- Description: Understanding strategies of resource partitioning within populations is a fundamental aspect of the study of ecology, and one which has been well documented across various taxa. In seabirds, sex-specific differences in foraging behaviour are often thought to reflect strategies of resource partitioning to avoid intra-specific competition. At present Bird Island in Algoa Bay hosts the world’s largest breeding population of gannets, with recent population estimates numbering well over 90 000 breeding pairs. Given the large size of this colony, intense levels of intra-specific competition are expected during the breeding season, particularly in years of poor feeding conditions. In addition, continuous foraging around the breeding colony may lead to the local depletion of prey as the season progresses, calling for flexibility in the foraging behaviour and diet of breeding individuals. Therefore, the aim of this dissertation was to investigate resource partitioning in Cape gannets breeding at Bird Island through strategies of sex-specific foraging and intra-seasonal variability in foraging behaviour and diet. To achieve this, the spatial and temporal aspects of Cape gannet foraging behaviour were assessed during the early stages of chick rearing, over two consecutive breeding seasons (2011/12, 2012/13) using GPS tracking and VHF monitoring. In addition, intra-seasonal variability in foraging behaviour and diet were investigated, taking into account the influence of sex and chick age, to provide a comprehensive assessment of foraging behaviour throughout the chick rearing period. GPS tracking data for 79 birds revealed limited evidence for sex-specific differences over spatial dimensions. An extension in the foraging range of females during a year of lower prey availability (2012/13) was, however, evident. This suggests a possible sex-specific response to prey limitation, likely reflecting resource partitioning strategies or differences in nutritional requirements. VHF data for 95 additional birds revealed a clear pattern in temporal foraging behaviour. In general, females were more likely to be on a foraging trip during the early and mid-day hours, with the probability of males being on a foraging trip increasing towards the late afternoon. Continuous tracking of individuals throughout the 2015/16 breeding season further revealed little evidence for sex-specific differences in spatial distribution and forging effort during both the guard and post-guard stages of chick rearing. A clear increase in foraging range was evident during the early stages of chick rearing as the season progressed. However, birds attending chicks older than 50 days showed an unexpected contraction in foraging range, which appeared to coincide with a dietary shift. During the early stages of breeding the diet was comprised almost exclusively of anchovy, the proportion of which decreased progressively in the diet throughout the breeding season. This was mirrored by a substantial increase in the proportion of saury in the diet. These results suggest that Cape gannets increase their foraging range in response to local prey depletion as the breeding season progresses. However, when alternative prey becomes available they may shift their diet, allowing them to access prey closer to the breeding colony. Overall, this dissertation provides insight into sex-specific behaviour in a monomorphic seabird, suggesting a marginal degree of spatial segregation. The results provide the first support for sex-related temporal foraging segregation in gannets. Such separation could potentially be driven by resource partitioning, but could also relate to differential nutritional requirements. In addition, this study also provided the first comprehensive assessment of foraging behaviour throughout the guard and post guard stages of chick rearing in gannets. These results suggest that a combination of foraging and dietary flexibility may allow Cape gannets to buffer the effects of prey depletion during the breeding season. In conclusion, resource partitioning through sex-specific foraging appears to be limited in the Cape gannet population at Bird Island. Segregation via alternative pathways may however be possible and as such, future studies should consider the influence of individual level segregation and habitat specialization.

A thorough understanding of marine predator foraging ecology is thus important to improve our understanding of ecosystem function, inform conservation and management measures, and to predict the response of predators to future environmental change. The Cape fur seal (Arctocephalus pusillus pusillus) comprises a major proportion of the southern African marine predator biomass. However, little is known about the foraging distribution and behaviour of the species, despite its obvious importance within the ecosystem and increasing concerns regarding seal-fisheries conflict. Using a variety of technologies, including, GPS and satellite-linked tags, dive behaviour recorders, accelerometers and stable isotope analysis, this thesis investigated the foraging ecology of female Cape fur seals breeding on the coast of South Africa. First, to identify important foraging areas and assess the influence of geographic, temporal and environmental factors on these areas, lactating females from three breeding colonies were fitted with GPS or Argos-linked tracking devices, while individuals from two of these colonies were also fitted with dive behaviour recorders. Results revealed geographic differences in the foraging distributions and ranges of individuals between breeding colonies, which likely reflects a combined influence of ecological setting and density dependant competition. - Description: This study focuses on the development of InAs/GaInSb strained-layer superlattice structures by metal organic chemical vapour deposition (MOCVD), and deals with two aspects of the development of InAs/GaInSb SLS’s by MOCVD viz. the deposition of nano-scale (~100 Å) GaInSb layers, and the electrical characterization of unstrained InAs. The first part of this work aims to study the MOCVD growth of GaInSb layers in terms of deposition rate and indium incorporation on the nano-scale. This task is approached by first optimizing the growth of relatively thick (~2 μm) epitaxial films, and then assuming similar growth parameters during nano-scale deposition. The GaInSb layers were grown as part of GaInSb/GaSb quantum well (QW) structures. By using this approach, the GaInSb QW’s (~100 Å) could be characterized with the use of photoluminescence spectroscopy, which, when used in conjunction with transmission electron microscopy and/or X-ray diffractomery, proves useful in the analysis of such small scale deposition. It is shown that the growth rate of GaInSb on the nano-scale approaches the nominal growth rates determined from thick (~2 μm) GaInSb calibration layers. The In incorporation efficiency in nano-layers, however, was markedly lower than what was predicted by the GaInSb calibration layers. This reduction in indium incorporation could be the result of the effects of strain on In incorporation. The choice of substrate orientation for QW deposition was also studied. QW structures were grown simultaneously on both (100) and 2°off (100) GaSb(Te) substrates, and it is shown that growth on non-vicinal substrates is more conducive to the deposition of high quality QW structures. The second part of this study focuses on the electrical characterization of unstrained InAs. It is long known that conventional Hall measurements cannot be used to accurately characterize InAs epitaxial layers, as a result of parallel conduction resulting from surface and/or interface effects. This study looks at extracting the surface and bulk electrical properties of n-type InAs thin films directly from variable magnetic field Hall measurements. For p-type InAs, the situation is complicated by the relatively large electron to hole mobility ratio of InAs which tends to conceal the p-type nature of InAs thin films from Hall measurements. Here, this effect is illustrated by way of theoretical simulation of Hall data.

