- Description: This study was undertaken to investigate the taxonomy, biogeography and biology of cow and frilled sharks (Chondrichthyes: Hexanchiformes). This taxon comprises two families, four genera and six extant species. The hexanchoids are a distinctive group of sharks characterized by six or seven paired gill openings, a single dorsal fin and an anal fin. Adult males of this group lack a siphon sac, but have in its place a clasper sac. This structure, which develops along the claspers, is unique to the Hexanchiformes. Hexanchoid sharks are widely distributed in area and depth. This group ranges from coastal bays and harbors along the open coast out across the continental shelf and down along the slopes to considerable depths. They occur from the equatorial zone to sub-polar regions. However, little is known about the ecology and life history of these sharks. Intraspecific variation of meristic counts were generally low for the Hexanchidae, but high for the Chlamydoselachidae, indicating that subpopulations, subspecies or even additional, new species exist within this family. Based on the indicators used in this study, maturity in male frilled sharks was attained at 916 mm TL, perlon sharks between 700 and 800 mm TL, sixgill sharks approximately 3140 mm TL, bigeyed sixgill sharks at about 1250 mm TL and sevengill sharks at approximately 1550 mm TL. Male reproductive success did not appear to be seasonal since males were found to contain viable sperm all year round. Female perlon sharks begin maturing between 950 mm and 1100 mm TL. Gravid females and newborns were absent from the other size classes and it is suspected that they aggregate in different locations to those of adult males and non-breeding females. Adult females are known at 4210 mm TL and immature at 3500 mm TL, However, a more accurate estimate of the size at maturity is wanting. Newborn sixgills were caught off southern Namibia during mid to late summer over three successive seasons. The occurrence of gravid females carrying term embryos during spring months and newborns during the summer months suggests a late spring or summer pupping period. Sixgill and sevengill sharks give birth in areas of high primary productivity. Energetically, this is advantageous for the newborns to be placed in an area with an abundant food source. The rapid growth rates of sixgill and sevengill sharks over the first year would enhance their survivorship since neither species has many predators. The number of female sevengills entering the breeding population is regulated to ensure that some portion of the population is reproductively active at any one time. The "staggering" of females which enter into the breeding population in any given year indicates a two year reproductive cycle. Fecundity estimates for 19 specimens with a largest egg diameter of at least 40 mm indicates a litter size of 67 to 104. The recapture of an adult female sevengill in approximately the same location in which it was tagged suggests that the same individual sharks may return to the same breeding grounds. As with any predators, sharks tend to exploit advantages over their prey. The hexanchoids, especially the sevengill, have evolved complex foraging strategies including social facilitation whereby they actively hunt in packs for large prey species. Sharks of the order Hexanchiformes, although lacking the diversity of the major shark orders, nonetheless play an integral role in the marine environment. The group's success can be attributed to their apical trophic position. In most habitats in which they occur, hexanchoids have no comparable competitors since equivalent sized sympatric squaloids and carcharhinoids feed at a lower trophic level.

- Description: The blacktail, Diplodus capensis, is an inshore sparid fish distributed from Mozambique to Angola. This species forms an important component of coastal fisheries within its distribution, one being the subsistence handline fishery in southern Angola. With this fishery being critically important to the livelihoods of local communities, a biological study and stock assessment was conducted to provide information for the management of this species in southern Angola. However, with molecular evidence suggesting that the Benguela current may have separated the southern African populations of many inshore fish species over two million years ago, a morphological, taxonomic analysis was considered necessary to first investigate whether there was evidence for allopatry in this species. A total of 46 morphometric measurements and 18 counts were carried out on specimens collected from various locations in southern Angola and South Africa. Results were analysed using multi-dimensional scaling (MDS) and the significance of clusters was tested using analysis of similarities (ANOSIM). Biological samples of D. capensis were collected monthly from an unexploited area from April 2008 to March 2009. Additional biological samples were collected from the subsistence fishers in an exploited area during May, June and December 2009. Standard biological laboratory techniques were employed for the lifehistory comparison between the exploited and unexploited area. A per-recruit analysis was conducted using the life-history parameters from both areas in order to assess the current status of the subsistence fishery and to investigate the potential short-falls of the per-recruit assessment approach. The morphometric comparison showed that there was not sufficient evidence for speciation between the southern Angolan and South African populations of D. capensis. There was, however, sufficient morphological evidence to suggest that these populations are separate stocks. This indicated that the existing reference points on which the management of the South African population is based are unsuitable for the Angolan population. Diplodus capensis in southern Angola is omnivorous, feeding predominantly on algae, barnacles and mussels. An ontogenetic shift from algae to barnacles and mussels was correlated with allometric growth patterns in their feeding apparatus. This species is a rudimentary hermaphrodite in southern Angola with peak spawning in June and July. The overall sex ratio (M: F) was 1: 4.7 in the unexploited area and 50% maturity was attained at 149.5mm FL and five years. Diplodus capensis in southern Angola exhibits very slow growth with the maximum age observed being 31 years (validated using mark recapture of chemically injected fish). Females [L(t) = 419.5(1-e⁻°·°⁴⁵⁽t⁻³·⁴ ⁾)] grew significantly faster (LRT, p < 0.05) than males [L(t) = 297.4(1-e⁻°·°⁷⁷⁽t⁻²·⁷⁾)], and females dominated the larger size classes and older age classes. In the exploited area, the length and age frequencies were severely truncated, the maximum observed age was greatly reduced (17 years) and the sex ratio was less female biased at 1: 2.2. Although there was no evidence for a physiological response to exploitation through alterations in growth or size/age at sexual maturity between the two areas, there was an increase in the proportion of small females in the exploited area, which may have been a compensatory response for the loss of large females. A combination of an underestimate of longevity, different estimates of the Von Bertalanffy growth parameters and overestimates of the natural mortality rate in the exploited population resulted in a 92% underestimate of the pristine spawner biomass-per-recruit (SBR) value. An assessment based on the actual pristine SBR estimate from the unexploited area revealed that the subsistence fishery had actually reduced D. capensis to 20% of its pristine SBR levels and highlighted the value of pre-exploitation life-history information for the application of per-recruit models. This study has shown that D. capensis in southern Angola displays life-history characteristics that render it susceptible to overexploitation, even at low levels of fishing pressure. The current lack of infrastructure and enforcement capacity in the fisheries department of Angola renders traditional linefish regulatory tools, such as size limits, bag limits and closed seasons, inappropriate. Therefore, suitably designed marine protected areas are recommended as the best management option for this species.

