on the west coast of Nosy Be, where foraging aggregations of large filter feeders (primarily R. typus and B. omurai) were frequently observed. Only SST had a significant effect on the relative abundance of animals in the predator feeding guild, which demonstrated a more longitudinal distribution along the continental shelf. The maximum entropy model (Maxent) was used for predicting habitat preference for the most frequently sighted species (Chapter 3). The distribution of whale sharks, mobulid rays and Omura’s whales were significantly overlapped as they were abundant in two main hotspots, Grand banc de l’entrée (~10 km of the west coast) and near Nosy Mitsio island, (~50 km northeast of Nosy Be). Suitable habitat for the dolphins was in relatively shallow waters in temperatures <28ºC near areas of steep changes in bathymetry. Bottlenose and spinner dolphins had a more widespread distribution across the continental shelf and humpback dolphin was closely associated with inshore reefs and occur in waters <10m. The coastal and inshore region of Nosy Be is an important habitat for a variety of megafaunal species, and the continental shelf provided essential areas for feeding and breeding. The results were similar to those presented for these species in other regions of the Indian Ocean (i.e., Eastern and Southern Africa; Western Australia; Seychelles) as well as outside of the Indian Ocean (i.e., Indonesia; Caribbean; North Atlantic). Thus the results from this thesis could be considered for management recommendations for marine conservation in the region. Evaluating the potential effects of microplastics at environmentally realistic concentrations in South African freshwater systems

- Description: Microplastic pollution is spatially broad, microplastics can be found in various habitats including freshwater systems. Microplastic exposure to aquatic organisms has been associated with several physical impacts on aquatic organisms from multiple trophic levels such as; increased immune response, a decrease in food intake, excessive loss of weight, reduced growth rate, reduced energy and adverse effects on successive generations. However, these significant effects of microplastics exposure have been observed mostly in studies that use concentrations of microplastics that exceed environmental concentrations. Therefore, there is an overall lack of research on the effects of microplastics on freshwater organisms using environmentally realistic concentrations, especially in the Southern Hemisphere. A series of toxicity tests were used to expose a range of taxa including Tilapia sparrmanii, Caridina nilotica, and Melanoides tuberculata to different environmentally realistic concentrations of microplastics of different polymers. The study results show that the environmentally realistic concentrations had no statistically significant effects on most of the chosen test organisms and selected study endpoints, except for T. sparrmanii microplastic particle egestion and growth in polyethylene exposures which produced significant results. Although this study showed that at environmentally realistic concentrations and 21 days of exposure, minute effects on the test taxa were detected, various studies have proven that with longer exposure to microplastics, significant effects on freshwater organisms can be detected. Additionally, studies using concentrations higher than the current environmental concentrations have recorded significant effects on organisms and therefore, with increasing concentrations in the environment, more significant effects may be observed. Therefore, plastic pollution in the environment should be reduced as microplastics are in continuous production and circulation, and microplastic concentrations in freshwater environments are predicted to increase. , Thesis (MSc) -- Faculty of Science, Institute for Water Research, 2023

Thermal physiology of juvenile red roman seabream, Chrysoblephus laticeps after long-term exposure to low pH conditions

- Description: Climate change has caused a combination of effects on the physiology of fishes. Of particular concern are the effects of thermal variability and ocean acidification. Organismal energy budgets change throughout ontogeny and research into the metabolic scope during early life stages is particularly useful in identifying potential bottlenecks. The first part of this thesis aimed to assess the absolute aerobic scope (AAS, described as the difference between the maximum and standard metabolic rates) of individual juveniles from a protected population of the endemic, commercially important seabream, Chrysoblephus laticeps, across a range of ecologically relevant temperatures (T = 11, 14, 18, 22˚C) under present-day conditions (pH = 8.03, pCO2 ≈ 420 μatm) using intermittent flow respirometry. The second component sought to investigate how long-term exposure (from fertilisation to juvenile, ~100 days exposure) to high-pCO2/hypercapnic conditions (pH = 7.63, pCO2 ≈ 1400 μatm), would affect the AAS of juvenile C. laticeps over a range of temperatures. Lower pH conditions were predicted to cause a decrease in the AAS of treatment animals due to additional energetic costs of acid-base regulation. The findings of the first data chapter demonstrated that juvenile C. laticeps reared under current CO2 conditions are tolerant to a wide range of thermal conditions, and individuals with a broad aerobic scope will be the best suited to coping with enhanced thermal variability. In contrast to the expected outcomes of the second data chapter, juvenile C. laticeps reared under high pCO2 conditions displayed greater AAS at high and low temperatures when compared with specimens from high pH conditions. Whilst a high degree of individual phenotypic variation was observed in the metabolic response of both groups, this was reduced at the lower and upper extreme temperatures for high pH and low pH animals respectively. Notably, the variation in treatment animal’s SMR was significantly diminished across all temperatures tested, compared to only a localised reduction in the SMR of high pH animals at cold temperatures. This may be indicative of compensatory pathways affecting energy restructuring and thermally-governed physiological trade-offs under hypercapnia. Given these results, juvenile C. laticeps appear to be more resilient to ocean acidification than anticipated, potentially owing to intrapopulation metabolic phenotypic diversity. This is likely attributed to the parental lineage originating in the Tsitsikamma MPA, which is thought to boast greater phenotypic diversity as a consequence of the refuge that these conservation areas offer from exploitation. Owing to the restriction imposed by the availability of surviving, captive-reared juveniles, the sample size used in this study was relatively low. However, owing to the repeated-measures nature of this research the sample size was sufficient to offer suitable statistical power for the polynomial mixed model used in the analysis. Future research should incorporate both physiological and behavioural responses to multiple environmental stressors to better understand covariation between these two traits, and to detect any behavioural trade-offs that might arise through compensation. An investigation into water and sanitation in the Eastern Cape Province and potential for implementation of biotechnology platforms