A gatherer’s paradise? early humans and plant foraging on the Cape south coast, South Africa

- Description: Humans were driven to refugia during the cold and dry glacial Marine Isotope Stage 6 (MIS6) (~195–125 ka); only a few places, including the Cape south coast, show archaeological evidence of continuous human occupation. It has been hypothesised that the Cape south coast provided the requisite shelter to ensure human’s earliest survival because it provided all the right ingredients. The shores offer abundant shellfish, the land a diverse array of plants, and the climate was ameliorated due to the proximity of the then exposed Palaeo-Agulhas Plain to the warm Agulhas current. The aim of this study is to determine whether the indigenous flora of this region could have provided a sufficient edible resource for early humans. Residents of the Cape south coast have genetic ancestry linking them to the Khoe-San, the original inhabitants of the area, and still have an extensive knowledge of the local edible plants. With their help, I set out to determine a) whether humans have been utilising the same plant species over time, b) what the foraging potential of the edible plants in the region is and c) how resilient these plants are to human foraging. If we know whether the plant species known and used today were the same as those that were utilised by past humans, we can then use the contemporary knowledge to make predictions about past utilisation. To answer this question, I collated two databases: archaeological (all plant species found in archaeological sites [dating 0 to 80,000 BP]) and contemporary (all plant species in the modern-day ethnographic literature [last 400 years]) that occur within the Greater Cape Floristic Region(GCFR). I found a significant number of plant species shared between the two databases, which suggests that at least some plant species have been used by humans over a long period of time. To determine the indigenous plant foraging potential of the region, I foraged for food in the Cape south coast (451 bouts) monthly over a two-year period with the help of local inhabitants. The findings show that edible plant resources are distributed patchily and focusing on specific vegetation types would greatly enhance chances of harvesting 2,000 kcal per day, which is considered the daily calorific requirements for a typical hunter-gatherer. I then sought to understand how resilient these plants [(with an emphasis on plants with an underground storage organ (USO)] would be to human foraging. To do this, I set out plots and harvested all edible foods for three consecutive years with the help of foragers. Results indicate that there was a significant reduction in edible weight only in the third year of consecutive harvesting. In conclusion, using various approaches, this study investigates the plant food potential of the Cape south coast from the perspective of early human consumers. The findings suggest that knowledge regarding useful plants dates back to at least 80,000 BP. Food resources are patchily distributed across the main vegetation types found within the Cape south coast and occur in hotspots, i.e. concentrated areas hosting high densities of edible plant foods, surrounded by areas with very low plant food densities. Foragers could have harvested their daily calorific quota more easily if they focused their harvesting efforts in specific vegetation types found in the Cape south coast. Furthermore, many USOs circumvent climatic fluctuations, herbivory or both by staggering their emergence over multiple years, which implies they have some resilience to human foraging.

- Description: Loggerhead and leatherback sea turtles nest on the beaches of the north-eastern portion of Kwazulu-Natal within the iSimangaliso Wetland Park. Loggerheads place ~60 percent of all nests within an 8 km stretch of beach, whereas leatherbacks tend to space their nests more evenly along the entire length of the monitoring area. The study aimed to determine nest site fidelity of loggerheads and leatherbacks (using four decades of nesting data housed by Ezemvelo KZN Wildlife) and the factors that influence nest site selection of both species within the 56 km of turtle monitoring area (32N to 100S) and the 5 km area of high-density loggerhead nesting (0N to 12N). The effectiveness of nest site selection was then determined through the hatching success of loggerheads over the 5km area (0N to 12N). Results showed that loggerheads show a high degree of nest site fidelity (~3 km) with nest site fidelity of individuals increasing over subsequent seasons of nesting, as well as these individuals using the same stretches of beach for nesting (the most popular area being 1N to 4N for repeat nesters). Leatherbacks displayed nest site fidelity of ~9 km and this did not increase over successive seasons of nesting. In terms of nest site selection, loggerheads and leatherbacks both avoided areas where low shore rock was present, whereas both species preferred nesting on beaches of intermediate morphodynamic state. Leatherback nesting was significantly higher in areas with wider surf zones. Both species were able to surpass the high water mark when nesting as nests below this point would be almost certainly doomed. Hatching success of loggerheads was comparative to high (83 %) relative to other studies, however, nest success varied across the beach from beacon 1N to 12N. Areas where highest nest success was observed were not areas of highest nest density presumably due to artificial lighting. Results from this study increase our understanding of the evolutionary biology of loggerhead and leatherback turtles in South Africa and the effectiveness of loggerhead nest site selection through hatching success.

