Research in this area has focussed on flies and beetles remain comparatively under-studied, partly because some adult carrion beetles are difficult to identify morphologically, as are their juvenile stages, often requiring specialist expertise in both cases. Molecular taxonomy has been proposed as a solution to these problems. DNA “barcodes" are short fragments of mitochondrial cytochrome oxidase I (COI) DNA that are anticipated to delineate species. This approach is becoming increasingly popular, but has been met with varying enthusiasm from taxonomists. This thesis examines their use in identifying forensically significant beetles.The DNA barcodes of 234 specimens of 25 forensically significant southern African beetle species from seven families (Cleridae, Dermestidae, Silphidae, Staphylinidae, Scarabaeidae, Trogidae and Histeridae) were obtained. Thirty-three initial barcode amplification failures were overcome by using primers other than the standard Folmer pair, undermining the barcode concept’s hope of universal primers that would allow even non-specialists to produce barcodes. Another 150 specimens (64%) entirely failed to yield barcodes, including 18 fresh specimens of three species of Trogidae, implying another lack of universality of the barcoding protocol. The majority of the beetles clustered with confamilials on neighbour-joining and maximum likelihood trees, but 1.3% of the barcodes failed to cluster with their respective families, raising questions concerning the associating power of barcodes. The identification tools of the GenBank and BOLD on-line DNA sequence databases identified 21% of the specimens to the species level, 6% of them correctly. There was evidence of a paralogous sequence in the Cleridae that, while supporting identification now that it has been associated with a morphological identification, would hamper attempts at identification by clustering or phylogenetic analysis.Distance and haplotype network analyses of the barcodes of six widespread species showed that they are not geographically structured. Barcodes are thus unlikely to be indicators of the region of origin of a species and will not determine whether a corpse has been relocated after death. To assess whether a different mitochondrial DNA fragment might address (some of) these problems, a 2.2 kb fragment extending from the 5’ end of the COI gene to the 3’ end of the Cytochrome Oxidase II (COII) gene was analysed for nine species. It was found that, for Dermestidae, Scarabaeidae and Histeridae, higher degrees of diversity occurred downstreamof the barcode region, but the region of highest diversity in the Cleridae was in the barcode region. Thus, finding a more reliable fragment along the COI-COII region for each family may make robust and guaranteed DNA-based identification of these beetles more likely. The possibility of a forensic specimen being incorrectly or not identified based on its barcode alone exists in about 40% of cases, even with the new barcodes reported here. Forensic science sets a very high bar in assessing the performance of its techniques, and it is concluded that barcodes currently have unsettling failure rates as court-worthy evidence.

Rather, the CPUE trends and catches from the area-disaggregated data were compared against those of the area-aggregated data, as a first attempt to discern any appreciable differences which would suggest the use of disaggregated data in future assessments. Both the trawl and jig CPUE trends from the area-disaggregated analysis differed only slightly from those of the area-aggregated data. Similarly, the spring and autumn biomass trends for the main spawning area (east of 22°E) followed the same trends as for the full area. It is therefore concluded that there is currently no need to assess the resource on an area-disaggregated basis.

