  • Description: publisher version , Passing the introductory accounting semester is often seen as a challenge for first year students. Being aware of both effective and ineffective ways of constructing knowledge in a discipline with a hierarchical knowledge structure will be of value to students and teachers alike in assisting in the development of effective styles of learning. This article, which is part of a larger body of research, analyses how students in an introductory financial accounting class at Rhodes University constructed knowledge. Previous research described the course as having a hierarchical knowledge structure. In this research, first year accounting students at Rhodes University were interviewed to gain an improved understanding of how they constructed knowledge in this course. This article describes how students who were successful in passing this semester course used similar, effective ways of constructing knowledge, while students who were not successful also employed similar but less effective ways of constructing knowledge. These different ways of constructing knowledge, both effective and ineffective, were analysed, using the Bernstein’s pedagogic device and Maton’s Legitimation Code Theory. This article provides those involved in teaching and learning in a discipline with a hierarchical knowledge structure, with a theoretical explanation of why some methods of constructing knowledge are more effective than others. Understanding and being explicit about more (and less) effective ways of constructing knowledge in a course with a hierarchical knowledge structure can guide those involved in teaching and learning to improve results.

An analysis of the fruit-sucking and fruit-piercing moth complex in citrus orchards in South Africa

  • Description: Fruit-piercing moths are a sporadic pest of citrus, especially in the Eastern Cape Province of South Africa, where the adults can cause significant damage in outbreak years. However, growers confuse fruit-piercing moths with fruit-sucking moths that do not cause primary damage. In this study we trapped these moths during the 2013–2015 growing seasons. A large number of diverse fruit-feeding moths were collected through weekly sampling in citrus orchards in the Eastern Cape and northern Limpopo provinces. Twenty-three species of fruit-feeding moth were trapped. However, only two were fruit-piercing species, capable of causing primary damage, namely Serrodes partita (Fabricius) (Erebidae) and Eudocima divitiosa (Walker) (Erebidae). Surprisingly S. partita, which has been reported as the main fruit-piercing moth pest of citrus in South Africa, comprised only 6.9 % of trap catches. The categorisation of moths as fruit-piercing or fruit-sucking (causing secondary damage) was confirmed by examining the morphological structures (tearing hooks and erectile barbs) of these moths’ proboscides. This study has shown that in non-outbreak seasons, S. partita comprised only a small percentage of fruit-feeding moths in citrus orchards. However, growers may misidentify the harmless fruit-sucking species as fruit-piercing species, and thus overestimate the density of fruit-piercing moths.

  • Description: The impact of formulation and process variables on the in-vitro release of prednisone from a multiple-unit pellet system was investigated. Box-Behnken Response Surface Methodology (RSM) was used to generate multivariate experiments. The extrusion-spheronization method was used to produce pellets and dissolution studies were performed using United States Pharmacopoeia (USP) Apparatus 2 as described in USP XXIV. Analysis of dissolution test samples was performed using a reversed-phase high-performance liquid chromatography (RP-HPLC) method. Four formulation and process variables viz., microcrystalline cellulose concentration, sodium starch glycolate concentration, spheronization time and extrusion speed were investigated and drug release, aspect ratio and yield were monitored for the trained artificial neural networks (ANN). To achieve accurate prediction, data generated from experimentation were used to train a multi-layer perceptron (MLP) using back propagation (BP) and the Broyden-Fletcher-Goldfarb-Shanno (BFGS) 57 training algorithm until a satisfactory value of root mean square error (RMSE) was observed. The study revealed that the in-vitro release profile of prednisone was significantly impacted by microcrystalline cellulose concentration and sodium starch glycolate concentration. Increasing microcrystalline cellulose concentration retarded dissolution rate whereas increasing sodium starch glycolate concentration improved dissolution rate. Spheronization time and extrusion speed had minimal impact on prednisone release but had a significant impact on extrudate and pellet quality. This work demonstrated that RSM can be successfully used concurrently with ANN for dosage form manufacture to permit the exploration of experimental regions that are omitted when using RSM alone.

