The use of a Roving Creel Survey to monitor exploited coastal fish species in the Goukamma Marine Ptrotected Area, South Africa

- Description: A fishery-dependant monitoring method of the recreational shore-based fishery was undertaken in the Goukamma Marine Protected Area (MPA) on the south coast of South Africa for a period of 17 months. The method used was a roving creel survey (RCS), with dates, times and starting locations chosen by stratified random sampling. The MPA was divided into two sections, Buffalo Bay and Groenvlei, and all anglers encountered were interviewed. Catch and effort data were collected and catch per unit effort (CPUE) was calculated from this. The spatial distribution of anglers was also mapped. A generalized linear model (GLM) was fitted to the effort data to determine the effects of month and day type on the variability of effort in each section. Fitted values showed that effort was significantly higher on weekends than on week days, in both sections. A total average of 3662 anglers fishing 21 428 hours annually is estimated within the reserve with a mean trip length of 5.85 hours. Angler numbers were higher per unit coastline length in Buffalo Bay than Groenvlei, but fishing effort (angler hours) was higher in Groenvlei. Density distributions showed that anglers were clumped in easily accessible areas and that they favored rocky areas and mixed shores over sandy shores. Catch documented between October 2008 and December 2009 included a total of 361 fish, of 27 species from 12 families. Sparidae had the highest contribution (12 species). A Shannon-Weiner diversity index showed that diversity was higher in Buffalo Bay (0.81) than Groenvlei (0.57). Catch composition of retained fish (336 individuals) showed that the six numerically most important species were blacktail (Diplodus sargus capensis) (66 percent of catch), followed by galjoen (Dichistius capensis) at 11 percent, Cape stumpnose (Rhabdosargus holubi), belman (Umbrina robinsonii) and strepie at 3 percent, and elf (Pomatomus saltatrix) at 2 percent. Catch composition of an earlier study in Goukamma (Pradervand and Hiseman 2006) was compared with the present study, as well as data from the De Hoop MPA, which is closed to fishing. A multi-dimensional scaling plot of catch composition showed tight clustering of the De Hoop samples, and high variability among the Goukamma samples. A bray-curtis similarity index and dendrogram of similarity between study sites and study periods showed that there was an 83 percent similarity among De Hoop samples and a 75 percent similarity among Goukamma samples (ignoring the two outliers). The two sites are different with respect to species composition, but this is expected because they are different areas. Differences between time periods in Goukamma (i.e. the previous study versus the present study) were not significant. The most significant result from the catch composition analyses is the high variability among the Goukamma samples. This can be explained by the variable fishing methods used by anglers in Goukamma, compared with the standardized fishing methods used by researchers in De Hoop, and the fact that fish are more abundant and populations are more stable in De Hoop – giving higher sample sizes which reduce the variability in the statistics. Species-specific CPUE was calculated for the six numerically most important species. The Goukamma MPA is a popular fishing destination and angler effort is high. It can be considered a node of exploitation for surf zone fish, for which it provides no protection. Even though the MPA allows shore angling, sustainable fishing practices should be incorporated in management plans if the MPA is expected to protect and conserve its stocks. Of noteworthy concern is the occurrence of illegal night fishing (the public may not enter the reserve between sunrise and sunset) which leads to underestimates of catch and effort (night surveys were not conducted because of safety concerns). It is recommended that more communication should take place between the angling community and the reserve management. Sign boards giving information on species which are under pressure, and why they are under pressure, with a short explanation on their life cycles, is advised. The roving creel survey method was suitable for the study area and delivered statistically rigorous results. I thus recommend that it is continued in the future by management. I make some recommendations for reducing costs of future surveys, as well as for altering the survey design if funds are very limited.

However, current methods for soil carbon determination are expensive, energy intensive, time consuming and potentially hazardous leading to a call for alternative methods, which should be cheap, fast, simple, accurate, safe and usable where resources and soil analysis laboratories are limited. To this end the student invented a novel rapid incineration field test (RIFT) for determining soil organic carbon and tested its validity in this study. This method incorporates principles found in dry combustion as well as loss-on-ignition and quantifying organic carbon through gravimetric analysis. In order to illustrate effectiveness and accuracy it was necessary to correlate RIFT with a reference method, in this instance dry combustion with a Leco device as well as another commonly used indirect method namely the Walkley-Black wet chemical oxidation method. Samples from eleven soil forms were collected from the Southern Cape region and they were subjected to the three testing methods. It was found that RIFT is indeed as effective and in 72% of the soil forms even more effective than Walkley-Black. Furthermore, it was ascertained whether the accuracy of RIFT can be improved by correcting for clay content. The correlation of RIFT with clay % was not very significant and clay % as a variable was therefore not used in this study to obtain further refinement of RIFT predictions. Another finding was also that RIFT illustrated less variability than both the Leco and Walkley-Black methods. The use and performance of recycling polypropylene in lead-acid battery cases

- Description: Polypropylene has proven to be the ideal material for the outer shell of the lead acid batteries. Due to its mold-ability and inert properties the material provides a capsule for the functioning components of the lead acid battery and can withstand a variety conditions encountered during its application, such as impact shock resistance, high and low temperatures and acid resistance. Polypropylene has however become of great concern with regards to environmental pollution since it is generally resistant to normal conditions of degradation and can only be properly disposed of by incineration. This factor has encouraged the industry to find ways to regenerate spent polypropylene. A good example of such a process is the recycling of lead acid batteries. This allows not only for the regeneration of lead, but also for the recycling of polypropylene in the manufacturing of battery cases. There are some cost advantages in using recycled polypropylene. However it does have its disadvantages in that the material does start to deteriorate after multiple processes. A common practice amongst battery manufacturers is to add virgin polypropylene to the recycled material in order to ensure performance consistency. The comparative study investigated the use of various ratios of virgin and recycled PP in the manufacturing of lead acid battery cases and their influence on the physical properties and performance of the final material. The degradation of PP was also investigated as the material was subjected to multiple manufacturing processes where the influence of stabilizers was further considered. A common technique of PP analysis such as MFI was shown to be an effective technique to maintain good quality control. The study further showed that it is important that the material grade of PP used in the manufacturing of the battery case and lid is compatible in order to allow for effective heating sealing of the two components. Polypropylene has a waxy surface finish and it is generally difficult to label or write on. Labels tend to fall off in application and make it difficult to maintain a track record of the manufactured batteries with time. This study showed successfully that a laser activated dye can be added to the PP without influencing its color or its performance. This allows for successful labeling of battery cases by various bar coding writers that can trace the battery through its manufacturing process. Lead acid batteries are often operated outside the specified temperature range that is determined by battery manufacturers resulting in premature failure. These failures can occur within the warranty period of the battery and result in illicit claims since the monitoring of the batteries in its application was not possible. A suitable temperature monitoring device was designed that would be incorporated into the vent cap or lid of the battery case. The device contained temperature sensitive indicators that would undergo a permanent color change at specified temperatures thereby giving the battery manufacturer an indication as to the maximum temperature the battery was exposed to.

