- Description: In this treatise a SEIR model for the transmission of tuberculosis was proposed. It assumes that the latent infected individuals progress to active tuberculosis through endogenous reactivation and exogenous re-infection and that the individuals who have recovered from the disease through treatment are not immune to tuberculosis re-infection. While most features of the original model were kept, the model was modified to incorporate two control measures in the form of post-exposure tuberculosis prophylaxis for the treatment of latent individuals and tuberculosis therapeutics for the treatmentof individuals with active tuberculosis. Mathematical analysis of the modelwas done under the assumption that the two control measures are positiveconstants. The disease-free equilibrium point was locally asymptotically stable when the basic reproduction number was less than unity and unstable when this epidemic threshold exceeds unity. An optimal control problem was formulated and the necessary conditions for optimality were derived.The numerical results based on data from Port Elizabeth suggest that using both forms of control measures simultaneously is more effective in reducing the total number of infected individuals than applying single controls separately.

While incineration is the simplest way to recycle the used tyres it isn’t an efficient way to dispose of tyres. This research looks at developing a method for characterizing the New Reclamation Group (NRG) reclaim with the main focus on Hi-Res™ thermogravimetric analysis. Good quantification was possible using this technique. A trade off was established between resolution, sensitivity and time. While increasing the resolution allowed greater separation to be achieved the time for each experiment increased rapidly. Although kinetic models do exist for quantifying rubber components in vulcanized sample, they were not suitable for this study. When devulcanization causes significant molecular changes, such as with NRG reclaim, the decomposition profiles no longer match those of virgin materials. Formulations developed focused on mixing, rheometry, crosslink density and tensile properties. The NRG reclaim acted as a processing aid which lowered the maximum torque. This provides mixing safety as the temperature is decreased as a result of the lower torque. This effect was not seen with conventional reclaim. Rheometry tests indicated that the addition of the devulcanizates decreased the extent of cure. It was demonstrated that this could be linked to crosslink density. Testing of the 100%, 200%, 300% moduli correlated the crosslink density to the maximum torque. Although the addition of NRG reclaim reduced the tensile strength of the formulation, a link between crosslink density and ultimate tensile strength (UTS) couldn’t be made. The decrease in the UTS and increase in extension at break is possibly caused by an increase in low molecular weight material present in the formulations and decrease in crosslink density. This could possibly increase the mobility of polymer chains which could increases flexibility.

Seasonal physiological responses in the Cape Rockjumper (Chaetops frenatus): a Fynbos endemic bird shows limited capacity to deal with temperature extremes

Of the six Fynbos-endemic passerines, Cape Rockjumpers (hereafter Rockjumpers; Chaetops frenatus) are most vulnerable to increases in temperature, with population declines correlated with warming, and low physiological heat thresholds. Rockjumper’s preferred mountain habitat is predicted to decrease as they lack opportunity to move to cooler regions as temperatures warm. As Rockjumpers currently occupy the coldest regions of the Fynbos, I hypothesized their thermal physiology would show cold adaptation at the expense of lowered ability to cope with higher temperatures. I aimed to determine the seasonal 1) maintenance metabolism and cold tolerance, and 2) thermoregulatory responses to high temperatures of Rockjumpers. I measured seasonal maintenance metabolic rate, thermal conductance, and maximum thermogenic capacity. I also measured seasonal resting metabolic rate, evaporative water loss, evaporative cooling efficiency, and body temperature at high air temperatures. In winter, Rockjumpers had higher maximum thermogenic capacity, lower maintenance metabolic rate, and lower thermal conductance. Lower maintenance metabolic rates (and thus, lower metabolic heat production) combined with the decreased thermal conductance, confers substantial energy savings in winter. The increased winter maximum thermogenic capacity of Rockjumpers was expected, although the mean seasonal values fell below those expected for a ~ 50 g bird using a global data set, suggesting Rockjumpers are not especially cold tolerant. I further show that in summer Rockjumpers had higher elevations in resting metabolic rates, evaporative water loss, and body temperature, denoting higher rates of heat production and lower heat thresholds in summer compared to winter. - Description: The dissertation details the experimental work on the attempt to develop rhenium(V)phthalocyanine complexes directly from its +7 oxidation state (perrhenate). Different reducing agents (PPh3, Na2S2O5 and NaBH4) were employed and consequently, different results were acquired, such as rhenium(V)-mediated oxidative hydrolysis of the phthalocyanines (Pcs), the formation of a rhenium-phthalocyanine complex and phthalocyanine-capped nanoparticles. The rhenium nanoparticles that were formed were optimized from a synthesis point of view and, cancer localizing ability of the rhenium nanoparticles was investigated. The complexes were synthesized through direct metalation of pre-formed metal-free phthalocyanines using the “cold isotopes” of the rhenium metal. Rhenium nanoparticles (Re NPs) were synthesized in aqueous saline medium so as to imitate the environment on which Re is produced from its reactor. The nanoparticles (NPs) were capped with phthalocyanines which were covalently biofunctionalized with a folic acid moiety to enhance the targeting ability of the Re NPs. These NP systems were characterised with techniques such as ultraviolet-visible UV-Vis spectroscopy and transmission electron microscopy TEM. Cytotoxicity of the NPs was tested against four different cell lines and subsequently their cytotoxicity profiles were elucidated, and the profiles shown a dose-dependent responsealthough the results in some cell lines were unclear. Their fluorescence properties were also studied to provide photophysical information for investigation of their tumor localization using human cancer cells lines via confocal fluorescence microscopy studies. Particle size effect on localization of NPs was also investigated using confocal fluorescence and TEM. Two sizes were chosen (10 and 50 nm), and the smaller NPs (10 nm) were found to exhibit stronger fluorescence properties than the 50 nm NPs, and they were also found to have a better localization ability than the 50 nm NPs. Finally, their tumor and organ biodistribution studies will be carried out using micro-SPECT kits and model mice (using the “hot” isotopes in a radiopharmacy laboratory).