  • Description: The quantification of the essential amino acid requirements of a species is a prerequisite to the formulation of biologically optimized diets. In this study, crystalline L-lysine was used in an attempt to determine the quantitative lysine requirement of juvenile Haliotis midae. Two microencapsulation techniques [gelatine/acacia and cellulose acetate phthalate (CAP)] were used to retard leaching of crystalline L-lysine incorporated into semipurified test diets. An assessment of the efficacy of the encapsulation techniques, revealed that despite effective lysine supplementation, H. midae fed semipurified test diets containing encapsulated crystalline L-lysine failed to promote significant improvements in either growth, feed or protein efficiency (P > 0.05). The failure of the crystalline L-lysine to illicit growth and nutritional responses is discussed.

  • Description: Natural resource management (NRM) education has the potential to improve the quality and relevance of rural education in South Africa. For this potential to be realised, the various educational resources that are commonly used by teachers and learners in rural schools need to incorporate natural resource management knowledge. Using Bernstein’s concepts of classification and his theories on recontextualisation, a content analysis study was carried out to compare the level of NRM integration within the Grade 10 Life Sciences syllabus, and a Grade 10 Life Sciences textbook. Results from the analysis of the syllabus showed that overall only 9% of all the knowledge statements analysed had a strong link to NRM and related issues. The highest percentage of such sentences was found in the Core Knowledge section of the syllabus (21%). For the textbook, only 8% of the analysed items had a strong link to NRM and related issues, with the highest percentage of such items occurring in the Suggested Activities section (16%). However, the level of NRM integration in both documents increased considerably when sentences that had only an implicit link to NRM and related issues were included. It was concluded that both documents provide ample opportunities for NRM learning, although the extent to which this occurs varies among their different sections. The recontextualising role of the Grade 10 Life Sciences textbook was reflected in its relatively higher level of NRM integration in the Suggested Activities category, and in the Glossary category. This study highlights the need for further strengthening of the position of NRM within the Grade 10 Life Sciences syllabus, and for more Bernstein-based research to inform South Africa’s curriculum reform initiatives in environmental education.

  • Description: A systems approach was previously developed for mitigating phytosanitary risk of Thaumatotibia leucotreta (Meyrick) in citrus fruit exported from South Africa, as an alternative to a standalone cold disinfestation treatment. The present study first tested the original systems approach by applying it on a semicommercial scale in 10 Nova mandarin orchards. Fruit were inspected at points in the production, packing, and simulated shipping process, to assess performance of the systems approach. Additional data were obtained from 17 Valencia orange orchards and six packinghouses. In the second part of this study, the systems approach was accordingly revised and improved, consisting of three measures: 1) preharvest controls and measurements and postpicking sampling, inspection, and packinghouse procedures; 2) postpacking sampling and inspection; and 3) shipping conditions. The model quantifying the effectiveness of the systems approach was improved by correcting errors in the original version, updating parameter values and adding a component that provides for comparison with the risk mitigation provided by a standalone disinfestation treatment. Consequently, the maximum potential proportion of fruit that may be infested with live T. leucotreta after application of the improved systems approach is no greater than the proportion of fruit that may be infested after application of a Probit 9 efficacy postharvest disinfestation treatment to fruit with a 2% pretreatment infestation. The probability of a mating pair surviving is also determined. The model enables a priori determination of the required threshold levels for any of the three measures, based on quantification of the other two measures.

  • Description: Coastal communities are some of the most at-risk populations with respect to climate change impacts. It is therefore important to determine the vulnerability of such communities to co-develop viable adaptation options. Global efforts to address this issue include international scientific projects, such as Global Learning for Local Solutions (GULLS), which focuses on five fast warming regions of the southern hemisphere and aims to provide an understanding of the local scale processes influencing community vulnerability that can then be up-scaled to regional, country and global levels. This paper describes the development of a new social and ecological vulnerability framework which integrates exposure, sensitivity and adaptive capacity with the social livelihoods and food security approaches. It also measures community flexibility to understand better the adaptive capacity of different levels of community organization. The translation of the conceptual framework to an implementable method is described and its application in a number of “hotspot” countries, where ocean waters are warming faster than the rest of the world, is presented. Opportunities for cross-cultural comparisons to uncover similarities and differences in vulnerability and adaptation patterns among the study’s coastal communities, which can provide accelerated learning mechanisms to other coastal regions, are highlighted. The social and ecological framework and the associated survey approach allow for future integration of local-level vulnerability data with ecological and oceanographic models.

