The conjugates were employed as fluorescence-based sensors for anion sensing. Among the anions that enhanced the fluorescence of the probe, fluoride ion was chosen as the test ion to test the efficacy of the probe. Förster resonance energy transfer from the QDs to the phthalocyanine was observed as an indication for the fluorescence quenching of the QDs upon binding to the phthalocyanine. The fluorescence of the linked QDs was progressively enhanced, and linearly proportional to increasing concentrations of fluoride ion. The type of substituent attached to the phthalocyanine ring influenced the efficiency of fluorescence enhancement. The proposed nanoprobe has been employed to detect fluoride ion in cell culture medium and tap water.

The objectives of the study were to identify the types of records that are kept by SMMEs; evaluate the infrastructure for the SMMEs records management; determine the compliance of SMMEs with the existing RM legal and regulatory requirements; and identify the challenges related to SMMEs records practices. Twenty three (23) respondents from SMMEs were interviewed. Findings revealed all the respondents could not confirm adoption of any systemic way of records management to support business functions and operations. Majority of the SMMEs due to their inability to maintain records could not identify potential benefit of business RM with an exception of three SMMEs. The respondents were not aware of any compliance and regulatory framework most especially on business records management. The challenges facing SMMEs include; inability to capture/create maintain, preserve business records systematically in its lifecycle. The SMMEs lack needed training especially in records management to allow them the benefit of maintaining business records. It is recommended that SMMEs should acquire skill that would enable them manage their business records that may help improved decision making. There should be legislative and regulatory framework that ensures compliance. We assessed the direction and periodicity of experimental learning of people in the Western Solomon Islands after a tsunami in 2007. We compared the results of marine science surveys with local ecological knowledge of the benthos across 3 affected villages and 3 periods before and after the tsunami. We sought to determine how people recognize biophysical changes in the environment before and after catastrophic events such as earthquakes and tsunamis and whether people have the ability to detect ecological changes over short time scales or need longer time scales to recognize changes. Indigenous people were able to detect changes in the benthos over time. Detection levels differed between marine science surveys and local ecological knowledge sources over time, but overall patterns of statistically significant detection of change were evident for various habitats. Our findings have implications for marine conservation, coastal management policies, and disaster-relief efforts because when people are able to detect ecological changes, this, in turn, affects how they exploit and manage their marine resources.

In a world in which coral reefs are increasingly threatened (Hughes et al. 2003, McClanahan et al. 2008), understanding how people perceive and use coral reefs is essential for predicting future ecological and social impacts, as well as for understanding human adaptation mechanisms to ecological change in these tropical marine ecosystems. This is particularly true for Oceanic islands, which are vulnerable socially and ecologically to deteriorating coral reefs, rising sea levels, and increasingly unpredictable climatic and geological phenomena (Lazarus 2012, McClanahan and Cinner 2012). Increasing human vulnerability to changing coral reefs, consequently, has resulted in numerous calls for comprehensive management using tools that include fishing regulations and quotas, marine protected areas (MPAs), and ecosystem-based management (EBM) for protecting coral reefs and other marine ecosystems. A Note on the (Continued) Ability of the Yield Curve to Forecast Economic Downturns in South Africa

- Description: In 2002-2003, the South African yield spread falsely signalled a downswing that never materialised. This paper provides two reasons for this false signal. First, while the Reserve Bank never actually officially declared the start of a downswing, by alternative measures a downswing did actually occur. It is this severe weakness in economic activity at that time that the yield curve pointed to. Second, short-term interest rates in 2003 were higher than they should have been because of a mistake made in measuring consumer price inflation. Because South Africa had recently introduced an inflation-targeting regime, policy interest rates were, as a result of this error, kept too high for too long. This policy mistake was rectified as soon as the error in the Consumer Price Index was discovered. Thus, the yield curve in 2003 pointed to the reality that short-term interest rates were too high and risked pushing the economy into full blown recession. This is demonstrated by the fact that it was a fall in long bond interest rates that caused the yield spread to turn negative, indicating expectations that short-term interest rates would need to be cut – as indeed they were.

Despite the acknowledged importance of family, the association between family functioning and individual well-being outcomes have remained unexplored in the current body of knowledge. This paper explores the association between family functioning and reported levels of life satisfaction and happiness in South Africa. The paper employs the Family Attachment and Changeability Index (FACI8) to measure family functioning, using data from the 2011 South African Social Attitudes Survey. Four measures of family functioning are utilised, namely the aggregate FACI8 scale, the attachment and changeability sub-scales, and family functioning type. Improvements in the level of family functioning as well as in the levels of attachment and changeability are positively associated with life satisfaction and happiness. In addition, individuals living in midrange or balanced family functioning types are more satisfied with life and happier compared to persons living in extremely or moderately dysfunctional families. The findings highlight the importance of supportive intra-family dynamics in fostering greater individual well-being. This in turn places emphasis on the investigation of likely correlates of family functioning and impact evaluations of family-focused social work interventions’ impact on family functioning as areas for future research.