- Description: The application of high rate algal ponds (HRAP) in the treatment of brewery effluent that met the South African Department of Water Affairs and Forestry's (DWAF) general limits for discharge into a natural water resource of 1998 were tested during a lO-month baseline phase, followed by an 11-month optimization phase. The objective of the baseline phase was to monitor the seasonal performance of HRAPs. The hydraulic retention time (HRT) fluctuated between 11.16 d and 12.00 d in HRAPs. The chemical oxygen demand (COD) increased from 130.12 ± 6.94 mg/L (post-AD), to 171.21 ± 7.99 mg/L (post-HRAP) . The presence of algal cells and evaporation contributed towards an increase in post-HRAP COD. The ammonia (NH₄-N) concentration decreased from 46.59 ± 2.47 mg/L (post-AD), to 1.08 ± 0.12 mg/L (post-HRAP). The nitrite (NO₂- N) concentration remained below 1.00 mg/L in post-pilot plant AD, post-PFP and post-HRAP effluent. The phosphate (PO₄-P) concentration decreased from 29.81 ± 1.39 mg/L (post-AD) to 17.30 ± 1.16 mg/L PO₄-P. The objective of the optimization phase was to manipulate the HRT to achieve the maximum treatment rate that met the DWAF general limits for discharge into a natural water resource of 1998. Nitrogen (as NH₄-N, NO₃-N, NO₂-N) removal efficiency was used as an indicator of nutrient removal success. HRT was influenced by season. The optimal HRT for autumn was 4.30 d at a temperature of 20.53ºC in HRAP A2 (heated) and 18.96ºC in HRAP B2 (ambient). The optimal HRT for summer was 2.74 d at 29.90ºC in HRAP A2 (heated) and 26.36ºC in HRAP B2 (ambient). The COD decreased from 152.33 ± 4.85 mg/L (post-AD) to 95 .00 ± 3.75 mg/L (post-HRAP A2), and to 100.82 ± 5.93 mg/L (post-HRAP B2). The incoming NH₄-N concentration decreased from 42.53 ± 1.38 mg/ L (post-AD), to 1.70 ± 0.81 mg/ L (post-HRAP) . The nitrate (NO₃-N) concentration post-HRAP was 12 - 14 mg/L. The main methods for NH₄-N removal were probably NH₄-N volatilization through algal uptake. HRAPs were able to lower nitrogen and phosphorous concentrations to within the DWAF limits under normal operating conditions. It is recommended that HRAP treated brewery wastewater be used for irrigation after salt removal, or alternatively, for groundwater recharge . Regulatory exemptions would be required for higher than permitted COD and EC concentrations to enable these actions.

- Description: Batoid populations are declining globally but the paucity of information makes management or conservation efforts difficult. Additionally, batoids are mesopredators and are hypothesised to play important ecological roles, but a comprehensive understanding of these roles is also limited. Therefore, information on batoid habitat use, foraging ecology, and resource partitioning is needed. St. Joseph Atoll is a relatively pristine ecosystem that hosts a sympatric dasyatid community (Pastinachus sephen, Urogymnus granulatus, and U. asperrimus). Passive acoustic telemetry identified high levels of long-term site affinity by both juvenile and adult dasyatids to St. Joseph Atoll. Dispersal from the atoll by larger juveniles was also evident. Juveniles displayed restricted horizontal movements in the atoll, but the extent of these movements differed seasonally. Stomach content and stable isotope analyses identified inter-specific prey partitioning (P. sephen juveniles were mollusc specialists and U. granulatus juveniles were crustacean specialists) and intra-specific prey partitioning. Juveniles were reliant upon a seagrass-based food web, whereas adults were reliant on phytoplankton-based food web. Adults fed at higher trophic levels compared to juveniles (mean of 4.6 and 3.4 respectively). Juvenile dasyatids preferentially selected the shallow reef at habitat of the atoll, where there was no evidence for inter-specific habitat partitioning (individuals co-occurred randomly with con-and hetero-specifics). Conversely, resident adults were more reliant on the deeper lagoon. Juveniles selected the reef at habitat likely because it provided refuge from predation and foraging opportunities. However, juveniles were also detected in the lagoon habitat and this appeared to be necessitated by physical factors as they were detected more frequently in the lagoon at low tides and when temperatures on the reef fats became too warm or too cold. All results together suggest that St. Joseph Atoll is a nursery area for these dasyatids. Juveniles may be limited by resources as prey was partitioned between species. However, top-down control by larger sharks was likely a significant influence on habitat selection, as was the physical effects of tide and temperature. Finally, St. Joseph Atoll is suitable for the designation of a Marine Protected Area, which would likely confer high conservation benefits to this dasyatid community.

- Description: Little information exists for the widely occurring but rare porcupine ray Urogymnus asperrimus. This is a concern given that it is a vulnerable species and likely plays an important ecological role in tropical atoll ecosystems. The aims of this thesis were to investigate the trophic and spatial ecology of juvenile porcupine rays within the St. Joseph Atoll, which is hypothesized to be functioning as a nursery for this species. Specific objectives were to determine i) the dietary composition and resource selectivity displayed by juvenile porcupine rays, ii) residency to the atoll, space use, and seasonality in movements, and iii) whether environmental factors affect short-term movements. Gastric lavage was used to collect stomach contents from 55 juveniles and sediment samples were collected. Thirteen juveniles were implanted with acoustic transmitters and monitored for a year by an array of 88 acoustic receivers situated in and around the St. Joseph Atoll. Porcupine rays appeared to be generalist and opportunistic feeders, foraging predominantly on annelids with the highest environmental availability. Polychaetes of the family Capitellidae were the most important prey item (Index of Importance = 35%). Rays only fed on deep infaunal prey, likely because of their foraging behaviour, suggesting they may influence this community. There was a size-related shift in the crustacean families consumed. The tagged rays displayed high residency to the atoll (mean residency index of 64%) and restricted movements, with small core use and activity space areas (mean of 0.5km² and 3km² respectively) and the majority of detections were recorded within 1km of their tagging locations. The rays favoured the sand ats where 88% of detections occurred. Transient use of the lagoon and fringe reef peaked in the north-west monsoon season, indicating a seasonal usage of these habitats. The effects of temperature and water depth on movements were manifested as diel and tidal cyclical patterns in presence. Evidence illustrates that the St. Joseph Atoll constitutes an important nursery for this species, which has been declared a Marine Protected Area. A major determinant behind the dependence of this nursery habitat is related to predator avoidance. Thermoregulatory behaviours were identified, suggesting that this species is vulnerable to climate change.

