The role of arbuscular mycorrhizal fungi in the biotransformation of coal and application in dump rehabilitation

  • Description: Fundamental processes underpinning the biotransformation of coal by fungal biocatalysts have been intensively investigated, however, limited large-scale industrial applications using such systems have been reported. The un-anticipated sporadic growth of Cynodon dactylon on the surface of un-rehabilitated discard coal dumps has been noted and this was found to be coupled with the breakdown of coal into a humic soil-like material in the top 1.5 metres of the dumps. Extensive fungal growth was observed to be associated with the Cynodon dactylon root system and examination of plant roots indicated the presence of mycorrhizal fungi. Analysis of the Cynodon dactylon plant roots around which coal biotransformation was occurring confirmed the presence of arbuscular mycorrhizal colonisation with the species Glomus clarum, Paraglomus occultum, Gigaspora gigantea and Glomus mosseae identified to be associated with the plants. Further molecular characterisation of non-mycorrhizal rhizospheric fungi showed the presence of fungal species with coal-degrading capabilities that most likely played a role in the coal biotransformation observed. The discard coal dump environment was simulated in pot and column studies and coal biotransformation was reproduced, with this process enhanced by the addition of mycorrhizal and non-mycorrhizal rhizospheric fungal inocula to the environment. Mycorrhizal and non-mycorrhizal species in the inoculum were re-isolated from the simulated environment fulfilling a number of Koch’s postulates and indicating a causal role in the biotransformation of coal. An inversion of conventional mycorrhizal colonisation was demonstrated in this system with reduction in extraradicular presence and an increase in intracellular colonisation compared to soil controls. A descriptive model was formulated suggesting a two-part fungal system involving organic carbon and nutrient exchange between the plant, mycorrhizal fungi and non-mycorrhizal coal-degrading rhizospheric fungi ultimately resulting in the biotransformation of coal. The biotransformation observed was comparable to reports of “rock-eating fungi”. Results suggest that the biological degradation of coal in situ with the production of a soil-like substrate could provide a feasible method of discard coal dump rehabilitation as well as provide a humic-rich substrate that can be utilised in further industrial applications.

  • Description: Over the past couple of decades traditional manufacturing techniques have been replaced by lean production around the world. This change in production methodology brought about the opportunity for numerous companies to reduce costs and customer lead time through the application of this lean philosophy. The worldwide shift from traditional manufacturing to lean manufacturing can be ascribed to the success of Toyota Motor Company. Lean manufacturing refers to a manufacturing improvement process in order to minimize or eliminate waste while maximizing production flow (Tapping, et al., 2002:30). A value stream includes all the operations and processes to transform raw material into finished products or services. Value stream is a management tool used for the planning of a production process involving lean techniques through systematic data capturing and analysis (Tapping 2002 et al., 41). Value stream is a proven process for planning the improvements that will allow companies to develop lean practices. One key to Toyota’s success that GMSA and many other South African companies have not been able to emulate is the transformation of their suppliers to apply the lean philosophy. This lack of supplier transformation is due to various reasons including supplier proximity, supplier relationships, supplier performance levels, and the ordering policies used for supplied parts. Even though many manufacturing organizations realize the importance of practicing lean manufacturing techniques, few organizations apply lean techniques with the required knowledge and tools to transform their organization from traditional manufacturing to lean manufacturing. This research project is based purely on the lean manufacturing principles and philosophies. The aim of the study is to make GMSA’s suppliers aware of the principles and processes of lean manufacturing and to develop a lean implementation strategy to assist organizations with the successful implementation of lean practices. In order to correctly implement and sustain lean manufacturing practices this study will also focus on supplier support and development and the behavior an organization must exhibit to make this transformation a success. The research methodology comprised the following steps: • A literature study was performed by the researcher to get a better understanding of the principles and philosophies of lean manufacturing; • A second literature study was also performed to get a better understanding of the continuous improvement philosophies of lean manufacturing including supplier support & development; and • To accomplish the objectives of this study, interview sessions were conducted with four senior managers of General Motors South Africa. The researcher selected the interviewees from the following departments within General Motors South Africa: Global Purchasing and Supply Chain; Supplier Quality and Development; Vehicle Assembly Operations and Material Supply. The interviewees from General Motors South Africa were identified as qualified data sources for this research project, as their professional opinions and viewpoints could best address the research questions.

