- Description: Complaints of crop damage by cranes on planted maize in the North Eastern Cape, South Africa, have been increasing since the mid-1990‘s, and in some instances severe losses have been reported. Crop damage by the Grey Crowned Crane Balearica regulorum regulorum near the town of Maclear (31º04´S 28º22´E), has been quantified over two growing seasons, and assessed relative to losses caused by foraging Cape Crows Corvus capensis and other feeding damage assumed to be caused by insects. Twelve fields were selected based on previous patterns of crop depredation. Maize seed in seven of the fields was treated with the chemical ‗Gaucho‘ and five fields were planted with untreated maize. In order to determine the source of losses, twenty quadrats (4 m x 4 m) randomly distributed within each field were visited on average every second day, for a period of up to twenty eight days. Results indicate that seed treatments do act as a deterrent to feeding by both cranes and crows, however crane damage is generally insignificant compared to other sources of damage. My study also reviewed past sightings data of the Grey Crowned Crane in an effort to determine if the conversion of former grassland to plantations in this region may have increased foraging activity in maize fields. The data did not allow for clear-cut conclusions regarding changes in distribution or population trends. Conclusions provide direct input into the management of agricultural areas by enabling landowners to take steps to mitigate crop damage. These mitigation measures may either involve the application of seed treatments, or the planting of low risk crops in high risk areas. Future studies should consider the ppossible detrimental effects of chemical seed treatments on crane biology.

- Description: This study looks at the impact of elephant feeding on the Xeric Succulent Thicket component of Eastern Cape Subtropical Thicket (ECST) in Addo Elephant National Park (AENP). Observations of elephant feeding were carried out and vegetation transects were surveyed for impact of elephant feeding. The results indicated that the Nyati elephants spent the majority of their time grazing (nearly 90%), particularly the cow-young herds, and especially when the herd gathered in larger numbers. Browsing events were concentrated on Acacia karroo (81%) and there was no significant difference between the sexes in their preference for this species. Despite being subjected to most of the browsing, the majority of A. karroo trees were undamaged and the effect of elephants was generally light. It appears unlikely that, three years after re-introduction to Nyati, the elephants have had an effect on community structure of the vegetation. Surveys were conducted on stands of the alien invasive weed prickly pear Opuntia ficus-indica, and it was recorded that elephants in Nyati have had a dramatic effect on prickly pear, utilising all adult plants assessed and destroying 70% of them. This level of destruction in such a short period of time suggests that prickly pear is a highly favoured species. The results from the present study suggest that elephants can play a role in the control of prickly pear. Results are discussed in terms of elephants as both megaherbivores and keystone species, and as agents of intermediate disturbance.

An understanding of the drivers of conflict is essential for any efforts to achieve coexistence. This study investigated the levels of conflict, livestock management and tolerance amongst the farming communities of the Southern and Western Kalahari in Botswana. A questionnaire survey was completed with 310 farmers throughout the region. The majority of respondents regarded coexisting with carnivores as a challenge, with losses due to depredation perceived as the greatest problem facing farmers. Conflict was widespread throughout the study area, with some spatial variations for certain species. Cattle management levels were low and while smallstock management was better, the use of improved levels of management could reduce current levels of conflict. Tolerance levels were generally low with few respondents seeing the benefits of coexistence with carnivore species. The results indicated that farm type, gender, education level, source of income, livestock numbers, location and land use all have an effect on perceived conflict and tolerance levels and strongly interact with each other. In general cattleposts were characterised by higher proportions of females, lower education levels, more benefits derived from wildlife and veld products and fewer livestock than fenced ranches. They also experienced more conflict, carried out more management and had better tolerance levels. It could be that closer connections to the land and deriving benefits from natural resources resulted in more tolerance and this is certainly worth further investigation. An improvement in the use of effective methods of livestock management, targeted environmental education programs to develop a greater awareness for the conservation value of carnivores and a diversification of livelihoods to include benefits from natural resources have the potential to reduce conflict and improve tolerance in the Kalahari region.

- Description: The re-introduction of cheetah onto small reserves in the Eastern Cape Province has created the opportunity to study this species in an environment (Valley Bushveld) in which it has not previously been studied and which is quite different from the less wooded habitats in which previous studies have occurred. Thus the aim of this study is to add to the growing number of studies of cheetah in more wooded habitats and to contribute a better understanding of the level of specialization or adaptability in space use, habitat selection, diet and hunting behaviour that the cheetah possesses. The research was conducted on Kwandwe Private Game Reserve from February 2003 to August 2004. Direct observations, scat analysis and continuous observations were used to avoid bias towards large sized prey found when only using direct observations to study diet. The cheetah killed 15 species and four of the five most important prey species (kudu, springbok, grey duiker and bushbuck) were hunted according to their abundance on the reserve, while impala, the fifth most important prey species, was avoided (i.e. preference index less than one) by the cheetah. The cheetah’s main prey was medium sized ungulates, although the three male coalition killed larger prey than females and females with cubs. The daily food intake per cheetah per day was calculated to range from 5.3kg/cheetah/day for the coalition to 8kg/cheetah/day for a solitary female. Home range areas ranged from 11.1 km² for female cheetah with cubs in a den to 65.6 km² for single female cheetah and core areas ranged from 6 km² for the three male coalition to 26.5 km² for independent cubs. Habitat selection by cheetah on Kwandwe varied between the social groups depending on their susceptibility to predation by lions, their need for cover and need for water, and was similar to what has been previously reported. The home range of the coalition incorporated the most open vegetation type (karroid shrubland) with surrounding denser vegetation, while females occupied areas of denser vegetation. Activity patterns and hunting behaviour varied between different cheetah social groups with female cheetah being more active during day light hours compared to males which made 38% of their kills after dark. The average chase distances for the various cheetah groups varied considerably, and the chase distance for successful hunts was longer than for unsuccessful for all groups except single female cheetah. The percentage of kills’ kleptoparasitised on Kwandwe was very low compared to other studies possibly due to the low density of direct competitors, which in turn lead to longer mean kill retention times. These results suggest that cheetah are more adaptable than previously thought and this adaptability may have important implications for their conservation.