- Description: Loggerhead and leatherback sea turtles nest on the beaches of the north-eastern portion of Kwazulu-Natal within the iSimangaliso Wetland Park. Loggerheads place ~60 percent of all nests within an 8 km stretch of beach, whereas leatherbacks tend to space their nests more evenly along the entire length of the monitoring area. The study aimed to determine nest site fidelity of loggerheads and leatherbacks (using four decades of nesting data housed by Ezemvelo KZN Wildlife) and the factors that influence nest site selection of both species within the 56 km of turtle monitoring area (32N to 100S) and the 5 km area of high-density loggerhead nesting (0N to 12N). The effectiveness of nest site selection was then determined through the hatching success of loggerheads over the 5km area (0N to 12N). Results showed that loggerheads show a high degree of nest site fidelity (~3 km) with nest site fidelity of individuals increasing over subsequent seasons of nesting, as well as these individuals using the same stretches of beach for nesting (the most popular area being 1N to 4N for repeat nesters). Leatherbacks displayed nest site fidelity of ~9 km and this did not increase over successive seasons of nesting. In terms of nest site selection, loggerheads and leatherbacks both avoided areas where low shore rock was present, whereas both species preferred nesting on beaches of intermediate morphodynamic state. Leatherback nesting was significantly higher in areas with wider surf zones. Both species were able to surpass the high water mark when nesting as nests below this point would be almost certainly doomed. Hatching success of loggerheads was comparative to high (83 %) relative to other studies, however, nest success varied across the beach from beacon 1N to 12N. Areas where highest nest success was observed were not areas of highest nest density presumably due to artificial lighting. Results from this study increase our understanding of the evolutionary biology of loggerhead and leatherback turtles in South Africa and the effectiveness of loggerhead nest site selection through hatching success.

- Description: This study reports the findings on the effect of Microcystin-LR (MCLR) on the gastrointestinal tract cells of mice and on two different cell lines, Caco2 and MCF-7. The cyanobacterium Microcystis aeruginosa produces the potent toxin, MCLR. This toxin has been implicated in a number of cases of ill-health. It was decided to investigate whether microcystin-LR induced apoptosis in the gastrointestinal tract of mice and also which possible mechanisms were involved in the induction in vitro. Balb/c mice were given a 75% LD50 intraperitoneal dose of pure microcystin -LR and sacrificed at 8, 16, 24 and 32 hours post-exposure. The small intestinal sections were stained with haematoxylin and eosin and examined for apoptotic cells. There was a time-dependent increase in the number of apoptotic cells with most in the duodenum and the jejunum. No change in glycogen content was evident at 24 hours post exposure when PAS-stained sections were examined. To determine that microcystin was the agent responsible for the changes, fluoroscein isothiocyanate (FITC) immunostaining for the toxin was done on the sections. Apoptosis in vitro was investigated in Caco2, a cell line that behaves like normal enterocytes when the cells are differentiated at confluency, and MCF-7, a breast cancer cell line deficient in pro-caspase-3, cells by 3-[dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) and lactate dehydrogenase (LDH) assays and by staining with DAPI and Rhodamine 123. MCLR exposure induced apoptosis, as seen in decreased cell viability and increased leakage of LDH, as well as mitochondrial damage shown by Rhodamine staining. The MCF-7 cells, deficient in pro-caspase-3, and Caco2 cells did not show cleavage of poly(ADP)ribose polymerase (PARP) after exposure to 50μM MCLR after 72 hours exposure. Both micro- and milli-calpain activity was however significantly increased in both cell lines exposed to the toxin. There was a significant increase in H2O2, one of the key reactive oxygen species, production during the first 30 minutes that the cells were exposed to 50 mM MCLR.

Curcumin and Nigella sativa have anti-cancer, anti-metastatic and antioxidant-properties and may be effective against breast cancer. This study focused on the effect of N. sativa extracts or thymoquinone and curcumin, individually and in combination, on breast cancer cells. An MTT assay showed that curcumin reduced cell viability by 50% (IC50) at 18 ± 2.63 μg/mL and thymoquinone (TQ) at 5 ± 0.95 μg/mL against the MDA-MB-231 cells. The IC50 values for curcumin and TQ were 35 ± 6.98 μg/mL and 4 ± 0.96 μg/mL against the MCF-7 cells, respectively. The IC50 value for the NSBE was determined to be 350 ± 55 μg/mL. The IC50 value of NSAE did not fall within the selected concentration range. Synergism was noted for combinations of NSBE with curcumin, and combinations of TQ with curcumin, against both MCF-7 and MDA-MB-231 cells. Two synergistic combinations per treatment per cell line, as determined by the combination index analysis, were chosen for further investigation. The combinations and individual treatments tested against the MCF-10A cells, were not significant, except for NSBE80:CURC20 combination. Curcumin had the most significant anti-oxidant activity; however, no link was noted between the anti-oxidant activity and the cytotoxicity of the combinations. The combination treatments induced apoptosis more effectively than the individual treatments. Caspase-3 dependent apoptosis was noted for NSBE10:CURC90 and TQ80:CURC20 combinations against the MDA-MB-231 cells, and the TQ60:CURC40 combination against the MCF-7 cells. The individual and combined treatments effectively reduced MDA-MB-231 cell adhesion to fibronectin, but not all reduced the cell adhesion to laminin. Effect of selected physical and production traits on the tick burdens of beef cattle

- Description: The objective of this study was to determine the effect of selected traits such as age, sex, body weight, body length and height, body condition score (BCS), coatscore (CS), skin thickness and average skin surface temperature on tick burdens in beef cattle. Bonsmara cattle (n= 143) were used to measure visible tick counts, body condition score, coat score, skin thickness, body height and length, body weight, body surface temperature, gender and inter calving period. Measurements were taken from April to December. All animals were managed extensively on natural and cultivated pastures near George in the Southern Cape. Female animals had significantly (p<0.05) greater tick infestation (37.98±2.7) compared to male animals (16.52±1.2). Age was a significant factor (p<0.001) with the younger animals below two years having (46.40±5.26) more ticks than those of two years and older (20.15±2.44). A significant negative correlation (p<0.001; -0.29) was reported between the infestation of ticks on the animals and the age of the animal. Animals with an average body weight below 250kg had 42% (p<0.05) more ticks compared to animals with a body weight above 250kg. Age of the animal and weight were highly correlated (r= 0.70); p<0.001), while the correlation between the number of ticks per cow and the mean weight was negatively correlated (r= -0.37; p<0.001). Skin surface temperature significantly influenced tick infestation on the animals (p<0.001). The degree of infestation increased as body surface temperature exceeded 30º C. Coat score, skin thickness, body condition score and inter calving period did not significantly influence tick infestation on the animals. The infestation of ticks on the animals were significantly influenced by body height (p<0.019) and body length (p<0.001). Animals smaller than a 130cm in height had a significantly (p<0.05) greater tick infestation (36.5±5) compared to animals taller than 130cm (21.2±1.5). This trend was also observed for body length. Animals with a body length shorter than 145cm had a greater (p<0.05) average tick infestation of 41.3±4.5 compared to 23.2±1.3 to animals longer than 145cm, indicating a 44% greater tick infestation in favour of the shorter animals. For increased production and tick resistance animals should have smoother coats and be able to dissipate heat effectively.

