- Description: The actinomycetes are gram-positive bacteria that produce more than two-thirds of the known biologically active microbial natural products, including many commercially important antibiotics, anti-cancer agents, other pharmacologically useful agents, animal health products and agrochemicals. The prevailing utilization of antibiotics continues to be the mainstay against microbial infections and a majority ofthe over six thousand antibiotics discovered thus far are from Streptomyces spp. One of the most well-characterized antibiotic biosynthetic pathway is the one involving the biosynthesis of the penicillins, cephalosporins and cephamycins. This pathway involves two initial steps which are common in filamentous fungi, lower eukaryotes and prokaryotes. The penam nucleus of penicillins and the cephem nucleus of both cephamycins andcephalosporins are formed by the condensation of the three precursor amino acids L-a-aminoadipic acid, Lcysteine and L-valine by a mechanism designated as 'non-ribosomal peptide synthesis', which involves activation and condensation of the three component amino acids and epimerization of the L- to D-valine to form a linear acyclic tripeptide called o-(L-a-aminoadipyl)-L-cysteinyl-Dvaline (ACV) by the action of a peptide synthetase. ACV is then cyclized to form isopenicillin N, an intermediate that contains an L-a-aminoadipyl side-chain attached to the penem nucleus (Fig. 1.2) by isopenicilin N synthase (IPNS or Cyclase) and this encompasses the creation of the Beta-lactam and thiazolidine rings. A broad range of ~-lactam producing Streptomyces spp were grown, the DNA extraction procedure optimised and total chromosomal DNA isolated. A bioinformatics analysis of known IPNS gene sequences allowed the synthesis of PCR primers for the iso-penicillin N synthase gene. IPNS genes and lPNS-like genes were successfully amplified from the total DNA of ten strains including two novel thermophilic strains, A. and B. Sequencing was carried out on the genes from S. hygroscopicus, S. tanashiensis and the two thermophiles A and B. This allowed development of the conditions for gene shuffiing of the IPNS gene which was carried out pairwise and resulted in the reconstitution of shuffied genes of the correct size. The resulting mixed gene sequences were cloned into the pTrcHis2-TOPO expression vector and the plasmid DNA screened and assayed for IPNS activity using HPLC which showed ten fold increase in IPNS activity as a result of the shuffiing.

- Description: Whole Effluent Toxicity (WET) testing has been identified as one of the tools in the management of complex effluents in aquatic ecosystems. In South Africa, toxicity testing has not been required for regulatory purposes. Recently, the Department of Water Affairs and Forestry has adopted WET testing as a tool to evaluate the suitability of hazardous effluent for discharge into receiving environments. This has necessitated suitable procedures to be established for use in the South African situation. With the implementation of the new National Water Act (No 36 of 1998), industries have to comply with set standards to protect the aquatic environment. However, the South African Water Quality Guidelines for Aquatic Ecosystems have been set using international toxicity data, and it is not known if they are comparable with South African conditions. The aim of this study was to investigate the tolerances of selected indigenous riverine invertebrates to complex saline effluents. The study investigated the effects of kraft mill effluent to Tricorythus tinctus, a tricorythid mayfly from the Sabie River, Mpumalanga, and the effects of a textile effluent to baetid mayflies of the Buffalo River, Eastern Cape. Indigenous riverine invertebrates were chosen as test organisms, as there is no toxicity data in South Africa which could be used to evaluate the level of protection afforded by the South African Water Quality Guidelines for Aquatic Ecosystems. The use of indigenous riverine invertebrates added the challenge of variability of a wild population, and the use of a complex effluent as toxicant added the variability of effluent composition. In this study, WET testing was used to determine the dilution of whole effluents required for discharge. Hazard-based guidelines were developed for the disposal of kraft and textile effluents. The level of environmental hazard posed by different effluent concentrations was ranked, and was related to the River Health Class. This indicated effluent concentrations that may be allowed to enter the aquatic environment, e.g. 3% effluent concentration guideline for both general kraft effluent and general textile effluent for the protection of a Class A river. This approach could contribute to the use of an Environmental Risk Assessment, approach for the management of complex effluents. A number of acute 96 hour toxicity tests were conducted following an unreplicated regression design, using kraft and textile effluents as toxicants, mayfly nymphs as test organisms, and river water as diluent and control. Test organisms were sampled from unimpacted, flowing-water riffle areas, and were exposed in recirculating artificial streams (or channels) to a range of effluent concentrations. Mortality was selected as end-point and observed twice daily. The experimental results showed the variability and acute toxicity of both kraft and textile mill effluents. Baetids were more sensitive (mean LC50=16% effluent concentration) to General Textile Effluent (GTE), but less sensitive to Post Irrigation Textile Effluent (PITE). Textile effluent (PITE) held in a holding dam were therefore less variable and less toxic; suggesting that stabilization of the effluent could have contributed to reduced toxicity. Effluent composition, e.g. higher calcium levels, may also have contributed to lowering toxicity. T. tinctus was sensitive to kraft effluents, but showed less variable responses to Irrigation Kraft Effluent than General Kraft Effluent. Toxicity test data indicated that GKE, IKE and GTE should not enter the aquatic environment without treatment, as they can cause adverse effects to aquatic biota. Both kraft and textile effluents must therefore be treated before discharge. Different responses to different effluent batches were probably due to effluent variability. The use of indigenous organisms, and not a standard laboratory organism, could also have contributed to variability. A hazard-based approach could be useful, as it will provide a consistent basis for deciding on the acceptability of impacts, while allowing natural site-specific differences to be taken into account.

