- Description: Game ranching offers an economic alternative to conventional stock farming and is a means of supplementing income derived from agricultural activities. Competition is intensifying as ranchers continuously look for ways to improve and expand their businesses and gain a competitive advantage. Ranchers consequently have to find strategies that can ensure growth. Since a reciprocal relationship and dependency exists between a game ranch and the environment in which it operates, it is essential that ranchers take cognizance of aspects in the marketing environment that can either limit, and hence encumber the choice of growth strategy, or stimulate the growth potential of game ranches. As the objective of marketing is to offer customers superior value, it is pertinent for ranchers to know and understand the needs, requirements and motivations of visitors. Failure to do so will cause visitors to patronise other game ranches, or alternative tourism offerings that meet their expectations. A literature review was conducted to define strategy and assist in identifying growth strategies available to a game ranch. The literature review assisted in categorising elements in the external, internal and customer analysis phases of strategic marketing management. Surveys were conducted by means of self-completion questionnaires which were distributed throughout South Africa. Two sets of questionnaires were distributed - one to respondents with prior experience of a game ranch, and one to respondents with no-prior experience. A qualifying question directed the fieldworker to decide which questionnaire should be completed by the respondent. The surveys were constructed in such a way as to determine to what extent various factors would persuade respondents to visit a ranch or extend their stay. Based on a factor analysis, an eight factor structure emerged which represented the personal values of respondents and their levels of expectation for each of the factors. The eight factors were stimulation, universalism, tradition, security, hedonism, power, socialisation, and self-direction. The study revealed the following. - The majority of respondents with no-prior experience of a game ranch is either unaware of the offerings of game ranches, or perceives them as too expensive. - The sustainable use of natural resources is not only essential for the existence of a game ranch, but is also an important motivator for respondents to either visit a game ranch or extend their stay at a ranch. - Staff play a critical role in the overall experience of visitors and act as an important motivator to either visit/revisit a game ranch or extend their stay at the ranch. - Clean and comfortable overnight facilities are deemed important by all respondents. - Respondents have a strong desire to relax and spend time with family and friends. A decision-making framework from which game ranches can select applicable growth strategies resulted from the study. The decision-making framework comprises a three-phase process; phase 1 is the identification of a game ranch's target market; phase 2 is the use of a simplistic framework to determine a possible growth strategy for a game ranch; phase 3 is the identification of ways to implement the selected strategy. As the game ranch industry makes significant contributions to the economic, social and ecological welfare of South Africa, this study added to the body of knowledge of game ranching in South Africa, particularly with regard to the growth options available to game ranches. Recommendations for consideration of future research included the following. - As the current study investigated hunting and tourism, future research on growth strategies for game ranches could also include other aspects of game ranching such as the sale of live game and game products. - More focused empirical research into external and internal strategic analysis should be considered to supplement the empirical research into customer analysis resulting from the current study. - Finally, additional information obtained on the external and internal strategic analysis process could lead to expanding the decision-making framework developed in this study into a more holistic tool to be used in selecting a growth strategy, or combination of strategies for a game ranch3.

- Description: Escherichia coli (E. coli) is a common inhabitant of surface waters in the developed and developing worlds. The majority of E. coli cells present in water are not particularly pathogenic to humans; however, there are some present in small proportion that possess virulence genes that allow them to colonize the digestive tract. Pathogenic E. coli causes acute and chronic diarrheal diseases, especially among children in developing countries and in travelers in these locales. The present study, conducted between August 2007 and July 2008, investigated the prevalence and distribution of virulent E. coli strains as either free or attached cells in the final effluents of three wastewater treatment plants located in the Eastern Cape Province of South Africa and its impact on the physico-chemical quality of the receiving water body. The wastewater treatment plants are located in urban (East Bank Reclamation Works, East London), peri-urban (Dimbaza Sewage Treatment Works) and in rural area (Alice Sewage Treatment Works). The effluent quality of the treatment plants were acceptable with respect to pH (6.9-7.8), temperature (13.8-22.0 °C), dissolved oxygen (DO) (4.9-7.8 mg/L), salinity (0.12-0.17 psu), total dissolved solids (TDS) (119-162 mg/ L) and nitrite concentration (0.1-0.4 mg/l). The other xii physicochemical parameters that did not comply with regulated standards include the following: phosphate (0.1-4.0 mg/L); chemical oxygen demand (COD) (5-211 mg/L); electrical conductivity (EC) (237-325 μS/cm) and Turbidity (7.7-62.7 NTU). Results suggest that eutrophication is intensified in the vicinity of the effluent discharge points, where phosphate and nitrate were found in high concentrations. Presumptive E. coli was isolated from the effluent samples by culture-based methods and confirmed using Polymerase Chain Reaction (PCR) techniques. Antibiogram assay was also carried out using standard in vitro methods on Mueller Hinton agar. The viable counts of presumptive E. coli for the effluent samples associated with 180 μm plankton size ranged between 0 – 4.30 × 101 cfu/ml in Dimbaza, 0 – 3.88 × 101 cfu/ml in Alice and 0 – 8.00 × 101 cfu/ml in East London. In the 60 μm plankton size category E. coli densities ranged between 0 and 4.2 × 101 cfu/ml in Dimbaza, 0 and 2.13 × 101 cfu/ml in Alice and 0 and 8.75 × 101 cfu/ml in East London. Whereas in the 20 μm plankton size category presumptive E. coli density varied from 0 to 5.0 × 101 cfu/ml in Dimbaza, 0 to 3.75 × 101 cfu/ml in Alice and 0 to 9.0 × 101 cfu/ml in East London. The free-living presumptive E. coli density ranged between 0 and 3.13 × 101 cfu/ml in Dimbaza, between 0 and 8.0 × 101 cfu/ml in Alice and between 0 and 9.5 × 101 cfu/ml in East London. Molecular analysis successfully amplified target genes (fliCH7, rfbEO157, ial and aap) which are characteristic of pathogenic E. coli strains. The PCR assays using uidA-specific primer confirmed that a genetic region homologous in size to the E. coli uidA structural gene, including the regulatory region, was present in 3 of the E. coli isolates from Alice, 10 from Dimbaza and 8 from East London. Of the 3 E. coli isolates from Alice, 1 (33.3%) was positive for the fliCH7 genes and 3 was positive for rfbEO157 genes. Out of the 10 isolates from Dimbaza, 4 were xiii positive for fliCH7 genes, 6 were positive for the rfbEO157 genes and 1 was positive for the aap genes; and of the 8 isolates from East London, 1 was positive for fliCH7 genes, 2 were for the rfbEO157 genes, 6 were positive for the ial genes. Antimicrobial susceptibility profile revealed that all of the E. coli strains isolated from the effluent water samples were resistant (R) to linezolid, polymyxin B, penicillin G and sulfamethoxazole. The E. coli isolates from Dimbaza (9/10) and East London (8/8) respectively were resistant to erythromycin. All the isolates were found to be susceptible (S) to amikacin, ceftazidime, ciprofloxacin, colistin sulphate, ceftriaxone, cefotaxime, cefuroxime, ertapenem, gatifloxacin, gentamycin, imidazole, kanamycin, meropenem, moxifloxacin, neomycin, netilmicin, norfloxacin and tobramycin. The findings of this study revealed that the Alice wastewater treatment plant was the most efficient as it produced the final effluent with the least pathogenic E. coli followed by the Dimbaza wastewater treatment plant. In addition, the findings showed that the wastewater treatment plant effluents are a veritable source of pathogenic E. coli in the Eastern Cape Province watershed. We suggest that to maximize public health protection, treated wastewater effluent quality should be diligently monitored pursuant to ensuring high quality of final effluents.

