- Description: This study investigated the information needs and information-seeking behaviour of practising medical doctors at Katutura and Windhoek Central hospitals in Namibia. Anchored on Wilson’s (1996) model of information-seeking behaviour, the purpose of the study was to establish the information required by practising medical doctors to carry out their work, how they seek information, the information sources and resources they use, and the factors that affect them as they do so. Understanding users’ information needs and the way information is disseminated in hospitals is necessary in developing an effective information provision system and ensuring the quality of information services in the hospitals. A mixed-methods research approach was used to meet the objectives of the study. Questionnaires were administered on 140 practising medical doctors at Katutura and Windhoek Central state hospitals, while fifteen doctors were interviewed telephonically, using a semi-structured interview guide. IBM SPSS (Version 25) was used to analyse quantitative data from the questionnaires, while content analysis was used to analyse qualitative data from the interviews. The findings of this research are that practising medical doctors need information mainly for improving clinical decision-making, keeping up-to-date, improving professional knowledge, and continuing education. The study also revealed that medical doctors use a variety of information sources and resources. However, the degree to which information sources are xiii used depends on the clinical context (outpatients, wards and casualty/emergency). Some contextual factors (organisational context, socio-cultural context, and information sources) can either support or hinder doctors in seeking the information they need. The patient is one of the primary information sources for medical doctors. However, language problems are a significant challenge in communicating with patients or patients’ relatives. Lack of patients’ understanding of medical terms and unavailability of properly run libraries in hospitals are some of the factors that affect doctors’ information-seeking behaviour. Participants recommended health education for patients, training of medical doctors on the use of the Internet and ICT, provision of properly run hospital libraries, and improved communication with other international hospitals as some of the ways to improve health information sources and services. Three related models were drawn from the study’s findings, based on Wilson’s (1996) model and literature, namely, the doctors’ decision-making model (DDmM), the clinical context information sources model (CCISM), and the model of information needs and information-seeking behaviour of medical doctors (MINISBMD). The thesis concludes with practical recommendations to enhance the provision of information at Katutura and Windhoek Central hospitals and suggestions for further research.

Institutions, Production and Marketing of maize in Zimbabwe. Case of Marondera District of Mashonaland East Province

- Description: Food security situation in Zimbabwe is deteriorating despite the efforts made by the government for embarking on agrarian reforms in terms of redressing colonial imbalances and taking on the major trading investment risks. The government assured the resettled farmers that if they invest in maize production, they would not be stranded by lack of marketing services as the markets are liberalized. However, this market liberalization policy occurring in maize sector has resulted in A1 resettled farmers becoming more vulnerable to traders preying on their weaknesses as the policy is not living up to expectations and is failing to bring dramatic increases in agricultural growth. This may mean that sending children to school is a nightmare for those who depend on farming for educating their children, inadequate food in the home, unpaid loans and subsequent impoverishment. The resettled A1farmers will remain trapped in the poverty cycle as long as institutions are not corrected. The general aim of the study was to evaluate institutional factors influencing maize production and marketing by A1 resettled farmers of Marondera district in Mashonaland East province of Zimbabwe. Using a sample of 343 farmers randomly selected in Marondera District, different econometric models like Data envelopment Analysis, Cob Douglas, Binary logistic, and Tobit as well as descriptive statistics were used to analyse the data. Institutional variables were captured in two ways: as a dummy showing whether farmers have access to institutional variables and as a continuous variable indicating importance of institutions on maize production and marketing. The study found out that, most of the A1 farmers are old, with a mean age of 47 years and a maximum of 55 years of farming experience. It is also dominated by male farmers who are literate. Results also showed that most of A1 farmers from the sample are married and very few separated. These farmers sell most their maize and keep little for consumption though they are still using convectional farming methods. Most of the farmers from the sample generate their incomes from farming activities only and they have good access to market and production information as well as extension xvi messages through training .They however have poor access to financial credit and weak social capital as well as low contract farming. Results also indicated that A1 farmers pursue the formal supply chains than informal supply chains. It is noted that, A1 farmers participate in a number of markets that include agricultural inputs markets seed, fertilizers, chemicals.; credit markets which includes banks as well as labour markets which can in the form of (maricho). In terms production and marketing efficiency, the study found out that A1 farmers are technically, allocatively and economically inefficient. The maximum efficient score is 1 while the minimum score is 0.01 for both production and marketing stage. The mean scores for production stage are 0.195, 0.504 and 0.072 for technical, allocative and economic efficient respectively. The mean scores for marketing are 0.103, 0.688, and 0.068 for technical, allocative and economic efficiency respectively. Farmers are performing better at production stage than at marketing technically while there is better allocative and economic efficiency at marketing stage. Results for determinants of technical, allocative and economic efficiency at production stage showed that, cattle ownership, farming experience, access to financial credit and (member of farmer organisation) which was used as a proxy indicator for social capital ,access to information, contract farming, distance to input market ,area planted to maize and quantity of seed used influences marketing efficiency of A1 maize farmers in Marondera District. Access to credit, Access to market, distance to market, road infrastructure, farming experience and access to information influences technical, allocative and economic efficiency at marketing stage. Cooperative membership, road infrastructure, contractual farming arrangements, and access to financial credit and transport cost to market influence choice of marketing channel respectively. Finally results from the non linear cob Douglas reveals that, maize production is significantly influenced by contract with value chain actors, access to market xvii information, access to financial credit, total topdressing used , total basal fertilizer used, total herbicides used, and total area put under maize production. The results showed that, future of A1 resettled farmers depends on correcting the institutions which are of paramount importance for maize production and marketing. The study therefore recommended the government to improve institutional environment that has direct link with institutional innovations necessary for sustainable maize production and marketing. This can be done through correcting inefficiencies and ineffective institutions by creating and maintaining both environment and arrangement that will transform maize production and marketing systems to operate efficiently. The study presents policy options to address both production and marketing of maize arguing that new direction of production and marketing of maize depends on correcting dysfunctional institutions. Policy options include increasing A1 farmers access to financial credit by removing the stringent collateral requirements which are not attainable by most A1 farmers as their asset endowment is limited to non capital. Addressing imperfections in the input markets by improving the supply of chemical fertilisers and seed at affordable prices as this influences total area put under maize production. The other policy is for private companies and NGOs to offer contract farming to A1 maize farmers as this will help them to increase total area put under maize production, access to market, access to credit, and access to information and reduces long distances to purchase inputs. Introducing focused training to motivate resettled A1 farmers on formation farmer organisations as this assists in social capital which is an important asset in maize production and marketing.Improving road infrastructure by creating a fund that would be used to maintain the road rather waiting for DDF only.

