- Description: This report on the diatoms (Bacillariophyta) of the Sundays and Great Fish rivers is divided into two sections. In Volume 1 the taxonomy of the diatoms from these two river systems is examined. A total of 331 taxa were studied, of which 5 are described as new species. Two taxa are given new names, since they were previously known under later homonyms. Twenty seven taxa were recorded for the first time from South Africa, while a number of species are given amended descriptions in the light of fresh information obtained concerning them. A few taxa (chiefly South African forms) have been united with previously described taxa. For the remainder of the taxa mentioned in the text, comments are passed with regard to their taxonomy and dimensions, based on facts gained through literature surveys and personal observations of the relevant taxa. As far as possible all taxa examined in Volume 1 are illustrated, and these illustrations are presented in Volume 3 of this study. Volume 2 deals with the ecology of the diatoms in the Sundays River. Special emphasis is laid on relating the composition of the diatom associations at the selected stations to the ambient salinity and ionic composition of the water. A new index, the Salinity Tolerance Index (STI), is proposed to summarize by means of a numerical value the information obtained from the analysis of a diatom association with respect to the character of the component taxa, and to indicate whether the association is derived from a freshwater, brackish water or marine source. The efficacy of the STI is evaluated in the text. The effect of the geological formations of the Sundays River valley and the underground waters emanating from them is discussed in the text. The consequences of a severe drought in the lower Sundays River, including hypersaline water in the upper estuary, on the diatom flora are also investigated.

- Description: Pontederia cordata L. (Pontederiaceae) is a tristylous invasive macrophyte – originating from North and South America – that has caused detrimental environmental, agricultural and socio-economic impacts in South Africa (SA). This novel study investigates the invasive ecology of P. cordata in SA by determining population genetics, pollination ecology and floral traits. Preliminary field surveys suggest that only one of three tristylous forms of P. cordata is invading SA and no seeds have been observed in any invasive populations. This study therefore determined the population genetics, mode of spread of P. cordata in SA and possible reasons for the lack of seed production, as well as providing suggestions for future control and management strategies. Inter Simple Sequence Repeats of leaf samples from invasive populations in SA and the native range of the United States of America (USA) were performed to determine the population genetics of P. cordata. The clarification of population structure of an alien invasive plant can provide insight into founder effects, introduction events and modes of spread and is important for the development of management plans such as biological control. Results from the genetic analyses indicated that P. cordata populations have low genetic diversity within and amongst invasive populations in comparison to native populations. This suggests that high gene flow and sexual reproduction is not present in invasive populations, and that only a single or very few introductory events have occurred in SA. Furthermore, invasive P. cordata populations shared the highest genetic similarity with native samples from Belle Haven, Virginia, USA, and thus further sampling and future genetic surveys should be conducted in this area to identify source populations to survey for potential biological control agents. Following these findings, I investigated whether sexual reproduction and seed production is absent from invasive P. cordata populations in SA as speculated. Floral traits from populations throughout all the invaded provinces were measured and, along with pollen grain measurements, it was determined that only short-morphed plants are present in SA. It was speculated that the absence of native pollinators in the invasive range may be responsible for the absence of sexual reproduction. However, a pollination study confirmed the presence of generalist insect pollinators. Thereafter, artificial pollination experiments on 8 865 flowers were conducted to determine whether an incompatibility system was present which prevented seed production. No seeds were produced and it was concluded that illegitimate pollination of the short-morphed plants prevented seed production and rhizomes are responsible for the invasion of P. cordata throughout SA. The implications of these findings and possible management strategies such as biological control is discussed in Chapter 4. These findings suggest that control programmes should target the plants rhizomes to prevent and reduce spread. Preventing the introduction of medium- and long-morphed plants into SA is crucial to prevent P. cordata from producing seeds and intensifying invasion further through both asexual and sexual spread.

It is the most distant area from the equator in which pineapples are grown commercially, most other areas lying between latitudes 25°N and 25°S (Collins, 1960). The prevailing climatic conditions may be considered as being adverse to the growth of a plant which originated in the tropics. Girton, (1962) considers the area to be unsuitable for commercial production because of the cold weather experienced during winter. Seasonal fluctuations in growth and nutrient levels of the plant have been referred to by van Lelyveld, (1964), but these have never been studied locally. Because of the profound effect of temperature on the growth and nutrient requirements of any plant, it was deemed necessary to investigate the effects of the cooler conditions on the growth and nutrient levels of the pineapple plant, the results of which may give some indications of seasonal nutrient requirements.