A standardised protocol for roadkill detection and the determinants of roadkill in the greater Mapungubwe Transfrontier Conservation Area, Limpopo Province, South Africa

- Description: Despite evidence suggesting that road traffic is a major threat to biodiversity loss, very little is known about its actual impact on wildlife populations in South Africa. Globally, road density and traffic volumes are increasing, and although huge budgets are devoted to the construction and upgrading of roads, there is little or no allocation to mitigation measures for protecting fauna in most countries, particularly Africa. Further, no global standardised protocol exists for the rapid assessment of roadkill or the most economical and efficient approach for assessing roadkill rates. Using vehicle field trials, the reliability of detecting artificially deployed roadkill was assessed. Roadkill detection rates decreased significantly at speeds >50 km/h and were also significantly influenced by light conditions (i.e. detection success was greater when the sun was high) and the position of the roadkill on the road (i.e. smaller roadkill on verges were often missed). These results suggest that roadkill sampling was most effective between 1.5 hours after dawn and 1.5 hours before dusk and that driving at slower speeds (<50 km.h⁻ₑ) was required to detect roadkill. This protocol was implemented across three ecological seasons on a 100 km paved road and a 20 km unpaved road in the Greater Mapungubwe Transfrontier Conservation Area, Limpopo Province, South Africa. Driven daily over a 120-day period (three periods consisting each of 40 days), a total of 1,027 roadkill were recorded. These comprised 162 species from all terrestrial vertebrate groups with birds being the most commonly encountered roadkill (50% of all incidents). The high numbers of vertebrates identified as roadkill suggests that road traffic could have potentially unsustainable impacts on wildlife populations and hence the biodiversity of the area. Seventeen variables were identified as possible determinants of roadkill occurrence with season, rainfall, minimum and maximum temperature, habitat type, grass height, grass density, fence type and vehicle type significantly influencing roadkill numbers. Significantly more roadkill were detected on the paved road (9.91/100km) than on the unpaved road (1.8/100km) probably because of greater traffic volumes and the increased speed that vehicles travelled on the paved road. Warmer temperatures and increased rainfall in the preceding 24 hours also increased road mortality numbers as animals tended to become more active during these times. Interestingly, more roadkill was detected in open roadside habitats compared to dense roadside habitats on both the paved and unpaved roads and when grass on the roadside verge was of intermediate height. Open habitat possibly may provide a natural corridor for wildlife which ultimately end up on the road. Roadkill numbers increased when certain other physical barriers, such as cattle fences, were present, probably because these barriers were more penetrable than electric fencing. A series of mitigation measures are proposed to reduce the impacts of roads on wildlife in South Africa. These mitigation measures highlight the need to address the balance between the development of a country’s transport infrastructure and the conservation of its fauna. - Description: Despite evidence suggesting that road traffic is a major threat to biodiversity loss, very little is known about its actual impact on wildlife populations in South Africa. Globally, road density and traffic volumes are increasing, and although huge budgets are devoted to the construction and upgrading of roads, there is little or no allocation to mitigation measures for protecting fauna in most countries, particularly Africa. Further, no global standardised protocol exists for the rapid assessment of roadkill or the most economical and efficient approach for assessing roadkill rates. Using vehicle field trials, the reliability of detecting artificially deployed roadkill was assessed. Roadkill detection rates decreased significantly at speeds >50 km/h and were also significantly influenced by light conditions (i.e. detection success was greater when the sun was high) and the position of the roadkill on the road (i.e. smaller roadkill on verges were often missed). These results suggest that roadkill sampling was most effective between 1.5 hours after dawn and 1.5 hours before dusk and that driving at slower speeds (<50 km.h⁻ₑ) was required to detect roadkill. This protocol was implemented across three ecological seasons on a 100 km paved road and a 20 km unpaved road in the Greater Mapungubwe Transfrontier Conservation Area, Limpopo Province, South Africa. Driven daily over a 120-day period (three periods consisting each of 40 days), a total of 1,027 roadkill were recorded. These comprised 162 species from all terrestrial vertebrate groups with birds being the most commonly encountered roadkill (50% of all incidents). The high numbers of vertebrates identified as roadkill suggests that road traffic could have potentially unsustainable impacts on wildlife populations and hence the biodiversity of the area. Seventeen variables were identified as possible determinants of roadkill occurrence with season, rainfall, minimum and maximum temperature, habitat type, grass height, grass density, fence type and vehicle type significantly influencing roadkill numbers. Significantly more roadkill were detected on the paved road (9.91/100km) than on the unpaved road (1.8/100km) probably because of greater traffic volumes and the increased speed that vehicles travelled on the paved road. Warmer temperatures and increased rainfall in the preceding 24 hours also increased road mortality numbers as animals tended to become more active during these times. Interestingly, more roadkill was detected in open roadside habitats compared to dense roadside habitats on both the paved and unpaved roads and when grass on the roadside verge was of intermediate height. Open habitat possibly may provide a natural corridor for wildlife which ultimately end up on the road. Roadkill numbers increased when certain other physical barriers, such as cattle fences, were present, probably because these barriers were more penetrable than electric fencing. A series of mitigation measures are proposed to reduce the impacts of roads on wildlife in South Africa. These mitigation measures highlight the need to address the balance between the development of a country’s transport infrastructure and the conservation of its fauna. - Description: Interactions between insects have been shown to be important regulators of population abundances and dynamics as well as drivers of spatial segregation and distribution. These are important aspects of the ecology of insects used in biological control and may have implications for the overall success of a particular programme. In the history of biological control there has been a tendency to release a suite of agents against a weed, which in some cases has increased the level of success, while in others little change has been observed. In most of these cases the implications of increasing the level of complexity of the system is not taken into account and there is little research on the effect of releasing another agent into the system. A brief meta-analysis was done on all the biological control programmes initiated in South Africa. Emphasis was placed on multi-species releases and the effects that overlapping niches were having on the number of agents responsible for the success of a programme. Where overlapping niches were present among agents released the number of agents responsible for success was lower than the number established. Water hyacinth, Eichhornia crassipes (Martius) Solms-Laubach in South Africa has more arthropod agents released against it than anywhere else in the world, yet control has been variable. If the biology and host utilisation of all the agents against water hyacinth is considered, a definite overlap of niches is apparent in at least one life stage of all the agents. Therefore the probability of these insects interacting is high, especially if they are established at the same site in the field. Three of the insects released in South Africa have been selected to investigate possible interactions. They are Neochetina eichhorniae Warner, Neochetina bruchi Hustache and Eccritotarsus catarinensis (Carvalho). Y-tube olfactometer bioassays were used to measure responses of these insects to water hyacinth with prior feeding damage by either conspecifics or heterospecifics. This was done to determine whether olfactory cues played a role in host acceptability and avoidance of conspecifics or heterospecifics. The insects were given a choice between damaged and undamaged plants in various combinations. There was a significant preference for the undamaged plants when given a choice between undamaged and damaged plants. However when the insects were given a choice between two damaged plants there was no discrimination between heterospecific or conspecific damaged plants. This may indicate that there is little or no ecological cost for the insect to share a plant with other insects utilising a similar resource. Insect – insect interactions were investigated in a common garden plot experiment to measure the impact that pairwise combinations of the insect may have on their performance. There was a significant interaction between the mirid E. catarinensis and the weevil N. eichhorniae, with the weevil not performing as well when in combination with the mirid than when alone. Interestingly there was a negative interaction between the two weevil species when in combination, however it was impossible to determine which species was being affected if not both. None of the insects performed significantly better when in combination with another insect. A field study on Wriggleswade Dam in the Eastern Cape, South Africa was initiated to determine whether the relationship between the mirid E. catarinensis and the weevil N. eichhorniae could be determined in the field. The performance of the insects at the different sites in the field suggests that there was an interaction between the agents. This interaction did not limit the establishment of either insect at a site, but it did result in one insect dominating at a site over another. Interactions between the three species of insect tested in this thesis suggest that there are both negative and neutral relationships between them. A basic comparison between the insect performances from 15 sites around the country was done to determine if the spatial segregation observed in the field could be extrapolated to the natural South African situation. The interaction observed between N. eichhorniae and E. catarinensis does seem to extrapolate to the general South African situation where there is definite spatial segregation on a landscape level. The co–occurrence of the two Neochetina weevils at these sites suggests that the negative relationship observed between them in the common garden experiment does not extrapolate to the field. The results from this thesis suggest that the interactions between the agents tested would not limit establishment or have significant ramifications on performance. However, there may be spatial and temporal segregation of these species in the introduced range.