The role of a symbiotic bryozoan in the chemical ecology of a marine benthic predator-prey interaction

- Description: The subtidal whelk Burnupena papyracea (Brugière) co-occurs with a voracious predator, the rock lobster Jasus lalandii (Milne Edwards), in situations where other potential prey are largely eliminated. This has been ascribed to a symbiotic bryozoan, Alcyonidium nodosum (O’Donoghue and de Watteville), which characteristically encrusts the shells of B. papyracea and deters feeding by Jasus. In this study it is shown that this is not due to physical effects of either induced physical defences in the bryozoan or increased shell strength due to the presence of the bryozoan. Neither spectroscopic screening of chemical extracts of the bryozoan nor analysis for volatile constituents revealed any apparent chemical components that are likely to deter feeding. Chemical extracts also failed to show larvicidal effects in a standard toxicity assay using the brine shrimp Artemia salina (Leach). Despite this, bioassays using individual Jasus indicated a chemical basis for feeding deterrence. The assays were run separately on three sets of Jasus and some repeats of assays gave contradictory results. However, assays showing no significant effect of treatment occurred with moulting Jasus, involved very low overall feeding rates and so gave a less convincing result. In other assays Jasus always avoided Burnupena papyracea with live Alcyonidium encrusting the shell, and food pellets containing Alcyonidium or an Alcyonidium extract. Significant preferences were shown for an unencrusted whelk, B. cincta (Röding), over B. papyracea; for B. papyracea with the bryozoan scraped off over natural B. papyracea; for B. papyracea on which the bryozoans had been killed with liquid nitrogen over untreated B. papyracea; and for food pellets prepared from ground, dried mussel over pellets prepared with dried mussel mixed with A. nodosum or its crude organic extract. It is concluded that the protection which Alcyonidium confers on Burnupena papyracea does have a chemical basis, but that the chemical responsible is either present in only trace quantities, or that it is a structurally unremarkable compound which is distasteful to Jasus. This work highlights both the advantages of using ecologically relevant bioassays (positive results when standard techniques give a negative result) and also the disadvantages (logistic constraints on sample sizes when using large test animals and individual variability in a relatively sophisticated test animal).

The Rhodes BioSure process in the treatment of acid mine drainage wastewaters

Both biological and physico-chemical processes, set in train by mining operations, give rise to the oxidation of sulphur species, and the resultant generation of AMD. The Vaal River system is most affected and receives large tonnages of mining related salinity as both direct discharges, and in diffuse runoff flows. The long-term burden of this problem, and sustaining ongoing treatment over the time-frames involved will almost certainly resort to the community inhabiting the area, notwithstanding progressive mine closure legislation and comprehensive regulation governing the polluterpays principle. The volume and time-frame of the AMD problem, and the need for a long-term and sustainable response has focused interest in biological treatment approaches. These have concentrated on active and passive treatment systems, both of which rely on microbial activity related to the biological sulphur cycle. Notwithstanding the reactor type, and the particular treatment approach used, widespread application of active AMD treatment has not yet been seen on any large scale. Singular factors constraining process development are bioreactor design, cost of bioreactor construction, and the cost of the carbon source and electron donor for the biological sulphate reduction process. The SRB are able to utilise only a limited range of small organic molecules. The studies reported here were motivated by the need to evaluate low-cost options and the treatment of high volume AMD flows. This has focussed research activity on bioprocess developments using complex organic compounds derived from waste streams as electron donor sources, and the integration of AMD treatment with other waste treatment objectives. The co-disposal of organic wastes with AMD treatment would enable the development of an 'integrated resource management' approach to the problem, including sustainability of treatment operations over the long time-frames involved. The refining of calcium using a sulfate reducing bacterial system

- Description: White lime is used in many industries in South Africa but is not produced locally and must be imported. Many technologies have been suggested for the large-scale manufacture of calcium carbonate but these are not necessarily suitable for application in South Africa. This study investigated a chemical preparation of calcium carbonate combined with biological purification Calcium containing materials from the Pretoria Portland Cement, Lime Division factory at Lime Acres in the Northern Cape were studied as the starting materials for the manufacture. Investigation showed that they contained various impurities, including iron and manganese compounds which were largely responsible for the brown-grey colour of the lime products. Complete dissolution of calcium hydroxide, the purest of the potential starting materials, and subsequent hydroxide precipitation was not successful in removing all iron and manganese. Precipitation with sulfide ions was successfill, decreasing levels of metals to below the detection limit of atomic absorption spectrophotometry. Studies of all potential starting materials revealed that the levels of impurities in the starting material did not have a large effect on levels of impurities in the calcium carbonate produced. It was therefore possible to convert the residual calcium oxide or hydroxide in waste lime dusts to white calcium carbonate, a marketable prciduct Recycling of the water and starting material used in the process served to increase, rather than decrease, the purity of the calcium carbonate product. This allows for water conservation as water is not consumed in the process but merely utilised. When waste lime dust was used as the starting material, sulfate was found in the product. While still a white lime, the calcium carbonate was not chemically pure. Sulfate removal was therefore investigated and the use of sulfate-reducing bacteria was studied as a novel application. A mixed sulfate-reducing bacterial population was isolated and found to be hIghly active at sulfate concentrations between 0.2 and 2 ~~~. They were capable of autotrophic growth and could reduce sulfate in solutions with elevated pH and in calcium carbonate suspensions, although they did not grow readily in these media. A process was designed for the production of bulk quantities of calcium carbonate making use of the facilities and materials available at Lime Acres. This was tested using a small scale bench-top reactor series, with favourable results. The process would allow automatic, continuous production of large quantities of white lime using waste lime dust. Provision was also made for manufacture of smaller quantities of pure calcium carbonate using sulfate-reducing bacteria to remove the sulfate impurity.