- Description: The removal of microalgae from high rate algal ponds (HRAP) in waste-water treatment systems remains a constraint to their use in effluent treatment systems. Conventional algae harvesting methods often have high energy demands, take up lots of space, are expensive to operate or are time consuming. The aim of the study was to determine if fish such as Clarias gariepinus and Oreochromis mossambicus, could be used to remove microalgae from waste-water treatment ponds (in the absence/presence of a flocculent in the former and in the absence or presence of pH moderation in the latter), and to investigate the subsequent influence of algae concentration on several water quality parameters. The age of Clarias gariepinus (3-12 months) had a positive relationship with the distance between gill rakers (98.27 to 163.34 μm; y=90.576+4.823*x: R²=0.549; F(1,18)=21.867; p<0.001) and these data suggested that these fish might be efficient at removing algae from HRAP effluent. However, this was not the case, even with flocculent application (but this result might have been confounded by very high pH readings, at which flocculation is less likely to occur). Oreochromis mossambicus removed some of this algae, but the pH was too high for tilapia culture. It was not possible to moderate the increase in pH by keeping tanks in the dark and thus preventing photosynthesis; but pH fluctuation in HRAP effluent could be moderated using CO2 sparging in an attempt to make the environment more hospitable for tilapia (the average pH that was moderated with CO2 was 8.43±0.06, whereas the unmoderated average was 10.65±0.06). However, pH moderation using CO2 sparging did not increase the rate at which algae were removed by O. mossambicus; rather, it compromised O2 concentration which dropped to 4.17±1.26 mg/l after five hours of CO2 sparging, whereas it increased to 20.50±1.41 mg/l in treatments with unadjusted pH over the same period. Fish can be used to remove algae from treated effluent, and Oreochromis mossambicus remains a recommended species. Future work needs to investigate moderating fluctuations in pH and O2 concentration to further facilitate this method of algae removal.

- Description: Physiological stress in farmed abalone can lead to immunosuppression and increase the susceptibility to bacterial, viral and parasitic disease, often followed by mortality. Thus, handling and poor water quality can reduce farm production efficiency. Probiotics in aquaculture have been effective in a wide range of species in enhancing immunity, survival, improving feed utilisation and growth. Three putative probionts identified as a result of in vitro screening had been beneficial to laboratory-reared abalone in a previous study. The aim of this study was to produce an abalone feed that contains a suite of probionts that may promote abalone growth and health under farming conditions. The objectives were to compare growth and physiological responses (i.e., haemocyte and phagocytosis counts) of abalone fed a commercial feed (Abfeed®S 34, Marifeed, Hermanus) supplemented with probiotics (i.e., the probiotic diet) to abalone fed the commercial feed without probiotic supplementation as a control treatment in a factorial design with handling method as an independent variable. This experiment was conducted at HIK Abalone Farm (Pty Ltd) for a period of eight months with initial weight and length 36.1 ± 0.05 g and 58.6 ± 0.06 mm abalone-1. Another experiment was carried out at Roman Bay Sea Farm (Pty) Ltd with initial weight and length 34.7 ± 0.17 g and 62.3 ± 0.18 mm abalone-1, but this experiment included one factor only, i.e. the presence and absence of the probionts in the feed. At HIK there was no significant interaction between diet and handling on average length and weight gain month-1 after four (p=0.81 and p=0.32) and eight (p=0.51 and p=0.53) months, respectively. Average length (additional handling = 73.9 ± 0.52 mm, normal farm handling = 75.8 ± 0.57 mm) and weight gain (mean: additional handling = 68.5 ± 1.20 g, normal farm handling = 74.3 ± 1.86 g) increased significantly in animals that were handled under normal farm procedure and were either fed probiotic or control diet after eight months (p=0.03 and p=0.02, respectively). There was no iii difference in length gain or weight gain of abalone fed the probiotic diet and those fed the control diet (ANOVA: F(1,16)=0.04, p=0.84; F(1,16)=0.14, p=0.71, respectively). After four months phagocytotic count was significantly different between dietary treatments with mean values of 74.50 ± 10.52 and 63.52 ± 14.52 % phagocytosis count per sample for the probionts and control treatment, respectively (p=0.04), there was no difference after eight months at HIK Abalone Farm. There was no effect of stressor application (p=0.14) and no interaction between dietary treatment and stressor application for this variable i.e., phagocytosis count (p=0.61). There was no difference in feed conversion ratio between treatments with values ranging from 2.9 to 3.8. At Roman Bay Sea farm, there was no significant difference in mean length gain between abalone fed the probiotic and control diet after eight months (repeated measures ANOVA: F(4,28)=16.54. Mean weight gain of abalone fed the probiotic diet was significantly greater than those fed the control diet after eight months (repeated measures ANOVA: F(4,28)=39.82, p(0.00001). There was no significant difference in haemocyte counts between animals fed either probiotic or control diet after four and eight months at Roman Bay Sea farm (p>0.05).

The use of treated brewery effluent as a water and nutrient source in crop irrigation

- Description: Brewery effluent (BE) needs to be treated before it can be released into the environment, reused or used in down-stream activities. Current technologies used to address this concern at the experimental wastewater treatment plant at Ibahyi Brewery (SAB Ltd) include anaerobic digestion (AD), primary facultative ponds (PFP), high rate algal ponds (HRAP) and constructed wetlands (CW). The aim of this work was to determine if BE treated in these systems might be suitable for crop irrigation. A test crop, cabbage (Brassica oleracea cv. Star 3301), grew best on post-AD and post-PFP BE compared to those irrigated with post-HRAP or post-CW effluent. However, the yield was 13% lower than cabbage plants irrigated with a commercial nutrient solution and fresh water. The relatively high conductivity (3019.05 ± 48.72 μs/cm2) of BE may be the main factor reducing the cabbage yields. Post-HRAP and post-CW BE were the least suitable for irrigated crop production due to the higher conductivity and lower nutrient content of these treated effluents. After three months, soils irrigated with post-AD and post-PFP BE had a significantly higher sodium content and sodium adsorption ratio (3919 ± 94.77 mg/kg & 8.18 ± 0.17) than soil irrigated with a commercial nutrient solution (920.58 ± 27.46 mg/kg & 2.20 ± 0.05; p<0.05). However, this was not accompanied by a deterioration in the soil’s hydro-physical properties, nor a change in the metabolic community structure of the soil (p>0.05). After prolonged irrigation with treated BE, sodium is likely to build up in the soil and this can be expected to be accompanied by a deterioration in the soil physical structure. However, crops species such as millet (Echinochloa esculenta), lucerne (Medicago sativa) and saltbush (Atriplex nummularia) reduced the build-up of sodium in the soil. The results suggest that sodium was mainly removed from the soil through plant-assisted leaching. Of the crops grown, lucerne showed the most promise because it improved the soil physical properties, is able to grow well in alkaline environments, is a popular fodder crop and can be harvested multiple times from a single stand. Brewery effluent is more suitable for soil production systems than hydroponic production systems because the soil was able to act as a buffer against the high pH of post-AD BE, whereas in a hydroponics systems the high pH reduced the availability of key minerals to plants. In conclusion brewery effluent contains sufficient plants nutrients to support the growth of cabbages, saltbush, lucerne and millet. However the sodium content of BE is a concern as it accumulates in the soil, and in the long-term it may lead to soil degradation. It is suggested that the brewery change the pH neutralising treatment of BE from sodium hydroxide to potassium hydroxide, or dolomitic lime (calcium and magnesium carbonate) because this would reduce the introduction of sodium into the system, and would increase the suitability of BE for crop production, given potassium and calcium are plant nutrients. The benefits of developing this nutrient and water resource could contribute to cost-reductions at the brewery, more efficient water, nutrient and energy management, create job opportunities with the potential of improving food security in the local community.