- Description: Human-wildlife conflict frequently involves carnivores, mainly because of their large home ranges and dietary requirements. As such, carnivores tend to be the first animals to be lost in human-dominated ecosystems. This is significant because the removal of carnivores can alter the functionality of ecosystems. However, the conservation of carnivores depends as much on the socio-political and socio-economic landscapes as it does on the ecological one. The consolidation of vast, un-fragmented conservation areas in Africa and the world is unlikely. Thus, unravelling the factors (both biological and sociological) responsible for and influencing human-predator conflict is critical for carnivore conservation. The Winterberg district in the Eastern Cape, South Africa has been a sheep (Ovis aries) farming stronghold for nearly 200 years. Consequently, conflict between farmers and predators is commonplace and depredation of livestock by caracals (Caracai caracal) remains a perennial problem. However, the extent of this human-predator conflict (including a reliable assessment of caracal density) has not been quantified. This study used camera trapping to estimate the density of caracals (a nonindividually recognisable species) in the Winterberg and a structured questionnaire to gauge the general attitudes of the farmers of the region. Caracal density was estimated to be 0.20 caracals/km². This estimate equates to a population of approximately 54 caracals across the entire district. Therefore, caracals do not appear to occur at high densities in the Winterberg. However, the survey of the residents of the Winterberg Conservancy revealed that predators were rated as the greatest problem faced by farmers in the area. Although not significant, it also showed that the size of a respondent's property and sheep mortality due to caracals had the greatest probability of affecting attitudes towards predators. The farmers lost less than 10% of their stock to caracal depredation on an annual basis. Therefore, the general perception that predators were the most important factor in stock loss in the area does not appear to be fully supported by my data. The Winterberg is a male-dominated, multigenerational society and it is likely this propagates certain perceptions towards predators which are a reflection of long-held family traditions and beliefs. Such situations can result in reactions which are disproportionate to the actual scale of the problem. However, my study was limited to a sub-set of the landowners in the Winterberg. Thus, more intensive assessments of caracal space use and community attitudes towards predators are recommended.