Job Engagement and Coping Strategies as Moderators of the Relationship between Occupational Stress and Burnout among Police Officers in the Eastern Cape, South Africa

- Description: Police work in South Africa is demanding and stressful. Literature reveals that occupational stress when fully manifest amount to emotional burnout. It was therefore the aim of this study to examine job engagement and coping strategies as moderators of the relationship between occupational stress and burnout among police officers. Using a sample of 377 police officers from the Eastern Cape, South Africa, data was collected using various scales namely; the Effort – Reward Imbalance (ERI) questionnaire (occupational stress), the Maslach Burnout Inventory – General Survey MBI-GS (emotional burnout), the Utrecht Work Engagement Scale (job engagement), and the Coping Orientation to Problems Experienced questionnaire (coping strategies). Correlation tests and regression analysis were ministered on the data to test the research hypotheses. The major finding of the study indicated that job engagement and coping strategies collectively significantly moderate the relationship between occupational stress and emotional burnout. However, job engagement on its own was reported to be an insignificant moderator of the relationship between occupational stress and emotional burnout.

- Description: Knowledge is now regarded as the most important resource surpassing other resources like land and capital. It is a foregone conclusion that knowledge flows out of organizations through various ways such as resignations, retirement and death yet, it does not seem as if organizations are employing strategies to retain such a vital component of production and service delivery. This study investigated the knowledge retention strategies in three Southern Africa Development Community (SADC) public broadcasting organizations of the South African Broadcasting Corporation in South African (SABC), the Department of Broadcasting Services (DBS) in Botswana and the Zimbabwe Broadcasting Corporation (ZBC) in Zimbabwe. The aim of this study was to establish how knowledge is captured and retained at such organizations. The objectives of the study were; to find out the types of strategies the three broadcasting organizations employ to capture and retain knowledge; establish how the three broadcasting organizational cultures enable knowledge acquisition, transfer, sharing and retention; establish the role of ICTs as tools and enablers of knowledge retention, creation, transfer and sharing in the three broadcasting organizations; and to propose recommendations on the best strategies for retention of knowledge in the three public broadcasting organizations as well as suggest areas for further and future research. v The study used the survey method and employed the triangulation design for data gathering. Data were collected from three SADC public broadcasting organizations. The population sample comprised 240 professional staff including 38 managers from different sections of the broadcasting organizations. The researcher collected data by administering a questionnaire to staff, interviews with managers and observations. Documentary analysis was also conducted in order to supplement data and fill in gaps. The researcher was mainly interested in gathering data that would give a comprehensive picture about knowledge retention methods in broadcasting organizations. There were four main findings. First, knowledge management as a relatively new concept and practice has not yet been properly embraced in the public broadcasting organization system in order to capture and retain knowledge that is acquired and generated in the organizations. There are no measures put in place in the organizations to retain the knowledge to ensure that once those who hold vital tacit knowledge are gone, the knowledge may still be available and accessible. Second, the study established that the respective organizational cultures impede on organizational knowledge retention. The study established that there was no culture of knowledge sharing. It also emerged that employees were not free to share their knowledge because of government regulations, prevailing political contexts, mistrust and general lack of incentive to share knowledge. Third, the study established that the organizations have no strategies or systems in place to capture the experts‟ knowledge or the knowledge of experienced staff and vi those approaching retiring age. The Human Resources Departments hold exit interviews that cover issues purely to do with why the person is leaving the organization instead of recording the work experiences and expertise of the departing employees. Fourth, the study revealed that the public broadcasting organizations still lag behind in collaborative and communicative technologies that facilitate knowledge transfer and sharing of tacit knowledge but retention of knowledge generally. The study also found that, some of the organizations studied did not have internet connectivity and websites were not yet functional, which hampered acquisition and sharing of knowledge for retention purposes. - Description: Aromatic pollutants are a diverse group of chemicals which are continuously produced from industrialization, urbanization and sophistication in technological advancement. Pristine water source polluted by these chemicls makes the water unsafe for human consumption, and as well disrupts the trophic structure of the aquatic milieu. Physico-chemical treatment techniques employed so far have been accompanied by major drawbacks which have overriden the relative successes recorded, hence, greener, simpler and more efficient methods of pollutant transformation is imperative. The prospect of enzymatic treatment of pollutants has gradually been receiving growing attention in contemporary times due to the their environmental friendliness and production economic feasiblity. Laccase, a multicopper oxidase has heightened its appeal towards environmental and biotechnological applications due to its broad substrate specificity and its requirement of atmospheric molecular oxygen as a cosubstrate and the discharge of water as the byproduct. Hence, this present study was designed to evaluate the biotechnological potentials of laccases produced by some bacteria species from some aquatic biomes of the Eastern Cape Province, South Africa. The laccase-producing bacteria were isolated from selected environmental samples by selective enrichment using selective aromatic compounds as sole carbon source and subsequently, laccase-screening phenolic substrates. The laccase-producing bacteria were identified by molecular techniques as proteobacteria belonging to the following genera: Achromobacter, Bordetella, Citrobacter, Pseudomonas and Stenotrophomonas. Optimisation of laccase production in a submerged fermentation was by traditional and statistical methods, where four isolates (Hb9c; Achromobacter xylosoxidans HWN16, Hb16c; Bordetella bronchisepta HSO16, Berl11b2; Stenotrophomonas maltophilia BIJ16, Ie1c; Citrobacter freundii LLJ16) were evaluated for the fermentative production of laccase from lignocellulosic agroindustrial residues. Predictions from statistical optimisation showed that weakly acidic conditions (pH 5) and low agitation speed (100 rpm) were required for maximum laccase production from mandarin peelings (0.5 g/200 mL) and NaNO3 (0.25 g/200 mL) in Hb9c, maize stover (0.50 g/200 mL) and NaNO3 (0.050 g/200mL) in Berl11b2 while a lower agitation speed (50 rpm) was required for maximum laccase output from 2.0 g/200 mL maize stover and 0.050 g/200 mL KNO3 in Ie1c. However, 2.50 g/200 mL wheat bran, 0.050 g/200 mL yeast extract and 50 rpm agitiation under acidic conditions (pH3) yielded maximum laccase titres in Hb16c. Further characterisation of Hb9c and Ie1c laccase secretions portrayed their polyextremotolerant capacities. They were active at a broad range of tempertaure (0-90 degreesC); with optima at 70°C (Hb9c) and 60°C (Ie1c), pH (3-11); with optima at pH 6 (Ie1c) and pH 8 (Hb9c), respectively, and were equally thermo- and pH-stable. Their activities were either improved or left unabated by high concentrations of cations, detergents, and chloride. In addition, catalytic activities of Hb9c and Ie1c laccase secretions increased when they were preincubated with 2 – 20 percent of fluoride, a potent inhibitor. Consequently, a molecular perspective depicted the isolates to have multiple homologous laccase encoding genes. The enzymes were successfully immobilised on solid supports comprising gelatin and Na-alginate with a recovery of cca. 85 percent residual activity after 8 cycles of oprertional stability experiments. The immobilised laccases were remarkable in the decolourisation of synthetic dyes, albeit, free forms also elicited satisfactory performances. Ultimately, the application of free laccases in denim bleaching, individually or with a blend of a mediator, ABTS, showed that denim colours could be bleached without the need for chemical bleaching agents. The results obtained suggest the bacteria laccases produced from lignocellulosic wastes may serve as potent degraders of phenolic pollutants in water and, may also contribute to the bioeconomy and promote greener techniques for industrial applications.

