- Description: Baylis-Hillman reactions of 2,2’-dithiodibenzaldehyde with the acyclic alkenes, methyl vinyl ketone (MVK) and methyl acrylate have afforded the thiochromene derivatives in moderate yields, and this approach has been extended to the use of the cyclic alkenes, 2-cyclohexenone and 2-cyclopentenone to afford the tricyclic analogues. In all cases, reduction of the disulphide link and intramolecular cyclisation occurred in situ, and a preliminary kinetic study of this reaction using the acyclic substrates MVK and methyl acrylate was undertaken with the aim of elucidating the mechanism involved. The results obtained showed that the consumption of both 2,2’-dithiodibenzaldehyde and MVK and/or methyl acrylate followed 1st-order kinetics during the initial stages of the reaction, but then deviated from 1st-order linearity. The reaction with methyl acrylate was much slower than with MVK, and the kinetic data indicates the mechanism to be more complex than anticipated. Conjugate addition reactions of methyl acrylate-derived 2-nitrobenzaldehyde Baylis-Hillman adducts with the amines, piperidine and benzylamine, afforded a range of conjugate addition products as diastereomeric mixtures in excellent yield (80-100%). Catalytic hydrogenation of the conjugate addition products using a Pd-C catalyst in ethanol, has afforded the corresponding, novel 3-amino-2-quinolone derivatives in lower yield (22-37%).The application of [superscript 13]C NMR prediction programmes to selected compounds synthesized in this study has revealed reasonable correlations between the experimental and predicted values.

- Description: Prevention of the accumulation of undesirable biological material i.e. biofouling upon a solid surface requires the use of antifouling systems. The solid surface may be a contact lens, an off shore oil rig or a living organism. When chemicals are employed as a mechanism of defense against biofouling, the agents involved are known as antifouling agents. Marine algae must protect themselves from fouling organisms and it is thought that one of the mechanisms used by these organisms is the production of secondary metabolites with an array of biological activities. In vitro studies have shown numerous compounds isolated from marine algae to possess antibacterial, antifungal and antimacrofouling activity. The aim of this study was to evaluate the secondary metabolite extracts of selected Southern African marine macro-algae as a potential source of compounds that inhibit biofilm formation and that could be used as antifouling agents. In this project, marine macro-algae were collected from various sites along the South African coastline. Their extracts were screened for antimicrobial activity against four ubiquitous microorganisms, Staphylococcus aureus, Klebsiella pneumoniae, Mycobacterium aurm and Candida albicans. Results of screening assays guided the fractionation of two Rhodophyta, Plocamium corallorhiza and Laurencia flexuosa. The algae were fractionated using silica gel column chromatography and compounds were isolated by semi-preparative normal phase HPLC. Compound characterization was performed using UV, IR and advanced one- and two-dimensional NMR (¹H, ¹³C NMR, COSY, HSQC, HMBC and NOESY) spectroscopy and mass spectrometry. Ten halogenated monoterpenes including four members of the small class of halogenated monoterpene aldehydes were isolated from extracts of P. corallorhiza. The compounds isolated included the known compounds 3,4,6,7-tetrachloro-3,7-dimethyl-1-octene; 4,6-dibromo-1, 1-dichloro-3,7 -dimethyl-2E,7 octadiene; 4,8-d ibromo-1,1,7 -trichloro-3, 7-dimethyl-2,5Eoctadiene;1 ,4,8-tribromo-3, 7 -dichloro-3,7-dimethyl-1 E,5E-octadiene; 8-bremo-6, 7-dichloro-3,7-dimethyl-octa-2E,4E-dienal; 4-Bromo-8-chloro-3,7-dimethyl-octa-2E,6E-dienal; 4,6- Dibromo-3,7-dimethyl-octa-2E,7-dienal; 2,4-dichloro-1-(2-chlorovinyl)-1-methyl-5-methylidene-cyclohexane and two new metabolites 4,8-chloro-3,7-dimethyl-2Z,4,6Z-octatrien-1-al and Compound 3.47. Methodology was developed for the chemical derivatization and mass spectrometric analysis of the aldehydic compounds, The aldehyde trapping reagent 0-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine hydrochloride was used to derivatize the molecules, stabilizing them and allowing for their complete characterization. From Laurencia flexuosa a new cuparene sesquiterpene 4-bremo-2-(5-hydroxy-1,2,2- trimethylcyclopent-3-enyl)-5-methylphenol was isolated along with two geometric isomers of the vinyl acetylene bromofucin , An halogenated monoterpene 3S*,4R*-1-bromo-3,4,8-trichloro-9-dichloromethyl-1-E,5-E,7-Z-octatriene was also isolated but was suspected to be a contaminant and an investigation into its biological source revealed that it originated from Plocamium suhrii. A third alga, Martensia elegans was extracted based on published reports of antimicrobial compounds in related species. A new a-alkyl malate derivative was isolated and characterized. Selected compounds isolated during the course of the study were employed in preliminary assays that tested their ability to inhibit biofilm formation by Pseudomonas aeruginosa. The halogenated monoterpenes isolated from the Plocamium species were the only active compounds. 3S*,4R*-1-bromo-3,4,S-trichloro-g-dichloromethyl-1-E,5-E,7-octatriene from P. suhrii inhibited biofilm formation through antibacterial activity on planktonic cells but could not prevent biofilm formation when employed as a film on the surface of microtitre plate wells. 1,4,8-tribromo-3,7-dichloro-3,7-dimethyl-1E,5E-octadiene and 4,6-dibromo-1,1-dichloro-3,7-dimethyl-2E,7-octadiene inhibited biofilm formation when applied as a film to the microtitre plate wells but had no significant antibacterial activity. - Description: Background: In recent years the incidence of Attention Deficit/Hyperactivity Disorder (ADHD) has increased. Objectives: The primary aim of the study was to determine the treatment of children and adolescents diagnosed with ADHD in South Africa and more specifically the Eastern Cape. Methods: The study consisted of two parts, a retrospective drug utilisation study and a questionnaire survey. The drug utilisation study was conducted on chronic medication records of a private sector medical scheme administrator and contained records from 1994 to 2006. A total of 42 250 records representing 8 274 patients and 4 001 families, were analysed. The questionnaire survey was conducted in the Eastern Cape. The questionnaires were distributed by responsible pharmacists to parents of children receiving medication for the treatment of ADHD from his/her pharmacy. Fifty one completed questionnaires were analysed. Results: The average age of patients in the drug utilisation study were 14.28 (SD = 5.50) years. Males represented 77.37 percent of patients. Ritalin® 10 mg was the most frequently prescribed methylphenidate-containing product, followed by Ritalin® LA 20 mg. The two most frequently prescribed active ingredients apart from methylphenidate were fluticasone and budesonide. The two most common comorbid conditions were allergic rhinitis and asthma. The average age of patients in the questionnaire survey was 10.26 (SD = 2.51) years. Males represented 73.58 percent of patients. A third of patients had relatives previously diagnosed with ADHD. Ritalin® 10 mg was the most frequently prescribed methylphenidate-containing product. Conclusion: ADHD is a complex life-long disorder. It does not only affect the child diagnosed with the disorder, but also has effects on the family, schooling system and peer relationship. Therefore, more studies regarding the incidence and treatment of ADHD need to be conducted in South Africa.

