- Description: South Africa is not well-endowed with indigenous forests and those that remain are known to be degraded and declining at an alarming rate. This constitutes a direct threat to the quality of life of the resource-poor rural households which are directly dependent on the resources indigenous forests offer, as well as to ecological integrity. It is also recognised that the declining tree resources, particularly the indigenous tree species of high value, are increasingly threatened by a number of growing subsistence demands. This emphasises the need to establish, cultivate and conserve high-value tree species. Community forestry is recognised as a viable option for optimising land productivity; reducing pressure on indigenous forests and woodlands; ensuring a sustainable supply of desired tree products and services; and improving the quality of life of the resource-poor rural households. The primary purpose of Community Forestry Development (CFD) in the Hlabisa District is the provision of an information service and of technical support to enhance the livelihoods of rural communities by means of tree-related resources. Community forestry mainly focuses on tree resources that form an integral part of or contribute to rural people’s livelihoods. This includes small-scale growers (woodlots), woodlands and indigenous forests. Community forestry can be defined as tree growing and managed by the people for the people. Community forestry encompasses a wide range of activities which include farm forestry, agroforestry, village planting, woodlots, woodland management and indigenous-forest management by rural people, as well as tree planting in urban and peri-urban areas. Natural forests in South Africa account for less than 1 percent of the total land area. It has been suggested that these natural forests will no longer be able to meet the demand for forest products by 2020. The KwaZulu Department of Agriculture and Forestry embarked on community forestry in 1976 (but only on an ad hoc basis) as a possible solution to this dilemma. Later, the two South African pulp and paper giants, SAPPI and Mondi, started the Project Grow and Khulanathi Schemes respectively. One of the objectives of out-grower schemes was to encourage rural communities to plant trees on their own lands for the economic, social and environmental benefits trees offer. For example, timber could be sold to forestry companies. Financial assistance was offered to these communities to encourage implementation of such schemes. This exercise was aimed at bringing economic activity, capacity- building and community empowerment to the rural areas. The growers would benefit from the financial assistance and the readily available market provided by the company, while the company would be able to satisfy its demand for timber.

- Description: Aspen Pharmacare is listed on the Johannesburg Securities Exchange South Africa (JSE) and is Africa’s largest pharmaceutical manufacturer. The company is a major supplier of branded pharmaceutical and healthcare products to the local and selected international markets. For decades, Aspen has manufactured a basket of affordable, quality, and effective products for the ethical, generic over-the-counter (OTC) and personal care markets. Aspen is also the leading supplier of generic medicines to the public sector, providing comprehensive coverage of the products on the Essential Drug List. Aspen continues to deliver on its commitment toward playing a role in social responsibility diseases such as HIV/AIDS, tuberculosis and malaria. In August 2003 Aspen developed Africa’s first generic anti-retroviral drug, namely Aspen-Stavudine. Aspen’s manufacturing facilities are based in Port Elizabeth (PE) and East London. Aspen has recently completed an Oral Solid Dosage (OSD) manufacturing facility worth approximately R150 million in PE. The Group manufactures approximately 20 tons of product daily and in excess of 400 tons of solid dosage pharmaceuticals, which equates to more than 2 billion tablets. In addition, more than 3 million litres of liquid pharmaceuticals and over 200 tons of pharmaceutical creams and ointments are produced per year [1]. Aspen excels at delivering quality products and services, exceeding customer expectations, complying with international standards in an environment that cultivates technical expertise and innovation. Following this philosophy through to the shop floor areas mean that there are always initiatives in continuous production improvement. One of these improvement projects introduced is called Six Sigma. 8 Ten members of the staff, selected from different expertise fields in the company were trained in Six Sigma. Knowledge gained from the two week training course were applied to different areas in the factory using Six Sigma principles. This dissertation focuses on the study undertaken in one of production areas, namely the filling process of the ointments and creams at the Aspen Port Elizabeth facility.

The development of an electrochemical process for the production of para-substituted di-hydroxy benzenes

- Description: The project was concerned with the investigation of the electrochemical oxidation of various phenols, and to develop a viable reaction system for the production of the respective hydroquinones. Current production routes utilizing phenol as starting material have the limitations of using large amounts of acids, having to be stopped at low conversions and producing a mixture of the hydroquinones and catechols. Of the possible routes to the respective hydroquinones from the various phenols, the electrochemical oxidation of these phenols offers commercial and environmental advantages and hence formed the theme of the investigation. The synthetic possibilities proved to be more prevalent in a system when the electrochemical oxidation of these phenols was performed in an aqueous medium utilizing an organic co-solvent. Results obtained during this investigation made it possible to make certain predications about the mechanism taking place. This was found to depend on the anode material used for the oxidation. The results showed that the process developed for the electrochemical oxidation of these phenols, yields mainly the para-isomers of the respective hydroquinones and benzoquinones in good yields and selectivities, with fair current efficiencies and good mass balances at high conversions. For example: * Phenol (batch) 8 F: 345% current efficiency, 70% hydroquinone, 6% catechol, 9% benzoquinone, 9% phenol and 94 mass balance. * 2-Tert-butylphenol (flow) 10 F: 37% current efficiency, 65% hydroquinone, 33% benzoquinone, 2% phenol and 100% mass balance. * 2,6-Di-tert-butylphenol (flow) 11 F: 23% current efficiency, 92 % hydroquinone, 6% benzoquinone, 1% phenol and 99% mass balance. The developed electrochemical oxidation system offers the following advantages over previous and current methods: simplified isolation and extraction procedures, smaller amounts of acid usage, reasonably selective synthesis of the para-isomer and a less corrosive system all at high conversions.