However, due to a variety of reasons, including the dominance of small-scale farming and challenges of land reform, agricultural productivity remains low. Moreover, despite land reform, including massive land redistribution, the process appears incomplete and inconclusive. This is so essentially because the land reform neither increased food production nor reduced poverty. Accordingly, sustainable development remains a distant reality, especially among rural small-scale farmers. The study aimed at assessing the impact of the fast track land reform (FTLRP) on poverty reduction and sustainable rural development in Zimbabwe. Despite undertaking one of Southern Africa’s largest post-colonial redistributive land reform to date, the provision of land on its own has not been sufficient to address rural poverty and achieving meaningful rural development. This is so because the land reform has not articulated integrated growth as remoteness and poverty remain endemic, which is connected to the land. So, resolving the land question still remains crucial for sustainable rural development in Zimbabwe.

Land rental policy and land market in Mashonaland East Province, Zimbabwe: implications on farmer decisions, efficiency and equity under A1 and A2 models

- Description: Government promulgated the land rental policy starting in the year 2007. The policy required that A1 (market residual) and A2 (indigenous commercial) farmers pay rentals to the state. Since then, there has also been an emerging trend in which farmers have been renting in and out land, resulting in an informal market for land rentals. The study therefore assessed farmers’ perceptions of land rental policy and evaluated the possible association of this policy to farmers’ decision making on land rental markets as well as its implications on farmers’ efficiency and equity. The study was carried out in Mashonaland East Province of Zimbabwe, covering two districts, Goromonzi and Marondera. The focus was specifically on A1 and A2 farmers, as the rental policy was directed at these resettlement models only and much of the informal land rental markets were prevalent among these categories of farmers. Data collection utilised the survey approach. This was supported by key informant interviews and focus group discussions, with a final sample of 339 households selected using multi-stage sampling method. Survey data were transcribed on CsPro 6, and analysed using Stata, SPSS and Frontier 4.1. Results showed that about a third of household respondents was from Marondera and twothirds was from Goromonzi, while 79% and 21% was A1 and A2 farmers respectively. About 80% was male headed households and the remainder was female headed households. About half of the households were not participating in land rental markets while the other half was split almost equally between those renting-in and renting-out land. In general, A2 farmers were better in agricultural productivity than A1 farmers. It was concluded that farmers had a fairly good knowledge of the agricultural land rental policy enunciated in the Finance Bills. A1 farmers were more knowledgeable than A2 farmers, but no vii significant differences were observed between male and female headed households and farmers involved and not involved in land rental markets. Farmers’ attitudes in relation to policy were categorised as fair, with significant differences based on gender and land market participation. Practice scores showed a poor adherence to policy for all categories of farmers. Overall perceptions of farmers on rental policy was inferred as fair with significant differences existing between land rental market participants and non-participants (autarky). The results of a bivariate Tobit model results showed that the decision to rent-in land was significantly influenced by gender, household income, permanent labour, cultivated area, tenure certainty, irrigable land size and crop diversification. On the other hand, age, permanent labour, irrigable land size and crop diversification significantly influenced farmers’ decisions to rent-out land. The conclusion was that household characteristics, land endowments and transaction costs significantly influenced the decision to rent-in land while the former two were strong in influencing renting-out decisions. Results from the linearized Cobb Douglas model showed that economic efficiency for most farmers was above 50%. Farmers renting-out land under A1 were found to be the most economically efficient, followed by those who were renting-in and least efficient were farmers not participating in land rental markets, though the differences were marginal. For the A2 model, renting-in farmers were the most efficient, followed by those in autarky position, while renting-out farmers were the least. Overall, the most efficient farmers were those renting-in, followed by those renting-out while farmers in autarky were the least efficient. Overall efficiency was higher for A2 farmers than for A1 farmers. For farmers not participating in rental markets, the sources of inefficiency were crop type, crop area and labour. For farmers renting-in, it was proportion of irrigable land, size of irrigable land, crop type, crop area and viii labour, while inefficiency drivers for renting-out farmers were crop type and associated area. Equity analysis showed that participation in land rental markets reduced inequality for farmers in the two districts and male and female headed households. Inequality was increased among A1 farmers and remained unchanged among A2 farmers. Overall, the emerging position was that participation in land rental markets resulted in higher efficiency and reduced inequality in land holding among the sampled farmers. It is recommended that government should be consistent on land rental policy and bring into place effective administration of land rental policy. Government may also consider formal acceptance of land rental markets in light of the marginal efficiency and equity benefits shown in the study. However, formalizing land rental markets alone may not be a panacea to improved efficiency and needs to be supported by other productivity measures given the average efficiency values for farmers. Crop and livestock production based on compatibility with the natural regions and defined minimum size of land should be encouraged to improve efficiency