Isolation, purification and characterization of a 'factor' from Fusarium oxysporum responsible for platinum nanoparticle formation

- Description: Nanoparticles are microscopic particles in the nanometre range of between 1-100 nm. A wide variety of metal nanoparticles have been found to be produced by prokaryotic and eukaryotic organisms including several fungal species, when exposed to solutions containing metal salts. Previous studies have suggested that this bioreduction of metal particles may occur via an active reductase/hydrogenase enzyme process where H2 is the electron donor and positively charged platinum species act as the electron acceptors becoming reduced to a neutral metal nanoparticle. In view of this on going research, the current study investigated the “factors” in the fungus Fusarium oxysporum which were responsible for platinum nanoparticle formation. The fungus F.oxysporum was used in this study as it has been previously shown to produce a variety of nanoparticles including gold and silver. During exposure of the biomass to H2PtCl6 the initial response to the platinum salts was metal internalisation and subsequent reduction of H2PtCI6 to produce platinum nanoparticles. The observed localization and distribution of platinum precipitates provided some evidence for a hydrogenase mediated bioreduction of platinum salts to produce nanoparticles. Factors secreted by the fungus into the extracellular fluids, were shown to be responsible for platinum nanoparticle formation. From the identification, purification and characterisation studies it was concluded that a hydrogenase and other “factors” were responsible for platinum nanoparticle formation in F.oxysporum. Purification of the hydrogenase by freeze-drying and Sephacryl S200 size exclusion- ion exchange chromatography revealed the enzyme to be a dimer with a 29.4 and 44.5 kDa when analysed by a 10 % SDS-PAGE. Characterisation of the enzyme revealed optimal activity at a pH of 7.5 and temperature of 38 °C while it exhibited a poor thermal stability with a half life of 36 minutes. The kinetic parameters Vmax and Km were 3.16 U ml-1 and 3.64 mM respectively. The purified hydrogenase was used in subsequent experiments for the reduction of platinum salts, H2PtCl6 and PtCl2. the results indicated an over 90 % reduction of the platinum salts and TEM micrographs indicated the production of platinum nanoparticles under the various experimental conditions.