- Description: Many network monitoring tools do not provide sufficiently in-depth and useful reports on network usage, particularly in the domain of application services data. The optimisation of network performance is only possible if the networks are monitored effectively. Techniques that identify patterns of network usage can assist in the successful monitoring of network performance. The main goal of this research was to propose a model to mine and visualise application services data in order to support effective network management. To demonstrate the effectiveness of the model, a prototype, called NetPatterns, was developed using data for the Integrated Tertiary Software (ITS) application service collected by a network monitoring tool on the NMMU South Campus network. Three data mining algorithms for application services data were identified for the proposed model. The data mining algorithms used are classification (decision tree), clustering (K-Means) and association (correlation). Classifying application services data serves to categorise combinations of network attributes to highlight areas of poor network performance. The clustering of network attributes serves to indicate sparse and dense regions within the application services data. Association indicates the existence of any interesting relationships between different network attributes. Three visualisation techniques were selected to visualise the results of the data mining algorithms. The visualisation techniques selected were the organisation chart, bubble chart and scatterplots. Colour and a variety of other visual cues are used to complement the selected visualisation techniques. The effectiveness and usefulness of NetPatterns was determined by means of user testing. The results of the evaluation clearly show that the participants were highly satisfied with the visualisation of network usage presented by NetPatterns. All participants successfully completed the prescribed tasks and indicated that NetPatterns is a useful tool for the analysis of network usage patterns.

- Description: Web Usage Mining (WUM) can be used to determine whether the information architecture of a web site is structured correctly. Existing WUM tools however, do not indicate which web usage mining algorithms are used or provide effective graphical visualisations of the results obtained. WUM techniques can be used to determine typical navigation patterns of the users of organisational web sites. An organisational web site can be described as a site which has a high level of content. The Computer Science & Information Systems (CS&IS) web site at the Nelson Mandela Metropolitan University (NMMU) is an example of such a web site. The process of combining WUM and information visualisation techniques in order to discover useful information about web usage patterns is called visual web mining. The goal of this research is to discuss the development of a WUM model and a prototype, called WebPatterns, which allows the user to effectively visualise web usage patterns of an organisational web site. This will facilitate determining whether the information architecture of the CS&IS web site is structured correctly. The WUM algorithms used in WebPatterns are association rule mining and sequence analysis. The purpose of association rule mining is to discover relationships between different web pages within a web site. Sequence analysis is used to determine the longest time ordered paths that satisfy a user specified minimum frequency. A radial tree layout is used in WebPatterns to visualise the static structure of the organisational web site. The structure of the web site is laid out radially, with the home page in the middle and other pages positioned in circles at various levels around it. Colour and other visual cues are used to show the results of the WUM algorithms. User testing was used to determine the effectiveness and usefulness of WebPatterns for visualising web usage patterns. The results of the user testing clearly show that the participants were highly satisfied with the visual design and information provided by WebPatterns. All the participants also indicated that they would like to use WebPatterns in the future. Analysis of the web usage patterns presented by WebPatterns was used to determine that the information architecture of the CS&IS web site can be restructured to better facilitate information retrieval. Changes to the CS&IS web site web were suggested, included placing embedded hyperlinks on the home page to the frequently accessed sections of the web site.

A comparison of programming notations for a tertiary level introductory programming course

- Description: Increasing pressure from national government to improve throughput at South African tertiary education institutions presents challenges to educators of introductory programming courses. In response, educators must adopt effective methods and strategies that encourage novice programmers to be successful in such courses. An approach that seeks to increase and maintain satisfactory throughput is the modification of the teaching model in these courses by adjusting presentation techniques. This thesis investigates the effect of integrating an experimental iconic programming notation and associated development environment with existing conventional textual technological support in the teaching model of a tertiary level introductory programming course. The investigation compares the performance achievement of novice programmers using only conventional textual technological support with that of novice programmers using the integrated iconic and conventional textual technological support. In preparation for the investigation, interpretation of existing knowledge on the behaviour of novice programmers while learning to program results in a novel framework of eight novice programmer requirements for technological support in an introductory programming course. This framework is applied in the examination of existing categories of technological support as well as in the design of new technological support for novice programmers learning to program. It thus provides information for the selection of existing and the design of new introductory programming technological support. The findings of the investigation suggest strong evidence that performance achievement of novice programmers in a tertiary level introductory programming course improves significantly with the inclusion of iconic technological support in the teaching model. The benefits are particularly evident in the portion of the novice programmer population who have been identified as being at risk of being successful in the course. Novice programmers identified as being at risk perform substantially better when using iconic technological support concurrently with conventional textual technological support than their equals who use only the latter form. Considerably more at risk novice programmers using the integrated form of technological support are in fact successful in the introductory programming course when compared with their counterparts who use conventional textual technological support only. The contributions of this thesis address deficiencies existing in current documented research. These contributions are primarily apparent in a number of distinct areas, namely: • formalisation of a novel framework of novice programmer requirements for technological support in an introductory programming course; • application of the framework as a formal evaluation technique; • application of the framework in the design of a visual iconic programming notation and development environment; • enhancement of existing empirical evidence and experimental research methodology typically applied to studies in programming; as well as • a proposal for a modified introductory programming course teaching model. The thesis has effectively applied substantial existing research on the cognitive model of the novice programmer as well as that on experimental technological support. The increase of throughput to a recommended rate of 75 percent in the tertiary level introductory programming course at the University of Port Elizabeth is attributed solely to the incorporation of iconic technological support in the teaching model of the course.

A statistical evaluation and analysis of mosquito repellent combination

- Description: The present product development project was aimed at studying the synergism and/or antagonism amongst various known insect repellent actives with the view to formulating a multi-active repellent product with improved properties when compared to current single-active commercial products. Advanced statistical methods were used to identify synergism between individual active substances and to define a formulation as close as possible to the “ideal” formulation. Several mosquito repellent samples were prepared and sent to the South African Bureau of Standards (SABS) in Pretoria to test for their efficiency in repelling mosquitoes. From the results of the repellency tests of the various active combinations, three actives were identified that showed promising signs of synergism. These actives were then studied in further detail to determine their optimum combination. In addition, it was shown that when using a natural flavourant as promoter and incorporating a slow-release agent into formulations for aerosols and lotions, a product is obtained that gives comparable levels of efficiency to current commercial products, but at much reduced levels of active loading. Accelerated stability tests performed on the final combination of the three actives used in the final formulation showed no adverse reactions over a three-week study. These tests shall be repeated once the final application form (lotion, aerosol, etc) and product packing have been decided.

