- Description: There are apprehensions that genetic modification of maize with Bacillus thuringiensis (Bt) may have negative effects on soil biodiversity, ecosystem processes and functions. This study aimed at determining the effect of Bt maize crop, Bt maize residues and its genetic modification on microbial biomass carbon (MBC), selected enzyme activities, vesicular arbuscular mycorrhizal (VAM) fungi and N and P release patterns. The study was conducted under field, glasshouse and laboratory conditions. In 2010/2011 season, four maize cultivars; DKC 61-25B (Bt), DKC 61-24 (non-Bt), PAN 6Q-321B (Bt) and PAN6777 (non-Bt) were planted. Determination of MBC, enzyme activities and fungal spore count was done at 42, 70, and 105 days after planting (DAP). A loam soil amended with Bt or non-Bt maize leaf residues from a study of 2009/2010 season was incubated to investigate effects of Bt maize residues on MBC and soil enzyme activities. Leaf residues of Bt and non-Bt maize cultivars (DKC 61-25B, DKC 61-24, PAN 6Q-321B and PAN6777) were used and soil without residues was used as a control. Samples were collected at 7, 28 and 56 days of incubation (DOI). An incubation study was also carried out in the laboratory to determine the effect of Bt maize residues (i.e. leaf, stem and root) and its genetic modification on N and P release patterns. - Description: The study was conducted in Eastern Cape Province to 1) investigate local people’s knowledge and perceptions on Euryops floribundus and 2) landscape distribution and effect of Euryops floribundus on herbaceous vegetation and soil. A total of 90 households who own livestock were randomly selected from the three communal areas, namely; Tsengiwe, Upper Mnxe and Manzimdaka for household surveys and group discussions. For landscape study, a total of about 5 ha of land were selected at Upper Mnxe communal area. Selection criteria included the presence of E. floribundus and different landscape gradients. A total of four 50mx50 plots were marked along the landscape gradients: Bottom, Middle, and Upper Slope and upland positions. To study the effect of E. floribundus invasion on the herbaceous vegetation and soil, a total of 4 ha communal land was selected in an accessible area which had a largely flat terrain. The area was selected to have adjacent sites with no invasion (< 5% shrub cover), light (5-15% shrub cover), moderate (>15-35) and heavy invasions (>35% shrub cover) of E. floribundus. The average household size in the study area was 6.8± 0.7. All people in the three communal areas unanimously ranked sheep as the most important species for their livelihood, but the ranking of cattle and goat varied among the communal areas. All elder groups agreed that Europs floribundus decreases both the quantity and quality of herbaceous forage as well as livestock production. Moreover, invaded patches create more bare areas which are responsible for the loss of top fertile soils and the formation of rills and gullies. The result showed that the total density of E. floribundus significantly increased from the topland (2301 plants ha-1) moving to the bottomland (4888 plants ha-1). Canopy cover was significantly lowest in the topland (17.9%), but the remaining gradients had similar cover. Grass dry matter yield was higher in the bottomlands and sloppy gradients than the toplands. Soil organic carbon was significantly higher (P<0.05) in the flat than the sloppy terrain. Soils from higher elevations (toplands and upper sloppy) had significantly higher N than the lower elevations (bottomlands and Middle sloppy). The lowest dry matter was observed at low invasion and non-invaded sites. Soil chemical properties were generally different from all density levels. In conclusion, E. floribundus encroachment was found to be major cause of decline in the peoples’ livelihood because as this reduces the vegetation diversity and livestock production, both of which are the major of their livelihoods.

Evaluation of range condition, soil properties, seed banks and farmer's perceptions in Peddie communal rangeland of the Eastern Cape, South Africa