An adaptive user interface model for contact centres

- Description: Contact centres (CC), are the primary interaction point between a company and its customers and these are rapidly expanding in terms of both workforce and economic scope. An important challenge for today's CC solutions is to increase the speed at which CCAs retrieve information to answer customer queries. CCAs, however, differ in their ability to respond to these queries and do not interact with the computer user interface (UI) in the same way as they each have different capabilities, experience and expertise. Studies have provided empirical support that user performance can be increased when the computer UI characteristics match the user skill level. Adaptive user interfaces (AUIs) are the key to creating personalised systems. Their sole task is to provide an interface most suitable to users' needs whilst facilitating the users' varying skill levels. The aim of this research was to develop an AUI model for CCs to support and improve the expertise level of CCAs. A literature review of CCs, user expertise, AUIs and existing AUI models resulted in the proposal of an AUI model for CCs. The proposed AUI model was described in terms of its architecture, component-level and interface design. An AUI prototype was developed as a proof-of-concept of the proposed AUI model. A literature review on existing AUI evaluation approaches resulted in an evaluation strategy for the proposed AUI model. The AUI prototype was evaluated according to the evaluation strategy that was identified. User testing incorporating eye-tracking and a post-test questionnaire was used to determine the usefulness and usability of the AUI prototype. Significant results were found with regards to user satisfaction ratings, the learnability of the AUI prototype and its effectiveness. This dissertation makes an important contribution in the design of an AUI model that supports and improves the expertise level of CCAs. The model could be used to assist the development of CC applications incorporating AUIs. Future research is however needed to evaluate the effect of the proposed AUI model in a larger CC environment.

- Description: Contact centres (CC), are the primary interaction point between a company and its customers and these are rapidly expanding in terms of both workforce and economic scope. An important challenge for today's CC solutions is to increase the speed at which CCAs retrieve information to answer customer queries. CCAs, however, differ in their ability to respond to these queries and do not interact with the computer user interface (UI) in the same way as they each have different capabilities, experience and expertise. Studies have provided empirical support that user performance can be increased when the computer UI characteristics match the user skill level. Adaptive user interfaces (AUIs) are the key to creating personalised systems. Their sole task is to provide an interface most suitable to users' needs whilst facilitating the users' varying skill levels. The aim of this research was to develop an AUI model for CCs to support and improve the expertise level of CCAs. A literature review of CCs, user expertise, AUIs and existing AUI models resulted in the proposal of an AUI model for CCs. The proposed AUI model was described in terms of its architecture, component-level and interface design. An AUI prototype was developed as a proof-of-concept of the proposed AUI model. A literature review on existing AUI evaluation approaches resulted in an evaluation strategy for the proposed AUI model. The AUI prototype was evaluated according to the evaluation strategy that was identified. User testing incorporating eye-tracking and a post-test questionnaire was used to determine the usefulness and usability of the AUI prototype. Significant results were found with regards to user satisfaction ratings, the learnability of the AUI prototype and its effectiveness. This dissertation makes an important contribution in the design of an AUI model that supports and improves the expertise level of CCAs. The model could be used to assist the development of CC applications incorporating AUIs. Future research is however needed to evaluate the effect of the proposed AUI model in a larger CC environment.

The mean concentrations of metals in the sediments(mg·kg⁻¹) showed ranges of 0.28 – 2.31 for Cd, 1.26 – 6.24 Co, 0.69 – 6.93 for Cu, 2119 – 14912 for Fe, 2.29 –14.01 for Ni, 4.81 – 22.20 for Pb and 5.77 – 21.75 for Zn. Mean normalized enrichment factors ranged between 0.75 – 6.19 for Cd, 0.53 – 2.71 for Co, 0.22 – 0.84 for Cu, 0.30 – 1.87 for Ni, 0.99 – 3.17 for Pb and 0.14 – 0.98 for Zn. All nine estuaries had average enrichment factors of greater than 1 for Cd. In general there was no enrichment of Cu and Zn in the sediments of any of the estuaries included in this study (EFs < 1). The Kariega, East Kleinemonde, West Kleinemonde, Riet and Great Fish Estuaries showed some degree of enrichment for Co (1 < EF < 4), Ni (1 < EF < 2) and Pb (1 < EF < 4), while the Mpekweni, Kasouga, Boknes and Kowie Estuaries were unenriched with these metals (EF < 1). Enrichment factors for Cd, Co and Pb typically followed the development gradient along the estuaries, suggesting anthropogenic enrichment. The concentrations of Cd and Pb in the water of the nine estuaries were also determined. The average concentrations of Cd and Pb in the water (μg·ℓ⁻¹) ranged between 0.05 – 3.32 and 0.75 – 34.13 respectively. On average the concentrations of Cd and Pb in the water of all the estuaries were below the South African recommended water quality guidelines for coastal marine waters. Variations in metal concentrations associated with changes in hydrology (wet vs. dry season) were determined in the water and sediment of the Kariega, East Kleinemonde and Riet Estuaries. Cobalt, Pb and Ni enrichment in the Kariega Estuary sediment was significantly higher during the dry season, and the mean concentrations of Pb and Cd in the water column were 19-fold and 66-fold higher in the dry season. The elevated concentration of metals during the dry season could be related to accumulation of diffuse pollution from human activities within the catchment area. Conversely, inflow of fresh water into the estuary had the net effect of reducing the concentration and enrichment of these metals within the Kariega Estuary due to scouring and outflow of estuarine water and sediment into the marine environment. The temporal variations in metal concentrations and enrichment factors were less pronounced in the temporary open-closed estuaries than the permanently open Kariega Estuary. The observed trend can probably be related to the low anthropogenic impact within the catchment areas of these systems, and the relatively smaller size of the catchments. Significant spatial variations existed in metal enrichment in the sediment of both the East Kleinemonde and Riet estuaries, with the highest degrees of enrichment occurring in the sediments from the marine environment and lower reaches.