Interactions between fig wasps and their host figs

- Description: Fig trees (Ficus spp.) and fig wasps (Hymenoptera: Agaonidae) are partners in an intimate mutualism. The trees provide ovules in which wasp larvae develop while the wasps pollinate the flowers and are therefore indispensible for fig seed production. Agaonid fig wasps oviposit down the styles of fig flowers and it has generally been accepted that they were unable to reach the ovules of "long" styled flowers , which would produce seeds, thus maintaining an evolutionary stable mutualism. African fig species were found to have unimodal style length frequencies, with no separation into long and short styled flowers. In several species the ovipositors of their associated agaonids were long enough to reach the majority of ovules. The number of foundress agaonids entering a fig influenced fig seed set and therefore was an important factor regulating the proportion of flowers producing seeds or pollinators. In the two Ficus species that were studied, entry of more than three agaonid foundresses into one fig resulted in competition for limited oviposition sites and less female - biased offspring sex ratios. It is hypothesised that sequential laying of male eggs followed by female eggs, under variable oviposition site limitation, results in sex ratio adjustment, as predicted by local mate competition theory. Evidence in support of this hypothesis is presented. A number of non - pollinating torymid and pteromalid fig wasps also oviposit into each fig species. The sycophagines and sycoecines oviposit down the styles from inside the fig inflorescences like their agaonid counterparts, while other species insert their ovipositors through the wall of the fig from the outside. Like the agaonids, sycophagines were characterised by being pro - ovigenic, with numerous fully developed eggs at emergence. Sycoecines were able to re - emerge from figs they had oviposited in and lay their eggs in more than one fig. They had short ovipositors, allowing access to a smaller proportion of flowers than agaonids or sycophagines. Externally ovipositing fig wasps were syn-ovigenic, able to develop eggs as adults and invested more energy and time during each oviposition event. - Description: Fig trees (Ficus spp.) and fig wasps (Hymenoptera: Agaonidae) are partners in an intimate mutualism. The trees provide ovules in which wasp larvae develop while the wasps pollinate the flowers and are therefore indispensible for fig seed production. Agaonid fig wasps oviposit down the styles of fig flowers and it has generally been accepted that they were unable to reach the ovules of "long" styled flowers , which would produce seeds, thus maintaining an evolutionary stable mutualism. African fig species were found to have unimodal style length frequencies, with no separation into long and short styled flowers. In several species the ovipositors of their associated agaonids were long enough to reach the majority of ovules. The number of foundress agaonids entering a fig influenced fig seed set and therefore was an important factor regulating the proportion of flowers producing seeds or pollinators. In the two Ficus species that were studied, entry of more than three agaonid foundresses into one fig resulted in competition for limited oviposition sites and less female - biased offspring sex ratios. It is hypothesised that sequential laying of male eggs followed by female eggs, under variable oviposition site limitation, results in sex ratio adjustment, as predicted by local mate competition theory. Evidence in support of this hypothesis is presented. A number of non - pollinating torymid and pteromalid fig wasps also oviposit into each fig species. The sycophagines and sycoecines oviposit down the styles from inside the fig inflorescences like their agaonid counterparts, while other species insert their ovipositors through the wall of the fig from the outside. Like the agaonids, sycophagines were characterised by being pro - ovigenic, with numerous fully developed eggs at emergence. Sycoecines were able to re - emerge from figs they had oviposited in and lay their eggs in more than one fig. They had short ovipositors, allowing access to a smaller proportion of flowers than agaonids or sycophagines. Externally ovipositing fig wasps were syn-ovigenic, able to develop eggs as adults and invested more energy and time during each oviposition event. This is especially important in cases where the morphology of invasive alien plants does not reflect genetic differences between populations. A previous study of the genetic structure of the invasive populations of N. mexicana in South Africa suggests the presence of hybrid forms of the plant in South Africa, with only one of these populations matching with samples from the native range. However, the study only used samples from two sites in the native range using amplified fragment length polymorphisms (AFLPs), so it was necessary to conduct further genetic analyses using samples from more sites in the native range. Hence, the first aim of this study was to develop a better understanding of the genetic structure of N. mexicana populations in the native and invaded range. Genetic samples were collected from sites in the native range during field surveys for potential biological control agents, and inter-simple sequence repeats (ISSRs) were used to compare the genetic structure of invasive and native populations of N. mexicana in South Africa. The results from these analyses suggest that seven of the 14 investigated invasive populations of N. mexicana in South Africa are genetically similar to populations in the native range, while the remaining seven populations are likely to be hybrid forms of the plant. This knowledge will be useful to target populations for biological control and highlights the need for further genetic analyses to determine the parentage of these hybrids so that biological control efforts are more likely to be successful. The initiation of a biological control programme requires that a series of steps are taken in order to maximise the likelihood that this form of intervention will be successful. The first few steps include: identification of the target weed and its genetic structure; exploration in the native range for potential biological control agents; and prioritisation of these agents based on factors such as climatic and genetic compatibility, feeding damage, abundance, and likely host range. Hence, the second aim of this study was to conduct surveys for potential biological control agents in the native range of N. mexicana, and to prioritise these agents. Field surveys were conducted between August and October in 2018 at 17 sites in Florida, Louisiana, and Texas, USA. Sites were selected based on climatic similarity of native sites compared to invasive sites by use of MaxEnt modelling. Native N. mexicana plants were searched for natural enemies, and these were prioritised based on feeding damage, abundance, incidence, and observations of field host range. Two species were prioritised: Bagous americanus LeConte (Coleoptera: Curculionidae) and Megamelus toddi Beamer (Hemiptera: Delphacidae). These species will be imported into quarantine facilities at Rhodes University for host specificity tests to be conducted. Understanding the factors that contribute to the successful establishment of biological control agents is important to improve the efficiency and reduce the costs incurred during the initiation of biological control programmes. Acquiring knowledge of the factors that predict the efficacy of biological control agents is similarly important, and these factors are discussed in the last chapter of this study. The challenges of the biological control of hybrids are also considered, and recommendations are made for the control of N. mexicana and other plants in South Africa.