The role of pacC in Aspergillus flavus

  • Description: Many microorganisms, and in particular fungi, are able to grow over a wide pH range. Thus, these microorganisms must possess some regulatory mechanism or system that senses the environmental pH signal and ensures that gene expression of certain molecules is tailored to the pH of the environment (Penalva and Arst, 2002). In Aspergillus species and several other fungi, pH regulation is mediated by seven genes viz. palA, palB, palC, palF, palH, palI and the global pH regulatory gene, pacC (MacAbe et al, 1996; Negrete-Urtasun, 1999; Denison, 2000). The activated form of the PacC protein activates genes that are required at alkaline pH, e.g. genes coding for alkaline phosphatases, and represses certain genes that are functional at acidic pH, e.g. genes encoding acid phosphatases (Negrete-Urtasun, 1999). PacC (and its homologues) also positively regulates genes involved in penicillin biosynthesis, e.g. the isopenicillin N synthase gene, ipnA, in A. nidulans (Penalva and Arst, 2002). It has also been hypothesised that pacC may negatively regulate aflatoxin biosynthesis, a carcinogenic secondary metabolite in several species of Aspergillus (Keller et al, 1997). To elucidate the role of pacC a novel method of post-transcriptional gene silencing known as RNA interference was utilized. This method involved the cloning of a partial pacC gene fragment first in the forward and then the reverse orientations in a fungal expression cassette to create an RNA interference (RNAi) vector. The unique structure of this vector would allow the cloned fragments to be expressed and the resulting RNA to immediately form a double stranded stem-loop structure or short hairpin RNA (shRNA; McDonald et al, 2005). The formation of this shRNA, in turn, would be responsible for activating the endogenous RNA degradation complexes that would lead to mRNA degradation and subsequent gene silencing (Liu et al, 2003; Kadotoni et al, 2003; McDonald et al, 2005). The results presented here have shown that confirmed pacC RNAi mutants produced aflatoxins irrespective of environmental pH (i.e. the mutants produce aflatoxins under acidic and alkaline conditions). Thus, pacC is essential for pH regulation of aflatoxin production in A. flavus. There are numerous other biological (e.g. presence of oxylipins, lipooxygenases) and non-biological factors (pH, carbon source etc.) which affect maize colonisation and aflatoxin production by A. flavus (Burrow et al, 1996; Wilson et al, 2001; Calvo et al; 2002; Tsitsigiannis et al, 2006). However, all the genetic mechanisms involved have as yet not been identified. It has been shown by Caracuel et al (2003) that pacC acts as a negative virulence regulator in plants and these workers have hypothesised that PacC prevents expression of genes that are important for infection and virulence of the pathogen. Therefore the physiological effects that pacC silencing had on the growth, conidiation and pathogenicity of A. flavus mutants were also investigated. The results of this study showed that pacC does not play a significant role in primary growth and development but does affect conidial production. SEM results showed that mutants have many “open ended” phialides and poorly developed conidiophores. This would suggest that pacC activation of conidial production genes is also required. Furthermore, pacC RNAi silencing severely impaired the ability of the A. flavus mutants to infect and cause damage on maize. The results obtained here are similar to that of pacC null mutants in A. nidulans, C. albicans and F. oxysporum which also exhibited low pathogenicity (Davis et al, 2000; Fonzi, W.A, 2002; Caracuel et al, 2003; Bignell et al, 2005 and Cornet et al, 2005). This study indicates that pathogenicity of A. flavus on maize is directly related to the structural integrity of conidia, which in turn is greatly influenced by PacC. This gene is a global transcriptional regulator and may either repress or activate one or many genes in each of the above pathways (Penalva and Arst, 2002). Studies on the genetic mechanisms of pacC regulation on these pathways are needed to elucidate the mechanisms of activation or repression of these genes.

  • Description: This investigation looks at the role played by school-based studies in the development of reflective practice among student teachers at a Namibian college of education. Literature reviewed indicates that reflective practice helps teachers continually grow and enhance their professional insights and practices. The study focused on the perceptions and experiences of a group of teacher educators, support teachers and student teachers involved in a particular teacher education programme. A case study was chosen, and built up using an interpretive paradigm. This enabled the researcher to make meaning of information often taken for granted. Triangulation was used to ensure the development of as broad and authentic a picture of the case as possible. Key findings reveal that despite claims that Namibia’s three-year Basic Education Teacher Diploma programme (BETD) is explicitly designed to facilitate the development of critical reflective and reflexive practice in student teachers, the way in which aspects of the programme are implemented frequently undermines this design intention. A lack of careful and explicit mentoring on the importance of reflection for enhancing teaching, plus a measure of distrust between student teachers and teacher educators prevents college students from fully exploiting the opportunities afforded by school-based studies for the development of reflective skills.