For more than a decade, the wild abalone stock of South Africa has suffered decline due to over-exploitation and illegal activities such as poaching. Prior to 1970, no regulations were in place concerning the annual landings. As a result the fishery was exploited as if it were an infinite resource. It is this initial uncontrolled harvesting (regardless of age) and poaching that has driven the abalone resource decline. Due to the slow growth rate exhibited by abalone as a species, natural replenishment of wild stock following exploitation and poaching was far below the rate of exploitation of this resource. Studies on the growth of abalone have mainly been conducted under laboratory conditions. The purpose of this study was to measure the growth of abalone, fed different diets, under commercial culture conditions. Three food types were used namely; commercial pellets, seaweed (Ulva spp.) and dried kelp bars (Ecklonia maxima). Four diets were obtained from the three food types namely; combination of commercial pellets and seaweed (Diet A), commercial pellets only (Diet B), seaweed only (Diet C) and dried kelp bars only (Diet D). The food types used in this study represent both artificial (Commercial pellets) and natural feeds (seaweed and kelp) used in commercial abalone culture. The growth of two cohorts (40-50 mm and 50-60 mm) was followed over a 426 day period, with data for the first 183 days being used for statistical analysis to determine performance of a given diet. The best growth rates were found in abalone fed Diet A (40-50 mm: 2.64 mm.month-1; 50-60 mm 2.78: mm.month-1) and B (40-50 mm: 2.20 mm.month-1; 50-60 mm: 2.35: mm.month-1). These (Diets A and B) gave higher growth rates when compared to Diets C and D (natural diets), whose growth rates ranged between 0.50 mm.month-1 and 1.71 mm.month-1 for both cohorts. Also observed in this study was that, the mixture of formulated diet and seaweed gave better growth than formulated diet given exclusively.

A total of 11 389 navel oranges were collected from various orchards in the Addo/Kirkwood area, and false codling moth larvae infested 36.09% of the fruit. A single parasitoid species, A. bishopi, was reared from these larvae. In 2006 the highest parasitism rate, 11.43%, was recorded in May and in 2007, the highest parasitism rate, 13.27%, was in April. Agathis bishopi parasitizes larvae in instars 2 and 3, possibly due to the accessibility of these younger instars to the female parasitoid and possibly due to the length of the life cycle of this koinobiont. Second instar hosts yielded the highest number of parasitoids, and there was no emergence of parasitoids from fifth instar larvae. Females of A. bishopi live for 18.5 days (n = 20; S.E. = 3.1) and males for 8.25 days (n = 20; S.E. = 1.23). Females produce an average of 23 offspring in a lifetime, while female false codling moths produce about 800 eggs each. A high number of parasitoids will be required per hectare to reduce the population of false codling moth. Captive rearing of A. bishopi proved difficult due to viral and fungal contamination. Agathis bishopi has potential for use in an integrated pest management programme once the hurdle of mass-rearing has been overcome.

- Description: The main aim of this study was to assess the “top-down” and “bottom-up” control of bacterial production in the small temporarily open/closed East Kleinemonde Estuary, situated on the south-eastern coastline of southern Africa. Spatial and temporal patterns in bacterial abundance, biomass and production and the importance of abiotic and biotic factors were investigated over the period May 2006 to April 2007. The trophic interactions between bacteria, phytoplankton, nanoflagellates (< 20 μm), microzooplankton (< 200 μm) and mesozooplankton (< 2 000 μm) were investigated during winter and summer. Bacterial abundance, biomass and production ranged between 1.00 × 10⁹ and 4.93 × 10⁹ cells 1⁻¹, 32.4 and 109 μg C 1⁻¹ and 0.01 and 1.99 μg C 1⁻¹ h⁻¹, respectively. With a few exceptions there were no spatial patterns in the values. Bacterial abundance, biomass and production, however, demonstrated a distinct temporal pattern with the lowest values consistently recorded during the winter months. Nanoflagellate and bacterial abundances were significantly correlated to one another (lower reaches: r = 0.818, p < 0.001; middle reaches: r = 0.628, p < 0.001; upper reaches: r = 0.484, p < 0.05) suggesting a strong predator-prey relationship. The frequency of visibly infected bacterial cells and the mean number of virus particles within each bacterial cell during this study demonstrated no temporal or spatial patterns and ranged from 0.5 to 6.1 % and 12.0 to 37.5 virus particles per bacterium, respectively. Viral infection and lysis was thus a constant source of bacterial mortality throughout the year. The estimated percentage of bacterial production removed by viral lysis ranged between 7.8 and 88.9% of the total which suggests that viral lysis represented a very important source of bacterial mortality during this study. The biological interactions between the selected components of the plankton community demonstrated that among the heterotrophic components of the plankton, the nanoflagellates were identified as the most important consumers of bacteria and small phytoplankton cells (< 20 μm). In the presence of microzooplankton the impact of the nanoflagellates on both the bacteria and phytoplankton was reduced, indicating that larger heterotrophs were preying upon the nanoflagellates. Mesozooplankton, however, appeared to exert the greatest impact on nanoflagellates. In the cascading experiments, the data suggest that mesozooplankton consume nanoflagellates, which resulted in a decrease in the predation impact of these organisms on the bacteria. This result is consistent with predator-prey cascades. The presence of the larger heterotrophs therefore, mediates the interactions between the primary bacterivores, the nanoflagellates, and the bacteria within the temporarily open/closed East Kleinemonde Estuary.