- Description: Cajanus cajan (L.) Millsp. (Pigeonpea) belongs to the Leguminosae genus Cajanus which is composed of 34 species. Pigeonpea is the only cultivated member of the genus, while the remaining species are wild relatives belonging mainly to the secondary gene pool. DNA sequence data from the nuclear ITS region and the chloroplast trnL-F spacer were utilized to investigate the phylogenetic relationships between Cajanus and five other allied genera in the subtribe Cajaninae. This study revealed the non-monophyly of Cajanus and Rhynchosia and supported the monophyly of Eriosema and Flemingia, but more sampling ,especially from the large genera of Rhynchosia and Eriosema, is recommend to adequately test the hypothesis of generic monophyly. The phylogenetic relationships within the genus Cajanus resolved Cajanus scarabaeoides (L.) Thouars as the most basal species in the Cajanus clade. The study also utilized Single Nucleotide Polymorphism (SNP) markers derived from low copy orthologous genes and genotyped using the high throughput SNP-OPA Illumina golden gate assay. The aim was to understand phylogenetic and domestication history, genetic structure, patterns of genetic diversity, gene flow and historical hybridization between Cajanus cajan (pigeonpea) and wild relatives. The neighbor-joining tree resolved well-supported clusters, which reflect the distinctiveness of species and congruence with their geographical origin. It supported the ITS based phylogeny and resolved C. scarabaeoides as basal to the Cajanus clade. The phylogenetic signal and genetic signatures revealed insights into the domestication history of pigeonpea. Our results supported Cajanus cajanifolius as the presumed progenitor of pigeonpea and we speculate that for pigeonpea there was a single major domestication event in India. Genetic admixture and historical hybridization were evident between pigeonpea and wild relatives. Abundant allelic variation and genetic diversity was found in the wild relatives, with the exception of wild species from Australia, as compared to the domesticated pigeonpea. There was a reduction of about 75% in genetic polymorphism in domesticated pigeonpea as compared to the wild relatives, indicating a severe “domestication bottleneck” during pigeonpea domestication. We discovered SNP markers associated with disease resistance (NBS-LRR) loci. The SNPs were mined in a comparison of BAC-end sequences (BES) of C. cajan and amplicons of the wild species, C. scarabaeoides. A total of ~3000 SNPs were identified from 304 BES. These SNPs could potentially be used in constructing a genetic map and for marker assisted breeding.

- Description: Chromolaena odorata (L.) King and Robinson is an alien invasive weed to most of the Old World tropical regions of the earth, including South Africa where it is morphologically distinct from most other C. odorata plants examined from both its native and invasive range. It is thought that these morphological differences are related to difficulties encountered in successful establishment of biological control agents on the South African population of C. odorata. It has been postulated that the source population of the South African population will harbour potential biocontrol agents that will be suited to successful establishment on the South African plants. Several morphological, cytological and isozyme studies have been attempted to identify the source population of the South African population, but these have failed to identify the origin of the South African population. In this dissertation two PCR-based methods were attempted, in an investigation into whether the morphological differences and difficulties in establishment of biocontrol agents have a genetic basis. The two techniques attempted were: Inter Simple Sequence Repeat (ISSR) amplification, and DNA sequencing. Results could not be obtained using the ISSR method, and the reason for this was not discovered despite extensive trials. The internal transcribed spacer region and the external transcribed spacer region sequences were obtained from five samples, and compared. It was found that the ETS region gave more phylogenetic signal at the intraspecific level than the ITS region. However, due to difficulties in amplification of the external transcribed spacer region, work here focussed on obtaining Internal Transcribed Spacer sequences for 61 samples. Each of the samples sequenced had a unique ITS sequence, displaying a high level of intraspecific genetic diversity. The degree of this diversity is discussed with reference to the possible influences of polyploidy and concerted evolution on genetic structure. The ITS data indicated that some of the physical traits used to define ‘morphotypes’ of C. odorata were not correlated to genotype. From discussion and comparison of morphological character distributions and the ITS-based phylogeography it is suggested that the geographical origin of the South African population is Greater Antilelan, rather than from the continents of North and South America, which is where the Australasian, West African and Mauritian infestations are suggested to have originated.

