- Description: Since the discovery of a new species of sabellid worm in 1993, the Californian and South African abalone industries have experienced increasing shell breakage and reduced growth rates of abalone, resulting in reduced market value of product. This polychaete worm can reach very high infestation levels. It was described as Terebrasabella heterouncinata in 1999, having originated from the South African coastline where it had been observed on several wild mollusc species. Although not a shell borer, it occupies tubes on the growing edge which are formed by deposition of nacreous shell over sabellid larvae by the host abalone. Heavy sabellid infestation levels have placed the economic viability of several South African farms under threat. Past research methods have not been successful in establishing effective means of eradication or control, partly due to the lack of knowledge of basic biological information on this species. This study forms the first part of an ongoing research project into the biology of terebrasabella heterouncinata. Abalone farms in South African utilise two main diets in abalone culture. An artificial formulated feed, Abfeed, is produced in South Africa and is used by the majority of farms as it increases abalone growth rate. Kelp, a natural diet of abalone, is harvested and also used as a feed. Farmers have observed higher sabellid infestation levels on Abfeed-fed abalone and suspect that diet may be an important factor influencing sabellid infestation. Abalone stocking density has also been identified as a potential factor influencing sabellid population growth on commercial farms. In order to better understand sabellid nutrition, the feeding behaviour of the sabellid in response to suspended solids of various particle size classes from both abalone diets were investigated. Both the effect of abalone diet and stocking density and their effects on sabellid morphometrics were quantified. The proximate composition and particle size distribution of suspended-solids present within kelp and Abfeed abalone raceways were also quantified. Results indicate that sabellid feeding follows no structured behavioural pattern for both abalone diets. The quantity of suspended solids in kelp raceways was greater than in Abfeed raceways (p<0.02) while there was no difference in particulate level between three abalone stocking densities (p>0.05). Protein (p<0.0001) and energy (p<0.0.1) levels of suspended solids were significantly greater in raceways dedicated to feeding Abfeed. The infestation level (number of tubes/centimetre on the grwoing edge) was significantly higher (p<0.001) in Abfeed-fed abalone. The morphometrics of the sabellids indicated that sabellids from Abfeed-fed abalone were larger in various body measurements: length (p<0.00001); neck width (p<0.001); base width (p<0.001); and surface area (p<0.001). This study suggests that abalone diet has an important influence on the infestation level and size of sabellids and that this is likely to be due to the fragmentation and leaching of nutrients from the artificially enriched commercial abalone diet.

- Description: Erratic and highly variable catches in the South African chokka squid Loligo vulgaris reynaudii fishery, cause socio-economic hardship for the industry and uncertainty for resource managers. Catch forecasting can reduce this problem as it is believed that catch variability is strongly influenced by environmental factors. In this study, data were collected at varying temporal and spatial scales. Data for the hourly time-scale study were collected from 1996-1998, aboard commercial vessels, whilst for the longer time-scales, data were extracted for Kromme Bay (a single fishing area) from existing databases (1991-1998) that were comprised of compulsory catch returns and oceanographic data. The environment-catch relationship for chokka squid on the inshore spawning grounds was then investigated using multiple correlation and regression analysis, analysis of variance, contingency table analysis and cross-correlation statistical techniques. This simple, direct, 'black box' statistical approach was relatively successful in developing a predictive capability. On a short time-scale (hourly), the regression model accounted for 32% of the variability in catch, with turbidity the main determinant (13%). On a daily monthly time-scale, the best prediction model was on a monthly scale, accounting for 40% of the variability in catch. The principal determinant, bottom temperature anomaly (11 %), was found to lag one month forward. Seasonal and diel catch variations induced changes in the relative importance of turbidity, water temperature and wind direction on catches. A strong, positive relationship was found between easterly winds (which cause upwelling) and catch, particularly in summer. Catch rates, however, decreased with an increase in turbidity. The correlation between temperature and catch was generally negative, however, higher catches were associated with a temperature range of 13-18°C. Highest catch rates were associated with easterly winds, zero turbidity conditions and sea surface temperatures from 15.0-16.9°C. Selected case studies (in situ observations) suggested that upwelling and turbidity events act as environmental triggers for the initiation or termination of the spawning process, respectively. A holistic approach is required to improve predictive capability of chokka squid abundance. Although short-term predictability remains essential (i.e. hourly-scale), future research should concentrate on long-term prediction models (e.g., monthly time-scales) involving greater spatial variation, which are the most important for management.

- Description: The principle aim of this study was to isolate a chemical for the "safe anaesthesia" of abalone under commercial farming conditions. "Safe anaesthesia" implied that the anaesthetic had no immediate detrimental or long term sublethal effect on the abalone, that it was safe for the farmer, the consumer and the environment. Four chemicals, magnesium sulphate (MgS0₄), ethylenediamine tetra-acetic acid (EDTA), 2-phenoxyethanol and procaine hydrochloride were shown to effectively inhibit the in vitro contraction of isolated tarsal muscle of Haliotis midae. This identified them as potential anaesthetics for abalone. Since abalone, like any other aquaculture species, would be subject to frequent size-sorting during the grow-out period, size related dosage tables were developed for the four chemicals at a temperature of 18⁰C. Dosage tables were also developed for benzocaine and carbon dioxide (C0₂), Three size classes (5-15, 20-50 and 60-90 mm shell length (SL)) of abalone were considered. Only three of the six chemicals, viz. MgS0₄, 2-phenoxyethanol and CO₂, met the criteria of an effective abalone anaesthetic in that they effected rapid and mortality-free anaesthesia. The other three chemicals caused mortalities and were considered to be unsuitable for commercial scale anaesthesia. Temperature related dosage tables were then developed for MgS0₄ and CO₂, MgS0₄ concentrations and CO₂ flow rates for effective anaesthesia in abalone were found to be inversely related to temperature. The three size classes of H. midae were intermittently exposed to MgS0₄ and 2-phenoxyethanol anaesthesia for an eight month period to determine the effect of the anaesthetics on growth rate. Because of an increased resistance to the efficacy of 2-phenoxyethanol and high monthly mortalities it was concluded that this chemical was unsafe and unsuitable for commercial use. MgS0₄, on the other hand, had no effect on growth of abalone and no significant effect on the rate of mortality. MgS0₄ also had no measurable effect on H. midae muscle ultrastructure and, by implication had no effect on flesh texture. The use of MgS0₄ as an anaesthetic would, therefore, not affect marketability. Moreover, no magnesium residues were found in H. midae muscle tissue after short term or intermittent long term exposure to MgS0₄ anaesthesia. It was found that the three size classes of H. midae used in this study could be safely exposed to the recommended MgS0₄ concentrations for up to 40 minutes without any mortalities. This is more than adequate for routine farming procedures. Medium size abalone (20-50 mm SL) were also safely exposed to 14 g.100 ml⁻¹ MgS0₄ for up to 6 hours without any mortalities. The results have shown that MgS0₄ was undoubtedly the best chemical that was evaluated for anaesthesia of H. midae in this study. It fulfils the requirements set forth by the U.S.A. Food and Drug Administration (FDA) in that it is safe for the abalone, the farmer, the consumer and the environment.