- Description: South African rangelands in combination with their surrounding homesteads occupy 13% of the entire land surface in South Africa. These rangelands are a source of forage for communal livestock. The rangelands in communal tenure system are degraded due to high human population and livestock numbers. The rangeland of Peddie was never evaluated since the introduction of Nguni Cattle Empowerment Project. Therefore, socio-ecological evaluation was conducted in order to interlink farmer‟s perceptions and scientific data to recommend appropriate rangeland management and restoration programme. Two structured questionnaires consisting of close and open ended questions were used to investigate farmer‟s perceptions on rangeland condition, dynamics, and their causes. Sixty households were randomly selected on the bases of livestock ownership and the membership in Nguni Cattle Project. In each household, any respondent of 20 years or greater, and a key informant of age greater than 40 years were selected. For scientific assessment of range condition, three homogenous vegetation units namely grassland, scattered and dense bushland were demarcated into four 100m x 50m replicates. In each replicate, two 100m transects were laid parallel to each other with 30m equidistant apart. The step point and harvesting method along each transect were employed for herbaceous species composition and biomass production. The point-to-tuft distance was also determined as a proxy for basal cover. Woody density, species composition and tree equivalents were determined in 200m2 belt transects in each HVU replicates. The germination method for soil seed bank evaluation was also employed to find plant species composition and density. The soil nutrients (OC, N, P, K, Na, Ca, Mg, Zn, Cu and Mn) and pH were analysed through solution preparation and observation under photospectrometer to determine functional capacity of the soil of Peddie rangeland. The farmer‟s perceptions comprised of 63% females and 37% males (n = 120) with a mean household of 8 people, 5 adults and 3 children. It was perceived by 93.3% respondents that the rangeland of Peddie have undergone changes over two decades. These changes were perceived by 83% respondents to be accompanied by decline in livestock numbers. Woody encroachment and overgrazing were perceived to be the major attributes of these vegetation changes. The scientific rangeland condition assessment confirmed that these changes were more pronounced as bush density increases. Dense bushland had a significantly high (p<0.05) encroached condition with 6650 trees ha-1 and 4909.5 TE ha-1 beyond the recommended thresholds of 2400 trees ha-1 and 2500 TE ha-1 respectively. Scattered bushland had a fair condition of 1950 trees ha-1 and 1198.1TE ha-1. Themeda triandra as a key species was significantly higher (p<0.05) in grassland (31.1%) than scattered (15.6%) and dense bushland (6.1%). There was a declining trend in biomass production from grassland to dense bushland. The summer biomass production was significantly higher (p<0.05) in grassland than scattered and dense bushland but winter biomass was not significantly different (p>0.05) from all homogenous vegetation units of Peddie rangeland. However, the soil fertility increased with an increase in bush density except organic carbon (OC) which was 1.61% in grassland, 1.46% in scattered and 1.53% in dense bushland respectively. Soil N, K, P, Mg2+ Na+, Ca2+, Cu, Zn, Mn and pH were significantly higher (p<0.05) in dense bushland than grassland and scattered bushland. High soil fertility in dense bushland may be attributed to by abscission of woody plants and litter decomposition. In the soil seed bank, the abundances of forbs were significantly higher than sedges (χ2 = 12, df = 1, p = 0.001) and grasses (χ2 = 8.333, df = 1, p = 0.004) in all homogenous vegetation units while sedges were not significantly different (χ2 = 3, df = 1, p = 0.083) from grasses. The Sorensen‟s index indicated that soil seed bank and extant vegetation were significantly different (p<0.05). Annual and biennial forbs and sedges had high abundances while perennial grasses formed a bulk in above ground vegetation. This provided an insight that a reliance on soil seed bank for restoration of Peddie rangeland would not be advisable because it can result in retrogression. The communal rangeland assessment provided clear qualitative and quantitative data when the combination of indigenous knowledge and scientific assessments was done. The rationale is that conclusions and recommendations of range assessment are relient on the farmer‟s perceptions pertinent to their livestock production systems and their rangeland management objectives. This study has shown that inclusion of communal farmers in policy making can provide better insight because those are the people experiencing the consequences of range degradation.

- Description: An increase in diarrhoea and upper respiratory tract infection has been noted for the past five years and this is associated with the prevalence of Human Immune-deficiency Virus and Acquired Immune Deficiency Syndrome. HIV infection is an increasingly common cause of childhood morbidity and mortality in South Africa. In some areas of the country, like the Eastern Cape, more than 30 percent of the women attending antenatal clinics are HIV infected. Without intervention, approximately one third of the babies born to these HIV infected mothers will get infection from their mothers (WHO, 2003). Aim and Objectives of the study: The study was meant to assess the specific knowledge and skills of professional nurses at the primary health facilities regarding management and integration of childhood illnesses, evaluate the quality of training of professional nurses on aspects of IMCI including integration of HIV within IMCI programme for children under five years in Primary Health Care facilities in Mdantsane Township. Methods: A quantitative descriptive method was used for this study. The questionnaire developed by the researcher was used for data collection. The items on the questionnaire were divided into four (4) subsections. The questionnaire was administered to all professional nurses on duty and those available at the time of data collection. A checklist for IMCI was used to collect data from Road to Health files of children who were HIV positive admitted in the pediatric ward at Cecilia Makhiwane Hospital. Results: The results of this study for all the variables examined showed that the highest score was 56 percent and the lowest was18 percent.These results indicated that the knowledge and skills of professional nurses were inadequate.