- Description: Approaching the ecological rehabilitation of an open-cast phosphate mine in the West Coast of South Africa during the post-operational phase presented some challenges. The area was extensively modified during the mining operations. Soils from different layers were mixed with topsoil being covered by subsoil, overburden dumps and tailing dams being constructed resulting in extensive cross-zoned soils. Large areas of subsoil areas were exposed on the mine floor with localized and small scale salinity being evident. The modified topography as well as the complex new surface material posed a challenge in terms of identifying suitable local species that could be used to rehabilitate the post-mining environment. The mine area was heavily infested with woody alien invasive plants, such as Acacia cyclops, established in an attempt to reduce the dust and little natural vegetation cover was present. In the arid west coast environment, the four-month-long winter growing season is followed by hot and windy dry summers (Chapter 2) presenting a challenge reestablishing local vegetation in modified soils. Moreover, little was known about the local vegetation in terms of their propagation and use in stabilization techniques as an alternative to exotic vegetation such as the A. cyclops, which had been more often used in revegetation projects. A study was conducted to determine the most efficient and cost-effective methods of vegetative rehabilitation of the Chemfos site (Chapter 1). A review of the literature available at the time as well as approaches that were successfully implemented in other Western Cape rehabilitation projects such as the Du Toitskloof Pass and the Sishen–Saldanha railway line (Chapter 3), were considered. Previous studies on the west coast dunes at Blaauwberg had shown using Marram grass as a dune stabilizer was most successful. Marram grass was tested in trials alongside local grasses, such as Chaetobromus dregeanus and Ehrharta villosa, that showed potential but had not been formally evaluated. The environmental context of Chemfos (Chapter 2) as well as the Conceptual Rehabilitation Plan that considered the modified environment, soils and closure objectives of the mine, were reviewed to determine the most pressing rehabilitationrelated questions that required answers. This led to the final experimental design that was implemented mid-winter in 1996 (Chapter 5). The trials were implemented in the areas perceived to be the most difficult to rehabilitate, namely the mobile sands in the tailings dam and the exposed subsoil or mine floor areas. The use of brushwood together with specific plants and seeds appropriate for the use in either sandy soil or subsoil were evaluated in a variety of combinations and application densities to determine the most effective treatment combination at the minimum effective density. The best initial cover of the tailings dam were recorded in the Ammophila arenaria trials but the use of the local grass species Ehrharta villosa, performed better from year two onwards and was much cheaper to establish. On the subsoil, the trials where a cover of topsoil was used performed better than the combination trials. This indicated that topsoil placement on post-operational phase shaped subsoils during the mining operation to be the most desirable treatment. Initial results of the trials were used as a basis for developing the rehabilitation techniques that were rolled out across the Chemfos landscape. The approaches were refined as indications of trial responses became evident. Lessons learned were incorporated in the adaptive management approach that was followed and the rehabilitation techniques (Chapter 6) were continually re-evaluated and adjusted. This resulted in a significant step towards achieving the overall research objective of finding cost-effective approaches to rehabilitation. Components such as refinement of the seed collection and processing techniques (Chapter 7) where the post-harvest processing cost was significantly reduced by introducing specially designed drying racks. Processing techniques were adjusted to suit the different species, and a variety of mechanical processing options were explored. The scale of the Chemfos project led to the development of new techniques of manufacturing a smoke-derived germination stimulant (Chapter 8) since commercial availability of these products was very limited. The development of FireGrow assisted in the overall aim of reducing cost by increasing germination of seed in the rehabilitation sites using a very cost-effective smoke concentrate. Socio-economical aspects were considered during the implementation as well as the post-closure phases of the rehabilitation and BHPBilliton invested through the agency of the SAMANCOR Trust. This led to the development of livelihoods of the staff that remained in the area and that lived in the mine village. The demography of the population has changed over time in the Green Village as well as the skills that the inhabitants have developed. Thus, the new economic opportunities that were pursued have brought a new lease on life beyond the lifespan of the mine (Chapter 9) once the closure certificate had been issued.

Contemporary pollen spectra from the Natal Drakensberg and their relation to associated vegetation communities

- Description: The research focuses on the contemporary pollen rain-vegetation relationship for vegetation communities within the Natal Drakensberg, a region which is recognized as having the potential for extensive palynological investigations. The objective was to investigate the fundamental assumption underlying palynology, viz: that the pollen rain of a particular region is indicative or representative of the existing vegetation of that region. If the modern pollen rain is indicative of and/or distinctive for a particular vegetation community then the principle of methodological uniformitarianism can be applied, which states that the present day patterns and relationships can provide a factual basis for the reconstruction of the past through the extrapolation of modern analogues backwards in time. A vegetation survey was conducted in thirteen communities identified as pertinent to the research and a two-year modern pollen rain sampling programme was carried out, using both surface soil and pollen trap samples as a means of recording and quantifying the pollen rain. Once the necessary sample preparation and pollen counts had been performed, descriptive and numerical/statistical methods were employed to determine and describe the existing pollen-vegetation relationship. Descriptive analysis of the data sets was carried out with the aid of spectra depicted as rotated bar graphs and representing the relative percentage frequencies of the collected/counted taxa. Annual and seasonal pollen influx values were calculated and presented. Analysis of variance was applied to test various hypotheses related to sampling strategy and pollen influx variation. Statistical methods employed were two-way indicator species analysis (a classification analysis technique), detrended correspondence analysis and principal components analysis (ordination techniques), canonical correlation analysis (for data set association) and multiple discriminant analysis (for determination of vegetation zonal indices). The latter technique allowed for the probability of modern analogues to be assessed which are necessary for accurate interpretation of fossil pollen assemblages if the assumption under investigation is correct. The findings of the study were that the modern pollen rain-vegetation assumption holds true. Recommendations are put forward regarding future contemporary pollen studies with regards to the number of soil and pollen trap samples required, the magnitude of the pollen count and the numerical/statistical techniques most appropriate to clearly interpret the results. The conclusions are that future fossil pollen spectra can be expected to provide a good indication of former regional vegetation patterns for the study region. The study has extended the limited understanding of the contemporary pollen rain-vegetation relationship in South Africa and enables the interpretation of fossil pollen spectra to be carried out with greater confidence. This in turn lends greater credibility to possible Quaternary environmental change models required to help understand present and possible future environmental change.

