- Description: Malaria is one of the three most deadly diseases in Africa. Although there are available treatments, their efficacy has been greatly reduced over the past two decades due to the development of resistance to currently available drugs. This has necessitated the search for new and effective antimalarial agents. This project approached the search for new antimalarial compounds in two ways: (i) by screening natural products isolated from marine algae against the Plasmodium parasite and (ii) by modification of selected isolated active compounds to target 1-deoxY-đ-xylulose 5-phosphate reductoisomerase (DXR), an enzyme found in the nonmevalonate isoprenoid biosynthetic pathway of Plasmodium Jalciparum. It was envisaged that such a compound would exhibit dual action on the Plasmodium parasite. Extracts obtained from 22 marine algae were prefractionated by solvent partitioning and were screened for anti plasmodial activity against the chloroquine sensitive (CQS) P. Jalciparum D 10 strain. Overall, 50% of the algae screened produced at least one crude fraction with activity against P. Jalciparum. Extracts of the algae Sargassum heterophyllum, Plocamium cornutum, Amphiroa ephedrea and Pterosiphonia cloiophylla gave the most promising results. Fractionation of S. heterophyllum afforded three tetraprenyltoluquinols (3.1, 3.2 and 3.5) and an all-trans-fucoxanthin (3.6). Three new compounds (4.5, 4.6 and 4.7) and two known halogenated monoterpenes (4.1 and 4.4) were isolated from P. cornutum. Each of the isolated compounds from both S. heterophyllum and P. cornutum showed antiplasmodial activity with IC₅₀ values ranging from 2.0 - 15.3 μM for S. heterophyllum and 13 - 230 μM for P. cornutum. Attempts to synthetically modify halogenated monoterpene 4.4 by dihydroxylation and phosphorylation in order to inhibit the DXR enzyme was unsuccessful. However, the hemiterpene analogue (5.42) of the halogenated monoterpenes was successfully phosphorylated and dihydroxylated to give compound 5.45 which showed promising activity against DXR. The result obtained indicated that the proposed phosphorylation and dihydroxylation of the halogenated monoterpene 4.4 would result in the synthesis of a potent DXR inhibitor and therefore a potential antimalarial agent with dual mode of action on the Plasmodium parasite.

Purification of the hydrogenase by freeze-drying and Sephacryl S200 size exclusion- ion exchange chromatography revealed the enzyme to be a dimer with a 29.4 and 44.5 kDa when analysed by a 10 % SDS-PAGE. Characterisation of the enzyme revealed optimal activity at a pH of 7.5 and temperature of 38 °C while it exhibited a poor thermal stability with a half life of 36 minutes. The kinetic parameters Vmax and Km were 3.16 U ml-1 and 3.64 mM respectively. The purified hydrogenase was used in subsequent experiments for the reduction of platinum salts, H2PtCl6 and PtCl2. the results indicated an over 90 % reduction of the platinum salts and TEM micrographs indicated the production of platinum nanoparticles under the various experimental conditions.