Complete dissolution of calcium hydroxide, the purest of the potential starting materials, and subsequent hydroxide precipitation was not successful in removing all iron and manganese. Precipitation with sulfide ions was successfill, decreasing levels of metals to below the detection limit of atomic absorption spectrophotometry. Studies of all potential starting materials revealed that the levels of impurities in the starting material did not have a large effect on levels of impurities in the calcium carbonate produced. It was therefore possible to convert the residual calcium oxide or hydroxide in waste lime dusts to white calcium carbonate, a marketable prciduct Recycling of the water and starting material used in the process served to increase, rather than decrease, the purity of the calcium carbonate product. This allows for water conservation as water is not consumed in the process but merely utilised. When waste lime dust was used as the starting material, sulfate was found in the product. While still a white lime, the calcium carbonate was not chemically pure. Sulfate removal was therefore investigated and the use of sulfate-reducing bacteria was studied as a novel application. A mixed sulfate-reducing bacterial population was isolated and found to be hIghly active at sulfate concentrations between 0.2 and 2 ~~~. They were capable of autotrophic growth and could reduce sulfate in solutions with elevated pH and in calcium carbonate suspensions, although they did not grow readily in these media. A process was designed for the production of bulk quantities of calcium carbonate making use of the facilities and materials available at Lime Acres. This was tested using a small scale bench-top reactor series, with favourable results. The process would allow automatic, continuous production of large quantities of white lime using waste lime dust. Provision was also made for manufacture of smaller quantities of pure calcium carbonate using sulfate-reducing bacteria to remove the sulfate impurity.

I investigated the mandibular gland profiles and ovarial development of queenright workers from 4 localities. Their queens were then removed to determine how these aspects changed upon queen loss. In addition, drifted bees were analysed in the same way to determine how these factors changed once they had gained entry to a foreign hive. The queenright bees, form all 4 localities were found to have 9HDA as the most abundant of the 5 fatty acids measured and all localities had small percentages of 9ODA in their mandibular gland secretions. This resulted in relatively high queenright 9ODA:10HDA and 9HDA:10HDAA ratios. Despite this the percentage of bees with undeveloped ovaries was consistent with their queenright status. The mean values of these 2 ratios were significantly higher in the bees from East London and Cradock than those from Port Elizabeth. Steynsburg's bees were intermediate in this regard. Upon queen loss, the bees from all 4 localities had an increase in the percentage of 9ODA but the other compound changes in varying ways. East London's bees were the only ones not to become significantly more queen-like after queen loss. After 14 days without a queen, the mean values of these 2 ratios were much higher in the bees from Port Elizabeth than those from the other localities. Certain individuals from Port Elizabeth had values of these ratios that exceeded those found in A. m. capensis queens. Port Elizabeth was the only locality to display any surrogate queens and exhibited the highest increase in the number of bees with partially or fully developed ovaries. - Description: Midazolam, used clinically as an anticonvulsant, anxiolytic, muscle relaxant and sedative is a photolabile imidazo-benzodiazepine which is marketed under the trade names Dormicum® and Hypnovel® as tablets and injectables. Because of an aqueous solubility of < 0.1 mg/ml above pH 4, the preparation of aqueous dosage formulations near physiological pH, requires a solubilizer. The aim of this study was thus to prepare a 10 mg/ml midazolam aqueous solution for topical application using randomly-methylated-pcyclodextrin (RAMEB), considered to be a suitable candidate as a solubilizer because of its absorption enhancing properties, and to investigate its effect on both the solubility and the photostability of midazolam. Solubility studies of midazolam (excess of 15 mg/ml) in the presence of 0, 5,10,20, 30% m/v of RAMEB at pH 5.0 and pH 5.8 (phosphate buffer) were undertaken and the results analyzed using a UV method validated for linearity, accuracy, precision and specificity. A stability-indicating HPLC method was developed and validated (precision and accuracy, linearity, range, limit of quantitation, specificity, robustness and ruggedness) for application to kinetic photostability studies and the identification of photodegradants by LC-MS. Forced degradation studies were carried out at concentrations of 0.5 mg/ml of midazolam instead of the target concentration of 10 mg/ml because of the acceleratory effect of the decreased concentration on the rate of photodegradation. The solutions of midazolam with and without RAMEB were irradiated at 550 W/m² for 12 hrs in order to degrade the drug to ± 10% of the original concentration. The UV method proved to be valid in terms of linearity with a correlation coefficient of 0.9998, precise and accurate, and specific for the determination of midazolam in the presence of RAMEB. The results of the phase solubility studies indicated that desired solubility of 10 mg/ml was achieved with 30% m/v RAMEB at pH 5.0. RAMEB slightly decreased the photostability of midazolam, the rate constants being 0.137 and 0.154 hr⁻¹ in the absence and presence of RAMEB, respectively. LC-MS analysis revealed that one of the major photoproducts in the presence and absence of RAMEB was N-desalkylflurazepam, a starting material in the synthesis of midazolam. RAMEB inhibited formation of some photoproducts and introduced two new photoproducts, a dimer and an addition product. The difference in the nature of these photoproducts formed may be attributed to the ability of RAMEB to provide conformational control and to stabilize free radicals. Although RAMEB improved the solubility of midazolam to the target concentration, photostability is decreased with the presence of different photoproducts. - Description: Melatonin is a product of the pineal gland and is postulated to play an antigonadotropic role in the reproductive system of mammals. The reproductive system of non-seasonally breeding mammals is believed to be not as responsive to melatonin treatment as that of seasonally breeding mammals. Recently, there has been increasing support from in vivo and in vitro studies, for the hypothesis that melatonin has negative effects on sperm physiology, especially on sperm motility. High and/or low seminal concentrations of melatonin have been associated with abnormalities in human sperm motility and concentration. In this study, I examined the effects of melatonin on the testis, epididymis and sperm physiology, using in vivo and in vitro experiments, in a non-seasonally breeding mammal. Treatment, in vivo, with exogenous melatonin for six weeks did not inhibit testosterone production or spermatogenesis, nor did it affect the mass of the testes and epididymides at dissection, the concentration the morphology of speimatozoa. However, melatonin in vivo had a small, but significant negative effect on sperm motility and sperm motility index. In vitro incubation of spermatozoa Fith melatonin reduced the percentage (%) of forward progressive movement (fpm), increased the % reduction in fpm, reduced the vigor or quality of sperm motility, reduced the sperm motility index, and delayed and/or prolonged the transition of one pattern of sperm motility to the subsequent patterns. Melatonin increased the pH of the culture medium, and the increased pH, and the ethanol utilized as a solvent for melatonin, both negatively affected all the sperm motility parameters that were assessed in my study. The effects of ethanol increased with time, and the effects of pH increased with both time and increasing pH. Melatonin in vitro did not inhibit capacitation and the acrosome reaction, but it delayed the onset and the progression of capacitation and the acrosome reaction. These results suggest that while melatonin did not inhibit spermatogenesis in the Wistar rat, it may influence sperm motility. Therefore, the presence of high concentrations of melatonin in the reproductive fluids may inhibit sperm motility. With further detailed research, melatonin may have a potential use as a contraceptive drug.

