- Description: The adsorption and separation of platinum(IV) and palladium(II) chlorido species (PtCl₆²⁻ and PdCl₄²⁻) on polystyrene-based beads and nanofibers as well as silica microparticles functionalized with polyamine centres derived from ethylenediamine (EDA), diethylenetriamine (DETA), triethylenetriamine (TETA) and tris-(2-aminoethyl)amine (TAEA) is described. The functionalized sorbent materials were characterized by using microanalysis, SEM, XPS, BET and FTIR. The nanofiber sorbent material functionalized with ethylenediamine (F-EDA) had the highest loading capacity which was attributed to its high nitrogen content (10.83%) and larger surface area (241.3m²/g). The adsorption and loading capacities of the sorption materials were investigated using both the batch and column studies in 1 M HCI. The adsorption studies for both PtCl₆²⁻ and PdCl₄²⁻ on the polystyrene-based sorbent materials fit the Langmuir isotherm while the silica-based sorbents fitted the Freundlich isotherm with R² values > 0.99. In the column experiment the highest loading capacity of Pt and Pd were 7.4 mg/g and 4.3 mg/g respectively on the nanofiber sorbent material based on ethylenediamine (EDA). The polystyrene and silica-based resins with triethylenetetramine (TETA) functionality (M-TETA and S-TETA) showed selectivity for platinum and palladium, respectively. Metal chlorido complexes loaded on the sorbent materials were recovered by using 3% m/v thiourea solution as teh eluting agent with quantitative desorption efficiency under the selected experimental conditions. The separation of platinum from palladium was partially achieved by selective stripping of PtCl₆²⁻ with 0.5 M of NaClO₄ in 1.0 M HCI with PdCl₄²⁻ was eluted with 0.5 M thiourea in 1.0 M HCI. The selectivity of the M-TETA and S-TETA sorbent materials was proved by column separation of platinum(IV) and palladium(II), respectively, from synthetic solutions containing iridium(IV) and rhodium(III). The loading capacity for platinum on M-TETA was 0.09 mg/g while it was 0.27 mg/g for palladium on S-TETA. , Acrobat PDFMaker 10.1 for Word , Adobe Acrobat 9.54 Paper Capture Plug-in

- Description: This thesis discusses guidelines for developers of Augmented User Interfaces that can be used by illiterate and semi-literate users. To discover how illiterate and semi-literate users intuitively understand interaction with a computer, a series of Wizard of Oz experiments were conducted. In the first Wizard of Oz study, users were presented with a standard desktop computer, fitted with a number of input devices to determine how they assume interaction should occur. This study found that the users preferred the use of speech and gestures which mirrored findings from other researchers. The study also found that users struggled to understand the tab metaphor which is used frequently in applications. From these findings, a localised culturally-relevant tab interface was developed to determine the feasibility of localised Graphical User Interface components. A second study was undertaken to compare the localised tab interface with the traditional tabbed interface. This study collected both quantitative and qualitative data from the participants. It found that users could interact with a localised tabbed interface faster and more accurately than with the traditional counterparts. More importantly, users stated that they intuitively understood the localised interface component, whereas they did not understand the traditional tab metaphor. These user studies have shown that the use of self-explanatory animations, video feedback, localised tabbed interface metaphors and voice output have a positive impact on enabling illiterate and semi-literate users to access information. , TeX , Adobe Acrobat 9.51 Paper Capture Plug-in

Development of techniques for the isolation of a granulovirus from potato tuber moth, phthorimaea operculella (Zeller)

- Description: Phthorimaea operculella, commonly known as the Potato Tuber Moth, is an economically important agricultural pest worldwide. The baculovirus, Phthorimaea operculella granulovirus (PhoGV) has been considered as a means of control alternative to chemical control because of its host specificity and harmless impact on other organisms and ecosystems. An isolate of PhoGV obtained from a South African PTM population would be beneficial in the production of a biopesticide, which is not yet available. An efficient and cost-effective rearing method would be advantageous for potential commercial production. Commercial table and seed potato plantations and storage facilities located in Patensie, Bathurst, Howick and Ivanhoe were surveyed for PTM infestations. Patensie was the only site where milky discoloured larvae were found, a potential symptom of PhoGV infection. TEM analysis revealed no virus in these samples. Since no virus was found in the field-collected samples, PTM insects were collected to initiate rearing in the laboratory. PTM was raised by three different methods in the laboratory. A cost/benefit analysis, survival rate, fertility and sex ratio were recorded for each rearing method. Rearing method one was deemed unsuccessful for efficient commercial rearing, as survival percentage and fertility were low. Rearing methods two and three had high survival rates and high fertility, and were efficient and less labour intensive than rearing method one. Rearing method three was the most productive technique, but for commercial production rearing method two was considered the most manageable and efficient. The sex ratio was 1:1 for all three cultures. The cost analysis revealed that rearing methods two and three were less expensive than rearing method one because less labour was required to monitor insects. The success of rearing PTM for 19 months will enable these cultures to be up-scaled to a large production facility for mass rearing. Virus was not found in the field surveys or in laboratory cultures, therefore chemical, temperature, humidity and carbon dioxide stressors were used in an attempt to initiate a baculoviral infection. Symptoms were exhibited in larvae subjected to chemical, temperature and humidity treatments, but these were confirmed by TEM analysis not to be a result of PhoGV infection. The success of rearing PTM in the laboratory suggests that the method could be used in the commercial rearing of the insects in a large mass-rearing facility. The data obtained from induction protocols have allowed for better understanding for future induction for PhoGV and other baculoviruses in other insect species. The failure to isolate a South African PhoGV strain for developing a biopesticide against PTM has motivated further studies in obtaining a baculovirus in order for South Africa to develop a commercial product against this pest.