Ostrich high molecular weight kininogen was purified from plasma and low molecular weight kininogen was partially purified. The N-terminal amino acid sequences of both high- and low molecular weight kininogens from ostrich plasma were determined. Ostrich plasma high molecular weight kininogen was purified as a 118 kD protein. The purified high molecular weight kininogen inhibits the cysteine proteinase papain at a ratio of one molecule HKG to two molecules of papain. Ornitho kinin-like molecules were detected in ostrich urine using reverse phase HPLC.

- Description: The African potato is used as an African traditional medicine for its nutritional and medicinal properties. Most research has been carried out on H. hemerocallidea, with very little or nothing on other Hypoxis spp. The main aim of this project was to provide scientific data on the anticancer, anti-inflammatory and antioxidant properties of H. hemerocallidea, H. stellipilis and H. sobolifera chloroform extracts and their active compounds. The hypoxoside and phytosterol contents of the three Hypoxis spp. were determined using TLC, HPLC and GC. H. hemerocallidea and H. sobolifera chloroform extracts contained the highest amounts of hypoxoside and β-sitosterol, respectively. For the anticancer properties, cytotoxicity of the Hypoxis extracts and its purified compounds were determined against the HeLa, HT-29 and MCF-7 cancer cell lines (using MTT), and PBMCs (using CellTiter-Blue®). H. sobolifera had the best cytotoxicity against the three cancer cell lines, whereas H. stellipilis stimulated HeLa and HT-29 cancer cell growth. IC50 values of hypoxoside and rooperol were determined. DNA cell cycle arrest (using PI staining) occurred in the late G1/early S (confirmed by increased p21Waf1/Cip1 expression) and G2/M phases after 15 and 48 hrs, respectively, when treated with Hypoxis extracts and rooperol. H. sobolifera and rooperol activated caspase-3 and -7 (using fluorescently labelled antibodies) in HeLa and HT-29 cancer cells, and caspase-7 in MCF-7 cancer cells after 48 hrs. Annexin V binding to phosphatidylserines in rooperol treated U937 cells confirmed early apoptosis after 15 hrs. The TUNEL assay showed DNA fragmentation in the three cancer cell lines when treated with H. sobolifera and rooperol for 48 hrs. A shift pass the G2/M phase has led to the investigation of endoreduplication, which was confirmed by cell/nucleus size, and anti-apoptotic proteins (Akt, phospho-Akt, phospho-Bcl-2 and p21Waf1/Cip1). U937 cell differentiation to monocyte-macrophages was optimized using PMA and 1,25(OH)2D3, which was confirmed by morphological and biochemical changes. For the anti-inflammatory properties, Hypoxis extracts and rooperol significantly increased NO production in monocyte-macrophages (pre-loaded with DAF-2 DA) and phagocytosis of pHrodoTM E. coli BioParticles®. The treatments had no effect on COX-2 expression in monocyte-macrophages. The phytosterols significantly increased IL-1β and IL-6 secretion xv (using the FlowCytomix Multiplex human Th1/Th2 10plex Kit I) in the PBMCs of one donor. For the antioxidant properties, Hypoxis extracts and rooperol significantly increased ROS production in undifferentiated and differentiated U937 cells, which were pre-loaded with DCFH-DA. Hypoxis extracts and purified compounds had ferric reducing activities, but only rooperol had ferric reducing activities significantly greater than ascorbic acid. β-sitosterol, campesterol and cholesterol significantly increased SOD activity in Chang liver cells, while H. stellipilis, H. sobolifera and rooperol decreased SOD activity. Anticancer, anti-inflammatory and antioxidant properties of the Hypoxis extracts may be attributed to the β-sitosterol content, because Hypoxis chloroform extracts contained very little or no hypoxoside. Unidentified compounds, and synergistic and additive effects of the compounds may have contributed to the biological effects. This study confirms previous reports that rooperol is the active compound. Results provide scientific data on the medicinal properties of one of the most frequently used medicinal plants in South Africa.

The TUNEL assay showed DNA fragmentation in the three cancer cell lines when treated with H. sobolifera and rooperol for 48 hrs. A shift pass the G2/M phase has led to the investigation of endoreduplication, which was confirmed by cell/nucleus size, and anti-apoptotic proteins (Akt, phospho-Akt, phospho-Bcl-2 and p21Waf1/Cip1). U937 cell differentiation to monocyte-macrophages was optimized using PMA and 1,25(OH)2D3, which was confirmed by morphological and biochemical changes. For the anti-inflammatory properties, Hypoxis extracts and rooperol significantly increased NO production in monocyte-macrophages (pre-loaded with DAF-2 DA) and phagocytosis of pHrodoTM E. coli BioParticles®. The treatments had no effect on COX-2 expression in monocyte-macrophages. The phytosterols significantly increased IL-1β and IL-6 secretion xv (using the FlowCytomix Multiplex human Th1/Th2 10plex Kit I) in the PBMCs of one donor. For the antioxidant properties, Hypoxis extracts and rooperol significantly increased ROS production in undifferentiated and differentiated U937 cells, which were pre-loaded with DCFH-DA. Hypoxis extracts and purified compounds had ferric reducing activities, but only rooperol had ferric reducing activities significantly greater than ascorbic acid. β-sitosterol, campesterol and cholesterol significantly increased SOD activity in Chang liver cells, while H. stellipilis, H. sobolifera and rooperol decreased SOD activity. Anticancer, anti-inflammatory and antioxidant properties of the Hypoxis extracts may be attributed to the β-sitosterol content, because Hypoxis chloroform extracts contained very little or no hypoxoside. Unidentified compounds, and synergistic and additive effects of the compounds may have contributed to the biological effects. This study confirms previous reports that rooperol is the active compound. Results provide scientific data on the medicinal properties of one of the most frequently used medicinal plants in South Africa.