- Description: The larval fish assemblage in the shallow, nearshore region of a proposed marine protected area (MPA) in eastern Algoa Bay, temperate South Africa was investigated. Current velocities and direction and the swimming abilities of late-stage larvae were further assessed to determine potential larval movement to and from the MPA. In total, 6045 larval fishes were collected along two depth contours (~5m and ~15m) in the shallow nearshore of eastern Algoa Bay using stepped-oblique bongo net tows, twice per season for two years (March 2005 – January 2007). These larvae represented 32 families and 78 species. The Gobiidae, Cynoglossidae, Clupeidae, Engraulidae and Sparidae were the dominant fish families. Catches varied significantly between seasons peaking in spring with a mean of 64 larvae/100m3. Preflexion stage larvae dominated catches (75%). All developmental stages of Diplodus capensis, Engraulis capensis, Heteromycteris capensis, Sardinops sagax and Pomadasys species were found in the study area. It appears that these species use the shallow nearshore as a nursery area. Analysis of 12 months (May 2006 – May 2007) data from a bottom-moored Acoustic Doppler Current Profiler within the study area showed that offshore south eastward (39%) and onshore north westward currents (33%) dominated. The south westward current (15%) and north eastward current (12%) occurred less frequently. Current velocity decreased with depth in the nearshore, with a mean velocity of ~29 cms⁻¹ recorded at a depth of 4 m and a mean velocity of ~11 cms⁻¹ recorded at a depth of 14 m. Understanding the dispersal and movement of marine fish larvae in coastal habitats requires knowledge of active swimming abilities. The critical speed and endurance swimming of late stage larvae of two common inshore species occurring in the study area, Diplodus capensis and Sarpa salpa (Family Sparidae), were measured in a laboratory swimming chamber. The mean U-crit value for D. capensis (18.6 cms⁻¹) was similar to that of S. salpa (18.0 cms⁻¹), whereas mean endurance (km swum) was greater in S. salpa (8.4 km) than D. capensis (5.9 km). These swimming abilities exceed the average current velocities observed in the shallow nearshore providing larvae with the ability to greatly alter their passive dispersal trajectories and ultimately influence their distribution in the nearshore.

- Description: Using a pseudo-bisymmetric enriched cl-premonoid as the underlying lattice, we examine different categories of lattice-valued spaces. Lattice-valued topological spaces, uniform spaces and limit spaces are described, and we produce a new definition of stratified lattice-valued uniform convergence spaces in this generalised lattice context. We show that the category of stratified L-uniform convergence spaces is topological, and that the forgetful functor preserves initial constructions for the underlying stratified L-limit space. For the case of L a complete Heyting algebra, it is shown that the category of stratified L-uniform convergence spaces is cartesian closed.

- Description: Oldenburgia grandis is a rare, long-lived woody paleoendemic of the Fynbos Biome of South Africa. Confined to quartzite outcrops, it has a small geographic range and narrow habitat specificity. O. grandis responds to its fire-prone environment by resprouting. Elasticity analysis of O. grandis reveals that growth and fecundity were traded off for persistence of adult, mature and sapling stages. Morphological adaptations such as a corky fire-resistant bark and the ability to resprout after fire are traits that O. grandis have evolved to persist in a frequently disturbed environment. Population growth rate for sites undisturbed by fire for a number of years (l = 1.01) and sites at various stages of recovery after fire (l = 1.00) were very similar. The highest variation in transition probabilities for all sites was seen in the persistence of the seedling stage and growth from seedling to sapling. Observed population structure and stable stage distribution determined by the matrix model show that sites recently undisturbed by fire had high abundances of the adult and sapling stages. A peak in sapling stages was seen for the stable stage distribution where similar peak in sapling numbers were seen for population structures of sites at various stages of recoveryafter fire. Favourable environmental conditions for the persistence of O. grandis populations include no fire with transition probabilities between the observed minimum and maximum and fire frequency at a 10 year interval where seedling protection from the fire is high and adult and mature mortalities during the fire are low. Stochastic environmental events that could put populations (particularly small populations) at an increased risk of extinction include high to moderate fire intensities where seedling protection from the fire is low and adult and mature mortalities are high as a result of the fire.

- Description: Design changes due to lack of constructability, cost overruns, delays and dissatisfied clients are but a few problems experienced in construction due to poor management of the design processes. Increased problems, challenges, demands and continuous criticism of the architectural profession has led to increased demand for research into the improvement of design processes. The aims of this research were to determine the adequacy of design management processes used by Eastern Cape (EC) architectural companies and compare these with the design management processes used in manufacturing in order to establish practices, theories, principles, technologies and deliverables that can be transferred from the manufacturing into the construction to improve efficiency of architectural design management. The quantitative research approach was implemented for this research, the questionnaire was designed to acquire primary, factual and attitudinal data from EC architectural companies and secondary data were acquired through a literature review. The results revealed that design management processes, continuous improvement philosophies, lean principles, and Information and Communication Technology (ICT) used by EC architectural companies are not similar to those used in manufacturing. Therefore EC architectural companies could increase their efficiency by adopting some of the design management processes, ICT, continuous improvement philosophies and lean principles originating from the manufacturing industry.