The testing of natural and synthetic adjuvants to reduce herbicide-use and/or improve efficacy for the control of difficult-to-kill forest weeds

- Description: Commercial plantations within South Africa occur across diverse sites in terms of climate and physiography. Intensive silvicultural practices (site preparation, fertilisation, weed management etc.) are carried out during the establishment phase to ensure that the actual production of timber approximates the potential capacity for a site. Of these silvicultural practices, weed management is regarded as one of the most important, as if not carried out, competition from weeds may negatively influence seedling survival, growth and uniformity. During the establishment phase (planting to canopy closure), weed management operations are generally effective, as most competing species are still in a juvenile growth phase, and easily controlled. Due to limited weed-tree competition following canopy closure, weed management operations are reduced to a single event per annum (if necessary). This reduction in weed management often allows semi-shade/shade tolerant species to become established, together with an increase in the effort required for their control. The South African forest industry makes use of independent international auditing bodies to ensure that plantations are managed in a responsible, yet sustainable manner. Integrated management practices are considered the most effective as they reduce reliance on a single control method, such as the use of herbicides alone. In conjunction with integrated management, would be the need to reduce the overall quantities of herbicides used, yet at the same time ensuring improved efficacy of those herbicides that are used. The testing of these two aspects (reduced herbicide use and/or improved efficacy) was conducted on three diverse weeds that occur during the post-establishment phase within South African plantations, namely: Setaria megaphylla (setaria); Hedychium gardnerianum (wild ginger); and Solanum mauritianum (bugweed). The management of these species is considered problematic, as current herbicides and rates of application are not always completely effective. Four trials were implemented in KwaZulu-Natal (Midlands). Setaria megaphylla (setaria): Two trials were initiated (spring and autumn), to test the seasonal application of glyphosate + adjuvant combinations for improved efficacy (and hence reduced herbicide-use). A control (no herbicide applied), Select® (clethodim cyclohexanedione 120 g a.i. ℓ-1), Roundup® (glyphosate isopropylamine salt 360 g a.i. ℓ-1), and five Roundup® + adjuvant combinations were tested in both trials. iii The five adjuvants included three natural and two synthetic products that exhibited different properties and modes of action. At both sites, the eight treatments were replicated three times and laid out in a randomised complete blocks design. Setaria cover was assessed prior to spraying (0 days relative to spraying), and then again after two months (64 days relative spraying), with absolute changes in regrowth analysed. Although the application of Roundup® on its own provided good control within both trials, complete setaria control could not be obtained. Select® and the tested adjuvants did not improve Roundup® efficacy at the rates at which they were applied. The application of Roundup® in autumn (x̄ = 5.2% live cover) was more effective than the spring application (x̄ = 8.5% live cover), indicating that future studies should focus more on a combination of a season of application and subsequent follow-up operations, rather than the role of the adjuvants tested. Hedychium gardnerianum (wild ginger): Currently no herbicides are registered for the control of wild ginger in South Africa. One trial was implemented to test the potential for the control of wild ginger with herbicides applied to the foliage, cut-stems or rhizomes. In addition to an untreated control (no herbicide applied), Roundup® (glyphosate isopropylamine salt 360 g a.i. ℓ-1 at 4%) was applied as a foliar spray, either alone or with five adjuvants (three natural and two synthetic adjuvants). Brush- Off® (metsulfuron-methyl 6 g a.i. ℓ-1) or Roundup® was also applied to the cut-stems and rhizomes. Herbicide phytotoxicity was assessed up till 229 days after treatment application in terms of mortality, reduced and/or abnormal growth (of the resprouts) and foliar necrosis and/or discolouration. Although the foliar application of Roundup® (alone or with adjuvants) resulted in limited phytotoxicity, these treatments were ineffective in terms of killing wild ginger. The application of Roundup® or Brush-Off® to either the cut rhizome or cut-stems were equally effective, however, the cut-stem application would be preferred due to the reduced effort required. Future studies should further optimize the application of Roundup® (lowered rates + adjuvants) to the cut-stems/rhizome. Solanum mauritianum (bugweed): One trial was implemented on a stand of mature bugweed (located within a pine stand). The potential to reduce rates and/or improve the efficacy of Garlon® (triclopyr butoxyethyl ester 480 g a.i. ℓ-1®) was tested through the use of four adjuvants (two natural and two synthetic products) when applied as either a cut-surface or basal bark application. Two untreated controls (cut-surface or iv basal bark) were included for comparative purposes. The trial consisted of a 2 x 5 factorial arrangement of 10 treatments replicated three times and laid out as a randomized complete block design (RCBD). Although the cut-surface treatments were more effective than the basal bark treatments, no treatment provided complete control of bugweed. Of the cut-surface treatments, Garlon® + Break-Thru® (polyether-polymethylsiloxane-copolymer 1 000 g ℓ-1), Garlon® + Orosorb® (orange oil 50 g ℓ-1), and Garlon® + Actipron Super® (mineral oil 820 g ℓ-1) were equally effective, with Garlon® + Nu-Film P® (poly-1-menthene 975 g ℓ-1) the least effective. As the adjuvants tested did not improve the efficacy of Garlon® at the rates applied, the use of the most cost-effective and environmentally acceptable product would be preferable. For effective long-term control of mature bugweed, a cutsurface application is advisable, together with follow-up operations required. The results from these trials indicate that for bugweed and setaria, the currently registered herbicides remain the products of choice, albeit not 100% effective on wellestablished plants. In addition, the use of the adjuvants tested did not improve herbicide efficacy (and hence the opportunity for reducing rates of application) for all three species tested. - Description: Commercial plantations