- Description: Aimed at replicating the results of an American study into "Burnout" in Group Home houseparents, this research had the following aims: 1) To ascertain the validity and reliability of the instrument used in the original study and presented as a "useful measure of burnout”. 2) To establish whether this line of research, within a highly problematic research field, can at present offer any guidelines in the resolution of the current staffing crisis faced by South African children's homes. 3} To describe more closely the burnout syndrome. 4) To study possible etiological factors within a local context. Sixty three houseparents completed questionaires and three independent measures of burnout were obtained. The results were regarded as having failed to replicate those of the original study. The reason for this was found to be the low validity of the original instrument. The main conclusion drawn was that the line of research adopted in the original study can offer only very tentative guidelines towards the resolution of the staffing crisis faced until such time as valid and reliable instruments to measure burnout have been developed. - Description: [Excerpt taken from Introduction] The purpose of this study is to take a look at one of these African leaders, inkosi Albert Luthuli through a biographical lens to assess whether he should be recognised as an African intellectual. Sifiso Mxolisi Ndlovu states that inkosi Luthuli is recognised as the father of South Africa’s non-racialism. He used his moral authority in a historic fashion to influence the liberation movement to adopt non-violent resistance. During his time as President-General, he became the beacon of non-violent resistance. As the president of the liberation organisation, he delivered speeches that steered the African National Congress (ANC) and the liberation movement when the State escalated its oppression against Africans. The State retaliated by deposing him as an elected Chief, imprisoned him, imposed multiple bans on him in attempts to silence him. His intellect proved to be a threat to the State. He spoke out boldly against the apartheid state and advocated for chiefs, African people, African women, sugar farmers, and all oppressed racial groups. Inkosi Luthuli used his speeches to deliver political concepts like non-racialism, multiracialism, African nationalism and democracy into the public space. He cemented ANC’s cooperation policy that created the environment for the existence of the Congress Alliance that produced the Freedom Charter. He spoke out against the oppression of not just South Africans but Africa and all oppressed groups internationally. He illustrated that he possessed geopolitics that would gain the attention of the world. He illustrated his geopolitics through his internationalism philosophy gained the international community’s attention. Inkosi Luthuli was revered and respected by his Groutville community, the African community, South Africans of all racial groups and the international community. His impact can be seen through him being the first African-born Nobel Peace Prize recipient. A biological assessment of the Cape knifejaw (Oplegnathus conwayi) an endemic South African teleost

- Description: The South African spearfishery forms a small component of South Africa’s complex coastal fishery. Although the smallest, this fishery sector has the largest proportion of data deficient species, and as a consequence is poorly managed. Oplegnathus conwayi (Cape knifejaw) is an example of one of these species for which there is very little and outdated biological and ecological data. This species is among the top target species by recreational spearfishers in South Africa. This thesis aimed to improve our socio-ecological knowledge of the South African spearfishery through the collection of biological and Fishers’ Ecological Knowledge (FEK) information on Oplegnathus conwayi, and human dimension information on the South African spearfishery. Monthly biological samples were collected through routine sampling (spearfishing) and augmented by recreational spearfishers. A total of 254 fish were collected from the warm-temperate coastal zone (East London – Cape Agulhas), 28 from the sub-tropical zone (Durban – East London) and 23 from the cool-temperate zone (Cape Agulhas – Cape Point). The results indicated that Oplegnathus conwayi is a slow-growing species, with a maximum recorded age of 27 years. The population sex ratio was skewed, with males dominant in the population (1M:0.6F). The length- and age-frequency distributions were, however, similar for both sexes. No significant differences were observed between male and female growth (LRT, p > 0.05), with the overall population growth curve being best described as L(t) = 697.15(1-e-0.06(t-6.30)). Males matured at a slightly larger size than females, however, no significant differences were observed (LRT, p > 0.05). The length- and age- at-50% maturity was 330 mm (FL) and 5.73 years for the full population, respectively. Histological analyses showed that Oplegnathus conwayi are asynchronous spawners with a gonochoristic reproductive style. Macroscopic staging and gonadosomatic index results indicated a protracted spawning season for Oplegnathus conwayi, with a peak in spring. A survey was designed and disseminated to collect FEK on the biology and population status of Oplegnathus conwayi and human dimension information on South Africa’s spearfishery. A total of 103 survey responses were received, of which 94 were regarded as specialised (spearfishers who had greater experience, skill and avidity, and maintained