- Description: School-based violence amongst students in South Africa is a growing concern. Such violence is a great concern to all stakeholders involved in the school system. The more violence escalates; the more students are less likely to concentrate on their academic activities which then reduces their chances of success. There are weekly report broadcasts on the media on the death of students across the globe due school-based violence. School-based violence needs to be curbed and schools should be a safe environment where learning and teaching takes place not a battlefield where students live in fear of being the next victim. The aim of this study was to examine students’ experiences of school violence in Sarah Baartman District Municipality, Eastern Cape Province, South Africa. A qualitative approach and an exploratory research design were used in conducting this study as these approached allowed participants to share first hand experiences regarding the issue under investigation. A purposive sample was employed to select participants for the study. This technique allowed the researcher to select participants who displayed most of the characteristics of the larger population and who had sufficient knowledge on the issue under investigation. Forty participants were selected for the study as follows: 25 learners; 5 social Workers; 5 educators; and 5 members of the Representative Council of Learners from schools mostly affected by school-based violence. Data was obtained through semi-structured interviews; focus group discussions with students, social workers and educators; and observations. Data was analysed thematically and supported by the relevant literature. The findings revealed that there are high levels of school-based violence in Sarah Baartman District Municipality, however, the whole-school approach as a measure of combatting school violence, has proved to work in some parts of the Eastern Cape, the Western Cape and Kwazulu Natal provinces. Also, retributive forms of punishment show more positive results against corrective /corporal punishment. It is, therefore, recommended that the state should employ school and place social workers in schools in order to assist in addressing social issues within the school system. Stamping out inequalities among students will ultimately decrease school violence.

Listening to silenced voices: The lived experiences of Xhosa mothers during the initiation period of their sons

- Description: The aim of this study was to explore and describe the lived experiences of Xhosa mothers during the initiation period of their sons. The male rite of initiation among the amaXhosa has various taboos that prohibit women from getting involved in decision-making regarding the ritual. This creates a challenge as the number of female-headed households are increasing. The rationale for this study was to give Xhosa mothers a voice to relate their narratives of how they experienced the initiation period of their sons. In order to carry out this aim, the paradigm of Afrocentricity was used to anchor the study. Afrocentricity is a paradigm that supports the agency of African people in a study conducted about their issues. It also aims to give the previously oppressed groups a voice so that they can speak for themselves. Interpretative Phenomenological Analysis (IPA) as set out by Jonathan Smith was used as a qualitative research methodology in this study. Data was generated, recorded and transcribed from semi-structured interviews with eight selected participants. The biological mothers of Xhosa initiates, that experienced the initiation of their sons, took part in this study. The over-arching inclusion criteria were that the mothers gave permission for their sons to be initiated, and that their sons did not experience any major complications during the ritual. They met the inclusion criteria as set out by the researcher. The findings of the study indicated that the mothers’ experiences were multifaceted. They included the spiritual, psychological and emotional experiences, as well as their experiences as women, and, more especially, as mothers. The challenges that mothers faced during this period included lack of feedback regarding the health of their sons. This led to the experiencing of somatic symptoms like lack of sleep, poor appetite, and anxiety. In order to deal with their symptoms, the main coping mechanism that the participants used, was prayer. - Description: The aim of this study was to explore and describe the lived experiences of Xhosa mothers during the initiation period of their sons. The male rite of initiation among the amaXhosa has various taboos that prohibit women from getting involved in decision-making regarding the ritual. This creates a challenge as the number of female-headed households are increasing. The rationale for this study was to give Xhosa mothers a voice to relate their narratives of how they experienced the initiation period of their sons. In order to carry out this aim, the paradigm of Afrocentricity was used to anchor the study. Afrocentricity is a paradigm that supports the agency of African people in a study conducted about their issues. It also aims to give the previously oppressed groups a voice so that they can speak for themselves. Interpretative Phenomenological Analysis (IPA) as set out by Jonathan Smith was used as a qualitative research methodology in this study. Data was generated, recorded and transcribed from semi-structured interviews with eight selected participants. The biological mothers of Xhosa initiates, that experienced the initiation of their sons, took part in this study. The over-arching inclusion criteria were that the mothers gave permission for their sons to be initiated, and that their sons did not experience any major complications during the ritual. They met the inclusion criteria as set out by the researcher. The findings of the study indicated that the mothers’ experiences were multifaceted. They included the spiritual, psychological and emotional experiences, as well as their experiences as women, and, more especially, as mothers. The challenges that mothers faced during this period included lack of feedback regarding the health of their sons. This led to the experiencing of somatic symptoms like lack of sleep, poor appetite, and anxiety. In order to deal with their symptoms, the main coping mechanism that the participants used, was prayer. Litter quality effects on soil stabiltiy and erodibility in the Ntabelanga Area, Eastern Cape, South Africa

