Smashing the crystal ball: post-structural insights associated with contemporary anarchism and the revision of blueprint utopianism

- Description: This thesis is an exploration of the images which define revolution's meaning. It suggests a possible shifting of emphasis from the scientific imaginary which centres on identifying the correct way to totalising revolution, towards a post-structuralist-anarchistic imaginary which privileges prefigurative radicalisations of social relations in the here and now. It looks specifically at how the field of post-structuralism intertwines with historically anarchist concepts to generate an horizon of social change animated by experimental and open-ended transformations. While the thesis offers positive characterisations of the types of contemporary movements, tactics and principles which embody the change from closed to open utopianism, it is chiefly a commentary on the role of theory in depicting the complexity of relations on the ground and the danger of proposing one totalising pathway from one state of society to another. It asks the reader to consider, given the achievements of movements and given the insights of post-structuralism, whether it is still worthwhile to proclaim certainty when sketching the possibilities for transcendence toward emancipation, an aim, which in itself, is always under construction. I engage this by firstly establishing a practical foundation for the critique of endpoints in theory by exploring the horizontal and prefigurative nature of a few autonomous movements today. Secondly I propose the contemporary theory of post-structuralist anarchism as concomitant with conclusions about transformation made in the first chapter. Finally I recommend a few initial concepts to start debate about the way forward from old objectivist models of transformation. The uncertainties of daily life, crumbling of economic powers and rapid pace of change in the twenty-first century have opened up fantastic spaces for innovative thought. Reconsidering old consensus around what constitutes a desirable image of revolution is of considerable importance given today's burgeoning bottom-up political energy and the global debate surrounding the possibilities for bottom-up revolutionisation of society. I submit that theories which portray stories of permanent, pure and natural end-points to revolution are deficient justifications for radical action.

The synthesis and breast cancer inhibitory activity of cinnamic acid analogues based on the halogenated monoterpene pharmacophore

It is estimated that 1.3 million women are diagnosed with the disease each year especially in the developing countries. Current chemotherapy relies on the use of high doses of non-specific toxic agents that possess adverse side effects and compromise patient’s compliance and adherence to treatment. Paclitaxel, one of the common drugs used in breast cancer chemotherapy results in sensory and motor neuropathy, whilst hormonal therapy e.g. Herceptin causes severe cardiovascular, gastrointestinal and cutaneous side effects. There has been a demand in developing newer cancer agents that demonstrate selective cytoxicity with minimal effect on normal body tissue. Numerous studies have shown that marine organisms produce a wide range of halogenated compounds that possess cytotoxic properties, and hence can be a source of new drug hits or leads for cancer therapy. Halomon, a polyhalogenated monoterpene from Portieria hornemannii, displayed interesting activity against brain, renal and lung cancer tumours with selective/differential cytotoxicity. This inspired us to focus our project on halogenated monoterpenes isolated from the same Rhodophyta class as P. hornemannii but with particular attention to Plocamium species. Several metabolites have been isolated from P. cornutum, P. corallorhiza and P. suhrii that possess interesting cytotoxicities against a breast cancer cell line (MCF7) and an oesophageal cancer line (WHCO1). The aim of the project was therefore centred at isolating target compounds for preliminary structure-activity studies against a breast cancer cell line, and use this information to synthesize a series of analogues that are more stable than the natural products and yet as active using a fragment-based type approach to map out pharmacophoric elements. Five metabolites were isolated from P. cornutum and five from P. corallorhiza. Cell-based assays were conducted using an MTT assay kit against MCF7 and MDA-MB-231 breast cancer cell lines and (1E,3E,5S,6R)-1,5,6-trichloro-2-(dichloromethyl)-6-methylocta-1,3,7-triene, isolated from P. cornutum was the most active with IC50 values of 3.0 μM and 6.15 μM respectively. Introduction of a terminal aromatic ring to enhance stability, together with varying substituents (H, CH3, CF3, Br, CN, CHO, CHCl2) on position 7 of the molecule, gave rise to a series of cinnamate ester derivatives inspired by (1E,3E,5S,6R)-1,5,6-trichloro-2-(dichloromethyl)-6-methylocta-1,3,7-triene. The analogues were synthesized from their benzaldehyde precursors via Aldol condensation, esterification and Wittig reactions. Their carboxylic acid counterparts were synthesized by hydrolysis of the parent esters in an attempt to promote water solubilities of the analogues. Biological activity assays were then conducted with the cinnamate analogues against the MDA-MB-231 breast cancer cell line using an MTT assay kit. Ester derivatives with -CHO and -CHCl2 functionalities had IC50 values of 43.45 μM and 100.01 μM respectively whilst the other ester derivatives were inactive. Bioaccumulation and histopathology of copper in Oreochromis mossambicus