- Description: Brewery effluent that had undergone treatment in an anaerobic digester (AD) was used as an alternative water and nutrient source for hydroponic crop production. Brewery effluent was demonstrated to contain sufficient nutrients to support the growth, flowering and fruiting of Lycopersicum escolentum "Moneymaker" tomato crops. The adjustment of the effluent pH with phosphoric acid to between pH 6.0 and 6.5 increased the development of the crops by around 100% compared to crops grown in unaltered effluent. The pH adjusted effluent-grown plants grew to a mean height of 831.4 ± 21.1 mm and a dry biomass weight of 42.34 ± 2.76 g compared to the unaltered pH effluent plants which grew to a height of 410.6 ± 20.5 mm and a weight of 7.65 ± 0.68 g after 49 days. Effluent treatment in high-rate algal ponds (HRAP) was determined to have no positive effect on the nutritional potential of the effluent for Moneymaker production. The effluent-grown plants did not perform as well as plants grown in inorganic-fertilizer and municipal water. Plants grown in effluent grew taller but did not produce significantly more fruit when phosphoric acid (height: 1573.3 ± 50.4 mm, 19.4 ± 1.4 fruit per plant) was compared to nitric acid (height: 1254.1 ± 25.4 mm, 15.6 ± 1.5 fruit per plant) as the pH adjustment over 72 days. Direct and secondary plant stresses from effluent alkalinity, ammonium nutrition, nitrogen limitation, sodium concentrations and heat stress among other factors were probably confounding variables in these trials and require further investigation. Considering the raw effluent composition and manipulating the AD operation is a potential opportunity to improve overall AD performance, reduce chemical inputs in the effluent treatment process, reduce the final effluent alkalinity, and increase available nitrogen content in the final effluent. The anaerobic digester discharging >1000 m³ of nutrient enriched effluent every day is a resource with considerable potential. The benefits of developing this resource can contribute to cost-reduction at the brewery, more efficient water, nutrient and energy management at the brewery, and offer opportunities for job creation and potentially benefit local food security.

- Description: Climate change has caused a combination of effects on the physiology of fishes. Of particular concern are the effects of thermal variability and ocean acidification. Organismal energy budgets change throughout ontogeny and research into the metabolic scope during early life stages is particularly useful in identifying potential bottlenecks. The first part of this thesis aimed to assess the absolute aerobic scope (AAS, described as the difference between the maximum and standard metabolic rates) of individual juveniles from a protected population of the endemic, commercially important seabream, Chrysoblephus laticeps, across a range of ecologically relevant temperatures (T = 11, 14, 18, 22˚C) under present-day conditions (pH = 8.03, pCO2 ≈ 420 μatm) using intermittent flow respirometry. The second component sought to investigate how long-term exposure (from fertilisation to juvenile, ~100 days exposure) to high-pCO2/hypercapnic conditions (pH = 7.63, pCO2 ≈ 1400 μatm), would affect the AAS of juvenile C. laticeps over a range of temperatures. Lower pH conditions were predicted to cause a decrease in the AAS of treatment animals due to additional energetic costs of acid-base regulation. The findings of the first data chapter demonstrated that juvenile C. laticeps reared under current CO2 conditions are tolerant to a wide range of thermal conditions, and individuals with a broad aerobic scope will be the best suited to coping with enhanced thermal variability. In contrast to the expected outcomes of the second data chapter, juvenile C. laticeps reared under high pCO2 conditions displayed greater AAS at high and low temperatures when compared with specimens from high pH conditions. Whilst a high degree of individual phenotypic variation was observed in the metabolic response of both groups, this was reduced at the lower and upper extreme temperatures for high pH and low pH animals respectively. Notably, the variation in treatment animal’s SMR was significantly diminished across all temperatures tested, compared to only a localised reduction in the SMR of high pH animals at cold temperatures. This may be indicative of compensatory pathways affecting energy restructuring and thermally-governed physiological trade-offs under hypercapnia. Given these results, juvenile C. laticeps appear to be more resilient to ocean acidification than anticipated, potentially owing to intrapopulation metabolic phenotypic diversity. This is likely attributed to the parental lineage originating in the Tsitsikamma MPA, which is thought to boast greater phenotypic diversity as a consequence of the refuge that these conservation areas offer from exploitation. Owing to the restriction imposed by the availability of surviving, captive-reared juveniles, the sample size used in this study was relatively low. However, owing to the repeated-measures nature of this research the sample size was sufficient to offer suitable statistical power for the polynomial mixed model used in the analysis. Future research should incorporate both physiological and behavioural responses to multiple environmental stressors to better understand covariation between these two traits, and to detect any behavioural trade-offs that might arise through compensation. - Description: Temperature changes due to the effects of climate change are evident on all continents and oceans. As a result, there is a growing concern over how marine ectotherms will respond to extreme or fluctuating environmental temperatures. Temperature changes have strong direct and indirect effects on individual, population, and ecosystem functioning traits. A multi-scale approach determining the thermal tolerance and performance of several marine ectotherms belonging to different coastal habitats is rarely considered in thermal physiology studies but is effective for an integrated ecosystem assessment. As such, for this thesis, I aimed to quantify and compare the thermal tolerance and performance of a range of coastal marine ectotherms (fish and macro-invertebrates) with different biogeographical distributions from estuarine, subtidal and rocky intertidal habitats to available and projected in situ temperature data. This was also undertaken to gauge the local vulnerability of each species across summer and winter in a warm-temperate region of South Africa. This was done using a multi-method physiological approach, which included the dynamic method (CTmax and CTmin), static respirometry and maximum heart rate fHmax). Results of the dynamic method on several fish and macro-invertebrate species indicated that there are differences in thermal tolerance according to taxonomy, biogeography and habitat for both summer and winter. Macro-invertebrate species generally had higher CTmax endpoints, lower CTmin endpoints, higher upper and lower breadths in tolerance, higher upper and lower thermal safety margins and higher thermal scopes than the fish species. This could be a result of the macro-invertebrate species studied being less mobile compared with fish species (which are able to move to more favourable conditions) as well as having broader geographical distributions. In addition, macro-invertebrates from the intertidal rock pool habitat (Palaemon peringueyi; Pernaperna) were more tolerant of high and low temperatures compared with the macro-invertebrates from the estuarine habitat (Clibanarius virescens; Parasesarma catenatum; Upogebia africana). Overall, macro-invertebrates, with the exception of Parechinus angulosus, investigated in this study indicated that current temperatures and projected climate change scenarios across seasons would not have a significant impact on them and that they are highly adaptable to changing temperature regimes. This sign of high tolerance was further supported by the heart rates of P. perna and P. catenatum under an acute increase in temperature (1.0 °C.h-1) which showed individuals of each species physiologically depressing their metabolism until a final Arrhenius breakpoint temperature was reached (TAB). Among the fish species investigated in this study, tropical species (Chaetodon marleyi; Kuhlia mugil) had the highest CTmax and CTmin endpoints when compared with the temperate (Diplodus capensis; Sarpa salpa), warm-water endemic (Chelon dumerili; Rhabdosargus holubi) and cool-water endemic (Chelon richardsonii) fishes. This suggests that due to their lower breadths in tolerance and thermal safety margins being small, tropical species may be less tolerant of cold temperatures and thermal variability, especially in the form of summer upwelling events which are expected to increase in intensity and frequency in this region as a result of anthropogenic climate change effects. On the other hand, however, if a temperature increase of 2.0 - 4.0 °C takes place at the end of the century as predicted by the Intergovernmental Panel on Climate Change (IPCC), it is likely that tropical species such as C. marleyi will become more common. Temperate species such as D. capensis and S. salpa were able to tolerate a wide range of temperatures (wide thermal scope) compared with the other fish species. These findings may suggest that D. capensis and S. salpa are thermally resilient and may be the least vulnerable to climate change effects and temperature variability. When evaluating the different life stages of D. capensis, however, using the dynamic method (juveniles and adults), static respirometry (juveniles) and maximum heart rate (adults), results suggested that juveniles of this temperate species will be more resilient to increases in ocean temperatures compared with the adults because they have a higher thermal tolerance (CTmax/TCRIT) and a greater metabolic scope (TOPT) at higher temperatures. For both juveniles and adults, temperatures beyond 28.0 °C (upper Tpej; Tarr) will have a significant impact on their physiology. Using a multi-scale and multi-method approach thus helped to identify which species or community may be vulnerable to the effects of climate change within shallow coastal environments in this warm-temperate climate change hotspot. Adopting this type of approach will assist policy makers in developing comprehensive climate change management frameworks for coastal ecosystems globally and around South Africa.

