- Description: A fluorescent nanofiber probe for the determination of Ni2+ was developed via the electrospinning of a covalently functionalized pyridylazo-2-naphthol-poly(acrylic acid) polymer. Fluorescent nanofibers with diameters in the range 230–800 nm were produced with uniformly dispersed fluorophores. The excitation and emission fluorescence were at wavelengths 479 and 557 nm respectively, thereby exhibiting a good Stokes' shift. This Ni2+ probe that employs fluorescence quenching in a solid receptor–fluorophore system exhibited a good correlation between the fluorescence intensity and nickel concentration up to 1.0 μg mL−1 based on the Stern–Volmer mechanism. The probe achieved a detection limit (3δ/S) of 0.07 ng mL−1 and a precision, calculated as a relative standard deviation (RSD) of more than 4% (n = 8). The concentration of Ni2+ in a certified reference material (SEP-3) was found to be 0.8986 μg mL−1, which is significantly comparable with the certified value of 0.8980 μg mL−1. The accuracy of the determinations, expressed as a relative error between the certified and the observed values of certified reference groundwater was ≤0.1%. The versatility of the nanofiber probe was demonstrated by affording simple, rapid and selective detection of Ni2+ in the presence of other competing metal ions by direct analysis, without employing any further sample handling steps.

- Description: Changes within ICT over the past decade required a review of the network solution deployed in the Siyakhula Living Lab (SLL), a joint venture between the Universities of Fort Hare and Rhodes, focused on finding blueprints for sustainable e-infrastructure in poor communities. The review found that the topology used in the SLL’s network is still viable but WiMAX is now outdated. Also, the in-network cache, Squid, is no longer effective, given the switch to HTTPS, the strong presence of advertising, and the data weight of current web pages. The solution to the first issue is outdoor Wi-Fi, a proven solution easily deployable in grass-roots fashion. The second issue can be cured leveraging Squid’s ‘bumping’ and splicing features; deploying a browser extension to make picture download optional; and using Pi-hole, a DNS sinkhole. Hopefully, the revised solution could become a component of South African Government’s broadband plan, “SA Connect”.

These studies give evidence of the formation of a complex between gold(III) and mannan in acid media as judged by the enhancement in the AdCSV currents and shift in the reduction peak of gold(III) in the presence of mannan. The AdCSV currents were linearly dependent on gold(III) concentrations ranging from 7.0 × 10−7 to 3.0 × 10−4 mol dm−3. A detection limit of 6.0 × 10−8 mol dm−3 was obtained. Interferences of copper(II) were observed in the presence of mannan, but there was no significant interference of silver(I).

In this study, crystalline L-lysine was used in an attempt to determine the quantitative lysine requirement of juvenile Haliotis midae. Two microencapsulation techniques [gelatine/acacia and cellulose acetate phthalate (CAP)] were used to retard leaching of crystalline L-lysine incorporated into semipurified test diets. An assessment of the efficacy of the encapsulation techniques, revealed that despite effective lysine supplementation, H. midae fed semipurified test diets containing encapsulated crystalline L-lysine failed to promote significant improvements in either growth, feed or protein efficiency (P > 0.05). The failure of the crystalline L-lysine to illicit growth and nutritional responses is discussed.

- Description: Purpose: To isolate the anti-cancer and anti-trypanosomal principles of Zanthoxylum leprieurii, a medicinally versatile wild tropical plant used for managing tumours, African trypanosomiasis, and inflammation in southeastern Nigeria. Methods: The pure compounds were isolated using chromatographic methods. The structural elucidation of the pure compounds was based on their NMR (1D and 2D) and mass spectral data as well as chemical test results. Structure-activity relationships were based on the structural differences among the compounds. The cytotoxicity of the extracts and compounds (1, 2, 3, and 4) was evaluated in HeLa (human cervix adenocarcinoma) cell line while the trypanocidal activities were evaluated on Trypanosoma brucei brucei. Results: Two acridone alkaloids, 1-hydroxy-3-methoxy-10-methylacridin-9 (10H)-one, named fabiocinine (1), and 1-hydroxy-2,3-dimethoxy-10-methylacridin-9 (10H)-one (arborinine, 2), together with a furoquinoline alkaloid, skimmianine (3), and a chelerythrine derivative, 6-acetonyl-5,6-dihydrochelerythrine (4) were isolated from the root bark of Zanthoxylum leprieurii. Skimmianine (3) exhibited cytotoxicity and anti-trypanosomal IC50 of 12.8 and 13.2 µg/mL respectively (p less than 0.05). Compound (1) and arborinine (2) were selectively cytotoxic to HeLa cells with cytotoxicity IC50 of 28.49 and 62.71 µg/mL, respectively, while (4) did not show significant activity (p less than 0.05). Conclusion: Zanthoxylum leprieurii root bark contains cytotoxic and trypanocidal compounds, and is thus a potential source of anti-cancer and anti-trypanosomal leads.