- Description: Design changes due to lack of constructability, cost overruns, delays and dissatisfied clients are but a few problems experienced in construction due to poor management of the design processes. Increased problems, challenges, demands and continuous criticism of the architectural profession has led to increased demand for research into the improvement of design processes. The aims of this research were to determine the adequacy of design management processes used by Eastern Cape (EC) architectural companies and compare these with the design management processes used in manufacturing in order to establish practices, theories, principles, technologies and deliverables that can be transferred from the manufacturing into the construction to improve efficiency of architectural design management. The quantitative research approach was implemented for this research, the questionnaire was designed to acquire primary, factual and attitudinal data from EC architectural companies and secondary data were acquired through a literature review. The results revealed that design management processes, continuous improvement philosophies, lean principles, and Information and Communication Technology (ICT) used by EC architectural companies are not similar to those used in manufacturing. Therefore EC architectural companies could increase their efficiency by adopting some of the design management processes, ICT, continuous improvement philosophies and lean principles originating from the manufacturing industry.

- Description: A great deal of research has demonstrated that Non-Timber Forest Products (NTFPs) play a crucial role in the livelihoods of the rural poor, and are particularly important to the most marginalised people throughout the developing world. However, these livelihood benefits are not without cost to the natural resource base that rural communities depend so heavily upon. The continued dependence on NTFPs as a major livelihood source must be contingent upon the minimisation of the level of disturbance created through this dependency. This study assesses the level of disturbance created through natural resource harvesting in eight study sites around South Africa, and applies a predictive conceptual model created by Shankaar et al. (2004b) to try and ascertain under what conditions the level of disturbance created through natural resource harvesting will be high. It assesses the three key factors that Shankaar et al. (2004b) identified (level of Local Ecological Knowledge (LEK), level of dependency and access to markets) in relation to the level of disturbance found at each of the study sites. It was found that there was a statistically significant relationship between the level of dependency and the level of disturbance, but there was no statistically significant relationship between either access to markets or the level of LEK and disturbance. Regulation of land use is a key issue, with weak local institutions in communal areas making effective resource management difficult. The significance of these findings is discussed, and priorities for future research are identified. This study adds to the body of knowledge related to NTFP harvesting and critically analyses the conflicts between the livelihood gains and the level of disturbance created through NTFP harvesting in an attempt to ascertain how livelihoods can be safeguarded. And in the longer-term, so that management strategies can be identified where resource extraction is not at the cost of undermining the very livelihoods that depend upon the natural resource base.

On the Processing of InAsSb/GaSb photodiodes for infrared detection

- Description: The objective of this dissertation is the development of the necessary processing steps needed to manufacture infrared photodiodes on InAs1-xSbx material. Preliminary surface preparation steps were performed on both InAs and InSb material, thus covering both possible extremes of the antimony mole fraction. The first experiments endeavoured to characterise the effect of several possible etchants with regards to etch rate, repeatability, limitations for photolithographic patterning and the resultant surface roughness. The etchants investigated include a lactic acid based etchant, a sulphuric acid based etchant, an acetic acid based etchant, an ammonium based etchant, a hydrochloric acid based etchant as well as an organic rinse procedure. These cleaning and etching steps were evaluated at several temperatures. Measurements were performed on an Alpha Step stylus profiler as well as an atomic force microscope. Metal-insulator-semiconductor capacitor devices were manufactured, on both InAs and InSb material, in order to investigate the effects of the above-mentioned etchants combined with surface passivation techniques in terms of surface state densities. Capacitance-versus-bias voltage measurements were done to determine the resultant surface state densities and to compare these to the surface state density of an untreated reference sample. The surface passivation techniques included KOH, Na2S as well as (NH4)2S anodisation. Auger electron spectroscopy measurements were done on InAs and InSb material in order to examine possible surface contamination due to the etchants as well as combinations of these etching and anodisation procedures. The extent of surface coverage by contaminants as well as by the intrinsic elements was measured. The results of the cleaning and etching as well as the surface passivation studies were used to manufacture photovoltaic infrared diodes on an MOCVD (metal oxide chemical vapour deposition) grown p-InAs0.91Sb0.09/i- InAs0.91Sb0.09/n-GaSb structure. Current-versus-voltage and electro-optical measurements were performed on the these diodes in order to evaluate the effect of sulphuric acid based etching combined with KOH, Na2S or (NH4)2S anodisation on the detector performance. The results of surface passivated structures were compared to those of an unpassivated reference detector.