- Description: The main aim of this project is to test novel naturally-derived plasticizers (bio-plasticizers) synthesized at Innoventon and to determine if they are a suitable replacement for conventional plasticizers used to plasticize polyvinyl chloride. Polyvinyl chloride (PVC) is one of the most common plastics used in the world because of its durability and low cost. PVC when blended with plasticizers is a versatile thermoplastic and is used in many applications. A plasticizer is a substance that is added to a material, usually a polymer, to increase flexibility, softness and workability. Many elastomers and polymers are tough and dry; therefore they need plasticizers to make them useful. A plasticizer must be thoroughly incorporated in the elastomer or polymer in order for it to be effective. There are many types of plasticizers used in the world. Phthalate plasticizers are the most commonly used plasticizers of the past century. They comprise 92% of the global production of plasticizers. DEHP (di-ethyl hexyl phthalate) is the most widely used phthalate plasticizer. Phthalate plasticizers have been proved to be harmful to the environment and human beings. They can cause many harmful diseases to humans e.g. kidney failure, heart disease, liver failure, fertility and growth disorders. They lower testosterone in baby boys that are exposed through breast milk. In light of the above, the use of certain phthalates has been banned in cosmetics and consumer products. Since there is sufficient evidence to raise fears over the use of phthalate plasticizers, there is a great opportunity to find a way to replace phthalate plasticizers in PVC with bio-plasticizers. The current study represents the first step in developing alternative plasticizers for PVC and other polymers. These alternative plasticizers are characterized by low toxicity, low migration and are less harmful to the environment and human beings. p-menthane-3, 8-diol citronellal acetal is one such proposed bio-plasticizer which has a similar structure to the phthalates and has been previously reported by Innoventon to have potential plasticizing characteristics. During this study PVC was plasticized with PMD-citronellal acetal and tested for plasticizing properties. The plasticizing effect of PMD-citronellal acetal in PVC films was compared to that of conventional plasticizer dibutyl phthalate (DBP). The two plasticizers‟ performance was evaluated by preparing PVC films with varying amounts of each plasticizer and PMD-citronellal acetal subjecting the films to a series of tests. Tensile strength and elongation tests were performed on the plasticized and unplasticized films but these tests did not produce consistent results due to premature failures on the films. However, tensile strength was reduced in some films for both plasticizers compared to unplasticized film. Elongation and flexibility were increased in some samples as the percentage of plasticizer in each film was increased. Glass transition temperatures were determined for the plasticized and unplasticized PVC films. Glass transition temperature of both plasticizers was reduced as the percentage of plasticizer in each film was increased. Multiple regression analysis was done on the data to compare the similarity of the two plasticizers. The statistical tests showed that DBP and PMD-citronellal acetal have the same plasticization property with regard to glass transition temperature of PVC. The thermal stability of the PVC films with the two plasticizers was evaluated. Similar thermal stability was observed on the PVC, PVC with DBP and PVC with PMD-citronellal acetal. The leaching of the PMD-citronellal acetal and DBP were compared. The plasticizer loss increased as the percentage of each plasticizer in the PVC increased. The relationship between the percentage of the plasticizers and leaching rate was further analysed using multiple linear regressions. The two plasticizers were found to have the same leaching rate. FTIR analysis was performed on the plasticized and unplasticized PVC films to identify functional groups present and to determine whether the plasticizers formed any bonds with PVC functional groups. The characteristics bands of PVC were observed in FTIR. The FTIR spectra indicated that the plasticizers were incorporated into the PVC matrix without bonding. The results of the tests showed that PMD-citronellal acetal can be a suitable bio-plasticizer for PVC since it exhibited similar plasticization properties to DBP with regard to glass transition temperatures, leaching rates and thermal stability. Regarding tensile strength and elongation, further testing has to be done as the preliminary results were inconclusive. PMDcitronellal acetal was shown to be a possible alternative for DBP.