- Description: The purpose of this dissertation was to provide a review of the theory of Optimization, in particular quadratic programming, and the algorithms suitable for solving both convex and non-convex quadratic programming problems. Optimization problems arise in a wide variety of fields and many can be effectively modeled with linear equations. However, there are problems for which linear models are not sufficient thus creating a need for non-linear systems. This dissertation includes a literature study of the formal theory necessary for understanding optimization and an investigation of the algorithms available for solving a special class of the non-linear programming problem, namely the quadratic programming problem. It was not the intention of this dissertation to discuss all possible algorithms for solving the quadratic programming problem, therefore certain algorithms for convex and non-convex quadratic programming problems were selected for a detailed discussion in the dissertation. Some of the algorithms were selected arbitrarily, because limited information was available comparing the efficiency of the various algorithms. Algorithms available for solving general non-linear programming problems were also included and briefly discussed as they can be used to solve quadratic programming problems. A number of algorithms were then selected for evaluation, depending on the frequency of use in practice and the availability of software implementing these algorithms. The evaluation included a theoretical and quantitative comparison of the algorithms. The quantitative results were analyzed and discussed and it was shown that the results supported the theoretical comparison. It was also shown that it is difficult to conclude that one algorithm is better than another as the efficiency of an algorithm greatly depends on the size of the problem, the complexity of an algorithm and many other implementation issues. Optimization problems arise continuously in a wide range of fields and thus create the need for effective methods of solving them. This dissertation provides the fundamental theory necessary for the understanding of optimization problems, with particular reference to quadratic programming problems and the algorithms that solve such problems. - Description: Introduction: A typical task in quantitative data analysis is to derive estimates of population parameters based on sample statistics. For manifest variables this is usually a straightforward process utilising suitable measurement instruments and standard statistics such the mean, median and standard deviation. Latent variables on the other hand are typically more elusive, making it difficult to obtain valid and reliable measurements. One of the most widely used methods of estimating the parameter value of a latent variable is to use a summated score derived from a set of individual scores for each of the various attributes of the latent variable. A serious limitation of this method and other similar methods is that the validity and reliability of measurements depend on whether the statements included in the questionnaire cover all characteristics of the variable being measured and also on respondents’ ability to correctly indicate their perceived assessment of the characteristics on the scale provided. Methods without this limitation and that are especially useful where a set of objects/entities must be ranked based on the parameter values of one or more latent variables, are methods of paired comparisons. Although the underlying assumptions and algorithms of these methods often differ dramatically, they all rely on data derived from a series of comparisons, each consisting of a pair of specimens selected from the set of objects/entities being investigated. Typical examples of the comparison process are: subjects (judges) who have to indicate for each pair of objects which of the two they prefer; sport teams that compete against each other in matches that involve two teams at a time. The resultant data of each comparison range from a simple dichotomy to indicate which of the two objects are preferred/better, to an interval or ratio scale score for e d Bradley-Terry models, and were based on statistical theory assuming that the variable(s) being measured is either normally (Thurstone-Mosteller) or exponentially (Bradley-Terry) distributed. For many years researchers had to rely on these PCM’s when analysing paired comparison data without any idea about the implications if the distribution of the data from which their sample were obtained differed from the assumed distribution for the applicable PCM being utilised. To address this problem, PCM’s were subsequently developed to cater for discrete variables and variables with distributions that are neither normal or exponential. A question that remained unanswered is how the performance, as measured by the accuracy of parameter estimates, of PCM's are affected if they are applied to data from a range of discrete and continuous distribution that violates the assumptions on which the applicable paired comparison algorithm is based. This study is an attempt to answer this question by applying the most popular PCM's to a range of randomly derived data sets that spans typical continuous and discrete data distributions. It is hoped that the results of this study will assist researchers when selecting the most appropriate PCM to obtain accurate estimates of the parameters of the variables in their data sets.

- Description: Aspen Pharmacare, one of the leading pharmaceutical manufacturers in South Africa has embarked on a programme of improving the production processes currently employed at their Port Elizabeth site. With the introduction of new technology at the site and the move towards globalization, it became imperative that Aspen remain competitive in the market. The product of interest in this research, Degoran Plus tablets, is one of the company’s leading brand sellers. Upon investigation, it became apparent that this product created opportunity for process improvement using the new technology. The manufacture of Degoran Plus entails granulation, compression and coating of the product. Most opportunity for improvement was possible in the granulation stage because of the laborious nature of the present process. Degoran Plus tablets had a history of analytical failures, especially with regard to the dissolution rate of the final product, as well as other quality related issues. The product was not considered to be a “through-runner”, which resulted in bad production output, due to continual repeats of not only analysis but also reworks in production. A strategic decision was taken to manufacture Degoran Plus using the Collette Gral granulator as the equipment offered superior mixing capability when compared to the Bear planetary granulator. It was assumed that the granulation process would result in more uniform distribution of the actives. Upon producing a better granule, a final product of superior quality would be attained. The validation protocol stipulates that three samples be taken and tested from the powder mix. Nine samples taken from granulated bulk are treated in the same manner. The validation protocol further stipulates that the first three batches manufactured utilise the new process, and tested according to the protocol. The results obtained from the analysis are evaluated statistically and a conclusion and recommendation were derived based on the evaluation.

