Invariably, the nature of research questions led the study to be located within the pragmatic paradigm. A mixed method approach and concurrent triangulation design was adopted to examine issues under study. The study adopted stratified random sampling and purposive sampling methods to identify its respondents and participants. Data were collected from respondents who were envisaged knowledgeable about critical issues under study. Several research instruments were used to solicit quantitative and qualitative data. These included: questionnaires, interview schedules, focus group discussions and documents. The sample of the study consisted of 20 lecturers, 50 pre-service teachers, 3 Department of Teacher Education lecturers and 2 Directors in the Ministry of Higher and Tertiary Education Science Innovation, Technology and Development. The study established that 95%of the teacher educators had the requisite STEM content knowledge. Pedagogical content knowledge, knowledge of organisation and education purpose, engineering content and pedagogical content was found lacking. Furthermore, the teacher educators employed 21st century STEM specific inquiry based and constructivist teaching strategies. More so, it was established that government, the Department of Teacher Education and other development partners rendered support to teacher education colleges to prepare for pre-service teachers for STEM education. In addition, the study revealed that preparation of pre-service teachers for STEM was impeded by several structural factors that obtained in teacher education colleges. Overall, the study concluded that teacher educators had requisite STEM knowledge and employed inquiry-based strategies to prepare pre-service teachers for STEM Education. Furthermore, the study recommended that teacher educators’ knowledge in engineering needs further strengthening through workshops and synergies with industry. An alternative model for effective STEM preparation was recommended for consideration.

The characterisation of trypanosomal type 1 DnaJ-like proteins

- Description: Trypanosomes are protozoans, of which many are parasitic, and possess complex lifecycles which alternate between mammalian and arthropod hosts. As is the case with most organisms, molecular chaperones and heat shock proteins are encoded within the genomes of these protozoans. These proteins are an integral part of maintaining the structural integrity of proteins during normal and stress conditions. Heat shock protein 40 (Hsp40) is a co-chaperone of heat shock protein 70 (Hsp70) and in some cases can act as a chaperone. These proteins work together to bind non-native polypeptide structures to prevent unfolded protein aggregrate formation in times of stress, translocate proteins across organelle membranes, and transport unsalvageable proteins to proteolytic degradation by the cellular proteasome. Hsp40s are divided into four types based on their domain structure. Analysis of the nuclear genomes of eight trypanosomatid species revealed that less than 10 of the approximate 70 Hsp40 sequences per genome were Type 1 Hsp40s, many of which contained putative orthologues in the other seven trypanosomatid genomes. One of these Type 1 Hsp40s from T b. brucei, Trypanosoma brucei DnaJ 2 (Tbj2), was functionally characterised in T brucei brucei. RNA interference knockdown of expression in T brucei brucei showed that cells deficient in Tbj2 displayed a severe inhibition of the growth of the cell population. The levels of the Tbj2 protein population in T brucei brucei cells increases after exposure to 42°c and the protein was found to have a generalized cytoplasmic subcellular localization at 37°c. These findings provide evidence that Tbj2 is an orthologue of Yeast DnaJ 1 (Y dj l), an essential S. cerevisiae protein. Hsp40s interact with their partner Hsp70s through their J-domain. The amino acids of the J-domain important for a functional interaction with Hsp70 were examined in Trypanosoma cruzi DnaJ 2 (Tcj2) (the orthologue of Tbj2) and T cruzi DnaJ protein 3 (Tcj3) by testing their ability to substitute for Y dj l in Saccharomyces cerevisae and for DnaJ in Escherichia coli. In both systems, the positively charged amino acids of Helix II and III of the J-domain disrupted the functional interaction of these Hsp40s with their partner Hsp70s. Substitutions in Helix I and IV of the J-domains of Tcj2 and Tcj3 produced varied results in the two different systems, possibly suggesting that these helices serve to define with which Hsp70s a given Hsp40 can interact. The inability of an Hsp40 and an Hsp70 to interact functionally does not necessarily mean a total absence of physical interaction between these proteins. The amino acid substitution of the histidine in the HPD motif (H34Q) of the J-domain of Tcj2 and Tcj3 removed the ability of these proteins to interact functionally with S. cerevisiae Hsp70 (Ssal) in vivo. However, preliminary binding studies using the quartz crystal microbalance with dissipation monitoring (QCM-D) show that Tcj2 and Tcj2(H34Q) both physically interact with M sativa Hsp70 in vitro. - Description: The majestic mountains of the uKhahlamba Drakensberg, formed many millennia ago were home to the Bushmen[footnote 1] or San people. They lived at these mountains for thousands of years before they were colonised by the Bantu speakers and the Europeans. Academic writings for many years have perpetuated the thinking that Bushman people were largely extinct. The dominance of this view in the academic writings was encouraged by historical evidence that Europeans and Bantu speakers hunted and killed Bushmen over the last several centuries. Researchers argue that the extermination of the Bushmen was because they were less human in the eyes of the foreigners, due to cattle raiding. There is still some element of this thinking amongst today’s academics, although research in the last decade is questioning this thinking. The question of whether descendants do exist is relevant to issues of rights of access to ancestral sacred sites, in particular rock art sites. At present, access to rock art sites is granted on qualification as an authentic fee-paying tourist (or affordability) rather than on group rights to a cultural heritage resource (cultural rights). Based on this, I argue that access to rock art sites is based on qualification rather than by right. This is largely driven by an approach that emphasises the physical conservation and financial sustainability of a site, rather than its spiritual maintenance. It has become clear that the interests in rock art by tourists and Bushman descendants are distinct from each other. Tourists have an aesthetic significance for rock art while Bushmen descendants have a spiritual significance for the paintings. Beyond any doubt, the physically based and financially driven approach has brought new challenges to today’s Bushmen descendants, whom in reaffirming their identities now have a new challenge to overcome. Not only are the rock art sites physically threatened but also they have lost much of their spiritual powers. Their fate lies in the hands of heritage officers who must determine access rights to the painted shelters. Both the National Heritage Resources Act and the KwaZulu-Natal Heritage Act acknowledge living heritage. However, the existence of this heritage is judged against the physical approach to rock art management. If the practises of descendants are perceived to be a threat to the rock art, they will not be approved. The case of the Duma is a classic example. Prior to the ritual ceremony at Game Pass Shelter, Kamberg, they were informed of the minimum standards for opening a rock art site to public and rules of how people should behave while visiting painted shelters. While it was evident that there are problems with the two approaches, the spiritual and physical approach, discussed in the thesis, it is important that solutions are identified. I do not believe that one approach on its own will be good enough, for reasons discussed in the thesis. Instead, the two approaches should be implemented together to compliment each other by identifying common grounds. I provide strategies as to how I believe that such a common ground can be reached. In addition, I provide my own analytical thinking as to how these strategies can be achieved. There is no general consensus over which term is appropriate. Both terms are considered by some academics to be derogatory or pejorative (Chennels 2003). San means vagabond and was given to the Bushmen by Khoi-Khoi people, because they considered themselves of a better social class, as they had domesticated animals and were more sedentary than Bushmen. However, according to WIMSA (Thoma 2003) the word San is derived from the Hai||om language meaning “people who gather”. It is normally written Saan but it has been accepted to write San. In 1993 the San requested to be called San when referred to as an entire group. If one refers to individual people/groups they like to be called by their language and cultural name i.e. Khwe, !Kung, !Xun, Ju|’hoansi, ‡Khomani, N|u, |’Auni, Hai||om, etc In this thesis, Bushmen is a preferred term, because it is a better-known term among the people who are central to this study. The geochemical evolution of three alkaline complexes in the Kuboos-Bremen igneous province, southern Namibia