- Description: Important to the evaluation of potential feed ingredients for inclusion in fish diets should be their digestibility and amino acid requirement so that feeds can be formulated on a digestible basis rather than a gross nutrient basis. This thesis established techniques for faecal collection, the validity of digestibility markers, it determined apparent digestibility coefficients for various novel feed ingredients, and it established the optimal lysine requirement for Argyrosomus japonicus, which was used to estimate each of the essential amino acid requirements by using the ideal protein concept. There were no significant differences in dry matter apparent digestibility coefficients when faeces were collected by stripping (77.0 %), dissection (80.1 %) or settlement (83.5 %). Faecal collection by the settlement method produced the most reliable digestibility data. Apparent digestibility coefficients for dry matter, crude protein and energy calculated using acid-insoluble ash were higher (84.0 %, 93.2 % and 93.0 %, respectively) than those using chromic oxide (55.7 %, 79.1 % and 78.2 %) and titanium dioxide (58.6 %, 79.7 % and 80.5 %). The magnitude of variation in digestibility coefficients obtained using acid-insoluble ash was always lower than that obtained with the other markers. Therefore, acid-insoluble ash was preferred as a dietary marker. Using the above protocol, protein and amino acid digestibility of some animal and plant protein ingredients were evaluated in a series of experiments. The first trial compared the apparent digestibility coefficients of some animal products included as single protein source in the test diets. Apparent protein digestibility values were 84.5 %, 83.8 %, 85.8 % and 83.1 % for fishmeal-prime, fishmeal-standard, poultry meal and pork meal, respectively. Apparent digestibility coefficients for poultry meal were comparable to those of fishmeal, which indicate its potential as a substitute for fishmeal in the diets of A. japonicus. The second trial determined the apparent coefficients of plant and animal protein sources included at 30 % into a practical reference diet (70 %). Apparent protein digestibility ranged from 92.4 % in sunflower meal to 85.5 % in corn gluten meal. Soybean meal is a promising feed ingredient in A. japonicus due to the high apparent digestibility of its protein (92.0 %) and essential amino acid digestibility (mean average 91.4 %). A fundamental assumption in fish feed formulation is that the digestibility of nutrients is additive, i.e., digestibility of a nutrient in one ingredient does not interact with the digestibility of the same nutrient in another ingredient. In the third trial, additivity of feed ingredients was tested using pork meal and poultry meal. The results indicate that the apparent digestibility coefficient of animal protein ingredients could be calculated from compound diets to accurately determine protein and amino acid digestibility in A. japonicus, and possibly other carnivorous fish species. Dietary essential amino acid requirements were determined for juvenile A. japonicus in two trials. A dose-response study was conducted using crystalline lysine to determine the optimal requirement of dietary lysine for A. japonicus. Optimal dietary lysine was estimated at 31.7 g kg-¹ dry diet, corresponding to 73.5 g kg-¹ of dietary protein, based on specific growth rate and broken-line segmented regression analyses. Dietary requirements for other essential amino acids ranged from 22 g kg-¹ (histidine) to 71 g kg-¹ (leucine) crude protein. The results of the present study provided a research tool that could be used to assess and verify the conclusions of earlier dietary work on A. japonicus and in further studies to develop least cost diet formulations for this species. The study also adds to the knowledge of the nutritional requirements of A. japonicus by providing information on the digestibility of plants and animal protein ingredients. It also contributes to future dietary research for this species because this study determines, for the first time, the most suitable methods for investigating the digestibility of raw materials for A. japonicus.