within South Africa occur across diverse sites in terms of climate and physiography. Intensive silvicultural practices (site preparation, fertilisation, weed management etc.) are carried out during the establishment phase to ensure that the actual production of timber approximates the potential capacity for a site. Of these silvicultural practices, weed management is regarded as one of the most important, as if not carried out, competition from weeds may negatively influence seedling survival, growth and uniformity. During the establishment phase (planting to canopy closure), weed management operations are generally effective, as most competing species are still in a juvenile growth phase, and easily controlled. Due to limited weed-tree competition following canopy closure, weed management operations are reduced to a single event per annum (if necessary). This reduction in weed management often allows semi-shade/shade tolerant species to become established, together with an increase in the effort required for their control. The South African forest industry makes use of independent international auditing bodies to ensure that plantations are managed in a responsible, yet sustainable manner. Integrated management practices are considered the most effective as they reduce reliance on a single control method, such as the use of herbicides alone. In conjunction with integrated management, would be the need to reduce the overall quantities of herbicides used, yet at the same time ensuring improved efficacy of those herbicides that are used. The testing of these two aspects (reduced herbicide use and/or improved efficacy) was conducted on three diverse weeds that occur during the post-establishment phase within South African plantations, namely: Setaria megaphylla (setaria); Hedychium gardnerianum (wild ginger); and Solanum mauritianum (bugweed). The management of these species is considered problematic, as current herbicides and rates of application are not always completely effective. Four trials were implemented in KwaZulu-Natal (Midlands). Setaria megaphylla (setaria): Two trials were initiated (spring and autumn), to test the seasonal application of glyphosate + adjuvant combinations for improved efficacy (and hence reduced herbicide-use). A control (no herbicide applied), Select® (clethodim cyclohexanedione 120 g a.i. ℓ-1), Roundup® (glyphosate isopropylamine salt 360 g a.i. ℓ-1), and five Roundup® + adjuvant combinations were tested in both trials. iii The five adjuvants included three natural and two synthetic products that exhibited different properties and modes of action. At both sites, the eight treatments were replicated three times and laid out in a randomised complete blocks design. Setaria cover was assessed prior to spraying (0 days relative to spraying), and then again after two months (64 days relative spraying), with absolute changes in regrowth analysed. Although the application of Roundup® on its own provided good control within both trials, complete setaria control could not be obtained. Select® and the tested adjuvants did not improve Roundup® efficacy at the rates at which they were applied. The application of Roundup® in autumn (x̄ = 5.2% live cover) was more effective than the spring application (x̄ = 8.5% live cover), indicating that future studies should focus more on a combination of a season of application and subsequent follow-up operations, rather than the role of the adjuvants tested. Hedychium gardnerianum (wild ginger): Currently no herbicides are registered for the control of wild ginger in South Africa. One trial was implemented to test the potential for the control of wild ginger with herbicides applied to the foliage, cut-stems or rhizomes. In addition to an untreated control (no herbicide applied), Roundup® (glyphosate isopropylamine salt 360 g a.i. ℓ-1 at 4%) was applied as a foliar spray, either alone or with five adjuvants (three natural and two synthetic adjuvants). Brush- Off® (metsulfuron-methyl 6 g a.i. ℓ-1) or Roundup® was also applied to the cut-stems and rhizomes. Herbicide phytotoxicity was assessed up till 229 days after treatment application in terms of mortality, reduced and/or abnormal growth (of the resprouts) and foliar necrosis and/or discolouration. Although the foliar application of Roundup® (alone or with adjuvants) resulted in limited phytotoxicity, these treatments were ineffective in terms of killing wild ginger. The application of Roundup® or Brush-Off® to either the cut rhizome or cut-stems were equally effective, however, the cut-stem application would be preferred due to the reduced effort required. Future studies should further optimize the application of Roundup® (lowered rates + adjuvants) to the cut-stems/rhizome. Solanum mauritianum (bugweed): One trial was implemented on a stand of mature bugweed (located within a pine stand). The potential to reduce rates and/or improve the efficacy of Garlon® (triclopyr butoxyethyl ester 480 g a.i. ℓ-1®) was tested through the use of four adjuvants (two natural and two synthetic products) when applied as either a cut-surface or basal bark application. Two untreated controls (cut-surface or iv basal bark) were included for comparative purposes. The trial consisted of a 2 x 5 factorial arrangement of 10 treatments replicated three times and laid out as a randomized complete block design (RCBD). Although the cut-surface treatments were more effective than the basal bark treatments, no treatment provided complete control of bugweed. Of the cut-surface treatments, Garlon® + Break-Thru® (polyether-polymethylsiloxane-copolymer 1 000 g ℓ-1), Garlon® + Orosorb® (orange oil 50 g ℓ-1), and Garlon® + Actipron Super® (mineral oil 820 g ℓ-1) were equally effective, with Garlon® + Nu-Film P® (poly-1-menthene 975 g ℓ-1) the least effective. As the adjuvants tested did not improve the efficacy of Garlon® at the rates applied, the use of the most cost-effective and environmentally acceptable product would be preferable. For effective long-term control of mature bugweed, a cutsurface application is advisable, together with follow-up operations required. The results from these trials indicate that for bugweed and setaria, the currently registered herbicides remain the products of choice, albeit not 100% effective on wellestablished plants. In addition, the use of the adjuvants tested did not improve herbicide efficacy (and hence the opportunity for reducing rates of application) for all three species tested. However, Roundup® or Brush-Off® applied to either the cut rhizome or cut-stems were equally effective in terms of wild ginger control, indicating an opportunity for further testing to optimise rates of application.