  • Description: Thirteen years in the new democratic South Africa, South Africa is still faced with socio-economic problems such as high rates of unemployment, shortage of housing, crime and HIV/Aids. Buffalo city falls within the Amathole District Municipality (ADM). ADM population is estimated at + 1, 67 million, being predominantly rural and living in low socio-economic conditions. The demographic trends of ADM population depict high poverty, illiteracy and unemployment rates, rendering them prone to high morbidity and mortality (www.amathole.gov.za, 2007). The aim of this research was to assess the role of the SMMEs in the socio-economic development of Buffalo City. This study is based on exploratory quantitative and qualitative research methodologies. Using a convenience sampling technique structured questionnaires were used to collect data amongst 28 SMMEs in Buffalo City. The findings of this study suggest that SMMEs play a vital role in the socio-economic development of Buffalo City. The SMMEs create employment and incomes; provide human capital investment in form of training programs and HIV/Aids programs; make donations to community structures; give sponsors to various sports clubs and food to the homeless. Finally SMMEs contribute towards tax revenues that in turn help reduce poverty and redistribute wealth. However, SMMEs in Buffalo City face a number of constraints, namely, lack of access to funding, lack of operating space, and high cost of property to lease and difficulty in finding trained competent staff. Further, the perceptions of the SMMEs about the adjudication of tenders was some biasness in the adjudication of tender in favour of those who were close to the public officials. There was no accountability and professionalism amongst the adjudicating officials. Finally the government was not doing enough to encourage SMME development in Buffalo City. In view of the socio-economic benefits of the SMMEs in Buffalo city, it is recommended that support programmes to the SMMEs should be enhanced. On the basis of the findings in this study, it is suggested that assistance to the SMMEs should go beyond institutional support such as Ntsika, Khula, DTI and/or SEDA but should be targeted to funding opportunities for the SMMEs. Commercial banks should be involved in ensuring that SMMEs obtain access to funding. Infrastructural facilities such as affordable business premises should be provided for the SMMEs. Affordable premises will reduce the overhead costs of the SMMEs and in turn increase the profits of these SMMEs. Increase the profits of the SMMEs will ensure the survival of the SMMEs and will in turn contribute towards the upliftment of the socio-economic status of the people who would have otherwise been unemployed, destitute and poor.

  • Description: Cardiovascular disease contributes to mortality and morbidity statistics worldwide and in South Africa. The current focus in health care revolves around activities aimed at preventing the development of cardiovascular disease, rather than the treatment of disease. The identification of risk factors that can predispose a patient to the development of cardiovascular disease is an essential component of any cardiovascular disease management programme. It is necessary that in the management of these risk factors, they are not considered to be isolated, but inter-related. Through the provision of point-of-care cardiovascular risk screening and monitoring services as well as disease-related counselling, the community pharmacist, as a readily accessible source of healthcare, can play an essential role in the cardiovascular disease management process. The aim of this study was to describe the nature of the services provided by community pharmacists with respect to cardiovascular risk and disease management in the Nelson Mandela Metropole. The research design was a non-experimental, descriptive study using a crosssectional survey method. Data was obtained through the utilisation of a questionnaire. The questionnaire consisted of three sections and was administered to community pharmacies in the Nelson Mandela Metropole, that provided cardiovascular point-of-care screening services. The community pharmacists correctly identified cardiovascular risk factors such as obesity (76.6 percent; 36, n=47) and smoking (27.7 percent; 13, n=47). Other cardiovascular risk factors such as abdominal obesity (4.2 percent; 2, n=47), gender (2.1 percent; 1, n=47) and family history (4.2 percent; 2, n=47) were largely ignored by the pharmacists. Point-of-care testing services were readily available in the pharmacies, with all of the pharmacies providing blood glucose and blood pressure measurements. Blood cholesterol measurements were only provided in 87.8 percent (36, n=41) of the pharmacies. The services were generally provided in a clinic facility, with 90.2 percent (37, n=41) of the pharmacies having a clinic facility available. Pharmacists were involved in the provision of point-of-care services, with 85.4 percent (35, n=41) of the pharmacies indicating that the pharmacists participated. Pharmacists readily provided counselling prior (70.7 percent; 29, n=41) to and after (80.5 percent; 33, n=41) the conduction of the screening services on areas such as lifestyle modification and treatment options. Only 15 percent (7, n=47) of the pharmacists indicated that they were aware of Cardiovascular Risk Calculator Tools and none of the pharmacists indicated that they had utilised such a tool. Pharmacists recommended frequent monitoring (60.5 percent; 26, n=43) and lifestyle modification (67.4 percent; 29, n=43) to patients, if the result of their screening service was within normal limits. However, the majority of the pharmacists indicated that they would refer patients, if the results obtained were out of the normal range. Conclusions based on the findings indicated that the pharmacists are readily providing cardiovascular risk screening services. The pharmacists were also able to identify the presence of any risk factors that can lead to the development of cardiovascular disease in the patients. However, active pharmaceutical involvement in further cardiovascular disease monitoring seemed to be lacking. Recommendations were made on areas such as reimbursement for pharmaceutical care services, increased utilisation of support staff and Continuing Professional Development events that could assist in improving the role of the community pharmacist in cardiovascular disease management.