The utilisation of Asolla filiculoides Lam. as a biofertiliser under dryland conditions

- Description: The response of wheat to soil fertilised with varying quantities of the water fern Azolla filiculoides was investigated. Experiments were conducted to differentiate between the effects of increased soil mineral status and water status. In the preliminary investigation, experiments were carried out in the greenhouse using potted wheat grown in sand with varying proportions of A. filiculoides that had been subjected tovarious pre-treatments. The pre-treatments were fresh, dry and heated A. filiculoides applied at 20%, 50% and 80% volume per 3000 ml. There were significant differences in the measured growth parameters between the plants grown in the various treatments. In addition, the grain yield of wheat plants varied with the different treatments. Results of the preliminary study showed that the addition of heated and dried A. filiculoides resulted in significantly better growth than the addition of fresh A. filiculoides in sand. For fresh biomass, grain weights, Leaf area ratio (LAR) and relative growth rate (RGR), the performance of dried A. filiculoides was as good as that of the heated A. filiculoides. Productivity of wheat in the heated treatments increased significantly with increasing proportion of A. filiculoides added to sand, while in dry treatments there were no significant increases in productivity in the preliminary study. This supported the hypothesis that A. filiculoides, a notorious water weed can be put to agricultural use under dryland conditions in poor nutrient soils. Further investigations using dried A. filiculoides in sand and topsoil showed that the use of the same amounts of the dried fern made no significant short term impact on topsoil grown winter wheat but significantly improved the productivity of wheat in sand. Results showed that the addition of dried 20% Azolla to sand improved the soil fertility to levels equalling the quality of the control topsoil, but the addition of 80% Azolla to sand led to significantly greater wheat productivity than all other treatments. The addition of dried 20% Azolla (8.14 × 10^3 kg ha^(-1)) in sand produced as much wheat biomass as the addition of the recommended NPK fertiliser (30 kg N ha^(-1)) to sand. A comparison between the topsoil and sand-grown plants showed differences in flowering time but these had no effect on the final grain and above ground biomass.

- Description: Rapid, anthropogenic climate change is a contemporary phenomenon which is threatening natural ecosystems. Mountains are globally important, housing high levels of plant and animal biodiversity, as well as being important suppliers of water with 50% of mountainous regions being essential for the provision of water to downstream inhabitants. Owing to their importance, an assessment of the historical, current and potential impacts of climate change on the south-eastern Great Escarpment encompassing South Africa and Lesotho is necessary and is the primary aim of this thesis. In order to assess the historical and current impacts of climate change, historical rainfall records were collected from 78 sites throughout the Eastern Cape. These records were used to determine rainfall trends and trends in the frequency of daily extreme events. Climate change projections predict increasing trends in summer and annual rainfall, and a marginal increase to a slightly decreasing trend in winter rainfall, depending on if statistically downscaled or dynamically downscaled projections are examined. Extreme events are also predicted to increase in frequency. Results showed trends which generally agree with the predictions, except for extreme events for which there was insufficient data to support the projections. This has implications for farmers in the area and validates the downscaled Regional Climate Models (RCMs) to be used for the predictive analysis determining the potential future impacts of climate change on plant species' distributions in the south-eastern Great Escarpment. In order to assess montane plant species to future climate predictions, occurrence records of 46 species were correlated to rainfall and temperature predictor variables and predictions for species richness and beta diversity were made for current and future climates. This predictive analysis used and compared two correlative species distribution modelling methods - one method used one correlating algorithm in a nuanced manner and the other method used five correlating algorithms in an ensemble. A trend of range restriction to higher elevations for the 46 species analysed was as expected from a warming climate. The nuanced, single algorithm modelling method produced less conservative models. The predicted trend of an elevational increase in montane vegetation in this region points to Lesotho and the Drakensberg highlands as being an important refugium for montane plant taxa of the southeastern Great Escarpment. The Maloti Drakensberg Transfrontier Conservation Area hence needs to be expanded and its mandate properly enforced to protect this region adequately.

