- Description: 1. The external morphology of all stages in the life cycle of B. decoloratus is described with the aid of scanning electron micrographs. 2. The biological activities of B. decoloratus during its parasitic cycle are examined with special reference to the course of feeding of all stages, and to the influence of the pharate nymph and adult conditions in accelerating the completion of the cycle. 3. The pattern of dropping of replete females is considered in relation to their weight and their time of drop-off. 4. The adult male population structure is described. Two distinct weight groups are reported and the taxonomic significance of these is discussed. 5. The preoviposition period duration of B .decoloratus was studied under laboratory conditions and found to be temperature dependent and humidity independent. Female size, as implied by their engorged weights, influenced the duration of the oviposition period: ticks weighing less than 20 mg possessed longer preoviposition periods. 6. The preoviposition period of B.decoloratus females under fluctuating macroclimatic conditions was studied and found to be long in duration. The implications of this are discussed in relation to laboratory findings and the summer build-up of this species in nature. 7. The duration of the oviposition period of B. decoloratus and the number of eggs produced during this period were studied under laboratory and field conditions. Both these parameters were found to be temperature dependent and uninfluenced by humidity. The significance of a temperature dependent oviposition period is discussed. 8. The minimum amount of 'blood' required by female ticks for oviposition of eggs was found to be approximately 16 mg. under laboratory conditions. 9. The oviposition behaviour of B. decoloratus females has been described and the water relations of ovipositing and non-ovipositing females were examined and discussed . 10. The effects of handling female ticks during their oviposition period was studied and females which were handled were found to produce significantly fewer eggs. The significance of this is discussed. 11. During preliminary experiments on the incubation period of B. decoloratus it was found that the viability of eggs laid during the first half of the oviposlting period was greater than for eggs laid in the second half of the period. The critical temperature of the 'wax' coating of eggs was found to be 42⁰C while the critical humidity was approximately 70% R.H. (7,53 mmHg) at 26⁰C. The implications of these findings are discussed. 12. The development of eggs, traced throughout the incubation period by monitoring the build-up of guanine spectrophotometrically, was studied under different humidity conditions . Successful development was found to be dependent on the water content of the eggs at the time of laying. As eggs were also found to be unable to take up water vapour from the atmosphere this finding is important evidence to support the suggestion that the survival of the egg stage largely determines the spacial distribution of larvae in the field. 13. Results of work done at constant, alternating and naturally fluctuating temperatures and humidities demonstrated that the duration of the incubation period of B.decoloratus is temperature dependent and humidity independent. The significance of this is discussed in relation to the findings of previous workers and to the biology of the species. 14. Egg viability (percentage hatch) was found to be humidity dependent and temperature independent. The implications of these findings are discussed. 15. Some of the problems associated with the assessment of the effects of climatic conditions on ticks are discussed. Special reference has been made to the advocation of using saturation deficit instead of relative humidity as the parameter indicative of environmental humidity in any biological study involving fluctuating humidity conditions. 16. The problems associated with having three variables (e.g. temperature, saturation deficit and incubation period duration) are discussed and reasons given for why contour diagrams have been used in this investigation. 17. A model of the interactions between the main abiotic factors of the environment (temperature and humidity) and the biological parameters studied during the second part of this investigation is presented. 18. The effects of climatic conditions on the life cycle and distribution of B.decoloratus are discussed. Special reference has been made to the available evidence explaining the summer build-up in tick numbers known to occur in nature and the way in which the present findings have helped in an understanding of the spacial distribution of B.decoloratus in the field.

- Description: A range of chromone-2-carboxylic acids has been prepared by condensing suitably substituted 2-hydroxyacetophenones with diethyl oxalate. pK₂ Studies of these acids revealed that 6- or 7-methoxy substituents decreased acidity while the 6-nitro group enhanced acidity; the strongest acid was the 3-chloro derivative, the increase in acidity being attributed to steric inhibition of acid-weakening delocalisation between the carboxyl group and the chromone system. Various chromone-2-carboxamides, derived from acid chloride precursors, were converted to polysubstituted acrylamides by nucleophilic ring-opening with selected amine nucleophiles. The main fragmentation patterns exhibited by these acrylamides were elucidated using a combination of low resolution, high resolution and meta-stable peak analysis, while the effect of substituents on the simultaneous internal rotation involving the carboxamide and enamine moieties were studied using dynamic NMR spectroscopy. Rotational barriers of ca. 67.1 kJmol ̄¹ and ca. 102 kJmol ̄¹ were found for the enamine and amide rotors, respectively. Several synthetic pathways were followed to prepare a series of 2-(N,N-dialkylamino)chromones which were subjected to detailed mass spectral analysis. In addition to substituent-specific fragmentations , the 2-aminochromones appear to fragment via 3 major pathways. The effect of substituents on the internal rotation of the amino moeity was investigated by variable temperature ¹H NMR spectroscopy and the resulting DNMR data was used to calculate the rotational barriers. Examination of the data reveals that the electron-releasing 6- and 7- substituents reduce the C-NMe₂ rotational barrier to ca. 43.5 kJmol ̄¹ , while the nitro analogue has the largest rotational barrier (ca. 46.1 kJmol ̄¹) because of the electron-withdrawing effect of this substituent.