The role of the euthecosome pteropod, limacina retroversa, in the polar frontal zone, Southern Ocean

  • Description: The aim of the present study was to assess the ecological role of the euthecosome pteropod, Limacina retroversa, in particular, and the mesozooplankton community, in general, in the pelagic ecosystem of the Polar Frontal Zone (PFZ), Southern Ocean. Data were collected from four oceanographic surveys to the Indian sector of the PFZ during austral autumn 2000, 2002, 2004 and 2005. Copepods, mainly Calanus simillimus, Oithona similis, Clausocalanus spp. and Ctenocalanus spp., typically dominated total mesozooplankton counts, accounting for, on average, between 75.5 % and 88.1 % (Mean = 77.4 %; SD = 13.4 %) of the total, during the present investigation. Results of the study indicate that L. retroversa may, at times, contribute substantially to total mesozooplankton abundances. During the study, L. retroversa contributed between 0.0 and 30.0 % (Mean = 5.3 %; SD = 7.1 %) to total mesozooplankton numbers. Significant small-scale variability in abundance and size structure of L. retroversa and abundance of copepods was minimal. Inter-annual variability, on the other hand, was significant between some years. Total pteropod numbers were greatest during April 2002 and 2004, while copepods exhibited greatest abundances during April 2004 only. Pearson’s Correlation analysis suggested that L. retroversa abundances were positively correlated to total surface chlorophyll-a (chl-a) concentrations. The significantly lower chl-a concentrations recorded during April 2005 may explain the reduced pteropod numbers observed during that survey. The size class structure of L. retroversa comprised mainly small and mediumsized individuals during all four surveys. This corresponds well with records from the northern hemisphere (sub-Arctic and Arctic waters) where Limacina spp. are reported to exhibit maximum spawning during mid to late-summer. Higher abundances of large individuals only occurred during April 2005, when chl-a concentrations were very low; possibly the result of delayed spawning, due to reduced food availability. Ingestion rates of the four most abundant copepods, determined using the gut fluorescence technique, ranged between 159.32 ng (pigm) ind⁻¹ day⁻¹ and 728.36 ng (pigm) ind⁻¹ day⁻¹ (Mean = 321.01 ng (pigm) ind⁻¹ day⁻¹; SD = 173.91 ng (pigm) ind⁻¹ day). Ingestion rates of L. retroversa were much higher, ranging from an average of 4 28.68 ng (pigm) ind⁻¹ day⁻¹ in April 2002 to 4 196.88 ng (pigm) ind⁻¹day⁻¹in April 2005 (Mean = 4157.36 ng (pigm) ind⁻¹ day⁻¹; SD = 35.37 ng (pigm) ind⁻¹day⁻¹). Average daily grazing rates for the pteropod varied between 0.39 mg (pigm) m⁻² day⁻¹ in April 2005 and 17.69 mg (pigm) m-2 day-1 in April 2004 (Mean = 6.13 mg (pigm) m⁻² day⁻¹; SD = 11.04 mg (pigm) m⁻² day⁻¹); corresponding average daily grazing impacts ranged between 8.4 % and 139.8 % of the phytoplankton standing stock in April 2005 and 2004, respectively (Mean = 48.5 %; SD = 84.5 %). Average daily grazing rates of the four copepods ranged from 4.58 mg (pigm) m⁻² day⁻¹ to 8.77 mg (pigm) m⁻² day⁻¹ -1, during April 2002 and 2004, respectively (Mean = 6.28 mg (pigm) m⁻² day⁻¹; SD = 5.94 mg (pigm) m⁻² day⁻¹). Collectively, the copepods removed an average of between 31.6 % and 89.8 % of the phytoplankton standing stock per day, during April 2002 and 2004, respectively (Mean = 70.8 %; SD = 86.7 %). The daily grazing impact of the copepods accounted for an average of between 40.4 % and 87.8 % of the total zooplankton grazing impact, during April 2004 and 2005, respectively (Mean = 75.0 %; SD = 65.5 %). L. retroversa was responsible for an average of 52.4 % and 59.5 % of the total zooplankton grazing impact, during April 2002 and 2004, respectively. However, during April 2005, when L. retroversa numbers were significantly lower than previous years, the pteropod contributed an average of only 7.5 % to the total zooplankton grazing impact. Thus, during the present investigation,the pteropod was responsible for removing a mean of 48.9 % of the available phytoplankton (SD = 74.9 %). The predation impact of the dominant carnivorous macrozooplankton and micronekton in the PFZ was determined during April 2004 and 2005 using daily ration estimates obtained from the literature. Additionally, gut content analysis was used to determine the contribution of L. retroversa to the diet of the dominant predators. Average predation impact ranged from 1.1 % and 5.7 % of the total mesozooplankton standing stock during April 2004 and 2005, respectively (Mean = 3.8 %; SD = 12.3 %). Chaetognaths and euphausiids dominated total carnivore numbers and made the greatest contributions to total predation impact during both years. Copepods appeared to be the main prey item of the dominant carnivorous macrozooplankton-micronekton in the region. L. retroversa was only detected in the gut contents of the amphipod, Themisto gaudichaudi, but not in either of the chaetognath species (Eukrohnia hamata and Sagitta gazellae) or the myctophid fish (Electrona spp.). The pteropod was found in 19 % of amphipod guts dissected. Pearson’s Correlation analyses showed that the four major predatory zooplankton groups found in the PFZ (chaetognaths, euphausiids, amphipods and myctophid fish) were positively correlated to abundances of L. retroversa, suggesting that the pteropod might be an important prey item for many of the carnivorous macrozooplankton/micronekton in the PFZ. To conclude, L. retroversa may play an important role in the pelagic ecosystem of the PFZ, in austral autumn. However, ocean acidification and calcium carbonate undersaturation (as a result of increased anthropogenic carbon dioxide emissions), that is predicted to occur within the next 50 – 100 years, will most likely have significant implications for the Sub-Antarctic pelagic ecosystem if L. retroversa cannot adapt quickly enough to the changes.