Electrochemical investigation of valve regulated lead acid batteries

- Description: One of the technical advances made by the lead-acid battery industry in the field of portable power supply was the development of the valve regulated lead-acid battery (VRLA). This battery reduced the necessity for periodic servicing in terms of having to replenish the cells with distilled water. Further, this new type of battery can now be installed near sensitive electronic equipment without the danger of acid spill or dangerous fumes being emitted. In addition, longer service performance is achieved in terms of life cycle capacity, when compared to the conventional flooded type batteries. However, the new type of battery requires the manufacturing of high precision electrodes and components with low tolerances for error. In order for the manufacturers to produce such a premium product, a thorough understanding of the electrochemistry of the inner components is necessary. None of the South African lead-acid battery manufacturers are currently making VRLA batteries to supply a very competitive global market, where a large range of sizes and capabilities are available. In order to introduce the VRLA battery into such a competing market in South Africa, a niche area for its application was identified in order to establish the viability of manufacturing such a battery locally. This is done by integrating the VRLA concept into an existing battery, such as the miners cap lamp (MCL) battery. Its application is specific with well-defined performance criteria in a relatively large consumable market in the South African mining industry. The study looked at various components within a local manufacturing environment that required a better understanding and modification of the processes to build VRLA MCL batteries. This included a detailed study of the manufacturing processes of the positive electrode. The study involved the investigation of the types of grid alloys used, the type of electrode design, such as tubular or flat plate, the addition of redlead to the paste mixing process and subjecting the batteries to accelerated life cycle testing. A better understanding of the oxygen recombination cycle was also performed in order to evaluate the correct use of certain design criteria in the manufacturing process. This included the study of the pressure release valve and the type of positive electrode used. The study also looked at developing an inexpensive analytical technique to evaluate the porosity of cured and formed electrodes using a glycerol displacement method. The monitoring of the state of health (SoH) of VRLA batteries on a continuous basis is an important parameter in unique applications such as remote power supply. A device was developed to monitor the SoH of VRLA batteries on a continuous basis. The working principle of the device was tested on a MCL VRLA battery. With the development of other types of VRLA batteries for specific applications such as in stand-by power supplies, the monitoring device would then be integrated in the battery design.

- Description: This research was directed at reducing arsenic levels in the effluents generated at the Canelands facility that manufactures monosodium methyl arsenate. Two effluent streams containing arsenic have to be considered, a raw water stream that is treated on site and a brine stream that is disposed of by sea outfall. Removal of arsenate from aqueous media by coagulation was investigated and models were developed describing selected variables that influence the removal of the arsenate. Three coagulant systems were investigated, namely aluminium(III) coagulation, iron(III) coagulation and binary mixtures of aluminium(III) and iron(III). Researchers have studied individual aluminium (III) sulphate and iron(III) chloride coagulation. No detailed research and modelling had, however, been carried out on the use of binary mixtures of aluminium (III) sulphate and iron (III) chloride coagulation of aqueous arsenate, nor had individual aluminium(III) sulphate and iron(III) chloride coagulation of arsenate been modelled at relatively high arsenate concentrations. The models that were generated were validated statistically and experimentally. The variables investigated in the aluminium(III) model included initial arsenate concentration, pH, polymeric flocculent concentration, aluminium(III) concentration and settling time. The variables modelled in the iron(III) coagulation were initial arsenate concentration, pH, polymeric flocculent concentration, and iron(III) to arsenic mole ratio. The modelling of the binary coagulant system included initial arsenate concentration, pH, iron (III) concentration, aluminium(III) concentration, and flocculent concentration as variables. The most efficient arsenic removal by coagulation was iron(III), followed by the binary mixture of aluminium(III) and iron(III) and the weakest coagulant was aluminium(III) sulphate. Scale-up coagulations performed on real raw water samples at a 50 litre volume showed that iron(III) was the most efficient coagulant (on a molar basis) followed closely by the binary mixture, while aluminium(III) coagulation was considerably weaker. The residual arsenic levels of the iron(III) and the binary coagulation systems met the effluent discharge criteria, but the aluminium coagulation system did not. Leaching tests showed that the iron(III) sludge was the most stable followed by the sludge of the binary mixture and the aluminium(III)-based sludge leached arsenic most readily. Settling rate studies showed that the flocs of the iron(III) coagulations settled the fastest, followed by binary mixture flocs and the aluminium flocs settled the slowest. The flocs of the binary mixture had the lowest volume, followed by the iron(III) flocs, while the aluminium(III) flocs were the most voluminous. Based on current operations of the raw water treatment plant the aluminium(III)-based coagulation is the most cost efficient. Given a relative costing of 1.00 for the aluminium(III) coagulation, the iron(III) chloride-based coagulation would be 2.67 times more expensive and the equimolar binary mixed aluminium(III)/iron(III) system would be 1.84 times the cost of aluminium(III) coagulation.

- Description: Currently, the approved anti-HIV drugs on the market only target the three HIV enzymes: reverse transcriptase, protease and more recently, integrase. Due to the limited nature of the current therapy, it is possible that a multi-drug resistant virus can emerge. The main concerns in developing countries however, are the expense and availability of the drugs and because of this, it is essential to investigate all alternatives. Traditional medicine offers many advantages as compared to allopathic treatment in so far as being relatively cheaper, accessible and it is broadly accepted in the population groups of the developing countries. Little is known though, of the exact efficacy and toxicity of these remedies so it is vital that these possible leads be investigated thoroughly. For the purpose of this study, two plants, Sutherlandia frutescens and Lobostemon trigonum were studied to ascertain their potential anti-HIV activity. Sutherlandia has received international attention as a possible cheap herbal remedy to improve the health of AIDS sufferers. Anecdotal evidence from health workers claim that HIV- infected patients on Sutherlandia treatment have shown improved CD4 counts, decreased viral loads and a general improvement in well-being. Extracts were prepared from dried leaves and flowers in methanol, ethanol, acetone, methylene dichloride or distilled water. Sulphated polysaccharides have been described extensively in literature with regards to their anti-HIV activity, so as a form of dereplication; an ethanol precipitation was performed on the aqueous extracts to remove sulphated polysaccharides. A toxicity study was performed on all crude extracts using uninfected peripheral mononuclear blood cells (PBMCs) isolated from whole blood. To measure anti-HIV activity, HIV-infected PBMCs were cultured with each of the crude extracts and cell viability measured using the tetrazolium salt, XTT. HIV-infected CEM-NKR-CCR5 cells were also used and supernatant from the viral studies was tested for the HIV antigen p24. xii Results varied greatly between assays but with the inclusion of a point-scale system to evaluate the extracts it was clear that overall the organic extracts of the Sutherlandia flowers, especially the acetone extract (SFA), showed great anti-HIV potential. SFA in every case decreased p24 levels and in the toxicity study did not decrease cell proliferation. With the HIV-infected PBMCs SFA actually helped improve cell proliferation despite the infection. To determine the specific anti- HIV activity, all crude extracts were tested for inhibition of HIV-I reverse transcriptase, the glycohydrolase enzymes: a-glucosidase, ß-glucosidase, ßglucuronidase, HIV-I integrase and HIV-II protease. No significant inhibition was seen with these experiments except for the HIV-I RT assay. The aqueous extract of the Lobostemon leaves produced an inhibitor of HIV-RT with a very low IC50 value of 0.049mg/ml. Some inhibitory effect was lost with the removal of the sulphated polysaccharides and the addition of BSA to the assay, but still 64% inhibition of the HIVRT remained, which confirmed that the inhibitor could be something novel, and not of the polysaccharide or tannin compounds.