- Description: The aim of this work was to investigate the nutrition of a popular ornamental fish, the swordtail (Xiphophorus helleri), under intensive aquaculture conditions. The study focused on the formulation and manufacture of artificial dry diets, as well as the influence of livefeed supplementation on growth and nutritional indices of both juvenile and broodstock fish. A combination of 65% fishmeal and 35% soya oil cake meal was found to closely approximate the essential amino acid profile of X helleri (r² = 94,3%) and lysine was identified as the first limiting amino acid. The influence of dietary protein and energy on the growth of X helleri was investigated by means of a growth trial comparing a range of nine test diets combining 3 crude protein (45,38 and 30%) and 3 digestible energy (16,5, 14,5 and 12,5 kJ/gram) levels. A high crude protein content (45% dry matter) and a protein to energy ratio of ≥ 27 mg protein per kJ digestible energy was shown to promote optimal growth rate and feed conversion ratio in juvenile X helleri. The final body protein and lipid content of the fish were significantly correlated with the dietary protein to energy ratio. The nutritional value of identical dietary formulations prepared by drum-drying or cold extrusion techniques and presented either as flake or crumble particles was evaluated in a growth trial and through analysis of vitamin C levels in the diets. Up to 27% of vitamin C activity was destroyed during the drum-drying of flake diets, while 80% of the remaining vitamin C was lost through leaching within 10 minutes of immersion in water. A crumble-type diet was shown to be acceptable to X helleri, and yielded significantly better growth rate and feed conversion than the equivalent flake feed. Almost 19% of fish fed the flake diets developed vitamin C deficiency symptoms including scoliosis and lordosis, while this was completely prevented by feeding crumbles, indicating a significant restriction of leaching losses from this type of particle. The use of livefeed (Daphnia spp.) as a daily supplement to dry feeds was shown to result in a significant improvement in growth rate and survival of juvenile swordtail fish. Furthermore, the synergistic action of nutrients contained in the livefeed resulted in an improvement in the feed conversion efficiency of the artificial formulation. While mortalities of 13,4% and 15% were recorded in fish fed exclusively on flake feed or Daphnia, no mortality occurred in the group fed the supplemented diet. Furthermore, the incidence of vitamin C deficiency symptoms was prevented by daily supplementation with Daphnia. Fish fed exclusively on Daphnia exhibited significantly more intense pigmentation due to carotenoids contained in the livefeed. In broodstock fish maintained primarily on flake diets, growth rate, feed conversion ratio as well as reproductive performance, as measured by fecundity, were significantly improved by daily livefeed supplementation. Weekly supplementation showed no measurable advantage over flake feed only. A direct correlation was demonstrated between fecundity and size of female fish with a mean fecundity of 190 embryos per female in those fed a daily Daphnia supplement. This was significantly higher than the yield from the non-supplemented or weekly supplemented treatments (133 and 140 embryos per female respectively). Despite these differences, the number of young harvested during the experiment did not differ between treatments and this discrepancy was ascribed to parental cannibalism of newborn juveniles. This phenomenon constitutes a major potential bottleneck in production with estimates of up to seventy percent of young lost in some exp experimental tanks. In conclusion, the present study demonstrated that established aquaculture nutrition principles and techniques are applicable to the tropical ornamental fish species, X helleri. It was demonstrated that by using balanced crumble feed formulations, and regular livefeed supplementation, production yields under intensive conditions may be significantly improved. Suggestions for future work on poeciliid production in South Africa include investigation of the economic implications of the recommendations stemming from this project, and further research into effective restriction of parental cannibalism of newborn fish.

- Description: The South African abalone Haliotis midae Linnaeus, 1758, is an important commercial, recreational and aquaculture mollusc species. It is the largest of the six haliotid species found in South African waters and has the second largest distributional range aside from Haliotis spadicea which is widely exploited by rock and surf anglers as bait. Analysis of population structure at Great Fish Point revealed that H. midae exhibited a high degree of microhabitat specificity, and that while dietary habits played a role in habitat selection, it was ultimately the activities of predators which confined size classes to particular niches and restricted all animals to nocturnal activities. Large (> 100 mm SL) exposed animals relied on shell thickness and adhesion to combat predators, while small (> 45 mm SL) sub-boulder animals and medium sized (50 - 95 mm SL) animals relied on their cryptic microhabitats and the protective spine canopies of co-resident urchins (Parechinus angulosus) for daytime protection. Populations of H. midae were discontinuously distributed along the coast, occupying small isolated reefs which offered a suitable array of microhabitats and a good food supply. They mostly inhabited shallow intertidal and subtidal reefs, but were occasionally encountered on deeper subtidal reefs at 4 - 5 meters. Mean length- and width-at-age were determined from growth rings composed of alternate conchiolin (dark) and aragonite (white) bands in the internal nacreous shell layer. Growth was described by the Special Von Bertalanffy growth equation: Lt(mm) = 176.998918 (1 - e⁻°·²⁴²⁴¹⁹⁽t ⁺ °·⁴⁹⁵⁴⁹⁴⁾) Wt(mm) = 159.705689 (1 - e⁻°·¹⁹⁵⁴³⁹⁽t ⁺ °·²¹¹⁶⁾) The ageing technique used was validated for animals from Great Fish Point and Mgwalana using independent tag-return data. The same data provided evidence that growth rates varied between animals from Great Fish Point and Bird Island. The growth data also showed that H. midae exhibited a high degree of individual variation in growth rate. Males and females exhibited similar growth rates. Exposed large animals showed a preference for red seaweeds, in particular Plocamium corallorhiza and Hypnea spicifera, while small sub-boulder cryptic animals included larger proportions of brown (Ralfsia expansa) and green (VIva spp.) algae in their diets. Exposed individuals also exhibited a higher degree of selectivity towards prey items, but in general, stomach contents reflected the most abundant seaweed types. Both drift and attached algal species were utilized by H. midae which was a nocturnal feeder. Pigments from red algae were incorporated into the shell layers giving the shells a pink or brick red colour. Haliotis midae is a dioecious broadcast spawner. Gonad Bulk Indices in combination with detailed histological examination of gonads showed that individuals were iteroparous, asynchronous spawners and that the breeding season extended from March through to October, although the peak spawning activity was between April and June. Males and females can spawn partially, totally or not at all, with atresia of residual gametes occurring after spawning. There is no resting stage, and gametogenesis is initiated directly after spawning. The structure of the ovary and testis and the process of gametogenesis is typical of haliotid species. AI: 1 sex ratio was observed from all populations studied. Sexual maturity was first attained in the 40 - 59 mm SL size class, although evidence for the smallest size at first spawning was recorded at 54.6 mm SL for females and 69 mm SL for males. Sizes at 50% sexual maturity were 72.5 mm SL (52.8 mm SW) at Great Fish Point, 72.5 mm SL (57.4 mm SW) at Mgwalana, 73.7 mm SL (51.2 mm SW) at Cape Recife, and 73.5 mm SL (53.8 mm SW) at Kelly's Beach. Haliotis midae was typically highly fecund, although a high degree of variation resulting in poor relationships between fecundity/shell length and gonad weight/shell length. The relationship between fecundity and gonad weight was linear. In the Eastern Cape, H. midae possessed a faster growth rate, smaller size at sexual maturity, smaller maximum size and lower longevity when compared to con specifics in Western Cape waters. A smaller minimum legal size of 93 mm SW is proposed for Eastern Cape animals and it is suggested that the closed season be moved to the peak spawning period between April and June. The benefit of a closed season during the spawning period is questioned, and the feasibility of closed areas as a management option for H. midae in the Eastern Cape is discussed.