- Description: The spatial and temporal patterns in the population demographs and feeding ecology of the caridean shrimp, Palaemon peringueyi, was investigated over a 12 month period in the permanently open Kariega Estuary situated on the south-east coastline of South Africa. Shrimps were sampled monthly from three stations corresponding to the lower, middle and upper reaches of the Kariega Estuary. The shrimps were collected at the different stations using a pull net. Temperature, salinity, dissolved oxygen and % cover of submerged vegetation were measured at each sampling station. Finally, the feeding ecology of the shrimp was investigated employing stable carbon (δ¹³C) and nitrogen (δ¹⁵N) isotopes analysis. Temperature, salinity, dissolved oxygen and vegetation cover showed distinct seasonal patterns. The shrimps attained total abundances and biomass of between 0 and 245.6 individuals per m² (ind.m⁻²) and between 0 and 13.6 grams wet weight per m² (g wwt.m⁻²), respectively. Mean abundance ranged between 2.3 (± 3.0) and 88 (± 27.3) ind.m⁻² in the lower reach, between 0.9 (± 0.4) to 21.5 (± 4.5) ind.m⁻² in the middle reach and 0 to 1.6 (± 0.5) ind.m⁻² in the upper reach. Mean biomass ranged between 0.4 (± 0.5) and 4.4 (± 1.4) g wwt.m⁻² in the lower reach, between 0.1 (± 0) to 4.3 (± 0.9) g wwt.m⁻² in the middle reach and between 0 to 1.6 (± 0.5) g wwt.m⁻² in the upper reach. Greatest abundances and biomasses were recorded in winter following heavy rainfall within the catchment area. Freshwater inflow which is due to rainfall is thought to provide a cue for the recruitment of marine breeding organisms. The spatial patterns in the distribution of the shrimps showed that juveniles occupied the lower and middle reaches and adults the entire length of the estuary. There were no significant relationships between the abundance and biomass of the shrimps and the selected physico-chemical variables (p > 0.05 in all cases). Results of the stable isotope study indicated that the mean δ¹³C of the shrimps in the lower, middle and upper reaches were -12.4 (± 1.3) ‰, -15.6 (± 0.5) ‰ and -17.5 (± 0.5) ‰, respectively. The δ¹⁵N isotopes of the shrimps in the lower reach was 11.6 (± 0.5) ‰ with middle and upper reaches having similar δ¹⁵N values, 13.5 (± 0.4) ‰ and 13.4 (± 0.4) ‰, respectively. There was no significant difference in the δ¹³C and δ¹⁵N isotope (p > 0.05) between the sexes of the shrimps. Two-way ANOVA indicated a significant difference in the δ¹³C isotope of the shrimps between the sites (p < 0.05). The difference in the δ¹³C and δ¹⁵N ratios of the shrimps suggests spatial variability in the diet of P. peringueyi within the Kariega Estuary. The diet of the shrimps in the lower reach is primarily composed of Nanozostera capensis epibionts. In the middle reach the diet of the shrimps is likely comprised of the detritus of benthic algae. In the upper reach of the estuary, P. peringueyi likely derives its carbon from a combination of epibionts, benthic algae and detritus derived from Codium.

Ericoid mycorrhizal fungi and potential for inoculation of commercial berry species (Vaccinium corymbosium L.)

- Description: Ericaceous plants are the richest growth form of the fynbos vegetation of South Africa. The fynbos is characterized by highly leached acidic soils, low mineral nutrients and climatically it is a winter rainfall and dry summer region. Ericoid mycorrhizal fungi associate with Erica species enhancing their ability to access essential nutrients for survival under unfavourable growth conditions. The aim of this study was to select local Ericaceae plant species and to isolate, identify and characterize the ericoid endophytes and assess these isolates as potential inocula for commercial berry species. Two ericaceous plants Erica cerinthoides L. and Erica demmissa Klotzsch ex Benth. were identified from the Mountain Drive area of Grahamstown, Eastern Cape. Root staining was used to confirm the mycorrhizal status of both plants. Hyphal coils typical of ericoid association were observed within the epidermal cells of the hair roots under a light microscope. The endophytes were successfully isolated in pure culture on 2% malt extract agar (MEA) and modified Fontana medium. Cultural morphology and microscopy were used for initial identification. Two slow growing isolates were selected. These isolates were further subjected to molecular identification; extracted DNA was amplified using ITS1 and ITS4 fungal primers. The rDNA gene internal transcriber spacer (ITS) was then sequenced and analyzed by comparison to sequences in the GenBank. On the basis of percentage sequence identity Lachnum Retz. species and Cadophora Lagerb. & Melin species were identified as the ericoid endophytes of E. cerinthoides and E. demmissa respectively. The optimum growth parameters of the fungal isolates were determined in 2% MEA incubated at varying temperatures and pH. It was established that both species had optimum growth at 27⁰C and pH 5. The Ericaceae species are sometimes found in metal contaminated sites were ericoid fungi have been proved to alleviate toxicity of their host. The fungal isolates were grown in increasing concentration of Cu²⁺ and Zn²⁺ in 2% MEA. The growth of Lachnum species decreased with increasing Zn²⁺ ions above 2.7 mM while Cadophora species showed a change in morphology and also decreased in growth with increased ion concentration. However there were no significant differences recorded in the growth of Cadophora and Lachnum species on increasing Cu²⁺ concentration. Lachnum and Cadophora isolates were formulated into a semi solid inoculum and inoculated onto micropropagated Vaccinni corymbosum L. plantlets of 5 different varieties. Colonization was low for all varieties, Elliott and Brightwell varieties recorded the highest colonization of 35% and 31% respectively. Lachnum species infected roots showed potential ericoid structures while the Cadophora inoculated plantlets had hyphal coils within the cortical cells typical of ericoid mycorrhizas. Inoculation significantly enhanced the shoot growth of Brightwell and Elliott varieties. The Chandler variety inoculated with Lachnum species showed improved shoot dry weight. The Bluecrop and Elliott varieties inoculated with Cadophora and Lachnum accumulated more root biomass. All inoculated Bluecrop plantlets had an improved canopy growth index. Brightwell plantlets inoculated with Lachnum species also had an enhanced canopy growth index. The growth responses were variable within varieties and between varieties. Treatments with the Cadophora and Lachnum have shown potential in the promotion of growth of the Blueberry species. The findings indicate the need to conduct trials under conditions which simulate the commercial growth conditions so as explore the optimum potential of the isolates.