- Description: In South Africa the pharmacy profession has experienced a number of changes around the turn of the century such as the introduction of the National Drug Policy (NDP), pharmacy ownership and price regulation. With this the role and earnings of the pharmacy profession, as well as to what extent the pharmacist adds value to the profession and society, are being questioned. Community pharmacists are thus faced with the challenge to prove that the value that they add to society is meaningful. Therefore, the aim of the study was to document community pharmacy availability and activities in South Africa and based on this to quantify the perceived value that the community pharmacist adds to society through the delivery of pharmaceutical services and pharmaceutical care. In order to determine the pharmacist’s true value added two surveys were conducted in 2006; an original pharmacist survey and a general public survey. The results obtained were verified by a follow–up pharmacist survey in 2009 to confirm or reject the results obtained in the original survey. The study was representative of both the community pharmacies and the general public in South Africa and was primarily quantitative in design and analysis. More than half of the responding pharmacies (63.16%) were open seven days a week. The average hours of service per day ranged from 10 hours (Monday to Friday) through to 6.45 hours on Saturdays and 3 hours on Sundays. Pharmacists continuously upgraded their professional knowledge. More than three quarters of pharmacies had the necessary equipment available to perform the services investigated in the study. The general public was not aware of all the services provided by pharmacists and as a result, depending on the service, many people did not make use of these services. The general public that made use of services delivered by community pharmacies mainly perceived the services delivered to be of good quality. The main barrier to practicing pharmaceutical care was indicated by pharmacists as not receiving payment for the advice given followed by pharmaceutical care being time consuming, and that there was not enough time to talk to patients. The general public indicated that they found it difficult to ask questions in pharmacies because other patients could hear what was discussed, or other patients had to wait longer if they asked something, and pharmacy staff being too busy. The results of the original pharmacist and the general public survey were confirmed by the results of the follow-up survey with the exception of dispensing prescription medicine (8 minutes 28 seconds), OTC medicine (7 minutes 23 seconds), counselling of prescription medicine (8 minutes 51 seconds) and OTC medicine (8 minutes) which on average took longer to conduct than in the previous analysis. The study highlighted the value added to the wellness and quality of life of the community of South Africa through the delivery of pharmaceutical care and pharmaceutical services by community pharmacy staff, and proved that pharmacists are committed to the provision of pharmaceutical care and pharmaceutical services.

- Description: β-N-methylamino-L-alanine (BMAA) is a non-proteinogenic amino acid associated with human neurodegenerative disease. Due to the cosmopolitan nature of cyanobacteria, detection of BMAA in cyanobacteria has caused concerns about human exposure risk. This study was therefore based on the hypothesis that BMAA poses a health risk to humans either by direct ingestion or by indirect exposure to BMAA from a cyanobacterial source via a freshwater food chain. A validated gas chromatography-mass spectrometry (GC-MS) BMAA analysis method and a confirmatory liquid chromatography-mass spectrometry (LC-MS) method, with improved sensitivity, were developed in addition to a LC-MS/MS method for analyte confirmation. These methods were used to quantify BMAA in South African cyanobacteria, isolated from various potable water reservoirs. The majority of the isolates tested, contained BMAA. Possible human exposure by direct consumption of BMAA released from cyanobacterial blooms was investigated by the development of a robust solid phase extraction (SPE) method used for BMAA concentration and quantification in raw and treated tap water. Despite the use of the SPE method that facilitated the concentration of BMAA from large quantities of water, no free dissolved BMAA was detected in raw or processed fresh water. The fate of exogenous BMAA was therefore investigated firstly by evaluating the efficacy of standard water treatment processes employed in South Africa and secondly by investigating the possibility of BMAA bioaccumulation and biomagnification in aquatic food chains. Standard water treatment processes proved highly efficient at removing free dissolved BMAA, explaining the absence of BMAA in treated tap water. However, the cause of the BMAA absence in raw potable water remained unknown. Uptake of BMAA by model aquatic organisms was investigated in controlled experiments. BMAA uptake was documented in both Ceratophyllum demersum and Daphnia magna, however, BMAA-protein association and biomagnification were not observed in D. magna. BMAA had an inhibitory effect on the oxidative stress enzyme acitivties of both organisms tested (as well as human S9 extracts), resulting in accumulation of detrimental reactive oxygen species (ROS) in the cells. Exposure of crop plants to BMAA in controlled experiments resulted in BMAA uptake, protein association, and subsequent inhibition of the antioxidative enzyme activities. However, BMAA was detected in neither free nor protein-associated form in natural crop plants irrigated with known BMAA-containing bloom water. Post-mortem liver samples of Clarias gariepinus (Catfish) and Crocodylus niloticus (Crocodile), from a natural fresh water ecosystem that experienced frequent cyanobacterial blooms, contained both free and protein-associated BMAA. Higher BMAA concentrations were found in crocodile liver samples compared to fish liver samples, strongly suggesting biomagnification from one trophic level to the next. BMAA concentrations corresponded to crocodile age. This is the first report of bioaccumulation and biomagnification in two trophic levels in a fresh water ecosystem. These findings strongly suggest possible human exposure via aquatic food chains of cyanobacterial origin. Direct BMAA exposure via drinking water is not plausible due to the efficiency of standard water treatment processes to remove BMAA. The use of raw water for agricultural and recreational use, however, remains a problem. The development of management strategies as well as daily tolerable levels for BMAA is urgently required.

- Description: This thesis creates a basic framework and provides the information necessary to create a more accurate description of the topside ionosphere in terms of the altitude variation of the electron density (Ne) over the South African region. The detailed overview of various topside ionospheric modelling techniques, with specific emphasis on their implications for the efforts to model the South African topside, provides a starting point towards achieving the goals. The novelty of the thesis lies in the investigation of the applicabilityof three different techniques to model the South African topside ionosphere: (1) The possibility of using Artificial Neural Network (ANN) techniques for empirical modelling of the topside ionosphere based on the available, however irregularly sampled, topside sounder measurements. The goal of this model was to test the ability of ANN techniques to capture the complex relationships between the various ionospheric variables using irregularly distributed measurements. While this technique is promising, the method did not show significant improvement over the International Reference Ionosphere (IRI) model results when compared with the actual measurements. (2) Application of the diffusive equilibrium theory. Although based on sound physics foundations, the method only operates on a generalised level leading to results that are not necessarily unique. Furthermore, the approach relies on many ionospheric variables as inputs which are derived from other models whose accuracy is not verified. (3) Attempts to complement the standard functional techniques, (Chapman, Epstein, Exponential and Parabolic), with Global Positioning System (GPS) and ionosonde measurements in an effort to provide deeper insights into the actual conditions within the ionosphere. The vertical Ne distribution is reconstructed by linking together the different aspects of the constituent ions and their transition height by considering how they influence the shape of the profile. While this approach has not been tested against actual measurements, results show that the method could be potentially useful for topside ionospheric studies. Due to the limitations of each technique reviewed, this thesis observes that the employment of an approach that incorporates both theoretical onsiderations and empirical aspects has the potential to lead to a more accurate characterisation of the topside ionospheric behaviour, and resulting in improved models in terms of reliability and forecasting ability. The point is made that a topside sounder mission for South Africa would provide the required measured topside ionospheric data and answer the many science questions that this region poses as well as solving a number of the limitations set out in this thesis.