- Description: The geelbek Atractoscion aeguidens is an important commercial and recreational linefish species along the South African eastern seaboard . Aspects of its biology and population dynamics were investigated using data collected from various sampling sites along the South African east coast and from catch statistics obtained from both the Sea Fisheries Research Institute (Cape Town), and dealer records. The biological aspects studied included age and growth, reproduction and feeding. It was established that A. aeguidens is a fast growing species with a relatively late age at sexual maturity and a 1:1 sex ratio. Feeding studies revealed that it has become specialized to feed on small pelagic fish. The South African geelbek stock consists of three distinct age related sub-populations, each representing a particular phase in the life cycle. The sub-adults (1-4 yrs) occur in the South West Cape and feed predominantly on anchovy, Engraulis capensis, which they follow, moving inshore in summer and offshore in winter. The adults (5-9 yrs) undergo a seasonal inshore migration to Natal where they spawn in spring. The dynamics of this migration are strongly influenced by the annual movement of their principal prey viz. , Sardinops ocellatus, to that region. As a result the geelbek first arrive in Natal during mid winter. Evidence is also presented to suggest that a significant number of "adolescent " fish (4-5 yrs) undergo limited gonad maturation and a partial migration to at least as far as the South East Cape. Spawning appears to occur offshore. It is postulated that eggs and larvae are transported southwards by the peripheral waters of the Agulhas Current. After spawning the adults probably remain offshore and use the current to aid their return to the South and South West Cape, where they spend summer feeding on the Agulhas Bank. Juveniles (<1 yr) first appear in the South East Cape between January and March. They apparently use this region as a nursery area for almost a year before moving southwards to join the sub-adults in the South West Cape. In the South East Cape, the juveniles feed initially on mysids whereafter they switch to anchovies and pilchards. A preliminary stock assessment using yield per recruit and spawner biomass per recruit models, indicated that despite the current legislation the South African geelbek population is already overexploited. This is largely attributed to high rates of fishing mortality of the migratory adult population. Further management recommendations are therefore proposed. These include a size limit (660 mm FL), derived from an investigation of the interrelationship between natural mortality and somatic growth in a theoretical unexploited cohort, as well as a uniform bag limit for all user groups (5 fish per person per day)

- Description: Several strategies used by the gobiid fish Caffrogobius caffer enabling it to colonise the harsh intertidal environment in the eastern Cape were studied. Q. caffer is opportunistic in its use of available food resources and feeds throughout the day with a slight tendency towards crepuscular peaks of feeding activity. The system of space utilisation used by C. caffer is so organised that all individuals of the population have access to patchily distributed food resources. Current velocities occurring over high tide limit the movements of Qo caffer and interpool movements occur only during the initial stages of a rising tide and in the final stages of an ebbing tide when the current velocities are below the tolerance limit of Q. caffer. The epifauna associated with the alga Ulva rigida, which forms an important constituent of the diet of the intermediate size classes of Q. caffer, was examined and it appears that this epifauna is an important source of food to Q. caffer. - Description: The doublesash butterflyfish, Chaetodon marleyi, indigenous to South Africa, is popular amongst marine aquarists. To provide a basis for the management of the species and assess its suitability for aquaculture, aspects of its biology were investigated. Specimens were collected between February 1996 and November 1997 from localities along the eastern Cape coastline of South Africa and from specimens in the RUSI fish collection. The dentition of C.marleyi is typical of non-coraline, benthic invertebrate feeding chaetodontids. The teeth are long, inwardly hooked and spatulate which allow easy grasping and manipulation of the prey. Stomach content analysis showed that the species fed predominantly on the tentacles of terebellid polychaetes with other polychaetes, crustaceans, hydro ids and ascideans contributing to the remainder of the diet. Juveniles had a larger volume of terebellid tentacles compared to adults which preyed more upon other polychaetes. The proximate composition (64.25% protein, 4.76% fat, 4.84% carbohydrate and 24.3 kJ/g total energy content) of the terebellid tentacles was determined suggesting a high energetic requirement of the fish or maximisation of feeding profit rates.

Aspects of their biology, including age and growth, reproduction, nursery areas and feeding, were Investigated to provide a basis for the rational exploitation of the resource. Both species are slow-growing and long-lived. Growth in S. durbanensls was described by the von Bertalanffy growth equation: Lt(mm) = 1021.2(1-e⁻°.°⁹°°⁽t ⁻° .⁷°⁹⁾). The von Bertalanffy growth model did not describe the total data set in P. grande, but by excluding the zero and one year old fish the data were described by: Lt(mm) = 461.1(1-e⁻°.¹⁵³⁽t ⁺¹.⁶⁴°⁾). Detailed histological examination of gonadal development showed that S. durbanensis and P. grande are rudimentary hermaphrodites. Both species had restricted breeding seasons which coincided with peak exploitation. Results indicated that both species are group spawners with pelagic eggs. Size at 50% maturity in S. durbanensis and P. grande was determined at 350 and 300mm fork length, respectively, corresponding to ages of 4.5 and 5.5 years. Intertidal pools functioned as nursery areas for S. durbanensis during the first year, whereas subtidal waters were more important for larger juveniles. P. grande juveniles were commonly observed on subtidal weed beds to a depth of 10m. A size-related change in the diet of S. durbanensis was observed. The juveniles were omnivorous, feeding predominantly on gastropods, echinoids and chlorophytes. The adults fed on a variety of large, reef-associated invertebrates. P. grande were omnivorous, with macroalgae being the principal dietary component. This species appears to utilise the storage and extracellular carbohydrates of the macroalgae. Results showed that macroalgal degradation by gut endosymbionts and the utilisation of macroalgal epibionts does not appear to occur in this species. A detailed analysis of catch composition and catch per unit of effort In the Port Elizabeth rock-angling fishery and the Eastern Cape spearfishery emphasised the recreational importance of S. durbanensis and P. grande. The localised distribution, slow growth and late maturation in both species results in their being susceptible to over-exploitation in these size-selective fisheries. Current legislation protecting these fish is discussed in relation to the findings of this study and additional management measures are proposed.