The taxonomic status of dolphins Tursiops spp. and Delphinus in South African waters

This poses a problem for cetaceans as basic information is frequently deficient and therefore not suitable for conservation planning purposes. This leads to difficulties in identifying and prioritizing areas for the monitoring, management and for the protection of species or populations. Morphological and geographic data used to be the primary means used by taxonomists to define species and subspecies, and due to this, most extant species have been classified by morphological characteristics. However, in the recent years, molecular analyses have been introduced and subsequently stable isotopes were also included to assist in interpreting results. Thereafter, it became a common approach to use more than one technique or a combination of independent datasets to resolve taxonomic uncertainties. This has resulted to a proposed term known as integrative taxonomy. Integrative taxonomy has recently been used in most studies that deal with taxonomy or population structure. The use of multiple lines of evidence, which is sometimes referred to as integrative taxonomy, raises the level of confidence in clearly delineating species. Cetaceans are the most taxonomically diverse group of aquatic mammals, with 93 living species accepted. Previously, their taxonomy was based on morphological characters. However, cetaceans have recently been subjected to intense phylogenetic investigations using both morphological and molecular data. These problems are not only prevalent in larger whales, but the small toothed whales also have these challenges, particularly family Delphinidae. This family is known to be the most diverse among the toothed whales. This is because this family diverged recently, and the divergence occurred relatively fast. This study aimed to clarify the taxonomic status of Tursiops spp. and Delphinus in the family Delphinidae, inhabiting the South African waters, using a combination of morphological and molecular methods. For skull morphology, geometric morphometrics were used, while for molecular methods, the D-loop region in the mitochondrial DNA was used to detect any differences within each genus. For the genus Tursiops, this study showed the consistency of results in all analyses used to investigate the presence of two species in the genus Tursiops in South African waters. This study only analysed sequences from the mitochondrial D-loop region and has re-confirmed the presence of the two species in South Africa. Skull and external morphology both show T. truncatus to be bigger and more robust, and to have a shorter rostrum compared with T. aduncus. These external observations are helpful when identifying the species in the field. Providing evidence supporting the presence of two Tursiops species in South Africa does not only contribute to the global context of the taxonomy of this genus, but also in the local context. This study also gives insight on the distribution of the two species in the region. For the genus Delphinus, molecular analysis did not reveal any clustering in South African Delphinus specimens, which suggested a single species, D. delphis, and that this species is morphologically highly variable. Both skull and external morphology did not reflect the molecular results, supporting suggestions that morphology in this genus does not reflect genetic differences. It has been shown that morphological variation reflects feeding specialization rather than speciation. This may also reflect local adaptations. Both skull and external morphology in this study showed no significant differences. This study did not investigate the phylogeographic details of the species in South Africa and further molecular work including a larger sample size from the west coast of South Africa is needed. By attempting to unravel the taxonomy of these dolphins in South African waters, the findings of this study will be very useful in the development of effective conservation and management strategies for these dolphins.

- Description: A series of core-functionalized borinic acids was synthesized from Grignard reagents and tributylborate and reacted with ethanolamine, amino acids, 2-pyridylmethanol as well as 2-(ethyl amino ethanol). Among the borinic acids synthesized were the ones bearing: para-(2, 15, 16, 22, 27)-methyl, (3, 20, 25)-chloro, (4, 23, 26)-fluoro, (5)-methoxy; meta-(7, 17)-methyl, (9)-fluoro; ortho-(6)-methyl, (8)-chloro, (10, 18, 19)-methoxy substituents on their respective aromatic systems. All compounds obtained by condensation of the borinic acids with the aforementioned nitrogen-containing substrates were characterized structurally by means of diffraction studies based on single crystals and found to constitute chelate esters/amides featuring covalent B-O and dative N→B bonds. Factors mentioned in the literature that has an influence on the hydrolytic stability on the heterocyclic borinate motif have been taken into consideration and verified for the systems researched in this study. The substitution at the nitrogen atom with one ethyl group caused the greatest increases in the N→B bond length (27) 1.714 Å, due to the increase in the nitrogen Lewis basicity, making it the least hydrolytically stable, these include compounds (24-26). Furthermore, DFT calculations were performed by employing Becke’s three parameter hybrid exchange functional with Lee–Yang–Parr functional (B3LYP) method using the Gaussian 16 program package (Rev. B01) employing the basis set 6-311++G(d,p), to corroborate and correlate the experimental findings, which gave a general R-squared value of 0.9978 for calculated vs experimental 13C chemical shifts. The synthesis, characterization, and application of peptide-capped magnetite nanoparticles for the targeting of cancer cells

