The methods currently in use are manual, but despite our lack of understanding of the existing demands, advances in forestry engineering have resulted in an introduction of semi-mechanised versions of these tasks. This project aimed to compare the task demands of silviculture tasks using the current manual techniques and the more modern, semi-mechanised techniques. Methods: A holistic investigation focused on the worker characteristics of a sample of black male pitters and black female planters from the Kwa-Zulu Natal forestry industry, as well as biomechanical (spinal kinematics and L5/S1 forces), physiological (heart rate, oxygen consumption and energy expenditure) and psychophysical (ratings of perceived exertion and body discomfort) responses associated with manual and semi-mechanised pitting and planting. Results: The pitting task saw significant improvements in the spinal kinematic measures as a result of the increased mechanisation, with eight of the 16 recorded variables decreasing to a lower level of risk classification. Physiologically, the manual task was associated with a mean heart rate of 157 bt.min⁻¹ and absolute energy expenditure of 11.27 kcal.min⁻¹, which were not found to be significantly different to the values of 143 bt.min⁻¹ and 9.8 kcal.min⁻¹ recorded during the semi-mechanised technique. Psychophysical responses indicated that the workers perceived manual pitting to be more physically demanding than the semi-mechanised method. The manual and semi-mechanised planting tasks were, in general, found to be acceptable from a spinal kinematics perspective, with the majority of variables classified as low risk. However, the maximum sagittal angle was reduced by more than 20 degrees as a result of the new equipment. The physiological and psychophysical demands associated with manual planting were found to be within acceptable limits. Conclusion: In terms of pitting, it can tentatively be concluded that the semi-mechanised technique is better than the manual one, based on the biomechanical and psychophysical findings, however physiological demands require further investigation. When considering the planting techniques, the semi-mechanised method showed a slight improvement from the biomechanical perspective, but further physiological and psychophysical investigations are needed.

The addition of an A₄ family symmetry and extended Higgs sector to the Standard Model can generate this mixing pattern, assuming the correct vacuum expectation value alignment of Higgs scalars. The effect of deviating this alignment is studied, for different types of A₄ models, with a phenomenological emphasis: the effect of perturbations on the model predictions for the neutrino oscillation and neutrino mass observables. The standard theoretical description of neutrino oscillations is presented, along with a summary of the past, present and future experimental efforts aimed at measuring the neutrino mixing parameters. Additionally, the current constraints on the sum of absolute neutrino masses and the amplitude for neutrinoless double beta decay, which is yet to be observed, are discussed. These constraints provide a model-independent test of family symmetery models. The Standard Model is reviewed, and extensions to the Standard Model such as the seesaw mechanism(s) are discussed: these are designed to endow neutrinos with mass, and can be incorporated into A₄ symmetry models. Chapter 6 contains the analysis of six models that assign all leptons to the 3 (underlined) representation, with four of these utilising the seesaw mechanism. The models are tested for any degree of fine tuning of the parameters that define the mass matrices. The effect of perturbations on the mixing angle observables, in particular sin² ∅₁₃ and sin² ∅₂₃, is studied, as well as the effect on the Jarlskog invariant, Jcp. - Description: The human skin blanching assay (HSBA) is a well-researched and validated method for the bioequivalence assessment of topical corticosteroids. Traditionally, visual assessment of skin blanching has been used. Such testing methods are not conducive for interlaboratory comparisons. Regulatory bodies prefer less subjective methods of analysis. The FDA released guidelines on the assessment of bioequivalence for topical corticosteroids that recommends the use of a chromameter as a reliable method to measure skin blanching although the use of visual assessment with acceptable validation is also provided for. However, the FDA does not elucidate on the manipulation and handling of the chromameter during skin blanching measurements. The purpose of this project was several fold, which included investigations to standardize the manipulation and handling of a chromameter. In particular, measures to avoid skin whitening resulting from the effects of pressure on the skin during chromameter use were investigated. Other methods of analysis should surpass or at least be comparable to the HSBA if such methods are to be used for the assessment of topical corticosteroids. Microdialysis is a relatively new technique for assessing the rate at which drug penetrates the skin. The advantage of using this method is that there are fewer restrictions for selection of an appropriate study population unlike those required for the HSBA where one has to be both a ‘responder’ and a ‘detector’ for their results to be used in