CD23 is a multifunctional type II transmembrane protein that binds its known ligands through its ectodomain either as a membrane-bound or soluble receptor generated in vivo by specific proteolytic cleavages. IgE production is primarily regulated by interactions between IgE, CD23 and CD21. Despite its importance for development of strategies to limit hypersensitivity, precise information about the molecular interactions remains limited. During this study, I engineered, expressed and purified from bacteria three soluble human CD23 fragments that are normally formed in vivo and shed from the cell surface (1) derCD23, amino acids 156-298 (2) sCD23, amino acids 150-321 and (3) the entire ectodomain, exCD23, amino acids 48-321 to examine the comparative binding of recombinant human CD21 SCR 1-2 and native human IgE to these fragments. Gel filtration HPLC revealed that derCD23 and sCD23 were monomeric whereas exCD23 assembled as a heterogeneous mixture that included trimers and monomers. At the concentrations utilized, CD23 fragments sCD23 and exCD23 bound CD21 with similar affinity, whereas interaction between derCD23 and CD21 was minimal when analyzed by surface plasmon resonance (SPR) spectroscopy. These findings suggest that penultimate “tail” amino acids between 298 and 321 stabilize CD21 attachment, although it cannot be ruled out, the region between Met 150 and Ser 156 may also play a role in binding CD21 SCR 1-2. In contrast, there is a progressive increment in the affinity of soluble fragments (exCD23>sCD23>derCD23) for IgE, upon increasing length of the proximal CD23 “stalk” domain. These findings highlight the differences in both the structural basis and affinity of the three physiological fragments of human CD23 for the ligands CD21 and IgE and underscore the complexity of CD23-mediated regulatory networks. It was found that B-cells only make up ~5% of the PBMC population, and that these cells were able to be activated, via STAT-6 phosphorylation, to enter class switch recombination (CSR) by the addition of switch factors (IL-4 and anti-CD40). Titration experiments dictated that 25 ng/mL of CD23 was the most efficient concentration to up-regulate IgE synthesis in PBMCs; furthermore, soluble CD23 proteins were incubated with PBMCs in the presence and absence of CD21 SCR 1-2 to investigate the effect that these recombinant proteins have on IgE synthesis. Results showed that the influence of recombinant proteins (both CD23 and CD21) on IgE synthesis was slight. It was shown that while derCD23 had no significant effect, monomeric sCD23 down-regulated, and the mixture of monomeric and oligomeric exCD23 up-regulated IgE synthesis. On addition of CD21 SCR 1-2 to the cells switched and treated with soluble CD23, it was found that in both cases for sCD23 and exCD23, IgE synthesis was increased, while for derCD23, there was no noticeable difference in IgE synthesis. This confirmed previous data showing the lack of binding between derCD23 and CD21 SCR 1-2. The exact binding site for CD21 on the CD23 molecule is unknown, and incompletely represented in the NMR and crystal structures. It is thought that CD21 binds to the C-terminal tail section, not present in derCD23. It is therefore likely that only a negative-feedback mechanism operates with derCD23 to regulate IgE synthesis. Further investigation of the binding of CD23 fragments to SCR 5-8 of CD21 and the effect of this on IgE synthesis may lead to a potential therapeutic role for derCD23 in the treatment of allergic disease. Data accumulated in this study suggests that investigating the modulation of oligomeric state and thus the activity of soluble CD23 fragments may be important in the construction of new regulators of IgE synthesis.

- Description: Seabirds are important marine sentinels, providing valuable information about ecosystem health and condition. Dietary studies of seabirds can inform on ecosystem functioning through identifying the condition and changes occurring in their surrounding environment. Dietary studies are also crucial as they can identify threats that seabirds face. Until recently, the diets of southern African endemic and Endangered Cape Gannets Morus capensis and African Penguins Spheniscus demersus were solely investigated through stomach content analyses, which allows for the identification and quantification of prey remains. To overcome some of the limitations associated with stomach content analysis, stable isotope and fatty acid analyses were proposed. However, these methods still require bird handling and thus impose some stress on the birds. DNA metabarcoding of faecal matter shows promise as a completely noninvasive method for seabird dietary studies. This thesis first examined whether DNA metabarcoding could be used to determine the diet of Cape Gannet and African Penguin breeding in Algoa Bay, South Africa. Then, this method was used to investigate the influence of intrinsic (both seabird species) and extrinsic (African Penguins) factors on the diet of the two seabird species. For the first data chapter, stomach content samples and faecal matter were collected concurrently from breeding Cape Gannets and African Penguins on Bird Island. DNA was successfully extracted from most faecal matter samples and the primers (18S_SSU, 16S_Fish) used for group, taxa and fish prey identification allowed amplification of prey DNA for both study species. A similar piscivorous diet was determined by both dietary methods for Cape Gannets with Sardine Sardinops sagax and Atlantic Saury Scomberesox saurus dominating the diet. Sardine also dominated the African Penguin diet in both dietary methods. In addition, DNA metabarcoding in African Penguins revealed a more diverse diet compared to the one determined by stomach content analysis. This was likely due to the limitations on the allowed number of stomach content samples to be collected for that Endangered species. Non-invasive DNA metabarcoding of faecal matter was then used to investigate whether the diet varied with breeding stages (incubation, chick-rearing) and age classes (chick-rearing adults, chicks) in both seabird species, but also breeding colonies (Algoa Bay: Bird Island, St Croix Island) in African Penguins. DNA metabarcoding revealed that breeding stages influenced the diet of Cape Gannets and African Penguins on Bird Island where prey species diversity was greater for incubating adults. On the contrary, the diet of incubating and chick-rearing penguins on St Croix Island was similar. When comparing the diet of African Penguins between Bird Island and St Croix Island, there was an increased overlap in diet composition between incubating and chick-rearing adults on St Croix Island when compared to Bird Island. Importantly, DNA metabarcoding indicated an increase in fishery interactions in Algoa Bay, particularly for Cape Gannets and the hake Merluccius spp. fishery, where the contribution of hake to chick-rearing adults and chick diet was considerably greater than incubating adult diet. In this study, I showed that DNA metabarcoding of faecal matter provides an opportunity for non-invasive dietary studies in two southern African endemic and Endangered seabird species. Devulcanization of model compounds by a variety of diphenyldisulfides