The isolation of the active constituents was undertaken by bio-autographic assays inconjunction with chromatographic techniques. The identification and characterisation of the isolatedcompounds were done using mass spectrometry (MS) and Fourier transformed infrared spectrometry(FTIR) as well as 1D- and 2D- nuclear magnetic resonance (NMR) analyses.Results:The leaves and stems ofA. cordifoliaexhibited varied antibacterial activity against all eight pa-thogens. Most of the MIC values ranged between 63 and 2000mg/ml. The highest activities for the crudeextracts (63mg/ml) were observed againstS. saprophyticus[stem (EtOAc, CHCl3and hexane), leaves(MeOH, EtOH, EtOAc and CHCl3)],E. coli[stem (MeOH and EtOH), leaves (MeOH, EtOH, EtOAc andCHCl3)],M. catarrhalis[leaves (EtOAc and CHCl3)],K. pneumoniae[stem (CHCl3), leaves (CHCl3)] andS.aureus[leaves (CHCl3)]. Seven constituents [stigmasterol (1), stigmasta-4,22-dien-3-one (2), friedelin (3),friedelane-3-one-28-al (4), 3-O-acetyl-aleuritolic acid (5), 3-O-acetyl-erythrodiol (6) and methyl-3,4,5-trihydroxybenzoate (methyl gallate) (7)] were isolated from the stem MeOH extract. All these com-pounds displayed some antibacterial activity against the eight pathogens with highest activity againstS.saprophyticus(2mg/ml). Furthermore, this is thefirst report of compounds1,2,3,4,6and7isolated fromA. cordifoliaand where a complete set of 2D-NMR data for fridelane-3-one-28-al (4) is presented.Conclusion:The study demonstrated that the antibacterial activities ofA. cordifoliaextracts may be dueto the presence of the seven isolated compounds, where compounds3–6showed the best activity. Theobserved activity against gastrointestinal, skin, respiratory and urinary tract pathogens supports thetraditional use for the treatment of such ailments.

- Description: Aerobic cathode microbial fuel cells (MFCs) have been widely researched to provide bioremediation of wastewaters, coupled to sustainable energy production. In order to effectively accomplish this aim, suitable catalysts and catalyst supports for oxygen reduction reaction (ORR) are required. While iron phthaloycanine (FePc), supported on multi-walled carbon nanotubes has previously been studied for this function, cost of industrial production may hinder this. Importantly, this study examines the use of several available grades of carbon black for their relative suitability to perform as supports for FePc in MFC formats. Voltammetric studies showed that the different grades of carbon black provided varying lowering of the ORR overpotential, between ∼160 and ∼270 mV relative to unmodified GCEs, and an optimum grade (N326) was selected for further study. Carbon black/FePC composite electrodes exhibited comparable lowering of the ORR overpotential (606 mV) to potentials previously reported to nanotube/FePc composites (620 mV), as well as lowered charge-transfer resistance compared to electrodes solely modified with FePc. When applied as cathode modifiers in dual chambered MFCs utilising Enterobacter cloacae, the combined use of carbon black and FePc provided greater power densities than either alone; composite electrodes obtaining ∼400% power density, compared to unmodified electrodes. Modification of the anode with carbon black further increased power density, generating power densities an order of magnitude larger than those obtained at unmodified electrodes. The ability of beer brewery waste water (BBWW) to generate power at these modified surfaces yielded permissible power densities (∼40% that of reinforced clostridial media). Differences observed, in particular under agitation, are attributed to variations in nutrient content and nutrient complexity, between the two fuel substrates.