- Description: Cu is one of the most toxic elements that affect fish populations when the fish are exposed to concentrations exceeding their tolerance. To investigate the effects of elementary Cu on aspects of bioconcentration, histology and behaviour, O. mossambicus were exposed to 0 and 0.75 ± 0.20 mg/l of Cu for 96 hours (short-term study), and 0, 0.11 ± 0.02, 0.29 ± 0.02, and 0.47 ± 0.04 mg/l of Cu for 64 days (longterm study) under controlled conditions in the laboratory. For the long-term study fish were sampled for gills, liver, and kidney Cu accumulation analysis after 1, 32 and 64 days of exposure and after 1, 2, 4, 16, 32, and 64 days for gills, liver and spleen histology analysis. Cu accumulation was concentration-duration dependent with the highest accumulation capacity in the liver. A multifactor linear model was developed for the relationship between exposure dose, exposure duration and Cu accumulation in the organs with the liver model: Log L = 3.35 + 0.85W + 0.31T (r² = 0.892) giving a better fit than the gills: G = −35.09 + 10.58W + 17.58T (r² = 0.632). Where L = Cu accumulation values in the liver, G = Cu accumulation values in the gills (both in μg/g dry mass); W = exposure dose in water (mg/l); and T = exposure time (days). Using this model Cu accumulation in organs can be estimated when exposure concentration and duration is known. This model should be tested under different conditions to determine the potential of the model in monitoring Cu toxicity in the environment. Lesions were observed in the liver, gills and spleen in all Cu treatments at all exposure concentration and exposure durations. However, the incidence and the degree of alteration was related to the concentration of Cu and duration of exposure. The sequential appearance of lesions in the order of, hepatic vacuolar degeneration, fatty degeneration and necrosis indicated a gradual increase in liver damage with larger duration of exposure time and increasing Cu concentration. The initial lesions in the gills were manifested as hypertrophy and hyperplasia of the gill epithelium causing increase in the thickness of the secondary lamellae, mucous cell hypertrophy and proliferation, mucous hypersecretion, proliferation of eosinophilic granule cells and hyperplasia of interlamellar cells. With increase in exposure time, necrosis of the eosinophilic granule cells, lamellar oedema, epithelial desquamation and increase in severity of lamellar hyperplasia were observed. These lesions indicated an initial defence mechanism of the fish against Cu toxicity followed by advanced histological changes that were related to Cu concentration and duration of exposure. Changes in the spleen were haemosiderosis, increase in the white pulp and macrophage centres, reduction in the red pulp, and necrosis suggesting that fish exposed to environmentally relevant levels of Cu may be histopathologically altered leading to anaemia and immunosuppression. Regression analysis was used to quantify the relationship between the total activity of the fish, and duration of exposure. There was a gradual decline in fish activity related to Cu concentration and duration of exposure before introducing food into the tanks. There was a constant activity after introducing food in the tanks at the control and 0.11 ± 0.02 mg/l Cu exposure levels irrespective of exposure time. Analysis of covariance (ANCOVA) was used to test for the difference in slopes between treatments. There was no significant difference (p > 0.05) between slopes of the control and 0.11 ± 0.02 mg/l Cu, and between 0.29 ± 0.02 and 47 ± 0.04 mg/l Cu before and after introducing food in the tanks. The slopes of both the control and 0.11 ± 0.02 mg/l Cu were significantly different from those of 0.29 ± 0.02 and 0.47 ± 0.04 mg/l Cu (p < 0.05). There were significant differences in the mean opercular movements per minute between treatments (p < 0.05). There was hyperventilation at 0.11 ± 0.02 mg/l Cu i.e. 87 ± 18 opercular movements per minute (mean ± standard deviation) and hypoventilation at 0.29 ± 0.02 and 0.47 ± 0.04 mg/l Cu i.e. 37 ± 34 and 13 ± 6 opercular movements per minute compared to the control. Hypo- and hyperventilation were related to the lesser and greater gill damage, respectively. In conclusion Cu accumulation and effects on histology of the liver, gills and were related to the concentration of Cu in the water and duration of exposure showing a gradual increase in incidence and intensity with larger duration of exposure time and increasing Cu concentration. However, as the exposure continued, the homeostatic mechanism appears to have failed to cope with the increasing metal burden causing advanced histological changes.