- Description: Improvements to the properties of recycled rubber, by developing a more selective breakdown process, is an important issue and a global challenge. Devulcanization is the most promising way to achieve this. It is a process which aims to totally or partially cleave monosulfidic (C-S-C), disulfidic (C-S-S-C) and polysulfidic (C-Sx-C) crosslinks of vulcanized rubber. In this study, the devulcanization of sulfur-vulcanized natural rubber with a variety of diphenyldisulfides has been studied using 2,3-dimethyl-2-butene and squalene as model compounds, vulcanized by sulfur and 2-bisbenzothiazole-2,2’-disulfide. 2-aminodiphenyldisulfide; 4-amino-diphenyldisulfide; bis(2-benzamido-)diphenyldisulfide and 2,2’-bithiosalicylic acid were used as aromatic disulfides devulcanization agents. Thermal analysis was used to investigate interactions between various combinations of curatives in the vulcanization systems in the absence of model compounds. Thermogravimetric analysis was used to detect mass changes during vulcanization and whether the samples decompose/evaporate so as not to contaminate differential scanning calorimetry instrumentation. Differential scanning calorimetry was used to investigate reactions occurring between curatives. Comparison of reversing and non-reversing signals in modulated experiments are suggestive of reactions between devulcanization agents and sulfur, and accelerators Vulcanization of the model compounds were performed in 10m evacuated sealed glass ampoules placed in an oil bath at 160 °C and agitated for the duration of vulcanization. After 60 min the ampoules were removed and quenched. These were then devulcanized by exposure to a devulcanization agent in a sealed ampoule at 180 for 60 min. Crosslinked 2,3-dimethyl-2-butene species were isolated and analysed by reversed phase high performance liquid chromatography. Particular attention was paid to changes in the number of sulfur atoms in the crosslinks. Results indicate that of the devulcanization agents tested 4-amino-diphenyldisulfide was the most effective. No detrimental effect on devulcanization efficiency was caused by the substitution of a basic amino group with an acidic carboxylic acid group Gel permeation chromatography was performed on devulcanized squalene crosslinks using a tetrahydrofuran eluent. GPC investigations with squalene were less effective than HPLC experiments with 2,3-dimethyl-2-butene at evaluating the relative efficiency of individual devulcanization agents. Results were, however, consistent with 4-amino-diphenyldisulfide again being the most efficient devulcanization agent.

No detrimental effect on devulcanization efficiency was caused by the substitution of a basic amino group with an acidic carboxylic acid group Gel permeation chromatography was performed on devulcanized squalene crosslinks using a tetrahydrofuran eluent. GPC investigations with squalene were less effective than HPLC experiments with 2,3-dimethyl-2-butene at evaluating the relative efficiency of individual devulcanization agents. Results were, however, consistent with 4-amino-diphenyldisulfide again being the most efficient devulcanization agent.