- Description: The symbiotic unit of an arbuscular mycorrhizal fungus and its host is able toachieve and maintain far higher inflow of nutrients than non-mycorrhizal roots. The colonization strategy of the mycobiont within the plant is intrinsic to the symbiosis with respect to both structural adaptations and nutrient exchange. An investigation into the effect of Glomus etunicatum colonization on the structure and phloem transport in Eragrostis curvula (Umgeni) allowed for greater insight into the dynamic of the symbiosis. The combined use of stains (such as Trypan Blue, Chlorazol Black, Safranin and Fast Green), and techniques, (such as freeze-microtome transverse sectioning and permanent slide preparations) contributed to a successful general observation of an intermediate colonization strategy using light microscopy methods. However, clarity into structural detail of mycorrhizal forms required electron microscopy studies. The SEM method used with freeze fracture was a relatively quick and simple method allowing for the observation of surface and internal features. The TEM method allowed for highresolution images providing insight into the variations in the apoplasmic compartmental form, and how this may relate to the function of the symbiosis with regard to fungal coils or arbuscules. The apoplasmic nature of mycorrhizas was substantiated and no symplasmic connections were found between symbionts. Fluorescence studies demonstrated that 5,6-carboxyfluorescein was transported through the phloem into the roots of E. curvula, but remained predominantly in the root phloem. Unloading only occurred in optimal nutrient exchange areas of meristimatic lateral or apical growth regions. It was not possible, using fluorescence techniques and related equipment available, to conclusively establish if there were symplasmic connections between the mycobiont and its host or if bidirectional transfer of nutrients occurred at the same interface.

- Description: The study was conducted to investigate the effects of: (i) varying the level of supply of the heavy metals iron, manganese, copper and zinc. (ii) varying the ratio between different pairs of metals in the medium. In particular the iron:manganese and zinc:copper ratios were studied. Initially the two fungi Aspergillus niger van Tieghem, (variety and strain) and Penicillium notatum Westling, were used. Penicillium notatum Westling was subsequently discarded, in favour of Penicillium glancum Link, because it did not sporulate freely in liquid culture. The fungi were grown in controlled nutrient solutions, and during the course of tho growth and development, the form and sporulation of the felts was noted. After a period of growth, the felts were removed, dried and weighed. The pH of the liquor was measured. the results were studied to determine the effect of varying levels of supply of the heavy metals, and of the varying heavy metal ratios in the culture solution. In the investigation of the effect of varying the level of supply of individual heavy metals, optimum concentrations were demonstrated for copper and manganese. Increaning the concentration of pairs of heavy metals cimultaneously was found to influence the appearance and degree of symptoms of toxicity. Cultures of Aspergillus and Penicillium were found to be able to tolerate concentrations of copper, in particular, considerably greater than the observed optima, when zinc was present in equal concentration. Citric acid, and subsequently ethylene- diaminetetra-acetic acid, were used as chelating agents, in order to prevent the precipitation of the metals in the culture solution during autoclaving. It was found that the use of chelating agents markedly reduced symptoms of toxicity. There was no conclusive evidence that the iron:manganese ratio in the culture medium was an important factor in the growth and development of cultures of Aspergillus and Penicillium. However there is considerable evidence that in cultures of Penicillium, the zinc:copper ratio in the medium is of some importance in the determination of the dry weight yield trends. This effect was not demonstrated in cultures of Aspergillus.

- Description: Namibia, Juttadinteria and Dracophilus, the three genera belonging to the subtribe Dracophilinae Schwantes (Aizoaceae: Ruschioideae) were revised. Macro-morphology, leaf anatomy and micromorphology of leaf epidermides, pollen, tapetal orbicules and seed were studied in order to test taxon limits, to determine relationships between taxa, and to improve knowledge of their characteristics as well as the taxonomy of the subtribe. The investigation was based on herbarium specimens, field observations and cultivated, living plants. Phenetic cluster analyses were used to confirm species while intergeneric and interspecific relationships of the taxa so indicated were later tested by means of a phylogenetic analysis. The existence of the three genera was provisionally upheld by this study although phylogenetic analysis showed Namibia nested as a strongly supported monophyletic group within a poorly supported luttadinteria. Further work is needed to clarify whether Namibia should be sunk into Juttadinteria. The latest treatments of Juttadinteria and Dracophilus by Hartmann (2001) were supprted. However, in contrast to her latest treatment of Namibia, N. pomonae was sunk into N. cinerea and N. ponderosa was reinstated and typified. Juttadinteria was found to be a poorly resolved genus with many intergrading and overlapping characters, possible due to recent speciation. Possible subspecific groups within J. deserticola and J. simpsonii were indicated by the phenetic study but further work is needed before any formal infraspecific taxonomic rank can be assigned to them. Detailed descriptions of the nine elucidated species and the three genera as well as new keys for their identification are provided in order to facilitate further work in this group. Distribution maps are provided for the genera and species, and the subtribe. Variation of character states within the subtribe, distribution area, ecology and biology are discussed.