- Description: In contemporary South Africa, a country in transition, destruction rather than reconstruction seems commonplace. Electricity supply is at an all-time low and ‘load shedding’ is an almost daily occurrence. Similarly, more fragility of water delivery and sanitation service is being reported with the likelihood of ‘water shedding’ a soon-to-be reality. In view of the ever-increasing reported mismanagement of South Africa’s water and sanitation infrastructure, which is likely nearing the point of collapse if not already collapsed, this thesis set out to interrogate at a provincial and municipal level the status of water and sanitation in Chris Hani District Municipality (CHDM), Eastern Cape Province. One major driver behind this study was to evaluate provincial and municipal water/wastewater infrastructure as a platform to support implementation of locally developed bioprocess technologies. First, publicly available historical data was used to derive a baseline or benchmark from which to determine gains/losses in compliance, water quality and efficiency. The period 2009-2013/4 during which Blue and Green Drop reporting was routine, was used as an appropriate start point. Data thus derived indicated that, at provincial and district municipality (DM) level, potable water supply was by bulk water schemes and, sanitation was typically by waste stabilization pond (WSP) treatment of municipal sewage. The derived benchmark for the period 2009-2013/14 indicated that most plants were not compliant (~75% of the Eastern Cape Province water treatment plants (WTP) operating between average performance to critical state), operated either at- or above design capacity (Eastern Cape Province, 16%; CHDM, 19%). A large number of plants for the province (62.4%) and district (81.25%) had hydraulic design capacity or average daily flows that were unknown (information not provided in available literature or plant reports) and final effluent did not always meet the general standard (70.3% of the CHDM wastewater treatment works (WWTW)). To determine the contemporary state (2020 - 2022) of water and sanitation within CHDM, a scoping exercise of WTP and WWTW in selected local municipalities was carried out. This was done along a west-east transect by appraising WTP and WWTW in the towns of Cradock, Tarkastad, Komani, Tsomo, Cacadu, and Ngcobo. Furthermore, targeted assessment of the Queenstown WTP and the Cradock WWTW was used to derive real-time data on the status of water and sanitation infrastructure. Results showed that water and sanitation services within CHDM had, in the years between 2009-2013/14 to date, deteriorated. Freshwater demand significantly exceeded capability of water supply schemes, where demand was as much as three times greater than reported available supply. For most WWTW, operation was in excess of capacity and between 1.52 and 12 times installed hydraulic loading. The targeted scoping exercise revealed that Queenstown WTP is a moderate risk plant (Cumulative risk rating, CRR = 11 and maximum risk rating, MRR = 47.8%), whereas Cradock WWTW was in working condition but with challenges and some infrastructural dysfunction. Assessment of water/effluent quality revealed that turbidity and TOC were above SANS 241 general limit. Non-compliance in terms of nitrate/nitrite-N, ammonium-N, phosphate-P, TSS, total coliforms, E. Coli, and free chlorine was evident at Cradock WWTW. Unstructured interviews with plant operators corroborated these findings. The overwhelming support for bulk schemes for potable water provision and WSP for sanitation indicated a partially water secure municipal district but derelict in terms of its sanitation services. The later, it was concluded, in particular provides an ideal opportunity for implementation of platform technologies to support bioprocesses for entrepreneurship, employment, economic benefit and to secure a closed circular economy for regional water and sanitation through valorisation of co-product streams. Pharmacists’ attitudes and perception of using pictograms as a communication tool in practice