It was found that although their professional contribution is vital in successful implementation, there are other obstacles namely, financial constraints, very high workload, lack of government support and inferior appointment procedures for District Education Officers which tend to hamper them to fulfil their duties.

  • Description: This study uses a systems thinking perspective to explore the role and meaning of participation, capacity development and project sustainability in the Zimbabwe Secondary Teacher Training Environmental Education Project (St²eep). Since there was no consistent critical reflection upon the different assumptions that underpin these important aspects of St²eep in the original project design, this study aims to articulate a theoretical framework for guiding the project. St2eep is a donorfunded project, located in the Ministry of Higher and Tertiary Education in Zimbabwe that seeks to integrate environmental education across the curriculum of secondary teacher education. The study first develops a theoretical framework drawing on systems thinking. In particular it uses the holistic and constructivist perspectives embedded in systems thinking to describe a number of analytic frameworks that are used as a guide to investigate participation, capacity development and project sustainability in the St²eep case study. The research methodology comprises a qualitative case study approach, which contains elements of an instrumental, evaluative and critical case study. Data-collection methods include document analysis, focus group discussions, focus group interviews, semi-structured face-to-face interviews and participant observation. Data analysis follows the constant comparative method of coding and categorising data as outlined by Strauss and Corbin (1998). The outcomes of this research show that participation processes in St²eep evolve around the interactions between a political dimension and a learning dimension. The political dimension relates to giving the project stakeholders a critical voice, allowing them to shape the project and involving them in the decision-making process. This approach has fostered an ongoing learning process in a small team of committed stakeholders based on the principles of collaborative learning, team learning and action research. Participation was shown to enhance capacity-development processes at individual and institutional level by the provision of support through learning teams, and by the renegotiation of responsibilities and power relations between lecturers involved in St²eep, donor representatives and college administrations. The strong operational role of the donor organisation was seen as a serious threat towards individual and institutional capacity development since it creates a functional but artificial and independent project system within the college system and takes over any local institutional support structure that it might seek to develop. Fostering continuous learning and capacity development, St²eep’s participatory approach was shown to contribute to a better understanding of the interconnectedness of factors that influence future sustainability of the project and the implementation of environmental education. This has assisted in the development of different scenarios on the sustainability of the project. The research shows that the project-ustainability planning process draws directly from St²eep’s ongoing learning process, with individual and institutional capacity development featuring strongly in the different scenarios, and with the external context such as the economic situation and the low priority of environmental education being recognised as important factors that need to be considered. Drawing on the findings from the case study, this study makes a tentative recommendation that donor organisations should focus more on capacity-development initiatives and avoid taking on a strong operational role in project activities. The research also recommends that there is need for a deliberate focus on both the political and learning dimensions of the participation process in order to foster local ownership. Making the learning aspect much more central in St²eep is presented as a possible strategy for motivating a larger number of college lecturers to become involved in the project and the implementation of environmental education. The study also urges St²eep to combine the different scenarios that have emerged during the project sustainability planning process and to focus on the benefits that stakeholders want to see sustained.