- Description: Wetlands provide a number of valuable functions to both the surrounding environment and society. The anaerobic conditions created by flooding in wetlands provide a habitat that supports unique assemblages of plant life. High altitude wetlands are amongst the most species-rich in South Africa. They house a number of rare species and play a vital role in the supply of water to lower lying areas. These are some of the reasons that mountain wetlands are of high conservation value. A phytosociological study was undertaken on the high altitude wetlands of the Great Escarpment with the aim of classifying the plant communities and identifying the environmental drivers of plant community patterns within these ecosystems. Data collection was focused in the Eastern Cape and was supplemented with data from existing studies to gain a more complete understanding of the wetlands of the Great Escarpment of South Africa. Using the Braun-Blanquet approach, Hierarchical Cluster Analysis and Indicator Species Analysis; five broad wetland groups were identified, comprised of 33 individual plant communities and 81 indicator species. Multivariate analysis, including Canonical Correspondence Analysis revealed that the effects of altitude, such as temperature and rainfall, are the most significant large-scale drivers of vegetation patterns. Smaller scale drivers include wetness and soil nutrients including nitrogen, phosphorus, electrical conductivity, sodium, and organic content. The identification of indicator species served to reveal potentially important wetland species across different areas of the Great Escarpment. The effects of altitude on plant community patterns highlights the susceptibility of the high altitude specific communities to upward temperature zone shifts resulting from global warming. Other threats include livestock trampling, water extraction, and land use change for agricultural purposes. The relative absence of alien species in these wetlands gives an indication of their pristine condition and therefore their importance as a reference from which they may be monitored. A large proportion of the wetlands studied here occur outside protected areas, and given the rate of wetland loss in South Africa, it is important that continued effective land management is practiced to ensure that these ecosystems are conserved in the future .

- Description: Rehabilitation is a dynamic process influenced by factors related to more than one field of ecology. It is therefore necessary to consider all these components when assessing the rehabilitation, although in the initial stages the successful revegetation of the disturbed areas is the most important criterion. Richards Bay Minerals, on whose mining site this project was carried out, is dredge mining heavy minerals on the north coast of Natal, where they have rehabilitated mined areas since 1978. This project has been carried out to establish: 1) The success of their dune forest rehabilitation using quantitative techniques. 2) The available seed bank in their rehabilitation stands. 3) The similarities in the succession taking place in rehabilitation stands compared to the revegetated stands in the vicinity of Richards Bay. 4) The best methods for creating alternative vegetation communities, especially grasslands, with a high species diversity on the mined tailings. This study reviews only the success of rehabilitation of the natural vegetation but other studies focusing on the insect, reptile, mammal and bird populations are also being undertaken by other researchers. No particular method of determining the success of vegetation rehabilitation has been chosen by restoration ecologists. Therefore in this study a broad range of quantitative techniques were used to show whether successional changes are occurring in the vegetation and physical environment. The results obtained from sampling the rehabilitated vegetation have shown that both the species richness and diversity are increasing as the returned vegetation matures. Levels of soil properties such as Sodium, Phosphate, Calcium and percentage organic matter have also risen with increasing stand age. Community complexity is also increasing with stand age, and TWINSPAN and DECORANA plots have separated out the differently aged stands based on their differences. A "pilot" study was done on the seed bank present in the rehabilitation stands. This has shown the presence of large amounts of early successional, mostly herbaceous species. Seeds of later successional and woody species were scarce which may be a result of the sampling intensity used. However seeds of late successional ground cover species were found in the older stands. Comparisons between the natural revegetation of disturbed areas in the vicinity of Richards Bay and the rehabilitation stands revealed similarities in both species composition and complexity. Species richness and diversity values are comparatively similar for the younger revegetated and older rehabilitation stands, and lWlNSPAN and DECORANA analysis techniques clustered the samples recorded from these areas in close proximity on their relative plots. The oldest revegetated sites contain a number of species found in the rehabilitated vegetation but as Acacia karroo has thinned-out in these stands many of these other species are now mature individuals. Attempts at rehabilitating an area of grassland at Richards Bay Minerals has not produced satisfactory species diversity and experimental manipulations were used to try and increase the diversity of the existing Eragrostis curvula dominated community. Of the several treatments used for the manipulation, a combination of burning and further topsoiling was the most successful in reducing Eragrostis importance and in increasing the species richness. Grassland topsoil spread directly onto the bare tailings produced an extensive vegetation covering over a short period but species richness was not significantly greater than for the existing Eragrostis dominated grassland, and further treatments and management needs to continue if this technique is to be employed. Only a limited amount of alien infestation of the rehabilitated areas was evident from the sampling undertaken during this research. As the removal of alien plants is an ongoing process and the rehabilitation stands are continuously monitored to identify any new invaders, this is not expected to become a problem. From the results of work done overseas and the rehabilitation carried out in South Africa it appears that it is possible to return natural vegetation communities on mined areas. That this is a lengthy process is to be expected but by manipulating the vegetation and continuously monitoring the process it may be possible to speed up development. Areas in need of further research have been identified based on the findings of this project. This will help to reinforce the undertaking of management proposals that will enhance the vegetation recovery and the success of the rehabilitation programme.