- Description: Anredera cordifolia (Basellaceae), Madeira vine, is a perennial, semi- succulent climber native from Paraguay to southern Brazil and northern Argentina. It has a history of weediness and difficulty of control once established. In South Africa Madeira vine has a wide range and distribution with altitudes ranging from 10-1800m above sea level. Described as a transformer species, its sheer weight is capable of breaking branches off trees, causing the potential collapse of forest canopies. Chemical and mechanical control methods are expensive, labour intensive and may provide only temporary relief. A biological control programme was therefore initiated in 2003. Cf Phenrica sp. 2 (Coleoptera: Chrysomelidae: Alticinae), was field collected from A. cordifolia in Brazil, SSW of Cascavel in the Paraná Province during a survey in November 2003. Eggs are laid in groups of 16 with the average fertility rate being 89%. After going though three larval instars, the larvae pupate in the soil with the adults eclosing after a period of 17 days. The total developmental time for a generation from egg to egg ranges between 7-8 weeks. Biological traits that favour the flea beetle as a possible biological control agent include long-lived adults (up to 5 months) and multiple generations during the summer period. Both adults and larvae feed extensively on leaves and stems and although developmental rates will slow down during the winter period, no indication of a definite diapause was found under the prevailing laboratory conditions. After completing the larval no-choice trials with twenty-six plant species from 14 plant families Phenrica sp. 2 proved to be adequately host specific, as larval development was only supported by 3 Basellaceae species (including the control A. cordifolia) and one Portulacaceae species. All of these are introduced species in South Africa. The only indigenous Basella species could not be tested as it has a very marginal distribution, and because it’s inconspicuous nature, it is seldom seen or collected. Adult multi-choice trials were restricted to species that could sustain larval development to give some indication of the acceptability of these species for adult feeding and oviposition. Although adult feeding was initially concentrated on B. alba, the oviposition preference was clear-cut as females only oviposited on A. cordifolia. In order to quantify the impact of Phenrica sp. 2 on plant biomass and to assess the incidence and intensity of foliar damage, a pair of adults was confined to the host plant, for 2 generations, with different levels of larval densities. The results indicated that the host plant, due to both larval and adult feeding, suffered leaf losses of up to 55%. Anredera cordifolia was however still capable of enlarging the root mass despite suffering huge leaf losses. This would imply that A. cordifolia has an effective re-growth capacity and it will only be vulnerable to attack of the storage organs that enable re-growth, or to repeated attack of other plant parts through which reserves are exhausted. Unfortunately the period of exposure (24 days) was too short to prove that Phenrica sp. 2 impacts on the below ground dry mass, but should the plant be completely defoliated, as was observed in the field, the host plant would be forced to deplete stored resources. Phenrica sp.2 has shown to be very host specific and although A.cordifoia loses its leaves during the winter period in most provinces in South Africa, the adults are long-lived and should be able to survive the leafless periods. - Description: African elephants (Loxodonta africana) were absent from large portions of the Eastern Cape Province of South Africa for more than 100 years following widespread hunting for ivory. However, recent shifts in land use practices have resulted in the establishment of many private game reserves throughout the region. Some of these reserves have reintroduced elephants, raising management concerns because of the perceived impact that elephants can have on vegetation and the animals that rely on it for resources. My thesis aimed to assess the role of elephants in determining the structure and complexity of the locally important Thicket Biome and how medium and large mammals are affected. I quantified the woody and succulent components of Albany Thicket across nine reserves with elephants between May 2016 and November 2017 using a modified Point-Centre-Quarter method. Camera traps were deployed at each site for the duration of a calendar year to measure the relative abundances of all medium and large mammals at the sites. Across all study sites, climatic conditions (specifically rainfall and temperature) were the primary drivers of woody vegetation structure and diversity. Elephants appeared to have little influence since they were reintroduced at low densities 20 years ago. The associated mammal communities were mostly influenced by the height and basal area coverage of the thicket across the sites. I conclude that because elephant populations have been maintained at relatively low densities across my study sites, negative effects on the thicket vegetation and the associated mammal communities were not observed. In fact, the establishment of private game reserves, even with elephants, present may offer sustainable conservation for the threatened Albany Thicket. However, these elephant populations are still relatively new and changes to the vegetation are likely to be cumulative. Thus, future research should focus on how the vegetation is affected over time. To this end, I recommend the establishment of permanent sampling stations across all reserves with elephants in the Eastern Cape Province.