- Description: The main aim of this study was to assess the “top-down” and “bottom-up” control of bacterial production in the small temporarily open/closed East Kleinemonde Estuary, situated on the south-eastern coastline of southern Africa. Spatial and temporal patterns in bacterial abundance, biomass and production and the importance of abiotic and biotic factors were investigated over the period May 2006 to April 2007. The trophic interactions between bacteria, phytoplankton, nanoflagellates (< 20 μm), microzooplankton (< 200 μm) and mesozooplankton (< 2 000 μm) were investigated during winter and summer. Bacterial abundance, biomass and production ranged between 1.00 × 10⁹ and 4.93 × 10⁹ cells 1⁻¹, 32.4 and 109 μg C 1⁻¹ and 0.01 and 1.99 μg C 1⁻¹ h⁻¹, respectively. With a few exceptions there were no spatial patterns in the values. Bacterial abundance, biomass and production, however, demonstrated a distinct temporal pattern with the lowest values consistently recorded during the winter months. Nanoflagellate and bacterial abundances were significantly correlated to one another (lower reaches: r = 0.818, p < 0.001; middle reaches: r = 0.628, p < 0.001; upper reaches: r = 0.484, p < 0.05) suggesting a strong predator-prey relationship. The frequency of visibly infected bacterial cells and the mean number of virus particles within each bacterial cell during this study demonstrated no temporal or spatial patterns and ranged from 0.5 to 6.1 % and 12.0 to 37.5 virus particles per bacterium, respectively. Viral infection and lysis was thus a constant source of bacterial mortality throughout the year. The estimated percentage of bacterial production removed by viral lysis ranged between 7.8 and 88.9% of the total which suggests that viral lysis represented a very important source of bacterial mortality during this study. The biological interactions between the selected components of the plankton community demonstrated that among the heterotrophic components of the plankton, the nanoflagellates were identified as the most important consumers of bacteria and small phytoplankton cells (< 20 μm). In the presence of microzooplankton the impact of the nanoflagellates on both the bacteria and phytoplankton was reduced, indicating that larger heterotrophs were preying upon the nanoflagellates. Mesozooplankton, however, appeared to exert the greatest impact on nanoflagellates. In the cascading experiments, the data suggest that mesozooplankton consume nanoflagellates, which resulted in a decrease in the predation impact of these organisms on the bacteria. This result is consistent with predator-prey cascades. The presence of the larger heterotrophs therefore, mediates the interactions between the primary bacterivores, the nanoflagellates, and the bacteria within the temporarily open/closed East Kleinemonde Estuary.

Finally, an enhanced framework is proposed, describing RFID adoption barriers within the South African retail sector. - Description: The greater photosynthetic nitrogen use efficiency (PNUE) of C4 compared with C3 plants may explain the relative success of C4 grasses in nutrient poor environments. This study compared the responses in photosynthetic parameters, leaf nitrogen and biomass allocation between the C3 and C4 subspecies of Alloteropsis semialata supplied soil nitrogen at three levels. Photosynthesis was assessed by means of CO2 response curves and the leaf nitrogen content assayed. Plants were destructively harvested, leaf areas determined and the dry biomass of functional plant components was measured. Results confirmed that the higher PNUE of C4 plants allowed them to accumulate more biomass than C3 plants at the high nitrogen level, despite smaller leaf areas. The greater productivity of C4 plants enabled them to invest more in storage and sexual reproduction than in leaves when compared to the C3 plants. In contrast the C3 plants invested biomass in less efficient and more nitrogen demanding leaves and bigger root systems. PNUE and photosynthetic rates were not significantly affected by nitrogen-limitation in either subspecies and the major response was a decrease in biomass accumulation and an increase in biomass allocation to roots. This altered root to shoot ratio was accompanied by a lowered allocation to sexual reproduction in the C4 subspecies, but an unaltered allocation to leaves, while in the C3 subspecies there was a decrease in leaf allocation. In a further experiment, the C4 subspecies was supplied three levels of nitrogen provided as nitrate, or alternatively as ammonium plus nitrate, and leaves were excised to within 5 cm of the ground at the start of treatment. Prior to flowering, photosynthesis was assessed by means of CO2 response curves and the plants were destructively harvested. Leaf areas and the dry biomass of functional plant components were determined, and at levels of nitrogen supply higher than those found in savanna soils the rate of photosynthesis was increased. Leaf re-growth was reduced by severe nitrogen limitation and co-provision of nitrate and ammonium had no significant effect other than increased tillering. Both subspecies of Alloteropsis semialata are adapted to nutrient poor environments and maintain photosynthetic rates by reducing leaf area. The C4 subspecies is likely to show greater resilience in disturbance-prone environments by exploiting its higher PNUE to allocate greater resources to storage and sexual reproduction, while the C3 subspecies is usually found in environments with closed canopies which favour vegetative growth, and allocate greater resources to leaves and roots.