spearfishing as an important component of their lifestyle) spearfishers. Based on the responses of the specialist spearfishers, the top four main species caught by spearfishers from this survey were Seriola lalandi (13.9%), Pachymetopon grande (11.7%), Oplegnathus conwayi (11.4%) and Sparodon durbanensis (11%), and the majority of respondents indicated that there had been no changes in abundance, size and catches of these species in the years that they had been spearfishing. Respondents indicated that Oplegnathus conwayi are most commonly targeted in the Eastern Cape and are found at depths of up to 40 m. Respondents also indicated that there may be a seasonal onshore (Summer/Winter) and offshore (Summer/Winter) migration with year-round spawning and a peak in November, December and January. The incorporation of spearfishers into the data collection, both through the collection of specimens and their FEK, was beneficial to this study. Besides providing samples from a broader geographical range than the primary collection area, the collaboration with spearfishers has promoted the inclusion of this group into the management system. The findings of this study also suggest that FEK data can be more reliable if the concept of recreational specialisation is incorporated into data collection. While the FEK suggested that the population was stable, a stock assessment is necessary to fully understand the population status and implement management strategies. Nevertheless, the key life history characteristics (slow growth and late maturation) observed in this study are characteristic of species that is vulnerable to overexploitation, and thus the precautionary approach should be applied. The reproductive information collected in this study has provided information for the implementation of an appropriate size limit regulation for Oplegnathus conwayi. Here, a minimum size limit of 400 mm TL, which corresponds approximately with the length-at-50% maturity of 330 mm FL, would be appropriate to allow fish to mature and spawn, and reduce the likelihood of recruitment overfishing. Reduction in the bag limit from five to two fish per person per day may also be appropriate as a precautionary measure until a stock assessment has been completed. Respondents also indicated that there may be a seasonal onshore (Summer/Winter) and offshore (Summer/Winter) migration with year-round spawning and a peak in November, December and January. The incorporation of spearfishers into the data collection, both through the collection of specimens and their FEK, was beneficial to this study. Besides providing samples from a broader geographical range than the primary collection area, the collaboration with spearfishers has promoted the inclusion of this group into the management system. The findings of this study also suggest that FEK data can be more reliable if the concept of recreational specialisation is incorporated into data collection. While the FEK suggested that the population was stable, a stock assessment is necessary to fully understand the population status and implement management strategies. Nevertheless, the key life history characteristics (slow growth and late maturation) observed in this study are characteristic of species that is vulnerable to overexploitation, and thus the precautionary approach should be applied. The reproductive information collected in this study has provided information for the implementation of an appropriate size limit regulation for Oplegnathus conwayi. Here, a minimum size limit of 400 mm TL, which corresponds approximately with the length-at-50% maturity of 330 mm FL, would be appropriate to allow fish to mature and spawn, and reduce the likelihood of recruitment overfishing. Reduction in the bag limit from five to two fish per person per day may also be appropriate as a precautionary measure until a stock assessment has been completed. - Description: Non-profit organisations (NPOs) contribute to our society by addressing needs through value-creation activities which are normally not addressed by normal for-profit organisations. These NPOs generate their income through donations, fundraising and market activities such as the sale of products or services. However, in the pursuit of organisational sustainability, these NPOs need to ensure that their income meets their expenditure needs over the long run, to allow continuous value creation and to pursue their mission. An organisation’s business model can be visualised through a business model framework (BMF), which have been illustrated to be useful tools for organisations to test new business models, plan and coordinate activities, and to communicate how an organisations business model works to stakeholders. The use of BMFs by NPOs may be useful for similar purposes. However, a key challenge for NPOs and their management is adapting for-profit tools to be more applicable to the NPO environment. Whilst there has been some progress within the literature on adapting these for-profit business model tools to NPOs, this area of research is relatively underrepresented in the literature. The single case study method, exploratory in nature, following a deductive approach with a theoretical framework was used to evaluate the NPO Business model framework for NPOs with multiple income streams (Sanderse, 2014) appropriateness for the Centre for Biological Control (CBC), a mission-driven research organisation located within Rhodes University. The study made use of an interpretivist paradigm through the lens of resource-based theory. The contextual environment of the CBC was similar to other NPOs in that