- Description: The blacktail, Diplodus sargus capensis and the zebra, Diplodus cervinus hottentotus, sparid fishes endemic to South Africa, are important components of the recreational shore-fishery. To provide a basis for the management of these fish, aspects of the biology of both species were investigated in the Tsitsikamma National Park (TNP). In particular, the role of the TNP was evaluated as a management option for the conservation of both species. Examination of stomach contents showed that juveniles of both species fed predominantly on amphipods, polychaetes and harpacticoid copepods. Adult !h. sargus were omnivorous and fed opportunistically on a wide variety of reef associated invertebrates and algae including echinoids, polychaetes, anthozoans, ascidians and rhodophytes. The diet of adult D. cervinus was more specialized with a preference shown for polychaetes and amphipods. Seasonal differences were apparent in the diets of both species where considerably more amphipods were consumed in winter than in summer. Feeding requirements of both species were reflected in their habitat preferences. Visual underwater assessment revealed that both species were most abundant on turbulent inshore reefs (1-10m). !h. sargus were observed over a wide variety of reef associated habitats. In the literature they have also been recorded in the surf zone of sandy beaches and in the lower reaches of estuaries. !h. cervinus, on the other hand, were more specific in their habitat requirements and were observed in greatest abundance on inshore reefs, often in close association with caves or overhangs. A comparison between the relative abundance and size structure of both species in the TNP with that of an exploited area immediately adjacent to the reserve, showed no significant differences. This was attributed to the low level of exploitation by rock-and-surf anglers occurring in the exploited study area, as well as the possibility of seeding of eggs and larvae, or emigration of adults from the TNP. An investigation of the reproductive biology of both fish showed that !h. sargus had an extended summer spawning season while in !h. cervi nus it was more restricted. Detailed histological examination of gonadal development showed that !h. sarqus were dygynous with partial protandry occuring in the male population. ~ cervinus were shown to be rudimentary hermaphrodites. Size at 50% maturity in ~ sargus and ~ cervinus was determined at 225 and 285mm fork length respectively, corresponding to ages of 4 and 6 years. An age and growth study based on the examination of sectioned otoliths showed that both species were slow growing capable of reaching ages in excess of 20 years. Growth in ~ sargus and ~ cervinus was described by the von Bertalanffy growth equations: L(t) = 309.44(1-e-0 . 247[t+l. 048 l) and L(t) = 396.85(1-e-0.146[t+2.148J) respectively. Life history characteristics of D. sargus and D. cervinus including slow growth, late maturation and occupation of a localised, demersal habitat showed that both species were vulnerable to the effects of over-fishing. Due to the present increase in the number of participants and the decrease in catch per unit effort in the recreational shore-fishery, more stringent management recommendations were proposed to ensure the adequate protection of both species. These included an increased minimum size limit and a decreased bag limit for both species. Based on the residency shown by both species and their high relative abundance in the TNP, marine reserves were considered to be a valuable addition to the suite of management options available for the conservation of these species . In this respect the reserve provided both protection for the spawner stock as well as the potential to seed adjacent areas.

- Description: This thesis represents a detailed investigation into aspects of the biology, ecology and population dynamics of two endemic ariid species, Galeichthys feliceps and G. ater, off the southeast coast of South Africa. The two species are exploited as a by-catch in the commercial ski-boat fishery off Port Alfred, a fishery dominated by highly fecund sparid and sciaenid species. They collectively constitute approximately 10% of the total annual catch in terms of landed mass G. feliceps outnumber G. ater in the catches by a ratio of 3:1. The investigation was designed to provide the biological data required for stock assessment and to determine optimum management strategies for the two populations. The implications of their K-selected life-history styles for exploitation received particular attention. While the two species were sympatric and had similar depth distributions they were found to be allopatric with respect to their foraging habitats. G. feliceps foraged over sandy and muddy substrata in marine and estuarine environments. G. ater fed only on reef-associated species and did not utilise estuaries. Their feeding-associated morphologies were identical and both species preyed primarily on crustaceans (brachyuran crabs and isopods), echiurids, molluscs and polychaetes. The diet of G. ater was broader in terms of the number of species consumed. The two species are mouth-brooders with low fecundity. G. feliceps and G. ater produced a mean of 49 and 32 eggs each, per annum. The buccal incubation period was determined to be in the region of 140 days for G. feliceps. Embryos hatched after approximately 75-80 days and the young began exogenous feeding thereafter. The young fed intra-buccally on detritus provided by the parent. Adult buccal mucus may also have been used as a food source. Young were released at a total length of ± 55mm. Adult males ceased feeding whilst mouth-brooding. Body musculature, abdominal fat and liver reserves provided energy for basal metabolism and males lost approximately 28% of their body mass during buccal incubation. Females expended less reproductive energy than males. Catches were dominated by mature fish (76% in G. feliceps and 97% in G. ater). Females were significantly more abundant in catches during the spawning and mouth-brooding period. The female to male sex ratios were 1.65:1 and 2.23:1 for G. feliceps and G. ater respectively. Age and growth studies revealed that the two species mature at advanced ages (10 and 9 years for G. feliceps and 9 and 7 years for G. ater males and females respectively). They are long-lived, reaching ages in excess of 18 years in G. feliceps and in excess of 15 years in G. ater. Females live longer than males and grow larger. Yield-per-recruit and spawner biomass-per-recruit analyses demonstrated that G. ater were exploited below FO.1 at a level where spawner biomass-per-recruit was reduced to between 45% and 65% of the unexploited level. The G. ater stock was not adversely affected by current levels of fishing effort. For G. feliceps, both sexes were exploited beyond F₀.₁ where spawner biomass-per-recruit was reduced to between 30% and 22% of the unexploited level. G. feliceps were shown to be sensitive to relatively low levels of exploitation, a phenomenon attributed to their highly Kselected life-history style. Should the species become targeted for in the future, effort restrictions in the form of a closed season during the spawning and mouth-brooding period would prove effective in reducing effort and conserving the population sex ratio.