  • Description: This special issue is the fourth decadal review of biological control of invasive alien plants (biocontrol of weeds) in South Africa, following those published in 1991, 1999 and 2011. Including this introduction, there are 24 papers covering the weed biocontrol programmes, or important developments in the science and practice, from the period 2011-2020. Seventy-two target weed species are covered, including 25 species on which projects were initiated during the past decade. Developments in regulations, mass-rearing and implementation, and community engagement are also reviewed. An updated catalogue of agents released, rejected and under consideration is presented and reflects the most recent methods of quantifying success in weed biocontrol. Key events over the last decade include the hosting of the XIV International Symposium on Biological Control of Weeds to celebrate 100 years of weed biocontrol in South Africa, as well as the establishment of the Centre for Biological Control at Rhodes University. The science and practice of weed biocontrol has expanded significantly in the past decade, with growth in the number of researchers and practitioners, increased funding, and an increased number of scholarly outputs. Unlike many other countries in the world, South Africa has largely avoided constraints due to restrictive and risk averse legislation and bureaucracy, and has continued to release new biocontrol agents at a similar rate to that in previous years. Much of the success of weed biocontrol in South Africa is due to the sustained and increasing support of the Natural Resource Management Programme of the Department of Forestry, Fisheries and the Environment (Working for Water Programme). However, gaps in funding, where no funds are available for months at a time, are a major concern as the weed biocontrol community loses human capital in these periods, and research programmes suffer significant set-backs. Weed biocontrol is an essential component of South Africa’s strategy to reduce the negative impacts of invasive alien plants and has contributed significantly towards the protection of the country’s ecosystems, indigenous biodiversity, water security, agricultural productivity, and society in general. If the trend of increasing support for weed biocontrol in South Africa continues, we can expect that the benefits for the country at large will increase substantially in the future.

An inventory of medicinal plants traded on the western boundary of the Kruger National Park, South Africa

  • Description: The World Health Organisation estimates that traditional medicine still plays a vital role in the lives of 70-80 of the populations of developing countries. Approximately 20 000 tons of medicinal plants are estimated to be traded in South Africa annually, resulting in considerable pressure on the wild populations from which these plants are harvested. In 1997, a study was initiated to assess the extent of trade in medicinal plants on the western boundary of the Kruger National Park, and to determine local perceptions of the availability and values of these plants. This paper presents an inventory of the species traded, including vendors' and traders' perceptions of current availability of species and consumer demand, as well as price/kg. In Mpumalanga, 176 species were identified (71 of the vernacular names encountered in the market place), representing 69 families. In Northern Province, 70 different species were identified (84 of the vernacular names recorded in the markets), representing 40 families. Perceptions of availability of a species varied considerably, often within the same markets. Perceptions of demand tended to be more consistent, although these also sometimes varied. Although monitoring markets is an efficient means of assessing the conservation status of medicinal plants, it is important to select appropriate parameters. Markets often differ from one another, and an understanding of local conditions is crucial. For example, in the medicinal markets on the western boundary of the Kruger National Park, price/kg and market perceptions of availability of species were highly variable and would thus not provide a consistent monitoring parameter. Perceptions of demand provide an indication of current and potential pressure on a species, particularly when combined with the monitoring of species traded, and the introduction of substitutes in the market place. The size of plant parts traded could also provide useful monitoring data. It is important to identify the plants utilised locally first hand where possible, due to the variation of vernacular names from one area to another.

  • Description: There is increasing evidence suggesting a protective role for anti-inflammatory medications in neurological disorders such as Alzheimer's disease (AD). While there has not been any direct evidence for this, a number of clinical studies indicate that those patients who have had a history of nonsteroidal anti-inflammatory use, have a lower incidence of AD. Since there is currently no evidence on the mechanism by which these agents offer possible neuroprotection, we investigated the potential neuroprotective properties of the nonsteroidal anti-inflammatory drug, ibuprofen, by examining whether this agent could reduce lipid peroxidation and superoxide radical generation. Quinolinic acid and cyanide, known neurotoxins, were used to induce lipid peroxidation and superoxide anion formation respectively, in rat brain homogenate. The results show that ibuprofen significantly (p more than 0.05) reduced quinolinic acid-induced lipid peroxidation and cyanide-induced superoxide production. The results of the present report therefore suggest a possible mechanism for the neuroprotective effect of ibuprofen.