Characterisation of dissimilar friction stir welds between 5754 Aluminium alloy and C11000 copper

- Description: Friction Stir Welding (FSW) is a solid state welding process invented and patented by The Welding Institute (TWI) in 1991, for joining ferrous and non-ferrous materials1. The FSW of Aluminium and its alloys has been commercialised; and recent interest is focused on joining dissimilar materials. However, in order to commercialise the process, research studies are required to characterise and establish process windows. This research work through material characterisation of the welded joints establishes a process window for the Friction Stir welding of 5754 Aluminium Alloy and C11000 Copper. Furthermore, preliminary studies83,85 on the FSW of aluminium and copper have revealed the presence of intermetallic compounds which are detrimental to the weld qualities. This research work is also aimed at establishing process parameters that will result in limited or no intermetallic formation in the weld. The joint integrity of the resulting welds will also be correlated with the input process parameters. Based on the preliminary investigations conducted, a final weld matrix consisting of twenty seven welds was produced by varying the rotational speed between 600 and 1200 rpm, and the feed rate between 50 and 300 mm/min using three different shoulder diameter tools – 15, 18 and 25 mm to compare the heat input into the welds and to achieve the best results. The welds were characterised through microstructural evaluation, tensile testing, microhardness profiling, X-Ray Diffraction analysis, electrical resistivity and statistical analysis – in order to establish the interrelationship between the process parameters and the weld qualities. viii Microstructural evaluation of the weld samples revealed that the interfacial regions are characterised by mixture layers of aluminium and copper; while 33 percent of the tensile samples are within the acceptable range (> 75 percent joint efficiency). High Vickers microhardness values were measured at the joint interfaces, which corresponded with the intermetallic compounds. The Energy Dispersive Spectroscopy analysis revealed the presence of thin layers of intermetallics in nanoscale at the interfacial regions. The diffractograms of the X-Ray Diffraction analysis showed small peaks for intermetallics in some of the welds. Low electrical resistivities were measured at the joint interfaces. The statistical analysis showed that the downward vertical force, (Fz) can significantly influence the resulting weld qualities. An overall summary of the analysis of the weld qualities - with respect to the shoulder diameter tools employed showed that the 18 mm shoulder diameter tool is most appropriate among the three shoulder diameters considered, and a process window of medium spindle speed of 950 rpm and low-to-medium feed rate between 50 and 150 mm/min is established for FSW of Aluminium and Copper. Welds produced at 1200 rpm and 300 mm/min with low heat input did not have intermetallics formed at the joint interface.

- Description: Variations in the yield, chemical composition, antibacterial, and antioxidant properties of the essential oils of Rosmarinus officinalis L. cultivated in Alice, Eastern Cape of South Africa over a period of 12 months using the solvent-free microwave extraction and traditional hydrodistillation methods were evaluated. The GC-MS analyses of the essential oils revealed the presence of 33 compounds with 1,8-cineole, a-pinene, camphor, verbenone, bornyl acetate and camphene constituting about 80 percent of the oils throughout the period of investigation, with the solvent-free microwave extraction method generally yielding more of the major components than the hydrodistillation method. Each of the major components of the oils varied in quantity and quality of yield at different periods of the year. The method of extraction and time of harvest are of importance to the quantity and quality of essential oil of Rosmarinus officinalis. Higher amounts of oxygenated monoterpenes such as borneol, camphor, terpene- 4-ol, linalool, a-terpeneol were present in the oil of SFME in comparison with HD. However, HD oil contained more monoterpene hydrocarbons such as a-pinene, camphene, β-pinene, myrcene, a-phellanderene, 1,8-cineole, trans- β-ocimene, γ-teprinene, and cis-sabinene hydrate than SFME extracted oil. Accumulation of monoterpene alcohols and ketones was observed during maturation process of Rosmarinus leaves. Quantitative evaluation of antibacterial activity, minimum inhibitory concentration values were determined using a serial microplate dilution method. The essential oils obtained using both methods of extraction were active against all the bacteria tested at a concentration of 10 mg mL-1. The minimum inhibitory concentrations for the SFME extracted oils ranged between 0.23 and 1.88 mg mL-1, while those of the HD extracted oils varied between 0.94 and 7.5 mg mL-1, thus suggesting that the oil obtained by solvent free microwave extraction was more active against bacteria than the oil obtained through hydrodistillation. The antioxidant and free radical scavenging activity of the obtained oils were tested by means of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH+) assay and β- carotene bleaching test. In the DPPH+ assay, while the free radical scavenging activity of the oil obtained by SFME method showed percentage inhibitions of between 48.8 percent and 67 percent, the HD derived oil showed inhibitions of between 52.2 percent and 65.30 percent at concentrations of 0.33, 0.50 and 1.0 mg mL-1, respectively. In the β-carotene bleaching assay, the percentage inhibition increased with increasing concentration of both oils with a higher antioxidant activity of the oil obtained through the SFME than the HD method. Thin layer chromatography (TLC) was used to analyze the chemical composition of the extracts using three eluent solvent systems of varying polarities i. e. CEF, BEA and EMW and sprayed with vanillin-sulfuric acid. The chemical composition of the different extracts was similar with the exception of methanol and water extracts which had only one or two visible compounds after treating with vanillin-spray reagent. To evaluate the number of antibacterial compounds present in the fractions, bioautography was used against two most important nosocomial microorganisms. S. aureus (Gram positive) and E. coli (Gram negative). Nearly all the crude serial extraction fractions contained compounds that inhibited the growth of E. coli. The hexane extract had the most lines of inhibition followed by ethyl acetate. Bioassay-guided fractionation against E. coli was used to isolate antibacterial compounds. The largest number of antibacterial compounds occurred in the hexane fraction. Furthermore we tried to complete the characterization by extracting and studying other biologically important plant metabolites such as phenolic compounds to evaluate the antioxidant capacity of Rosmarinus extracts.

Contrary to modernists‟ arguments (that indigenous African political institutions, of which the chieftainship is part, are incompatible with liberal democracy since they are, inter alia, hereditary, they compete with their elective counterparts for political power, they threaten the democratic consolidation process, and they are irrelevant to democratising African systems), this study finds that these arguments are misplaced. Instead, chieftainship is not incompatible with liberal democracy per se. It supports the democratisation process (if the governing parties pursue friendly and accommodative policies to it) but uses its political agency in reaction to the policies of ruling parties to protect its survival interests, whether or not this undermines democratic consolidation process. The chieftainship has also acted to defend democracy when the governing party abuses its political power to undermine democratic rule. It performs important functions in the country. Thus, it is still viewed by the country‟s political leadership, academics, civil society, and councillors as legitimate and highly relevant to the Lesotho‟s contemporary political system. Because of the inadequacies of the government policies and the ambiguous chieftainship-councils integration model, which tend to marginalise the chieftainship and threaten its survival, its relationship with the councils was initially characterised by conflict. However, this relationship has improved, due to the innovative actions taken not by the central government, but by the individual Councils and chiefs themselves, thus increasing the prospects for democratic consolidation. I argue for and recommend the adoption in Lesotho of appropriate variants of the mixed government model to integrate the chieftainship with the elected councils, based on the re-contextualised and re-territorialised conception and practice of democracy, which eschews its universalistic EuroAmerican version adopted by the LCD government, but recognises and preserves the chieftainship as an integral part of the Basotho society, the embodiment of its culture, history, national identity and nationhood.