- Description: In this study, a comparative analysis was performed upon three co-precipitation techniques for the synthesis of capped magnetite nanoparticles as to optimize the approach to the highest quality nanoparticles. Three techniques were applied whereby the capping agent either introduced before the stage of nanoparticle precipitation, simultaneously to the stage of precipitation, or after the stage of precipitation. The resultant nanoparticles were tested in terms of their size, dispersity, crystallinity, and magnetic characteristics. The three techniques gave nanoparticles of varying sizes and characteristics. Out of the three synthetic techniques, the post precipitation introduction method gave the highest quality nanoparticles in terms of size distribution, crystallinity and magnetic character. Three novel peptides were synthesized, incorporating amino acids to varying degrees. Structure was confirmed via IR and NMR spectroscopy. The peptides were studied potentiometrically to explore their acid nature and were explored computationally as to discern possible modes of interaction with the nanoparticles. These three peptides were further used in the capping of magnetite nanoparticles. For this set of nanoparticles, a higher synthesis temperature and larger iron content were used as to obtain larger nanoparticles. For the capping procedure, the post precipitation technique was used due to its previous positive results, once again yielding high quality nanoparticles with low size dispersity, high crystallinity, and high magnetic saturations. - Description: In this study, a comparative analysis was performed upon three co-precipitation techniques for the synthesis of capped magnetite nanoparticles as to optimize the approach to the highest quality nanoparticles. Three techniques were applied whereby the capping agent either introduced before the stage of nanoparticle precipitation, simultaneously to the stage of precipitation, or after the stage of precipitation. The resultant nanoparticles were tested in terms of their size, dispersity, crystallinity, and magnetic characteristics. The three techniques gave nanoparticles of varying sizes and characteristics. Out of the three synthetic techniques, the post precipitation introduction method gave the highest quality nanoparticles in terms of size distribution, crystallinity and magnetic character. Three novel peptides were synthesized, incorporating amino acids to varying degrees. Structure was confirmed via IR and NMR spectroscopy. The peptides were studied potentiometrically to explore their acid nature and were explored computationally as to discern possible modes of interaction with the nanoparticles. These three peptides were further used in the capping of magnetite nanoparticles. For this set of nanoparticles, a higher synthesis temperature and larger iron content were used as to obtain larger nanoparticles. For the capping procedure, the post precipitation technique was used due to its previous positive results, once again yielding high quality nanoparticles with low size dispersity, high crystallinity, and high magnetic saturations. The quality and quantity of the carbon spheres had an overall improvement as the synthesis temperatures increased from 800 oC to 950 oC. TGA and XRD analysis indicated a direct relationship between an increase in synthesis temperatures with an increase in the crystallinity of the carbon spheres. Anastrozole is an important drug used for the treatment of breast cancer. In the first step, mesitylene was brominated using N-bromosuccinimide to obtain 3,5-bis(bromomethyl)toluene. Selectivity became an issue due to the formation of two by-products; namely 1,3,5-tris(bromomethyl) benzene (the tribrominated by-product) and 1-(bromomethyl)-3,5-dimethylbenzene (the monobrominated by-product). Since the reaction parameters can be more precisely controlled in flow chemistry systems, we were able to optimize the formation the desired product 3,5-bis(bromomethyl)toluene. The reaction was initially optimized ina 15μLChemtrix glass micro reactor resulting in 100%conversion with 95%selectivity towards the desired product 3,5-bis(bromomethyl)toluenein 15 seconds, with a throughput of 0.006 g/h. The reaction was then scaled up in a 1.7 mLLTF reactor, equally yielding 100%conversion with 95%selectivityin 4minutes, with a throughput of2.01g/h. The bromination of mesitylene was also attempted in a homemade photochemical reactor consisting of a 3.02 polytetrafluoroethylene coil reactor and BLE-6T365 UV lamp. In this photochemical reactor, although 100%conversionwas again obtained only75%selectivity was achieved in 20minutes, with a throughput of 0.025g/h. This drop in selectivity was attributed to poorer mixing in the larger polytetrafluoroethylene coil reactor and possibly insufficient light penetration. The next step, involving the cyanation of 3,5-bis(bromomethyl)toluene to obtain 2,2'-(5-methyl-1,3-phenylene) diacetonitrilegave 100%conversion in 1 minute at 190 °C in a 1.7 mLLTFreactor, with a throughput of 5.2g/h. The subsequent methylation of 3,5-bis(cyanomethyl)toluene to obtain3,5-bis(1-cyano-1-methylethyl) toluene was investigated using a1mLpolytetrafluoroethylenecoil reactor, ultimatelyachieving99%conversionat40 °Cin 8minutes, with a throughput of 0.045g/h. - Description: In this work, the host capabilities of two structurally related compounds, N,N’-bis(9-phenyl-9- thioxanthenyl)ethylenediamine (H1) and N,N’-bis(9-phenyl-9-xanthenyl)ethylenediamine (H2) were compared in the presence of a wide variety of guest species. Additionally, the selectivity displayed by these host compounds were examined when exposed to mixtures of guests in order to ascertain whether it would be feasible to employ them in alternative separation strategies for the purification of industrially relevant chemicals. H1 and H2 were synthesized by reacting thioxanthone and xanthone with phenylmagnesium bromide. The resultant alcohol was then treated with perchloric acid and, finally, two of these molecules were effectively linked by utilizing ethylenediamine to afford the two host compounds. Initially, H1 and H2 were investigated for their inclusion abilities by recrystallizing each from a number of potential isomeric and non-isomeric guest compounds such as the xylenes and ethylbenzene, methylanisoles and anisole, methylpyridines and pyridine, methylcyclohexanones and cyclohexanone, heterocyclic five- and six- membered ring compounds, alkylsubstituted benzenes, anilines, and dihaloalkanes. H1 displayed excellent inclusion ability when presented with the above-mentioned compounds, and a 1:1 H:G ratio was consistently preferred in each case. H2 also proved to be successful in this regard but did not include the methylcyclohexanones and cyclohexanone nor the heterocyclic five-membered ring solvents. Furthermore, varying host:guest ratios were observed for the complexes formed with H2. Mixed competition experiments were carried out in the presence of either isomeric or related but non-isomeric guest species. When H1 and H2 were independently recrystallized from mixtures of the former, selectivity orders correlated for both hosts, but it was observed that H2 exhibited an enhanced selectivity for the preferred guests in each case, compared with H1. Interestingly, in mixtures of the latter, host behaviours were distinctly opposing (with the exception of the dihaloalkanes). H1, and even more so H2, demonstrated very high selectivities for p-xylene, aniline and N,Ndimethylaniline from the xylene and aniline guest series, respectively, where selectivities were found to be ~90% or higher for host recrystallization experiments from respective mixtures of these guests. Single crystal X-ray diffraction, Hirshfeld surface and thermal analyses were employed in order to elucidate the reasons for any selectivity observations. The inclusion of these guests was, in most cases, found to be as a result of interactions between host and guest species, which included π∙∙∙π stacking, C‒H∙∙∙π, hydrogen bonding and various other short contact types. Guest compounds were accommodated in either cavities or channels and this was dependent on the nature of the guest. The host molecule conformations showed H1 to adopt a bent tricyclic fused ring system with the N atoms of the linker in a synclinal arrangement, while in complexes with H2, the fused ring system was near-planar and the N atoms adopted an antiperiplanar geometry. These key differences resulted in a very ordered host‒host packing for H2 as a direct result of the more planar O-containing ring and linear linker; for H1, on the other hand, the buckled S-containing ring and gauche-orientated N atoms resulted in a less ordered packing, which ultimately related to the differences in the behaviour of the two host species. Hirshfeld surface analyses, in general, did not provide much information to explain the host selectivities, with the exception of complexes containing the five-membered ring guest heterocyclics. - Description: This investigation focussed on the inclusion and resolution ability of (2R,3R)-1,1,4,4-tetraphenylbutane-1,2,3,4-tetraol (TETROL), studies on the stoichiometry of its derivatives and the formation of inclusion compounds for single crystal analysis. The guest compounds that featured in the main study were cyclohexanone, 2-methylcyclohexanone, 3-methylcyclohexanone and 4-methylcyclohexanone. It was found that 3- and 4-methylcyclohexanone were trapped in their energetically unfavourable axial conformations in the inclusion crystal. Resolution experiments proved that (2R,3R)-1,1,4,4-tetraphenylbutane-1,2,3,4-tetraol can be used as a resolving agent for 2- and 3-methylcyclohexanone, with ee values of 13% and 22%, respectively (according to the method of Hiemstra), in favour of the R-enantiomer. Single crystal X-ray diffraction (SCXRD) studies, however, showed that 2-methylcyclohexanone was resolved with an ee of 30% in R. An ee of 56% in R was obtained for 3-methylcyclohexanone. Enantiomer enrichment of 2-methylcyclohexanone was achieved in basic medium (ee of 18% according to the method of Hiemstra) and showed that by using the host in either half or double the molar ratio of the guest, a higher ee was obtained than for a 1:1 ratio. The following TETROL derivatives were also synthesized and their stoichiometries with various guest compounds were determined: (2R,3R)-1,1,4,4-tetra(naphthalen-1-yl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(naphthalen-2-yl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(p-anisyl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(p-tolyl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(m-tolyl)butane-1,2,3,4-tetraol and; (2R,3R)-1,1,4,4-tetra(o-tolyl)butane-1,2,3,4-tetraol.