- Description: Global trends in teleost fisheries indicate significant population declines. Thus, alternative fisheries are being developed to meet the growing economic and nutritional demands of the expanding human population. Recently, it has been established that elasmobranch fisheries may fulfill these demands. As many elasmobranchs possess life-history characteristics that make them particularly vulnerable to overfishing, it is imperative to develop management strategies prior to the inception of these fisheries to ensure sustainable resource utilisation. In South Africa, elasmobranchs have been commercially exploited since the 1930s. Although generally considered an under-exploited resource, the potential for growth within these fisheries has been recognized. In 2005, the commercial shark fishery will undergo a transition from medium to long-term rights allocations. This represents an ideal opportunity for scientists and managers to develop precautionary species-specific management plans for commercially exploitable elasmobranch species. The soupfin shark (Galeorhinus galeus) is one of the principal target species in South Africa’s shark fisheries. Given its inherent susceptibility to overexploitation, G. galeus was selected as a management priority by South Africa’s regional fisheries organisation. The purpose of this study was to examine and describe the stock status of G. galeus in South Africa, and to develop a precautionary fishery management plan to ensure the sustainability of this resource. Age, growth, and mortality calculations for G. galeus were made from research survey data collected between 1996 and 1999. A small sample size precluded independent analyses of females. The maximum recorded age for G. galeus was 33 years. Estimated von Bertalanffy growth parameters from observed length-at-age for males and combined sexes were: L∞ 1542.8 mm TL, K 0.21 year⁻¹ , t₀ -2.79 year⁻¹ and L∞ 1560.3 mm TL, K 0.19 year¹, t₀ -3.03 year⁻¹, respectively. The age-at-50% maturity was determined to be 6 years, corresponding to 1011 mm TL for males and 1100 mm TL for combined sexes. Natural mortality was calculated as 0.126 yr⁻¹. The rate of instantaneous total mortality was calculated as 0.27 yr⁻¹. Catch trend analysis showed that catches and CPUE of G. galeus are increasing in the demersal longline fishery, and decreasing in the handline fishery. Decreasing catches and CPUE were observed in fishery-independent research survey data. The status of the soupfin shark stock was modelled using per-recruit analysis. The SB/R model indicated the soupfin shark is being optimally exploited and spawner biomass is at 43% of preexploitation levels. Current fishing levels ( F = 0.14 yr⁻¹) approximate the Fsb₄₀ level (F = 0.17 yr¹); thus, an increase in fishing pressure may lead to stock collapse. It was determined that the current age-at-capture (7.9 years) should be increased to 10 years, or 1420 mm TL, to maximize yield and minimize the possibility of recruitment failure. The results of this study indicate a need for immediate scientific and management intervention in South Africa’s soupfin shark fishery. An assessment report and fishery management plan for G. galeus was compiled, and several management options were proposed. These include the implementation of licence and size restrictions, as well as seasonal/area closures. The potential for an experimental gillnet fishery should be investigated.

- Description: The three major species Lethrinops ''pinkhead'', Otopharynx argyrosoma "red" and Copadichromis. cf virginalis which used to dominate the catches (by weight) of Lake Malombe in the past decade were investigated. In order to make rational recommendations regarding size and boundary of the existing sanctuary area (lightly fished), distribution and abundance, population structure and reproductive biology of the three species were related to habitat types of the southern part of the lake (south western side and south eastern side). Analysis of catch composition showed that species richness was higher in the south eastern side than in the south western side of the lake. Rare species such as Labeo mesops and Opsaridium microiepis were observed in the south eastern side and there was visual absence of such species in the south western side of the lake. The overall fish abundance was significantly greater in the south eastern side than in the south western side of the lake. In the south eastern side, the catch per unit effort (CPUE) was twice that of the south western side (P< 0.05). Seasonal fluctuations in CPUE were also prominent, with the highest peak occurring during September-October period. However, there was an indication of weak relationship among CPUE, phytoplankton biomass, temperature and water depth. The length-weight relationships revealed that growth of the three species closely followed the cubic law for isometric growth in both sides of the lake. Differences in regression coefficients between sides were not significant (ttest, P>0.05). The length-frequency distribution analysis revealed that the three species had the same growth rate regardless of habitat type. However, females grew faster than males. An investigation of reproductive biology showed that the three species have low fecundity and they are asynchronous spawners, with a breeding peak during August and September period. Furthermore, the length-fecundity relationships for L. ''pinkhead'', and 0. argyrosoma "red" indicated that fecundity was more closely related to length in the south eastern side than in the south western side. Based on the above characteristics of L. ''pinkhead'', 0. argyrosoma "red" and C. cf virginalis, and the substrate types, it was strongly suggested that the existing sanctuary (lightly fished) in the south eastern side of the lake could immediately be enlarged to conserve the fish stocks, favourable substratum and aquatic animals that may be of tourists concern.

- Description: There is a paucity of published information on fish utilization of salt marshes outside North America. This dissertation represents the first intensive examination of the ichthyofauna associated with salt marshes in southern Africa and examines the species composition, diversity, size structure, distribution and estuarine dependence of fishes that utilize salt marshes in the Kariega Estuary. The research was concentrated on Taylor's salt marsh, with comparative studies being conducted on other salt marshes and habitats within the marine dominated Kariega Estuary. The possible importance of salt marshes as feeding and refuge areas for fishes was examined, as was the role of salt marshes in the food web of the estuary. The fishes frequenting salt marshes in the Kariega Estuary were predominantly the juveniles of marine species, with Mugilidae being the dominant family. The ichthyofauna was distributed primarily in the intertidal creeks with very few specimens captured on the vegetated Spartina maritima and Sarcocomia perennis flats. The different reaches of the intertidal creek were characterised by distinct fish assemblages. The fish assemblages associated with the intertidal salt marsh creeks were significantly different from those found in the eelgrass beds, the other dominant intertidal habitat in the Kariega Estuary. The eelgrass beds were dominated by estuarine fish species and had a higher density and standing stock of fishes when compared to the salt marsh creeks. The diversity of fishes in the two habitats was however similar. The intertidal salt marsh creek ichthyofauna also differed significantly from that found in the main estuary channel. The creek ichthyofauna was dominated by 0+ juveniles while the main channel had many subadult and adult fishes. The channel habitat also had numerous large piscivorous fishes which were absent from the salt marsh creeks. The low number of piscivorous fishes, together with limited fish predation from other sources, may be the reason why salt marshes provide a refuge for juvenile fishes that frequent these habitats. Unlike previous studies on North American, Australian and European salt marshes, the dominant fish species that frequented Taylor's marsh were not recorded feeding extensively on the marsh, and those that did had a limited distribution. The role of fishes in the transfer of energy off the Kariega salt marshes is therefore likely to be minimal. A stable carbon isotope study on the dominant primary producers and consumers within the Kariega Estuary revealed that detritus originating from the high lying salt marsh plants Sarcocornia perennis and Chenolea diffusa was not utilized by fishes in the Kariega Estuary. Preliminary results indicated that the cord grass Spartina maritima may be an important energy source to the fishes in the Kariega Estuary, but further research is needed to confirm this.