their organisational sustainability was at risk due to uncertainty about the availability of long-term funding. The CBC is actively diversifying their income streams by establishing new partnerships, which increase the capacity and resource base of the organisation to improve its flexibility in meeting current and potential funders’ needs. The NPO Business model framework for NPOs with multiple income streams was found to be a suitable BMF to visualise the holistic operations of the CBC. However, the role of governance-related matters being represented in this BMF was found to be lacking. An amended framework including governance as part of the Business model framework for NPOs with multiple income streams was developed. This improved the BMF, by allowing for further contextual insight into the CBCs business model as governance played a major role in the key activities and income-generating strategies they participate in. The role a BMF has in assisting an organisation with resource management activities was also explored. A suitable BMF, which allows for a holistic and explicit display of an organisation’s business model, would assist in determining how resources could be better managed or orchestrated to pursue different potential income-generating strategies or improve the efficiency of how resources are used in the current business model. This study contributed to business model theory by further testing the Business model framework for NPOs with multiple income streams as a tool to visualise an NPOs business model and identifying that once the governance block was added, this BMF became more suitable within the CBCs context. Groen Sebenza is an environmental education ‘incubator’ programme driven and implemented by the South African National Biodiversity Institute (SANBI) to unlock green jobs and bridge the gap between education and job opportunities in the biodiversity sector in South Africa. The programme is a key intervention to strengthen biodiversity human capacity development in the biodiversity sector in South Africa, seeking to contribute to transformation of the biodiversity sector, and also address issues of youth unemployment in the country. The young ‘interns’ in the programme were called ‘pioneers’ at the start of the project. This research project explores how a host institution operating as a community of practice within a landscape of practice managed to implement the Groen Sebenza programme by absorbing and appointing all their pioneers into sustainable jobs beyond the pilot project. I sought to better understand the process of supporting and empowering unemployed youth into sustainable green jobs within the Groen Sebenza partnership programme. I drew on Community of Practice (CoP) theory, and its value creation framework to develop this understanding, and I under-laboured the analysis with a social realist analysis of enabling and constraining factors. The unit of analysis of a Community of Practice was a useful focus for the study, as these mentors, managers, and administrators were all involved in supporting the empowerment and retention of the young pioneers in the host institution. To develop deeper insight into the learning and knowing, and value created in and by the Groen Sebenza CoP in the Host Institution, I also sought insight into enabling and constraining factors and how these shaped and contributed to empowerment and retention of the pioneers in sustainable green jobs. The research addressed the main question of ‘How do processes of learning, knowing and value creation contribute to empowerment and retention of unemployed youth in a successful Host Institution in the Groen Sebenza programme, and what enabled or constrained the empowerment and retention processes and outcomes?’. Three sub-questions were used in the study, which focussed on the mentoring, training and workplace experiences and how they contribute to the process of learning and knowing within the Groen Sebenza Community of Practice in the Host Institution? [Addressed in Chapter 4], the value creation elements that emerged in the implementation of the programme in support and empowerment of the pioneers in the Host institution’s Groen Sebenza CoP? [Addressed in Chapter 5], and the enabling and constraining factors that shaped and contributed to the uptake of the Pioneers into sustainable green jobs at the Host Institution within the Groen Sebenza Programme? [Addressed in Chapter 6]. The research was conducted as a qualitative case study, in which I used semi-structured interviews as a key data source, as well as document analysis, and a questionnaire. The study drew on inductive, abductive and retroductive modes of inference since I sought to explore an understanding of the practices and learning that occurred that contributed and led to the successful uptake of Pioneers into jobs, as well as the enabling and constraining factors. The study was interpretive at the epistemic level, and had a social realist under-labouring at the ontological level. Key findings of the study point to the development of enabling cultures of mentoring in workplaces, and the provision of a diversity of workplace learning experiences including formal training. It also points to the importance of personal emergent properties amongst mentors and pioneers that embrace a willingness to work together and build strong relationships, and to learn together. Learning in the community of practice was shown to develop identity and a sense of belonging as pioneers were given meaningful tasks to do and their training and interactions with mentors was experienced as meaningful and