- Description: An increase in diarrhoea and upper respiratory tract infection has been noted for the past five years and this is associated with the prevalence of Human Immune-deficiency Virus and Acquired Immune Deficiency Syndrome. HIV infection is an increasingly common cause of childhood morbidity and mortality in South Africa. In some areas of the country, like the Eastern Cape, more than 30 percent of the women attending antenatal clinics are HIV infected. This is a period of adjustment, stress, growth and development and the transitional nurse is likely to feel uncomfortable, fearful and may experience feelings of inadequacy. This study was undertaken to explore and describe the experiences of final year nursing students at a public college of nursing in the Eastern Cape regarding their preparedness to become registered nurses, with the aim to identify gaps and make recommendations on strategies to close the gaps. The objectives were to explore and describe the experiences of final year nursing students at the public college of nursing in the Eastern Cape Province with regard to their preparedness to become registered nurses and to recommend strategies to improve the preparation of nursing students for transition to become registered nurses. To answer the research question “What are the experiences of final year nursing students at a public college of nursing in the Eastern Cape Province with regard their preparedness to become registered nurses”, a qualitative, explorative and descriptive design was used as a framework for the study. Data were collected by means of unstructured focus group interviews with a purposively selected sample of 27 final year nursing students at the particular college of nursing. Data were analysed using Tesch’s method of analysis for qualitative research. Two themes emerged, revealing that participants at the college experienced preparedness and lack of preparedness to assume the role of a professional nurse. This related to certain aspects that had an impact on the preparation of the final year nursing students for practice and included: curriculum-related aspects; clinical teaching and learning support; learning opportunities; interpersonal relationships between lecturers, students and clinical staff; equipment; and library resources. It was concluded that the learning needs of the nursing students were not adequately catered for, leading to lack of preparedness. Through the involvement of nursing students in the evaluation of their learning, shortfalls in both education and practice areas could therefore be detected. Recommendations regarding strategies to be used to promote preparedness of final year nursing students included: continuous feedback on student performance in the form of exit evaluations on the part of students to identify learning needs; writing of progress reports on the part of clinical practice; and the employment of clinical preceptors with clear role specifications between the lecturers, ward sisters and preceptors to avoid role confusion.

Some negative experiences were observed, like issues of confidentiality, depression and stress. Conclusion: University support groups can be improved through structured programs that includes various stakeholders like health professionals (wellness centre), university management, and others including academics within the university. Also, expanding their borders via the use of social networks, and expanding their activities to include music, sports, vegetable gardening, group visitations, skills training, and other useful projects shall help to improve lives of victims and fight against the infection of other community members.

The Land Bank of South Africa has shown keen interest to provide funds to set up an irrigation scheme in Guquka. Absence of peoples’ involvement and farmer participation has been identified as one of the causes of poor performance of smallholder irrigation schemes in South Africa. The poor performance of smallholder irrigation projects in South Africa provided a good basis for exploring factors that could possibly affect farmer participation and the success of the proposed irrigation scheme. This knowledge would be useful to providers of extension services, capital providers, policy makers and the recipients of the proposed irrigation scheme. The main objective of this study was to investigate the factors affecting the willingness of farming households to participate in the proposed irrigation scheme. This investigation helped to analyse demographic; socio economic situation; farming practices; economic activities; water sources, uses and management in the study area. The major tool of enquiry in this study was the questionnaire which was used to collect data from the households. Household and farm characteristics were collected using structured questionnaires with the help of locally recruited and trained enumerators. The data used for the empirical analysis was obtained from a survey of 50 farming households in Guquka, in the Eastern Cape Province of South Africa. The following variables were selected to determine the scope of their influence on farmer willingness to participate in the proposed scheme: age; membership in local group; investment in agriculture, educational level of the farmers, knowledge, household size, labour and gender. A logit model was used to determine the extent to which these selected characteristics influence the willingness of farmers to participate in the proposed irrigation scheme. The results showed that willingness to participate in the scheme was significantly influenced by the afore mentioned variables as well as knowledge about the planned irrigation scheme which is mainly provided by extension agents; and total household income. Age was found to decrease the willingness of farmers to participate in irrigation. A positive association existed between participation and the following variables; membership in local group, investment in agriculture, educational level of the farmers and knowledge. On the other hand, household size, labour and gender were not significant variables affecting willingness to participate in the proposed irrigation scheme. Based on the results from the logit regression model, it can be concluded that membership in a local group, investment in agriculture, education, knowledge and total household income are responsible for increasing the probability of participation. The negative relationship on age and participation indicates that the older the person is, the lesser the chances of participation. It is recommended that stakeholders should improve access to education and farmer training programmes. Support systems to disseminate information, training and knowledge should be enhanced. Improving institutional support and access to financial services should be prioritised by the stakeholders in the study area. Further studies on building institutional capacity and a cost benefit analysis of irrigation options in the study area are recommended.