Aspects of the feeding ecology of three common Sparid fish in the littoral zone at Clayton's Rocks in the Eastern Cape, with notes on their biology

- Description: The feeding interrelationships of three sparid fish was studied in the littoral zone during low tide at Clayton's Rocks in the Eastern Cape. The three species appeared in the study area only when between 9 and 10 mm long (SL), and remained resident until a length of approximately 120 fmm was reached. No sexually mature specimens occurred in the study area as large adults only use the intertidal zone for feeding purposes at high tide. From first recruitment into the littoral zone at 9 mm SL until some 35 mm long, the diet of Diplodus sargus consists of harpacticoid copepods and amphipods. Ingestion of amphipods and green algae increases with increased size of fish. Chironomid larvae, cirripede nauplii and an unidentifiable planktonic larva are important food items of small juveniles ( 50 mm), while diatoms are significant in larger juveniles (25 to 80 mm) in the spring/early summer season. The gut is short and the teeth are incisiform, backed by several rows of molars which increase in size and number with age. D.cervinus was almost completely carnivorous in the size range studied. The juveniles found on harpacticoid copepods and chironomid larvae while between 10 and 20 mm long. The diet then consists mainly of the shrimp Palaemon pacificus until the fish are 50 mm long and then amphipods until 100 mm long. All fish larger than this feed prodominantly on polychaetes. The gut is short and the teeth are similar to those of Q. sargus although there are fewer molars. From its first appearance at 10 mm, Sarpa salpa feeds mainly on harpacticoid copepods until 25 mm long. Diatoms then predominate in the diet, red algae also being taken until 75 mm long. Larger size classes are herbivores, feeding almost equally on red and green algae. Corresponding changes in gut length and dentition are reported, juveniles having a short gut and conical teeth and adults a long intestine and cuspidate, incisiform teeth. Marked ecological separation was observed between the three species. Spatial, temporal , behavioural and dietary differences were found . Q. sargus has continuous recruitment of juveniles into the littoral zone, S..salpa appears between July and early September and Q.cervinus is recruited from late September to November. Competition between small juveniles is reduced by cyclic abundances of food items important to each species and the sub-adults feed on different foods. Some competition exists between large juveniles of Q. sargus and Q.cervinus, but separation is maintained as individuals of the two species feed at different levels of the water column and in different parts of the littoral zone.

- Description: Past and current research on the biology and culture of the sharptooth catfish (Clarias gariepinus) has stimulated the development of its commercial production. At the inception of this project in 1985, it was decided that the development of an optimal dry feed was the most important step in developing a more complete culture technology for the species. The objectives of the project were, therefore, to investigate aspects of the nutritional physiology and dietary requirements of the species to provide the necessary information for the formulation of optimum economical feeds. A review of the natural feeding biology of the species provided valuable information with regard to its food preferences. From this information, and by estimating the nutritional composition of its preferred natural diet, it could be concluded that the animal requires a relatively high dietary protein content. This conclusion was substantiated by studies of the functional morphology of the digestive system (gross and fine structure). The characteristics of the digestive system and external anatomy were found to be typical of an opportunistic, omnivorous predator. The ontogenetic development of the digestive system is relatively fast, and enables the utilization of a variety of diets efficiently from an early stage. Since the ability of an animal to digest a given substance is predominantly dependent on the presence of appropriate enzymes the quantification and characterization of C. gariepinus digestive enzyme activities were investigated on an experimental basis. This work revealed that the animal has relatively high levels of digestive enzyme activities, and that its proteolytic enzyme activities correspond with those of other carnivorous fishes, while its starch digesting abilities correspond with those of specialized herbivores. Also, the high levels of lysozyme and alkaline phosphatase indicate an adaptation to detritivory, making this fish a truly opportunistic omnivore. Evidence was found to suggest that sharptooth catfish are able to re-absorb secreted digestive enzymes in the posterior section of its intestinal tract. It was shown that digestive enzyme activities were induced by food intake, and that no inherent rhythmicity in digestive activities seemed to occur. The development of digestive enzyme systems in the early life history of this animal was found to be also extremely rapid (complete within 10-16 days after hatching). It was also shown that artificial dry feed stimulates higher levels of digestive enzyme activities in larvae and juveniles than natural food. Although increased cellulase activity was found in larvae and juveniles that were fed on live zooplankton, the relative contribution of exogenous prey enzymes to protein digestion could not be ascertained. Specific nutritional requirements were investigated by means of feeding trials. The results of these trials confirmed the above indications regarding the dietary requirements of sharptooth catfish. It was concluded that for optimal growth and production performance, this animal requires a dietary protein content of 40-42%, a dietary lipid content of 10-12% and that diets should contain 14-16 kJ/g digestible energy. The optimum protein-to-energy ratio was found to be 26-29mg protein per kJ of digestible energy. Dietary requirements are evidently the same for juveniles and adults, except that younger fish require higher relative feeding levels. Finally, practical feed formulations were evaluated in terms of their economic feasibility. The results of these trials are presented along with recommendations on the formulation of commercial feeds for C. gariepinus. A computer program with which to implement least cost feed formulation by means of linear programming is provided.