data analysis. Microdialysis was investigated by initially conducting experiments in which microdialysis probes were embedded into topical formulations containing mometasone furoate (MF) and the initial results revealed that relatively low drug was released from the formulations. These results indicated that should microdialysis be applied to measure the in vivo release of MF from such topical formulations following application to the skin, even lower concentrations of MF would likely result in the dialysate, necessitating the need for ultra-high sensitive methods of analysis. Typically, the availability of an appropriate analytical technique such as liquid chromatography coupled with mass spectrometry (LCMS) would be a pre-requisite for such in vivo studies. However, only high-pressure liquid chromatography (HPLC) and other less sensitive equipment was available in the laboratories. The study objectives were therefore focussed on in vitro assessment of the release of MF from topical formulations using microdialysis and Franz cells. In addition, the in vivo release of MF was also studied using the HSBA. Data obtained from the microdialysis experiments were compared with the data obtained from the Franz cell diffusion studies in order to provide information on the pharmaceutical availability of MF from the various topical MF dosage forms. Subsequently, pharmaceutical equivalence was investigated from the comparative pharmaceutical availability data using statistical analysis. An additional objective was to attempt to correlate in vitro with in vivo data (IVIVC) to establish a model that could be used to assess safety and efficacy of generic topical drug products. The in vivo data obtained from the HSBA were processed according to the FDA requirements and these pharmacodynamic data were subsequently compared with the microdialysis and Franz cell results. In summary the objectives of this project were: 1. To develop a system to improve the reproducibility of the use of a Minolta® chromameter and compare this with the standard/normal manipulation and handling of such instruments. 2. To develop and validate an HPLC method for the analysis of MF for use with in vitro diffusion studies using microdialysis and Franz cells. 3. To conduct a comparative HSBA on proprietary MF topical creams from two different countries in accordance with the FDA guidance. 4. - Description: The Nelson Mandela Metropole is the largest contributor to the Eastern Cape Province’s economy and its share of the production of the Eastern Cape is about 41, 3 % per annum. This requires that the competitiveness of the Metropole be improved in order to enable it to meet the challenges of the Province. The objective of the GEAR policy strategy places a focus on the leading coastal industrial locations of South Africa. Furthermore, the national government has shifted the responsibility of service delivery to local governments in an effort to reinforce the integrated development planning process in municipalities. To meet those challenges, the Metropole has focused its attention on improving local resources. To enhance the performance of the manufacturing industry of the Metropole, it is necessary to ensure that the Metropole has locational attributes to attract new investment. Such attributes include creation of closely located industries. This will help the firms to engage in competition as well as engage in co-operative activities among themselves. The study employs a variety of theories to highlight the need to enhance productivity of industries in order to attract new investment. These theories include the new (endogenous) growth theory that argues that productivity growth is determined by introduction of new technologies. Such technologies accrue because ideas that contribute to their development are nonrival, and thus their creation has a fixed cost and zero marginal cost. The property of fixed cost in the creation of ideas results in the emergence of increasing returns to scale. The Porter’s Diamond framework is used in the study to take the issue of productivity growth further. Its thrust is that in particular nations some industries experience high productivity growth rates. It further argues that the locational attributes are responsible for these industries in registering high productivity growth levels. These attributes include the creation of advanced resources such as a skilled labour force. The data obtained from the survey of the research on the manufacturing industries forms part of this study. The findings of the survey reveal that although the manufacturing sector of the South African region has registered some significant success, there is still some room for improving its competitiveness. It revealed that development of the local markets through competition and cooperation among the industries would help to render these industries internationally competitive. This study concludes with some recommendations. These recommendations place emphasis on improvement of infrastructure, quality of labour force, and development of the region’s market. In order to carry out the recommendations effectively, government policy has to be repositioned so as to enhance its visibility among the stakeholders in the economy. Of importance is to ensure the promotion of policy that supports geographically concentrated businesses.