- Description: The South African mariculture industry is developing the rearing technology of indigenous fish species. Dusky kob (Argyrosomus japonicus) has high-quality flesh and a good market demand. Research is needed to determine the environmental requirements under which growth of dusky kob is optimised. This study assessed the effect of temperature, light intensity, feeding frequency and stocking density, respectively, on growth, feed conversion ratio (FCR) and survival of juvenile dusky kob within a weight range of 10-60 g fish⁻¹) in a series of four growth trials. The effect of temperature on growth and FCR was assessed at 14 temperatures from 17 to 28°C. The temperature for best growth was estimated to be 25.3 °C, while 21.4 °C was the temperature at which the best FCR was achieved. A growth trial testing the effect of light intensity on growth showed that light intensity did not affect growth in the range of 23 – 315 lx. Fish fed a restricted ration of 3.6% body weight per day (chapter 2) or 3.41% body weight per day (chapter 4) had a better FCR than fish fed to satiation. A trial to assess the effect of feeding frequency on growth and FCR showed that fish fed both two or three times daily grew better than those fed once daily. FCR was best in fish that were fed once or twice daily. Preliminary analysis of the results from a stocking density trial showed that stocking density in the range of 10 – 50 kg m⁻³ did not affect growth of juvenile dusky kob. It is recommended to culture dusky kob at a temperature of 25.3 - 21.4 °C at stocking densities up 50 kg m⁻³ to maximise growth. However, a study is needed to determine the environmental conditions needed to maximise profit under commercial conditions. The results allow the South African industry the opportunity to assess the culture potential of this species. In addition, the results will help develop protocols that can be used in other South African candidate aquaculture species such as silver kob, yellowtail, white-stumpnose and sole.

- Description: Under the guidance of the new coastal management policies within South Africa this thesis advocates a more integrated, co-operative approach to local coastal management. The project aimed to acquire baseline information on the local nearshore fishery and resource state and to propose a set of indicators that could be incorporated into the new management strategy. To gather the required information the project was split into two parts: 1) An assessment of the local linefisheries and 2) A comparative study of the reef fish community structure between exploited and unexploited reefs. The local linefisheries were assessed through the use of launch records, commercial catch records, access point and boat based surveys. A questionnaire was used to gather data on catch and effort, fisher demographics, fisher attitudes towards and knowledge of the current management regulations, assess the efficacy of the fisheries inspectorate and highlight spatial areas of fishing pressure. A total of 252 interviews and catch inspections were conducted. Total effort for the ski-boat fishery estimated from the access point survey was 890 boat days.year⁻¹ or 3560 fisher days. year⁻¹ compared to 736 boat days.year⁻¹ or 2944 fisher days. year⁻¹ recorded in the launch records. Effort was seasonal with greater pressure occurring over the summer holiday period. Analysis of the catch showed that Merluccius capensis, Argyrozona argyrozona, Argyrosomus inodorus, Chrysoblephus laticeps and Atractoscion aequidens were the most frequently caught species. The overall CPUE was 3.00±5.54kg.fisher⁻¹.day⁻¹ or 4.71±4.117 fish.fisher⁻¹.day⁻¹. Estimated targeted CPUE was 0.91±0.67kg.fisher⁻¹.day⁻¹ or 0.97±0.77 fish.fisher⁻¹.day⁻¹ for C. laticeps, 8.47±8.57kg.fisher⁻¹.day⁻¹ or 1.24±1.16 fish.fisher⁻¹.day⁻¹ for A. aequidens and 2.05±3.78kg.fisher⁻¹.day⁻¹ or 1.10±1.80 fish.fisher⁻¹.day⁻¹ for A. inodorus. Issues identified included poor fisher knowledge regarding linefish regulations, the low occurrence of fishery inspections and a limited degree of noncompliance. Although most fishers supported the current linefish management regulations, when tested on the size limits, bag limits and closed seasons of their target species a high proportion of fishers did not know the regulations (recreational 64%, charter 53%, commercial 42%). Furthermore only 27% of fishers had had their catch inspected whilst fishing in Plettenberg Bay and the majority of these had only been inspected once. Just over half the interviewees (60%) indicated that fishing had deteriorated within Plettenberg Bay with fewer and smaller fish being caught. The most common causes cited for this decline were commercial and recreational overfishing respectively. Underwater point counts and experimental angling were used to rapidly assess the state of the reef fish resource in Plettenberg Bay through a comparative study of the community structure between two exploited sites in Plettenberg Bay and two protected sites within the Tsitsikamma National Park. Generalized linear modeling showed that relative density of certain species was significantly different between the protected reefs inside the TNP and those exploited reefs within Plettenberg Bay. Both P. rupestris and C. laticeps had greater densities within the protected area whilst Chisquared tests showed that the size frequency distributions were significantly different with larger size-classes and the maximum size of fish greater within the reserve. These trends were noted with both the underwater visual surveys and the experimental angling. Multi-dimensional scaling and cluster analysis showed that there was an overall difference in the community structure between the study sites. It is hypothesised that through removal of key reef species and larger individuals that fishing has directly and indirectly affected the overall community structure. Within a simple framework based on ecological, institutional and social sustainability domains along with the results of the study area, a set of indicators is proposed and the sustainability of the local fishery scored within a rapid assessment matrix. The socio-economic domain scored the highest (83%) whilst the institutional domain scored the lowest (8.3%) and the ecological domain scored 25%, giving a total sustainability score of 38.8%. The results of this matrix show that at present the local fishery is non-sustainable and in need of greater management. By synthesizing papers dealing with and based on the concepts involved in Integrated Coastal Management, a structured approach is proposed to developing and implementing more holistic local coastal governance. It is envisaged that the framework to implement such an approach should be through the development of a local Coastal Management Plan and a subsidiary Bay Management Plan. Although stakeholder participation and representation is an essential component in the development of these plans, it is recommended that the local municipality should be the lead agent and incorporate the plans into the local Integrated Development and Spatial Plans thereby gaining long term local government support.