- Description: The aims of this study were to determine the potential use of biomarkers at multiple levels of biological organisation together with a fish community bioindicator to assess the estuarine health status of three temporarily open/closed estuaries. The estuaries investigated were the East Kleinemonde (EK), Old Woman's (OW) and Mtana (MTN), all of which are situated in the Eastern Cape Province. Three biomarkers, the acetylcholinesterase (AChE) assay, lipid peroxidation (LPx) assay, liver histopathology and a condition factor were used to determine sub-organism health and one bioindicator, the Estuarine Fish Community Index (EFCI), was used as a bioindicator of community health. The estuarine-dependent marine species Rhabdosargus holubi was selected as an indicator species for the sub-organism level analyses. The results from the community analyses indicated that the EK and OW estuaries were in 'good' condition, while the MTN was found to be in 'moderate' condition. Histological analyses revealed that R. holubi from all three estuaries showed signs of pathological changes to the liver, with the fish from the MTN eliciting the highest occurrence of these changes. The LPx assay found that R. holubi from both the OW and MTN showed signs of oxidative damage in the liver tissue, but those from the EK did not appear to be affected. The AChE assay showed that only the fish from the OW had been affected by anticholinesterase compounds. A laboratory study was undertaken using R. holubi as a positive control for the AChE and LPx assay. The fish were exposed to 3 μg/L chlorpyrifos, a known cholinesterase inhibitor, for six hours and their tissues were examined for changes to LPx levels and AChE activities. AChE activity was significantly inhibited (Mann Whitney U test, Z = 3.65, n = 38, P < 0.001) by the exposure, but LPx levels were not significantly affected. A composite index incorporating the biomarkers at different biological levels of organisation was developed. The index was designed to assist managers and scientists to determine whether the ichthyofauna of a system was being affected by environmental stressors and what management interventions could be undertaken to ameliorate the water quality in an estuary. The index was applied to the three estuaries investigated during the present study and both the OW and MTN were assessed to be in need of immediate management intervention. The fish in the OW were found to be stressed at all the sub-organism levels measured and the reason for this was hypothesised to be as a result of golf course activities in this adjacent estuary. A number of management actions are proposed to reduce the sub-organism stress observed in the fish from the OW. The livers of fish from the MTN were shown to be under stress; however the causative agent of this stress was unknown because there is no formal development in the MTN catchment. However, a possible contaminant source is proposed and management interventions to alleviate the stress on the biota of the MTN are suggested. The EK does not require immediate management intervention, however, continuous routine monitoring is recommended to ensure that conditions do not deteriorate. Shortcomings of the index were outlined and a number of suggestions were made in terms of other measures of biological health which could be incorporated into the index.

This research proposes a novel mechanism which is able to overcome hidden node problems and provide fair bandwidth sharing among nodes on ad-hoc 802.11 networks, and can be implemented on existing network devices. The scheme uses TDM (time division multiplexing) with slot reservation. A distributed beacon packet latency measurement mechanism is used to achieve node synchronisation. The distributed nature of the mechanism makes it applicable to ad-hoc 802.11 networks, which can either grow or fragment dynamically.

Biological intervention can range from application of biological products to enhance the nutritional value of crops or to control plant pathogens. Biostart, a biological product that demonstrated growth enhancement when applied in lettuce crops is currently in the market. The product is comprised of a consortium of bacterial isolates (Bacillus licheniformis, Brevibacillus laterosporus and Bacillus laterosporus) but the contribution of the individual isolates to growth enhancement had not been elucidated. Green house experiments on lettuce seedlings with individual and mixed treatments were commissioned to determine such contribution. There was either no or marginal growth enhancement observed in the experiments. The results showed that the product was effective as a consortium and not as individual isolates. Further isolation and screening for potential Bacilli with antifungal properties was undertaken. An isolate identified as Bacillus subtilis that demonstrated inhibition against a wide spectrum of fungi, and especially the phytopathogenic Verticillium dahliae and Fusarium oxysporum, was successfully identified. The isolate was cryo-preserved and cultivated to significant levels at bench scale. A characterized comparison of different putative products with known systematic fungicide showed potential application even of heat treated products. The product showed control V. dahliae when tested in green houses with potatoes and tomatoes as test crops. This isolate has been targeted for further development as a biological control product.