The significance of the safety-net role of NTFPs in rural livelihoods, South Africa

  • Description: This study was conducted in two rural villages in South Africa with the purpose of investigating the safety-net role of non-timber forest products (NTFPs). The use of NTFPs as a safety-net is reported as a common feature of rural livelihoods however empirical data supporting this function is limited. Therefore, policy makers and land-use planners underappreciate the probable role and consequent value of the safety-net function. The findings show that poor, rural households are vulnerable to a range of risks. Over a two year period 100% of households reported experiencing crises including unexpected idiosyncratic risks (e.g. illness, retrenchment, crop failure and death) as well as expected expenses and periods of vulnerability (e.g. the payment of school fees and seasonal crop shortfalls). Households are prone to these risks irrespective of wealth or gender of the de jure household head. To secure their livelihoods households in both villages rely on a variety of livelihood strategies including waged labour (53%), self-employment (30%), government grants (60%), arable agriculture (56%), animal husbandry (64%) and the use (100%) and sale (22%) of NTFPs. In terms of the proportion of households involved, the findings suggest land-based strategies make an important contribution. Formal employment and old-age pensions distinguish wealthy households from poor and allow for investments in assets and saving schemes (62%). In response to the risks experienced households employed a variety of coping strategies. Generally the most commonly used strategies were kinship (85%), reduced household spending (72%), changes to food consumption and saving (72%) and relying on NTFPs (70%). Kinship and NTFPs show no differences for wealth or gender of the de jure household head. The remaining strategies are influenced by wealth. The use of NTFPs as a safety-net manifested predominantly through the increased use of products, then the substitution of commercial goods by NTFPs and lastly through the sale. Of those using NTFPs as a rural safety-net 41.4% used of wild foods, 40% used medicinal plants, 25.7% substituted paraffin with fuelwood, 10% sold fuelwood and 8.6% sold other products.

  • Description: The United States of America has in its custody several hundred Taliban and Al Qaeda combatants who were captured after the September 11, 2001 attack and during the war in Afghanistan. These prisoners are incarcerated at the Guantanamo naval base in Cuba. The treatment given to these detainees has elicited widespread criticism, as well as unprecedented intellectual and legal debates regarding prisoners of war. In order to fully understand the position of the Guantanamo Bay detainees, one has to be aware of the origins of the prisoner-of-war phenomenon. From biblical times, through the countless conflicts that were waged across the globe through the ages, the concept of “prisoner of war” gradually evolved. Growing concern for the plight of prisoners of war was paralleled by the development of the laws of war, which sought to regulate the conduct of combatants during an armed conflict. The laws of war that have bearing on modern day States are those documented in the Geneva Conventions. The Geneva Conventions regulate armed conflicts and set out the requirements for prisoners of war, as well as their trial rights. The United States, in declaring the Guantanamo Bay detainees “unlawful combatants” or “illegal enemy combatants”, terms which are undefined in International Law, have sought to evade the prescripts of the Geneva Conventions. In direct contravention of the Geneva Conventions, the Guantanamo Bay detainees are denied the right to humane treatment, a fair trial and due process of the law. Prior to Hamdan vs Rumsfeld, the United States’ position was challenged with very little success. The Supreme Court, in Hamdan vs Rumsfeld, directed the president to accord the detainees the protections of the Third Geneva Convention. The relief brought by this decision was very short lived. In September 2006 the United States Congress passed the Military Commissions Act of 2006. This Bill gives the president of the United States unfettered power in dealing with anyone suspected of being a threat to the State, as well as the authorisation to interpret and apply the Geneva Conventions according to his sole discretion.

  • Description: It is becoming increasingly apparent that heat shock proteins play an important role in the survival of Plasmodium falciparum against temperature changes associated with its passage from the cold-blooded mosquito vector to the warm-blooded human host. Interest in understanding the possible role of P. falciparum Hsp70s in the life cycle of the parasite has led to the identification of six HSP70 genes. Although most research attention has focused primarily on one of the cytosolic Hsp70s (PfHsp70-1) and its endoplasmic reticulum homolog (PfHsp70-2), further functional insights could be inferred from the structural motifs exhibited by the rest of the Hsp70 family members of P. falciparum. There is increasing evidence that suggests that PfHsp70-1 could play an important role in the life cycle of P. falciparum both as a chaperone and immunogen. In addition, P. falciparum Hsp70s and Hsp40 partners are implicated in the intracellular and extracellular trafficking of proteins. This review summarizes data emerging from studies on the chaperone role of P. falciparum Hsp70s, taking advantage of inferences gleaned from their structures and information on their cellular localization. The possible associations between P. falciparum Hsp70s with their cochaperone partners as well as other chaperones and proteins are discussed.