- Description: This study explored the balancing out of the rights associated with terrorist and counter-terrorist attacks by using descriptive case studies of the US 9/11 attacks; and the counter-terrorist attacks on Afghanistan and Iraq. The research was conducted within a critical theory paradigm, drawing on the ideas of Habermas and other Critical Theorists. The research design was influenced by the securitive and ideological nature of the topic and it was decided that an extensive review of literature would be more suitable than a field study. A small number of interviews added to the richness of the data. Human rights, needs and international relations were investigated to serve as a theoretical starting-point for the study (Chapter 2). The case studies were subsequently explored against the background of this theoretical approach. This thesis therefore assessed the impact of human rights law on terrorism and counter-terrorism attacks under the Human Rights Act (1998). It considered how the provisions of the Human Rights Act have influenced the formulation and interpretation of anti-terrorism laws, and it examined the role of the judiciary in adjudicating disputes between the individual and the state. It further discussed human needs and the progress on human rights, terrorist attacks, as well as counter-terrorism attacks. Extensive data was gathered on the 9/11 attacks, and it was concluded that these attacks fall within the definition of crimes against humanity under international human rights jurisprudence. To bring about a truly secure world we must adopt a new paradigm that shifts priority to the security of the individuals and of communities to achieving human security, the honouring of human rights, and respect for the rule of law. This will obviously require a renewed commitment by all individuals and a shared sense of responsibility for all people, all over the world. What we need now is a major course correction – a new iii A. Fabbriciani approach which begins with a broader understanding of what defines human rights and the rule of law (Wilson, 2007). The study also focuses on counter-terrorist attacks in Afghanistan and Iraq (Chapters 3 and 4). It was shown that counter-terrorist attacks had an effect on the global economic system and development policies, which have been dominated by ideological strategies for many years. However, resistance has come from Islamic states, which have realised that new-liberal economic practices are incompatible with their theological and economic traditions. This has caused a situation to rethink global development programmes by political leaders, and to move away from new-liberal schemes towards true global development strategies. One of the main findings of the study was that the crimes of persecution and torture on the basis of political or religious views have been perpetrated by both parties, namely Al-Qaeda, and the US and its allies. It has been shown that the explored acts of terrorism and counter-attacks represent crimes against humanity, as defined by the relevant provisions of international law.

- Description: The major socio-economic development challenges facing most African countries include economic diversification, poverty, unemployment, diseases and the unsustainable use of natural resources. The challenge of quality healthcare provisioning is compounded by the HIV/AIDS pandemic in Sub Saharan Africa. However, there is a great potential in using electronic healthcare (e-health) as one of the supportive systems within the healthcare sector to address these pressing challenges facing healthcare systems in developing countries, including solving inequalities in healthcare delivery between rural and urban hospitals/clinics. The purpose of this study was to compile a Provincial E-health Framework (PEHF) based on the feedback from electronic healthcare readiness assessments conducted in selected rural and urban hospitals/clinics in the North West Province in South Africa. The e-healthcare readiness assessment was conducted in the light of effective use of ICT in patient healthcare record system, consultation among healthcare professionals, prescription of medication, referral of patients and training of healthcare professionals in ICT usage. The study was divided into two phases which were phases 1 and 2 and a qualitative design supported by a case study approach was used. Data were collected using different techniques to enhance triangulation of data. The techniques included group interviews, qualitative questionnaires, photographs, document analysis and expert opinions. The outcome of the assessment led to the compilation of the PEHF which was based on Service Oriented Architecture (SOA). SOA was chosen to integrate the hospitals/clinics‟ ICT infrastructure yet allowing each hospital/clinic the autonomy to control its own ICT environment. To assist hospitals/clinics integrate their ICT resources, this research study proposed an Infrastructure Network Architecture which clustered hospitals/clinics to share common ICT infrastructure instead of duplicating these resources. Furthermore, processes of the e-health services (e-patient health IV record system, e-consultation system, e-prescription system, e-referral system and e-training system) were provided to assist in the implementation of the PEHF. Finally, a set of guidelines were provided by the research study to aid the implementation of the PEHF.

- Description: The global number of obese people has reached pandemic proportions. High caloric diets and reduced physical exercise are to blame for this growing epidemic. Obesity has a very complex association with several other metabolic disorders, such as insulin resistance (IR), diabetes mellitus type 2 (DMT2) and cardiovascular disease. This puts a huge burden on health care systems world wide and claims many lives. Sutherlandia frutescens is a traditionally used herb, which is known to have anti-diabetic properties. However, the direct mode of action of S. frutescens still remains to be elucidated. The aim of this study was to investigate the developmental stages of high fat diet (HFD)-induced IR, to illuminate the pathogenesis of IR with a focal point on modifications in the lipid metabolism. Furthermore, the effects of S. frutescens as a treatment or prevention drug for IR and associated metabolic changes were examined. Two sets of experiments were conducted on male Wistar rats. In the first experiment rats, one week post weaning received a low fat diet (LFD), high fat diet (HFD) or HFD supplemented with S. frutescens (50mg/kg BW/d). Rats were sacrificed at week 0, 1, 2, 4, 8 and 12 in the feeding regime. In a second experiment rats were fed with a LFD or a HFD for 12 weeks and treated thereafter with S. frutescens (50mg/kg BW/d), metformin (13mg/kg BW/d) or water (control) for 28 days. Rats in the second experiment were sacrificed at week 12 to confirm IR while concurrently run rats were sacrificed after 28 days of treatment. For all the experiments rats were anaesthetized, blood was removed and rats were dissected. Plasma samples were analyzed for insulin, glucose, blood lipid parameters and cytokines. Liver, muscle and adipose tissue were analyzed for glucose uptake, total lipid content, lipid profile and fatty acid profile. It was shown that the intake of HFD caused IR and hyperinsulinaemia. The developmental stages in experiment one confirmed that an increase in plasma free fatty acids preceeded the onset of IR. Plasma and tissue lipid parameters (free fatty acid-, triglyceride- and cholesterol concentrations) showed pathological modifications in the HFD group. An ectopic accumulation of fat was observed in muscle and liver, as well as a change in membrane fatty acid profile. The results for circulating cytokines were somewhat inconclusive. Rats supplemented with S. frutescens did not develop HFD-induced IR (study one) or IR was reversed (study two). S. frutescens treatment also resulted in positive changes in plasma and tissue lipid parameters. In summary, an animal model for HFD-induced IR was established and the detrimental effect of elevated plasma FFA on glucose and lipid metabolism was observed. A novel discovery suggests that the anti-diabetic mode of action of S. frutescens is through modulation of lipid metabolism. It was also established that S. frutescens has the potential to prevent IR in vivo.

- Description: In today’s dynamic technological landscape, wireless communication networks have become an important part of economic development. The emergence of wireless technologies raises hopes to extend communication to remote areas that have not seen any tangible deployment to date. As developing nations pin their hopes to wireless technologies, cost models for wireless communication networks are becoming vital to support the emerging technologies. However, varying cost changes raise critical challenges to the estimation of both capital expenditure and operational expenditure. The network deployment process has numerous events that may cause adjustments to initially estimated project costs. These adjustments are necessary for a cost management plan and this plan includes monitoring cost performance and ensuring that only appropriate changes are made to the network project. The incidents that may cause cost changes can not be entirely predicted as their distribution tend to change dynamically from time to time. Estimating network deployment costs in such a dynamic environment necessitates cost models that can adapt to random occurrence of cost changes. Widely used cost models are usually performed by experienced personnel whose engineering experience is derived from deploying similar networks. In this approach experienced personnel add a certain percentage to the cost estimate to cater for contingency costs. Certainly such an approach depends on individual opinion, making it subjective and void of mathematical estimating relationships which are of paramount importance in ensuring that estimated deployment costs are sufficient to deal with cost uncertainties. We observe that existing approaches can only explore a limited solution space and hence can lead to cost overruns if implemented in dynamically cost changing environments. This thesis presents a wireless communication network deployment cost model that incorporates uncertainties into the final cost estimate. The model is adaptive to unpredictable cost changes since it allows adjustments of confidence levels when calculating contingency costs. This allows dynamically updating the cost changes without the cost model being reconstructed from scratch. We make use of the Poisson process in modeling the occurrence of incidents that are responsible for causing cost changes during network deployment. We also show that the occurrence of the incidents causing cost change are random and tend to follow the Poisson distribution. Using different levels of confidence we model various cost contingencies and make sensitivity analyses to identify the probability of cost overrun when given different contingencies. The dynamic adaptive cost model can be used either at the strategic level to understand the cost of a particular technique or at the operational level, as a way to show how Poisson process in network deployment can compare with engineering experience and other estimating techniques. We believe that the model is useful for remote areas where deployment costs are volatile and the distribution of incidents causing cost change to original cost estimates are diverse and dynamically changing. Further we expect that our research improves the knowledge base of information about the costs for rural communities to connect to the Internet, consequently providing useful input to future policy debates. This work is further poised to be a utility function to help those planning internet infrastructure deployments in least developed regions.