- Description: Zinc oxide (ZnO) is a II-VI semiconductor material that offers tremendous potential as a light emitter in the blue-to-UV range. It has a wurtzite structure, and a direct band gap that can be tuned from 3.0 to 4.0 eV by alloying with Cd or Mg, respectively. In this work, ZnO thin films were grown by metalorganic chemical vapor deposition (MOCVD) on n-Si 2 ° off (100), amorphous glass, n-GaAs (100), and c-plane sapphire substrates. Diethyl zinc (DEZn) and tert-butanol (TBOH) were chosen as precursors. For the first time, Second Harmonic Generation Imaging was applied to the mapping of ZnO epilayers. The images obtained highlighted the polycrystalline character of the thin films, and provided insight as to the growth mode of ZnO on Si. The influence of substrate temperature on the structural properties of the epilayers was investigated by X-ray diffraction and optical microscopy. Grain sizes as high as 54 nm were measured. The optimum temperature range for this system proved to be 450 – 500 °C. The influence of the VI:II ratio during growth on the optical properties of the epilayers was studied by UV-vis-near IR spectroscopy. The lowest Urbach tail E0 parameter was measured for material grown at a VI:II ratio of 18:1. The films’ free electron concentration was shown to decrease by over two orders of magnitude, from 1019 to 1017 cm-3, as the VI:II ratio increased from 10 to 60:1. This decrease in carrier concentration with rising VI:II ratio was paralleled to the surge at 12 K of a photoluminescence (PL) emission band characteristic of p-type ZnO. The band gap energies extracted from room temperature transmission spectra ranged between 3.35 and 3.38 eV, in agreement with the value of 3.35 eV measured by room temperature PL. Moreover, variable temperature PL spectra were recorded between 12 and 298 K on ZnO grown on Si. The 12 K spectrum was dominated by a donor-bound exciton (D°X) at 3.36 eV, while the 298 K scan displayed strong free exciton emission (FX) at 3.29 eV. The width of the D°X band proved to be as narrow as 7 meV. The intensity ratio between the room temperature near-band edge emission and the defect-related green band was as high as 28:1, highlighting the optical quality of the layers deposited in this work. The electrical properties of the thin films were studied by Hall measurements (van der Pauw configuration), and a maximum room temperature mobility of 11 cm2/Vs was recorded. Furthermore, a palladium (Pd) Schottky barrier diode on ZnO was fabricated. The barrier height and ideality factor were calculated from current–voltage measurements to be 0.83 eV and 1.6, respectively. The capacitance–voltage curve of the diode yielded a carrier concentration in the depletion region of 8·1017 cm-3. This study has shown that the optical and electrical properties of ZnO depend strongly on the growth conditions employed. A suitable choice of growth parameters can yield high quality ZnO that may be used for various devices. - Description: Chemical synthesis is not complete without waste treatment. There is no chemical process that does not produce waste. The untreated effluent discharged by chemical industries poses a major environmental concern. This dissertation describes the results of an investigation carried out on the formation of by-products (impurities) during the synthesis of atrazine by neutralization of the reaction mixture during the actual synthesis procedure, and prior to disposal using carbon dioxide or hydrochloric acid. The main objectives for the said investigation were: {u100083} To optimise the reaction parameters to obtain a higher yield and purity of atrazine. {u100083} To evaluate the use of carbon dioxide and hydrochloric acid to neutralise the reaction mixture. {u100083} To study the solids present in the effluent. Synthetic reactions were carried out in the presence of an organic solvent, methyl isobutyl ketone (MIBK). The synthesis of atrazine was optimised in terms of addition sequence and rates of amine reagents and base (proton acceptor) additions by means of bench scale reactions. It was found that addition of a neutralising step (hydrochloric acid or carbon dioxide) to neutralise the basic reaction mixture prior to and after addition of the secondary amine and the base minimizes the formation of by-products. Of particular importance in ensuring high yields and selectivity to the atrazine product was temperature control, pH control, reagent ratios, addition rates and mixing. The latter was important in view of the rapid reaction of amine/NaOH with cyanuric chloride, as well as the possible reaction of mono-i, the reaction intermediate, with IPA in cases of excesses of reagent. Under optimum conditions, an atrazine composition of >97% was achieved where the resultant product was well within stipulated product specifications.