- Description: African elephants (Loxodonta africana) were absent from large portions of the Eastern Cape Province of South Africa for more than 100 years following widespread hunting for ivory. However, recent shifts in land use practices have resulted in the establishment of many private game reserves throughout the region. Some of these reserves have reintroduced elephants, raising management concerns because of the perceived impact that elephants can have on vegetation and the animals that rely on it for resources. The Brazilian population remains Eccritotarsus catarinensis (Carvalho), while the Peruvian population is now known as Eccritotarsus eichhorniae (Henry). The aim of this project was to investigate the mating behaviour and other behavioural traits of the two species that have resulted in reproductive isolation, and which could have led to speciation. In addition, investigations involving analysis of chemical compound compositions of the two species aimed to determine the extent to which the compounds played a role in the development and maintenance of reproductive isolation. To achieve the aims, behavioural-observation experiments were conducted in the form of no-choice, bi-choice and multi-choice tests in 1:1, 2:1 and 3:1 sex ratio assessments, both within and between species. Chemical compound compositions of E. catarinensis and E. eichhorniae were also assessed using Nuclear Magnetic Resonance (NMR), Solid-phase micro-extraction (SPME) and Gas-Chromatography Mass-Spectrometry (GC-MS) techniques. In no-choice experiments, the highest number of single and multiple copula incidences, and average total copula duration was found within species while copulation between species was much rarer. In bi-choice experiments, E. eichhorniae females and E. catarinensis males only chose to mate with their respective conspecifics, and within species copulations continued to have higher average total copula duration. In multi-choice experiments, the highest number of single and multiple copula incidences and average total copula duration was also found within species. GC-MS analysis suggested that E. catarinensis females and E. eichhorniae males have unique chemical compounds missing in their conspecifics and same sex of the other species. Further analysis suggested that E. catarinensis females and E. eichhorniae males have similar chemical compound compositions, whereas as E. eichhorniae females and E. catarinensis males have similar chemical compound compositions. These results suggest that there are behavioural differences that led to the development and maintenance of prezygotic reproductive isolation mechanisms, and that this is probably driven by pheromones in chemical compound compositions. These two species were geographically isolated in the native range and the populations have diverged to the point that they are now reproductively incompatible and therefore, distinct species. The main driver of the speciation is most likely mate recognition and attraction, as only reproductively important traits such as pheromones, genitalia, the scent glands and antennae have changed, while other traits, including host range and morphology, have remained remarkably stable. This provides evidence that differences in sexual selection in isolated populations may be important drivers of speciation and reproductive isolation in cryptic species.

Immune responses of dogs to primary challenges were poor. A second challenge or relapse was important in the development of a resistant immunity. Heterologous challenges showed that dogs which were immune to the ZIM1, QTN1 and DBN1 stocks were partially immune to the JHB1 stock. The dog that was immune to the GHT1 stock was the only one that was totally resistant to the JHB1 stock. Western blots revealed interstock antigenic variation between the JHB1 and the other four stocks. These differences were, however, not great enough to separate the stocks taxonomically.

- Description: Phylogeographic patterns of coastal organisms with different life histories and breeding strategies may reveal patterns not consistent with the current delineation of the biogeographic provinces around South Africa. The subdivision of the South African coastline into these three main climatological or biogeographic regions: namely the cool temperate west coast, the warm temperate south coast and the subtropical east coast, is based on average seawater temperatures and hydrological conditions. Genealogies of two estuarine fish species Atherina breviceps, a marine breeder, and Gilchristella aestuaria, an estuarine spawner, were reconstructed using mitochondrial DNA (mtDNA) control region sequences. The study comprised two components, an assessment of a small dataset of both fish species to compare their population structure along the South African coastline and a more comprehensive investigation of the phylogeography of G. aestuaria collected from 21 estuaries around the coast. The comparative study of A. breviceps and G. aestuaria indicate different population distribution patterns along the South African coastline. Results of the A. breviceps analysis demonstrate substantial gene flow due to the random mixing of alleles, while the comparative G. aestuaria dataset indicates a more structured population and considerably less gene flow. The G. aestuaria population demonstrates geographic separation into four groups, namely the west coast (Great Berg), Bot (south coast), Seekoei (south coast) and east coast (Bushmans, Kasouga and Cefane). Results from the larger G. aestuaria dataset indicate that the phylogeographic patterns observed during this study do not conform to existing biogeographic boundaries along the southern African coastline. The delineation identified during this study between the warm temperate and subtropical regions is further south than originally perceived and this southward extension can be ascribed to the prevailing hydrology. The life history patterns and ecology of these two estuarine fish species appears key to understanding their population structure. These factors interact with environmental characteristics such as physical oceanography and the distribution of estuaries (along the coastline) to explain the observed distribution patterns and population structure of A. breviceps and G. aestuaria.