- Description: Like orchids, the complexity of flowers found in asclepiads (Asclepiadoideae, Apocynaceae) and the fact that pollen is presented as pollinaria, offers excellent opportunities to study various aspects of plant-pollinator interactions. In this thesis I investigated two broad themes: ecological aspects of the pollination biology of hymenopteran and fly-pollinated asclepiads as well as the degree of specialization to certain pollinators in these species. Colonizing plants often reproduce through self-pollination, or have highly generalized pollination systems, or both. These characteristics facilitate establishment in small founding populations and generates the prediction that reproductive success should be independent of population size in these species. Chapter one examines the pollination biology of Gomphocarpus physocarpus, an indigenous, weedy species and investigates the relationship between reproductive success and population size. In this species, there is no evidence of an Allee effect and reproductive success is not correlated with population size. In addition G. physocarpus is not capable of self-pollination, suggesting it is completely reliant on pollinators for seed set. The lack of a relationship between pollination success and population size is therefore likely explained by the generalized wasp pollination system of this species. Several milkweeds are invasive outside of their native ranges. Invasive species either need to co-opt native pollinators in order to reproduce or reduce their reliance on pollinators through having the ability to self-pollinate. Co-opting native pollinators is expected to be easier in species that have generalized pollination systems, alternatively species with specialized flower morphologies need to rely on similar functional groups of pollinators to be present within the invaded range. Chapter two investigates the pollination biology and pollination success of the invasive milkweed, Araujia sericifera, and finds that in South Africa, this species is visited mainly by native honeybees and nocturnal moths. Moths however contribute little to pollen removal, and deposition. Based on the apparent morphological mismatch between the flower of A. sericifera and native honeybees, I propose that the native pollinators of this species are likely to be larger Hymenoptera (e.g. Bumblebees). Data from a breeding system study, indicated that this species is not capable of automatic self-pollination, but could set fruit from geitonogamous self-pollinations pointing to the importance of native pollinators for successful reproduction. The pollinaria of milkweeds can accumulate on pollinators to form pollen masses large enough to physically interfere with the foraging behaviour of pollinating insects. In chapter three I describe the pollination biology of Cynanchum ellipticum and find that this species is mainly pollinated by honeybees although this species is visited by several other members of Hymenoptera, Lepidoptera and Diptera. Due to the structure of the pollinaria, these chain together relatively efficiently and frequently form large pollinarium loads on the mouthparts of honeybees. However there is little evidence that these pollinarium loads influence the foraging times of pollinators and only a few individual honeybees exhibited longer foraging times and most honeybees were unaffected by the presence of large pollinarium loads. Within the genus Cynanchum there is large variation in the gynostegium structure that may influence the pattern of pollinarium loading on pollinators as well as pollen reception as shown in chapter three. In Chapter four, the pollination biology of Cynanchum obtusifolium is examined, and like that of C. ellipticum, this species is visited by a wide diversity of pollinators but honeybees appear to be the primary pollinators. More importantly this species is shown to be andromonoecious and produces two morphologically different flower types, that may be distinguished based on differences in the gynostegium structure. These two types of flower could mainly be distinguished by the length of the anther wings. I found that flowers with short anther wings function as male flowers by only exporting- and rarely receiving pollinia. Flowers with longer anther wings function as hermaphrodite flowers and can both export and receive pollinia. The ratio of male to hermaphrodite flowers varied at different times during the flowering season, but preliminary data suggested that this was not related to levels of pollination success. The genera Stapelia and Ceropegia are well known for their intricate floral adaptations that mimic the brood and feeding substrates of pollinating flies. Despite several studies that have documented the various adaptations to fly pollination in different species, there is a lack of natural history studies documenting different flower visitors, pollen loads and long term levels of pollination success in these species. In Chapter six I document the pollination biology of Ceropegia ampliata by documenting different pollinators and quantifying average levels of pollination success and the nectar reward. I also experimentally manipulated the trapping hairs of this species to determine whether trapping hairs influence average levels of pollen export and receipt. I show that Ceropegia ampliata is pollinated by a generalist guild of flies (mainly Tachinidae, Sarcophagidae, Muscidae and Lauxaniidae) and produces minute quantities of relatively dilute nectar as a reward. Pollination success was generally low in this species and increases periodically suggesting that the abundance of pollinators is patchy. I found that flowers with trapping hairs that had already wilted had higher levels of pollinarium removal than flowers with erect hairs, however experimentally removing the hairs had no significant effect on pollen export and receipt. In Chapter seven, I document the pollinators, pollen loads and long term levels of pollination success in Stapelia hirsuta var. bayllissi, a rare sapromyiophilous stapeliad. I find that, in contrast to C. ampliata, this species was specialized to pollination by small flies of the family Anthomyiidae. Similar to the results from Chapter seven, I find that long term levels of pollination success were typically low but could increase periodically, although such increases were generally unpredictable. There are currently very few records documenting pollinator interactions in the Periplocoideae. Many species within this subfamily exhibit open-access flowers suggestive of pollination by short-tongued insects. I investigated the pollination biology of Chlorocyathus lobulata, a rare species with a highly localized distribution. I aimed to determine the pollinators, average levels of pollination success and demography of this species in order to determine whether this rare species is suffering from the collapse of a highly specialized pollinator mutualism. I also quantified the high incidence of flower herbivory caused by larvae of the moth, Bocchoris onychinalis. I find that C. lobulata has a highly generalized fly pollination system and average levels of pollination success suggested that a large proportion of flowers had pollen removed and deposited suggesting that this species is not experiencing pollination failure. The large numbers of juveniles present also indicated that recruitment is taking place.

- Description: This thesis offers a critical reflection of a study I initiated to enhance self-esteem for self-actualization. The research process involved enhancing my own self-esteem before facilitating the enhancement of the self-esteem of my participants. This endeavour included the simultaneous exposure given to researcher and researched, with the aim of helping to attain the respective transformative potentials of the latter. An action research methodology was used, intending to improve my own practice and to involve and learn from the participants in order for all relevant stakeholders to capitalize from the learning experience. Whilst undertaking this study, I learned to reflect on my values, attitudes and relationships with my learners. I also became aware of how individuals‟ perceptions of themselves can influence other aspects of their development. This awareness enabled me to become more sensitive to the emotional needs of the learners in my care and also led to a greater understanding of their individuality. I came to comprehend the impact of a positive self-esteem on the learning process that aided me in developing an understanding of the positive impact an enhanced self-esteem has on how my learners view themselves. In the course of the research, I developed an awareness of the need to create a caring practice centred on values of gentleness, respect, kindness, awareness and recognition of individual strengths. These afore-mentioned values are premised on the recognition and acknowledgement of basic human rights, inclusivity, equality and social justice; principles reflecting my own living standards. This heightened vii awareness influenced the ways I organized the learning and teaching process in my classroom. Enhancing my own self-esteem and those of my learners basically involved interchangeable interventions such as encouraging involvement, allocating responsibilities, creating opportunities for assertiveness and being generous with affirmation, praise and motivation. These endeavours occurred within a social constructivist approach, enabling participants to construct their own epistemologies within their respective ontologies. My findings offer new conceptualizations about how an enhanced self-esteem can help individuals realize their fullest potential.