Peripheral and non-peripheral 3-hydroxypyridine tetrasubstituted gallium(III)phthalocyanines (5a, 6a) and their quaternarized derivatives (5b and 6b) have been synthesized and characterized by elemental analysis, IR, 1H NMR spectroscopy, electronic spectroscopy and mass spectra. The quaternarized compounds (5b and 6b) are soluble in water and not aggregated (in water and in organic solvents) within a wide concentration range. General trends are described for singlet oxygen quantum yields, fluorescence quantum yields and fluorescence lifetimes of these compounds. These complexes showed better singlet oxygen quantum yields in water than the related quarternarized porphyrazine complexes.

  • Description: Many research papers have documented the positive relationship between the slope of the yield curve and future real economic activity in different countries and different time periods. One explanation of this link is based on monetary policy. The forecasting ability of the term spread on economic growth is based on the fact that interest rates reflect the expectations of investors about the future economic situation when deciding about their plans for consumption and investment. This thesis examined the predictive ability of the term structure of interest rates on economic activity, and the effects of different monetary policy regimes on the predictive ability of the term spread. The South African experience offers a unique opportunity to examine this issue, as the country has experienced numerous monetary policy frameworks since the 1970s. The study employed the Generalised Method Moments technique, since it is considered to be more efficient than Ordinary Least Squares. Results presented in this thesis established that the term structure successfully predicted real economic activity during the entire research period with the exception of the last sub-period (2000-2004) when using the multivariate model. In the periods of financial market liberalisation and interest rates deregulation the term structure was found to be a better predictor of economic activity in South Africa. These findings emphasise the importance of considering the prevailing economic environment in testing the term structure theory.

  • Description: This haunting photograph (Fig. 1) from the 1976 Soweto Uprisings in South Africa is often referred to as the single most important photograph to emerge from the struggle against apartheid (Purtilo 1999:22). According to South African film director Feizel Mamdoo, there are particular moments in history that are defined by photographic, celluloid, or television images, such as the world famous photograph of the Saigon girl, naked and burning from napalm (Worsdale 1998; see also Richards 2001). He argues that the iconic photograph by Sam Nzima depicting Hector Pieterson being carried in the arms of Mbuyisa Makhubu, with his distraught sister Antionette1 running alongside, is comparable in the way that “it marks history, both social and personal” (Worsdale 1998).

  • Description: The study investigates how rural Grade 4 teachers and learners experience the transition from Oshikwanyama to English as a medium of instruction. The study was conducted at a rural school in Oshana region in northern Namibia. The research site and participants were purposefully selected. Grade 4 teachers and learners were interviewed and observed. Relevant documents were also analyzed. The purpose was to understand participants’ experiences, perceptions and practices in relation to their experience of the transition to English medium instruction. The study revealed out that both Grade 4 teachers and learners are struggling to teach and learn through the medium of English. This was borne out by participants’ responses, classroom practices and document analysis, especially learners’ oral presentation and written work. The conclusion is drawn that limitations in English proficiency overshadow the good intentions of the curriculum. This was evident in the fact that Grade 4 learners at a selected school found it difficult to cope with the demands of the curriculum. In addition, most of the learners have poor literacy skills even in their mother tongue, Oshikwanyama. In the linguistic sense, proficiency in the first language can pave the way for second language to flourish. If the reverse is the case then the learner might find it difficult to acquire linguistic skills in the second language. The study concludes that for the sake of effective learning, an intervention on professional development needs to be in place. This might help rural lower primary teachers to upgrade their existing skills particularly in terms of the English language proficiency as well as how to prepare learners for the transition to English.