- Description: The purpose of this project was to investigate the role of melatonin in the pathophysiology of anxiety in humans. The literature study confirmed the intimate relationship between serotonin and melatonin. Melatonin is not only able to act as an agonist (in physiological concentrations) and an antagonist (at higher concentrations) on serotonin receptors but via control of brain pyridoxal kinase activity might have an effect on GABA, serotonin, dopamine and norepinephrine synthesis. A clinical trial to investigate melatonin's effect on anxiety in humans was conducted as a pilot study. Thirty patients complaining of anxiety participated in a liN of 1" double blind placebo controlled trial. During the experiment each subject was thus exposed to melatonin and a placebo for a week at a time on two occasions. During the first phase of the experiment, (Pair '1) patients showed a statistically significant reduction in their anxiety levels during the first period (P1P1), which was not the case during the second period (P1P2). The improvement however continued during the second phase of the experiment (Pair 2) so that there was also a statistically significant improvement during P 2 P 2 (Period 2 / Pair 2) when placebo was administered. It could not conclusively be shown that melatonin was responsible for the improvement in the patients' anxiety. The explanation for these results suggests thelt the improvement was due to a: 1) placebo effect throughout, 2) psychotherapeutic effect due to contact with a clinician, 3) melatonin induced phase shift in the patient's endogenous melatonin response curve, 4) combination of all 3 options. This pilot study lays the groundwork for a much more exhaustive study in which the melatonin of the patients is determined before melatonin is administered, the role of the clinician is clarified and the most appropriate time for melatonin administration is sought .

A few Dunaliella species failed to grow in this effluent. A correlation was observed between the lack of growth displayed by these species in this organio-rich medium and their failure to utilise organic compounds. Glycine, a major component of this effluent, possibly stimulates the growth of Dunaliella. Studies on the mechanism of growth stimulation by glycine revealed that an algal-bacterial relationship existed whereby the bacteria mineralised the amino acid, releasing ammonia which was then utilised by the alga. Results of this work revealed significant variations in the intracellular glycerol content amongst the Dunaliella species under study. Large differences were also observed between the glycerol contents of effluent-grown and control Dunaliella cells, where the effluent-grown cells were characterised by greatly reduced intracellular glycerol content. These reduced glycerol levels are assumed to have arisen from the glycine-induced stimulation of glycerol release which was observed in this study, where the high glycine content of the hide soak liquor is proposed to have induced glycerol release. This enhanced glycerol release in tatmery effluent could play a central role in the fimction of Dunaliella-based High Rate Oxidation Ponding systems, by stimulating bacterial activity. Observed glycerol productivities were therefore proposed to be a fimction of the type and concentration of the organic constituents of the medium. A similar medium-induced phenomenon was observed in the starch content of Dunaliella cells.

All Besshi-type deposits are contained within sequences of clastic sedimentary rock and intercalated basalts in a marine environment. The basalts and amphibolites are principally tholeiitic in composition. Besshi-type deposits characteristically form stratiform 1enses and sheet-like accumulations of semi-massive to massive sulphide. The main ore assemblage consists dominantly of pyrite and/or pyrrhotite with variable amounts of chalcopyrite, sphalerite and trace galena, arsenopyrite, gold and e1ectrum, barite being absent in general. The median Besshi-type deposit (n=75) contains 1.3 million tonnes (Mt) of massive sulphide with a Cu grade running at 1.43%. It is suggested that Besshi-type deposits form by both exhalative and synsedimentary replacement processes when considering geological features and comparisons with modern analogues in the Guaymas Basin, Middle Valley and Escanaba Trough. The currently forming metalliferous sediments in the Red Sea provide for a brine pool model explaining the lack of footwall feeder zones below sheet-like deposits. Where thick sulphide lenses are contained in some Besshi-type deposits, combinations of exhalative precipitation and sub-sea-floor replacement of permeable sediments and/or volcanic rocks, take place in the upper parts of submarine hydrothermal systems.