- Description: Marine Protected Areas (MPAs), the Ecosystem Approach to Fisheries management (EAF) and Integrated Coastal Management (ICM) have been identified as possible alternatives to traditional linefish management measures, which have largely failed. Monitoring and assessment of fish communities on a long-term basis is necessary, and will provide a means to evaluate the effectiveness of such management measures. Therefore, standardised protocols and optimal sampling methods for long-term monitoring (LTM) and assessment of coastal fish communities are essential. This study aimed to identify suitable methods and develop a protocol for assessment of inshore reef fish communities. A suitable location for evaluation of proposed methods was identified in the warm temperate biogeographical region of South Africa, encompassing the well-established Tsitsikamma Coastal National Park MPA and an adjacent exploited area. Chrysoblephus laticeps (roman) was identified as an indicator species for the study, as it has been well-studied and is well represented in the area. Underwater visual census (UVC) and controlled fishing were identified as suitable methods. UVC transects were found to be superior to point counts, in terms of sampling efficiency, variability, bias and required sample size. An effort of two angler hours per fishing station was shown to provide low catch variability, while at the same time a representative catch and low overall cost and required time. The methods were incorporated in a proposed sampling protocol, and evaluated. The methods were able to detect known differences between protected and exploited communities. It is recommended that LTM within protected areas, for detection of natural change, be focused on community-level indicators, while LTM in exploited areas, aimed at detection of anthropogenic change, be focused on species-level indicators. The proposed protocol with standardised methods will allow for comparisons across a network of LTM sites and provide the opportunity for a broad-scale assessment of the effects of environmental variables on reef fish stocks. The protocol developed in this study has application in other biogeographical regions in South Africa, and other parts of the world. Shift in the focus of much marine research, in South Africa and elsewhere, to LTM, highlights the relevance and timeous nature of this study.

- Description: The greater photosynthetic nitrogen use efficiency (PNUE) of C4 compared with C3 plants may explain the relative success of C4 grasses in nutrient poor environments. This study compared the responses in photosynthetic parameters, leaf nitrogen and biomass allocation between the C3 and C4 subspecies of Alloteropsis semialata supplied soil nitrogen at three levels. Photosynthesis was assessed by means of CO2 response curves and the leaf nitrogen content assayed. Plants were destructively harvested, leaf areas determined and the dry biomass of functional plant components was measured. Results confirmed that the higher PNUE of C4 plants allowed them to accumulate more biomass than C3 plants at the high nitrogen level, despite smaller leaf areas. The greater productivity of C4 plants enabled them to invest more in storage and sexual reproduction than in leaves when compared to the C3 plants. In contrast the C3 plants invested biomass in less efficient and more nitrogen demanding leaves and bigger root systems. PNUE and photosynthetic rates were not significantly affected by nitrogen-limitation in either subspecies and the major response was a decrease in biomass accumulation and an increase in biomass allocation to roots. This altered root to shoot ratio was accompanied by a lowered allocation to sexual reproduction in the C4 subspecies, but an unaltered allocation to leaves, while in the C3 subspecies there was a decrease in leaf allocation. In a further experiment, the C4 subspecies was supplied three levels of nitrogen provided as nitrate, or alternatively as ammonium plus nitrate, and leaves were excised to within 5 cm of the ground at the start of treatment. Prior to flowering, photosynthesis was assessed by means of CO2 response curves and the plants were destructively harvested. Leaf areas and the dry biomass of functional plant components were determined, and at levels of nitrogen supply higher than those found in savanna soils the rate of photosynthesis was increased. Leaf re-growth was reduced by severe nitrogen limitation and co-provision of nitrate and ammonium had no significant effect other than increased tillering. Both subspecies of Alloteropsis semialata are adapted to nutrient poor environments and maintain photosynthetic rates by reducing leaf area. The C4 subspecies is likely to show greater resilience in disturbance-prone environments by exploiting its higher PNUE to allocate greater resources to storage and sexual reproduction, while the C3 subspecies is usually found in environments with closed canopies which favour vegetative growth, and allocate greater resources to leaves and roots.