In South Africa (SA), most soils have low (< 4 percent) soil organic matte (SOM) contents, making them susceptible to soil erosion. Little is known about SOM quality effects in soil stabilization in the SA context, it is therefore difficult to advocate specific organic matter sources in order to stabilize a particular soil. This study evaluated effects of different litter quality sources (Vachellia karoo (sweet thorn) leaf litter and Zea mays (maize) stover) on soil aggregate stabilization at a soil micro-level (soil aggregate), guided by the following specific objectives; (i) to characterize soil properties that influence the erodibility of soils in the Ntabelanga area, Eastern Cape (EC) Province, SA (ii) to assess fractionation of organic carbon during decomposition of different litter sources (iii) to determine rate of soil macroaggregate re-formation under different litter amendments (iv) to determine the influence of different litter quality on splash soil erosion and (v) to delineate the Ntabelanga area into different soil erosion sensitivity zones. Soil samples collected from a proposed Ntabelanga dam site, EC Province, SA, were characterized for factors influencing erodibility. Soil physical and chemical properties were analyzed, soil erodibility (K-factor) estimated using SOC content and surface properties and soil loss rates by splashing determined under rainfall simulations at 360 mm h-1 rainfall intensity. Soil loss was then correlated with the chemical and physical soil properties. SOC showed significant (P< 0.05) inverse relationship with soil loss. Soils samples were bulked to seven composite samples according to major soil horizons (orthic A, melanic A, pedocutanic B, red apedal B, saprolite, G horizon and prismacutanic B) in the areas of soil associations, macroaggregate (> 0.250 mm) were removed by passing through a 0.250 mm sieve. The remaining soil microaggregates (< 0.250 mm) were then mixed with high quality Vachellia karoo leaf litter (C/N = 23.8) and low quality Zea mays stover (C/N = 37.4) and incubated at 23oC for 30 weeks. Jars were arranged as a 7 × 3 factorial laid in completely randomized design (CRD) with three replicates. Data was repeatedly measured at 1, 3, 8, 14, 23 and 30 weeks after incubation: to determine the stabilization of SOM by fractionating the soil particulate organic matter (POM), rate of soil macroaggregate re-formation and influence of litter quality on splash soil erosion along a decomposition continuum. Free light fractions (Free LF), coarse particulate organic matter (CoPOM) and fine particulate organic matter (FiPOM) within macroaggregates (> 250 μm) decreased exponentially from week 1 to 30 and were significantly (p < 0.05) influenced by litter quality × soil horizon × time interactions. A similar trend for FiPOM fractions in macroaggregates was observed in the 53-250 μm microaggregates (p < 0.05). The greatest influence on changes in POM fractions occurred within the first 3 weeks of incorporation. Both V. karroo leaf litter and Z. mays stover significantly stabilized the macroaggregates within the first 3 weeks after incubation. Cumulative macroaggregates yields, mean weight diameter (MWD), percentage water stable aggregates (%WSA) and whole soil stability index (WSSI) in litter amended soils increased up to week 8 of incubation and thereafter gradually declined in all soils. An increase in macroaggregation resulted in increased MWD, WSSI values and large and small aggregates distribution. Aggregation was significantly higher in soils with higher clay content than sand content, suggesting that soil texture was highly influential in litter decomposition. Rate of litter decomposition was influenced by soil type × time interactions which determined extent and macroaggregation dynamics along a decomposition continuum. The amount of splashed sediments was determined from each soil horizon at 360 mm h-1 simulated rainfall intensity applied as either single 8minute rainstorm (SR) or 4 × 2minute intermittent rainstorms (IR) separated by a 72 h drying period. Results showed a reduction in splashed sediments under IR and SR storms in litter-amended soils during the first 8 weeks of incubation with gradual increase thereafter. More sediments were splashed under IR than SR and litter quality had no influence on splashed sediments per soil horizon. Soil horizons profile with more clay than sand particles enhanced the litter effects on the soil resistance against detachment. The amount of SOC loss was influenced by primary particle size distribution and initial SOC content of the soil. Rainfall pattern and the initial SOC content were the main factors by which different soils influenced SOC loss. More rainstorm patterns should be investigated on these soils.

Livelihood and unemployment transitions : drivers and effects on welfare in Nigeria

- Description: Hunger, unemployment and social crisis are increasing in Nigeria. Statistics supports it that most of the able-bodied youth, both male and female, are not in employment, school or training in Nigeria while poverty rate is as high as a decade ago. A number of research efforts have looked at the micro and macroeconomic paths of the problem with less attention on dynamism of unemployment vis-a-vis the labour market transition, hence this study. This study investigates dynamism of labour market by assessing pattern and transition in labour market and out of labour, as well as determining the effect size of households head’s unemployment on household welfare. To achieve this objective, the General Household Survey (GHS) panel data for 2010/211 and 2012/2013 for Nigeria was used coupled with panel regression analyses to identify determinants of transitions from unemployment to productive sectors, and determinants of transition from productive sectors to unemployment, as well as effect of unemployment on household welfare while controlling for potential endogeneity in unemployment. Across the productive sectors and between 2010 and 2013, the descriptive results of transition show that workforce in the services sector (68 percent) are the most persistent, followed by agriculture (59 percent) and least is mining and manufacturing (44 percent). Transition from nonemployment occurred the most to services sector (22 percent), followed by out-of-labour force (18 percent), agriculture (15 percent) and least to mining and manufacturing (5 percent). Further, the results show that there are both locations and gender differences in the extent of transitions in the labour market. The econometric analyses reveal that job search characteristics, age, educational levels, gender, family ties, locations and time significantly determine transition from unemployment to productive sectors (agriculture or non-agriculture). Likewise, job characteristics, demographic variables, social network, location and time significantly determine switch from one productive sector to another or to unemployment. Moreover, it explicitly reveals as well from the analysis that household head month of unemployment reduces household total consumption and food consumption by 6.4 percent with gender and sectorial differences. The study recommends a combined and swift intervention on three sides: one, there is a need to stimulate education either through scholarship, or adult education from both public and private sectors; two, need to create jobs for youth to reduce potential social and economic crisis; and lastly there is a need to provide social support either in terms of public workfare, skills for jobs and unemployment benefits.