- Description: The study deals with the formulation development and thermorheological properties of crumb rubber/ethylene vinyl acetate (EVA) modified bitumen with the aim of optimizing the amount of crumb rubber and EVA loadings in bitumen based on thermorheological parameters complex shear modulus, phase angle and rutting parameter. In the modified binders EVA offers plastomer properties whereas the crumb rubber confers elasticity to the bitumen. 13.75% crumb rubber and 2.5% EVA loadings based on aging indices gave the best optimized mixture. The thermorheological behaviour of the best optimized mixture was compared to industrial grade EVA (AP-1) and crumb rubber (AR-1) modified bitumen. Improving aging behaviour of the bitumen was evaluated using four antioxidants: carbon black, hydrated lime, Irganox 1010 and Irgafos 168. The least aging indices at 58˚C and 64˚C were obtained from a 1:1:1 Irganox 1010/hydrated lime/carbon black mixture. SARA (saturates, aromatics, resins and asphaltene) analysis gave the chemical background for application of antioxidants to reduce the propensity of short term aging. This was done to match the EVA content and maintain the original maximum loading of 2.5% in the optimized mixture. The phase change abilities of FT wax owing to the high latent heat enabled co-crystallization with the EVA with the elastomeric backbone of crumb rubber acting as support material. The energy-sustainability nexus was found to have worked at less than 1% loading of FT wax in the EVA/crumb rubber modified bitumen. Response surface methodology approach to all the sections of work was used to come up with the optimized mixtures based on rheological parameters complex shear modulus, phase angle and rutting parameter at test temperature 64˚C. The interaction chemistry of bitumen, crumb rubber, EVA and FT wax was found to be first and second order in all cases involving individual contributions and co-interaction amongst the factors. Project costing for each section of work (optimizing section, antioxidant section and FT wax section) was conducted involving the raw materials used, equipment used, labour involved and other costs incurred for all the project work.

The aim of this study was to generate aptamers against the two highly conserved linear epitope regions (amino acid 421-440 and amino acid 601-620) of the EBOV nuclear protein (NP) using site directed SELEX. Such aptamers can be used as target recognition elements in the development of a LFD for the diagnosis of EBOV infection. Four aptamers that can potentially bind to the linear epitope spanning from amino acid 421 to 440 of the EBOV NP and four aptamers that can potentially bind to the linear epitope spanning from amino acid 601 to 620 of the EBOV NP were identified in this study. An in silico analysis of the predicted secondary structure of the putative aptamers was performed before and after the truncation of nucleotide sequences from the 5’ and 3’ ends of the aptamers to remove excess nucleotide sequences. Although this study did not characterise the interaction between the aptamers and linear epitope regions, the study succeeded in optimising the buffer conditions for future interaction studies using the SPR Biacore 3000 instrument.

The functionalized sorbent materials were characterized by microanalysis, SEM-EDS and FT-IR. Palladium selectivity of the sorbent materials was achieved by stripping of rhodium, iridium and platinum chlorido species with 0.5 M of NaClO4 in 1 M HCl while [PdCl4]2 was eluted with 3% w/v thiourea. The desorption efficiency of thiourea was confirmed by the SEM-EDS analysis of the materials after Pd(II) elution. Palladium loading capacity of the sorbents were in the order S-NSSN (23.85 mg/g) > S-NSN (12.70 mg/g) > S-TETA (4.97 mg/g). The extraction patterns on the sorbent materials were explained by considering the coordination chemistry of the ligand with [PdCl4]2ˉ and ionic interactions of [PtCl6]2ˉ and [IrCl5(H2O]ˉ. The square planar complexes, [Pd(HNSSNH)Cl2]Cl2 and [Pd(NSNH)Cl2][PdCl4]2, were isolated, analyzed by spectroscopy and single crystal X-ray, whereas evidence of the fraction of Pd(II)-TETA complex was obtained by HPLC studies. This provided proof of the inner sphere coordination mechanism as the mode of interaction of these ligands with [PdCl4]2ˉ. Trace amounts of Brˉ anions in ligands resulted in the inadvertent isolation of bromide coordinated Pd(II) NSN and NSSN complexes. The ion-pair salts of [TETAH4]4+ with [PtCl6]2ˉ, [IrCl6]3ˉ and [RhCl6]3ˉ were also isolated and characterized by microanalysis and IR to further explain the extraction patterns.