Purification and characterisation of 20S proteasome from ostrich skeletal muscle and its role in meat tenderisation

- Description: The proteasome is renowned for its high molecular weight, multisubunit and mulicatalytic nature. One of its many suggested roles is the degradation of myofibrillar proteins, and therefore it has been proposed to play a role in the meat tenderisation process. The aim of this study was therefore to isolate, purify and characterise the 20S proteasome from ostrich skeletal muscle, with a view to ultimately investigating its role in the tenderisation process of ostrich meat. The 20S proteasome was successfully isolated and purified from ostrich skeletal muscle using Toyopearl Super Q-650S, Sephacryl S-300, hydroxylapatite and Mono Q chromatographies. The intact molecule showed a molecular weight of 725 K and a pI of 6.67. The subunits showed a molecular weight range of 22.2-33.5 K and a pI range of 3-9. 2D-PAGE revealed at least 14 polypeptides. The amino acid composition of the intact enzyme and of each of the eight subunits separating on SDSPAGE, as well as the N-terminal sequences of five of the eight subunits, were determined. The trypsinlike (Tr-L), chymotrypsin-like (ChT-L), peptidylglutamyl peptide hydrolase (PGPH) and caseinolytic activities showed pH optima of 11, 9, 7-8 and 10.3, and temperature optima of 40, 60, 70 and 60oC, respectively. The pH stability range for all four activities was 5-12. The ChT-L and PGPH activities showed thermostabilities up to 60oC, whereas the Tr-L and caseinolytic activities were stable up to 40o C. The enzyme showed complex kinetics. It was inhibited by the peptide aldehyde Z-LLL-CHO and cysteine protease inhibitors. Cations had negligible effects on the enzyme, excepting for Ca2+ and Mg2+. Of the detergents tested, SDS had the most potent stimulatory effect, particularly on the PGPH and caseinolytic activities. The fatty acid studies showed that unsaturation enhanced the ChT-L and the caseinolytic activities, while it completely suppressed the Tr-L activity. Heating at 60oC for 1-2 min stimulated the caseinolytic and PGPH activities. The studies on the role of ostrich skeletal muscle 20S proteasome in ostrich meat tenderisation suggested a definite but minor role of this enzyme, based on the fact that it remained active throughout the 12 days of storage of ostrich M. iliofibularis meat at 4oC and that it participated in myofibril degradation of post-mortem muscle, but to a small degree. These results support the proposal that the proteasome comes into play after the calpains have initiated degradation. However, there was a lack of improvement in tenderness values and minimal myofibrillar degradation over the 12-day storage period of the ostrich M. iliofibularis meat, leading to the conclusion that the tenderisation of this meat was incomplete after 12 days.