- Description: The use of controlled release dosage forms has increased significantly in recent years as they result in increased patient compliance and higher therapeutic efficiency. This research focused on the development of a once daily dosage form that could be used for the treatment of hypertension. Both a separate sustained release dosage of metoprolol tartrate and a combination dosage form that included both an immediate release hydrochlorothiazide and a sustained release metoprolol component, were developed and evaluated. A matrix tablet, consisting of an ethylcellulose ranulation of metoprolol tartrate compressed into a hydrophilic hydroxypropyl methylcellulose polymer matrix, effectively sustained metoprolol release over a 22-hour experimental period. A multiparticulate combination dosage form that consisted of six coated mini matrix tablets of metoprolol and a powder blend of hydrochlorothiazide packed into a gelatin capsule, displayed zero order release kinetics for metoprolol release over 22 hours (r2=0.9946). The release of hydrochlorothiazide was found to be comparable to that of a commercially available product tested. Differential Scanning Calorimetry was used to identify possible incompatibilities between MPTA and excipients initially, and long term stability testing was used to assess to behaviour of the dosage form. Dissolution testing of the dosage forms was performed using USP Apparatus III, which was found to be more discriminating between the batches assessed. Dissolution curves were evaluated for similarity and difference using f1 and f2 fit factors. Samples were analyzed using a high performance liquid chromatographic method that was developed and validated for the simultaneous determination of the compounds of interest. Various factors influencing drug release from the developed dosage forms were assessed and recommendations for further optimization of the formulation are made. Factors evaluated included the quantity of granulating fluid, matrix polymer content, drug load and process variables, including drying time and compression force. The influence of various coating levels on drug release was assessed and none of the levels assessed were found to adequately retarded drug release over a 22-hour period. Combinations of tablets coated to different levels allowed for the successful development of a sustained release metoprolol component, which could be included into the combination dosage form.

- Description: The use of controlled release dosage forms has increased significantly in recent years as they result in increased patient compliance and higher therapeutic efficiency. This research focused on the development of a once daily dosage form that could be used for the treatment of hypertension. Both a separate sustained release dosage of metoprolol tartrate and a combination dosage form that included both an immediate release hydrochlorothiazide and a sustained release metoprolol component, were developed and evaluated. A matrix tablet, consisting of an ethylcellulose ranulation of metoprolol tartrate compressed into a hydrophilic hydroxypropyl methylcellulose polymer matrix, effectively sustained metoprolol release over a 22-hour experimental period. A multiparticulate combination dosage form that consisted of six coated mini matrix tablets of metoprolol and a powder blend of hydrochlorothiazide packed into a gelatin capsule, displayed zero order release kinetics for metoprolol release over 22 hours (r2=0.9946). The release of hydrochlorothiazide was found to be comparable to that of a commercially available product tested. Differential Scanning Calorimetry was used to identify possible incompatibilities between MPTA and excipients initially, and long term stability testing was used to assess to behaviour of the dosage form. Dissolution testing of the dosage forms was performed using USP Apparatus III, which was found to be more discriminating between the batches assessed. Dissolution curves were evaluated for similarity and difference using f1 and f2 fit factors. Samples were analyzed using a high performance liquid chromatographic method that was developed and validated for the simultaneous determination of the compounds of interest. Various factors influencing drug release from the developed dosage forms were assessed and recommendations for further optimization of the formulation are made. Factors evaluated included the quantity of granulating fluid, matrix polymer content, drug load and process variables, including drying time and compression force. The influence of various coating levels on drug release was assessed and none of the levels assessed were found to adequately retarded drug release over a 22-hour period. Combinations of tablets coated to different levels allowed for the successful development of a sustained release metoprolol component, which could be included into the combination dosage form.