- Description: This study examined the current plant diversity status and the impact of drivers of change on the peat basins of the Kromme River peatland. It was conducted at six sites over sixty one years in the Eastern Cape Province of South Africa. I reviewed the rapid habitat and biodiversity loss of wetlands globally and discussed the distribution of wetlands and specifically peatlands in South Africa. Plant species diversity was assessed using Modified- Whittaker plots. The influence of environmental variables on floristic composition and distribution was investigated using ordination techniques (DCA and CCA). Land use dynamics were assessed by applying GIS techniques on orthorectified aerial images. Six different peat basins were subjectively classified into good, medium and poor condition peat basins. The good condition peat basin (Krugersland) was the most diverse in plant species (4.1 Shannon-Weiner’s index) (p> 0.20; F = 11.04; df = 2), with the highest mean number of plant species (32.5 ± 3.4). This was followed by the medium condition class (Kammiesbos) (26.5 ± 9.0) and poor condition class (Companjesdrift) (22.5 ± 8.9). On average, species composition was not evenly distributed across the peat basins (p> 0.21; F = 0.94; df = 2), since 77.8% of the Shannon-Weiner evenness index obtained were less than one. However, there were variations in plant species richness across six peat basins as confirmed by Oneway ANOVA test (p= 0.0008, F = 1241.6, df = 4). Key environmental variables that influenced plant species distribution and structure were erosion and grazing intensity, potassium, phosphorus, soil pH and calcium. Total species variance accounted for in the first two axes for ground cover and plant height were 40.7% and 56.4% respectively. Alien species (e.g. Acacia mearnsii and Conyza scabrida) were common in degraded peat basins, whereas good condition peat basins supported indigenous species (e.g., Cyperus denudatus, Chrysanthemoides monolifera and Digitaria eriantha). Analysis of aerial images revealed a general progressive decrease in the peatland area between 1942 and 1969 in the good (Krugersland) and poor (Companjesdrift) condition class, with a marginal increase from 1969 to 2003. Peatland area in the good and poor condition class decreased by 5.3% and 8.3% respectively between 1942 and 1969, with a marginal increase of 1.5% and 4.1% respectively from 1969 to 2003. Annual net rate of change in peatland area over the 61 year period was -0.32% (good condition class) and - 0.79% (poor condition class). Transformed lands were impacted by drivers of change such as alien invasives, agricultural activities, erosion and sediment transport. The area under alien invasives increased by 50% between 1942 and 2003, with an annual net rate of change of +0.82 (good condition class) and +1.63% (poor condition class).

Pneumocystis jiroveci and respiratorey bacterial pathogens in cases of pneumonia at hospitals in Port Elizabeth