- Description: The most common treatment used for cancer is chemotherapy. Chemotherapeutic agents have a greater affinity for rapidly dividing cells which is a characteristic of tumour cells. Although anti-cancer agents have their advantages in providing anti-cancer effects, they can be seen as highly toxic molecules posing a threat to normal healthy tissue within the human body. However, these toxic therapies need to be delivered to tumour sites without damaging healthy tissue. Liposomes can serve as a delivery system for these toxic molecules and be delivered to the tumour site via the EPR effect. Hence, liposomes that fuse with tumour cell line membranes are advantageous in delivering payloads of drugs directly into the tumour cell without damaging normal healthy tissue. The aim of the study was to formulate an optimised liposome preparation in order to enhance cellular uptake by MCF-7, Caco-2 and C3A cancer cell lines via membrane fusion. The optimal liposome formulation was aimed to be prepared utilising a statistical design approach in order to determine the ranges of the parameters that were furthermost optimal in formulating an ideal liposome preparation. The primary screening design was conducted using a 24-1 fractional factorial design that took into account the four parameters that were used to determine the optimisation of the liposomal preparation. The four variables used in the liposome preparation were the phospholipid type (PS or DOPE), the concentration of cholesteryl hemisuccinate (CHEMS) (10 – 40 %), the concentration of PEG2000-PE (0.5 – 4 %) and liposome size (100 or 200 nm). Liposomes were prepared using thin film hydration method and characterisation for size and zeta potential was carried out using photon correlation spectroscopy (PCS). Visual characterisation of liposome size was carried out using atomic force microscopy (AFM). Liposomes were exposed the cancer cell lines with visualisation and uptake being measured using fluorescent microscopy and flow cytometry, respectively. An optimal liposome preparation was prepared following the statistical design method. The optimal liposome preparation consisted of phospholipid type PS, 22.91 % of CHEMS, 4 % of PEG2000-PE and a liposome size of 200 nm. AFM analysis has shown that optimal liposome sizes ranged between 130 and 170 nm. Flow cytometry analysis indicated high level of liposome uptake with actual values falling below the predicted values set out by the statistical design. Fluorescence microscopy captured images of the fluorescent liposomes concentrated on the membrane of cells. The objective of the study was to determine from literature which variables would be desirable in preparing an optimal non-targeted liposome preparation. This was achieved by identifying four such variables and utilising them in a statistical design approach which was screened in order to determine the ideal parameters in preparing the optimised liposome batch. Largely as a result, South African portfolios are net recipients of returns and volatility shocks emanating from major world markets. In light of these, and other, sources of risk, this dissertation proposes a methodology to improve risk management systems in funds by building a contemporary asset allocation framework that offers practitioners an opportunity to explicitly model combinations of hypothesised global risks and the effects on their investments. The framework models portfolio return variables and their key risk driver variables separately and then joins them to model their combined dependence structure. The separate modelling of univariate and multivariate (MV) components admits the benefit of capturing the data generating processes with improved accuracy. Univariate variables were modelled using ARMA-GARCH-family structures paired with a variety of skewed and leptokurtic conditional distributions. Model residuals were fit using the Peaks-over-Threshold method from Extreme Value Theory for the tails and a non-parametric, kernel density for the interior, forming a completed semi-parametric distribution (SPD) for each variable. Asset and risk factor returns were then combined and their dependence structure jointly modelled with a MV Student t copula. Finally, the SPD margins and Student t copula were used to construct a MV meta t distribution. Monte Carlo simulations were generated from the fitted MV meta t distribution on which an out-of-sample test was conducted. The 2014-to-2015 horizon served to proxy as an out-of-sample, forward-looking scenario for a set of key risk factors against which a hypothetical, diversified portfolio was optimised. Traditional mean-variance and contemporary mean-CVaR optimisation techniques were used and their results compared. As an addendum, performance over the in-sample 2008 financial crisis was reported. The final Objective (7) addressed management and conservation strategies for the NMBM. The NMBM wetland database that was produced during this research is currently being used by the Municipality and will be added to the latest National Wetland Map. From the database, and tools developed in this research, approximately 90 wetlands have been identified as being highly vulnerable due to anthropogenic and environmental factors (Chapter 6) and should be earmarked as key conservation priority areas. Based on field experience and data collected, this study has also made conservation and rehabilitation recommendations for eight locations. Recommendations are also provided for six more wetland systems (or regions) that should be prioritised for further research, as these systems lack fundamental information on where the threat of anthropogenic activities affecting them is greatest. This study has made a significant contribution to understanding the underlying geomorphological processes in depressions, seeps and wetland flats. The desktop mapping component of this study illustrated the dominance of wetlands in the wetter parts of the Municipality. Perched wetland systems were identified in the field, on shallow bedrock, calcrete or clay. The prevalence of these perches in depressions, seeps and wetland flats also highlighted the importance of rainfall in driving wetland formation, by allowing water to pool on these perches, in the NMBM. These perches are likely to be a key factor in the high number of small, ephemeral wetlands that were observed in the study area, compared to other semi-arid regions. Therefore, this research highlights the value of multi-faceted and multi-scalar wetland research and how similar approaches should be used in future research methods has been highlighted. The approach used, along with the tools/methods developed in this study have facilitated the establishment of priority areas for conservation and management within the NMBM. Furthermore, the research approach has revealed emergent wetland properties that are only apparent when looking at different spatial scales. This research has highlighted the complex biological and geomorphological interactions between wetlands that operate over various spatial and temporal scales. As such, wetland management should occur across a wetland complex, rather than individual sites, to account for these multi-scalar influences.