- Description: The restructuring of South African education, after the country’s first non-racial democratic elections in 1994, coincided, with the development of inclusive education in international education. The implementation of inclusive education, internationally, was guided by the Salamanca Statement of 1994 and the international “Education for All” movement. Education White Paper 6 (2001), serves as the blueprint for implementing inclusive education in South Africa. This investigation argues that the successful implementation of the inclusive education system is dependent on the development of an effective, quality education support structure. Such an educational support structure is one that should reflect a holistic, integrated and community based approach to support. Based on this presupposition, the primary research aim of this investigation was the establishment of a framework for a holistic, integrated, community based education support structure to do justice to learners with mainly external barriers to learning (LSEN) and educators finding themselves in an inclusive classroom in Nelson Mandela Metropole and surrounding areas. In order to understand how such a framework can be established, the following guiding secondary research questions were posed: • What was the international perspective on the implementation of inclusive education? • What was the nature of educational provisioning for learners with barriers to learning (LSEN) before 1994, and the implementation of inclusive education policies thereafter? • What were the practical implications of implementing Education White Paper 6 (Department of Education, 2001) for the support roles of education support providers in South African schools? • What is the support challenges facing learners and educators within inclusive classrooms in Nelson Mandela Metropole and surrounding areas? • What support structures are currently available at the different levels of the education system? This investigation was completed from a phenomenological perspective. A constructivist approach to data collection and data analysis was followed, as the investigator did not attempt to prove or disprove theory, but rather to understand the phenomena under investigation from the viewpoint of participants. iv The sample, selected by means of non-probability purposive and theoretical sampling techniques, included 120 educators from 85 different schools in Nelson Mandela Metropole and surrounding areas. In addition, 4 education officials, 4 members of community organisations and 2 teacher union representatives were selected to the sample. Through the process of data analysis, accomplished through the principles inherent to grounded theory, two themes and sub-themes were identified. The investigator presented a discussion on the two themes and sub-themes. This discussion was followed by a proposal for the establishment of a framework for a holistic, integrated, community based educational support structure. The investigation was completed by recommendations related to the primary and secondary research questions. The investigation concluded that the implementation of inclusive education is severely hampered by strong exclusionary factors that are principally related to socio-economic backlogs that have not been successfully addressed by the current government.

- Description: The current interest in conservation agriculture (CA) technologies is a result of the need to reduce excessive land degradation in most crop producing areas as well as to enhance sustainable food production. Cover crops that are usually grown under CA to provide soil cover, may offer secondary benefits, depending on the farming system. The concept of growing cover crops is a relatively new phenomenon to smallholder farmers. Production of large biomass yields and weed suppression from cover crops were major challenges affecting success and uptake of CA technologies by smallholder irrigation farmers. Coupled with this, low soil fertility limit maize productivity and reduce water use efficiency on smallholder irrigation schemes in what is largely a water strained agro-ecology in South Africa. While cover cropping can increase maize productivity, benefits of different types of mulch are not well understood, leading to challenges in selecting the most appropriate cover crop species to grow in the Eastern Cape Province (EC) of South Africa (SA) which has a warm temperate climate. With respect to any new technology, smallholder farmers are more interested in the economic benefits. Cover crops have been defined as leguminous or non-leguminous plants used for ground cover in various temporal and special configurations used in crop or animal production systems. The purpose of these cover crops is to improve on or more of the following: soil erosion, availability and cycling of N, P, K, Ca and other nutrients, soil moisture and water infiltration, and weed or pest control (Eilitta et al., 2004).. Improvement of animal or human diet may be additional goals. This definition accommodates diverse systems which may include intercrop and sole-cropping systems. In the Eastern Cape Province of South Africa, a government initiative has promoted the growing of winter cover crops in smallholder irrigation schemes (Allwood, 2006). In other parts of Africa, legume food crops have been simultaneously grown with cereal staples to improve both soil cover and human diet (Eilitta et al., 2004). Winter experiments were undertaken in 2007 and 2008 to evaluate biomass accumulation, C and N uptake, weed suppression and response to fertilization. Winter cover crops planted included; oats (Avena sativa), grazing vetch (Vicia dasycarpa), faba bean (Vicia faba), forage peas (Pisum sativum) and lupin (Lupinus angustifolius). After cover crops were terminated, the effects of residues on weeds, fertility, moisture conservation and maize productivity were undertaken in the 2007/08 and 2008/09 summer seasons. Field studies were also done in the 2007/08 and 2008/09 summer seasons to investigate effects of strip intercropping patterns (3:2; 4:2; and 6:2 patterns) of maize (cv. PAN 6479) with mucuna (Mucuna pruriens) or sunnhemp (Crotalaria juncea) on maize productivity and summer cover crop biomass production. In a separate experiment effects of relay intercropping sunnhemp, mucuna and sorghum (Sorghum bicolor) on biomass accumulation and maize productivity were investigated. Decomposition, N and P release from both winter cover crops and summer cover crops were also assessed in laboratory incubation experiments. Oats, grazing vetch and forage peas cover crops produced mean dry mass of 13873 kg/ha, 8945.5 kg/ha and 11073 kg/ha, respectively, while lupin had the lowest dry mass of 1226 kg/ha over the two seasons. Oats responded to fertilization while, there was little or no response from the other winter cover crops. Oats and grazing vetch also reduced weed density by 90 % and 80 % respectively while lupin only reduced weed density by 23 % in relation to the control plots. Nitrogen uptake was 254 kg N/ha for oats while it was 346 kg N /ha for grazing vetch. In the subsequent summer season, grazing vetch and forage pea residues significantly (P < 0.01) improved soil inorganic N. Oat and grazing vetch residues significantly (P < 0.05) reduced weed dry masss and weed species diversity compared to plots with lupin residues and the control. Lack of maize fertilization tended to reduce maize yields but not for maize grown on grazing vetch residues. From an economic perspective, grazing vetch resulted in the highest returns. Decomposition of winter cover crops was much faster for grazing vetch followed by forage peas and lastly oats. Oats had 40 % ash free dry mass remaining after 124 days while grazing vetch and forage peas had 7 % and 16 % respectively. Maximum net mineralized N and P were greater for grazing vetch (84.8 mg N/kg; 3.6 mg P/kg) compared to forage peas (66.3 mg N/kg; 2.7 mg P/ha) and oats (13.7 mg N/kg; 2.8 mg P/kg). In the strip intercropping trials, sunnhemp achieved the highest biomass yield of 4576 kg/ha in the 3:2 pattern while mucuna achieved 1897 kg/ha for the same strip pattern. The 3:2 strip intercropping pattern slightly depressed yields, however, yield reduction was more pronounced in the first season where water stress was experienced. Growing maize on previous cover crop strips failed to increase maize productivity probably due to weed growth during the fallow reducing mineral N in these strips. Decomposition was faster in sunnhemp leaves and mucuna compared to sunnhemp stems. Sunnhemp stems had about 65 % of ash free dry mass remaining after the end of the experiment at 132 days while just over 10 % of mucuna and sunnhemp leaves still remained. Mucuna mineralized 60 mg N/kg and 3.2 mg P/kg and sunnhemp mineralized 45 mg N/kg and 3.5 mg P/kg. Relay intercropping did not significantly (P > 0.05) affect maize biomass and grain yield. Sorghum experienced the largest drop in biomass when relay-intercropped with maize. Mucuna resulted in the highest N uptake (271 kg N/ha) in sole cropping while sorghum had the lowest (88 kg N/ha). Grazing vetch results in high biomass yields with minimal fertilizer application in a warm-temperate climate. Grazing vetch mulch is also the most cost effective mulch for better early weed control, improving soil mineral N status, water conservation and ultimately enhanced maize productivity in smallholder irrigation maize-based systems. The 3:2 pattern maximizes summer cover crop biomass yields compared to the 6:2 and 4:2 patterns. However, the 3:2 pattern may slightly depress yields in a water stressed environment. Relay intercropping mucuna, sunnhemp and sorghum into a maize crop at 42 days after maize sowing has no effect on maize productivity while cover crop biomass yields are low. Having a long winter fallow period after maize harvesting, a common practice in the study area, reduces the positive impact of legume cover crops on soil mineral N. Results suggest that winter cover crops may result in weed control, soil fertility and maize yield improvement benefits while a long fallow period may cancel-out these benefits for summer cover crops. Grazing vetch is a cost effective cover crop that produces high maize yields with minimal fertilizer input. Maize growing on oat mulch requires more fertilizer application than crops growing on grazing vetch mulch. Conservation agriculture systems in which summer cover crops are grown alongside the maize crop with a long winter fallow period do not produce the intended CA benefits.