Gasification characteristics of sugarcane bagasse

- Description: Sugarcane is a major crop in many countries. It is the most abundant lignocellulosic material in tropical countries such as South Africa. It is one of the plants with the highest bioconversion efficiency. The sugarcane crop is able to efficiently fix solar energy, yielding some 55 tons of dry matter per hectare of land annually. After harvest, the crop produces sugar juice and bagasse. Sugarcane bagasse is a residue that results from the crushing of sugarcane in the sugar industry. It is a renewable feedstock that can be used for power generation and manufacturing cellulosic ethanol. As biomass, sugarcane bagasse holds promise as a fuel source since it can produce more than enough electricity and heat energy to supply the needs of a common sugar factory. However, in the sugarcane industry the bagasse is currently burnt inefficiently in boilers that provide the heating for the industry. This project seeks to investigate the possibility of gasifying sugarcane bagasse as an efficient conversion technology. The investigation is necessary because fuel properties govern the gasifier design and ultimately, the gasification efficiency. Proximate and ultimate analysis of sugarcane bagasse was conducted after which the results were used to conduct a computer simulation of the mass and energy balance during gasification. The kinetic investigation undertaken through the TGA and DTG analyses revealed the activation energy and pre – exponential factor which were obtained by the model – free Kissinger method of kinetic analysis and were found to be 181.51 kJ/mol and 3.1 × 103/min respectively. The heating value of sugarcane bagasse was also measured and found to be 17.8 MJ/kg, which was used in the calculation of the conversion efficiency of the gasification process. Fuel properties, including moisture content and gasifier operating parameters were varied in order to determine optimum gasifier operating conditions that results in maximum conversion efficiency. The highest conversion efficiency was achieved at low moisture content after computer simulation of the gasification process. Moisture content also affected the volume of CO and H2 as the former decreases with increasing moisture content while the latter increases with increasing moisture content, accelerating the water – gas reaction. Scanning electron microscope fitted to an Energy dispersive X – ray spectroscopy was also used in order to view the shape and size distribution as well as determine the elemental composition of sugarcane bagasse. The results obtained established that the fuel properties and gasification conditions affect the conversion efficiency. During computer simulation, it was established that smaller particle size resulted in higher conversion efficiency. The smaller throat diameter also resulted in higher conversion efficiency. The throat angle of 25° also resulted in higher conversion efficiency. The temperature of input air was also found to be one of the major determining factors in terms of conversion efficiency. The dissertation presents the proximate and ultimate analysis results as well as the kinetic analysis results. The SEM/EDX analysis as well as the computer simulation results of the gasification process is also presented. The major contribution of this project was on the investigation of the gasification characteristics of sugarcane bagasse and the utilization of these in the design of a laboratory scale sugarcane bagasse gasifier with enhanced conversion efficiency through computer simulation.

- Description: Sugarcane is a major crop in many countries. It is the most abundant lignocellulosic material in tropical countries such as South Africa. It is one of the plants with the highest bioconversion efficiency. The sugarcane crop is able to efficiently fix solar energy, yielding some 55 tons of dry matter per hectare of land annually. After harvest, the crop produces sugar juice and bagasse. Sugarcane bagasse is a residue that results from the crushing of sugarcane in the sugar industry. It is a renewable feedstock that can be used for power generation and manufacturing cellulosic ethanol. As biomass, sugarcane bagasse holds promise as a fuel source since it can produce more than enough electricity and heat energy to supply the needs of a common sugar factory. However, in the sugarcane industry the bagasse is currently burnt inefficiently in boilers that provide the heating for the industry. This project seeks to investigate the possibility of gasifying sugarcane bagasse as an efficient conversion technology. The investigation is necessary because fuel properties govern the gasifier design and ultimately, the gasification efficiency. Proximate and ultimate analysis of sugarcane bagasse was conducted after which the results were used to conduct a computer simulation of the mass and energy balance during gasification. The kinetic investigation undertaken through the TGA and DTG analyses revealed the activation energy and pre – exponential factor which were obtained by the model – free Kissinger method of kinetic analysis and were found to be 181.51 kJ/mol and 3.1 × 103/min respectively. The heating value of sugarcane bagasse was also measured and found to be 17.8 MJ/kg, which was used in the calculation of the conversion efficiency of the gasification process. - Description: The contamination of heavy metals in the environment is a looming concern worldwide. Egeria densa (Planch) (Submerged aquatic plant) from two ponds: Site A with co-ordinates (32º 48’22.04”S; 26°48’58.79” E) and Site B with co-ordinates (32°48’33.25”S; 26°48’33.25”S) in Alice (Eastern Cape) was evaluated for its ability to absorb heavy metals, phytochemical constituents, antimicrobial activity and ultra-structure using standard analytic procedures. Cadmium (Cd), copper (Cu), iron (Fe), lead (Pb), manganese (Mn), and zinc (Zn) were measured in water, sediments and plant. The concentrations of these metal elements were determined with use of Inductively Coupled Plasma- Optical Emission Spectrometry (ICP-OES). In sediments, the heavy metals (mg/kg) decreased in the order of their average concentration as follows: Fe (40.320) > Zn (1.259) > Pb (0.564) > Mn (0.186) > Cu (0.037) in Pond 1 whereas in Pond 2 Fe (61.527) > Cd (0.999) > Mn (0.648) > Pb (0.586) > Zn (0.156) > Cu (0.045). The highest concentration of Fe was detected in both sites and Cu being the least. The concentrations of the metals in the plants sample (from Pond 1) were found in order of Mn > Pb > Cu > Fe whereas cadmium and zinc were not detected, while the concentration in Pond 2 decreases in order of Zn > Mn > Pb > Cd > Fe > Cu. In the water samples, concentrations of heavy metals (mg/L) decreased in the order of their average concentrations as follows: Pb (35.36) > Fe (3.07) > Mn (0.238) > Cu (0.104), both cadmium and zinc were below the limit of detection in Pond 1, whereas in Pond 2 the concentrations decreased as follows: Pb (13.033) >Fe (1.69) > Cu (0.270) > Mn (0.248) > Cd (0.004) and Zinc was not detected. Phytochemical analyses of the plant extracts revealed the presence of phenols, flavonoids, proanthocyanidin, flavonols, saponins, alkaloid and tannins in all the extracts (water, acetone and n-hexane). Both acetone and water extracts, showed high concentration of proanthocyanidin, while tannin was the lowest in acetone extract. Antimicrobial evaluation using, Gram positive (Staphylococcus aureus, Bacillus pumilus, Bacillus cereus, Streptococcus pyogenes, Enterococcus faecalis) and Gram negative (Klebsiella pneumonia, Escherichia coli, Pseudomonas aeruginosa, Proteus vulgaris and Serratia marcescens) bacteria showed negative results for all the strain, except Streptococcus pyogenes which was inhibited at MIC of 0.1 mg/ml. Scanning electron microscopy (SEM) of ultra-structure of Egeria densa, showed that certain bacteria attached to the leaf, However more work has to be done on E. densa to verify the mechanism by which it accumulates heavy metals. The study shows that E. densa has a potential of accumulating heavy metals especial Manganese in plant.