The World Food Programme (WFP) and the government in Zimbabwe are providing food aid to approximately 1.6 million people across the 37 districts in the country. Measures have been undertaken to counter food insecurity in Zimbabwe but to no avail. Food related public policies such as donor aid and government subsidies have also been unworkable for such a long time. In this study a comprehensive analysis of the role of decent rural employment in counteracting food insecurity will be provided. The study also seeks to assess how humanitarian work psychologists can work together with various stakeholders in society so as to curb household food insecurity. The Study population included all the households in Bindura rural district in Zimbabwe. Both quantitative and qualitative research designs were used. The researcher used cluster sampling procedure in which households were divided into groups (clusters) and a sample of the groups was selected. A random sample was then drawn from the elements within each selected group. The results confirmed that those that are not decently employed face a broad range of decent work deficits, exploitation at work and are food insecure, a finding that is similar to other countries. Thus, the researcher concluded that Decent Rural employment do have an impact on food security. Taking a look at the basic tenets of decent employment such as full time employment, a decent income, reasonable working hours and employee benefits among others, the results showed that they all impact on food security. The information obtained will be fundamental for communities at risk and dealing with the issue may help improve food insecurity situation in the country. This information will also help in formulating special measures in the form of tailor made strategies specifically made for the district and helps in doing away with what is called a one size fits all approach to food security as it pays particular attention to the households in that particular district. , Thesis (MA) -- Faculty of Management and Commerce, 2014

- Description: The Hsp70/Hsp90-organising protein (Hop) is a 60 kDa co-chaperone that acts as an adaptor molecule, facilitating the transfer of client proteins between the Hsp70 and Hsp90 chaperone systems. Hop functions both intracellularly and extracellularly and has been implicated in many processes involved in cancer progression, including cell migration and invasion. Little is known about the mechanisms or domains by which extracellular Hop functions. In addition, little is known about the effects of Hop on signalling molecules involved in cell migration and invasion through regulation of actin dynamics. It was hypothesised that both extracellular and intracellular pools of Hop would regulate distinct cell migration processes by activation of cell signalling pathways or direct interactions with signalling intermediates. HS578T cells were treated with recombinant full length and truncated murine Hop proteins (overexpressed and purified in this study) to determine the effects of extracellular Hop and the independent domains on cell migration processes. Additionally, RNA interference (RNAi) techniques were used to determine the effect of Hop knockdown on cell migration related signalling intermediates and cell morphologies. A short hairpin RNA (shRNA) system for the stable knockdown of Hop was developed and used for a number of these studies. Treatment of HS578T cells with the TPR2A2B and TPR1 domains of Hop resulted in a significant decrease in cell migration and caused changes in the actin cytoskeleton and extracellular matrix proteins, gelatin and fibronectin. RhoC immunoprecipitated in a common complex with Hop and Hsp90. Hop knockdown reduced levels of actin and total RhoC, as well as active RhoC. In addition, knockdown of Hop resulted in a reduced migratory phenotype. We interpreted these data to indicate that intracellular Hop played a role in cell migration through regulation of RhoC activity, either through a direct interaction between Hop and RhoC, or an indirect interaction of RhoC with the Hsp90 multichaperone heterocomplex. Taken together, the data suggested that extracellular and intracellular Hop played distinct roles in extracellular and intracellular processes that lead to actin dynamics and cell migration. We draw on four examples of social learning research in the environmental and sustainability sciences from sub-Saharan Africa to reflect on possible reasons behind the preponderance of case study research in this field, and to identify common elements that may be significant for social learning research more generally. We find that a common interest in change oriented social learning, and therefore processes of change, makes case studies a necessary approach because long term process analyses are required that are sensitive to social-ecological contexts. Common elements of the examples reflected upon included: a focus on initiating, tracking and/or understanding a process of change toward sustainability; long term research; an action research agenda that involves reflecting on data with research participants; and temporal, process based analysis of data coupled with in-depth theoretical analysis. This paper highlights that there is significant scope for exploratory research that compares case studies of social learning research to generate a deeper understanding of social learning processes, and their relationship to human agency and societal change.