The local web newspapers framing suggests that there are numerous channels such as natural food production, livestock production, smallholder farming that can enhance availability and access to affordable food to people in the Eastern Cape Province. The findings revealed that local web newspapers frame increased agricultural production as an important issue that demand serious and urgent attention of the Provincial government as it has the potential of not only ensuring food security but also impacting positively on the income of local farmers. The local web newspapers frame farm attacks, land redistribution and reinstatement as important factors that hinder food production and, ultimately, food security in the Eastern Cape v Province. Furthermore, this study discovered that the frame of references used by the local web newspapers, especially the emphasis on local farming, land redistribution and private sector help to empower and mobilize individuals and communities to participate in food security initiatives. This study concludes that Eastern Cape Local Web Newspapers can be veritable tools to enhance food security channels in the Eastern Cape Province

Mechanism and synchronicity of wheat (Triticum aestivum) resistance to leaf rust (Puccinia triticina) and Russian wheat aphid (Duiraphis noxia) SA1

- Description: Wheat (Triticum aestivum and T. Durum) is an extremely important agronomic crop produced worldwide. Wheat consumption has doubled in the last 30 years with approximately 600 million tons consumed per annum. According to the International Maize and Wheat Improvement Center, worldwide wheat demand will increase over 40 percent by 2020, while land as well as resources available for the production will decrease significantly if the current trend prevails. The wheat industry is challenged with abiotic and biotic stressors that lead to reduction in crop yields. Increase knowledge of wheat’s biochemical constitution and functional biology is of paramount importance to improve wheat so as to meet with this demand. Pesticides and fungicides are being used to control biotic stress imposed by insect pest and fungi pathogens but these chemicals pose a risk to the environment and human health. To this effect, there is re-evaluation of pesticides currently in use by the Environmental Protection Agency, via mandates of the 1996 Food Quality Protection Act and those with higher perceived risks are banned. Genetic resistance is now a more environmental friendly and effective method of controlling insect pest and rust diseases of wheat than the costly spraying with pesticides and fungicides. Although, resistant cultivars effectively prevent current prevailing pathotypes of leaf rust and biotypes of Russian wheat aphid from attacking wheat, new pathotypes and biotypes of the pathogen/pest may develop and infect resistant cultivars. Therefore, breeders are continually searching for new sources of resistance. Proteomic approaches can be utilised to ascertain target enzymes and proteins from resistant lines that could be utilised to augment the natural tolerance of agronomically favourable varieties of wheat. With this ultimate goal in mind, the aim of this study was to elucidate the mechanism and synchronicity of wheat resistance to leaf rust (Puccinia triticina) and Russian wheat aphid (Duiraphis noxia) SA1. To determine the resistance mechanism of the wheat cultivars to leaf rust infection and Russian wheat aphid infestation, a proteomics approach using two-dimensional gel electrophoresis was used in order to determine the effect of RWA SA1 on the wheat cultivars proteome. Differentially expressed proteins that were up or down regulated (appearing or disappearing) were identified using PDQuestTM Basic 2-DE Gel analysis software. Proteins bands of interest were in-gel trypsin digested as per the protocol described in Schevchenko et al. (2007) and analysed using a Dionex Ultimate 3000 RSLC system coupled to an AB Sciex 6600 TripleTOF mass spectrometer. Protein pilot v5 using Paragon search engine (AB Sciex) was used for comparison of the obtained MS/MS spectra with a custom database containing sequences of Puccinia triticina (Uniprot Swissprot), Triticum aestivum (Uniprot TrEMBL) and Russian wheat aphid (Uniprot TrEMBL) as well as a list of sequences from common contaminating proteins. Proteins with a threshold of ≥99.9 percent confidence were reported. A total of 72 proteins were putatively identified from the 37 protein spots excised originating from either leaf rust or Russian wheat aphid experiments. Sixty-three of these proteins were associated with wheat response to stress imposed by RWA SA1 feeding while 39 were associated with infection by Puccinia triticina. Several enzymes involved in the Calvin cycle, electron transport and ATP synthesis were observed to be differentially regulated suggesting greater metabolic requirements in the wheat plants following aphid infestation and leaf rust infection. To this effect, there is re-evaluation of pesticides currently in use by the Environmental Protection Agency, via mandates of the 1996 Food Quality Protection Act and those with higher perceived risks are banned. Genetic resistance is now a more environmental friendly and effective method of controlling insect pest and rust diseases of wheat than the costly spraying with pesticides and fungicides. Although, resistant cultivars effectively prevent current prevailing pathotypes of leaf rust and biotypes of Russian wheat aphid from attacking wheat, new pathotypes and biotypes of the pathogen/pest may develop and infect resistant cultivars. Therefore, breeders are continually searching for new sources of resistance. Proteomic approaches can be utilised to ascertain target enzymes and proteins from resistant lines that could be utilised to augment the natural tolerance of agronomically favourable varieties of wheat. With this ultimate goal in mind, the aim of this study was to elucidate the mechanism and synchronicity of wheat resistance to leaf rust (Puccinia triticina) and Russian wheat aphid (Duiraphis noxia) SA1. To determine the resistance mechanism of the wheat