- Description: Pneumocystis jiroveci, Mycoplasma pneumoniae and Mycobacterium tuberculosis are respiratory pathogens associated with pneumonia, with increasing prevalence of Pneumocystis pneumonia (PcP) and tuberculosis (TB) in AIDS patients. Increased resistance of M. tuberculosis has emphasized the need for rapid susceptibility testing, such as flow cytometry. Sputum specimens (102) were assessed by PCR employing primers directed at the following genes: P. jiroveci: mitochondrial large subunit ribosomal RNA (mtLSUrRNA), dihydropteroate synthase (DHPS) and dihydrofolate reductase (DHFR), and for M. pneumoniae: 16S rRNA and P1 adhesin. Positive P. jiroveci samples were genotyped by PCR-SSCP (single-strand conformation polymorphism) targeting the: internal transcribed spacer region (ITS), intron of the nuclear 26S rRNA gene (26S), variable region of the mitochondrial 26S rRNA gene (mt26S) and β-tubulin gene (β-tub). Multi-drug resistant (MDR-TB) cultures grown in the presence and absence of four antibiotics (rifampicin, isoniazid, ethambutol and ofloxacin) were heat killed, stained with SYTO16 and Propidium Iodide and analysed using flow cytometry. Rifampicin resistance gene mutations were screened by PCR and DNA sequencing. Details of patient’s gender, age, HIV and M. tuberculosis status were provided by the hospitals. Women were seen to be at high risk for community-acquired P. jiroveci colonisation. Overall, prevalence of P. jiroveci was 55.1 percent (54/102 patients). P. jiroveci was mainly associated with HIV (25/102 P. jiroveci positive patients for which clinical data was available) and co-colonisation with M. tuberculosis was observed in 11 cases. Sequence analysis of DHPS and DHFR products found no resistance associated mutations. M. pneumoniae was detected in one patient. Four simple SSCP patterns were identified and there were no co-infections with other P. jiroveci strains. Nine M. tuberculosis samples [8 MDR-TB isolates (NHLS) and M. tuberculosis ATCC® 27294TM] were tested. There was a 53 percent (19 out of 36 tests) agreement of flow cytometry with the BACTEC MGIT 960. Mutations (at two specific codons, namely 516 and 531) in the rifampicin resistance-determining region (RRDR) of the rpoB gene were observed in eight M. tuberculosis isolates. Evaluation of methods for genotyping and drug susceptibility testing of PcP and TB are imperative for epidemiology and drug resistance studies, and impact on treatment protocols.

- Description: The Session Initiation Protocol (SIP) has in recent years become a popular protocol for the exchange of text, voice and video over IP networks. This thesis proposes the use of a class of structured peer to peer protocols - commonly known as Distributed Hash Tables (DHTs) - to provide a SIP overlay with services such as end-point location management and message relay, in the absence of traditional, centralised resources such as SIP proxies and registrars. A peer-to-peer layer named OverCord, which allows the interaction with any specific DHT protocol via the use of appropriate plug-ins, was designed, implemented and tested. This layer was then incorporated into a SIP user agent distributed by NIST (National Institute of Standards and Technology, USA). The modified user agent is capable of reliably establishing text, audio and video communication with similarly modified agents (peers) as well as conventional, centralized SIP overlays.

Purification and partial characterization of a Myofibril-Bound Serine Protease and its endogenous inhibitor from skeletal muscle of the ostrich

- Description: The ostrich is becoming an important source of meat for humans in developed and developing countries. This study was conducted to purify and characterize myofibrilbound serine protease (MBSP) and its endogenous inhibitor (MBSPI) from skeletal muscle of the ostrich. It is well documented that MBSP is tightly bound to myofibrils and its endogenous inhibitor has been purified from the same tissue of other studied mammalian species. Literature supports an association of MBSP and its endogenous inhibitor with the degradation of myofribrillar proteins, resulting in the softening of muscle that lead to the conversion of muscle into meat with the control of the inhibitor. MBSP was successfully dissociated from washed myofibrils by 40 percent ethylene glycol at pH 8.5. Following centrifugation, MBSP was partially purified in two chromatographic steps, namely Toyopearl Super Q 650S and p-aminobenzamidine-Agarose. On the other hand, MBSPI was fractionated from the sarcoplasmic fraction using 75 percent ammonium sulfate saturation, followed by centrifugation and partially purified by three chromatographic steps, namely Toyopearl Super Q 650S, Superdex 200 and HiTrap SP HR. Ostrich MBSP was physicochemically and kinetically characterized, while MBSPI was only physicochemically characterized. Ostrich MBSP revealed an Mr of 21 kDa, cleaving synthetic fluorogenic substrates specifically at the carboxyl side of arginine residues. Optimum pH and temperature of ostrich MBSP were 8.0 and 40˚C, respectively. Kinetic parameters (Km and Vmax values) were calculated from Lineweaver-Burk plots. The characteristics of ostrich MBSP were compared to the values obtained for commercial bovine trypsin in this study, as well as that obtained for MBSP from various fish species and mouse. The results suggest that ostrich MBSP is a trypsin-like serine protease, thereby confirming the existence of MBSP in ostrich skeletal muscle. Partially purified ostrich MBSPI (Mr 17 kDa) (one form) shares 100 percent identity to myoglobin from the same species, while 2 other forms of MBSPI (Mr values of 35 and 36 kDa) exhibited high sequence identity to glyceraldehyde 3- phosphate dehydrogenase (GAPDH) (76 percent) from human and rat.

