Development of a translation device for axiological-semantic density in political news articles: Wording and charging

- Description: The concept of axiological-semantic density from Legitimation Code Theory (LCT) is extremely helpful in analysing political knowledge-building, as it describes the strength of relations between various people, political stances and moral judgements, enabling these to be positioned in relation to each other. We present a multi-level translation device designed to identify strengths of axiological-semantic density in political news articles from the Daily Sun, South Africa’s most popular tabloid newspaper. This translation device was devised through analysis of selected texts from a corpus of 516 articles published between January and June 2015. It was developed through a collaborative process involving the first author and a team of student research assistants. The final translation device has five tools, of which two, the wording and charging tools, are described in this article, and then illustrated using an example analysis of a Daily Sun political news article. Both tools reveal insights into South African political discourses and ways in which axiological-semantic density can be enacted in future research. Making axiological-semantic density visible using such a translation device also has practical applications in assisting readers to understand the ways in which publications such as the Daily Sun position political parties, enabling them to engage more constructively in discussions on the country’s future.

Ethnobotanical survey, phytoconstituents and antibacterial investigation of Rapanea melanophloeos (L.) Mez. bark, fruit and leaf extracts

- Description: Rapanea melanophloeos is traditionally used in South Africa in the treatment of ailments of the skin, pulmonary and gastro intestinal tract. This study was aimed at giving an overview of these traditional uses and comparing the phytochemicals and antibacterial activities of various crude extracts of the leaves, fruits and bark in order to validate these uses. The three plant parts were extracted using petroleum ether (PE), ethyl acetate (EtOAc), methanol (MeOH) and water. Various phytochemicals were compared using TLC, while alcohol precipitable solids (APS), non-polar terpenes and amino acids were analysed by GC-MS. Antibacterial activity was determined against three Gram-positive and three Gram-negative strains by microdilution assays. Caryophyllene oxides, α-cadinol and (−)-spathulenol were identified in the PE extracts. All nine essential amino acids were present in fruit extracts in significantly higher levels than in the leaves and bark; 255.1, 23.4 and 21.3 mg/g respectively. Most of the extracts showed good antibacterial activity, especially against the Gram-positive pathogens (MIC of ≤1 mg/mL), the EtOAc extracts exhibited the best activity with the fruit having an MIC values of 0.1 ± 0.2 mg/mL against Staphylococcus epidermidis and Enterococcus faecalis, 0.05 mg/mL against Bacillus cereus. Results from this study validate the ethnomedicinal uses of R. melanophloeos extracts for ailments of bacterial etiology. The plant had a rich supply of secondary metabolites, APS and amino acids and TLC and antibacterial activities of the extracts showed slight variations in chemical composition due to geographic distribution.

“Ha! Relationships? I only shout at them!”: Strategic management of discordant rapport in an African small business context

- Description: This study demonstrates how and why interactants at a tyre fitment centre in Grahamstown, South Africa, manage discordant interpersonal relationships in strategic ways. Individuals in a post-apartheid small business respond to their social and economic context and exercise agency to their advantage in doing so. This study draws on linguistic ethnography (Rampton 2007) and the Rapport Management Framework (RMF, Spencer-Oatey 2000b, 2011), itself a development of politeness theory (Brown and Levinson 1987). An initial RMF analysis ran into difficulties around interactions that at first glance appeared to be oriented toward Rapport Challenge and Neglect. Upon closer examination, it appeared that discordant rapport was being actively maintained in this business. This led us to address underdeveloped areas of RMF that were not responsive enough to describe naturally occurring small business interactions, and propose an Enhanced Rapport Management Framework to overcome its inadequacies. We conclude that people may deliberately maintain discordant relationships when it is in their best interests to do so. Thus, contrary to a common-sense belief that harmonious social relations are an intrinsic good, we found that promoting discordant social relations can be understood as a rational response to individuals’ social and economic contexts, particularly in conditions such as those in many postcolonial African societies.

Selective modulation of plasmodial Hsp70s by small molecules with antimalarial activity

- Description: Plasmodial heat shock protein 70 (Hsp70) chaperones represent a promising new class of antimalarial drug targets because of the important roles they play in the survival and pathogenesis of the malaria parasite Plasmodium falciparum. This study assessed a set of small molecules (lapachol, bromo-β-lapachona and malonganenones A, B and C) as potential modulators of two biologically important plasmodial Hsp70s, the parasite-resident PfHsp70-1 and the exported PfHsp70-x. Compounds of interest were assessed for modulatory effects on the steady-state basal and heat shock protein 40 (Hsp40)-stimulated ATPase activities of PfHsp70-1, PfHsp70-x and human Hsp70, as well as on the protein aggregation suppression activity of PfHsp70-x. The antimalarial marine alkaloid malonganenone A was of particular interest, as it was found to have limited cytotoxicity to mammalian cell lines and exhibited the desired properties of an effective plasmodial Hsp70 modulator. This compound was found to inhibit plasmodial and not human Hsp70 ATPase activity (Hsp40-stimulated), and hindered the aggregation suppression activity of PfHsp70-x. Furthermore, malonganenone A was shown to disrupt the interaction between PfHsp70-x and Hsp40. This is the first report to show that PfHsp70-x has chaperone activity, is stimulated by Hsp40 and can be specifically modulated by small molecule compounds.