- Description: Iron-formations are ferruginous sedimentary rocks which have their source from fumarolic activity associated with submarine volcanism, with deposition of iron as oxides, hydroxides, and hydrous oxide-silicate minerals in shallow and/or deep marine sedimentary systems. The Precambrian ironformations of southern Africa have a wide age range, but are more prominently developed before 1.SGa. These iron formations occur in greenstone belts of the Kaapvaal and Zimbabwean cratons, in the Limpopo mobile belt, in cratonic basins and in the Damara mobile belt. The Archaean-Proterozoic sedimentary basins and greenstone belts host iron ore deposits in iron-formation. Iron formations have a lengthy geological history. Most were subjected to intense, and on occasions repeated, tectonic and metamorphic episodes which also included metasomatic processes at times to produce supergene/hypogene high grade iron ores. Iron-formations may be enriched by diagenetic, and metamorphic processes to produce concentrating-grade ironformations. Uplift, weathering and denudation, have influenced the mineral association and composition of the ores, within which magnetite, haematite and goethite constitute the major ore minerals. The iron resources of the southern Africa region include the Sishen deposits, hosting to about 1200 Mt of high grade direct shipping ore, at >63% Fe. Deposits of Zimbabwe have more than 33 000 Mt of beneficiable iron-formation. The evaluation of an iron ore prospect involves many factors which must be individually assessed in order to arrive at an estimate of the probable profitability of the deposit. Many of these are geological and are inherent in the deposit itself. Other factors are inherent aspects of the environment in which the ore is formed. Although the geological character of the ore does not change, technological advances in the processing techniques may have a great effect on the cost of putting the ore into marketable form. Geochemical, geophysical and remote sensing methods would be used for regional exploration. Chip sampling and drilling are useful for detailed exploration. Purely geological exploration techniques are applicable on a prospect scale in the exploration of iron ore deposits. Regional exploration targeting should choose late Archaean greenstone belts containing oxide facies iron-formation or Early Proterozoic basins located at craton margins as they are both known to host high-grade haematite orebodies formed by supergene/hypogene enrichment. Most types of iron ore deposits in southern Africa are described and classified. An attempt is made to emphasize the major controls on mineralisation, in the hope that these may be applicable to exploration both in the southern African region and within analogous settings around the world.

Mineralization varies from massive and matrix to disseminated, and is nearly always concentrated at the base of the host unit. Primary ores have a relatively simple mineralogy, dominated by pyrrhotite-pentlandite-pyrite, and to a lesser degree millerite. Metamorphic grades tend to range from prehnite-pumpellyite facies through to lower and upper amphibolite facies. Genesis of Group I and II deposits is explained by the eruption of komatiites into rift-phase greenstone belts, as channelized flows, which assimilated variable amounts of footwall rocks during emplacement. Sulphide saturation was dependent on the mode of emplacement and, the amount of sulphidic sediments that became assimilated prior to crystallization. This possibly accounts for variations in ore tenor. The Six Mile Deposit (SMD) in Western Australia, is an adcumulate body of the Group IIB-type, exhibiting disseminated mineralization. The ore has been "upgraded" due to hydration and serpentinization. A profound weathering sequence exists, which was subsequently utilized during initial exploration. Exploration techniques has been focused on Western Australia, as it is here that the most innovative ideas have emerged.

A method to correct for the effects of the contamination has been developed and applied to these kimberlites. Using the corrected data, the four kimberlite types in each group appear to be related to each other by crystal/liquid fractionation models. However the two groups cannot be related to each other. The De Beer Mine has two varieties of kimberlite, a monticellite apatite and calcite rich variety which intruded first, and a phlogopite rich type forming a discrete cylindrical body within the earlier kimberlite. These two kimberlites do not appear to be related by any of the fractionation models discussed. An examination of the data from this work and published sources, suggests that kimberlites are derived from below the low velocity zone by small degrees of partial melting involving garnet lherzolite with subordinate phlogopite and carbonate. Diamonds are probably incorporated as xenocrysts in the magma. Upward movement and emplacement of kimberlite appears to have been very rapid. The diatremes were probably eroded and shaped by gas, derived from the kimberlite magma, escaping to surface along weak zones in the earth's crust. Xenoliths of crustal material incorporated in the kimberlite on intrusion have also been studied and various features due to alteration by the magma are described, including the formation of natrolite and cebollite.