RFID technology provides great potential in many industries and a wide spectrum of possible uses. Areas of application include person identification, logistics, pharmaceutical, access control, security guard monitoring and asset management. One of the areas where RFID is being used and where it promises excellent results is the retail industry. While RFID systems have the potential to revolutionise the way products and goods are tracked and traced in the retail supply chain, barriers to its widespread adoption exist: for example; technical constraints, return on investment constraints, a lack of awareness and education and as well as privacy and security issues. The research aims to identify the barriers to the adoption of RFID and to investigate the perceptions of RFID held by members of the retail sector in South Africa (SA). Current research and available literature are used to identify RFID adoption barriers and a conceptual framework on this subject is proposed, which is then verified by SA retailers’ perceptions, established by means of a survey. Initial barriers to widespread adoption include a shortage in skills, a lack of standards, high costs associated with RFID devices, the difficulty of integrating with current legacy systems, and a lack of familiarity with the system. Finally, an enhanced framework is proposed, describing RFID adoption barriers within the South African retail sector. In summary, the framework is an outline of the barriers impacting RFID adoption in the SA retail sector that need to be considered and addressed. The framework identifies six categories of RFID adoption barriers, with each category containing two or more barriers relating to that particular category. These categories are Technological, Cost and return on investment, Privacy and security, Implementation, Organisational factors and People.