From the database, and tools developed in this research, approximately 90 wetlands have been identified as being highly vulnerable due to anthropogenic and environmental factors (Chapter 6) and should be earmarked as key conservation priority areas. Based on field experience and data collected, this study has also made conservation and rehabilitation recommendations for eight locations. Recommendations are also provided for six more wetland systems (or regions) that should be prioritised for further research, as these systems lack fundamental information on where the threat of anthropogenic activities affecting them is greatest. This study has made a significant contribution to understanding the underlying geomorphological processes in depressions, seeps and wetland flats. The desktop mapping component of this study illustrated the dominance of wetlands in the wetter parts of the Municipality. Perched wetland systems were identified in the field, on shallow bedrock, calcrete or clay. The prevalence of these perches in depressions, seeps and wetland flats also highlighted the importance of rainfall in driving wetland formation, by allowing water to pool on these perches, in the NMBM. These perches are likely to be a key factor in the high number of small, ephemeral wetlands that were observed in the study area, compared to other semi-arid regions. Therefore, this research highlights the value of multi-faceted and multi-scalar wetland research and how similar approaches should be used in future research methods has been highlighted. The approach used, along with the tools/methods developed in this study have facilitated the establishment of priority areas for conservation and management within the NMBM. Furthermore, the research approach has revealed emergent wetland properties that are only apparent when looking at different spatial scales. This research has highlighted the complex biological and geomorphological interactions between wetlands that operate over various spatial and temporal scales. As such, wetland management should occur across a wetland complex, rather than individual sites, to account for these multi-scalar influences.

Results reveal that tremendous urban development had taken place in the study area along the Jukskei River area for the past sixteen years. It was observed that there was a sharp decrease in vegetation from 237ha (1997) to 134ha (2006) to 68ha (2013). This may had a negative impact on the environment around this area by decreasing surface runoff. The trend however, shows that bare surface and vegetation land-cover class has no potential to recover. Questionnaires aimed at all the residents in the study area were used to assess the effect of periodic flooding on communities. Community leader and City of Johannesburg Disaster Management Unit (CoJDMU) interviews were also conducted to get more insight about floods management in the study area. Finally, strategies for alleviating flood risk in the study area were discussed and some recommendations were made to help the government and municipal authorities to improve and development sustainable flood mitigation measures.

Alternative agricultural production methods which are environmentally less taxing, are affordable and yield nutritious food, need to be investigated and adopted. A long-term trial investigating the differences in yields between conventional and organic farming systems was established at the NMMU George Saasveld campus. A baseline study preceded the cropping seasons to establish pre-treatment soil conditions before the two farming systems were be implemented. The trial was a randomized complete block design split into organic, conventional and control plots. In the first cropping season (summer) three crops cabbage (Brassica oleracea var. capitata), sweet potato (Ipomea batatas) and cowpea (vigna ungucuilata) were planted under the different treatments. Soil fertility changes after application of chemical and organic fertilizer were measured. Soil pH and phosphorus increased whilst potassium and soil carbon decreased from baseline levels. The organic plots were found to have higher soil pH, potassium and carbon, whilst P was higher in the conventional plots. The organic cabbages had a yield 12% lower than conventional cabbages, organic cowpeas were 51% lower than conventional cowpeas. Baboons damaged sweet potato plots before maturity. Broccoli (Brassica oleracea var. italica), radish (Raphanus sativa) and green pea (Pisum sativum) were planted in the second cropping season (winter). Organic broccoli yields were 51% lower than conventional broccoli, organic green peas were 18 % lower than conventional green peas. The organic radish yields were 36% lower than conventional yields and this was statically significant. The yield differences were statically significant between control and conventional cabbages (25%) and broccolis (68%). The results indicate that there is on average an initial 25% yield difference between organic and conventional farming systems. Similar studies have shown that the yield gap can be reduced within three to four years. This study presents preliminary results of trials that are to continue for ten years, during which time the yield differences may vary.