- Description: The Beattie Magnetic Anomaly is the world’s longest terrestrial magnetic anomaly with a strike length of over 1000 km and a wavelength in excess of 100 km. Collinear with this is a large belt of elevated crustal conductivities called the Southern Cape Conductive Belt. Historical crustal interpretations proposed a common source of serpentinized ophiolite as an explanation for both the anomalous crustal magnetic susceptibility and electrical conductivities. Spreading between the Western and Eastern Cape of South Africa the mid- to lower crust that hosts these anomalies is obscured by the overlying Cape and Karoo Supergroups. Between 2003 and 2006, three high resolution geophysical experiments were completed across the surface maximum of the Beattie Magnetic Anomaly (BMA) and the Southern Cape Conductive Belt (SCCB). These included a magnetotelluric (MT) survey and near vertical reflection and wide angle refraction seismic profiles. Within the MT inversion model the SCCB appeared as a composite anomaly, which included a mid-crustal conductor which is spatially associated with the BMA and a laterally continuous upper crustal conductor which is located at depths equivalent to the lower Karoo Supergroup. Subsequently; the upper crustal conductor was identified in northern and eastern extensions of the magnetotelluric profile; a distance in excess of 400 km. Historical magnetometer and Schlumberger Sounding experiments have previously identified elevated conductivities in the Karoo sequences which were attributed to the Whitehill and Prince Albert formations. These carboniferous, transgressive sediments are known to be conductive from borehole conductivity surveys and direct measurements at surface. In order to constrain the conductive properties of these sediments, impedance spectroscopy (IS) experiments were completed on core samples collected from a historical borehole drilled near to the MT profile. Part One of this thesis presents the results of these experiments, which support the proposition that the Whitehill and Prince Albert Formations are responsible for the laterally continuous, sub-horizontal, upper crustal conductor visible in the MT inversion model. Vitrinite reflectance studies were performed on the same samples by the Montanuniversität, in Leoben, these results corroborate the proposition that elevated organic carbon, of meta-anthracite rank, is the primary conductive phase for the Whitehill and Prince Albert formations. Part two of this thesis completed forward modelling exercises using historical aeromagnetic data previously collected across the Beattie Magnetic Anomaly. Preliminary models were unable to fit the geometry of any single magnetic model with conductors present in the MT inversion model discounting the proposition that the SCCB and BMA arise from a single crustal unit. Two constrained models were arrived at through an iterative process that sought a best fit between the measured data and the NVR crustal interpretations. The first model, proposes a largely resistive unit which incorporates portions of elevated crustal conductivity; these conductors are spatially correlated to crustal portions also characterised by high seismic reflectivity. The size of this modelled body suggest the likely host of the BMA is an intermediate plutonic terrane, analogous with the Natal sector of the Namaqua Natal Mobile Belt as well as the Heimefrontfjella in Dronning Maud Land, Antarctica, with magnetite hosted within shear zones. This is in agreement with previous studies. The second model proposes a lower crustal sliver imaged in the NVR data at depths proximal to the Curie Isotherm for magnetite and hematite as the source of the BMA. At these depths geomagnetic properties such as burial magnetisation or thermo-viscous remanent magnetism (TVRM) can potentially be linked to regional scale tectonic processes and can theoretically elevate a body’s net magnetic susceptibility. TVRM has been proposed for long wavelength crustal anomalies elsewhere.

Having a system that notifies the user of important messages using the most appropriate medium and device will benefit users that rely on time critical information. There are several existing systems and models for mobile notification as well as for adaptive mobile notification using context awareness. Current models and systems are typically designed for a specific set of mobile devices, modes and services. Communication however, can take place in many different modes, across many different devices and may originate from many different sources. The aim of this research was to develop a model for adaptive mobile notification using context awareness. An extensive literature study was performed into existing models for adaptive mobile notification systems using context awareness. The literature study identified several potential models but no way to evaluate and compare the models. A set of requirements to evaluate these models was developed and the models were evaluated against these criteria. The model satisfying the most requirements was adapted so as to satisfy the remaining criteria. The proposed model is extensible in terms of the modes, devices and notification sources supported. The proposed model determines the importance of a message, the appropriate device and mode (or modes) of communication based on the user‘s context, and alerts the user of the message using these modes. A prototype was developed as a proof-of-concept of the proposed model and evaluated by conducting an extensive field study. The field study highlighted the fact that most users did not choose the most suitable mode for the context during their initial subscription to the service. The field study also showed that more research needs to be done on an appropriate filtering mechanism for notifications. Users found that the notifications became intrusive and less useful the longer they used them.

Physiological and non-physiological induction of gastrointestinal differentiation

- Description: The human colonic carcinoma cell lines HT-29 and Caco-2 both exhibit structural and functional differentiation under appropriate culture conditions. HT-29 can be induced to differentiate by treatment with short-chain fatty acids or acetoacetate. Caco-2 cells differentiate spontaneously upon contact inhibition. In this study HT-29 cells were treated with 5 mM acetate, propionate, butyrate and acetoacetate (physiological inducers) to assess their effects on the expression of carbonic anhydrase 1, sucrase-isomaltase and alkaline phosphatase which are reported to be markers of gastrointestinal differentiation. The maturation induction observed was compared to that of the spontaneous differentiation observed in Caco-2 cells. Assays were performed over an 18 day period. Results showed a close correlation (p < 0.05) between HT-29 and Caco-2 cell on days 4 and 12. These results indicate that differentiation reported in both cell lines is comparable and can be used as a basis for further comparative studies. In addition, parallel experiments to the above were conducted using a selection of nine rationally designed cyclic dipeptides (CDPs) potential drug entities which were chosen as non-physiological inducers. The results showed that the cyclic dipeptides were able to induce the gastrointestinal phenotype as observed in HT-29 cells treated with physiological inducers. Studies on the effects of energy-related metabolism in HT-29 and Caco-2 cells as induced by physiological and non-physiological inducers indicated that energy metabolism is a significant role player in gastrointestinal differentiation. The results reported show a decrease in ATP concentrations indicating that the cyclic dipeptides, like physiological inducers, affect the energy state of the HT-29 cells and thus may effect the differentiation of these cells. A positive correlation was found between histone phsophorylation and differentiation confirming that histone phsophorylation was partly responsible for the decrease in ATP concentrations. It is suggested that the induction of differentiation in HT- 29 cells could be either due to non-specific transcription of genes by activation of a chromatin switch or specific by the activation of signal transduction pathways based on the flux of ATP through the cells. Differential display RT-PCR is probably the most sensitive method that could be used to validate the suggestion of either a nonspecific transcription of genes or a specific differentiation reported for HT-29 cells.