The gendered appropriation of the mobile phone for online health information by youths in Zimbabwean tertiary learning institutions

- Description: The study uses domestication of technology and Cultural Studies theories to investigate how youth in three institutions of tertiary learning in Harare, Zimbabwe are accessing health-related information online using their smart phones. The study critically examines how youths deploy these digital media technologies to construct their identities in a context where social and political power is unevenly distributed. To understand these issues, the study uses a triangulated research design comprising a social survey, in-depth individual interviews and field observation for data collection. Results from this data gathering showed that the use of digital media technologies differ across gender, at least in terms of the distribution of online health seeking practices between male and female youths. According to the survey results, more women tend to use their mobile phones to access health-related information. Data from the individual in-depth interviews showed that the most significant site of social change (social disruption) relates to how digital media facilitates emerging new identities (i.e. identity in the broader sense as well as health-related and sexual health identities). For both young men and young women, mobile phones are used precisely for image management, peer acceptance and the desire to define respective feminities and masculinities within their social networks. Both male and female youths assert they are able to access a variety of information online, and some of this information would not easily accessible when sought from traditional structures such as their parents. This way, youth feel that digital media technologies allow them to cultivate their own preferred identities outside the purview of parental authority and social control. Drawing on postmodern literature, such emerging identities are predicated on cultural volatility, unpredictability, decentralisation and refusal to fixation and conformity to socially constructed identities about being a youth, or being a young Zimbabwean woman or man, for example. Thus, the use of mobile phones and mobile Internet by youths in Zimbabwe to access health-related information has sociopolitical significance, because it allows young people to fashion preferred identities that resists entrenched regimes of social power. For young people in Zimbabwe, online health seeking practices precisely reflects attempt towards negotiating with and circumvent the structural limitations of either an expensive health care system or the general curiosity associated with growing up. That way, it is arguable that youth use digital media technologies to help them exercise some level of social autonomy and agency in dealing with everyday life. Individual in-depth interviews demonstrated that mobile phones and mobile Internet can thus be seen as opening up more spaces for youth to learn more about issues about growing up, sexuality and adolescents that a conservative society such as Zimbabwe traditionally consider as ‘inappropriate’ for youth consumption. They argued that the inability of parents to discuss with them issues about growing up often result in them “finding out on our own” using digital media technologies to satisfy the desire to wean themselves from what they view as arbitrary and asymmetrical social power. The study demonstrated that youth use the mobile phone to challenge the social world of adults and to show resistance to it, thereby strengthening a subculture as well as constructing an identity. However, despite the positive attributes of social capital, connectivity and personalised experience afforded by the mobile phone, the mobile phone is sometimes a source of conflict in relationships between young men and women; that is, between unmarried partners. Young women reported that their boyfriends often force them to disclose who they communicate with using their mobile phones. They also reported that their boyfriends often did some random surveillance of their social media contacts and activities. Thus, whilst one of the mobile phone’s most powerful attribute is its ability to offer personalised experience, as well as offer synchronised and unlimited access to distant connections, results from in-depth interviews showed that some unwritten expectations and norms dictated that young men closely watch their ‘girlfriends’ social media activities, including their online search activities. As a result, privacy as well as the much touted relationship between mobile phone and women’s autonomy becomes contested arenas. Even at young ages, and before marriage, women are socialised to show subservience to their partners by allowing them access to their mobile phones.