An investigation into the time-saving benefits of using a computerised taxation program

  • Description: Software programs designed to calculate the tax liability of individuals and other types of tax entity are being designed and used with increasing frequency. Research done in the United States of America appears to indicate that these programs do not achieve any saving in time and, in fact, may take longer to use than to complete a tax return by hand. As the South African revenue collection system appears to be moving closer to a self-assessment system, where the calculation of tax owing will be the responsibility of the taxpayer, the research essay sets out to determine whether there is a saving in time when calculating a tax liability using a tax software program, instead of calculating it by hand. In addition, the research aims to determine how much time, on average, is saved or lost, using such a program, and whether there is any correlation between the time taken to perform a calculation by hand and that using the software program. It does so by comparing the average estimated time it would take to complete tax calculations for individuals by hand with the average time taken to complete the same tax calculations using a tax software program. The average time taken to do the calculations by hand is based on the time allocation given for questions by the authors of a published question bank for university students. The time taken using software is determined by using a stop-watch to time each question being processed. The results, subject to assumptions made in carrying out the research, show that there is a substantial saving in time using the software program. Based on the data, however, the results indicate a weak correlation between the estimated time taken to do a calculation by hand and the estimated time using the software program. Possible reasons for the weak correlation are discussed. A recommendation is also made for the standardization and certification of existing tax calculation software.

The octabrominated dye has a relatively high singlet oxygen quantum yield of 0.78 in DMF and exhibits favorable photodynamic activity against MCF-7 cells with an IC50 value of 10.7 μM and a 5.74 log reduction value (5 μM) towards S. aureus under illumination at 660 nm for 60 min with a Thorlabs M660L3 LED (280 mW cm−2).

  • Description: Paracetamol (PA), being an analgesic and antipyretic medicine, can cause fatal hepatotoxicity and nephrotoxicity when overdosed. It is therefore important to develop electrochemical sensors that can monitor and quantify it in aquatic environments. In this study, rare-earth sandwich-type phthalocyaninato chelates based on neodymium (1 a) and samarium (1 b) were employed as electrocatalysts to modify glassy carbon electrodes (GCE) for the first time. It was found that 1 a-modified GCE (herein referred to as 1 a-GCE) is less conductive than 1 b-modified counterpart (1 b-GCE). A larger rate constant was also obtained for 1 b-GCE. It was established that a faster oxidation rate efficiency was responsible for lower limit of detection value obtained for 1 b-GCE as compared to 1 a-GCE.

  • Description: Purpose: To isolate the anti-cancer and anti-trypanosomal principles of Zanthoxylum leprieurii, a medicinally versatile wild tropical plant used for managing tumours, African trypanosomiasis, and inflammation in southeastern Nigeria. Methods: The pure compounds were isolated using chromatographic methods. The structural elucidation of the pure compounds was based on their NMR (1D and 2D) and mass spectral data as well as chemical test results. Structure-activity relationships were based on the structural differences among the compounds. The cytotoxicity of the extracts and compounds (1, 2, 3, and 4) was evaluated in HeLa (human cervix adenocarcinoma) cell line while the trypanocidal activities were evaluated on Trypanosoma brucei brucei. Results: Two acridone alkaloids, 1-hydroxy-3-methoxy-10-methylacridin-9 (10H)-one, named fabiocinine (1), and 1-hydroxy-2,3-dimethoxy-10-methylacridin-9 (10H)-one (arborinine, 2), together with a furoquinoline alkaloid, skimmianine (3), and a chelerythrine derivative, 6-acetonyl-5,6-dihydrochelerythrine (4) were isolated from the root bark of Zanthoxylum leprieurii. Skimmianine (3) exhibited cytotoxicity and anti-trypanosomal IC50 of 12.8 and 13.2 µg/mL respectively (p less than 0.05). Compound (1) and arborinine (2) were selectively cytotoxic to HeLa cells with cytotoxicity IC50 of 28.49 and 62.71 µg/mL, respectively, while (4) did not show significant activity (p less than 0.05). Conclusion: Zanthoxylum leprieurii root bark contains cytotoxic and trypanocidal compounds, and is thus a potential source of anti-cancer and anti-trypanosomal leads.