An isolate identified as Bacillus subtilis that demonstrated inhibition against a wide spectrum of fungi, and especially the phytopathogenic Verticillium dahliae and Fusarium oxysporum, was successfully identified. The isolate was cryo-preserved and cultivated to significant levels at bench scale. A characterized comparison of different putative products with known systematic fungicide showed potential application even of heat treated products. The product showed control V. dahliae when tested in green houses with potatoes and tomatoes as test crops. This isolate has been targeted for further development as a biological control product.

The National Department of Public Works (NDPW) became the focus of the study because it has the largest property portfolio in the southern hemisphere. It should be the leader in FM. The collected primary data (being quantitative) and secondary information provided the necessary basis to understand the application of FM in the NDPW. The study revealed shortcomings which are contributing to the problems outlined: namely, that no senior manager has been appointed to manage the portfolio and assist top management in decision-making; that neither a policy nor a FM framework are in place to guide the FM portfolio; that there is a lack of knowledge about the discipline, particularly by management and that there is no computer-aided FM in the entire department, let alone a FM helpdesk. FM is the coordination of workplace, people, physical infrastructure, processes and technology in order for an organization to meet its objectives. It is a wide field encompassing models that tend to differ considerably from one organization to another as it has to respond to the particular needs of each organization. Cellulosomal composition may vary according to the microbe’s nutritional requirement and allows for the anaerobic degradation of complex substrates. The complex substrates of interest in this research study were sugarcane bagasse and pineapple fibre waste, as they represent two important lignocellulosic, South African agricultural crops. The effective degradation of complex plant biomass wastes may present a valuable source of renewable compounds for the production of a variety of biofuels, for example bioethanol, and a variety of biocomposites of industrial importance. The identification of renewable energy sources for the production of biofuels is becoming increasingly important, as a result of the rapid depletion of the fossil fuels that are traditionally used as energy sources. An effective means of completely degrading lignocellulose biomass still remains elusive due to the complex heterogeneity of the substrate structure, and the fact that the effective degradation of the substrate requires a consortium of enzymes. The cellulosome not only provides a variety of enzymes with varying specificities, but also promote a close proximity between the catalytic components (enzymes). The close proximity between the enzymes promotes the synergistic degradation of complex plant biomass for the production of valuable energy products. Previous synergy studies have focused predominantly on the synergistic associations between cellulases; however, the synergy between hemicellulases has occasionally been documented. This research project established the synergistic associations between two Clostridium cellulovorans hemicellulases that may be incorporated into the cellulosome and a cellulosomal endoglucanase that is conserved in all cellulosomes. This research study indicated that there was indeed a synergistic degradation of the complex plant biomass (sugarcane bagasse and pineapple fibre). The degrees of synergy and the ratio of the enzymes varied between the two complex substrates. The initial degradation of the bagasse required the presence of all the enzymes and proceeded at an enhanced rate under sulphidogenic conditions; however, there was a low production of fermentable sugars. The low quantity of fermentable sugars produced by the degradation of the bagasse may be related to the chemical composition of the substrate. The sugarcane contains a high percentage of lignin forming a protective layer around the holocellulose, thus the glycosidic bonds are shielded extensively from enzymatic attack. In comparison, the initial degradation of the pineapple fibre required the action of hemicellulases, and proceeded at an enhanced rate under sulphidogenic conditions. The initial degradation of the pineapple fibre produced a substantially larger quantity of fermentable sugars in comparison to the bagasse. The higher production of fermentable sugars from the degradation of the pineapple fibre may be explained by the fact that this substrate may have a lower percentage of lignin than the bagasse, thus allowing a larger percentage of the glycosidic bonds to be exposed to enzymatic attack. - Description: In recent years, increased demands have been placed on hydrologists to find the most effective methods of making predictions of hydrologic variables in ungauged basins. A huge part of the southern African region is ungauged and, in gauged basins, the extent to which observed flows represent natural flows is unknown, given unquantified upstream activities. The need to exploit water resources for social and economic development, considered in the light of water scarcity forecasts for the region, makes the reliable quantification of water resources a priority. Contemporary approaches to the problem of hydrological prediction in ungauged basins in the region have relied heavily on calibration against a limited gauged streamflow database and somewhat subjective parameter regionalizations using areas of assumed hydrological similarity. The reliance of these approaches on limited historical records, often of dubious quality, introduces uncertainty in water resources decisions. Thus, it is necessary to develop methods of estimating model parameters that are less reliant on calibration. This thesis addresses the question of whether physical basin properties and the role they play in runoff generation processes can be used directly in the estimation of parameter values of the Pitman monthly rainfall-runoff model. A physically-based approach to estimating the soil moisture accounting and runoff parameters of a conceptual, monthly time-step rainfall-runoff model is proposed. The study investigates the physical meaning of the model parameters, establishes linkages between parameter values and basin physical properties and develops relationships and equations for estimating the parameters taking into account the spatial and temporal scales used in typical model applications. The estimationmethods are then tested in selected gauged basins in southern Africa and the results of model simulations evaluated against historical observed flows. The results of 71 basins chosen from the southern African region suggest that it is possible to directly estimate hydrologically relevant parameters for the Pitman model from physical basin attributes. For South Africa, the statistical and visual fit of the simulations using the revised parameters were at least as good as the current regional sets, albeit the parameter sets being different. In the other countries where no regionalized parameter sets currently exist, simulations were equally good. The availability, within the southern African region, of the appropriate physical basin data and the disparities in the spatial scales and the levels of detail of the data currently available were identified as potential sources of uncertainty. GIS and remote sensing technologies and a widespread use of this revised approach are expected to facilitate access to these data.