- Description: Pictograms, when used in conjunction with verbal and written information, are known to be effective in improving comprehension and recall of medicines information and in supporting communication between healthcare professionals and patients. However, pictograms are seldom used in routine pharmacy practice, and little is known about pharmacists’ opinions of pictograms and their intention to possibly incorporate pictograms into routine practice. This study aimed to investigate pharmacists’ opinions relating to pictograms as a communication tool, and, by applying the Theory of Planned Behaviour (TPB), to explore their intention to use pictograms in pharmacy practice as well as the barriers to their use. The quantitative study design involved a two-phase approach. Phase 1 was a descriptive, cross-sectional online national survey of pharmacists. The 70-item, four-section survey was primarily based on the constructs of TPB, which included attitude, intention, subjective norm, and perceived behavioural control. The last survey section recruited pharmacists for a follow-up Phase 2 survey. Following a pilot study, the survey was emailed to all pharmacists registered with the South African Pharmacy Council. Descriptive statistics for survey items were generated. Pearson correlation investigated the correlation between participant characteristics, familiarity with pictograms and use of pictograms in practice. Structural Equation Modelling (SEM) determined if there was a significant relationship between attitude, subjective norm and perceived behavioural control with intention to use pictograms. A total of 426 pharmacists responded to the Phase 1 survey. Most pharmacists were familiar with the term ‘pictogram’; however, over three-quarters of pharmacists had never observed pictograms being routinely used in a pharmacy setting. When presented with pictograms designed for a low health literate population, most pharmacists thought the design and overall look of the pictograms would be easy for most patients in South Africa to understand (71.6 ± 24.0). Two-thirds of pharmacists (65.0 ± 30.6), felt that pictograms should be used for all patient populations. More than 85% of pharmacists agreed that pictograms should be used for dosage instructions, auxiliary or additional information, warnings, and storage instructions. However, fewer (58-68%) felt that indication, side effects and risk communication information should be accompanied by pictograms. Pharmacists demonstrated positive attitudes towards using pictograms in practice (mean = 4.2 ± 0.9; range: 1 - 5), while perceived behavioural control (mean = 3.0 ± 1.2; range: 1 - 5), subjective norm (mean = 3.8 ± 1.0; range 1 - 5) and intention (mean = 3.3 ± 1.0; range 1 - 5) were all neutral. Attitude (β = -0.25, p < 0.117), however, was not a significant predictor of intention while perceived behavioural control (β = -0.83, p < 0.000) presented with a significant negative correlation with intention. Subjective norm (β = 0.57, p <0.000) was the strongest predictor of intention. Scale reliability ranged from 0.770 to 0.865 for the TPB constructs. Phase 2 aimed to expand on, and further investigate Phase 1 findings relating to current and intended pharmacist behaviour and opinions concerning pictogram usage. As Phase 2 looked to investigate issues in greater depth, questions included open-ended response options. The survey link was emailed to all pharmacists who had voluntarily offered to participate in Phase 2. Frequency data for all questions were generated, and content analysis was undertaken for the free-response comments offered by pharmacists. A total of 35 pharmacists responded to the Phase 2 survey. Most pharmacists who routinely used pictograms initiated their use with support from pharmacists’ colleagues (8/12) and their supervisor/manager (6/7). Pharmacists who stated their intention to use pictograms anticipated receiving support from their pharmacists' colleagues (18/23) and manager/supervisor (15/23). All 12 pharmacists who were routinely using pictograms reported a positive effect on patient communication, with almost all having encountered no negative aspects of using pictograms (11/12). Almost all pharmacists intending to use pictograms could foresee benefits from their use (22/23). Misinterpretation of pictograms was a prevalent barrier common to both pharmacists routinely using pictograms and to those intending to use pictograms. Increased workload was regarded as a prevalent barrier only by pharmacists intending to use pictograms. This study was the first national study of pharmacists to investigate their opinion of pictograms and their use and to adopt a theoretical approach to consider pharmacist intention to use pictograms in routine pharmacy practice. Pharmacists generally expressed positive attitudes to pictograms but showed inadequate understanding of pictogram use. Pharmacists using pictograms reported the positive effect of pictograms on their patient communication, whereas those planning to use pictograms could foresee the benefits of using pictograms despite regarding increased workload as a barrier.