- Description: This investigation focussed on the inclusion and resolution ability of (2R,3R)-1,1,4,4-tetraphenylbutane-1,2,3,4-tetraol (TETROL), studies on the stoichiometry of its derivatives and the formation of inclusion compounds for single crystal analysis. The guest compounds that featured in the main study were cyclohexanone, 2-methylcyclohexanone, 3-methylcyclohexanone and 4-methylcyclohexanone. It was found that 3- and 4-methylcyclohexanone were trapped in their energetically unfavourable axial conformations in the inclusion crystal. Resolution experiments proved that (2R,3R)-1,1,4,4-tetraphenylbutane-1,2,3,4-tetraol can be used as a resolving agent for 2- and 3-methylcyclohexanone, with ee values of 13% and 22%, respectively (according to the method of Hiemstra), in favour of the R-enantiomer. Single crystal X-ray diffraction (SCXRD) studies, however, showed that 2-methylcyclohexanone was resolved with an ee of 30% in R. An ee of 56% in R was obtained for 3-methylcyclohexanone. Enantiomer enrichment of 2-methylcyclohexanone was achieved in basic medium (ee of 18% according to the method of Hiemstra) and showed that by using the host in either half or double the molar ratio of the guest, a higher ee was obtained than for a 1:1 ratio. The following TETROL derivatives were also synthesized and their stoichiometries with various guest compounds were determined: (2R,3R)-1,1,4,4-tetra(naphthalen-1-yl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(naphthalen-2-yl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(p-anisyl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(p-tolyl)butane-1,2,3,4-tetraol; (2R,3R)-1,1,4,4-tetra(m-tolyl)butane-1,2,3,4-tetraol and; (2R,3R)-1,1,4,4-tetra(o-tolyl)butane-1,2,3,4-tetraol.

The priority species identified were: Acacia cyclops (rooikrans), Acacia longifolia (long-leaved wattle), Acacia mearnsii (black wattle), Acacia melanoxylon (blackwood), Acacia saligna (Port Jackson willow), Eucalyptus camaldulensis (red river gum), Pinus pinaster (cluster pine) and Pinus radiata (radiata pine). The Random Forest classifier on SPOT-6 data achieved an overall accuracy of 62.5% and this method was consequently deemed ineffective at separating invasive tree species from other tree species in the Garden Route. The overall accuracy of the WorldView-3 classifier was higher (78.9%) but the cost of the data limited the use of more images for the detection of the focal species throughout the Garden Route. Therefore, to identify priority areas for invasive tree management, criteria derived from existing literature were input into spatial conservation planning software. The analysis identified the: Saasveld section of the Garden Route National Park, the Wilderness Lakes, Knysna Forest, Knysna Estuary, Tsitsikamma Forest around Stormsriver and a disturbed area of fynbos southeast of Kareedouw as management priorities. Currently spatial conservation planning software proved to be cost-affordable and useful tool and is recommended for invasive tree management in the Garden Route.

The need for research into MPEG-4 authoring tools arose from the reported lack of knowledge of the MPEG-4 standard and the limited implementations of MPEG-4 authoring tools available to content authors. In order for MPEG-4 to reach its full potential, it will require authoring tools and content players that satisfy the needs of its users. The theoretical component of this dissertation included a literature study of the MPEG-4 standard and an investigation of relevant multimedia authoring systems. MPEG-4 was introduced as a standard that allows for the creation and streaming of interactive multimedia content at variable bit rates over high and low bandwidth connections. The requirements for the prototype 2-D MPEG-4 authoring system were documented and a prototype system satisfying the requirements was designed, implemented and evaluated. The evaluation of the prototype system showed that the system successfully satisfied all its requirements and that it provides the user with an easy to use and intuitive authoring tool. MPEG-4 has the potential to satisfy the increasing demand for innovative multimedia content on low bandwidth networks, including the Internet and mobile networks, as well as the need expressed by users to interact with multimedia content. This dissertation makes an important contribution to the understanding of the MPEG-4 standard, its functionality and the design of a 2-D MPEG-4 Authoring tool. The spatiotemporal aspects of predation on the Cape gannet Morus capensis population at the Lambert’s Bay gannet colony, Western Cape, South Africa