The trouble with culture : Plato's critique of poetry

  • Description: In this thesis I argue that Plato's critique of poetry, taken in its proper context, is a serious and relevant critique of popular culture. In the first chapter I argue that the poetic reforms proposed in Books 2 and 3 and Book 10 of Republic stand at the front of a total reform of Greek culture. I argue for the consistency of Plato's whole argument and then I claim that if we get Plato's targets right, not fine art or literature, and focus on appropriate modern analogues then we can see why his critique is still important. If we share his claim that we are influenced by popular culture in important and often insidious ways and agree that culture can promote corrupt values, then we have accepted the core of Plato's challenge. If we find his solution distasteful, then the task is to come up with a democratic alternative. In the remaining two chapters I focus specifically on the challenge to the poets, putting the other reforms to the side. In the second chapter I consider a possible reply to the challenge focusing on the worth of the poetry that was expelled. I first look elsewhere in the Platonic oeuvre at the account of beauty in Symposium and Phaedrus but I argue that neither of them gives anything like aesthetic value that could be usefully applied to poetry. Next I look to some modern accounts of aesthetic value. I argue that while they might go some of the distance against Plato's challenge, they face a difficult task because it is not sufficient positing the value, an account is needed of their positive benefit. In the third chapter I turn to a more direct response to the challenge. Arguably Aristotle offers such a response in the Poetics, in terms of the notion of katharsis. I consider two interpretative candidates for katharsis. The first takes the benefit of poetry to be psychological - katharsis is a purgation of otherwise pathological emotions. I argue that this fails because it misunderstands precisely what Plato's concerns about poetry are, and, furthermore, this account could even be compatible with Plato's worries. The second interpretation takes the benefit of poetry to be ethical- katharsis is a type of ethical clarification which is beneficial in training our emotional responses.   • Description: Geographic Information System technology today allows for the rapid analysis of vast amounts of spatial and non-spatial data. The power of a GIS can only be effected with the rapid collection of accurate input data. This is particularly true in the case of the South African National Working for Water (WFW) Programme where large volumes of spatial data on alien vegetation infestations are captured throughout the country. Alien vegetation clearing contracts cannot be generated, for WFW, without this data, so that the accurate capture of such data is crucial to the success of the programme. Mapping Invasive Alien Plant (IAP) data within WFW is a perennial problem (Coetzee, pers com, 2002), because not enough mapping is being done to meet the annual requirements of the programme in the various provinces. This is re-iterated by Richardson, 2004, who states that there is a shortage of accurate data on IAP abundance in South Africa. Therefore there is a need to investigate alternate methods of data capture; such as remote sensing, whilst working within the existing WFW data capture standards. The aim of this research was to investigate the use of Landsat ETM imagery as a data capture source for mapping alien vegetation for the WFW Programme in terms of their approved mapping methods, for both automated and manual classification techniques. The automated and manual classification results were compared to control data captured by differential Global Positioning Systems (DGPS). The research tested the various methods of data capture using Landsat ETM images over a range of study sites of varying complexity: a simple grassland area, a medium complexity grassy fynbos site and a complicated indigenous forest site. An important component of the research was to develop a mapping (classification) Ranking System based upon variables identified by WFW as fundamental in data capture decision making: spatial and positional accuracy, time constraints and cost constraints for three typical alien invaded areas. The mapping Ranking System compared the results of the various mapping methods for each factor for the study sites against each other. This provided an indication of which mapping method is the most efficient or suitable for a particular area.

The use of thermal modelling in improving rural subsistence aquaculture

  • Description: There has been a long history of attempts to promote subsistence rural aquaculture within South Africa. Many of these attempts have failed due to inadequate support from government and support agencies. There has been a recent revived interest in promoting rural aquaculture development. Presented in this project are six scenarios demonstrating the potential application of thermal modelling techniques to improving subsistence rural aquaculture. These scenarios were based on a model validated against a real life system, and run with environmental data for the year 2005. Bovine manure, grass thatch, cereal straw and Typha spp. water reeds were tested for thermal performance. These results were then applied to the scenarios. The open and kraal enclosed pond scenarios yielded the widest deviation in temperatures throughout the year, with an average temperature of 3.8oC and 6.0oC above ambient and a temperature range of 10.6oC and 12.4oC for the year respectively. Eliminating solar radiation and reducing wind speed resulted in an average difference of 1.5oC below ambient and a temperature range of 7.5oC for the year. When completely enclosed in a well-sealed traditional Xhosa style hut, the pond had an average difference of 5.6oC below outside ambient temperature, with a range of 7.8oC for the year. A passive solar heating scenario added to the sealed hut scenario, which when run continuously maintained an average difference of 34.7oC above outside ambient temperatures, with a range of 56oC for the year. When a hypothetical 25oC temperature control was included in the model, the system maintained an average of 7.1oC above ambient outside temperatures, with a temperature range of 0.8oC throughout the year. The wider range of temperatures made available through the use of thermal modelling approaches provides a more diverse range of species available for subsistence rural aquaculture at any site, significantly improving its potential.