Arg. are used as traditional medicine in many African countries for the management ofgastrointestinal, respiratory and urinary tract infections as well as for the treatment of wounds.Aim of the study:To determine the in vitro antibacterial activity of the crude extracts of leaves and stemsofA. cordifoliaon gastrointestinal, skin, respiratory and urinary tract pathogens and to identify thecompounds in the extracts that may be responsible for this activity.Materials and methods:The antibacterial activities of crude extracts [hexane, chloroform (CHCl3), ethylacetate (EtOAc), ethanol (EtOH), methanol (MeOH) and water (H2O)] as well as pure compounds isolatedfrom these extracts were evaluated by means of the micro-dilution assay against four Gram-positivebacteria, i.e.Bacillus cereusATCC 11778,Enterococcus faecalisATCC 29212, Staphylococcus aureusATCC25923 andS. saprophyticusATCC 15305,as well as four Gram-negative bacterial strains, i.e.EscherichiacoliATCC 25922, Klebsiella pneumoniaeATCC 13883, Moraxella catarrhalisATCC 23246 andProteus mir-abilisATCC 43071. The isolation of the active constituents was undertaken by bio-autographic assays inconjunction with chromatographic techniques. The identification and characterisation of the isolatedcompounds were done using mass spectrometry (MS) and Fourier transformed infrared spectrometry(FTIR) as well as 1D- and 2D- nuclear magnetic resonance (NMR) analyses.Results:The leaves and stems ofA. cordifoliaexhibited varied antibacterial activity against all eight pa-thogens. Most of the MIC values ranged between 63 and 2000mg/ml. The highest activities for the crudeextracts (63mg/ml) were observed againstS. saprophyticus[stem (EtOAc, CHCl3and hexane), leaves(MeOH, EtOH, EtOAc and CHCl3)],E. coli[stem (MeOH and EtOH), leaves (MeOH, EtOH, EtOAc andCHCl3)],M. catarrhalis[leaves (EtOAc and CHCl3)],K. pneumoniae[stem (CHCl3), leaves (CHCl3)] andS.aureus[leaves (CHCl3)]. Seven constituents [stigmasterol (1), stigmasta-4,22-dien-3-one (2), friedelin (3),friedelane-3-one-28-al (4), 3-O-acetyl-aleuritolic acid (5), 3-O-acetyl-erythrodiol (6) and methyl-3,4,5-trihydroxybenzoate (methyl gallate) (7)] were isolated from the stem MeOH extract. All these com-pounds displayed some antibacterial activity against the eight pathogens with highest activity againstS.saprophyticus(2mg/ml). Furthermore, this is thefirst report of compounds1,2,3,4,6and7isolated fromA. cordifoliaand where a complete set of 2D-NMR data for fridelane-3-one-28-al (4) is presented.Conclusion:The study demonstrated that the antibacterial activities ofA. cordifoliaextracts may be dueto the presence of the seven isolated compounds, where compounds3–6showed the best activity. Theobserved activity against gastrointestinal, skin, respiratory and urinary tract pathogens supports thetraditional use for the treatment of such ailments.

  • Description: The MeOH/CH2Cl2 (1:1) extracts of the roots and leaves of Beilschmiedia louisii and B. obscura showed potent antitrypanosomal activity during preliminary screening on Trypanosoma brucei brucei. Phytochemical investigation of these extracts led to the isolation of a mixture of two new endiandric acid derivatives beilschmiedol B (1) and beilschmiedol C (2), and one new phenylalkene obscurene A (3) together with twelve known compounds (4–15). In addition, four new derivatives (11a–11d) were synthesized from compound 11. Their structures were elucidated based on their NMR and MS data. Compounds 5, 6, and 7 were isolated for the first time from the Beilschmiedia genus. Additionally, the NMR data of compound 4 are given here for the first time. The isolates were evaluated for their antitrypanosomal and antimalarial activities against Tb brucei and the Plasmodium falciparum chloroquine-resistant strain Pf3D7 in vitro, respectively. From the tested compounds, the mixture of new compounds 1 and 2 exhibited the most potent antitrypanosomal activity in vitro with IC50 value of 4.91 μM.