- Description: The Makganyene Formation forms the base of the Postmasburg Group in the Transvaal Supergroup of the Northern Cape Province. The Makganyene Formation has diamictite as the main rock type, but siltstone, sandstone, shale, and iron-formations are also present. A glacial origin has been proposed in the past due to the presence of dropstones, faceted and striated pebbles. Typically, the Makganyene Formation contains banded iron-formations interbedded with clastic rocks (shale, siltstone, sandstone and diamictites) at the contact with the underlying iron-formations. This transitional zone is generally overlain by massive or layered diamictites which contain poorly sorted clasts (mainly chert) within a shaly matrix. Striated pebbles have been found during field work, and dropstones have been observed in diamictites and banded iron-formations during the study. The top of the Makganyene Formation contains graded cycles interbedded with diamictites and thin layers of andesitic lavas from the Ongeluk Formation. The basal contact of the Makganyene Formation with the underlying Koegas Subgroup was described as unconformable by previous workers. However field work localised in the Rooinekke area shows a broadly conformable and interbedded contact with the underlying Koegas Subgroup. As described above, banded iron-formations are interbedded with the clastic rocks of the Makganyene Formation. Moreover, boreholes from the Sishen area display the same interbedding at the base of the Makganyene Formation. This suggests that no significant time gap is present in the whole succession between the Ghaap and Postmasburg Group. The Transvaal Supergroup in the Northern Cape displays the following succession : carbonates-BIFs-diamictites/ lava-BIFs-carbonates. The Makganyene Formation is thus at the centre of a symmetrical lithologic succession. Bulk rock compositions show that the diamictites have a similar composition to banded iron-formation with regard to their major element contents. Banded iron-formations acted as a source for the diamictites with carbonates and igneous rocks representing minor components. Differences in bulk composition between the Sishen and Matsap areas emphasize that the source of the diamictite was very localised. The Chemical Index of Alteration (CIA) has been calculated, but since the source dominant rock was iron-formation, this index cannot be usefully applied to the diamictites. ACN, A-CN-K, and A-CNK-FM diagrams confer a major importance in sorting processes due to the separation between the fine and coarse diamictites. The interbedded iron-formations display little clastic contamination indicating deposition in clear water conditions. However, dropstones are present in one borehole from the Matsap area, indicating that iron-formation took place under ice cover, or at least under icebergs. Stable isotope studies show that the iron-formations, interbedded towards the base of the Makganyene Formation, have similar values to the iron-formations of the Koegas Subgroup. As a result of the above observations, new correlations are proposed in this study, relating the different Transvaal Supergroup basins located on the Kaapvaal Craton. The Pretoria Group of the Transvaal Basin has no correlative in the Griqualand West Basin, and the Postmasburg Group of the Northern Cape Basin has no lateral equivalent in the Transvaal Basin. These changes have been made to overcome problems present in the current correlations between those two basins. The Makganyene Formation correlates with the Huronian glaciations which occurred between 2.4 and 2.2 Ga ago in North America. Another Precambrian glaciation is the worldwide and well-studied Neoproterozoic glaciation (640 Ma). At each of these glaciations, major banded iron-formation deposition took place with associated deposition of sedimentary manganese in post-glacial positions. The central position of the Makganyene Formation within the Transvaal Supergroup in the Northern Cape emphasizes this glacial climatic dependence of paleoproterozoic banded iron-formation and manganese deposition. However these two Precambrian glaciations are interpreted in paleomagnetic studies as having occurred near to the equator. The controversial theory of the Snowball Earth has been proposed which proposes that the Earth was entirely frozen from pole to pole. Results from field work, sedimentology, petrography and geochemistry were integrated in a proposed depositional model of the Makganyene Formation occurring at the symmetrical centre of the lithologic succession of the Transvaal Supergroup. At the beginning of the Makganyene glaciation, a regression occurred and glacial advance took place. The diamictites are mostly interpreted as being deposited from wet-based glaciers, probably tidewater glaciers, where significant slumping and debris flows occurred. Any transgression would cause a glacial retreat by rapid calving, re-establishing the chemical sedimentation of banded iron-formations. These sea-level variations are responsible for the interbedding of these different types of rocks (clastic and chemical). The end of the Makganyene glacial event is characterised by subaerial eruptions of andesitic lava of the Ongeluk Formation bringing ashes into the basin. Banded iron-formation and associated manganese accumulations are climate-dependant. Glacial events are responsible for the build up of metallic ions such as iron and manganese in solution in deep waters. A warmer climate would induce a transgression and precipitation of these metallic ions when Eh conditions are favourable. In the Transvaal Supergroup, the climatic variations from warm to cold, and cold to warm are expressed by the lithologic succession. The warm climates are represented by carbonates. Cold climates are represented by banded iron-formations and the peak in cold climate represented by the diamictites of the Makganyene Formation. These changes in climate are gradual, which contradict the dramatic Snowball Earth event: a rapid spread of glaciated areas over low-latitudes freezing the Earth from pole-to-pole. Therefore, to explain low-latitude glaciations at sea-level, a high obliquity of the ecliptic is most likely to have occurred. This high obliquity of the ecliptic was acquired at 4.5 Ga when a giant impactor collided into the Earth to form the Moon. Above the critical value of 54° of the obliquity of the ecliptic, normal climatic zonation reverts, and glaciations will take place preferentially at low-latitudes only when favourable conditions are gathered (relative position ofthe continents and PC02 in the atmosphere).

Abstract The constructivist perspectives on learning have helped enhance science educators' understanding of how students make sense of their lived experiences. Constructivism purports to be a transformation of the traditional curriculum. As such this article starts with a brief overview of behaviorism: the scientific approach to education. The main tenets underlying constructivism, how constructivism guides educators to change their classroom practice, and the implications to science teaching have been reviewed. 2 RESEARCH PROJECT TWO (Empirical study): Being Constructive: College students' learning of work and heat as aspects of the energy concept based a constructivist approach. Abstract This study is an extension of a literature review on constructivism as an alternate teaching and learning approach discussed in research project one. It is an empirical study concerning the use of a learning module based on a constructivist approach to develop pre-service student teachers' understanding of work and heat as aspects of the energy concept. The data consisted mainly of transcripts of students' interviews, written responses to questionnaires designed in the form of a worksheet, and comments from non-participant observers and students. The results seem to suggest that a carefully designed learning module based on a constructivist teaching and learning approach may be a valuable tool in developing pre-service student teachers' understanding of work and heat. 3 RESEARCH PROJECT THREE (Empirical study): A College in transition: A case study of the readiness of a college in the Eastern Cape province to implement Outcomes-Based Education in an Education Development centre. Abstract Curriculum 2005 premised on Outcomes-Based Education is the new curriculum framework for South Africa. It signifies a paradigm shift in education from the traditional 'telling-listening' relationship between the teacher and the learner to one that emphasises leamer-centred approach to the teaching process. Teachers, though recognized as crucial to the educational transformation process in the country have also being identified as ill-equipped to meet the challenges posed by Outcomes-Based Education. This study starts with a brief overview of the South African curriculum and the main tenets underlying Outcomes-Based Education. The institutional conditions and whether the lecturers at a college in the Eastern Cape province perceive the need for a change in their classroom practice were also investigated. Bearing in mind the need for further research to validate the findings of this study, positive indicators that emerged from the study suggest the readiness of the college to implement Outcomes-Based Education at the proposed Education Development Centre.