An investigation into the influence of the environment on spawning aggregations and jig catches of chokka squid Loligo Vulgaris reynaudii off the south coast of South Africa

- Description: Erratic and highly variable catches in the South African chokka squid Loligo vulgaris reynaudii fishery, cause socio-economic hardship for the industry and uncertainty for resource managers. Catch forecasting can reduce this problem as it is believed that catch variability is strongly influenced by environmental factors. In this study, data were collected at varying temporal and spatial scales. Data for the hourly time-scale study were collected from 1996-1998, aboard commercial vessels, whilst for the longer time-scales, data were extracted for Kromme Bay (a single fishing area) from existing databases (1991-1998) that were comprised of compulsory catch returns and oceanographic data. The environment-catch relationship for chokka squid on the inshore spawning grounds was then investigated using multiple correlation and regression analysis, analysis of variance, contingency table analysis and cross-correlation statistical techniques. This simple, direct, 'black box' statistical approach was relatively successful in developing a predictive capability. On a short time-scale (hourly), the regression model accounted for 32% of the variability in catch, with turbidity the main determinant (13%). On a daily monthly time-scale, the best prediction model was on a monthly scale, accounting for 40% of the variability in catch. The principal determinant, bottom temperature anomaly (11 %), was found to lag one month forward. Seasonal and diel catch variations induced changes in the relative importance of turbidity, water temperature and wind direction on catches. A strong, positive relationship was found between easterly winds (which cause upwelling) and catch, particularly in summer. Catch rates, however, decreased with an increase in turbidity. The correlation between temperature and catch was generally negative, however, higher catches were associated with a temperature range of 13-18°C. Highest catch rates were associated with easterly winds, zero turbidity conditions and sea surface temperatures from 15.0-16.9°C. Selected case studies (in situ observations) suggested that upwelling and turbidity events act as environmental triggers for the initiation or termination of the spawning process, respectively. A holistic approach is required to improve predictive capability of chokka squid abundance. Although short-term predictability remains essential (i.e. hourly-scale), future research should concentrate on long-term prediction models (e.g., monthly time-scales) involving greater spatial variation, which are the most important for management.

A strong, positive relationship was found between easterly winds (which cause upwelling) and catch, particularly in summer. Catch rates, however, decreased with an increase in turbidity. The correlation between temperature and catch was generally negative, however, higher catches were associated with a temperature range of 13-18°C. Highest catch rates were associated with easterly winds, zero turbidity conditions and sea surface temperatures from 15.0-16.9°C. Selected case studies (in situ observations) suggested that upwelling and turbidity events act as environmental triggers for the initiation or termination of the spawning process, respectively. A holistic approach is required to improve predictive capability of chokka squid abundance. Although short-term predictability remains essential (i.e. hourly-scale), future research should concentrate on long-term prediction models (e.g., monthly time-scales) involving greater spatial variation, which are the most important for management.

Unfavourable conditions during fermentation may alter the performance of the yeast strain Saccharomyces cerevisiae, resulting in a "poor" end-product. It has been established that high concentrations of extracellular ethanol, when added to the fermentation medium inhibit yeast activity. It has been recently suggested that increased carbon dioxide pressure could inactivate the yeast activity adding to further brewing problems. The aim of this study was to investigate the effect of extracellular carbon dioxide pressure and ethanol addition, on yeast performance when added to a fermentation medium, and to establish whether an inhibitory relationship existed between ethanol and carbon dioxide pressure, when combined and added to the fermentation medium. Dissolved C0₂ in the medium, medium pH and substrate utilisation were analysed daily during a fermentation, as were membrane fatty acid composition. These parameters were used to assess the effect of ethanol and carbon dioxide on the yeast performance and consequently the final end-product. Supplementing the medium with extracellular ethanol, even as low as 5%, was shown to inhibit yeast performance during fermentation. This effect was even more marked as the ethanol concentration was increased, with almost total inhibition of yeast activity occuring after the addition of 15% ethanol (v/v). A similar effect was observed when elevated C0₂ pressures were applied to the medium, and although low C0₂ pressures initially induced the synthesis of saturated yeast membrane fatty acids, elevated C0₂ pressures (greater than 1,0 atm.) was shown to follow a similar inhibitory trend, if not as dramatic, as ethanol. A combination of both ethanol and C0₂ pressure showed a further increase in the level of yeast inhibition, although the low C0₂ pressure appeared to initially inhibit the toxicity of ethanol on the yeast. Increasing the levels of the C0₂/ethanol treatment (1,0 atm.), showed a synergistic effect on yeast performance. The results of this study indicate that both extracellular ethanol and carbon dioxide do appear to inhibit yeast performance and affect membrane fatty acid composition of the cells by inhibiting the synthesis of the respective fatty acid. This affect has a significant bearing on the general metabolism of the yeast cell.