Impact of human dimensions on smallholder farming in the Eastern Cape Province of South Africa

- Description: Considering the backward and forward linkages, the agro-industrial sector contributes about 12 percent of South Africa‘s GDP, and employs approximately 8.5 million people. In the Eastern Cape Province, the sector contributes about 1.9 percent of the Provincial GDP, and over 3 million people derive their livelihoods from subsistence smallholder farming. Despite its importance, agricultural productivity has stagnated for several years across the Eastern Cape rural communities. There have been several attempts by the government to improve the agricultural productivity on smallholder farms since the end of apartheid, especially through the establishment of small-scale irrigation schemes, subsidization of farm inputs, and provision of credit facilities and enacting a number of land reform policies. In spite of the government support, most rural communities like Qamata and Tyefu are still faced with high levels of poverty affecting 76 percent and 91 percent of the population, respectively. This research evaluated the current smallholders‘ production efficiency, and the link between smallholder farmers‘ human dimensions (entrepreneurial spirit and positive psychological capital, goals and social capital, and other efficiency related variables) with production efficiency and household commercialisation index/level. The study used participatory approaches for site selection, sample selection and data collection. The analysis was based on both information from informal interviews and formal primary data collection. The Data Envelopment Analysis and Stochastic Production Frontier techniques were used to determine the relative efficiencies of individual farmers and to identify the major factors that influence the efficiency of production. Overall, 158 farmers were interviewed both at Qamata and Tyefu irrigation schemes. Descriptive statistics of this study indicated that most of the farmers were men with an average age of 61 years, and mean household size of 4 persons with the household head having at least obtained some primary school education. Farming is the major source of livelihood for smallholders with an average income of R4527.49 per crop season. Smallholders use improved seeds, fertilizers and tractor for ploughing with less use of pesticides and herbicides. Although smallholder irrigators generate more gross margins from maize and cabbage enterprises, generally both categories of farmers exhibited a low average household commercialization index for maize and cabbage at 0.41 and 0.22, respectively. Both Data Envelopment Analysis and Stochastic Production Frontier results indicate that farmers are about 98 percent technically efficient in maize and cabbage enterprises, respectively. However, farmers were allocatively inefficient as they were under-utilizing seed and pesticides while over-utilizing inorganic fertilizers. Factors that are positively associated with technical efficiency in maize production included household size, farming experience, off-farm income, use of agro-chemical; gross margins and commercialisation level of maize output. Determinants of technical efficiency in cabbage enterprise included farming experience, amount of land owned, use of agro-chemicals, group membership and gross margins accrued to cabbage sales. Farmers‘ human dimensions that could be more positively and significantly associated with production, efficiency and household commercialisation level included risk taking (hope), innovativeness (confidence) and optimism for entrepreneurial/positive psychological capital. Farmers‘ goals included self-esteem and independence, and only external social capital which were identified to be more positively and significantly associated with farmers‘ production efficiency and commercialization level. The transition from homestead subsistence to commercial oriented small-scale irrigation farming is inevitable since smallholder irrigators earn more incomes from maize and cabbage and are relatively food secure. However, the key policy options that must be considered to address inefficiencies and improved commercialization level to aid the transition include: agricultural policies geared toward attracting youth in farming, improved quality of extension services, speeding up the land reform process, and formation of cooperatives and participatory policy formulation that takes full cognizance of the farmers‘ human dimensions. Since farmers‘ human dimensions as defined in the literature and this study are not things that are amenable to direct policy intervention, they can only be modified indirectly through policy actions that affect their determinants. This means that a number of the demographic and socio-economic characteristics such as age, sex and education level of household head, farming experience, size of land owned, crop incomes, source of water for irrigation and location of the irrigation scheme that govern the way people perceive reality and respond to them must be the focus of concerted policy actions over the medium to long term.