Removal of copper and nickel from solution by the non-viable biomass of the water fern Azolla filiculoides in an upscaled fixed-bed column system

- Description: The potential of non-viable Azalia filiculaides for the removal of Cu and Ni from aqueous solutions and the possibility of scaling up existing lab scale Azalia column systems was investigated. The effects of factors such as metal starting concentration, pH and two metals in solution on the removal of Ni and Cu from aqueous solution by dried and crushed Azalia biomass were studied in batch systems. Aqueous solutions of Ni with starting concentrations between 1000 and 2000J.lmolll gave the most efficient Ni removal by Azalla biomass. For Cu the optimum starting concentration for adsorption was 50J.lmol/l. The adsorption capacity of both eu and Ni increased as the starting pH of the sorption media increased. The optimum pH for Ni adsorption was found at pH 7 and for Cu, at pH 5. - Awlla biomass had a higher. maximum binding capacity (qrnax) for Cu than for Ni at pH 5. The removal of both Cu allct Ni showed little or no variation with the presence another metal in solution. Kinetic studies show that both Cu and Ni adsorbed rapidly onto the Azalia biomass. The removal of Cu and Ni from aqueous solutions using non-viable Azalia biomass was investigated in a lab scale fixed-bed column and an upscaled 4L column system. The nonviable Azalla filiculaides biomass when dried and used in a column for adsorption of Cu and Ni showed good physical stability under many different conditions. Preparation of the biomass before it could be used in the columns was very simple and did not involve any significant pretreatment steps. Prolonged exposure to UV light decreases Azalia biomass capacity for Ni and Cu adsorption. Column adsorption of Cu and Ni from aqueous solutions was successfully upscaled approximately 100 times. Relative to the lab scale column, the 4L column performed better for the uptake of Cu and Ni per gram of biomass. The larger column was also able to operate at relatively higher flow rates. The biomass showed good reusability with little change in the amount of Ni adsorbed in 10 consecutive cycles. Electron micrographs showecf little or no change in the physical structure and integrity of the Azolla biomass after exposure to mineral acids, Ni solution and high flow rates over 10 consecutive adsorption and desorption cycles. As much as 80% Ni and 70 % Cu was recovered when desorption profiles were generated using O.lMHCI as a desorption agent. The 4L column system was also tested using a highly concen~rat:~ Ni plating bath solution.(Nicrolyte 1). Only 18 % of the Ni could be removed from the expended Nicrolyte 1 pla~Jng solution after treating only 25L, indicating that Azolla biomass is more suited for removal of metals from more dilute industrial effluents.

- Description: The potential of non-viable Azalia filiculaides for the removal of Cu and Ni from aqueous solutions and the possibility of scaling up existing lab scale Azalia column systems was investigated. The effects of factors such as metal starting concentration, pH and two metals in solution on the removal of Ni and Cu from aqueous solution by dried and crushed Azalia biomass were studied in batch systems. Aqueous solutions of Ni with starting concentrations between 1000 and 2000J.lmolll gave the most efficient Ni removal by Azalla biomass. For Cu the optimum starting concentration for adsorption was 50J.lmol/l. The adsorption capacity of both eu and Ni increased as the starting pH of the sorption media increased. The optimum pH for Ni adsorption was found at pH 7 and for Cu, at pH 5. - Awlla biomass had a higher. maximum binding capacity (qrnax) for Cu than for Ni at pH 5. The removal of both Cu allct Ni showed little or no variation with the presence another metal in solution. Kinetic studies show that both Cu and Ni adsorbed rapidly onto the Azalia biomass. The removal of Cu and Ni from aqueous solutions using non-viable Azalia biomass was investigated in a lab scale fixed-bed column and an upscaled 4L column system. The nonviable Azalla filiculaides biomass when dried and used in a column for adsorption of Cu and Ni showed good physical stability under many different conditions. Preparation of the biomass before it could be used in the columns was very simple and did not involve any significant pretreatment steps. Prolonged exposure to UV light decreases Azalia biomass capacity for Ni and Cu adsorption. Column adsorption of Cu and Ni from aqueous solutions was successfully upscaled approximately 100 times. Relative to the lab scale column, the 4L column performed better for the uptake of Cu and Ni per gram of biomass. The larger column was also able to operate at relatively higher flow rates. The biomass showed good reusability with little change in the amount of Ni adsorbed in 10 consecutive cycles. Electron micrographs showecf little or no change in the physical structure and integrity of the Azolla biomass after exposure to mineral acids, Ni solution and high flow rates over 10 consecutive adsorption and desorption cycles. As much as 80% Ni and 70 % Cu was recovered when desorption profiles were generated using O.lMHCI as a desorption agent. The 4L column system was also tested using a highly concen~rat:~ Ni plating bath solution.(Nicrolyte 1). Only 18 % of the Ni could be removed from the expended Nicrolyte 1 pla~Jng solution after treating only 25L, indicating that Azolla biomass is more suited for removal of metals from more dilute industrial effluents.