relevant. The contributions of the pioneers to the institutional mandate was appreciated by the mentors and therefore also well supported within an empowerment orientation. Various structural factors contributed to this enabling situation, most notably strong support from management as well as good co-operation across divisions. Constraining factors included the physical distances in the province, as well as financial and technical issues such as poor ICT communication systems. Overall, though the study showed that a strong approach to learning in communities of practice supported by empowering mentoring can lead to the integration of young pioneers into sustainable green jobs in the environmental sector. A whole institution approach to this process is, however, needed, and the organisation needs to develop a culture of social learning. As recently as September 2020 as this study was being finalised, the Presidential Employment Stimulus Plan (Office of the President, 2020) following the initial economic shocks emanating from the COVID-19 pandemic, made yet another commitment to using the Groen Sebenza model to create and support sustainable job creation for young people in South Africa today in the environmental sector. This study has been developed and designed to understand those processes and enabling conditions that can support retention and empowerment of young people to take up jobs in the environmental sector today. - Description: This research investigation is based on continuous improvement changes arising from - and related to - ISO 9001 implementation at Sundays River Citrus Company (SRCC), which is one of the biggest packers, marketers, and exporters of citrus fruit in South Africa. The Board of SRCC adopted ISO 9001 to ensure organizational efficiency and sustainability while improving quality control, customer service, teamwork and leadership. However, research indicates not all organizations that have Implemented ISO 9001 realize the intended benefits. While no research was available on the citrus agriculture industry, research in the tourism industry indicates that not only did some companies not realize the expected benefits, but also incurred substantial investment costs. It is estimated that approximately thirty to ninety percent of change initiatives fail to meet their objectives, and research studies also indicate that middle managers play a critical role that can influence the outcomes of a change project. However, Balogun (2003) indicates that middle managers play a complex role and are exposed to role conflict, which can influence the outcomes of change initiatives. In this context, this research study aimed to investigate the role conflicts experienced by middle managers during the process of an ISO 9001 continuous improvement change. The study draws on role theory, applying it to their management of change. The following role conflict types were investigated: (1) intra-sender conflict, (2) inter-sender role conflict, (3) inter-role conflict, (4) role ambiguity and (5) role strain. The research approach is qualitative, and has adopted a post-positivist paradigm, utilizing a deductive qualitative method. The study adopted a case study approach. Data was gathered mainly from interviews and supported by organizational documents. Semi structured interviews were conducted with questions formulated through the use of the coding manual (See Appendix C) to ensure alignment of data collection with the research propositions derived from literature. A deductive thematic analysis method was used to analyze the interview data. The research findings confirmed that during continuous improvement change, as middle managers strived to satisfy the incompatible expectations of role senders (mainly senior and junior managers), they experienced the five role conflict types. The study findings also indicate that middle managers experience conflicts due to the incompatible expectations of other role senders such as quality and marketing departments. The findings suggest that middle managers are managing these conflicts, but notes that they do require some assistance and support from senior management. The study showed that the wind farm unintentionally applies some components of responsible leadership among its stakeholders. Openness, transparency, and communication are critical actions that this organisation undertakes to cultivate trust among both its internal and external stakeholders. External stakeholders experience this organisation differently. There are mixed feelings regarding how the wind farm conducts its stakeholder engagement activities, particularly relating to landowners and government institutions. The T. brucei genome encodes three copies of mtHsp70 which are 100% identical. MtHsp70 self-aggregates, a property unique to this isoform, and an Hsp70 escort protein (Hep1) is required to maintain the molecular chaperone in a soluble, functional state. This study aimed to compare the solubilizing interaction of Hep1 from T. brucei and Homo sapiens (H. sapien). The recently introduced Alphafold program was used to analyze the structures of mtHsp70 and Hep1 proteins and allowed observations of structures unavailable to other modelling techniques. The GVFEV motif found in the ATPase domain of mtHsp70s interacted with the linker region, resulting in aggregation, the Alphafold models produced indicated that the replacement of the lysine (K) residue within the KTFEV motif of DnaK (prokaryotic Hsp70) with Glycine (G), may abrogate bond formation between the motif and a region between lobe I and II of the ATPase domain. This may facilitate the