Monitoring and assessment of fish communities on a long-term basis is necessary, and will provide a means to evaluate the effectiveness of such management measures. Therefore, standardised protocols and optimal sampling methods for long-term monitoring (LTM) and assessment of coastal fish communities are essential. This study aimed to identify suitable methods and develop a protocol for assessment of inshore reef fish communities. A suitable location for evaluation of proposed methods was identified in the warm temperate biogeographical region of South Africa, encompassing the well-established Tsitsikamma Coastal National Park MPA and an adjacent exploited area. Chrysoblephus laticeps (roman) was identified as an indicator species for the study, as it has been well-studied and is well represented in the area. Underwater visual census (UVC) and controlled fishing were identified as suitable methods. UVC transects were found to be superior to point counts, in terms of sampling efficiency, variability, bias and required sample size. An effort of two angler hours per fishing station was shown to provide low catch variability, while at the same time a representative catch and low overall cost and required time. The methods were incorporated in a proposed sampling protocol, and evaluated. The methods were able to detect known differences between protected and exploited communities. It is recommended that LTM within protected areas, for detection of natural change, be focused on community-level indicators, while LTM in exploited areas, aimed at detection of anthropogenic change, be focused on species-level indicators. - Description: The larval fish assemblage in the shallow, nearshore region of a proposed marine protected area (MPA) in eastern Algoa Bay, temperate South Africa was investigated. Current velocities and direction and the swimming abilities of late-stage larvae were further assessed to determine potential larval movement to and from the MPA. In total, 6045 larval fishes were collected along two depth contours (~5m and ~15m) in the shallow nearshore of eastern Algoa Bay using stepped-oblique bongo net tows, twice per season for two years (March 2005 – January 2007). These larvae represented 32 families and 78 species. The Gobiidae, Cynoglossidae, Clupeidae, Engraulidae and Sparidae were the dominant fish families. Catches varied significantly between seasons peaking in spring with a mean of 64 larvae/100m3. Preflexion stage larvae dominated catches (75%). All developmental stages of Diplodus capensis, Engraulis capensis, Heteromycteris capensis, Sardinops sagax and Pomadasys species were found in the study area. It appears that these species use the shallow nearshore as a nursery area. Analysis of 12 months (May 2006 – May 2007) data from a bottom-moored Acoustic Doppler Current Profiler within the study area showed that offshore south eastward (39%) and onshore north westward currents (33%) dominated. The south westward current (15%) and north eastward current (12%) occurred less frequently. Current velocity decreased with depth in the nearshore, with a mean velocity of ~29 cms⁻¹ recorded at a depth of 4 m and a mean velocity of ~11 cms⁻¹ recorded at a depth of 14 m. Understanding the dispersal and movement of marine fish larvae in coastal habitats requires knowledge of active swimming abilities. The critical speed and endurance swimming of late stage larvae of two common inshore species occurring in the study area, Diplodus capensis and Sarpa salpa (Family Sparidae), were measured in a laboratory swimming chamber. The mean U-crit value for D. capensis (18.6 cms⁻¹) was similar to that of S. salpa (18.0 cms⁻¹), whereas mean endurance (km swum) was greater in S. salpa (8.4 km) than D. capensis (5.9 km). These swimming abilities exceed the average current velocities observed in the shallow nearshore providing larvae with the ability to greatly alter their passive dispersal trajectories and ultimately influence their distribution in the nearshore.

- Description: The larval fish assemblage in the shallow, nearshore region of a proposed marine protected area (MPA) in eastern Algoa Bay, temperate South Africa was investigated. Current velocities and direction and the swimming abilities of late-stage larvae were further assessed to determine potential larval movement to and from the MPA. In total, 6045 larval fishes were collected along two depth contours (~5m and ~15m) in the shallow nearshore of eastern Algoa Bay using stepped-oblique bongo net tows, twice per season for two years (March 2005 – January 2007). These larvae represented 32 families and 78 species. The Gobiidae, Cynoglossidae, Clupeidae, Engraulidae and Sparidae were the dominant fish families. Catches varied significantly between seasons peaking in spring with a mean of 64 larvae/100m3. Preflexion stage larvae dominated catches (75%). All developmental stages of Diplodus capensis, Engraulis capensis, Heteromycteris capensis, Sardinops sagax and Pomadasys species were found in the study area. It appears that these species use the shallow nearshore as a nursery area. Analysis of 12 months (May 2006 – May 2007) data from a bottom-moored Acoustic Doppler Current Profiler within the study area showed that offshore south eastward (39%) and onshore north westward currents (33%) dominated. The south westward current (15%) and north eastward current (12%) occurred less frequently.