- Description: Hermannia incana Cav. (Sterculiaceae), known as sweet yellow bells, is a medicinal plant used by the people of the Eastern Cape for the treatment of stomach-ache and diarrhoea. It has purgative and diaphoretic effects. It is a prostrate herb with yellow flowers and sparsely hairy and slightly glandular leaves, occurring in grassland and marshes in the Eastern Cape Province of South Africa. Based on the ethnomedical uses of this plant, the research project was designed to evaluate its antidiarrhoeal and toxicological properties. An ethnobotanical study of plants used for the treatment of diarrhoea in the Eastern Cape Province was carried out, using a questionnaire which was administered to herbalists, traditional healers and rural dwellers. This survey indicated a total of 17 plant species from 14 families. Elephantorrhiza elephantine (Burch.) Skeels, Hermannia incana Cav., Pelargonium reniforme Curt., Alepidea amatymbica Eckl. & Zeyh. and Bulbine latifolia (L.f.) Roem. et Schult. were the most frequently mentioned and highly recommended plants for the treatment of diarrhoea by both the traditional healers and rural dwellers. The root, bark and leaves are the common parts of plants used, while decoctions and infusions are the main methods of preparation. The agar dilution method was used to study the antimicrobial activity. The methanol extracts of the plant showed appreciable activity against Gram-positive and Gram-negative bacteria at concentrations ranging from 0.5 to 7.0 mg/ml. The acetone and water extracts of both the leaves and the roots showed moderate activity against Gram positive bacteria and less activity against Gram negative bacteria. All the extracts inhibited the growth of the fungi Aspergillus flavus, Aspergillus niger, and Mucor hiemalis with growth inhibition ranging from 54.31 percent to 96.67 percent at 0.1-10 mg/ml. None of the extracts suppressed the growth of Candida albicans at the maximum concentration (10 mg/ml) tested. iii In the in vivo antidiarrhoeal evaluation using Wistar rats, the aqueous extract at all the doses tested, significantly prolonged the time of induction of diarrhoea and also reduced the frequency of diarrhoeal episodes and fecal parameters (total number, number of wet, fresh and dry weight and water content of the faeces). The percentage inhibition of defecation and intestinal content (enteropooling) were increased in dose dependent manner. The doses also reduced the intestinal transit time of charcoal, masses and volumes of intestinal fluid (gastrointestinal motility). These results are indications of antidiarrhoeal property of H. incana leaf extract with the 600 mg/kg body weight of the extract being the most effective. In the toxicological evaluation using Wistar rats, the oral administration of the extract did not produce any significant effect on the liver and kidney body weight ratios, RBC, HB, PCV, MCV MCH, MCHC, RCDW, WBC, neutrophils, monocytes and basophils cholesterol, triacylglycerol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol and atherogenic index. The extract also did not affect the levels of sodium, potassium, chloride, inorganic phosphorus, urea, creatinine, total protein, globulin, albumin, total and conjugated bilirubin. The activities of alkaline phosphatase, gamma glutamyl transferase and alanine aminotransaminase in the serum were increased by the extract whereas aspartate aminotransaminase was decreased. The levels of LUC, platelets, lymphocytes and eosinophils were significantly affected at 600 mg/kg body weight. The available evidence in this study suggests that the extract of H. incana leaf is mild, parameter and dose specific. The structure and distribution of foliar appendages on the leaves of this plant were investigated with the JEOL (JSM-6390LV) scanning electron microscope (SEM). Both glandular and non-glandular trichomes were observed. Long stalked glandular trichomes were present on both the abaxial and adaxial surfaces while short stalked glandular trichomes were present only on the adaxial surface. Glandular trichomes were capitate while nonglandular trichomes were stellate with many arms. Energy dispersive X-ray spectroscopyiv SEM showed that Al, Ca, K, Na, Ti and Si were the major constituents of the crystals analyzed from the leaf surfaces. The phytochemical screening of H. incana revealed the presence of bioactive antidiarrhoeal agents such as alkaloids, tannins, saponins, phenolics, triterpenes, cardiac glycosides, flavonoids, cardenolides and dienolides. Two flavonoids, epicatechin and 3, 5, 7, 2’ tetra-hydroxy flavone-3- O--D-glucopyranoside were isolated from the leaves of the plant through bio-active guided fractionation. Both these compounds were screened against diarrhoea causative organisms (Echerichia coli, Shigella flexneri, Bacillus cereus and Staphylococcus aureus) and exhibiting minimum inhibitory concentrations ranging from 12.5 to 100 μg/ml. The findings from this research have generally justified the traditional use of this plant for the treatment of diarrhoea in this province.