- Description: We reply to the article, ‘Why are black South African students less likely to consider studying biological sciences?’, authored by University of Cape Town (UCT) researcher Nicoli Nattrass1 and published in the South African Journal of Science on 27 May 2020. At the time of writing our reply the article had already received sharp criticism from the Black Academic Caucus at UCT and in a statement released on 5 June 2020, the UCT executive distanced itself from the content of the paper, inviting rigorous, respectful review of the published research. Nattrass’ article1 follows two papers published in 2019 by scholars associated with South African universities, both of which were widely denounced for the racist undertones of their content. The first article was authored by Stellenbosch University researchers who reported on low cognitive functioning of coloured women linked to education levels and lifestyles2 ; see comment by Le Grange3 . The second article was co-authored by an adjunct professor at UCT and examined the role of cognitive ability or intelligence on slave exports from Africa4 . These publications gave rise to a broader debate on enduring racism in science and the re-ascendency of race science internationally. In this reply, we focus on the methodology of attitudinal survey used for the study reported by Nattrass (and by many other scientists). There are two lines of argument that we weave together: the quality of the research as reported, and problems inherent to comparative attitudinal survey research (regardless of how well it is executed). It should be noted that the Black Academic Caucus at UCT has also critiqued the research design, identifying many of the points we make here, and they have additionally included a discourse analysis of the paper (circulated via e-mail). To demonstrate how problematic a survey design can be, particularly when it is not rigorously executed, we share data and observations that counter the apparent findings of Nattrass’ study. We then argue that the publication of this research, in this manner, is detrimental to the biodiversity sector in which the tropes being perpetuated, can cause enormous harm. In the process we hope to encourage scientists to be more reflexive about their methodology, and we encourage the South African Journal of Science to publish works that are worthy of the young people of this country - research in which they can recognise themselves, rather than being forced to look at reductionist portrayals, legitimised under the banner of science.

- Description: The photoelectrochemistry of metallo-octacarboxyphthalocyanines (MOCPc, where M = Zn or Si(OH)2) integrated with MWCNTs for the development of dye-sensitized solar cells (DSSCs) is reported. The DSSC performance (obtained from the photo-chronoamperometric and photo-impedimetric data) decreased as ZnOCPc > (OH)2SiOCPc. The incorporation of the MWCNTs on the surface of the TiO2 film (MOCPc–MWCNT systems) gave higher photocurrent density than the bare MOCPc complexes. Also, from the EIS results, the MOCPc–MWCNT hybrids gave faster charge transport kinetics (approximately three times faster) compared to the bare MOCPc complexes. The electron lifetime was slightly longer (ca. 6 ms) at the ZnOCPc systems than at the (OH)2SiOCPc system (ca. 4 ms) meaning that the presence of the MWCNTs on the surface of the TiO2 film did not show any significant improvement on preventing charge recombination process.

Adipic acid was also formed after long reaction times. The selectivity for 2-cyclohexene-1-one is favoured when Cl16PcFe or PcCo catalysts are employed, while PcFe is selective towards the formation of 2-cyclohexene-1-ol. The Cl16PcFe catalyst is transformed into a μ-oxo dimer (Cl16PcFeIIIOIIIFePcCl16) during the oxidation process. The catalytic process using the unsubstituted PcCoII and PcFeII catalysts involved PcMIII species as an intermediate. The active form of the Cl16PcFe catalyst was stable to degradation in that it was still active even after 4 weeks of continued catalysis.

- Description: Polybenzimidazole fibers, with an average diameter of 262 nm, were produced by the process of electrospinning. These fibers were used as a solid support material for the immobilization of oxovanadium(IV) which was achieved via a reaction with vanadyl sulfate. The oxovanadium(IV)-functionalized nanofibers were used as heterogeneous catalysts for the oxidation of thioanisole under both batch and pseudo-continuous flow conditions with great success. Under batch conditions near quantitative oxidation of thioanisole was achieved in under 90 min, even after four successive catalytic reactions. Under continuous conditions, excellent conversion of thioanisole was maintained throughout the period studied at flow rates of up to 2 mLh−1. This study, therefore, proposes that electrospun polybenzimidazole nanofibers, with their small diameters, impressive chemical and thermal stability, as well as coordinating benzimidazole group, may be a desirable support material for immobilization of homogeneous catalysts.