As a result of multiple human-induced threats, seabird species including the Cape gannet Morus capensis which is endemic to southern Africa population experience significant declines and is now classified as Endangered. The Cape gannet faces predation threats from a variety of different species. The current predators of concern at Lambert’s Bay gannet colony are both the Cape fur seal Arctocephalus pusillus pusillus and the Kelp gull Larus dominancus. The former predates on fledglings and the latter predates on eggs. In the 2005/06 breeding season the entire Cape gannet population deserted the Lambert’s Bay gannet colony due to predation by the Cape fur seal. Although the Cape gannets have subsequently recovered to approximately 8000 pairs, seal predation on Cape gannet fledglings continues, together with predation by Kelp gulls. This has caused concern over the impact of predation on this species at the Lambert’s Bay colony, particularly given the species’ conservation status, its small breeding range and its current population trend. CapeNature, the conservation agency involved at this colony has been monitoring and managing the predation on Cape gannets and are interested in evaluating the success of their efforts. Therefore the aim of this study was to determine the spatial and temporal aspects of predation on the Cape gannet population at Lambert’s Bay gannet colony. When examining the influence of fish availability on predation intensity, we found that in years with an increase in fish biomass there was a decrease in predation probability by Cape fur seals. This increase in fish biomass not only led to decreased predation on the fledglings it also led to increased breeding success of the Cape gannets. In determining the effect of predation within the Cape gannet breeding colony we found that the probability of predation by Kelp gulls decreased in years with increased adult gannet density. Indirectly, Cape gannet population growth should lead to a decrease in Kelp gull predation as a higher gannet density increases the risk of injury to the Kelp gulls. This suggests that if Cape gannet populations continue to decline, they may become increasingly vulnerable to Kelp gull predation which can result in a further population decline. In terms of past management actions, the selective culling of predatory Cape fur seals and Kelp gulls, was evaluated. It was found that in the years in which culling of Cape fur seals and Kelp gulls was implemented, a decrease in predation was observed. This indicated that culling reduced predation in the short-term. Predation is inevitable, but due to the Cape gannet’s conservation status we suggest that additional, long-term, and more sustainable methods to reduce predation should be explored. Our research emphasises the need for improving food availability to Cape gannets as food security is paramount to their long-term survival and population growth. Colony management in isolation from other management strategies, is not efficient for conserving a species. We therefore suggest that sound management of predation at the terrestrial habitats are required but that effective management of fisheries are also required for the successful conservation of the Cape gannet. Food security for Cape gannets could be improved by, for example, establishing some ways that we suggest improving the food security for Cape gannets would be to establish Marine Protected Areas (MPAs) with dynamic boundaries around Cape gannet colonies, and to have locate the MPAs close to breeding and foraging hotspots where these areas are closed to fishing thus reducing the competition between commercial fisheries and marine predators, such as the Cape gannet. Another way to improve food security for the gannets are to promote the efforts of focussing on developing and implementing an ecosystem approach to fisheries management in which fishing quotas account for the needs of marine predators. The findings of this study have not only aided the understanding of the dynamics of predation on the Cape gannets and highlighted the extent that predation takes place on them but can also guide future management decisions regarding predation on this endemic and endangered species.

This species provides important ecological services yet faces numerous anthropogenic threats. An emerging threat in the south eastern part of South Africa is that of wind energy development. Understanding the movement and roosting behaviour of the Cape Vulture may mitigate potential collisions if areas of high use are avoided. Juvenile Cape Vultures, who naturally suffer high mortality rates, are known to forage extensively over a wider landscape and as a consequence may face a greater assortment of threats. The overall aim of this study therefore was to determine the ranging and roosting behaviour of juvenile Cape Vultures in the Eastern Cape Province, South Africa. In order to achieve this aim, i) the ranging behaviour and habitat use were determined and ii) roost sites and factors that influence roost site selection were determined. Using five juvenile Cape Vultures tagged with Global Positioning System (GPS/GSM) transmitters, home range sizes, distance travelled from the nest and habitat use were determined. Home range was determined through kernel density estimates and distance travelled from the nest was determined through the Euclidean distance. Habitat use was determined by overlaying the home range size onto a merged layer of all protected areas and the National Land Cover Database of South Africa. To determine roost sites, data from the tagged vultures were split into encamped and exploratory movements using a mixture model in a cluster analysis setting. Encamped movements were associated with roost sites. Roost density was determined around the natal colony using predetermined buffer sizes and a generalised linear mixed model (GLMM) fitted to the data. Conditions considered favourable for roost sites were based on previous research conducted on cliff-nesting species and a GLMM conducted. Juveniles increased their home range progressively for the first two months, then exhibited a rapid increase in size associated with dispersal from the colony. Distance from the nest increased rapidly following the dispersal period. Protected areas and woody vegetation were areas of preferred habitat. The highest density of roosts for juveniles was located within 20 km’s from the breeding colony and decreased further away. Roost sites that were favoured for juveniles and adult birds were those that were located close to colonies, had low accessibility to terrestrial predators and were in areas of high wind speed. The orientation of the cliff into the prevailing wind direction was also a determining factor. Roosting sites and foraging areas are important spatial determinants of Cape Vulture behaviour and the identification of such areas can help with conservation management. With the additional threat of wind development in areas highly utilised, wind farms located too close to colonies could have a devastating impact on the Cape Vulture population. Therefore, this study highlights the importance of conservation buffer zones as no-go areas for wind energy development around vulture colonies.

Juvenile Cape Vultures, who naturally suffer high mortality rates, are known to forage extensively over a wider landscape and as a consequence may face a greater assortment of threats. The overall aim of this study therefore was to determine the ranging and roosting behaviour of juvenile Cape Vultures in the Eastern Cape Province, South Africa. In order to achieve this aim, i) the ranging behaviour and habitat use were determined and ii) roost sites and factors that influence roost site selection were determined. Using five juvenile Cape Vultures tagged with Global Positioning System (GPS/GSM) transmitters, home range sizes, distance travelled from the nest and habitat use were determined. Home range was determined through kernel density estimates and distance travelled from the nest was determined through the Euclidean distance. Habitat use was determined by overlaying the home range size onto a merged layer of all protected areas and the National Land Cover Database of South Africa. To determine roost sites, data from the tagged vultures were split into encamped and exploratory movements using a mixture model in a cluster analysis setting. Encamped movements were associated with roost sites. Roost density was determined around the natal colony using predetermined buffer sizes and a generalised linear mixed model (GLMM) fitted to the data. Conditions considered favourable for roost sites were based on previous research conducted on cliff-nesting species and a GLMM conducted. Juveniles increased their home range progressively for the first two months, then exhibited a rapid increase in size associated with dispersal from the colony. Distance from the nest increased rapidly following the dispersal period. Protected areas and woody vegetation were areas of preferred habitat. The highest density of roosts for juveniles was located within 20 km’s from the breeding colony and decreased further away. Roost sites that were favoured for juveniles and adult birds were those that were located close to colonies, had low accessibility to terrestrial predators and were in areas of high wind speed. The orientation of the cliff into the prevailing wind direction was also a determining factor. Roosting sites and foraging areas are important spatial determinants of Cape Vulture behaviour and the identification of such areas can help with conservation management. With the additional threat of wind development in areas highly utilised, wind farms located too close to colonies could have a devastating impact on the Cape Vulture population. Therefore, this study highlights the importance of conservation buffer zones as no-go areas for wind energy development around vulture colonies.