The material’s resistance to creep damage due to its microstructure can be quantitatively described by the back-stress. There are four microstructural contributions to the back-stress: Precipitate Hardening, Sub-Boundary Hardening, Solid-Solution Hardening and Dislocation Hardening. Fusion welding is performed on creep aged materials when a component needs to be replaced. This high temperature process results in the formation of different microstructural regions within the weldment. These creep damaged components have a weldability limit as set by the life management strategy of the power plant company. Measuring techniques capable of quantifying the microstructural contributions (precipitates, subgrains and dislocations) were developed and evaluated in this study. These techniques were then used to characterise the different microstructural regions within a new and creep aged X20 steel weldment. Differences in the microstructure of the new and creep aged X20 steel was illustrated by the results of this study. The measured size and number densities of the precipitates in the creep aged X20 material showed that there is a decrease in PH during creep exposure. There was a decrease in SBH and DH stress for the creep aged X20 material due to coarsening of the subgrains and annealing of dislocations during creep exposure. The quantitative techniques demonstrated in this study opens up the possibility to perform life assessment on weldments with inhomogeneous microstructures by following a microstructural based approach.

In particular, the technique is used to investigate the sub-bandgap response associated with the incorporation of GaSb quantum rings into the active region of a conventional GaAs p-i-n solar cell. These GaSb/GaAs quantum ring solar cells are a class of third generation cells, with the potential to exceed the Shockley-Queisser efficiency limit of single junction devices. Wavelength-modulation spectroscopy (WMS) techniques involve the modulation of the wavelength of a pseudo-monochromatic light source, with the resulting variation in the measured photocurrent then being a measure of the differential optical response of the solar cell. Although the conventional photocurrent spectrum of a solar cell is a good measure of the optical response characteristics, the differential technique gives supplemental detail related to the absorption spectrum. In addition to the basic WMS setup, we also developed an in situ flux correction module to ensure that a constant excitation intensity is maintained during the wavelength modulation. The excitation source inherently has a spectral dependence that leads to an undesirable contribution to the photocurrent signal. The operation of the flux corrected WMS setup has been demonstrated by photocurrent and photo-capacitance response measurements to obtain the differential quantum efficiency and charging characteristics of the quantum ring solar cells.

Synthesis and characterisation of lanthanide complexes with nitrogen- and oxygen-donor ligands