Incidence of Staphylococcus species in bovine milk: their antimicrobial sensitivity in selected antibiotics and Usnea barbata lichen extracts

- Description: This study was done in order to assess the incidence of Staphylococcus species from milk of cows with subclinical mastitis and their antimicrobial sensitivity in some selected antibiotics and Usnea barbata lichen extracts. The study was conducted in two different commercial dairy farms. Staphylococcus species isolates were identified using several biochemical tests which included Gram’s staining test, catalase test and oxidase test. A commercial API® staph kit (bioMerieux, France) was used to confirm the bacterial organisms to their species level. The antimicrobial sensitivity of individual species was determined according to the Clinical Laboratory Standard Institute (CLSI) for the selected antibiotics. Agar well diffusion method and the broth micro-dilution technique were used to determine the sensitivity of Staphylococcus species in U. barbata extracts. A total of 467 milk samples were screened for bacterial identification from the two farms. Fifteen different Staphylococcus species isolates were identified from all milk samples that were examined. The most frequently isolated species included Staphylococcus xylosus (54.34%), Staphylococcus hominis (24.78%), Staphylococcus aureus (16.38%), Staphylococcus saprophyticus (16.12%) and Staphylococcus haemolyticus (11.63%). Most Staphylococcus species were resistant to Penicillin (75.35%), Nalidixic acid (72.55%) and Ampicillin (63%). Furthermore, the bacterial sensitivity evaluation of U. barbata lichen extracted with methanol and ethyl-acetate against selected Staphylococcus species isolates showed 92.31% and 53.85% susceptibility, respectively. The minimum inhibitory concentration (MIC) of the methanol and ethyl-acetate extracts ranged between 0.0390 to 10 mg/ml. There was a relatively high incidence of Staphylococcus species identified in milk of cows with subclinical mastitis from both farms. Conversely, Staphylococcus species isolates were resistant to antibiotics (mostly penicillin and ampicillin) commonly used in the farms. Furthermore, the study investigated the antimicrobial sensitivity of U. barbata extract in-vitro which may validate its use in traditional medicine for treatment of cows with mastitis.

- Description: This study was done in order to assess the incidence of Staphylococcus species from milk of cows with subclinical mastitis and their antimicrobial sensitivity in some selected antibiotics and Usnea barbata lichen extracts. The study was conducted in two different commercial dairy farms. Staphylococcus species isolates were identified using several biochemical tests which included Gram’s staining test, catalase test and oxidase test. A commercial API® staph kit (bioMerieux, France) was used to confirm the bacterial organisms to their species level. The antimicrobial sensitivity of individual species was determined according to the Clinical Laboratory Standard Institute (CLSI) for the selected antibiotics. Agar well diffusion method and the broth micro-dilution technique were used to determine the sensitivity of Staphylococcus species in U. barbata extracts. A total of 467 milk samples were screened for bacterial identification from the two farms. Fifteen different Staphylococcus species isolates were identified from all milk samples that were examined. The most frequently isolated species included Staphylococcus xylosus (54.34%), Staphylococcus hominis (24.78%), Staphylococcus aureus (16.38%), Staphylococcus saprophyticus (16.12%) and Staphylococcus haemolyticus (11.63%). Most Staphylococcus species were resistant to Penicillin (75.35%), Nalidixic acid (72.55%) and Ampicillin (63%). Furthermore, the bacterial sensitivity evaluation of U. barbata lichen extracted with methanol and ethyl-acetate against selected Staphylococcus species isolates showed 92.31% and 53.85% susceptibility, respectively. The minimum inhibitory concentration (MIC) of the methanol and ethyl-acetate extracts ranged between 0.0390 to 10 mg/ml. There was a relatively high incidence of Staphylococcus species identified in milk of cows with subclinical mastitis from both farms. Conversely, Staphylococcus species isolates were resistant to antibiotics (mostly penicillin and ampicillin) commonly used in the farms. Furthermore, the study investigated the antimicrobial sensitivity of U. barbata extract in-vitro which may validate its use in traditional medicine for treatment of cows with mastitis.

Given that sensitivity and adaptive capacity of farming systems to climate change is shaped by both socioeconomic and institutional factors, a multiple regression model was used to test the relationship between indicators of vulnerability and household socioeconomic and institutional characteristics. Indicators were selected based on significant statistical relationships. This means that the statistical procedure for selecting indicators involved relating a large number of variables to vulnerability in order to identify statistically significant factors. The results showed reliability of income and reliability of water resources to be good indicators of vulnerability. Many statistically significant variables as well as respective R2 of 0.988 and 0.825 confirm the foregoing. Another indicator was the Simpson index that measures diversification of agricultural production. The results show that vulnerability to climate change was highest for the households near the river and lowest for the inland village. Moreover, the results confirmed that most blacks that are practicing agriculture receive little if any support largely because available resources are highly skewed towards certain farmers rather than others.