aggregation reaction of mtHsp70 orthologues and provides a residue of interest for future studies. Both TbHep1 and HsHep1 reduced the thermal aggregation of TbmtHsp70 and mortalin (H. sapien mtHsp70) respectively, however, TbHep1 was ~ 15 % less effective than HsHep1 at higher concentrations (4 uM). The HISIG-CF is developed from literature and qualitative data collected using an expert reviews method from practitioners in the healthcare sector who evaluated the constructs of the HISIG-CF. - Description: The research presented in this thesis is based on my interest and experience in forms of African music, ethnomusicology, and studying mbira with Professor Emeritus Andrew Tracey. When I arrived in South Africa in 2019 to join Rhodes University’s Applied Ethnomusicology programme, I chose to study the mbira with Tracey as the idea of learning more about Zimbabwe through music was important to the formation of my identity. Through the lens of embodied learning and a practice-based approach in this research, I evaluate how Tracey’s numerous contributions to African music pedagogy have improved prospects for African music scholars and students in terms of contributing to the goals of applied ethnomusicology. The primary purpose of this thesis is to respond to the absence of serious scrutiny of existing pedagogical approaches to African music at universities across South Africa. The contribution this research makes will be valuable to African music programmes across the continent as well as to practitioners of African traditional instruments, such as the marimba, mbira, timbila xylophones, nyanga pan pipes, and valimba xylophones. The thesis comprises five chapters. The first presents an introduction to the research, and its goals, procedures and approaches, along with an outline of the subsequent chapters. Tracey’s biography is covered in the second chapter. A consideration of the state of African music teaching in other African countries such as Ghana, Kenya, and Zimbabwe; the history of African music; and the state of African music pedagogy in tertiary institutions in South Africa constitutes the third chapter. Chapter 4 comprises an analysis of Tracey’s articles and data gathered from interviews, as well as my personal reflections as Tracey’s student. The final chapter presents a summary of the preceding chapters, the study’s findings, and suggestions for further research. A multidisciplinary approach was used for this thesis. The results finds that Tracey’s articles had six common themes which he wrote about and are a contribution to African music pedagogy. - Description: The aim of the present research was to investigate whether, as claimed by the South African Revenue Service in the media release issued in April 2018, the normal income tax provisions could apply to cryptocurrency transactions. To achieve this aim, a literature review was undertaken to describe the nature of cryptocurrencies and related crypto mining activities, providing definitions of cryptocurrencies, blockchains and crypto mining, as well as describing the functioning of the system. The research then proceeded to analyse the provisions of the definition of “gross income” in section 1 of the Income Tax Act, 58 of 1962, as amended, and the requirements of the “general deduction formula” in terms of the preamble to section 11, section 11(a) and section 23(g), as applying to cryptocurrency transactions. The application of other provisions in the Act to cryptocurrency transactions was analysed, including trading stock in terms of section 22, and capital allowances in terms of sections 11(e), 12C and 13quin of the Act, together with capital gains tax consequences in terms of the Eighth Schedule to the Income Tax Act. The regulation for income tax purposes of cryptocurrency transactions in Australia was discussed, with a view to making similar recommendations in South Africa. The research was situated in the interpretative paradigm, a doctrinal methodology was applied, together with a qualitative analysis of documentary data. The discussion was limited to the income tax consequences of cryptocurrencies as applying to individuals. - Description: This thesis interrogates the relationship between the Dispute Settlement Understanding and Africa Continental Free Trade Area dispute settlement mechanism, in light of the conflict of jurisdiction. The conflict of jurisdiction is an adverse effect of the fragmentation of international law. The uncoordinated proliferation of international treaties has increased occurrences of overlapping memberships and overlapping subject matter regulation amongst treaties. Whenever the overlaps mentioned above exist, and a dispute arises concerning matters of overlap, that dispute can be heard in more than one tribunal, giving rise to a conflict of jurisdiction. Jurisdictional conflicts are a problem because they breed uncertainty in the adjudication of disputes; they increase the risk of forum shopping, conflict of rulings, protracted litigation, and waste resources. There is a significant risk for jurisdictional conflicts between the World Trade Organisation and Africa Continental Free Trade Area agreements, because of membership and subject matter overlaps. To mitigate the problems caused by jurisdictional conflicts, the Africa Continental Free Trade Area agreement has incorporated a fork-in-the-road clause. Fork-in-the-road provisions allow parties to choose their preferred forum, and once the forum is chosen, the parties are prohibited from bringing the same dispute to another tribunal. Unfortunately, fork-in-the-road clauses are insufficient in resolving