- Description: The relationship between leaf architecture, vein anatomy and phloem ultrastructure, and that of possible routes from mesophyll cells to phloem and potential phloem loading method was investigated using species adapted to the southern African climate. The research was based on the hypothesis of Gamaiei and Van Bel, using nothern hemisphere species only (Gamalei 1985a, b, 1989, 1991, VanBeletal. 1988, Van Bel 1992, 1994, 1996, Van Bel & Gamalei 1991, 1992, Gamalei et al. 1992, 1994, 1996, Van Bel 1992a-c, 1993a, b, 1996). The thesis commenced with a survey ofleafarchitecture of the Ranunculaceae, Apocynaceae and Asclepiadaceae. Anatomical and ultrastructural studies followed. Leaf architecture was described according to Hickey (1973). Within the Ranuncuiaceae, leaf architecture was found to be marginally actinodromous. Venation pattern consisted of a widely spaced reticulum of delicate veins, especially in Ranunculus. Leaf architecture of the Apocynaceae was described as pinnate, camptodromous and brochidodromous. The Asclepiadaceae showed less uniformity in terms of leaf architecture, being pinnate and camptodromous, with mostly brochidodromous and, unexpectedly, eucamptodromous patterns of secondary venation. A predominantly common leaf architecture supported the move to amalgamate the two families. As the less advanced eucamptodromous arrangement could represent a more primitive branch of this huge family, the phylogenetic classification of the new amalgamated family is eagerly awaited for discussion. Allocation of vein order allowed comparisons between species and families to be drawn. Reticulum density and vein order anatomy was used to indicate potential routes from mesophyll to phloem. A definite contrast was obvious between the loose arrangement of mesophyll and veins in the mesic Ranunculus, and the close mesophyll and dense venation of the xeric apocynate and asclepiad species, and was related to habitat. Ultrastructural characteristics of companion cells, together with plasmodesmatal abundance, were considered especially important for the determination of minor vein configuration. Descriptions of plasmodesmatal distribution did not consider functional status. In this thesis, vein structure and ultrastructure were considered in relation to phloem loading, not as a demonstration thereof. All three families were designated minor vein configuration type 2a. Two interesting examples that did not adhere to the familial norm, viz. few plasmodesmata and normal companion cells, occurred in the Asclepiadaceae. Secamone alpinii had abundant aggregated plasmodesmata, forming a potential symplasmic continuum from mesophyll to companion cells. The question of plasmodesmatal functionality remained open. Ceropegia carnosa showed folding of the companion cell membrane, but no accompanying wall ingrowths. The folds were suggested to increase surface area for apoplasmic phloem loading in the noted absence of plasmodesmata. Loading routes and methods suggested were based on anatomical and ultrastructural evidence only. Whilst these results were supported by published data for other species of these families, the prediction of the Gamalei and Van Bel hypothesis did not hold true. The relatively primitive Ranunculaceae were expected to have the least advanced type 1 minor vein configuration, with abundance plasmodesmata providing a symplasmic phloem loading pathway. The relatively advanced Apocynaceae and Asc1epiadaceae were predicted to have the most progressive minor vein configuration, type 2b, with specialised transfer cells to maximise apoplasrnic uptake. As families with type 2a minor vein configurations, the Ranunculaceae were more advanced than expected and the Apocynaceae and Asc1epiadaceae less so.

- Description: Introduction: R factor mediated transfer of antibiotic resistance between Enterobacteriaceae has been reported to occur in the mammalian gastrointestinal tract (Farrar et al, 1972; Guinée, 1970; Kasuya, 1964; Reed et al, 1969; Wiedemann et al, 1970). In vivo conjugal transfer of genetic material has also been demonstrated with F¹, F⁺ and Hfr Escherichia coli strains (Jones & Curtiss, 1970). The environment in the lower gastrointestinal tract, where bacteria are abundant, is mainly anaerobic. This is demonstrated by the dominance of obligately anaerobic bacteria such as Bacteroides species (Finegold, 1969; Moore et al, 1969) and direct studies of intestinal gas composition (Askevold, 1956). However, most laboratory investigations of the incidence of R factors and their transfer frequencies have been performed under aerobic conditions using faecal facultative strains. The only investigation of resistance transfer under anaerobic conditions in vitro is that of Mitsuhashi (1965), who reported complete inhibition of transfer of an R factor from a Shigella flexneri donor to an Escherichia coli recipient. In addition, Fisher (1957) reported restriction of chromosomal transfer by an E. coli Hfr strain under anaerobic conditions in various media. On the basis of these results, it could be questioned whether in vivo R factor transfer is in fact possible (Chabbert et al, 1969). The contradictory situation prompted a reexamination of conjugation in facultative strains under anaerobic conditions. Both Fisher (1957) and Mitsuhashi (1965) obtained anaerobic conditions by evacuation. In this investigation, both mating and selection of recombinants were performed under stringent anaerobic conditions using methods developed for the isolation of obligate anaerobes (Hungate, 1969) to obtain a degree of anaerobiosis similar to that found in vivo.