In recent years the problem of poor solubility of paracetamol in water has been overcome and an intravenous formulation of paracetamol developed. In South Africa this is marketed by Bristol-Myers Squibb as Perfalgan®. Each 500 mg or 1g ampoule of Perfalgan® is recommended for single use only and is to be discarded once opened (Medicines.org, n.d.). This is most likely due to concerns about degradation of paracetamol or other ingredients in the solution once exposed to air and sterility issues. However in South Africa where this drug is expensive, some centers use one ampoule for multiple cases over the course of 12-24 hours. No obvious clinical adverse effects have been reported. 5.2 Objective of study The aim of this study was to examine the practice of using a single vial of Perfalgan® in divided doses over the course of a day for different patients by way of assessing the stability of Perfalgan® on exposure to air. The study has three components, namely: 1. In vitro assessment of paracetamol levels on exposure to air and stressors over time. 2. In vitro assessment of levels of the excipients, namely mannitol and cysteine on exposure to air and stressors over time. 3. Assessment of the lipid permeability of paracetamol in Perfalgan® on exposure to air and stressors over time. C Curran 2014 Degradation of paracetamol and other constituents in Perfalgan® 8 5.3 Methodology High performance liquid chromatography (HPCL-UV) was used to determine the concentration of paracetamol and the presence of degradation products in samples taken at set time periods following exposure of Perfalgan® to air and stressors. Initial work was done using pure paracetamol to determine optimal measurement conditions prior to analysing Perfalgan®. The concentrations of the additives and their degradation products were measured using nuclear magnetic resonance (1 H NMR). Appreciable changes in these could indicate a safety hazard or decreased bioavailability. Finally ultraviolet spectroscopy was performed to assess samples at a wider spectrum of wavelengths, instead of the single wavelength used in HPLC. This was used to assess for degradation products which absorb at different wavelengths and therefore may be missed if HPLC was used alone. Ultraviolet spectroscopy was also used to test bioavailability of the drug via the octanol:water partition coefficient under stressed and unstressed conditions. 5.4 Results The paracetamol in Perfalgan® did not degrade on exposure to air over 24 hours. Neither did it degrade on exposure to acid, alkali, oxidative or heat stress. The HPLC retention time was constantly between 3.17-3.29 minutes. 1 H NMR revealed no change in the formulation of Perfalgan® except for the conversion of the oxygen scavenger cysteine to cystine. The octanol:water partition coefficient likewise stayed constant and was in agreement with the value of 0.46-0.49 quoted in the literature (International Programme on Chemical Safety, 2008; McNeil Consumer Healthcare, 2010). 5.5 Conclusion The paracetamol in Perfalgan® did not degrade on exposure to air and other stressors over 24 hours. - Description: Urban forestry is advocated worldwide as a means of enhancing the liveability of towns and cities, and mitigating some of the anticipated impacts of climate change. Optimisation of the benefits of trees in urban areas is dependent upon knowledge of tree form, growth, and the products and benefits that trees provide. Growth rates are a vital variable for modelling benefits, yet there is a significant gap in knowledge pertaining to growth rates of trees in urban areas, especially indigenous species in developing world countries. Here we report on growth rates of indigenous street and garden trees in Grahamstown, South Africa, using two approaches; tree ring counts on increment cores and mean rates from trees of known planting age. Growth equations for both street and garden trees were derived. There was no significant difference in mean growth rates determined via the two methods. For both methods street trees grew approximately 30% slower than trees in gardens.

- Description: Urban forestry is advocated worldwide as a means of enhancing the liveability of towns and cities, and mitigating some of the anticipated impacts of climate change. Optimisation of the benefits of trees in urban areas is dependent upon knowledge of tree form, growth, and the products and benefits that trees provide. Growth rates are a vital variable for modelling benefits, yet there is a significant gap in knowledge pertaining to growth rates of trees in urban areas, especially indigenous species in developing world countries. Here we report on growth rates of indigenous street and garden trees in Grahamstown, South Africa, using two approaches; tree ring counts on increment cores and mean rates from trees of known planting age. Growth equations for both street and garden trees were derived. There was no significant difference in mean growth rates determined via the two methods. For both methods street trees grew approximately 30% slower than trees in gardens.

- Description: In this study, we report on the covalent linking of carboxylic acid functionalized silica nanoparticles with zinc phthalocyanine mono-substituted non-peripherally and peripherally with either a 4-amino phenoxy (1, peripheral and 2, non-peripheral) or an amino group (3 peripheral). The grafting is achieved via the formation of an amide bond between the carboxylic acid of the silica nanoparticles and the amino group of the phthalocyanine complexes. The hybrid nanoparticles retained the amorphous nature of silica nanoparticles after conjugation. A slight decrease in fluorescence and a general improvement in triplet quantum yields compared to free Pcs were observed. Triplet lifetimes for 2-SiNPs and 3-SiNPs also improved when compared to the free phthalocyanine. The changes in singlet oxygen quantum yields upon conjugation were minimal.

- Description: The purpose of the study was to evaluate the effect of different homogenization speeds and times, anchor speeds and cooling times on the viscosity and cumulative % clobetasol 17-propionate released per unit area at 72 h from pilot scale cream formulations. A 24 full factorial central composite design for four independent variables were investigated. Thirty pilot scale batches of cream formulations were manufactured using a Wintech® cream/ointment plant. The viscosity and in vitro release of CP were monitored and compared to an innovator product that is commercially available on the South African market, namely, Dermovate® cream. Contour and three-dimensional response surface plots were produced and the viscosity and cumulative % CP released per unit area at 72 h were found to be primarily dependent on the homogenization and anchor speeds. An increase in the homogenization and anchor speeds appeared to exhibit a synergistic effect on the resultant viscosity of the cream whereas an antagonistic effect was observed for the in vitro release of CP from the experimental cream formulations. The in vitro release profiles were best fitted to a Higuchi model and diffusion proved to be the dominant mechanism of drug release that was confirmed by use of the Korsmeyer–Peppas model. The research was further validated and confirmed by the high prognostic ability of response surface methodology (RSM) with a resultant mean percentage error of (±SD) 0.17 ± 0.093 suggesting that RSM may be an efficient tool for the development and optimization of topical formulations.