- Description: This work has focussed on the preparation of a variety of tridentate ligands, designed to form ruthenium complexes as potential metathesis catalysts. Various approaches to the tridentate, malonate-tethered imidazolidine system have been investigated, and a promising route to accessing ligands of this type is discussed. A tridentate malonate-tethered pyridine ligand has been successfully prepared and its dithallium salt has been accessed by hydrolysis with thallium carbonate; approaches to a longer-chain analogue have also been investigated. A thallium pyridine-2,6- dicarboxylate ligand has been has been successfully prepared, as have a range of pyridine diamine ligands, with various alkyl and aromatic substituents on the amine donor atoms. Preliminary investigations into the potential of these compounds as ligands for alkylidene ruthenium complexes are reported using molecular modelling techniques. The geometries and steric energies of the ligands and their corresponding complexes have been analysed, and results obtained from two different software packages are compared. Finally, some preliminary complexation studies have been undertaken.

Aspects of the biology of two inshore sparid fishes (Diplodus sargus capensis and Diplodus cervinus hottentotus) off the south-east coast of South Africa

- Description: The blacktail, Diplodus sargus capensis and the zebra, Diplodus cervinus hottentotus, sparid fishes endemic to South Africa, are important components of the recreational shore-fishery. To provide a basis for the management of these fish, aspects of the biology of both species were investigated in the Tsitsikamma National Park (TNP). In particular, the role of the TNP was evaluated as a management option for the conservation of both species. Examination of stomach contents showed that juveniles of both species fed predominantly on amphipods, polychaetes and harpacticoid copepods. Adult !h. sargus were omnivorous and fed opportunistically on a wide variety of reef associated invertebrates and algae including echinoids, polychaetes, anthozoans, ascidians and rhodophytes. The diet of adult D. cervinus was more specialized with a preference shown for polychaetes and amphipods. Seasonal differences were apparent in the diets of both species where considerably more amphipods were consumed in winter than in summer. Feeding requirements of both species were reflected in their habitat preferences. Visual underwater assessment revealed that both species were most abundant on turbulent inshore reefs (1-10m). !h. sargus were observed over a wide variety of reef associated habitats. In the literature they have also been recorded in the surf zone of sandy beaches and in the lower reaches of estuaries. !h. cervinus, on the other hand, were more specific in their habitat requirements and were observed in greatest abundance on inshore reefs, often in close association with caves or overhangs. A comparison between the relative abundance and size structure of both species in the TNP with that of an exploited area immediately adjacent to the reserve, showed no significant differences. This was attributed to the low level of exploitation by rock-and-surf anglers occurring in the exploited study area, as well as the possibility of seeding of eggs and larvae, or emigration of adults from the TNP. An investigation of the reproductive biology of both fish showed that !h. sargus had an extended summer spawning season while in !h. cervi nus it was more restricted. Detailed histological examination of gonadal development showed that !h. sarqus were dygynous with partial protandry occuring in the male population. ~ cervinus were shown to be rudimentary hermaphrodites. Size at 50% maturity in ~ sargus and ~ cervinus was determined at 225 and 285mm fork length respectively, corresponding to ages of 4 and 6 years. An age and growth study based on the examination of sectioned otoliths showed that both species were slow growing capable of reaching ages in excess of 20 years. Growth in ~ sargus and ~ cervinus was described by the von Bertalanffy growth equations: L(t) = 309.44(1-e-0 . 247[t+l. 048 l) and L(t) = 396.85(1-e-0.146[t+2.148J) respectively. Life history characteristics of D. sargus and D. cervinus including slow growth, late maturation and occupation of a localised, demersal habitat showed that both species were vulnerable to the effects of over-fishing. Due to the present increase in the number of participants and the decrease in catch per unit effort in the recreational shore-fishery, more stringent management recommendations were proposed to ensure the adequate protection of both species. These included an increased minimum size limit and a decreased bag limit for both species. Based on the residency shown by both species and their high relative abundance in the TNP, marine reserves were considered to be a valuable addition to the suite of management options available for the conservation of these species . In this respect the reserve provided both protection for the spawner stock as well as the potential to seed adjacent areas.

- Description: The thermal decomposition of silver permanganate, pre-irradiated in BEPO and in a ⁶°C₀ Ϫ 'hot spot' has been investigated in the temperature range 100 - 125°C. The results are similar to those for irradiated KMn0₄ and the mechanism proposed for the latter is again suggested. The activation energy for the migration of point defects over the induction period is 1.03 ev. The decompositions of unirradiated and irradiated crystals differ in that the latter undergo physical disintegration over the acceleratory period. X-ray studies immediately prior to disintegration show strain and fragmentation in the irradiated crystal. An explanation involving the annealing of point defects at dislocation is advanced to explain the changes produced in the p/t plots with increased dosage, and fixed decomposition temperature. Summary, p. 94.