An investigation of the aquatic macroinvertebrate fauna of the southern Great Escarpment (South Africa): insights from ecological and genetic studies

- Description: Biological diversity in freshwater biomes is vital to maintain healthy, functioning ecosystems with resilience to disturbance and the impacts of climate change. Freshwater ecosystems provide essential resources to life on Earth. However, as increasing pressure is being placed on the environment by human population growth, the quality of freshwater resources and the health of these ecosystems are at risk. Mountain streams provide an important source of water and are usually less affected by anthropogenic stressors, compared to lowland freshwaters. These montane streams are therefore of important conservation value and due to their untransformed nature serve as ideal ecosystems for biodiversity studies and as reference sites for studies on environmental change. This study explores aquatic macroinvertebrate biodiversity of the southern Great Escarpment in South Africa. Species assemblages and the environmental variables of each site were sampled from first order streams across five different mountain blocks along the Great Escarpment. Additionally, mitochondrial DNA of three mayfly species (Afroptilum sudafricanum, Demoreptus natalensis and Demoreptus capensis), commonly occurring in the study area, was analysed to compare the genetic diversity between habitat specialist and habitat generalist species. A total of 2 595 macroinvertebrate specimens from 47 families and 86 species were collected with several interesting and potentially new species being discovered. Partitioned diversity analyses indicate that macroinvertebrate diversity varies across mountain blocks. Multivariate analyses indicate that differences in assemblages could be attributed to differences in environmental variables between sites, particularly water velocity, total dissolved solids and salinity. As these environmental variables reflect of the topography of the sites, differences in species assemblages was attributed to difference in topography and therefore biotopes present. Habitat-restricted mayfly species (D. natalensis and D. capensis), occur in distinct populations confined to mountains blocks. Isolation-by-distance analyses further emphasis that these species are genetically restricted by their habitat preference for mountain streams. In contrast, A. sudafricanum, a habitat generalist, showed no indication of genetic structure due to location or distance. Possible cryptic taxa and new species were identified within A. sudafricanum and Dermoreptus respectively. This study provides an important contribution to the baseline data of freshwater macroinvertebrate diversity for the southern Great Escarpment region and provides insights into the considerable genetic diversity of selected aquatic taxa across this region.

- Description: Species possess characteristics that are considered adapted to burning and these allow them to outcompete species and dominate in fire prone environments. It has therefore been proposed that fire might have played a critical role in the observed expansion of the grasslands, during the late Miocene. The aim of this study was (i) to investigate whether plant response to fire was a result of physiology or (ii) whether it was due to phylogenetic history. This was achieved by doing a pair-wise comparison between Panicoideae (and Panicoideae) and non-Panicoideae (Danthonioideae and Aristidoideae) species. Pre-fire characteristics, that would enhance fire frequency and assist with plant recovery after burning, were compared across phylogenies and photosynthetic type. Post fire plant recovery was then followed in a field and pot comparison which examined the re-growth of the leaf canopy area, leaf mass, above-ground biomass and the cost of this to the below-ground biomass. The pre-fire characteristics showed both a photosynthetic and phylogenetic response. It was found that the species showed a greater canopy death during winter and had a lower moisture content than the species. These characteristics would potentially contribute towards a larger fuel load in the species. However, the comparison of the dead standing biomass at the end of winter and the below-ground biomass, showed a phylogenetic response with the Panicoideae having a proportionally larger dead standing biomass and below-ground biomass than the non-Panicoideae. These results suggest that not only did the Panicoideae have a larger potential fuel load but that they also shunted carbon below-ground, enabling a fast recovery after being burned. The post-fire results were more strongly determined by phylogeny than by photosynthetic type. The Panicoideae recovered faster and more completely than the non-Panicoideae grasses, possibly contributing to their success and expansion under conditions of increased fire frequency. Although recovery of the and Panicoideae were similar, frequently burnt grasslands are dominated by the Panicoideae. Hence, this dominance cannot be explained by differences in their fire responses and may be determined by the post-fire environmental conditions that potentially advantage species possessing the photosynthetic pathway. Panicoideae dominance is limited to mesic environments where fire is the likely driver of grassland expansion while more arid environments are dominated by non-Panicoideae species. Representative species from these non-Panicoid subfamilies showed poor recovery after fire. This suggests that factors other than fire were the likely drivers of these xeric grassland expansions. The ability of these subfamilies, and particularly the species, to cope with drought remains a likely selective mechanism that requires further research.