In addition, concerns about costs of solvents and the long-term environmental impact that can potentially result when solvents are discarded after their use have led to focused investigations into solvent-free procedures, as reported in recent literature. We have set out to explore the extent to which these advantages could be realized within our research. Non-volatile, non-flammable imidazolium ionic liquids [bmim][I], [bmim][BF4] and [bmim][PF6] were used as green solvents in ferrocene chemistry. Ferrocenoate esters were synthesised efficiently by the respective DCC/DMAP-promoted reactions of ferrocenecarboxylic acid and substituted benzoic acids or, alternatively, the DMAP-promoted reactions of ferrocenoyl fluoride with a range of substituted phenols in [bmim][BF4] and [bmim][PF6]. High yields and short reaction times were achieved. In addition, the ionic liquid was reused several times without a reduction in product yields. Under solvent-free conditions, DCC/DMAP-promoted reactions provided high yields within 3 min of reaction. The possible rearrangement of one of the intermediates in these reactions was modelled theoretically using density function theory (DFT) at the B3LYP/6-31G* level of approximation. Catalyst-free esterification was achieved by the application of microwave radiation to the reaction of ferrocenoyl fluoride and a range of substituted phenols. All the reactions were complete after 1 min of irradiation and products were isolated in high yield. DPAT, HfCl4, Sc(OTf)3 and Al(OTf)3 were screened as catalysts for esterification in [bmim][BF4] and under solvent-free conditions at various temperatures. All attempts at esterification of ferrocenecarboxylic acid with alcohols and phenols were unsuccessful. The Suzuki cross-coupling reaction was carried out in [bmim][BF4]. The isolated yields are, however, poor and suffer from poor reproducibility with different batches of [bmim][BF4] used.

The current study presents evidence indicating that indigenous vegetables provide a sustainable yet inexpensive answer to several of these challenges. The research was aimed at determining the socio-economic impact of indigenous vegetables in the Ntuze community of northern KwaZulu-Natal. A mixed-methods methodology was employed to achieve a holistic understanding of the relationship between the use of indigenous vegetables and socio-economic influences. The study discovered that indigenous vegetables play an important role in the livelihood strategy of this rural farming community. Findings revealed that the utilisation of indigenous vegetables makes an essential contribution to the livelihoods and well-being of the Ntuze people, especially in terms of curbing food poverty, income generation and medicinal benefits. However, the value of these vegetables was found to be appreciated mostly by the elderly. Conclusions therefore indicate a possible decline in future production and use of indigenous vegetables if indigenous knowledge is not passed on to the next generation. Integration of indigenous knowledge in agricultural technology transfer programmes is therefore vital to promote production of indigenous vegetables as a sustainable food resource. Actively addressing the stigma attached to indigenous vegetables as being “low-status food” is also crucial to enhance perception and understanding of the value of these vegetables. This would contribute to both preserving cultural heritage and conserving valuable indigenous flora. Such intervention would safeguard this sustainable and renewable resource in its pivotal socio-economic role in terms of rural food security as is substantiated by this study.

However, this great effort does not appeal to the principles that policies have set out in terms of equality. Along with the delivery of this high level of state funded housing, many of the RDP dwellers feel as though they have been marginalised and forced out of the area of opportunity, namely, the CBD and the northern suburbs (Huchzermeyer, 2003). Many researchers believe that the South African housing delivery pattern resembles that of apartheid segregationist planning, which exacerbated the social inequalities in the country (Huchzermeyer, 2003; Donaldson, 2001; Harrison et al., 2003). To gain an in-depth understanding of the lived experiences of residents of a low-cost housing settlement, an interpretative phenomenological analysis was undertaken. The emergent themes emanating from the research study revealed that social problems were pertinent factors to the lack of formulation of community social cohesion. Without this aspect, community building will be hampered, and thus cohesive neighbourhoods will be lacking. To this end, social innovation is proposed as a solution to the Zanemvula development. The issue of social development needs to placed back on the housing agenda, as is stated in the latest Housing Act.

The separation of platinum and gold from an industrial feed solution

- Description: In this thesis, the aim was to develop resins which are platinum and gold specific to be utilized for the early removal of these metals from the industrial feed. Efforts were therefore directed towards the synthesis of silica based resins with active centra which were designed for platinum and gold specificity respectively. The large chlorometallate ions in the feed stream were characterized in terms of physical parameters relevant to phase distribution namely distortability (RD), charge density, softness (σ) etc. Matching cations for each of the types were investigated. In order to attempt the design of platinum specific resins different structural amines were used to aminate the silicone precursor and subsequently to fix these onto the silica framework. Two different solvents i.e. alcohol and dmf were used for this process, resulting in two sets of resins with different properties. For gold specific resins, various polyethers were attached to a different type of silicone precursor, which was attached to the silica framework. The design was based on previous experience with these ions with reference to their behaviour towards different types of cations. The platinum species PtCl6 2- and PtCl4 2-, the gold species AuCl4 -, as well as the most important contaminants in the feed stream were typified bearing in mind size, charge, charge density and distortability. Different types of cationic centra having differences in charge density, stereochemical crowding and extent of hydrophobicity were synthesized and tested both as solvent extractants (where possible) and silica based resins. The results indicated that partly screened secondary ammonium cationic resin species, which could be regarded as “intermediate”, proved to be satisfactory both in their high percentage extraction for PtCl4 2- and rejection of contaminants like chlororhodates, chloroiridates(III) and FeCl4 -. It was, however, necessary to work at a redox potential where iridium(IV) in the form of IrCl6 2- is absent. Various 2-aminoalkane resins were prepared with variation in the length of alkane group and synthesized in the two different solvents. The latter resulted in two sets of resins with different compactness also having significantly different properties with reference to platinum specificity, HCl effect and stripping potential. The 2- aminobutane and 2-aminoheptane resins both proved to be very satisfactory platinum specific resins with respect to selectivity, platinum capacity and stripping potential. The various physical parameters could be utilized to accommodate the chemical behaviour. To obtain gold specific resins, experiments were performed with resins having oxygen-donor atoms which can readily be protonated to form onium type cations for example amides and ether oxygen atoms. In the case of the latter, various polyethers with a different number of ether groups (polyether groups linked by ethylene and propylene groups) and variations of hydrophobicity (by substitution) have also been studied. Linked to the polyether groups were alkane and aryl groups. Those having 8 to 10 ether groups and aromatic tail ends proved to be moderately successful in terms of gold capacity and sharp breakthrough curves of their columns, however, platinum could not be very effectively rejected.