jurisdictional conflicts because they do not bind the Dispute Settlement Understanding. It is only bound to enforce World Trade Organisation obligations and not non-World Trade Organisation obligations. The extent to which non-World Trade Organisation norms apply in the Dispute Settlement Understanding is unsettled, making it difficult to conclude whether a fork-in-the-road provision will be effective an effective solution to potential jurisdictional conflicts. In this thesis, the researcher investigates the prospects of the World Trade Organisation applying the AfCFTA fork-in-the-road clause, directly, as a potential solution to the conflict of jurisdiction. In addition, the researcher will also investigate an alternative means of applying the AfCFTA fork-in-the-road provision, indirectly, using the World Trade Organisation procedural good faith provisions. A distributed approach to leadership in an academic department in a South African university: an exploratory case study

However, due to the demanding nature of such a position, it is not sought-after, as it remains exclusive and unappealing to many academics. Moreover, it is a position that does not encourage inclusive leadership. Tension and role ambiguity are known to arise between the scholarly project on the one hand and management and administrative matters on the other. To address challenges associated with this singular leadership position, an academic department at a South African university adopted a distributed leadership approach in their department as a research experiment. My study was based on this research experiment. I designed this research as an exploratory case study, guided by a socio-cultural conceptualisation of distributed leadership that included a leader-plus and a practice aspect, defined as a product of the interactions of school leaders, followers and their situation. This qualitative case study aimed to describe and explain how a distributed leadership approach was understood and practised in an academic department. It also investigated the enablements and constraints of the approach. Data were generated through document analysis, observation, and individual and focus group interviews. The participants in this study included the departmental leadership team and the department’s academic and administrative staff. Unfortunately, my study took place during the global COVID-19 pandemic and national lockdowns. I had no choice but to adapt my data generation methods due to lockdown restrictions. As a result, most data generation was done via online communication. The study used inductive and abductive analysis to make the data meaningful to the reader. The findings revealed that different participants had different ideas about distributed leadership. Distributed leadership was understood as a socio-cultural practice rather than an individual practice with multiple leaders in relational practice. In addition, this approach was understood as a way of developing and sharing expertise by encouraging teamwork, collegiality and collective decision-making which aligns with the notion of democratic decision-making which creates a platform for the enablement of leadership in others. The study further explored how distributed leadership was practised. The findings were that the HOD position, usually a one-person role, was reconceptualised as a HOD team comprised of three academics and the departmental administrator. The HOD team divided the work among themselves, and this was done according to each individual’s expertise. In addition, leadership within the academic department was not limited to the HOD team but stretched across the department; thus, multiple leaders were evident. Therefore, based on the data, this study discovered that a distributed approach values leadership expertise in others. Consequently, it can be used to promote an inclusive environment in which any organisation member can lead. Inclusivity in the decision-making process was also regarded as a strong practice in the academic department. As a result, this study contends that those in formal positions can develop leadership in others through a distributed leadership approach. Through that, lecturer leadership was enacted through formal faculty and university structures and informally as and when the situation required it. The enablements of this departmental leadership approach included the structural innovation of the Friday check-in as well as the buddy system. These two innovations provided the space for collegiality and the development of voice and leadership. The consultative nature of the HOD team was also viewed as an enablement. Certain factors constrained the distributed leadership approach, one of which was the tension between the hierarchical structure of the university and the more horizontal, distributed leadership approach being piloted in the academic department. In addition to that, another constraint was also very real with the transition to Zoom as an online teaching, learning and supervision platform as a consequence of the COVID-19 pandemic. The final constraint was experienced concerning the department’s history, as it was within a historically White university in South Africa. The effects of this history impacted the departmental culture and, as we know from the literature, institutional culture is extremely difficult to change. Finally, the study concluded that conceptualising distributed leadership as a sociocultural practice with leader-plus and practice aspects provides descriptive language and a solid theoretical and analytical framework for a distributed leadership study. The study