- Description: The focus of this study is to explore and expand farmer learning processes in sustainable agriculture workplace contexts. It examines change oriented learning processes in the context of three sustainable agriculture practices. The study begins by discussing the history and emergence of environmental discourses and approaches; sustainable agriculture; and the histories of three kinds of sustainable agriculture practices: Permaculture, Organic Farming and Machobane Farming System. It also traces the evolution of agricultural extension approaches within the wider context of education for sustainable development. The main focus of the study is an exploration of how farmer learning can be mediated through an expansive learning process. The study methodology surfaces some of the contradictions in sustainable agriculture and learning activity systems that farmers encounter in learning and practising sustainable agriculture. It uses these contradictions as sources of expansive learning in and between the respective activity systems of farmers, sustainable agriculture facilitators, agricultural extension workers (conventional) and organic entrepreneurs. As shown in the study, the expansive learning processes result in the modelling, implementation and reviewing of solutions to contradictions being faced in the learning and practice of sustainable agriculture. The study also proposes a number of tools that can be adapted and used by development farmers and agricultural trainers to examine and expand learning as well as build farmer agency. The study was conducted in three case study sites in Lesotho, South Africa and Zimbabwe. In Zimbabwe the study is located in Hwedza district in the St Margaret Primary School and community that learn, practise and facilitate the learning of Permaculture within the Schools and Colleges Permaculture Programme (SCOPE). The second study site is in South Africa: Durban urban and peri-urban areas where a community of organic farmers, facilitators and entrepreneurs coordinate the marketing of their produce through Isidore Farm and Earth Mother Organic and support each other to learn and practise organic farming. The third study site is based in the Mafeteng and Mohale‟s Hoek districts of Lesotho where the focus was on farmers who learn and practise the Machobane Farming System (MFS) and are supported in this by the Rural Self Development Association (RSDA) and the Machobane Agricultural Development Foundation (MADF). Drawing on three sensitising concepts of dialectics, reflexivity and agency, the study worked with Cultural Historical Activity Theory (CHAT) underpinned by critical realism to reveal how farmer learning is mediated and expanded. The theory of practice/habitus also provided a useful theoretical lens with which to examine data generated. Using a two-phased, multiple embedded case study approach, the study worked within the broad framework of social learning. It used semi-structured individual and group interviews, observations and document analysis to explore learning processes and generate „mirror‟ data. This data was then used in Change Laboratory Workshops, within the Developmental Work Research methodology, where double stimulation and focus group discussions contributed to expanding learning processes. Drawing on critical realism the study used inductive, abductive and retroductive modes of inference to analyse data in each case study as well as across case studies. The findings of the study reveal that farmer learning is influenced by both intrinsic motives, such as identity, and extrinsic motives which are primarily associated with economic, ecological and health benefits. Farmers learn through scaffolding and mediating tools that link everyday and scientific knowledge. They also learn from fellow farmers through observation, practising and experimentation. Some of the issues that were raised in connection with farmer learning processes are: language; time to learn, practice and appropriate concepts; time to improve the natural resource base while at the same time improving income generation; and responses to climate change. The study also found that farmer learning and practice of sustainable agriculture in the case studies investigated, is influenced by past and current agricultural and educational policies; societal values and attitudes; social and cultural backgrounds; work affordances and gender relations; quality of training offered; poverty; and, HIV and AIDS. In the second phase of the study, which built on the problematic situations being encountered by research participants (sustainable agriculture farmers, sustainable agriculture facilitators, extension workers, and organic marketers) to surface contradictions, the main finding was that the expansive learning process has potential to enhance farmer learning and practice of sustainable agriculture. It does this by mobilising distributed cognition among participants as well as their preparedness to act. Through the expansive learning processes in each case study, research participants were able to question their practices, surface contradictions, model solutions and implement them, and thus build individual, collective and relational agency reflexively. Observation of this required micro-analysis of agentive talk and reflective talk. The study contributes in-depth insight into participatory research and learning processes, especially within the context of people-centred learning and innovation in the agricultural development arena. It provides empirical and explanatory insight into how change oriented social learning can emerge and be expanded in Education for Sustainable Development, explaining learning and change relationships in three sustainable agricultural practices. It also provides learning and extension tools to work with contradictions that arise from intentionality, experience, context and history in farming and training activity systems. Its key contribution lies in providing in-depth insight into mobilisation of human agency and reflexivity in change oriented sustainable agriculture learning and development, processes that are critical for responding to contemporary socio-ecological issues and risks.

- Description: Angiotensin-converting enzyme (ACE) inhibitors are some of the most commonly prescribed medications for hypertension. They are cited in many papers as the treatment most often recommended by guidelines and favoured over other antihypertensive drugs as first-line agents especially when other high-risk conditions are present, such as diabetic nephropathy. The development of captopril (CPT) was amongst the earliest successes of the revolutionary concept of structure-based drug design. Due to its relatively poor pharmacokinetic profile or short half-life of about 1 hour, the formulation of sustained-release microcapsule dosage form is useful to improve patient compliance and to achieve predictable and optimized therapeutic plasma concentrations. Currently, CPT is mainly administered in tablet form. One of the difficulties of CPT formulation has been reported to be its instability in aqueous solutions. CPT is characterized by a lack of a strong chromophore and, therefore, not able to absorb at the more useful UV–Vis region of the spectrum. For this reason, an accurate, simple, reproducible, and sensitive HPLC-ECD method was developed and validated for the determination of CPT in dosage forms. The method was successfully applied for the determination of CPT in commercial and developed formulations. Possible drug-excipient and excipient-excipient interactions were investigated prior to formulating CPT microcapsules because successful formulation of a stable and effective solid dosage form depends on careful selection of excipients. Nuclear magnetic resonance spectroscopy, Fourier transform infra-red spectroscopy (FT-IR), differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) were used for the identification and purity testing of CPT and excipients. The studies revealed no thermal changes during stress testing of binary and whole mixtures which indicate absence of solid state interactions. There were no shifts, appearance and disappearance in the endothermic or exothermic peaks and on the change of other associated enthalpy values on thermal curves obtained with DSC method. Characteristic peaks for common functional groups in the FT-IR were present in all the mixtures indicating the absence of incompatibility. The techniques used in this study can be said to have been efficient in the characterization and evaluation of the drug and excipients. The technique of microencapsulation by oil-in-oil was used to prepare CPT microcapsules. The effects of polymer molecular weight, homogenizing speed on the particle size, flow properties, morphology, surface properties and release characteristics of the prepared CPT microcapsules were examined. In order to decrease the complexity of the analysis and reduce cost response surface methodology using best polynomial equations was successfully used to quantify the effect of the formulation variables and develop an optimized formulation thereby minimizing the number of experimental trials. There was a burst effect during the first stage of dissolution. Scanning electron microscopy (SEM) results indicated that the initial burst effect observed in drug release could be attributed to dissolution of CPT crystals present at the surface or embedded in the superficial layer of the matrix. During the preparation of microcapsules, the drug might have been trapped near the surface of the microcapsules and or might have diffused quickly through the porous surface. The release kinetics of CPT from most formulations followed Fickian diffusion mechanism. SEM photographs showed that diffusion took place through pores at the surface of the microcapsules. The Kopcha model diffusion and erosion terms showed predominance of diffusion relative to swelling or erosion throughout the entire test period. Drug release mechanism was also confirmed by Makoid-Banakar and Korsmeyer-Peppas models exponents which further support diffusion release mechanism in most formulations. The models postulate that the total of drug release is a summation of a couple of mechanisms; burst release, relaxation induced controlled-release and diffusional release. Inspection of the 2D contour and 3D response surfaces allowed the determination of the geometrical nature of the surfaces and further providing results about the interaction of the different variables used in central composite design (CCD). The wide variation indicated that the factor combinations resulted in different drug release rates. Lagrange, canonical and mathematical modelling were used to determine the nature of the stationery point of the models. This represented the optimal variables or stationery points where there is interaction in the experimental space. It is difficult to understand the shape of a fitted response by mere inspection of the algebraic polynomial when there are many independent variables in the model. Canonical and Lagrange analyses facilitated the interpretation of the surface plots after a mathematical transformation of the original variables into new variables. In conclusion, these results suggest the potential application of Eudragit® / Methocel® microcapsules as suitable sustained-release drug delivery system for CPT.

- Description: Today information can be seen as a basic commodity that is crucial to the continuous well-being of modern organizations. Many modern organizations will be unable to do business without access to their information resources. It is therefor of vital importance for organizations to ensure that their infor- mation resources are adequately protected against both internal and external threats. This protection of information resources is known as information security and is, to a large extent, dependent on the behavior of humans in the organization. Humans, at various levels in the organization, play vital roles in the pro- cesses that secure organizational information resources. Many of the prob- lems experienced in information security can be directly contributed to the humans involved in the process. Employees, either intentionally or through negligence, often due to a lack of knowledge, can be seen as the greatest threat to information security. Addressing this human factor in information security is the primary focus of this thesis. The majority of current approaches to dealing with the human factors in information security acknowledge the need to foster an information security culture in the organization. However, very few current approaches attempt to adjust the "generic" model(s) used to define organizational culture to be specific to the needs of information security. This thesis firstly proposes, and argues, such an adapted conceptual model which aims to improve the understanding of what an information security culture is. The thesis secondly focuses on the underlying role that information security educational programs play in the fostering of an organizational information security culture. It is argued that many current information security edu- cational programs are not based on sound pedagogical theory. The use of learning taxonomies during the design of information security educational programs is proposed as a possible way to improve the pedagogical rigor of such programs. The thesis also argues in favor of the use of blended and/or e-learning approaches for the delivery of information security educational content. Finally, this thesis provides a detailed overview demonstrating how the various elements contributed by the thesis integrates into existing trans- formative change management processes for the fostering of an organizational information security culture.