In the present study, the potential of seven proline-based CDPs to inhibit cancer cell growth was investigated in HT-29 (colon), HeLa (cervical), MCF-7 (breast) and WHCO3 (oesophageal) cancer cell lines. The CDPs used in this study were cyclo(Phe-Pro), cyclo(Tyr-Pro), cyclo(Gly-Pro), cyclo(Pro- Pro), cyclo(His-Pro), cyclo(Leu-Pro) and cyclo(Thr-Pro). The sulforhodamine B (SRB) cell growth assay was used in an initial screening phase to investigate the effects of the CDPs in HT-29, HeLa and MCF-7 cells. After exposing the cells to 10mM of the respective CDPs for 48 hours, the SRB assay results showed that only cyclo(Phe-Pro) exhibited more than 50% growth inhibition (p<0.01) in the three cell lines. The other CDPs showed comparatively marginal growth-inhibitory effects, except for cyclo(Tyr-Pro), which exhibited a pronounced effect in MCF-7 cells compared to HT-29 and HeLa cells. The MTT assay was used to confirm the SRB assay results for cyclo(Phe-Pro) and cyclo(Tyr-Pro), extending the investigation to the use of the fourth cell line WHCO3 and using a longer exposure time of 72 hours. The MTT assay demonstrated a dosedependent (0.008-10 mM) growth inhibition by cyclo(Phe-Pro) with an IC50 value of 4.04 ± 1.15 mM for HT-29 cells. Cyclo(Phe-Pro) was subsequently used to investigate whether the growth-inhibitory effects of this CDP were related to the induction of apoptosis in HT-29 cells. Hoechst 33342 staining showed that 5mM cyclo(Phe-Pro) induced characteristic chromatin condensation and nuclear fragmentation in 18.3 ± 2.8% (p<0.01) of HT-29 cells after 72 hours. Furthermore, annexin V binding revealed that HT-29 cells treated with 5 mM cyclo(Phe-Pro) displayed phosphatidylserine externalization after 48 hours. In addition, it was shown that 10 mM cyclo(Phe-Pro) induced poly(ADP-ribose)polymerase PARP cleavage, one of the hallmark events of apoptosis. The use of the broad-range caspase inhibitor Z-VAD-FMK, showed that this PARP cleavage was caspase-dependent, which in turn was confirmed by demonstrating an increase in caspase-3 activity (p<0.01) in cyclo(Phe- Pro)-treated HT-29 cells. In conclusion, these findings demonstrate that cyclo(Phe-Pro) inhibited the growth of HT- 29, MCF-7, HeLa and WHCO3 cells, and induced apoptosis in HT-29 colon cancer cells, suggesting the potential antitumour activity of cyclo(Phe-Pro)-related CDPs.

Hoechst 33342 staining showed that 5mM cyclo(Phe-Pro) induced characteristic chromatin condensation and nuclear fragmentation in 18.3 ± 2.8% (p<0.01) of HT-29 cells after 72 hours. Furthermore, annexin V binding revealed that HT-29 cells treated with 5 mM cyclo(Phe-Pro) displayed phosphatidylserine externalization after 48 hours. In addition, it was shown that 10 mM cyclo(Phe-Pro) induced poly(ADP-ribose)polymerase PARP cleavage, one of the hallmark events of apoptosis. The use of the broad-range caspase inhibitor Z-VAD-FMK, showed that this PARP cleavage was caspase-dependent, which in turn was confirmed by demonstrating an increase in caspase-3 activity (p<0.01) in cyclo(Phe- Pro)-treated HT-29 cells. In conclusion, these findings demonstrate that cyclo(Phe-Pro) inhibited the growth of HT- 29, MCF-7, HeLa and WHCO3 cells, and induced apoptosis in HT-29 colon cancer cells, suggesting the potential antitumour activity of cyclo(Phe-Pro)-related CDPs.

Environmental cues and sensory preferences directing the nesting process in loggerhead turtles, caretta caretta, nesting in Maputaland, South Africa

- Description: Animals use their senses for everything on an immediate, and day to day fashion – detecting danger, finding food, finding mates among other activities. In sea turtles and other migratory species senses are used for long-distance migrations. Senses such as smell, vision and hearing, have been studied experimentally under laboratory conditions but seldom have been investigated in the field. This thesis takes a combination of field and laboratory experiments and investigates some of the hypotheses involved in natal homing and nest site selection. The loggerhead’s nesting distribution in Maputaland is distinct with characteristic high and low density nesting areas which are consistent from year to year. Investigations by earlier researchers on these beaches suggested that beach characteristics, such as beach morphodynamic type and beach width, do not appear to influence the beaches at which loggerheads emerge to nest. The high density nesting area (with approximately 440 loggerhead emergences/km) have similar beach characteristics as the low density nesting area (with <50 loggerhead emergences/km). It is therefore suggested that there is another cue that drives nest site selection. It can either be related to a physical characteristic not yet realised, or is a non-physical (but chemical or biological) cue. This dissertation aims to identify the sensory inputs received during the nest selection process, as well as sea-finding ability after nesting. To investigate the mechanism causing the high-density as opposed to the low-density nesting area, three potential drivers were investigated namely: chemical imprinting (as a natal beach cue), ambient and artificial light (as deterrents) and social facilitation (as a learned behaviour). It was also attempted to identify the strength of the most common senses – vision, hearing and smell. As animal ethics restricts interfering with emerging or nesting turtles, the strengths of these senses were tested during sea-finding by adult loggerheads. The results indicate that sulfide concentrations appear to be used as chemical cues for nesting as these concentrations are elevated (>150 percent) in the high density nesting beaches compared to the low density nesting beaches within and among seasons however further investigations are required. Artificial light (range: 0.045–0.5 lux) is an active deterrent of female emergences while ambient light, even under extreme conditions such as lightning during electric storms (up to 8.2 lux), appears to have no observable influence on the spatial or temporal distributions of emergences. Social facilitation appears unlikely as a primary nest site selection factor for loggerheads. It may however, play a minor secondary role to preferred areas or hotspots. Sea-finding in post-nesting female loggerheads appears to be driven exclusively by visual cues such as the light horizon, with minimal to no influence from other cues (the sound of the breakers, slope or smell of the ocean) which solidify the visual system’s use in sea-finding. This research on the nest site selection of loggerheads and the sensory systems involved in this process has added valuable information to the limited pool of knowledge already present and has created a solid framework on which further investigations can be based. Future work in this field should focus on integrating a suite of sensory stimuli and cues to receive a greater understanding of the sensory systems used in nest site selection.