makes an important knowledge contribution in the African Higher Education context as limited research has been carried out in this area. This study used Action Design Research to develop a guide based on both theory and the results gathered from the educator participants at a local secondary government school in Makhanda, South Africa. The finalised guide presents recommendations to educators, principals, and the DBE to promote acceptance and adoption of technology in the classroom and for distance learning. These recommendations include the provision of data, technological training and pedagogical support, a functioning community of practice, an online learning community of practice, and support from the DBE. - Description: Background: Sunglasses are popularly worn by outdoor athletes such as cricketers. They are primarily worn to preserve ocular health due to the length of time exposed to the sun’s glare on the field. More recently, sunglasses are now worn for their purported performance-enhancing benefits supposedly allowing athletes to “see clearly, react faster and perform with confidence”. Fielders typically wear sunglasses, but it is rare to see the bowler or the batter wearing sunglasses although they are exposed to the same conditions. If sunglasses provide visual clarity for better reaction time, it is assumed this would also be beneficial for both batsmen and bowlers alike. It is interesting that although fielding plays a key role in the winning of a match, it has received very little research in comparison to the areas of batting and bowling. Research concerning sunglasses in the past has been concerned with its effect on ocular parameters such as colour vision and contrast sensitivity, with minimal research on their effect on performance, specifically on how different colour tints may affect performance such as catching in fielding. Objective: The purpose of this study was to compare the catching performance of fielders when wearing three different colour tints of sunglasses. Methods: Thirty male cricketers currently playing for Rhodes University or country districts sides in Makhanda, South Africa, who were non-habitual sunglass wearers were recruited for this study. An optician administered a pre-screening test to ensure participants did not have any visual defects that would affect the results, determining who was eligible to continue with the research project. Twenty-five participants qualified with a further four withdrawing during the experimentation phase due to personal reasons. Eligible participants were required to complete four testing sessions on separate days thereafter in which they wore a different colour tint at each session (clear, blue, G30 and red). These tints were randomised among participants. Participants were required to catch 18 balls projected from a bowling machine in each session. The number of balls caught as well as the quality of catch was recorded. At the end of the fourth session, participants were presented with a questionnaire investigating their subjective experience of the protocol and preference for any particular colour of lens. Results: Statistically significant results were found between the different colour lenses for the ocular parameters of contrast sensitivity and stereopsis tested by the optician. The red lens was found to be the best for contrast sensitivity with the blue lens performing the worst on this test. With regard to stereopsis, the G30 lens was the worst, with the red lens performing the best. The performance measures resulted in no significant difference between the different colour lenses both concerning the total number of catches taken as well as the quality of catch taken. The same result was found irrespective of the colour of lens used in each session. Data gathered from the questionnaire regarding visual comfort and target visibility resulted in the red lens being rated statistically worse than the other three conditions. In terms of personal preference, the G30 lens was rated statistically as the best coloured lens. Conclusion: It can be concluded that objectively there is no difference in performance between the three coloured lenses. Although from an ocular perspective, the red lens was expected to perform the best in terms of catching performance, there was no difference. - Description: Heterocyclic structures have found extensive utility in the field of medicinal chemistry, as prominent regions of pharmacophores resulting in numerous drug treatments for many diseases. Accordingly, in this project we explored the respective antimalarial and anticancer activity exhibited by compounds featuring nitrogen containing indole and tetrazole heterocycles respectively. This thesis therefore comprises of two distinct parts. Part 1. Following the development of resistance towards traditional antimalarial therapy such as chloroquine and emerging resistance towards artemisinin combination therapies, the WHO reported the urgent need for new, effective drugs and identification of new drug targets to combat the Plasmodium falciparum parasite. In 2015 the parasite was the cause of 429 000 deaths, the majority occurring in the sub-Saharan region of Africa. This highlights the failing effectiveness of vector control strategies, reiterating the need to develop alternative control and treatment strategies. In response to this need we wanted to expand and further describe the SAR of the indole based series, indolyl-3-ethanone-α- thioethers, previously synthesized in our laboratory. These compounds were found to exhibit antimalarial activity with compounds 2.26 and 2.27 exhibiting activity