cultivars to leaf rust infection and Russian wheat aphid infestation, a proteomics approach using two-dimensional gel electrophoresis was used in order to determine the effect of RWA SA1 on the wheat cultivars proteome. Differentially expressed proteins that were up or down regulated (appearing or disappearing) were identified using PDQuestTM Basic 2-DE Gel analysis software. Proteins bands of interest were in-gel trypsin digested as per the protocol described in Schevchenko et al. (2007) and analysed using a Dionex Ultimate 3000 RSLC system coupled to an AB Sciex 6600 TripleTOF mass spectrometer. Protein pilot v5 using Paragon search engine (AB Sciex) was used for comparison of the obtained MS/MS spectra with a custom database containing sequences of Puccinia triticina (Uniprot Swissprot), Triticum aestivum (Uniprot TrEMBL) and Russian wheat aphid (Uniprot TrEMBL) as well as a list of sequences from common contaminating proteins. Proteins with a threshold of ≥99.9 percent confidence were reported. A total of 72 proteins were putatively identified from the 37 protein spots excised originating from either leaf rust or Russian wheat aphid experiments. Sixty-three of these proteins were associated with wheat response to stress imposed by RWA SA1 feeding while 39 were associated with infection by Puccinia triticina. Several enzymes involved in the Calvin cycle, electron transport and ATP synthesis were observed to be differentially regulated suggesting greater metabolic requirements in the wheat plants following aphid infestation and leaf rust infection. The second objective was to assess the therapeutic value of the four most frequently used plant species by THs by (a) analysing the phytochemical profile of the plant extracts, (b) their antioxidant activities, and (c) their antibacterial activities. Information linked to the social and ethnobotanical surveys was gathered via a semi-structured questionnaires, supplemented by field observations during face-to-face interview with 240 conveniently selected THs practicing in the municipalities of Capricorn, Sekhukhune and Waterberg districts of the Limpopo Province, South Africa. Data were generally analysed using Microsoft Excel and Statistical Package for the Social Sciences (SPSS). In addition to these softwares, ethnobotanical data were analysed via quantitative ethnobotanical indexes namely use mention (UM), fidelity level (FL), use value (UV) and preference ranking (PR). For laboratory experiments, qualitative phytochemical constitutes of the crude extracts were determined using the standard screening methods. The antioxidant activities of acetone, dichlomethane, hexane, methanol and water crude extracts were evaluated using qualitative and quantitative 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assays. Antibacterial activities of these extracts was assessed using microdilution and bioautography assays. Six types of RIs; tuberculosis (84.1%), sore throat (65%), asthma (58.3%), sinusitis (55%), pneumonia (53.3%) and rhinitis (43.7%) were treated by THs practicing as herbalists (58.7%), herbalists and diviners (40.5%), and diviners (0.4%). The identification of these RIs was based on the patients’ presentation of certain clinical symptoms, with THs practicing both herbalism and divination additionally using bone casting. There was a broad overlap between the clinical signs used to diagnose most of the RIs, and the specificity of symptoms followed when ascertaining an individual infection was low. A total of 224 plant species (83% indigenous and 16.9% exotics) belonging to 177 genera and 85 botanical families, mainly the Fabaceae (25 spp.) and Asteraceae (16 spp.) were used by THs as herbal medicines for the aforesaid RIs and perceived related symptoms (RSs). Most of the documented plant species were used for tuberculosis (82.1%) and asthma (46.4%). Roots (64%) and leaves (13%) formed an important mainstay for medicinal recipes preparations. Different routes of administering these recipes were noted, with oral (75.6%) and nasal (18.8%) being the most preferred. The most widely used species by THs to treat and manage all the six RIs with high UM, FL, UV and PR values included Clerodendrum ternatum, Cryptocarya transvaalensis, Enicostema axillare, Lasiosiphon caffer and Stylochaeton natalensis. Overall larger numbers of species used by Bapedi THs including the mentioned ones are recorded for the first time in scientific literature as remedies for RIs and RSs, thus reflecting a need for more similar studies amongst other ethnic groups of South Africa and elsewhere. The phytochemical screening of four most used plants C. ternatum, C. transvaalensis, E. axillare and L. caffer crude extracts revealed the presence of alkaloids, anthraquinones, cardiac glycosides, flavonoids, phlobatannin, saponins, steroids, tannins and terpenes. All plants including those that did not display any antioxidant activities using qualitative DPPH assay displayed a certain level of scavenging activity when quantified, with the mentioned extracts from E. axillare (water and hexane), L. caffer (water and acetone) and C. ternatum (acetone) showing excellent activity almost comparable to a standard antioxidant drug (ascorbic acid). Plant extracts from all used solvents were active against Mycobacterium smegmatis, Klebsiella pneumoniae, Staphylococcus aureus and Pseudomonas aeruginosa with miniumum bactericidal concentration values ranging between 0.019 and 2.5 mg/ml depending on the plant species. Some of plant extracts exhibited minimum inhibitory concentration values ranging between 0.63 to 2.5 mg/ml. This study showed that most of the diagnosis methods used by Bapedi THs and their herbal medicines have the potential to contribute towards the management and treatment of RIs and RSs. iii | Page Future endeavour should be focused on the following: (a) possibility of collaboration between questioned THs and local western health care practitioners in the treatment of these diseases, and (b) isolation, purification and characterization of the biologically active compounds from extract of the above-stated plants, amongst other inquiries.