- Description: The overall aim of this thesis, as required by the funders, was to provide research that would contribute towards the development of a bay management plan, specifically information on the local fisheries. The thesis therefore includes an assessment of the local shore-based linefishery in terms of catch and effort and socio-economics, and the development of indicators from this information with which to assess the sustainability of the fishery. Finally an implementation strategy for an integrated coastal management approach for local resource governance is proposed The shore-based linefishery of Plettenberg Bay was assessed through a combination of roving creel and access point surveys which included questions aimed at the quantification of total effort, total catch and cpue, as well as the spatial trends in catch and effort. Angler demographics as well as opinions and knowledge of current fisheries regulations were obtained and the efficiency of the fishery inspectorate was assessed. The survey period extended from August 2003 to September 2004 during which 1189 angler interviews were conducted. Catch data obtained during this study was compared to catch data gathered during a research tagging program conducted in an area closed to fishing (Tsitsikamma National Park) and a semi-open area (Rebelsrus – Cape St. Francis), thereby highlighting differences in species composition, abundance and size distribution of certain species between areas of differing fishing pressures. The fishery surveys revealed that the shore-based fishery is primarily recreational (99%) and dominated by white males. Most anglers agreed with the current fisheries regulations (60%) and admitted to abiding by them, however when tested on the regulations of their target species, their knowledge was poor. Effort in the fishery was highly seasonal with peaks coinciding with major school holidays. Total annual effort was estimated at 102 566 angler-hours, with distinct spatial patterns in effort. Total annual catch for the Plettenberg Bay shore-based linefishery was estimated at 31 217 fish.year⁻¹ with a total mass of 13.6 tons. Thirty-six fish species (26 teleost species and 10 elasmobranchs species) were identified during the survey period with Blacktail Diplodus capensis, Strepie Sarpa salpa, Red tjor-tjor Pagellus natalensis, Shad Pomatomus saltatrix and Sand steenbras Lithognathus mormyrus being the five most commonly caught species. Only 32% of interviewed anglers were successful in catching a fish with just 22% catching their primary target species. In addition, a large proportion of the anglers (69%) reported a decline in catch rate, with most blaming the commercial sector as a reason for the decline. The overall catch rate was (cpue) 0.374 fish.angler.hour⁻¹ or 170 grams.angler.hour⁻¹, considerably lower than that obtained from the fishery exempt area (TNP = 1. 02 fish.angler.hour⁻¹) and the partially exploited area (Rebelsrus = 0.91 fish.angler.hour⁻¹). In terms of species composition the most obvious difference between the three areas was the low proportion of non-migratory reef-associated species like red roman, poenskop, John brown, santer and bronze bream in Plettenberg Bay. Size comparisons revealed that the majority of species (particularly reef-associated species) were larger in the TNP than both the semi-exploited (Rebelsrus) and exploited area (Plettenberg Bay). Collectively theses findings suggest that certain species have been locally depleted, that recreational fishing has impacted heavily on the fish stocks and that even partial closure or limited access to an area can offer protection. From the results key issues were identified and sustainability indicators proposed according to the three sustainability domains (ecological, institutional and social) proposed by Pajak (2000). Within the ecological domain the most pertinent issue was the low catch rate, particularly for reef-associated species. Indicators to track these issues included: percentage of successful trips (where fish were caught), percentage of anglers that reached their daily bag limits and the proportion of the overall catch that comprised larger, more desirable species. Institutionally the most pertinent issue was the apparent inability of management institutions to manage effectively marine resources within Plettenberg Bay. Indicators within this domain included the proportion of anglers who were inspected, the existence of a localised management plan and an associated monitoring programme. Socially the fishery proved to be fairly sustainable. Indicators included in this domain were the proportion of subsistence anglers in the local fishing community, the percentage of correct answers pertaining to current fishery regulations, the level of noncompliance and the percentage of undersize fish retained. Aggregation of all the indicators within a sustainability matrix revealed that the Plettenberg Bay shore-based linefishery was unsustainable and thus requires increased local management effort. Since there is no local management strategy in place, an integrated coastal management approach for the governance of the coastal resources in Plettenberg Bay has been proposed. The development of a Coastal Management Plan and a subsidiary Bay Management Plan provide the frameworks within which management strategies can be put into operation.