- Description: The reactions of Ln(NO3)3∙6H2O (Ln = Pr, Nd or Er) with the potentially tridentate O,N,O chelating ligand 2,6-pyridinedimethanol (H2pydm) were investigated, and complexes with the formula, [Ln(H2pydm)2(NO3)2](NO3) (Ln = Pr or Nd) and [Er(H2pydm)3](NO3)3 were isolated. The ten-coordinate Pr(III) and Nd(III) compounds crystallise in the triclinic space group P-1 while the nine-coordinate Er(III) complex crystallises in the monoclinic system (P21/n). The reaction of PrCl3∙6H2O with H2pydm yielded the compound, [Pr(H2pydm)3](Cl)3, that crystallises in the monoclinic system, space group P21/c with α = 90, β = 98.680(1) and γ = 90°. The nine-coordinate Pr(III) ion is bound to three H2pydm ligands, with bond distances Pr-O 2.455(2)-2.478(2) Å and Pr-N 2.6355(19)-2.64(2) Å. X-ray crystal structures of all the H2pydm complexes reveal that the ligand coordinates tridentately, via the pyridyl nitrogen atom and the two hydroxyl oxygen atoms. The electronic absorption spectra of complexes show 4f-4f transitions. Rare-earth complexes, [Ln(H2L1)2(NO3)3] [Ln = Gd, Ho or Nd], were also prepared from a Schiff base. The X-ray single-crystal diffraction studies and SHAPE analyses of the Gd(III) and Ho(III) complexes shows that the complexes are ten-coordinate and exhibit distorted tetradecahedron geometries. With proton migration occurring from the phenol group to the imine function, complexation of the lanthanides to the ligand gives the ligand a zwitterionic phenoxo-iminium form. A phenolate oxygen-bridged dinuclear complex, [Ce2(H2L1)(ovan)3(NO3)3], has been obtained by reacting Ce(NO3)3∙6H2O with an o-vanillin derived Schiff base ligand, 2-((E)-(1-hydroxy-2-methylpropan-2-ylimino)methyl)-6-methoxyphenol (H2L1). Hydrolysis of the Schiff base occurred to yield o-vanillin, which bridged two cerium atoms with the Ce∙∙∙Ce distance equal to 3.823 Å. The Ce(III) ions are both tencoordinate, but have different coordination environments, showing tetradecahedron and staggered dodecahedron geometries, respectively. The reaction of salicylaldehyde-N(4)-diethylthiosemicarbazone (H2L2) in the presence of hydrated Ln(III) nitrates led to the isolation of two novel compounds: (E)-2[(ortho-hydroxy)benzylidene]-2-(thiomethyl)-thionohydrazide (1) and bis[2,3-diaza4-(2-hydroxyphenyl)-1-thiomethyl-buta-1,3-diene]disulfide. The latter is a dimer of the former. For this asymmetric Schiff base, 1 and the symmetric disulfide, classical hydrogen bonds of the O–H∙∙∙N as well as N–H∙∙∙S (for 1) type are apparent next to C–H∙∙∙O contacts. 4-(4-Bromophenyl)-1-(propan-2-ylidene)thiosemicarbazide was also prepared upon reacting 4-(4-bromophenyl)-3-thiosemicarbazide with acetone in the presence of ethanol and La(NO3)3∙6H2O. Sandy beach ecosystems are at the interface between land and sea and thus receive nutrients from both land/seascapes. The seasonal nesting of sea turtles introduces large inputs of eggs, and so nutrients, onto sandy beach ecosystems, but little is known about the effects of these spatially and temporally variable nutrient input pulses on the dynamics of consumers in the recipient system. In this study, I examined the ecological role of sea turtles as vectors of nutrients that introduce large amounts of nutrients (in the form of eggs) from distant foraging grounds into nutrient-poor beach ecosystems. Although some of the nutrients return to the sea in the form of hatchlings, nutrients from unhatched and depredated eggs, dead and predated hatchlings, as well as chorioallantoic fluid and egg shells remain on the beach and presumably enter sandy beach food webs. I hypothesized that turtle nutrients significantly increase the availability of nutrients to sandy beach ecosystems and that those nutrients are incorporated by both terrestrial and marine food webs. These hypotheses were tested by comparing isotopic signatures of 13C and 15N of consumers on beaches with high and low turtle nest densities. The response of meiofauna to the decomposition of turtle eggs was also investigated. I predicted that meiofaunal abundance is positively affected by turtle nutrients and that higher meiofaunal abundances will be obtained in decomposing, depredated nests. I tested this hypothesis by comparing meiofaunal abundance in naturally predated nests to densities away from turtle nests (as a control). An in situ experiment that mimics conditions of naturally predated sea turtle nest, was set up to test meiofaunal community responses to turtle nutrients over time. The study indicates that sea turtle eggs represent a short pulsed resource subsidy that increases the nutrient and energy budget of sandy beach ecosystems. The results show that of the five potential nutrient pathways tested, ghost crabs appear to consume egg nutrients in measurable quantities, altering their diet and feeding behaviour according to food availability. The study also showed that there was a strong, but short-lived positive response of meiofauna to the introduction of nutrients, with increased abundance of all taxa in predated nests and experimental treatments. This response was particularly strong for nematodes which peaked in abundance after seven days. I conclude that turtle-derived nutrients represent a pulsed resource subsidy that makes significant contribution to the energy budget of sandy beach/dune ecosystems.