Chemical composition of leaf essential oils of Lantana camara varieties in South Africa and their effect on the behavioural preference of Falconia intermedia

- Description: Lantana camara L. (Verbenaceae) is one the most problematic invaders in South Africa invading forest edges, sand dunes, and shorelines by forming impenetrable thickets. Lantana camara invasions degrade natural biodiversity, reduce the value of land and consequently it has been a target for biological control, over the last 50 years in South Africa. Studies that have reported on chemical profile of Lantana camara have been conducted around the world but not in South Africa. Hence, the first aim of the current study was to identify the chemical baseline of L. camara varieties in the Eastern Cape, South Africa. Recent studies have shown that feeding by one of the agents released against L. camara, Falconia intermedia (Distant) (Hemiptera: Miridae), induces anti-herbivory response through increased leaf toughness and trichome density. A preliminary study conducted also reported the production of volatile chemicals by one variety, Whitney Farm, due to feeding by the mirids. Therefore, the second aim was to determine the induced changes in chemical compounds of L. camara varieties after feeding by F. intermedia. A third aim was to determine the effect these chemical compounds have on the behaviour of F. intermedia. To identify the chemical baseline of L. camara varieties, the essential oils of four L. camara varieties (East London, Port Alfred, Whitney Farm and Heather Glen) were analysed using gas chromatography mass spectrometry (GC-MS) and that resulted to the identification of 163 constitutive and 75 induced chemicals across the varieties tested. Lantana camara varieties showed different chemical classes but were highly dominated by terpenes. A great variation in the number of constitutive chemical compounds was found in all the varieties. There were 56 constitutive chemical compounds in the Whitney Farm variety, 41 in the East London variety, 36 in the Heather Glen variety and 30 in the Port Alfred variety. The Whitney Farm variety had the highest number (22) of unique constitutive chemicals identified when compared with other varieties. This indicates the chemical distinctiveness of the Whitney Farm variety from the other varieties. In the varieties tested, there were common chemical compounds identified in constitutive and induced (discussed below) states of the plants such as caryophyllene, hexane, naphthalene, copaene and a-caryophyllene. Besides naphthalene, the majority of chemical compounds in South African L. camara varieties were similar to compounds that have been identified across the world, suggesting that they are closely related. The expression of naphthalene in these varieties may be due to changes in the chemicals expressed over evolutionary time as predicted by the Novel Weapons Hypothesis. Amongst the varieties, a great variation in chemical compounds and their concentrations was shown in the induced states of the plants. The concentration of constitutive caryophyllene ranged from (3.13 - 15.7) %, to (4.02 - 11.10) % after feeding. The concentration of constitutive hexane ranged from (6.13 - 71.19) %, to (33.3 - 75.8) % after feeding. The concentration of constitutive naphthalene ranged from (0.21 - 4.79) %, to (0.92 - 2.11) % after feeding. The concentration of constitutive copaene ranged from (0.57 - 1.57) %, to (1.20 - 2.72) %. Lastly, the concentration of constitutive a-caryophyllene ranged from (1.18 - 9.03) %, to (0.78 - 5.48) % after feeding. The changes in chemical concentrations in lantana varieties indicated that feeding by the mirid on L. camara varieties causes an induction by either reducing or increasing the chemical concentrations. To determine the effect of the identified compounds on the behaviour of F. intermedia adults, olfactometer bioassays were conducted using a Y-tube technique. A significantly higher proportion of F. intermedia were attracted to undamaged leaves over damaged leaves and purified air. Undamaged leaves attracted 52 % of F. intermedia from the East London variety, 62.5 % from the Port Alfred variety, 56 % from the Whitney Farm variety, 58 % from the Lyndhurst variety and 54.5 % from the Heather Glen variety in dual choice trials versus damaged leaves. Furthermore, a significantly higher proportion of F. intermedia were attracted to damaged leaves over purified air. Damaged leaves attracted 67 % of F. intermedia from the East London variety, 67 % from the Port Alfred variety, 65.9 % from the Whitney Farm variety, 65.3 % from the Heather Glen variety and 64.5 % from the Lyndhurst variety. Olfactometer bioassays were also conducted using purified standard compounds of four chemical compounds identified from essential oils, hexane was used as a positive control as it is reported to be an insect attractant in literature. Hexane was highly attractive to the mirids compared to three standard compounds caryophyllene, caryophyllene oxide and naphthalene at the rate of 80 %, 73 % and 80 %, respectively. The standard compounds tested against F. intermedia are major compounds contained by L. camara varieties and they have proven to have a repellent effect. This may indicate that after feeding by F. intermedia, the major compounds expressed by the plant varieties repel F. intermedia contributing to the invasiveness of this weed. The increased expression of hexane and caryophyllene after feeding may also indicate increased attraction to some insects, opening up the potential for third trophic level interactions in varieties where this is the case. This is the first study on the chemical composition of essential oils of L. camara in South Africa. Therefore, we recommend that where appropriate chemical profile studies of the invasive alien plants should be considered during host specificity testing, and the vital role of chemical compounds on agent-weed interactions must be taken into consideration with other factors before and after the biological control agents are released.