  • Description: Objectives: Several mushrooms species have been reported to be nematophagous and antiprotozoan. This study reported the antiplasmodial and cytotoxic properties of the n-hexane extract from the edible mushroom Pleurotus ostreatus and the isolation of a sterol from the extract. Materials and Methods: Antiplasmodial and cytotoxicity assays were done in vitro using the plasmodium lactate dehydrogenase assay and human HeLa cervical cell lines, respectively. The structure of the isolated compound from the n-hexane extract was elucidated using spectroscopic techniques. Results: The n-hexane extract (yield: 0.93% w/w) showed dose dependent antiplasmodial activity with the trend in parasite inhibition of: chloroquine (IC50=0.016 μg/mL) > n-hexane extract (IC50=25.18 μg/mL). It also showed mild cytotoxicity (IC50>100 μg/mL; selectivity index >4) compared to the reference drug emetine (IC50=0.013 μg/mL). The known sterol, ergostan-5,7,22-trien-3-ol, was isolated and characterized from the extract. Conclusion: This study reporting for the first time the antiplasmodial activity of P. ostreatus revealed its nutraceutical potential in the management of malaria.

  • Description: Melamine formaldehyde and melamine resorcinol formaldehyde microspheres were decorated with Au and Pd nanoparticles and applied as heterogeneous catalysts in the oxidation of 1-phenylethanol. The catalysts showed similar activities irrespective of the support employed. Moderate conversion activities of 48–50% were achieved when using acetonitrile as solvent; however, when employing water as solvent, the supported catalysts formed a three-phase, emulsion system which facilitated the catalytic conversion of 1-phenylethanol to acetophenone at much higher conversions of around 83%. The oxidant, TBHP, decomposed rapidly in acetonitrile, whilst it remained stable in aqueous solution, leading to the enhanced activities observed when using water as solvent. These systems also proved to be recyclable for up to five cycles, with only slight loss of activity observed; this can be attributed to the physical loss of catalyst during the workup procedure conducted between each cycle.

  • Description: Graphene quantum dots (GQDs) and zinc phthalocyanine (ZnPc) were separately modified with thymine to obtain thymine-functionalized GQDs (T-GQDs) and ZnPc (T-ZnPc). T-GQDs and nanoconjugates of T-ZnPc with pristine GQDs (represented as pristine GQDs–T-ZnPc) or T-GQDs (represented as T-GQDs–T-ZnPc) were employed as fluorescent probes for the detection of mercury(II) ions (Hg2+). The as-synthesized T-GQDs alone demonstrated a highly sensitive and selective fluorescence “turn-OFF” process for Hg2+ detection due to the specific interaction between the thymine functionality on the T-GQDs with Hg2+. On the other hand, the fluorescence of pristine GQDs and T-GQDs was quenched (“turn-OFF”) upon coordination with T-ZnPc. However, the fluorescence emission was selectively restored (“turn-ON” process) in the presence of Hg2+ resulting in the sensitive detection of Hg2+ in the nanomolar concentration range (limit of detection = 0.05 nM, for the pristine GQDs–T-ZnPc probe). The probe containing pristine GQDs and the T-ZnPc complex demonstrated a higher specific and sensitive recognition of Hg2+ as compared to the T-GQDs alone or T-GQDs–T-ZnPc probes which are ascribed to the fluorescence “turn-ON” process of the former. Screening of different metal ions and counter ions proved that the probes are specifically suited for Hg2+ detection.

  • Description: Lead (II) pyridyloxyphthalocyanine (PbTpyPc) and its quaternized form (PbTepyPc) were synthesized and the photophysical behavior examined. Low fluorescence quantum yields (ΦF) of 0.01 and 0.02 were observed for PbTepyPc and PbTpyPc, respectively. The singlet oxygen quantum yields (ΦΔ) were 0.60 and 0.68, for PbTepyPc and PbTpyPc, respectively using DPBF as a quencher in DMF. Singlet oxygen production of the embedded sensitizers in electrospun fiber were quantified using ADMA and were found to be ΦΔ = 0.41 and ΦΔ = 0.21 for PbTepyPc and PbTpyPc, respectively. Photodynamic inactivation of Escherichia coli (E. coli) with the quaternized photosensitizer at 5 μM, totally inactivated the E. coli (with log CFU = 10 decrease). Only 0.4 log CFU decrease was obtained with PbTpyPc. The embedded non-quaternized photosensitizer (PbTpyPc) was less active on the gram negative bacteria but the quaternized photosensitizer (PbTepyPc) was effective towards inactivation of E. coli.