Knowledge is required to conserve these systems, but ephemeral wetland research in South Africa is lacking. As primary producers, microalgae provide a key link between the biotic and abiotic components of aquatic ecosystems. Despite their importance, there is a paucity in information concerning microalgal dynamics in ephemeral wetlands. The aim of this study was to understand the spatial and temporal dynamics of microalgae in ephemeral wetlands of the Nelson Mandela Bay Metropole. The research followed a funnel-based approach that assessed 35 wetlands at a regional level, followed by a temporal assessment of six wetlands, monitored biweekly, monthly or quarterly. Thereafter, a case study at local scale was carried out to address wetland connectivity and Hydrogeomorphic unit trends. The research culminated in a benthic flux chamber experiment, addressing the influence of microalgal-mediated processes on wetland development for a month post inundation. Phytoplankton biomass showed strong regional associations with rainfall zone, with Chl a concentrations of 17; 4.6; and 25 µg.L-1 in the high, intermediate and low rainfall zones, respectively. The MPB communities in Seeps, Depressions and Wetland flats, comprised between 45 and 60 % diatoms, whilst other microalgal groups were dominant in the water columns (> 90 %). Phytoplankton and MPB interactions facilitated significant night-time NH4+ and SRP effluxes, (1.5 and 0.4 mg.m-2.h-1, respectively) four days post inundation. High Silica uptake in the latter stages, was attributed to an increase in diatom abundance. Within the chambers, MPB growth was continuous whilst phytoplankton exhibited cyclical growth as the system approached dynamic equilibrium. This study demonstrated the importance of nutrient cycling in structuring aquatic food webs, and indicated the sensitivity of ephemeral wetlands to environmental perturbations.

Morphospecies A, B, C and D were therefore delimited according to the Phenetic Species Concept. None of the Eurydice individuals examined exhibited the same suite of characters as E. longicornis. Molecular sequences of two mitochondrial markers, cytochrome c oxidase 1 and 16S rRNA were analysed using both distance- and monophyly based methods. Uncorrected p-distances, a Minimum Spanning Haplotype Network and Maximum Likelihood phylogenetic trees were investigated to determine genetic diversity and phylogenetic relationships in the six Eurydice morphospecies. Results from the genetic analysis made it possible to delimit morphospecies A B, C and D as phylospecies A, B, C and D according to the Phylogenetic Species Concept, thereby validating the species status of four new Eurydice species, provisionally called Eurydice A, Eurydice B, Eurydice C and Eurydice D. This increases the total number of Eurydice species from South Africa to seven, of which six form part of the macrofauna of sandy beaches. The outcomes of this study represent an important step in the resolution of the taxonomy of the genus Eurydice in South Africa and enhance current knowledge of the biodiversity of sandy beach macrofauna, thus facilitating better-informed decisions on conservation policies and management to the benefit of conservation of biodiversity on sandy beaches in South Africa.

Efficiency analysis of electricity distribution by South African Municipalities: a cost frontier approach

- Description: Efficiency analysis in the public sector assesses the performance of municipal and government entities in their provision of services to the public. South Africa, in particular, has experienced a large degree of negative feedback with respect to the ability of the government and its municipal departments to provide basic services to citizens. This has led to a number of service delivery protests throughout the country. To remedy this, the ability of the municipality to provide basic services needs to be assessed in order for improvements to be made. A first step in this process would be to determine the efficiency with which municipalities are providing these services. This study focuses on a particular municipal service, namely electricity distribution. Primarily, the efficiency with which municipalities provide electricity to consumers is determined. This is achieved using the parametric cost frontier approach, which is appropriate since municipalities aim to reduce the costs incurred in distributing electricity. The municipalities are compared to a frontier (theoretical best practice) from which inferences on their performances can be made. Those municipalities whose performances are not optimal, deviate from the frontier. The deviations (errors) are then assumed to be caused by two factors, namely stochastic random noise and inefficiency. This composition accounts for effects for which municipalities cannot control (stochastic random noise) and those for which it can (inefficiency). The parametric nature of the cost frontier approach allows for distributional assumptions to be made on these factors. Stochastic random noise is always assumed to be normally distributed, while inefficiency is assumed to be one-sided and positively skewed.

- Description: Mysticetes, or baleen whales, make up one of two sub-orders of the Order Cetacea within the Class Mammalia (Drikamer et al, 2013). It contains three families, each characterized by baleen plates (Drikamer et al, 2013). These baleen plates consist of keratin that replaced the tooth structures for feeding in these mammals (Drikamer et al, 2013). Other biological characteristics of these animals include paired external nares (blowholes), a sternum consisting of a single bone, symmetrical skull and the most conspicuous of all, the body length of between ten to thirty metres (Drikamer et al, 2013). It is thought that the size characteristic was probably the main factor when categorising them as cetaceans, with the Latin word ‘Cetus’ referring to ‘sea monster’ (Carnaby, 2006). In this literature review, the focus will fall on two species of mysticetes from separate families, namely that of the southern right whale, Eubalaena australis of the Balaenidae family, and the humpback whale, Megaptera novaeangliae of the Balaenopteridae family. Emphasis in this review will fall on the aspect of site fidelity of the mother-calf pairs.