- Description: Africa is currently experiencing a movement toward more democratic systems of government. The causes of such changes are numerous, but the literature on African democratization, like that on similar changes elsewhere in the world, places emphasis on the role of internal or domestic factors. The role of international pressures toward democratization is almost completely ignored. The case of Mozambique illustrates the dangers of such an omission. During the past decade Mozambique has undergone considerable political change. The single-party, Marxist-Leninist oriented state has been replaced by a multi-party system, devoid of explicit references to any guiding ideology. The government has also expanded its contacts with the West, particularly by means of its assuming membership of the International Monetary Fund and the World Bank. These changes in Mozambique's political orientation have been accompanied by economic reforms, designed to arrest the precipitous decline in the Mozambican economy. In this dissertation I argue that the causes of both the economic and political reforms lie in this decline and in the government's need to secure capital and debt relief internationally. In order to do this, the Mozambican government had to change the aspects of its political system which were seen as being unacceptable by the West, in particular the lack of multi-party competition and its overtly Marxist orientation and close ties to socialist countries. Because the reforms had their primary genesis in Mozambique's need for international acceptance and not in the growth of popularly based democratic organisations, the reforms are fragile and their meaningfulness questionable.

The digestive mechanisms of an intertidal grazer, the sea urchin Parechinus angulosus

- Description: Echinoids are important grazers in the near-shore ecosystem and can significantly effect their ecology. The sea urchin Parechinus angulosus occurs inter- and subtidally along the southern African coast. Within this range it consumes an extremely wide variety of algae. Since algal cell walls have an almost species specific chemical composition, the question arises as to how it can digest the algae that it consumes. In order to investigate the digestive mechanisms employed by P. angulosus, an ultrastructural study of the gut was undertaken to characterize the tissue and identify functional regions in the gut. Ten structural and storage polysaccharides commonly found in macroalgae were used as substrates to assay the digestive polysaccharidases of the sea urchin. The enteric bacteria of the sea urchin were isolated and tested separately for polysaccharidase activity using the same substrates. The results shown that the gut of Parechinus angulosus is regionally specialized, with the foregut primarily responsible for the production of hydrolytic enzymes, while the hindgut is primarily absorptive. The occurrence of lamellar bodies, heterolysosomes, cytoplasmic blebs and paddle cilia among other characteristic features of the digestive epithelium are described and discussed. Two levels of enzyme activity are apparent. Generally the urchin could hydrolyze the reserve polysaccharides, but only partially hydrolyze the structural polysaccharides, of red and green algae. P.angulosus was unable to digest alginic acid, the main structural polysaccharide of brown algae. Mixed cultures of bacteria utilized only the reserve polysaccharides of red and green algae. Significantly, the bacteria were able to hydrolyze alginic acid. Enteric bacteria also showed agarolytic activity. Parechinus angulosus has the ability to digest red and green algae. No lysozyme activity was detected. The enteric bacteria can digest the same algal reserve polysaccharides and so may compete for carbon in the gut. However, in the case of brown algae, bacteria have a potentially important endosymbiotic role as agents of digestion. These results correspond with food preference studies which have shown that, although P.angulosus consumes the kelp Ecklonia maxima, in the western Cape, it is amongst its least preferred food species. The reasons for this are its unpalatability and the urchin's inability to digest brown algae. The digestibility of algal material can be an important factor in determining algal-herbivore interactions.

Further it is known that the concentration of potassium ions in the haemolymph of phytophagous insects is far higher than that in predatory forms; this has led to the suggestion that the nerve sheath in plant feeding insects should be more highly developed than that of entomophagous insects. In this work the structure of the nerve sheath in phytophagous and predatory insects has been studied and this assumption has been shown to be groundles. However, preliminary experiments on the effects of ions and drugs on the peripheral nerves of phytophagous and predatory insects have shown that there is a definite difference in susceptibility between the nerves of these two forms and this has led to the postulate of a diffusion barrier beneath the level of the nerve sheath, which is more highly developed in phytophagous than in predatory forms. The properties of this second barrier are discussed. Part 1. , During the course of the work which has been described in Part 1- an outbreak of large saturniid moths Nudaurelia cytherea capensis Stoll.) occurred in the Grahamstown area. It was felt that an investigation into the properties of the flight motor of this moth, which has an extremely low wing beat frquency, might be rewarding as our knowledge of the flight motor in insects is limited to those with very much higher wing beat frequencies than that of this moth. The anatomy, innervation and histology of the flight muscles of Nudaurelia are described and it is shown that the flight motor of this moth is functionally different to that of other insects which have been investigated. Further, Nudaurelia shows a characteristic warm-up fluttering of the wings prior to flight - this phenomenon has also been examined in the following investigation. This study has yielded information about the location of a warm-up centre in the central nervous system of this moth. Part 2.