Furthermore, putative plant PEPs showed similar secondary structures to known PEPs and when a rice PEP was modelled onto porcine brain PEP structure, a high degree of similarity was found. Germination studies of wheat seed showed an increase of PEP activity over time with maximum PEP activity reached after 4 days of germination, which remained at this level until 9 days of germination, implying a function for PEP in plant seed germination. Wheat PEP was purified using ion exchange and gel filtration chromatography with a final yield of less than 1 percent and a relative purity (only 2 bands detected by SDS-PAGE). The purified wheat PEP had a molecular weight of approximately 55kDa, substrate specificity for chymotrypsin-like substrates (N-Suc-Ala-Ala-Pro-Phe-pNa, Km value of 0.58 mM, Kcat of 29.37 s–1; Kcat /Km 50813.14s–1 M–1); a pH optimum of 7.9; temperature optima of 37oC and a high sensitivity to temperature as indicated by loss of activity at temperatures above 40oC. Inhibition studies using E64, Leupeptin and PMSF confirmed that the wheat PEP is from the Serine protease family and is most likely a trypsin-like protease.

- Description: The purpose of this study was to explore and to describe experiences of learner nurses regarding clinical supervision at private hospitals in East London in the Eastern Cape. These were Life Healthcare hospitals. A qualitative, contextual approach was used and the exploratory, descriptive design was seen as most appropriate in this study. The subjects were the learner nurses in the Bridging Course Programme leading to registration as a General Nurse. The research questions were: How did you as a learner nurse, doing the Bridging Course, experience clinical supervision at Life Healthcare hospitals? A pilot study was conducted in a clinical setting involving respondents with similar characteristics to the study population. Fourteen participants were selected for this study by using a purposive, non-probability sampling method. Data were collected by means of individual interviews during which participants were able to describe their experiences during clinical supervision; data were collected until data saturation was reached.During data analysis, the researcher used the process of bracketing and remained neutral, setting aside previous knowledge and beliefs about the phenomenon under investigation. The researcher listened to the audiotapes used for data collection several times until she was completely satisfied with the interpretation of the verbatim data. Transcriptions were made within three days of the interview. The research was done in an ethically reflective manner and the researcher ensured the trustworthiness of the study at all times. - Description: To proffer a sustainable solution to ecological degradation in rangeland ecosystems as a consequence of fire, grazing and browsing, an understanding of rangeland ecological processes is vital. Due to the complexity of ecological processes and their interrelationships, it is usually difficult or expensive to directly measure status of ecological processes. Therefore, biological and physical characteristics are often used to indicate the functionality of ecological processes and site integrity. Long-term effects of fire, grazing and browsing on characteristics of the vegetation and organic matter and their subsequent effects on selected rangelands ecosystem ecological processes was conducted at Honeydale section of the University of Fort Hare farm in the Eastern Cape Province of South Africa and Matopos Research Station in Zimbabwe. In this study, attributes of biotic community integrity (species richness, composition and diversity), soil stability (basal cover, standing dead grass biomass, tuft to tuft distance, tufts diameter, canopy distance and stem to stem distance), productivity and plant vigour (grass yield, total canopy volume, plant height, canopy height, canopy diameter, main stem diameter, sprouts diameter and number of sprouts) and hydrologic function and nutrient cycling (grass litter biomass, soil organic carbon and microbial biomass carbon) were used to estimate long-term effects of burning, grazing and browsing by goats on the functionality of ecological processes in the rangeland ecosystem. Burning did not have differential effect on grass species richness (P>0.05), woody species diversity as well as compositional percentage for D.eriatha, C.plurinodis, S.fimbriatus, A.karro and E.rigida. Burning increased decreasers and increaser II species proportions and reduced (P ≤ 0.05) grass yield, total canopy volume, tree height, canopy height main stem diameter and sprouts diameter. Long-term burning, grazing, and goats browsing had differential effects on site stability. The effects on basal cover, tuft to tuft distance, tufts diameter, canopy distance and basal distance as a consequence of long-term burning, grazing, and goats browsing were not significantly different, whereas the effects on standing dead grass biomass as a result of long-term burning frequencies were significantly different. Long-term effects of burning followed by ten-year period of fire exclusion had significantly different effects on tuft-tuft distance but did not have statistically different effects on tufts diameter, canopy distance and basal distance. Long-term burning grazing and browsing had significantly different effects on attributes of hydrologic functions and nutrient cycling in the rangeland ecosystem (grass litter biomass, SOC and BMC). Long-term effects of burning followed by ten-year period of fire exclusion had significantly different effects on grass litter biomass, and SOC. Through their effect on vegetation and organic matter characteristics, burning, grazing and browsing could influence functionality of selected rangeland ecological processes such as biological community integrity, productivity and plant vigour, site stability, hydrologic function and nutrient cycling.