The most effective water-soluble photocatalyst is ZnPcS[subscript mix] in terms of the high quantum yield obtained for 4-Np degradation (Φ[subscript 4-Np]) as well as its photostability. While ZnPc(COOH)[subscript 8] has the highest Φ[subscript 4-Np] value relative to the other water-soluble complexes, it degrades readily during photocatalysis. The Φ[subscript 4-Np] values were closely related to the singlet oxygen quantum yields Φ[subscript Δ] and hence aggregation. The rate constants for the reaction with 4-Np were kr = 0.67 x 10[superscript 6] mol[superscript -1] dm[superscript 3] s[superscript -1] for ZnPcS[subscript mix] and 7.7 x 10[superscript 6] mol[superscript -1] dm[superscript 3] s[superscript -1] for ZnPc(COOH)[subscript 8]. ClAlPc is the most effective photocatalyst relative to the other heterogeneous photocatalysts for the phototransformation of 4-Np, with 89 ± 8.4 % degradation of 4-Np achieved after 100 min. The least effective catalysts were ZnPcCl[subscript 16] and MgPc. The final products of the photocatalysis of 4-Np in the presence of the homogeneous photocatalysts include 4-nitrocatechol and hydroquinone, while degradation of 4-Np in the presence of the heterogeneous photocatalysts resulted in fumaric acid and 4-nitrocatechol. ClAlPc was employed for the heterogeneous photocatalysis of the non-systemic insecticide, methyl paraoxon. Complete degradation of the pesticide was confirmed by the disappearance of the HPLC trace for methyl paraoxon after 100 min of irradiation with visible light. The removal of 4-Np from an aqueous medium using commercially available Amberlite[superscript ®] IRA-900 modified with metal phthalocyanines was also investigated. The metallophthalocyanines immobilised onto the surface of Amberlite[superscript ®] IRA-900 include Fe (FePcS[subscript 4]), Co (CoPcS[subscript 4]) and Ni (NiPcS[subscript 4]) tetrasulphophthalocyanines, and differently sulphonated phthalocyanine mixtures of Fe (FePcS[subscript mix]), Co (CoPcS[subscript mix]) and Ni (NiPcS[subscript mix]). Adsorption rates were fastest for the modified adsorbents at pH 9. Using the Langmuir-Hinshelwood kinetic model, the complexes showed the following order of 4-Np adsorption: CoPcS[subscript mix] > NiPcS[subscript 4] > NiPcS[subscript mix] > FePcS[subscript 4] > FePcS[subscript mix] > CoPcS[subscript 4]. - Description: In recent years, increased demands have been placed on hydrologists to find the most effective methods of making predictions of hydrologic variables in ungauged basins. A huge part of the southern African region is ungauged and, in gauged basins, the extent to which observed flows represent natural flows is unknown, given unquantified upstream activities. The need to exploit water resources for social and economic development, considered in the light of water scarcity forecasts for the region, makes the reliable quantification of water resources a priority. Contemporary approaches to the problem of hydrological prediction in ungauged basins in the region have relied heavily on calibration against a limited gauged streamflow database and somewhat subjective parameter regionalizations using areas of assumed hydrological similarity. The reliance of these approaches on limited historical records, often of dubious quality, introduces uncertainty in water resources decisions. Thus, it is necessary to develop methods of estimating model parameters that are less reliant on calibration. This thesis addresses the question of whether physical basin properties and the role they play in runoff generation processes can be used directly in the estimation of parameter values of the Pitman monthly rainfall-runoff model. A physically-based approach to estimating the soil moisture accounting and runoff parameters of a conceptual, monthly time-step rainfall-runoff model is proposed. The study investigates the physical meaning of the model parameters, establishes linkages between parameter values and basin physical properties and develops relationships and equations for estimating the parameters taking into account the spatial and temporal scales used in typical model applications. The estimationmethods are then tested in selected gauged basins in southern Africa and the results of model simulations evaluated against historical observed flows. The results of 71 basins chosen from the southern African region suggest that it is possible to directly estimate hydrologically relevant parameters for the Pitman model from physical basin attributes. For South Africa, the statistical and visual fit of the simulations using the revised parameters were at least as good as the current regional sets, albeit the parameter sets being different. In the other countries where no regionalized parameter sets currently exist, simulations were equally good. The availability, within the southern African region, of the appropriate physical basin data and the disparities in the spatial scales and the levels of detail of the data currently available were identified as potential sources of uncertainty. GIS and remote sensing technologies and a widespread use of this revised approach are expected to facilitate access to these data.

Preliminary work was conducted to highlight the dominant risks and areas which elicited higher numbers of MSDs within the chosen automotive plant. An area of concern was highlighted through medical record analysis and the use of risk assessment tools, thereby prioritising the need for ergonomic intervention. In particular, the effects of varying restricted and overhead work heights on the biomechanical, physiological and psychophysical responses of an individual were investigated. Twenty-eight subjects were required to complete sixteen conditions. The conditions consisted of the adoption of restricted and upright overhead static postures, with half requiring the holding of four kilograms of weight in the hands and the remaining eight conditions having no weight. Testing was carried out using an electromyography unit, ergospirometer and a perceptual Body Discomfort Map and Scale. This involved a habituation and testing session. The results of the testing revealed the biomechanical and physiological responses were dependant on the change in height. Body discomfort was also shown to be variable over the changing height conditions. This indicates that there is a significant effect of height on an individual’s responses during overhead work. The extreme restricted (-200mm and -100mm) and upright (+300mm and +400mm) overhead conditions within this study were limiting, as they elicited the highest muscle activation, physiological responses and body discomfort ratings. Positions that are preferable to adopt, which were identified from the results in this study, indicate conditions closer to head height (0mm and +100mm) were favourable. The results therefore illustrate how awkward working postures during work are likely to elicit higher demands from an individual, which could lead to an increased risk for the development of a musculoskeletal disorder.