These pipes have been labelled the South, Centre and North Lobes. The kimberlites intruded a thick sequence of Proterozoic shales, dolomites and sandstones and a thin veneer of consolidated to poorly consolidated mudstones and siltstones of the Karoo Supergroup. Although the shapes of these pipes are comparable to other southern African pipes, they are filled predominantly with crater facies volcaniclastic kimberlite. No tuffisitic kimberlite breccia, the characteristic rock type of the diatremes of other southern African pipes, has yet been identified. The Jwaneng kimberlite thus represents an exception to the standard model for southern African kimberlites, implying that different processes need to be invoked to explain its fonnation. The present study involves a detailed volcanological and sedimentological analysis of the volcaniclastic fill of the Jwaneng South Lobe. Two principal and distinct lithofacies have been identified: the quartz-free RVK facies and the quartz-bearing QRVK facies. Both facies include fine to coarse grained, predominantly massive and subordinate chaotically bedded deposits. The volcaniclastic rocks have been classified as resedimented volcaniclastic kimberlite (RVK) , since their deposition is ascribed to mechanisms dominated by mass flow processes. Based on certain characteristics and differences between the two principal facies, and their spatial distribution within the pipe, they are interpreted as being the products of at least two separate eruption episodes. Certain characteristics (e.g. }hape, granularity~ of the juvenile· magma clasts III the volcaniclastic kimberlite suggest complete crystallisation and devolatilisation of the magma at depth prior to explosive fragmentation. A scenario in which this might have occurred, and which led to catastrophic explosive eruption and pipe excavation is proposed. Explosive eruption and associated tuff cone formation is followed by resedimentation of the material back into the pipe by mass flow processes. Mass flow processes are dominated by debris flow, with lesser grain flow, hyperconcentrated flow and subaqueous mud flow and suspension settling of muddy kimberlitic sediments. Geochemical analyses of the latter indicate a high degree of contamination and weathering, and mixing between pristine kimberlite and silicic shale/mud compositions. Failure and collapse of parts of the underlying pipe walls yielded megablocks of poorly consolidated Permian Karoo mudstone in the peripheral zone of the pipe. This source of the megablocks is supported by their bulk chemical composition. Minor phreatic/phreatomagmatic eruptions are suggested by the presence of rare accretionary and armoured lapilli within both the QRVK and RVK facies. Subsidence of the volcaniclastic pipe fill, inferred mainly from the oversteepened dips of the bedded QRVK and RVK facies, may be related to gravity-induced compaction, late-stage phreatomagmatic eruptions or eruption ofthe adjacent Centre Lobe.

- Description: Metallophthalocyanine complexes containing non-transition metals are very useful as sensitizers for photodynamic therapy, a cure for cancer that is based on visible light activation of tumour localized photo sensitizers. Excited sensitizers generate singlet oxygen as the main hyperactive species that destroy the tumour. Water soluble sensitizers are sought after for the convenience of delivery into the body. Thus, phthalocyanine (pc), tetrapyridinoporphyrazines (tppa) and tetramethyltetrapyridinoporphyrazines (tmtppa) with non-transition central metal atoms of Ge, Si, Sn and Zn were studied. First was the synthesis of these complexes, followed by their characterisation. The characterisation involved the use of ultraviolet and visible absorption spectroscopy, infrared spectroscopy, nuclear magnetic resonance spectroscopy, electrochemical properties and elemental analysis. Photochemical properties of the complexes were then investigated. Photolysis of these macrocycles showed two processes; -reduction of the dye and photobleaching, which leads to the disintegration of the conjugated chromophore structure of the dye. Photobleaching is the reductive quenching of the excited state of the sensitizers. The intensity of the quenching decreased progressively from tmtppa, tppa to pc metal complexes with photobleaching quantum yields, 6.6 x 10.5⁻¹, 1.8 x 10.5⁻¹ and 5.4 x 10⁻⁶ for Zntmtppa, Zntppa and Znpc, respectively. Efficiency of singlet oxygen sensitization is solvent dependent with very different values obtained for the same compound in different solvents, for example, 0.25 and 0.38 were observed as singlet oxygen quantum yields for Gepc complex in DMSO and DMF respectively. In DMSO the efficiency of ¹O₂ generation decrease considerably from pc to tppa and finally tmtppa. In water Getmtppa exhibits much higher singlet oxygen quantum yield, hence promising to be effective as a sensitizer for photodynamic therapy.

Novel approaches to the synthesis of quinoline derivatives

- Description: The investigation has been concerned with the application of the Baylis-Hillman methodology to the synthesis of quinoline derivatives. An extensive range of novel Baylis-Hillman products has been prepared, typically in moderate to excellent yields, by condensing 2-nitrobenzaldehyde derivatives with various vinyl ketones and acrylic esters in the presence of diazabicyclo[2.2.2]octane (DABCO). Reduction of the nitro group in the Baylis-Hillman products was effected by catalytic hydrogenation in ethanol using a 10% palladium-on-carbon catalyst to afford quinoline, quinoline-N-oxide and quinolone derivatives. In all cases, it is apparent that cyclisation involves exclusive attack of nucleophilic nitrogen at the carbonyl centre, with acrylic ester derivatives affording quinolones and vinyl ketone derivatives affording quinolines and the corresponding quinoline-N-oxides. No products arising from a conjugate addition pathway were observed. The use of stannous chloride as an alternative reagent to effect reductive cyclisation of the Baylis-Hillman products has been explored, and found to favour the formation of 1,2- dihydroquinoline derivatives, with cyclisation occurring via a conjugate addition pathway. Isolation of the products, following work-up of the stannous chloride reactions, however, presented some difficulty. All compounds were characterised by spectroscopic (NMR and IR) and, where appropriate, elemental (high-resolution MS) analysis. Interconversion of the quinoline and quinoline-N-oxide derivatives has been explored and finally achieved in quantitative yields. Reduction of 2,3-dimethylquinoline-N-oxide to the corresponding quinoline was effected using phosphorus tribromide in DMF, and the reverse transformation with meta-chloroperbenzoic acid (MCPBA) in CHCl₃. Laboratory and occupation-simulating isokinetic and psychophysical responses of military personnel

The study aimed to evaluate the strength capabilities of South African infantrymen and establish benchmark data on a population not previously tested. “Work-simulation” packages have not been widely exploited and this study further aimed to approximate how effectively occupation simulating tasks could identify the capabilities of soldiers. Testing was carried out using a CYBEX 6000 isokinetic dynamometer and involved six laboratory tests (LTs) and four occupation-simulating tests (OSTs). Subjects were required to complete two testing sessions with the order of tests randomized. The LTs consisted of ankle, elbow, hip, knee, shoulder and trunk. In the OSTs, gripping, valve-tightening, wrench-turning and pulling/pushing responses were collected. Slow, medium and fast test speeds were used for each bout. Cardiac responses were measured using heart rate monitoring and perceptual measures assessed using Borg’s (1971) RPE scale. The results of the testing showed significant differences in agonist and antagonist responses at all three testing speeds, the only exception being slow speed trunk values (peak torque). Upper- to lower-extremity ratios highlighted a possible weakness in the elbow flexors group, while correlations between LTs and OSTs highlighted the specificity of strength principle, as poor relationships were observed.