- Description: Insect pests, especially lepidopterous stem borers, cause significant losses to cereal production at local and global scales. The borer species associated with cultivated grain cereals and alternative wild host plants were identified at three elevation gradients in the former Transkei area of the Eastern Cape Province, South Africa. Stem borer sampling was undertaken during maize crop and non-crop seasons in Port St Johns (PSJ), Mthatha and Elliot, sites that differ in elevation above sea level, annual precipitation and temperature. Morphologically identical borers retrieved from each sampling unit were recorded, processed and reared to adult stage for identification at an ambient temperature range of 25–28°C and a relative humidity range of 65–80%. Specimens were sorted first into morphospecies (MS1–MS6) based on external morphological characteristics. Six stem borer species (Busseola fusca, Sesamia calamistis, Eldana saccharina, Chilo orichalcocili, Chilo sacchariphagus and Chilo partellus) were collected during the survey. E. saccharina and C. orichalcocili were absent in PSJ; in Elliot, E. saccahrina occurred in very low numbers whereas C. partellus was absent. Busseola fusca was more abundant at high altitude (i.e. Elliot). Borer community structure in the study showed similarity between Mthatha and Elliot. Species richness and abundance across study sites were not statistically significant (P > 0.05). The study provides information on borer species occurrence in the study area, and techniques to identify larval stages for effective control by local farmers and extension workers. When designing strategies for borer control, elevation may not be a major concern at this spatial scale.

- Description: Insect pests, especially lepidopterous stem borers, cause significant losses to cereal production at local and global scales. The borer species associated with cultivated grain cereals and alternative wild host plants were identified at three elevation gradients in the former Transkei area of the Eastern Cape Province, South Africa. Stem borer sampling was undertaken during maize crop and non-crop seasons in Port St Johns (PSJ), Mthatha and Elliot, sites that differ in elevation above sea level, annual precipitation and temperature. Morphologically identical borers retrieved from each sampling unit were recorded, processed and reared to adult stage for identification at an ambient temperature range of 25–28°C and a relative humidity range of 65–80%. Specimens were sorted first into morphospecies (MS1–MS6) based on external morphological characteristics. Six stem borer species (Busseola fusca, Sesamia calamistis, Eldana saccharina, Chilo orichalcocili, Chilo sacchariphagus and Chilo partellus) were collected during the survey. E. saccharina and C. orichalcocili were absent in PSJ; in Elliot, E. saccahrina occurred in very low numbers whereas C. partellus was absent. Busseola fusca was more abundant at high altitude (i.e. Elliot). Borer community structure in the study showed similarity between Mthatha and Elliot. Species richness and abundance across study sites were not statistically significant (P > 0.05). The study provides information on borer species occurrence in the study area, and techniques to identify larval stages for effective control by local farmers and extension workers. When designing strategies for borer control, elevation may not be a major concern at this spatial scale.

- Description: Thiazoles have shown a broad range of biological activities and are found in many potent biologically active molecules such as Sulfathiazol (antimicrobial drug), Ritonavir (antiretroviral drug), Abafungin (antifungal drug), and Tiazofurin (antineoplastic drug) [1]. They have exhibited some degree of plant growth regulatory and antifungal activities [2], whilst some thiazoles have shown anti-infective [3] as well as antibacterial activities [4]. The regio-controlled synthesis of 2,5-disubstituted and 2,4,5-trisubstituted thiazoles from ethyl-2-bromo-5-chloro-4-thiazolecarboxylates using sequential palladium-catalyzed coupling reactions has been reported [5]. An efficient general method for the preparation of 2,4-di- and trisubsituted thiazoles is via P–TsOH. H2O-Catalyzed cyclization of trisubstituted propargylic alcohols with thioamides has been accomplished with moderate to excellent product yields under mild and standard conditions [6]. In the presence of triethylamine, (Z)-(2-acetoxyl-1-alkenyl) phenyl-λ3 iodanes reacts with thioureas or thioamides in methanol to afford 2,4- disubstituted thiazoles in good yields. The reaction is thought to proceed by the generation of highly reactive α-λ3 iodanyl ketones through ester exchange of the β-acetoxy group with liberation of methyl acetate, followed by nucleophilic substitutions with thioureas or thioamides [7].

- Description: We report on the synthesis of Ni nanoparticles (NiNPs) and their application in electrocatalysis in comparison with nickel phthalocyanine (NiPc). UV–vis spectroscopy, powder X-ray diffraction, transmission electron microscopy and electron paramagnetic resonance were used in the characterization of NiNPs. Cyclic voltammetry and electrochemical impedance spectroscopy were used in electrocatalytic studies of amitrole on the glassy carbon electrode modified with NiNPs. The apparent and catalytic rate constants for amitrole on the NiNP-GCE were found to be 2.58 × 10−5 cm s−1 and 1.11 × 103 M−1 s−1, respectively.

The formation of the conjugates was confirmed by X-ray diffraction and transmission electron microscope techniques. Single walled carbon nanotube conjugates of TaPc complexes showed the best catalysis as well as less passivation for bisphenol A detection and significant recovery of ∼98% compared to gold nanoparticle conjugates.