- Description: The source of abalone for human consumption has shown a dramatic shift away from wild-capture fisheries in the last 30 years, with over 90% of global production now coming from aquaculture. Farmers initially relied on the natural food of abalone (macroalgae) as a culture feed, though in regions where macroalgae availability was limiting, the need to develop formulated feeds was evident. Extensive research effort has led to the development of a number of formulated feed products currently employed in the industry. These feeds, however, differ markedly from the mixed macroalgal diets that abalone have evolved to utilise, particularly in terms of protein content and carbohydrate structure. The degree to which the nutritional physiology of abalone responds to these novel formulated diets, with and without macroalgal supplementation, was investigated in the current study. A multifaceted approach, combining growth trials, stable isotope nutrient tracers and metabolic experiments, was employed to gain insight into the post-absorption dynamics and utilisation of dietary nutrients under varying dietary regimes of fresh macroalgae and formulated feed. Growth trials conducted with both Haliotis rufescens and H. midae showed significantly higher growth and protein utilisation efficiency for abalone fed macroalgal diets compared to formulated feeds. Furthermore, when formulated feeds were supplemented with macroalgae to form combination diets, growth and the utilization of protein was improved compared to the formulated-feed-only diet. The poor utilisation of protein by H. midae fed the formulated feed could be traced, using a method combining stable isotope bio-markers with a Bayesian mixing model (SIAR), to the low incorporation of the fishmeal component of protein in the diet. The marked postprandial drop in the O:N ratio on abalone fed formulated feeds indicate that the protein was being diverted into catabolic metabolic pathways. The metabolic cost of digestion, termed specific dynamic action (SDA), was negated as a factor in the improved growth of abalone fed macroalgal diets, with the SDA coefficient 2.1 times that observed for formulated feed. Furthermore, the postprandial haemolymph glucose concentration (HGC) in H. midae was elevated when fed formulated feed compared to macroalgae. The high levels of circulating glucose are likely a result of the structure the carbohydrate source in formulated feeds and stimulate the deposition of glycogen through the allosteric control of glycogen synthase. Formulated feeds produced higher cooked meat yields in canning simulation trials, suggesting that muscle glycogen content may indirectly play a role in increasing canning yields through the displacement of collagen. The results of these empirical studies are synthesised under key themes, discussed within the context of their potential commercial relevance and future research directions are highlighted.

- Description: A physiological study was carried out to determine the effects of temperature, quantity of food fed, feeding activity and size class on the oxygen consumption and ammonia production of the South African abalone Haliotis midae L.. A set of intermittent flow respirometer chambers were used in a recirculating system to measure the oxygen consumption rates of fed (postprandial) and unfed (postabsorptive) abalone for three different size classes (15, 30, and 50mm) at three different temperatures (16°, 20° and 23°C) over a 24 hour period. Ammonia production rates and food consumption rates were simultaneously determined. Oxygen consumption and Ammonia production rates per gram abalone were linear functions of abalone size, temperature, and mass of food consumed. Oxygen consumption and ammonia production rates were found to increase proportionally to temperature and in inverse proportion to abalone size. These rates were significantly higher for fed as opposed to unfed abalone. Equations were derived to predict oxygen consumption and ammonia production for fed and unfed animals for a range of temperatures and size classes of abalone. A 96h LC50 lethal toxicity test for exposure to ammonia established 1.08mg.1⁻¹ of unionised ammonia as the lethal limit. A test on the long term effects on growth of acute exposure (12h) to various sublethal concentrations of ammonia showed no significant effects on growth for concentrations below 0.88 mg.1⁻¹. A safe chronic exposure level of 0.02 mg.1¹ NH₃-N was used to predict optimal flow rates required per kilogram of abalone in a rearing tank over a range of size classes and temperatures. The physiological and biological requirements of H. midae and the implications of this knowledge for rearing tank management and optimal design are discussed.

- Description: Afromontane regions represent some of the highly threatened ecosystems on the planet as they harbour endemic and often relic freshwater fauna. These ecologically sensitive ecosystems have been altered by multiple impacts, including invasion by non-native fishes, replacement of diverse indigenous vegetation with monoculture plantations, agricultural and mining activities, hydrological modifications, and degradation of instream habitats, with potential detrimental effects on aquatic community structures and food web dynamics. The aim of the present study was to compare spatio-temporal patterns of macroinvertebrate and fish communities as well as food web dynamics in three differently impacted headwater tributaries of the Kat River in the Amathole-Winterberg freshwater ecoregion in the Eastern Cape Province of South Africa. The upper Kat River catchment was the ideal model for purposes of the present study as it contains streams with varying degrees of human impact. The streams considered in the present study were (i) the Eyre River which was considered to closely represent near-natural conditions as it is the least impacted stream in this catchment, with the riparian zone dominated by relatively intact and diverse native woody vegetation, (ii) the Elands River whose catchment has been altered by agricultural activities, and (iii) the Lushington River, whose riparian zone has been heavily invaded by black wattle. The three headwater streams were generally distinguished based on the physical-chemical variables. The Elands River was characterised by high conductivity and total dissolved solids (TDS). In addition, the Elands River was more alkaline and relatively warmer than the other two rivers. This suggested the negative influence of agriculture activities on the water quality in the Elands River. In general, the Lushington and Eyre rivers had comparable physical and chemical variables. However, the Lushington River was generally characterised by low streamflow, likely as a consequent of black wattle which is known for altering hydrological regimes of streams .The Eyre River and Lushington River were comparable in terms of macroinvertebrate richness and diversity, whereas the Elands River was characterised by low macroinvertebrate diversity and richness. Redundancy analysis indicated that the macroinvertebrate communities were mostly influenced by seasonality, with land-use accounting for a small but significant difference in community composition. In comparison, the generalised linear mixed-effects models (GLMMs) showed that chubbyhead barb abundance was significantly influenced by land-use patterns. Evaluation of food web dynamics using stable isotope analysis revealed that the food web structure in the three rivers differed substantially as a result of land-use. Specifically, the Eyre River was characterised by less variation in stable isotope values of basal food sources and consequently, the consumer groups had a narrow isotopic breadth. In contrast, the Elands River was characterised by a wide variation in basal food sources and therefore, a wider isotopic breadth for consumers. The Lushington River was spatially variable in terms of basal sources and isotopic breadth for consumers. The study concluded that food webs and trophic interrelationships were a more informative indicator of land-use than community structure was in evaluating the impact of land-use patterns on aquatic communities. In addition, future studies should seek to investigate food webs interrelationships in addition to community structure to infer a more conclusive river assessment.