- Description: The Kuboos-Bremen Igneous Province comprises a linear zone of alkaline complexes that intrude Proterozoic and Pan-African rocks and trends in a northeast direction from the northwest of the Cape Province in South Africa into southern Namibia. Of the three most southerly complexes in Namibia. two comprise silicate rocks ranging from nepheline syenite to alkali-granite and are called the Grootpenseiland and Marinkas Kwela Complexes (GPC and MKC). The Marinkas Kwela Carbonatite Complex is the third and most northerly of the complexes. Isotopic age determinations on a number of rock types from both the silicate complexes yield ages around 520Ma and are consistent with published Pan-African ages for the Province. Each silicate complex shows a migrating locus of intrusion from Siundersaturated rocks in the southwest to Si-oversaturated rocks in the northeast. The complexes overlap in outcrop. The rocks are moderately to highly felsiC and none reflects primary magma compositions. The Si-undersaturated rocks from both complexes include side-wall cumulates formed from magmas that fractionated alkali-feldspar, clinopyroxene and amphibole. Foyaites also occur in the MKC and have a compositional range reflecting alkali-feldspar fractionation and, probably, some interaction with dolomite country rocks. Major and trace element data suggest that critically saturated alkali syenites occurring in both complexes evolved via protracted feldspar fractionation, and that critically saturated alkali-feldspar syenite occurring only in the GPC is a cumulate. The two rock types cannot be related genetically. Of the SI-oversaturated rocks in both complexes, those in the compositional range monzonite to granite were intruded before alkali-granites. Compositional diversity amongst the former reflects fractionation of feldspar and of mafic phases, but that process cannot genetically link the rocks to the alkali-granites. Isotopic compositions of Sr and Nd indicate that the silicate magmas were derived from an upper mantle source region characterised by low time-integrated Rb/Sr ratios and high time-Integrated Sm/Nd ratios, However, the evidence of Sr and 0 isotopic data is that the Si-oversaturated melts possibly interacted with a crustal component. presumably the Proterowlc rocks of the Namaqua Metamorphic Province. This interaction may explain the occurrence of apparently co-genetic rock series that evolved on opposite sides of the feldspar join in Petrogeny's Residua System. The Marinkas Kwela Carbonatite Complex was emplaced before the final intrusive phases of the MKC and exhibits unusually pronounced late-stage enrichment in manganese. The earliest intrusive rocks in the complex were nepheline syenites which were fenitised by later intrusions of sôvites. Although the commonly occurring magmatic sequence of sôvite-beforsite-ferrocarbonatite is observed at Marinkas Kwela, sôvites do not appear to have been parental to beforsites. Removal of apatite and early crystallisation of magnetite distinguish magnetite-rich beforsite from co-genetic apatite-rich beforsite. Two further magmatic sequences. the first from apatite-rich beforsite through ferrocarbonatite to Mn-rich ferrocarbonatite (high Fe/Mn) and the second from magnetite-rich beforsite to Mn-rich ferrocarbonatite (low Fe/Mn). reflect fractionation of dolomite and of dolomite+magnetite respectively.

Although the commonly occurring magmatic sequence of sôvite-beforsite-ferrocarbonatite is observed at Marinkas Kwela, sôvites do not appear to have been parental to beforsites. Removal of apatite and early crystallisation of magnetite distinguish magnetite-rich beforsite from co-genetic apatite-rich beforsite. Two further magmatic sequences. the first from apatite-rich beforsite through ferrocarbonatite to Mn-rich ferrocarbonatite (high Fe/Mn) and the second from magnetite-rich beforsite to Mn-rich ferrocarbonatite (low Fe/Mn). reflect fractionation of dolomite and of dolomite+magnetite respectively.