against P. falciparum 3D7 in the nanomolar range. Based on these compounds we synthesized compounds 3.21 and 3.24 – 3.32 following a three step reaction pathway. Our results in this study, indicate that compound 3.28, a pnitrothiophenol analogue of 2.27 was the most active of the compounds we synthesized and furthermore was superior in activity against Plasmodium compared to 2.27. This result indicated that the presence of p-NO2 is important in enhancing anti-plasmodial activity. Comparing compounds 3.25 and 3.26 with an oxygen on the ether bridge to compounds 3.29 and 3.30 with a sulfur, we observed an increase in hydrophilicity coupled to a decrease in anti-plasmodial activity in the compounds, thus, highlighting the importance of sulfur for enhanced activity. Furthermore, we investigated bioisosteric replacement of the 5-chloro substituent present in hit compounds 2.27 and 3.28, with an electron withdrawing nitrile (3.27) and electron donating methyl (3.29) and methoxy (3.31) substituents. These substituents decreased anti-plasmodial activity, confirming that a chlorine substituent is optimal for biological activity. This study furthered our understanding of the SAR of indolyl-3-ethanone-α- thioethers for the development of potent anti-plasmodial lead compounds. Part 2. Triple negative breast cancer (TNBC), which disproportionately affects women of sub-Saharan Africa, is unresponsive to hormone-based therapies. This emergence presents a population of patients devoid of effective drug treatment, signaling the urgent need to develop new effective therapies with novel drug targets. Therefore, we identified our target in TNBC cells as the protein-protein interaction between the co-chaperones HOP and HSP90. We reasoned that a disruption of this interaction would ultimately result in cancer cell death via the degradation of essential oncogenic client proteins. Following a fragment screening campaign, which identified several acid and tetrazole containing hits (4.56 – 4.58) which bound to HOP, with low anticancer activity, we sought to develop synthetic methodology to elaborate our fragment hits synthesizing tetrazole containing fragments to target TNBC cell lines. We therefore proceeded to synthesize a range of multi substituted fragments (4.59 – 4.63), utilizing a nitrile (4.66) to access tetrazoles via 1,3-cycloaddition and an acid by nitrile hydrolysis. Following the development of resistance towards traditional antimalarial therapy such as chloroquine and emerging resistance towards artemisinin combination therapies, the WHO reported the urgent need for new, effective drugs and identification of new drug targets to combat the Plasmodium falciparum parasite. In 2015 the parasite was the cause of 429 000 deaths, the majority occurring in the sub-Saharan region of Africa. This highlights the failing effectiveness of vector control strategies, reiterating the need to develop alternative control and treatment strategies. In response to this need we wanted to expand and further describe the SAR of the indole based series, indolyl-3-ethanone-α- thioethers, previously synthesized in our laboratory. These compounds were found to exhibit antimalarial activity with compounds 2.26 and 2.27 exhibiting activity against P. falciparum 3D7 in the nanomolar range. Based on these compounds we synthesized compounds 3.21 and 3.24 – 3.32 following a three step reaction pathway. Our results in this study, indicate that compound 3.28, a pnitrothiophenol analogue of 2.27 was the most active of the compounds we synthesized and furthermore was superior in activity against Plasmodium compared to 2.27. This result indicated that the presence of p-NO2 is important in enhancing anti-plasmodial activity. Comparing compounds 3.25 and 3.26 with an oxygen on the ether bridge to compounds 3.29 and 3.30 with a sulfur, we observed an increase in hydrophilicity coupled to a decrease in anti-plasmodial activity in the compounds, thus, highlighting the importance of sulfur for enhanced activity. Furthermore, we investigated bioisosteric replacement of the 5-chloro substituent present in hit compounds 2.27 and 3.28, with an electron withdrawing nitrile (3.27) and electron donating methyl (3.29) and methoxy (3.31) substituents. These substituents decreased anti-plasmodial activity, confirming that a chlorine substituent is optimal for biological activity. This study furthered our understanding of the SAR of indolyl-3-ethanone-α- thioethers for the development of potent anti-plasmodial lead compounds. Part 2. Triple negative breast cancer (TNBC), which disproportionately affects women of sub-Saharan Africa, is unresponsive to hormone-based therapies. This emergence presents a population of patients devoid of effective drug treatment, signaling the urgent need to develop new effective therapies with novel drug targets. Therefore, we identified our target in TNBC cells as the protein-protein interaction between the co-chaperones HOP and HSP90. We reasoned that a disruption of this interaction would ultimately result in cancer cell death via the degradation of essential oncogenic client proteins. Following a fragment screening campaign, which identified several acid and tetrazole containing hits (4.56 – 4.58) which bound to HOP, with low anticancer activity, we sought to develop synthetic methodology to elaborate our fragment hits synthesizing tetrazole containing fragments to target TNBC cell lines. We therefore proceeded to synthesize a range of multi substituted fragments (4.59 – 4.63), utilizing a nitrile (4.66) to access tetrazoles via 1,3-cycloaddition and an acid by nitrile hydrolysis.