The role of the queen in wax secretion and comb building in the Cape honeybee, Aps mellifera capensis (Escholtz)

- Description: The role of the queen in wax secretion and comb building was studied in the the Cape honeybee, Apis mellifera capensis (Escholtz). The percentage of bees bearing wax and the amount of wax borne by these bees did not differ between the experiments. This meant that the queenless and queenright colonies had the potential to construct equal amounts of comb as the amounts of wax available for comb building was the same. Contrary to this prediction, queenright colonies constructed 8 times more comb than their queenless counterparts. Queenright Apis mellifera scutellata colonies constructed 4 times more comb than their queenless counterparts. The increased amount of 9-oxo-2-decanoic acid (90DA) in the A.m.capensis mandibular gland secretions could not alone account for this difference. In fact, A.m.capensis and A.m.scutellata colonies constructed similar amounts of comb when they were given their own queens or queens from the other race. Worker bees need to have direct contact with their queen for comb building to be enhanced. Even when the queen had her mandibular glands extirpated and tergite glands occluded large amounts of comb were constructed than when access to the queen was limited. Direct access to the head of a mated queen proved to be the stimulus enhancing comb building. No comb was constructed when the workers had access to the abdomen of the queen. Virgin queens did not stimulate comb building. The relatively large amounts of 90DA and 9HDA from the mandibular glands of Cape virgin queens had not influenced comb building. Pheromones from other glands on the head may contribute towards the enhancement of comb building, and they are not present in virgin queens

- Description: The role of the queen in wax secretion and comb building was studied in the the Cape honeybee, Apis mellifera capensis (Escholtz). The percentage of bees bearing wax and the amount of wax borne by these bees did not differ between the experiments. This meant that the queenless and queenright colonies had the potential to construct equal amounts of comb as the amounts of wax available for comb building was the same. Contrary to this prediction, queenright colonies constructed 8 times more comb than their queenless counterparts. Queenright Apis mellifera scutellata colonies constructed 4 times more comb than their queenless counterparts. The increased amount of 9-oxo-2-decanoic acid (90DA) in the A.m.capensis mandibular gland secretions could not alone account for this difference. In fact, A.m.capensis and A.m.scutellata colonies constructed similar amounts of comb when they were given their own queens or queens from the other race. Worker bees need to have direct contact with their queen for comb building to be enhanced. Even when the queen had her mandibular glands extirpated and tergite glands occluded large amounts of comb were constructed than when access to the queen was limited. Direct access to the head of a mated queen proved to be the stimulus enhancing comb building. No comb was constructed when the workers had access to the abdomen of the queen. Virgin queens did not stimulate comb building. The relatively large amounts of 90DA and 9HDA from the mandibular glands of Cape virgin queens had not influenced comb building. - Description: Breast cancer is a leading cause of cancer death in Africa. Hsp90 has been identified as a target for anti-cancer treatments as its inhibition results in the disruption and ubiquitin–proteasome degradation of activated oncoproteins. Currently, there are no US Food and Drug Administration approved Hsp90 inhibitor drugs and existing Hsp90 inhibitors such as geldanamycin and novobiocin are hepatotoxic and display a low affinity for Hsp90, respectively. Therefore, there is a need for the development of Hsp90 inhibitors with improved inhibitory properties. In this study twelve natural compounds bearing a quinone nucleus were screened and characterised for the modulation of Hsp90. The compounds analysed formed three series; the sargaquinoic acid (SQA), naphthoquinone, and pyrroloiminoquinone alkaloid series. Certain compounds exhibited half maximal inhibitory concentrations of between 3.32 μM and 12.4 μM, while others showed no antiproliferative activity at concentrations of up to 500 μM in the MDA-MB-231 breast adenocarcinoma cell line. Immunofluorescence and Western analyses indicated that the modulation of Hsp90 and partner proteins by SQA was more similar to that of novobiocin. Isothermal titration calorimetry analyses suggested that SQA interacted with Hsp90β with a low affinity, and saturation-transfer difference nuclear magnetic resonance confirmed that this interaction with Hsp90β occurred through the methyl moiety bound to 1, 4 benzoquinone of SQA. Pulldown assays indicated SQA disrupted the association between Hsp90 and Hop dose-dependently, more similarly to novobiocin. Immunofluorescence and Western analyses performed on naphthoquinone and pyrroloiminoquinone alkaloid compounds indicated modulation of Hsp90 and Hsp90 partner proteins by the compounds. Naphthoquinone compounds were prioritised for analysis for binding to Hsp90β over the pyrroloiminoquinone alkaloid compounds. Lapachol interacted with Hsp90β with a low affinity however; this interaction was thought to be too weak to disrupt the association of Hsp90 and Hop. The remaining naphthoquinone compounds showed no interaction with Hsp90β, thus allowing the determination of a preliminary structure-activity relationship for these compounds.