- Description: The aim of the first study was to determine if barley could replace maize as an energy source in concentrates fed to dairy cows grazing on kikuyu/ryegrass pasture without affecting the milk production, milk composition, or cause metabolic disorders. Sixty Jersey cows, in early to mid lactation were randomly allocated to one of five treatments (n = 12) based on feeding concentrates with different ratios of maize to barley, ranging from 100:0, 75:25, 50:50, 25:75 and 0:100, respectively. Concentrates contained 12 MJ ME kg -1 and 130g CP kg-1 DM and cows were fed 3 kg (as-is) concentrate after each milking for a period of 42 days (14-day adaptation and 28-day measurement). Cows strip-grazed the irrigated kikuyu/ryegrass pastures (15.7 ± 1.8 percent DM; 20.2 ± 4.3 percent CP; 44.7 ± 3.5 percent NDF). Milk weights were recorded daily and milk samples collected weekly and analyzed for milk fat and protein content. Body condition score and live weight were recorded at the start and end of the experimental period. Data of all the studies were subjected to a one-way ANOVA. Daily milk yield, FCM, MUN, milk fat yield, milk fat percent, protein yield, protein percent, live weight change, or body condition score change were not affected by treatment and values were 15.8 kg, 17.2 kg, 14.9 mg dl-1, 0.72 kg, 4.56 percent, 0.59 kg, 3.77 percent, 6.67 kg, and 0.15 BCS; 15.6 kg, 17.4 kg, 15.2 mg dl-1, 0.73 kg, 4.3 percent, 0.57 kg, 3.71 percent, 1.33 kg and 0.04 BCS; 17.2 kg, 17.9 kg, 15.2 mg dl-1, 0.74 kg, 4.36 percent, 0.63 kg, 3.71 percent, 0.33 kg and 0.08 BCS; 15.6 kg, 16.4 kg, 15.5 mg dl-1, 0.67 kg, 4.33 percent, 0.60 kg, 3.83 percent, -1.46 kg and 0.11 BCS; and 15.0 kg, 16.0 kg, 15.5 mg dl-1, 0.67 kg, 4.57 percent, 0.57 kg, 3.85 percent, 8.86 kg, and 0.05 BCS, respectively for the cows fed 100:0, 75:25, 50:50, 25:75 and 0:100 maize to barley ratio concentrate. According to these results, barley can replace maize without significantly affecting the milk production or milk composition. None of the cows presented any visible symptoms of acidosis. As it was clear from the results in the first study that maize could replace barley the aim of study 2A was therefore focused on determining the effect of feeding different levels of such a barley-based (2.4, 4.8 or 7.2 kg cow-1day-1) concentrate, on milk production, milk composition and live weight change of Jersey cows on kikuyu/ryegrass pasture (23.1 ± 2.95 percent DM, 11.1 ± 0.11 percent CP, 60.8 ± 0.58 percent NDF). Forty-five Jersey cows (early- to mid lactation), were randomly allocated to one of three treatments (n = 15) involving different levels of concentrate (12 MJ ME and 130g CP kg-1 DM) feeding for a period of 42 days (14-day adaptation and 28-day measurement). Milk weights were also recorded daily, and milk samples collected weekly, and analyzed for milk fat and protein. Body condition score and live weight were recorded at the start and end of the experimental period. The results of this study indicated that increasing the concentrate level from 2.4 to 4.8 and 7.2kg cow-1day-1 did not increase the milk yield (14.0 kg, 15.2 kg, 14.4 kg; P = 0.19). The FCM production increased from 15.8 to 17.5kg (P = 0.04) as the concentrate level increased from 2.4 to 4.8kg cow-1day-1. Increasing the concentrate from 4.8 to 7.2kg cow-1day-1 did not result in a significant increase in FCM. The milk protein percent increased significantly from 3.4 - 3.6 percent when the concentrate feeding level was increased from 2.4 to 7.2kg cow-1day-1. The MUN levels were 17.09 mg dl-1, 16.03 5 mg dl-1, and 16.36 mg dl-1 for the 2.4, 4.8 and 7.2kg cow-1day-1 concentrate levels, respectively. This is well within the recommended MUN levels (12 – 18 mg dl-1) indicating that sufficient protein was fed to cows. Increasing the concentrate level from 4.8 to 7.2 kg cow-1day-1 did not increase production, probably due to a higher pasture substitution rate. Supplementing large quantities of rapidly fermentable grains, such as barley, can suppress rumen pH and may have a negative effect on the rate and extent of fibre digestion in the rumen. Therefore the aim of study 2B was not only to determine the effect of feeding different levels of a barley-based concentrate, on milk production, milk composition and live weight change, but was also to determine the effect of a low (4.8 kg cow-1day-1) versus a high (7.2 kg cow-1day-1) level of barley-based concentrate supplementation on ruminal DM and NDF degradability of Westerworld ryegrass sampled from the pastures that these cows were grazing on. Sixty Jersey cows (early- to mid lactation), were randomly allocated to one of three treatments (n = 20) involving different levels of concentrate feeding. Concentrate (12 MJ ME, 130g CP kg-1 DM) was fed at 2.4, 4.8 or 7.2 kg cow-1day-1 for a period of 74 days (14-day adaptation and 60-day measurement). These cows stripgrazed irrigated kikuyu/ryegrass pastures (14.7 ± 4.37 percent DM, 25.1 ± 1.53 percent CP, and 44.4 ± 2.58 percent NDF) at a daily pasture allocation of 10 kg DM cow-1. Milk weights were recorded daily and milk samples collected weekly and analyzed for milk fat and protein. Body condition score and live weight were recorded at the start and end of the experimental period. Twelve Jersey cows, fitted with ruminal cannulae, were randomly allocated to two of the three treatments in the production study and received either 2.4 or 7.2 kg cow-1 day-1, of the same barley-based concentrate, in a two-period crossover design. These cows strip-grazed the same irrigated kikuyu/ryegrass pastures as the sixty cows in the production study. Each period consisted of 21 days for adaptation and seven days for data collection. Rumen liquor samples were collected every 4 hours within a 24-hour cycle and repeated once. Rumen pH was measured immediately, recorded, and the supernatant fluid preserved and frozen, pending VFA analysis. The in situ nylon bag technique was used to determine DM and NDF degradation and dried samples of Westerworld ryegrass pasture were incubated for 0, 4, 8, 12, 20, 30, 48, 72 and 96 hours. The data were fitted in the non-linear model p = a + b (1-exp-ct) (Ørskov & McDonald, 1979). Daily milk production, fat corrected milk, milk fat yield and milk fat percent were not affected by treatment and values were 17.3 kg, 18.4 kg, 0.76 kg and 4.42 percent; 19.0 kg, 20.0 kg, 0.82 kg and 4.35 percent; and 18.1 kg, 19.1 kg, 0.79 kg and 4.37 percent for the 2.4, 4.8 and 7.2 kg cow-1 day-1 concentrate treatments, respectively. Milk protein percentage of cows on the 7.2 kg concentrate cow-1 day-1 was significantly higher than that of cows on 4.2 kg concentrate cow-1 day-1 feeding level. Live weight increased significantly as the level of concentrate feeding increased and values were 17.9 kg; and 28.9 kg on the 2.4 and 7.2kg concentrate treatment, respectively. There was a significant increase in the live weight of cows that were fed 7.2 kg cow-1 day-1 (as-is) in comparison to those cows that were fed 2.4 kg concentrate cow-1 day-1 (as-is). This may have resulted from more nutrients being partitioned to live weight gain rather than milk production. No further response in milk production was observed when concentrate daily feeding was increased from 4.8 to 7.2 kg cow-1 day-1. It is postulated that the higher concentrate allowance resulted in a higher substitution rate and lower DMI intake from pasture. 6 There was no significant decline in the rumen pH (6.2 ± 0.4 and 6.2 ± 0.5) when the concentrate level was increased from 2.4 to 7.2 kg cow-1 day-1 (as-is). The total VFA (118.1 ± 45.9 and 139.4 ± 45.6 mmol L-1) and isovalerate (0.009 ± 0.07 and 0.248 ± 0.52 mmol L-1) increased significantly when the concentrate was increased from 2.4 to 7.2 kg cow-1day-1. No other rumen parameters were affected by treatment. Ruminal DM and NDF degradability of the Kikuyu/ryegrass pature were not affected by the level of concentrate supplementation. An increase in the concentrate level from 2.4 to 7.2 kg cow-1day-1 did not reduce degradability of either DM (94.67 ± 5.97, 94.49 ± 5.09; P = 0.919) or NDF (92.15 ± 8.69, 94.4 ± 11.73; P = 0.451), respectively. Results of rumen parameters and PD values were within the range reported by Bargo et al., (2003), viz. pH 5.76 – 6.29, NH3-N concentration 8.7 – 32.2 mg dl-1, total VFA concentration 90.3 - 151.4 mmol L-1 and PD values 89.5 – 93.5 % reported by Bargo et al. (2003). According to these authors, there is no simple relationship between any amount of the concentrate supplemented, and the ruminal pH and concentrate feeding only affects the in situ ruminal digestion of pasture when it is fed, at quantities higher than 8 kg DM cow-1day-1 (Bargo et al., 2003).