Long-term effects of burning followed by ten-year period of fire exclusion had significantly different effects on tuft-tuft distance but did not have statistically different effects on tufts diameter, canopy distance and basal distance. Long-term burning grazing and browsing had significantly different effects on attributes of hydrologic functions and nutrient cycling in the rangeland ecosystem (grass litter biomass, SOC and BMC). Long-term effects of burning followed by ten-year period of fire exclusion had significantly different effects on grass litter biomass, and SOC. Through their effect on vegetation and organic matter characteristics, burning, grazing and browsing could influence functionality of selected rangeland ecological processes such as biological community integrity, productivity and plant vigour, site stability, hydrologic function and nutrient cycling.

- Description: The Tuberculosis infection rate has been generally escalating due to poor health conditions in the Gweru district of Zimbabwe. The study therefore seeks to identify the risk factors that affect TB treatment in the Gweru district. A cross sectional study was carried out in which a questionnaire was employed for data collection on 113 respondents. A binary logistic regression model was employed for data analysis. A total of 98 TB patients were interviewed: [50 respondents (44.0%) had Multi-drug resistant Tuberculosis and 63 respondents (56.0%) had general Tuberculosis). Before being enrolled into the study, an informed consent form was given to each of the participants. The data was then put into excel and later transferred to SPSS for analysis. Out of the 14 potential risk factors of TB treatment, only 6 variables (side effects, gender, alcohol use, HIV status, smoking during the treatment period and having been pre-exposed to TB drugs) were statistically significant in their association with treatment failure.

- Description: Tuberculosis (TB) is an ancient infectious disease that has been infecting different populations around the globe and it has also been considered as one of the most successful human and animal disease. TB found in animals such as cattle and other known bovids is known as bovine tuberculosis. Bovine tuberculosis (BTB) is an infectious disease found in cattle mainly caused by Mycobacterium bovis. M. bovis is a member of the Mycobacterium tuberculosis complex (MTC) together with M. tuberculosis, M. africanum, and M. canetti where the natural host is humans; whereas M. caprae, M. microti and M. pinnipedii usually have animals as their natural host. In this study the molecular characterization of the MTC from cow milk in the Eastern Cape was investigated. One hundred and twenty samples (40 ml each) were collected from three dairy farms in the Eastern Cape, South Africa. These samples were processed using a modified Petroff decontamination method. Sample processing was followed by DNA isolation using a Zymo Bacterial/Fungal DNA Kit and the amplification and detection of the MTC was done using the Seeplex MTB Nested ACE assay. The drug susceptibility tests were done using GenoTypeMTBDRplus assay which detects mutations and resistance to INH (isoniazid) and RMP (rifampicin). The milk isolates were further analyzed using a spoligotyping method which is based on the PCR amplification of a highly polymorphic direct repeat locus in the M. tuberculosis genome which detects and types the MTC. A percentage of 20.8 % samples were found to be positive for MTC using the Seeplex MTB Nested ACE assay. There were 42.1 % samples that were resistant to both INH and RMP with the rest sensitive to either INH or RMP. The spoligotyping method showed that 78.3 % samples resembled Family 33 strains and the rest (21.7 %) resembled a spoligotyping signature known to be that of M.africanum. Both these strains belong to the Ancestral lineage with Indo-Oceanic and West Africa 2 lineage. The outcomes of our study showed that molecular methods for detection of MTC can be applied directly on milk samples without the need for culturing.

Morphological and genetic variation in samango monkeys (Cercopithecus albogularis) in Southern Africa

- Description: My aim was to resolve the taxonomy of the South African forms of Cercopithecus albogularis by exploring morphological and genetic variation in the two samango subspecies described for the region: C. a. erythrarchus and C. a. labiatus. In addition, I estimated their geographic distributions and habitat requirements from the provenance data I collected during my study of museum specimens. My analysis has shown clear morphological differences between C. a. labiatus and C. a. erythrarchus. The two subspecies differ in pelage coloration, but also in cranial shape and tail lengths. Furthermore, C. a. labiatus is slightly smaller, shorter-tailed and stocky compared with the long-tailed slender northern forms of C. albogularis, which may be adaptations to cold environments like montane forest. The northern C. albogularis subspecies are distinguishable from the southern taxa with a high degree of reliability (98.5%), suggesting that C. albogularis consists of more than one species. The most appropriate name for the southern species is C. labiatus. My genetic study did not detect variation among the 10 animals sampled in Hogsback; while it clustered the different taxa, it could not resolve relationships between them, with the exception of the outgroup. The lack of resolution of the deeper nodes could be a result of the fact that our sequence was very short (274 bp). The mt 12S rRNA gene was not an ideal gene for this study, which should have involved a less conserved section of the mtDNA molecule, like the rapidly evolving D-loop. More genetic work is clearly needed to resolve the phylogenetic relationships within the C. mitis supergroup. However, preliminary genetic data indicate that the southern samangos are distinct from the C. mitis of West Africa, while my morphometric study suggests they may also be distinct from C. albogularis in East Africa. Molecular and karyological studies comparing the genomes of Hogsback “C. a. labiatus” with the neighbouring “C. a. erythrarchus” and Zanzibar C. albogularis would be extremely enlightening.