- Description: The activity of fishing can be traced back to prehistoric times. However, only in the last century has there been a focus on the management of fisheries. Fishery regulations are tools used by resource managers with the aim of protecting the long-term sustainability of fishery resources. Although there is an overwhelming amount of evidence demonstrating the decline of fisheries, non-compliance with these regulations by fishers continues to manifest, which can exacerbate the negative ecological impacts of fisheries. Popular methods towards the measurement of noncompliance in fisheries derived from previous human dimensions literature may be flawed. Theories on improving compliance behaviour have typically relied on theory, which has at times proved paradoxical. Addressing the issues of non-compliance within a fishery of interest requires measuring the levels of non-compliance within the fishery and determining the relevant sociopsychological drivers behind the non-compliant behaviour. The data collection methods used during these assessments are limited in human dimensions research and are often case and context specific, requiring researchers to identify which approach is most practical for the specific fishery of interest. By identifying relevant behavioural drivers of non-compliance, a more effective approach aimed at improving compliance can be tailored. The recreational marine-based shore fishery (MBSF) in South Africa is not impervious to noncomplaint behaviour. In fact, it has been estimated to have relatively high rates of non-compliance. This high level of non-compliance makes the South African MBSF a unique and optimal context in which to undertake research that aims to formulate a framework towards compliance assessments and that develops a suitable approach for improving compliance rates. Using surveys to obtain compliance data can provide a range of details about violators, however they are susceptible to social desirability bias (SDB). Choosing the best method for controlling SDB required an assessment of existing methods for doing so. In this first part of the study, only fishers who were covertly observed breaking the rules were surveyed, using one of three methods for reducing SDB, to ground-truth the responses. Ground-truthing was done to determine which method would be most effective for a large-scale study within the same fishery. Of the methods used, which include the direct questioning method (DQM), the random response technique (RRT) and the ballot box method (BBM), all contained some level of SDB. However, the BBM provided a significantly higher level of response accuracy (79.6% ± 11.9) than the DQM (46.5% ± 14.9) and the RRT (44.3% ± 12.5). Random-stratified roving creel compliance surveys that employed the BBM were then undertaken at various locations along South Africa’s coastline to estimate current rates of non-compliance, and the face-to-face results were compared to results from an identical online survey to determine the suitability of online surveys as a replacement. The results indicated that online surveys only represent a subgroup of the fisher population within the MBSF, suggesting that face-to-face survey methods are required to obtain a more comprehensive sample and a more robust estimate of noncompliance. The results, based on 453 face-to-face surveys, showed a high level of overall self-reported noncompliance (48.3%) within the fishery. Responses to Likert scale survey questions on various aspects of the fishery, including angler motivations for fishing, were then modelled to test the relationship between the anglers’ responses and their compliance behaviour. In the South African MBSF context, the most significant behavioural drivers behind non-compliance related to normative concepts. Specifically, the poor perceptions of management and value-based legitimacy as well as low levels of moral obligation to adhere to the regulations appeared to contribute most to the observed non-compliant behaviour. Angler motivations for fishing also played a significant role in determining the compliance behaviour of anglers, with those fishing for food being more likely to violate regulations. In most countries, regardless of economic context, interventions to improve recreational fishery compliance have been developed around the instrumental concept. However, these findings suggest that for recreational fisheries, managers would do well to evaluate the impact of normative concepts on compliance and to design interventions aimed at addressing these. In the case of the South African MBSF, interventions that address angler perceptions of legitimacy and aim to correct misperceptions about social norms of compliance may provide a more practical and cost- effective method for improving poor compliance behaviour.

Towards an alternative spatial-based management approach for estuarine fisheries in South Africa, with a case study from the Sundays Estuary

- Description: Estuaries are productive habitats and biologically important ecosystems which serve as juvenile nursery areas and feeding grounds for adults from a host of fish species. They are, however, threatened habitats, increasingly exposed to human disturbance and exploitation. The stocks of several South African estuary-dependent linefish species are now considered as either overexploited or collapsed. It is clear that their dependence on estuaries would warrant the inclusion of these ecosystems into marine reserve planning exercises. Since traditional management strategies (e.g. bag and size limit restrictions) have proven ineffective for estuarine fisheries, there is a need for alternative management measures, such as spatial and temporal restrictions, to ensure increased survival of juveniles and recovery of adult breeding populations. This thesis explored the potential for an ecosystem-based approach through the application of a rapid sustainability assessment technique, and a spatial-based management approach for an important fishery species, using conservation planning software. The Sundays Estuary, Eastern Cape, South Africa falls within the footprint of the Addo Elephant National Park, with a proposed expansion to include a marine protected area (MPA). However the estuaries resources were not considered during the planning of the proposed MPA. This study conducted an indicator-based sustainability assessment based on the principles of sustainable development. The results showed that present levels of exploitation, due to non-compliance and a lack of law enforcement are unsustainable. The sustainability of the Sundays Estuary had a low overall sustainability score of only 23.8%. With limited enforcement of estuarine fisheries regulations in South Africa, alternative management measures such as spatial regulations may provide a viable option forward. The sustainability of fishery resources depends on the comprehensive understanding of the fishery resource. Acoustic telemetry is a technique that has been widely adopted to infer habitat and area use patterns of fish species. The second component of this study made use of high resolution telemetry data collected on juvenile dusky kob Argyrosomus japonicus movements within the Sundays Estuary to conduct a scenario-based approach using Marxan conservation planning software. The best solution given by Marxan, in the form of a protected area for the conservation of juvenile A. japonicus in the Sundays Estuary was identified in the middle (starting 7km from the mouth) to the upper reaches (approximately 16km from the mouth) of the estuary, ultimately providing protection to tagged individuals for 61% of their time in the estuary. Although Marxan presented a best solution, the Sundays Estuary’s small size and shape, and minimal features used, was too simplistic to be included into a Marxan analysis. However, new methods and tools to analyse and plan spatial-based management options at this scale are currently being developed. Using the Sundays Estuary as a case study, a decision tree was then developed as a protocol to assist management address the challenges of effective estuarine management depending on the unique biological and socio-economic characteristics of individual estuaries in South Africa.

- Description: The harvest of Loligo reynaudii, or "chokka," represents a critical source of revenue and job creation in the historically impoverished Eastern Cape Province of South Africa. Due to the importance of visual stimuli in the reproductive processes, it has been hypothesized that a primary driver of successful reproduction is the clarity of the water column. The presence of increased particulate matter concentrations within the water column generates turbid conditions near the seafloor (visibility < 1m), that are proposed to restrict spawning activity. This benthic nepheloid layer (BNL) contains both organic and inorganic components, with the BNL intensity a function of bottom turbulence, substratum type, and detritus level. However, the spatial and temporal resolution of BNL intensity on the Eastern Agulhas Bank (EAB) and the environmental drivers thereof remain unknown. Here we show that benthic turbidity events are a common but highly variable occurrence on the EAB. Results from a 17-month time-series of in-situ and remote sensing data between 2002 – 2004 in Algoa Bay, supplemented by experiments in other bays important for spawning, show that turbid conditions existed for ∼ 30 % of the sample period. Exploration of environmental drivers, including the influence of wind, altimeter-derived significant wave height (Hs), sea surface temperature (SST), and chlorophyll-a (Chl-a) concentrations indicate that BNL intensity does not conform to a "one-size-fits-all" approach. Rather, complex local hydrological and physiochemical parameters control the BNL characteristics on the EAB. Global warming is likely to increase the frequency and intensity of extreme westerly-wind and storm events, promoting BNL events on the Eastern Agulhas Bank and possibly causing a shift in the reproductive strategy of chokka squid to the cooler mid shelf region. This is likely to have consequences for both the species in terms of reproductive success and the fishery, which is concentrated on inshore spawning aggregations. Future research needs to quantify and characterize the constituents, source particles and spatial-temporal variability of BNL events in order to build a predictive capacity. Through incorporating the qualitative analysis of the dynamics of nepheloid layers on the EAB into Regional Oceanographic Models (ROMS), General Linear Models (GLM) and particle distribution models such as DELFT-3D, it is possible to move toward predicting the timing and intensity of these events. , Thesis (MSc) -- Faculty of Science, Ichthyology and Fisheries Science, 2022