The thickness of the Karoo sediment unit ranges from 30m to 80m. The hydraulic parameters of the Karoo sediment aquifer were characterised in the field by constant discharge pumping tests and slug tests. Pumping tests indicated unconfined conditions and thus the Neuman's method of analysis has been used. Transmissivities from pumping tests are within the range 4.7 m²/d to 13.6 m²/d with an average of 8.9m²/d. The low transmissivities seem to be a major limiting factor in the exploitation of the groundwater resources. Thus the sustainable borehole yields tend to be small, mean values ranging from 33 m²/d to 253 m²/d. Specific yield could not be determined from the pumping tests due to the lack of observation boreholes. Low chemical concentrations render the water suitable for irrigation of all crops, while neither total nor any individual concentrations present health hazards to human or livestock. An average recharge value of 47.7 mm/y was inferred from water table fluctuation method. Chloride mass balance technique in the same area indicates recharge value in the order of 67.4 mm/y. Because the chloride mass balance gives a long-term mean annual recharge, the recharge figure of 67.4 mm/y was adopted for the study area. Based on the abstractable proportion of recharge, the sustainably exploitable volume of groundwater of the order of 2.68 x 10⁷ m³/y was established. This volume is more than 100 times the estimated current demand for groundwater (1.35 x 10⁵ m³/d), implying that there are large volumes of surplus water, which can be utilised for irrigation.

Specific yield could not be determined from the pumping tests due to the lack of observation boreholes. Low chemical concentrations render the water suitable for irrigation of all crops, while neither total nor any individual concentrations present health hazards to human or livestock. An average recharge value of 47.7 mm/y was inferred from water table fluctuation method. Chloride mass balance technique in the same area indicates recharge value in the order of 67.4 mm/y. Because the chloride mass balance gives a long-term mean annual recharge, the recharge figure of 67.4 mm/y was adopted for the study area. Based on the abstractable proportion of recharge, the sustainably exploitable volume of groundwater of the order of 2.68 x 10⁷ m³/y was established. This volume is more than 100 times the estimated current demand for groundwater (1.35 x 10⁵ m³/d), implying that there are large volumes of surplus water, which can be utilised for irrigation.

- Description: Ibuprofen is a non-steroidal anti-inflammatory drug with three major types of effect: anti-inflammatory, analgesic and antipyretic. Ibuprofen may be administered in a number of different forms via the oral as well as the topical route. Published evidence suggests that topical, unlike oral, non-steroidal anti-inflammatory drugs are associated with few systemic side effects as plasma concentrations are low compared to oral therapy. In some countries it is particularly difficult to obtain human skin for in vitro experimentation and it is therefore important to have alternate biological or synthetic membranes which mimic human skin for diffusion experiments. Synthetic membranes serve as predictive models for topical drug release and in South Africa, shed snake skin is easily obtainable from the many snake parks present in the country. The FDA guidelines were considered when choosing the apparatus to be used in the comparative diffusion study on proprietary ibuprofen-containing topical preparations from three countries and the verification of the usefulness, or otherwise, of shed snake skin as a biological membrane for the assessment of the permeation of ibuprofen. Two diffusion techniques were considered appropriate for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing. One was the Franz diffusion cell, as modified by Keshary and Chien (88,169) and the other was the European Pharmacopoeia diffusion cell (187). High performance liquid chromatography was used as the analytical technique for the analysis of ibuprofen in aqueous solution using ultraviolet detection at 222 nm. The validated method was applied to the determination of the diffusion of ibuprofen from topical ibuprofen-containing formulations (gels, creams and mousse) through synthetic silicone membrane and shed snake skin biological membrane from four different species. In a study of fifteen topical ibuprofen-containing formulations (gels, creams and mousse) from three countries (South Africa, United Kingdom and France) it was found that there was a trend of products from two countries consistently exhibiting superior diffusion characteristics as well as products from the same two countries consistently exhibiting the lowest diffusion of ibuprofen. Interpretation of the results of these studies demonstrated the importance of employing a combination of statistical analyses and peak integration values when drawing conclusions regarding comparative diffusion characteristics. Shed snake skin has been described as a 'model' membrane, i.e. a membrane which shows similar permeability to human stratum corneum. The results reported here show clearly that, for ibuprofen, the four species of snake produce shed skin with completely different diffusion characteristics when all other conditions are identical. It may well be that there is one particular species of snake which produces shed skin of identical permeability to human stratum corneum, but to describe shed snake skin in general as a model membrane seems incorrect. It is therefore important that if shed snake skin is used as a membrane, the species, skin site and orientation should be reported. The European Pharmacopoeia diffusion apparatus was judged to be the better of the two diffusion techniques assessed for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing using silicone membranes and for the measurement of the amount of ibuprofen diffusing across the ventral outside orientation of shed skin during in vitro testing, whereas the Franz diffusion apparatus was judged to be better for the measurement of the amount of ibuprofen diffusing across the dorsal outside orientation of shed skin during in vitro testing. However, the choice of this diffusion apparatus must be weighed against the relatively poor reproducibility as compared with the European Pharmacopoeia diffusion apparatus.