There were 56 constitutive chemical compounds in the Whitney Farm variety, 41 in the East London variety, 36 in the Heather Glen variety and 30 in the Port Alfred variety. The Whitney Farm variety had the highest number (22) of unique constitutive chemicals identified when compared with other varieties. This indicates the chemical distinctiveness of the Whitney Farm variety from the other varieties. In the varieties tested, there were common chemical compounds identified in constitutive and induced (discussed below) states of the plants such as caryophyllene, hexane, naphthalene, copaene and a-caryophyllene. Besides naphthalene, the majority of chemical compounds in South African L. camara varieties were similar to compounds that have been identified across the world, suggesting that they are closely related. The expression of naphthalene in these varieties may be due to changes in the chemicals expressed over evolutionary time as predicted by the Novel Weapons Hypothesis. Amongst the varieties, a great variation in chemical compounds and their concentrations was shown in the induced states of the plants. The concentration of constitutive caryophyllene ranged from (3.13 - 15.7) %, to (4.02 - 11.10) % after feeding. The concentration of constitutive hexane ranged from (6.13 - 71.19) %, to (33.3 - 75.8) % after feeding. The concentration of constitutive naphthalene ranged from (0.21 - 4.79) %, to (0.92 - 2.11) % after feeding. The concentration of constitutive copaene ranged from (0.57 - 1.57) %, to (1.20 - 2.72) %. Lastly, the concentration of constitutive a-caryophyllene ranged from (1.18 - 9.03) %, to (0.78 - 5.48) % after feeding. The changes in chemical concentrations in lantana varieties indicated that feeding by the mirid on L. camara varieties causes an induction by either reducing or increasing the chemical concentrations. To determine the effect of the identified compounds on the behaviour of F. intermedia adults, olfactometer bioassays were conducted using a Y-tube technique. A significantly higher proportion of F. intermedia were attracted to undamaged leaves over damaged leaves and purified air. Undamaged leaves attracted 52 % of F. intermedia from the East London variety, 62.5 % from the Port Alfred variety, 56 % from the Whitney Farm variety, 58 % from the Lyndhurst variety and 54.5 % from the Heather Glen variety in dual choice trials versus damaged leaves. Furthermore, a significantly higher proportion of F. intermedia were attracted to damaged leaves over purified air. Damaged leaves attracted 67 % of F. intermedia from the East London variety, 67 % from the Port Alfred variety, 65.9 % from the Whitney Farm variety, 65.3 % from the Heather Glen variety and 64.5 % from the Lyndhurst variety. Olfactometer bioassays were also conducted using purified standard compounds of four chemical compounds identified from essential oils, hexane was used as a positive control as it is reported to be an insect attractant in literature. Hexane was highly attractive to the mirids compared to three standard compounds caryophyllene, caryophyllene oxide and naphthalene at the rate of 80 %, 73 % and 80 %, respectively. The standard compounds tested against F. intermedia are major compounds contained by L. camara varieties and they have proven to have a repellent effect. This may indicate that after feeding by F. intermedia, the major compounds expressed by the plant varieties repel F. intermedia contributing to the invasiveness of this weed. The increased expression of hexane and caryophyllene after feeding may also indicate increased attraction to some insects, opening up the potential for third trophic level interactions in varieties where this is the case. This is the first study on the chemical composition of essential oils of L. camara in South Africa. Therefore, we recommend that where appropriate chemical profile studies of the invasive alien plants should be considered during host specificity testing, and the vital role of chemical compounds on agent-weed interactions must be taken into consideration with other factors before and after the biological control agents are released.

- Description: During the last 25 years, the Eastern Cape Province of South Africa has seen the establishment of many small to medium sized (≤ 440km2) game reserves. These reserves have reintroduced many of the larger indigenous wildlife that had been extirpated by the early 20th century. As such, these reserves and wildlife introductions have created many research opportunities, including investigations on the ecology of reintroduced carnivores in the Thicket biome. Brown hyaenas (Hyaena brunnea) are one of the large carnivore species that have been reintroduced into the area. As these animals have predominantly been studied in more open, arid systems, their reintroduction has provided an excellent opportunity to study the species in an alternate natural habitat. Information gathered from such investigations adds to our knowledge of the species and also provides information for the management of brown hyaenas within small, enclosed reserves. Data were collected over the period of one calendaryear, from February 2013 to February 2014 at Kwandwe Private Game Reserve in the Eastern Cape, South Africa. Brown hyaena population estimates were calculated using capture-recapture methods from individually identifiable images captured during a three month camera trapping survey. Images of brown hyaenas were separated into left- and right-side profiles. Twenty-eight individuals were positively identified from left-side images and 27 from right-side images. Non-spatial and spatially explicit capture-recapture analyses were both run in the program DENSITY 5.0. Density estimates ranged from 14 to 20 individuals/100km2 (equivalent to a total abundance of 26-37 individuals) depending on the method used. Despite the range of estimates, all are considerably higher than in other areaswhere densities have been calculated. Satellite/GPS collars were fitted to three individual brown hyaenas (two males and one female) to measure their home range size and use of space. Home range size was calculated using two different methods, Kernel utilisation distributions and Brownian bridges. Home range estimates were similar using both methods; however Brownian bridge methods appeared to exaggerate the use of space by individuals. Kernel home range sizes for the three individuals ranged between 42.62km2 and 79.88km2. These estimates are considerably smaller than previous findings from other parts of Africa and suggest that sufficient resources may be available within this enclosed system. The results from this study suggest that brown hyaenas are successful generalists in this enclosed system and are able to persist at high densities and occupy relatively small home ranges. This information is important for the managers of small reserves who wish to reintroduce brown hyaenas. Should brown hyaenas be introduced into reserves in the Thicket biome with sufficient resources, the numbers are likely to increase rapidly. If high numbers are not desired then preventative measures (e.g. contraception) should be investigated before release.