- Description: Increased CO2 production and the consequent ocean acidification (OA) have been identified as one of the greatest threats to both calcifying and non-calcifying marine organisms. Traditionally, marine fishes, as non-calcifying organisms, were considered to have a higher tolerance to near-future OA conditions owing to their well-developed ion regulatory mechanisms. However, recent studies provide evidence to suggest that they may not be as resilient to near-future OA conditions as previously thought. In addition, earlier life stages of marine fishes are thought to be less tolerant than juveniles and adults of the same species as they lack well-developed ion regulatory mechanisms for maintaining homeostasis. This study follows up on previous studies examining the effects of near-future OA on larval Argyrosomus japonicus, an estuarine-dependent marine fish species, in order to identify the tipping point of tolerance for the larvae of this species. These previous studies showed that elevated pCO2, predicted for the year 2100, had negative effects on growth, development and metabolism and ultimately, survival of larval A. japonicus from post-flexion stage. Larval A. japonicus in the present study were reared from egg up to 22 DAH (days after hatching) under three treatments. The three treatments, (pCO2 353 μatm; pH 8.03), (pCO2 451 μatm; pH 7.93) and (pCO2 602 μatm; pH 7.83) corresponded to levels predicted to occur in year 2050, 2068 and 2090 respectively under the Intergovernmental Panel on Climate Change (IPCC) Representative Concentration Pathways (IPCC RCP) 8.5 model. Size-at-hatch, growth, development and metabolic responses (standard and active metabolic rates and metabolic scope) were assessed and compared between the three treatments throughout the rearing period. Five earlier larval life stages (hatchling – flexion/post-flexion) were identified by the end of the experiment. There were no significant differences in size-at-hatch (P > 0.05), development or the active metabolic (P > 0.05) or metabolic scope (P > 0.05) of fish in the three treatments throughout the study. However, the standard metabolic rate was significantly higher in the year 2068 treatment but only at the flexion/post-flexion stage which could be attributed to differences in developmental rates (including the development of the gills) between the 2068 and the other two treatments. Overall, the metabolic scope was narrowest in the 2090 treatment, but varied according to life stage. Although not significantly different, metabolic scope in the 2090 treatment was noticeably lower at the flexion stage compared to the other two treatments, and the development appeared slower, suggesting that this could be the stage most prone to OA. The study concluded that, in isolation, OA levels predicted to occur between 2050 and 2090 will not negatively affect size-at-hatch, growth, development, and metabolic responses of larval A. japonicus up to 22 DAH (flexion/post-flexion stage). Taken together with the previous studies of the same species, the tipping point of tolerance (where negative impacts will begin) in larvae of the species appears to be between the years 2090 and 2100.

Towards determining the dietary lysine requirement in the South African abalone, Haliotis midae

- Description: Animals generally do not have a requirement for protein, but instead have a requirement for specific essential amino acids (EAAs) and non-essential amino acids (NEAAs). The NEAAs are those that can be synthesised by the animal, however, EAAs cannot be synthesised and must therefore be supplied as part of the diet. When these amino acids (AAs) are supplied in the correct ratios and with the correct level of digestible energy, nutritionists can maximise somatic growth from proteins. This has resulted in increased research into the use of crystalline AAs as a tool in feed formulation research in order to quantify the AA requirements in aquaculture species, and allow for alternative protein sources (other than fishmeal) to be utilised. In common with other water soluble nutrients, leaching of crystalline AAs from diets prior to ingestion is of concern in an aquatic environment. Microencapsulation techniques have been successfully employed to restrict micronutrient leaching, and improve ingestion rates. In this research, LysiPEARL™ (Kemin®) was used as a means to determine the lysine requirement in Haliotis. midae. This encapsulated lysine product is used in the dairy cattle industry as an effective source of rumen bypass for intestinal release of lysine. It has previously been proposed that crystalline AAs are not suitable for AA studies in H. midae due to the slow feeding rates of the species as well as the solubility of these AAs. However, 90.00 % of supplemented lysine was maintained in this study after a six hour period of leaching, showing that if effective microencapsulation techniques are used, it is possible to use crystalline amino acids to supplement protein bound lysine in abalone feeds. Six isoenergetic (15.90 MJ/kg), isolipidic (6.00 %) and isonitrogenous (29.00 %) diets enriched with 5.52, 6.40, 7.28, 8.14, 9.00 and 9.86 % lysine (as a % of protein) were fed to triplicate groups of 20 H. midae (20.41 ± 1.95 mm SL 1.51 ± 0.44 g w.wt) for 90 days. Wet weight and shell length measurements were taken every 30 days and specific growth rate (SGR) (% body weight.day-1), feed conversion ratio (FCR), protein efficiency ratio (PER), feed consumption (% body weight.day-1) and condition factor were calculated for each dietary treatment. Linear regression showed that FCR increased as dietary lysine increased (Regression analysis, p=0.031), and that PER reduced as dietary lysine increased (Regression analysis, p=0.026). Feed consumption also increased as dietary lysine increased (Regression analysis, p<0.001). The inclusion of lysine at 7.28 % of the total protein in the diet resulted in significantly superior SGR (0.57±0.01 % body weight.day-1) to that of 5.52 % (0.42±0.05 % body weight.day-1), FCR (1.51±0.05) to that of 8.14 % (1.99±0.21) and PER (2.45±0.07) to that of 8.14 % (1.99±0.18; ANOVA, p<0.05). There was a significant difference found in feed consumption (% body wt.d-1), with consumption increasing significantly between the first three dietary treatments and the last three dietary treatments (ANOVA, p<0.001). There was no significant improvement in SGR when dietary lysine increased above 7.28 % of the dietary protein in the diet, indicating that dietary lysine requirement was being met at 7.28 %, after which excess lysine promoted no growth response. The diet producing the best SGR, PER and FCR in this study was diet 3 which had a measured lysine content of 6.90 %. The results of the present study suggest that the lysine requirement in H. midae is in the range of 6.00 - 7.00 % of dietary protein. From these data amino acid ratios were used to estimate optimum inclusion levels of other essential amino acids. However, lysine availability in LysiPEARL™ may have resulted in over estimations due to the lipid encapsulation technique used, and haliotids limited ability to efficiently digest lipids. For this reason EAA requirements were suggested based on three different hypothetical scenarios of lysine availability from LysiPEARL™.