Serotonin binding in vitro by releasable proteins from human blood platelets

- Description: Among the substances released from human blood platelets are serotonin and various proteins. It was hypothesized that one of these proteins binds serotonin and that serotonin might be important to the protein's function or that the protein might be important to serotonin's function. Two platelet-specific proteins, platelet factor 4 (PF4) and ß-thromboglobulin ( ßTG) were found to bind serotonin in vitro. Endogenous PF4 was isolated by serotonin-affinity chromatography and was identified by radioimmunoassay. Purified l¹²⁵ I] -PF4 and native PF4 bound to and eluted from a serotonin-affinity column similarly. Ultrafiltration of the homologous protein, (ßTG, with [¹⁴C]-serotonin demonstrated binding of about 8 moles serotonin per mole tetrameric ßTG with a dissociation con stant of about 4 x 10-8ThesisThesis⁻⁸ M. Equilibrium dialysis of PF4 with radiolabelled serotonin was attempted, but no binding constant values were obtained because serotonin apparently bound to the dialysis membrane. Since EDTA was one of the two agents that eluted PF4 from the serotonin-affinity gel, calcium binding by -PF4 was investigated by equilibrium dialysis. Evidence was obtained for positively cooperative binding of calcium ions by PF4. - Description: Among the substances released from human blood platelets are serotonin and various proteins. It was hypothesized that one of these proteins binds serotonin and that serotonin might be important to the protein's function or that the protein might be important to serotonin's function. Two platelet-specific proteins, platelet factor 4 (PF4) and ß-thromboglobulin ( ßTG) were found to bind serotonin in vitro. Endogenous PF4 was isolated by serotonin-affinity chromatography and was identified by radioimmunoassay. Purified l¹²⁵ I] -PF4 and native PF4 bound to and eluted from a serotonin-affinity column similarly. Ultrafiltration of the homologous protein, (ßTG, with [¹⁴C]-serotonin demonstrated binding of about 8 moles serotonin per mole tetrameric ßTG with a dissociation con stant of about 4 x 10-8ThesisThesis⁻⁸ M. Equilibrium dialysis of PF4 with radiolabelled serotonin was attempted, but no binding constant values were obtained because serotonin apparently bound to the dialysis membrane. Since EDTA was one of the two agents that eluted PF4 from the serotonin-affinity gel, calcium binding by -PF4 was investigated by equilibrium dialysis. Evidence was obtained for positively cooperative binding of calcium ions by PF4. The taxonomy and osteology of fishes of the family Tripterygiidae (Perciformes : Blennioidei) of South Africa

- Description: This study is divided into two parts. The first deals with the taxonomy of the South African fishes of the Tripterygiidae. The second part describes the osteology of one genus of the family, and draws comparisons with the other genera discussed in this study. Five genera of Tripterygiidae are recognized from South African waters. Cremnochorites, a monotypic genus, is described as new. The single species, C. capensis, has been recorded only from the southern and south-eastern coast of South Africa. It is distinguished from other genera by a combination of features which includes scalation, dorsal and anal fin spine counts, and various osteological characters. Three genera, Norfolkia Fowler, Helcogramma McCulloch & Waite, and Enneapterygius Rüppell occur throughout most of the Indo-Pacific. A single species of Norfolkia, N. springeri Clark (in press) is found in Zululand. Two species are ascribed to Helcogramma, H. obtusirostre (Klunzinger) and H. fuscopinna sp.n. Parallels are drawn between two species of Tripterygion Risso, T. tripteronotus and T. delaisi from the Mediterranean. The two South African Helcogramma species show similar depth preferences to the two Tripterygion species, resulting in similar morphological differences between the two species of each pair. The genus Enneapterygius Rüppell is divided into two genera, Enneanterygius and Scoliosolen gen.n. The division is based on the form of the supraoccipital sensory canal and associated osteological characters. Sooliosolen has a crescent-shaped supraoccipital canal and cranial osteology similar to the majority of other tripterygiid genera, whereas Enneapterygius has a 'U'-shaped supraoccipital canal which curves around the first dorsal fin, a comparatively long, concave supraoccipital bone which extends anteriorly between the parietal and between the posterior ends of the frontals. Two species are referred to Scoliosolen, S. abeli (Klausewitz) and S. conspicuus (Clark), and two new species are described for Enneapterygius, E. pulcherrimus and E. trianeulus. A literature survey revealed little consistency in ascribing species to any particular genus. Thus, throughout this study an attempt is made to define the genera so that future confusion can be avoided. In the light of these definitions an assessment is made of the original descriptions of a large number of species to determine which of the species can be ascribed to Norfolkia and to Helcogramrna. This has been possible to a lesser degree for Enneapterygius and Scoliosolen, for the major external feature separating these two genera, the shape of the supraoccipital sensory canal, is described only for Red Sea (Clark, in press) and South African species (this study). This study places four species in Enneapterygius, and six in Scoliosolen. To provide a firmer foundation for defining the genera, an investigation was made of the osteology of Scoliosolen conspicuus. Enneapterygius was originally chosen for the osteolofical study as it is reputedly the largest genus of the family, and thus likely to be the most generalized. Once comparisons had been made with other Enneapterygius species, it became apparent that this genus had to be divided into two genera, Enneapterygius and Scoliosolen. It is not known whether Scoliosolen is the largest genus, but it is likely to be one of the largest once a complete survey of the species originally ascribed to Enneapterygius has been undertaken. Finally, an osteological comparison is made of the five genera which occur in South African waters to provide firmer bases for the generic definitions. Only those characters which appear to be constant within a genus are used. Reference is made to a number of genera which do not occur in South African waters, to ensure that the characters chosen cannot be applied to other genera