- Description: This project is concerned with the development of a search algorithm for a large archival database. The Port Elizabeth Genealogical Information System (PEGIS) contains a database consisting of almost 600000 individuals. The standard search algorithms are no longer sufficient to locate individuals in the database. A new algorithm was required that allows searches on any of the words or dates in the database, as well as a means to specify where in the desired record a word should occur. A ranking function of retrieved records was also required. A literature study on the field of Information Retrieval and on algorithms designed specifically for the PEGIS was done. These algorithms were adapted and hybridized to yield a search algorithm that allows for the boolean formulation of queries and the specification of the structure of search words in the desired records. The algorithm ranks retrieved records in assumed relevance to the user. The new algorithms were evaluated with regards to retrieval speed and accuracy and were found to be very effective.

- Description: Although the marine environment has generally been considered to be less in need of attention from conservationists, politicians and managers than the terrestrial environment, there has been an increasing appreciation that there is little left in the marine environment that can be considered unaltered by humans. Conservation action is therefore urgently needed to protect marine populations, species and environments from anthropogenic pressures. Since overharvesting seems to be the main cause of the collapse in coastal ecosystems, areas need to be protected so that stocks can recover. However, the proclamation of such marine protected areas (MPAs) need to be well planned to ensure that species are well represented and that their biological requirements are met in the long term. While terrestrial conservation planners have started to incorporate biological, social and economic information into so-called systematic conservation plans, marine planners have been more hesitant to use these approaches due to the greater openness of the ocean (with the associated greater dispersal of marine organisms and propogules, lower levels of endemism, and lack of ownership) and the paucity of available information. The current study aimed to investigate the applicability of systematic approaches to the process of selecting sites for a network of potential replenishment zones for seabream species that could supplement protection already afforded by existing no-fishing reserves. Since many of these seabream species are popular targets in both the recreational and commercial linefishery sectors, they have been overexploited and are in need of additional protection. Information on these species and the diversity of their associated reefs were obtained through consultations with scientists, managers and resource-users (namely SCUBA divers, spearfishers as well as recreational and commercial fishers). These data were obtained during workshops where Participatory Rural Appraisal (PRA) scoring and mapping methods were applied. Although ordinal-scale, rather than interval-or ratio-scale data were obtained, these data could be used in an interactive selection process. Because these methods require relatively cheap materials and few personnel, they could be used to collect information where limited budgets are available. Quantitative targets for the protection of individual species were set on the basis of their status in the Marine Living Resource Act, since this reflected their different requirements for protection. A preliminary investigation indicated that the current no-fishing reserves effectively achieved most of these goals for seabream species. The selection of sites for the replenishment network was based on irreplaceability analyses, while decisions between sites with lower irreplaceability values were based on explicit design rules. A total of 26 sites were required to achieve targets for all species. These replenishment zones could potentially be established, managed and monitored by voluntary community groups. This study therefore indicated that systematic conservation planning can successfully be applied to the marine environment, while the involvement of stakeholders is crucial to ensure that conservation action is supported.

The concentration of ten metals in the tissues of shark species Squalus megalops and Mustelus mustelus (Chondrichthyes) occuring along the Southeastern coast of South Africa

- Description: The southeastern coast of South Africa is deemed unpolluted and with the exception of a few localized areas, is believed to be pristine. This study aimed to (1) assess the concentration of metals in the water and sharks (Squalus megalops, Mustelus mustelus) from different sites along the southeastern coast, (2) determine the effects that metals have on each other and investigate the accumulation and storage of metals in different shark tissues, (3) determine the effects of length and sex on metal concentration and the presence of metallothioneins in the liver. Metal concentrations in water from seven sites along the southeastern coast revealed concentrations below the detection limit of the extraction method, except for iron at St. Francis Bay which was below the recommended levels as supplied by DWAF. Met al concentrations in S. megalops from the seven sites revealed significantly elevated levels in individuals from Algoa and St. Francis Bay when compared to the other 5 sites. This was observed for all metals except manganese, which had a higher concentration at Cape Agulhas. After comparing the metal levels in muscle tissue (S. megalops) with data from the Department of Health it was concluded that the sharks caught at Algoa and St. Francis Bay were unsuited for human consumption. Liver and vertebral tissues from both S. megalops and M. mustelus were deemed unsuited for human consumption but further research should be conducted especially on vertebral metal concentrations. Comparing the metal concentration in different tissue types of S. megalops and M. mustelus significant difference were observed and attributed to differences in the maturity (thus age) of the two species. Significant differences in the metal concentration of males and females for both the species were observed. A higher number of differences were however observed in M. mustelus than in S. megalops, most probably due to the differences in maturity. Length and weight vs. metal concentration regressions in the males and females of both species revealed more negative than positive trends. This was found to be in accordance with other studies from around the world. Using the life stages of S. megalops, significant differences in the average concentration of metals were observed between the life stages, with embryos usually having the highest concentration. The metal concentration series determined for S. megalops were similar to that which have been reported in the literature in that iron and zinc occupied the highest concentration positions while cadmium, copper and manganese occupied the last three. The spleen usually had the highest metal concentration except for aluminium in skin and arsenic in muscle tissue. Pearson correlation revealed significant negative correlations between cadmium and the mentioned metals suggesting that cadmium replaces essential metals such as manganese, iron and copper as. Negative correlations between essential metals were observed most probably due to the competition for binding positions on ligands. Positive correlations were observed between metals in sp ecific tissue types and may be indicative of synergistic effects, e.g. aluminium may result in the accumulation of other metals such as cadmium. Positive and negative correlations between a specific metal and different tissue types have also been observed and it appears that there are movement of metals between tissue types. Positive correlation for essential metals between yolk and other tissue have been observed, although no correlations for non-essential metals were observed. This could potentially indicate a mechanism that prevents the accumulation of nonessential metals in embryos. Though no metallothioneins were isolated form the liver, the presence of cadmium in other proteins indicate that that cadmium is substituted for essential metals, and as this may result in deleterious physiological effects, further studies should be conducted. In conclusion it can be said that although some of the results were expected there are still quite a few unanswered question and further studies should be conducted.