The study adopted the human centered mineral resource governance approach to assess the mineral governance structure in Ghana and identified that, the current system is a colonial legacy and does not promote human development in Ghana. The study, whilst recognizing the legacies of colonialism, argues that in order for mineral resources to be managed responsibly to promote human development, there should be a paradigm shift to focus on how mineral resources are governed. As a result, the study, relying on the human centered development approach investigates how mineral resources can be governed to promote development in Ghana, with specific focus on the mineral wealth Western Region of the country. Due to the finite nature of mineral resources, the human centered development approach argues that revenues from mineral resources should be invested in human capacity building and medical care delivery. Further, the mining sector should be integrated with other non-mining sectors of the economy to create jobs and provide alternative livelihoods which will go a long way to assist in the general expansion and growth of the country‟s economy. Although this thesis is a case study of mineral governance in Ghana, it can serve as a springboard to appreciate the role mineral governance can play in promoting people centered development in other sub – Saharan mineral wealth countries.

Molecular characterization of Mycobacterium bovis isolated from cattle and sputum specimens from humans in the Eastern Cape province, South Africa : public health and economic implication

- Description: Tuberculosis (TB) is a disease caused by the members of the Mycobacterium tuberculosis complex (MTBC), which infects both animals and humans. The primary host of Mycobacterium bovis is cattle. It remains a major global health problem. About 2 billion people are thought to be infected with TB and about 1.3 million decease each year from the disease. This study is aimed at determining the prevalence of the disease, characterizing the MTBC isolates and increasing the understanding of the genetic diversity MTBC in the Eastern Cape Province of South Africa, from studying both samples from both humans and cattle. A total of 376 cattle lymph nodes were collected from two commercial abattoirs for investigations. Suspected 6 000 sputum specimens were also collected from humans (cattle farmers). All collected samples were cultured, and isolates were confirmed with a multiplex polymerase chain reaction (PCR) targeting the mpb64 and IS6110 genes. Positive isolates were tested for resistance against anti-TB drugs using the standard Lowenstein Jensen proportion method. Isolates from cattle lymph nodes were further characterized to species level, using the region of deletion 1 (RD1) and oxyR restriction fragment length polymorphism analyses. MIRU-VNTR typing and spoligotyping assays were used to further examine the genetic diversity of all strains. Obtained spoligotype patterns were matched with isolates published in the universal spoligotyping database (SITVIT2). MTBC was detected in 162 (43.09 percent) cattle lymph nodes and 156 sputum specimens. Some isolates 25.9 percent (42/162) from the cattle lymph nodes samples were resistant to rifampicin (RIF) and 98.7 percent (154/156) were resistant to isoniazid (INH) and rifampicin (RIF) (of the human sputum specimens). All cattle lymph nodes isolates exhibited the RD1 gene, and the oxyR analyses classified 96.9 percent of the isolates as Mycobacterium tuberculosis complex and 3.1 percent as Mycobacterium bovis. Twenty seven spoligotype patterns were isolated from cattle lymph nodes, with 10 shared types (SIT) consigned to 5 lineages, including Bov_4-Caprea (1.9 percent), Microti (0.6 percent) and Beijing (17.9 percent) constituting East Asian, Latin-American-Mediterranean (LAM) (3.7 percent), X (4.3 percent), MANU (3.1percent), S (0.6 percent) and 67.9 percent were orphans that were further categorized to their designated lineages (Indo-Oceanic, Euro-American and West-African 2). Ninety one percent of the isolates from humans (farmers) were grouped into 7 clusters by spoligotyping, while the remaining 9 percent was unclustered. SITs were observed in 89.6 percent of the isolates from the Beijing, LAM, X, T and S families. The Beijing family was the most prevalent strain from both human isolates (77.2 percent) and cattle lymph nodes isolates (17.9 percent), and the T (0.6 percent) and Microti (0.6 percent) families were the least detected genotypes in both humans and cattle lymph nodes. In the cattle lymph nodes, spoligotyping showed a higher clustering rate of 82.1 percent, with the lowest (HGDI) = 0.485. The 12 MIRU-VNTR clustering rate was 64.8 percent, showing a higher HGDI of 0.671. However, a combination of both methods showed a decreased clustering rate of 53.7 percent; however these methods resulted in a higher HGDI (0.676). Isolates from human sputum specimens spoligotyping showed a higher clustering rate of 89 percent with an HGDI = 0.822. The 12 MIRU-VNTR showed the lowest clustering rate of 69 percent and an HGDI = 0.940 and the combination of the methods showed a clustering rate of 78 percent and an HGDI = 0.951. The results of this study show the diversity of MTBC strains in the Eastern Cape Province and the low clustering rate indicates continuing transmission in the province. The detection of MTBC strains from slaughtered cattle lymph nodes in abattoirs confirms the risk of providing TB infected meat in markets which has serious implications for the control of TB in South Africa. The continuing spread of MTBC in South Africa threatens both the public and economic health.

- Description: Streptococcus agalactiae (S. agalactiae) also known as group B Streptococcus (GBS) is one of the leading causes of bacterial morbidity and mortality among neonates worldwide. It is the cause of invasive Early Onset Disease (EOD), which occurs in the first 7 days of life and characterised by sepsis, pneumonia and meningitis and Late Onset Disease (LOD) occurring between 7 and 89 days of life. Late onset disease is characterised by meningitis and long term neurological sequelae such as cerebral palsy, hearing impairment and cognitive challenges. S. agalactiae does not only infect neonates, it also infects the elderly, immunocompromised individuals and pregnant and non-pregnant women, causing invasive disease. In the world, 10-40 percent of healthy women are rectally or vaginally colonised with GBS and they face the risk of passing it to their babies during the process of childbirth. During parturition, a GBS colonized pregnant woman transfers the bacterium to her new-born as the baby passes through the ruptured membrane, thus infecting the child. However, GBS has been reported to be transferred even without rupture of membranes. Once it infects the membranes, it is transferred into the amniotic fluid and subsequently infects the baby. It can be aspirated into the lungs causing pneumonia or it can infect the blood stream and disseminated round the body causing septicaemia, meningitis and other infections. Once in the neonate’s body, the bacteria is able to evade the immune system as the host immune system is not yet fully developed. Bacterial evasion of the immune system is enhanced by its various virulence factors which are deployed to help it escape the immune system. These include the polysaccharide capsule, haemolysin and the release of complement inactivating factors such C5a peptidase. The World Health Organisation (WHO) (2010) recommends universal screening of pregnant women to identify those colonised and who are at risk of passing the bacterium to their babies during birth. WHO also recommends identification of at risk women and providing Intrapartum Antibiotic Prophylaxis (IAP) using penicillin. However, problems arise in penicillin allergic women and while alternatives for IAP include erythromycin and clindamycin, there is increasing resistance to these drugs thereby limiting therapeutic options. Antimicrobial susceptibility testing is also not always possible in most resource constrained countries due to poor infrastructure, limited access to health care and the logistical problems in implementing the WHO guidelines. Alternative therapeutic options to GBS infection include developing new and potent antibiotics, development of a vaccine, use of medicinal plants and the use of bacteriophage therapy. While these look like better alternatives there is massive scientific work to be carried out to ensure proper characterisation and efficiency of such alternatives. This process should be followed by in vitro diagnostic testing, experiments with animal models and clinical trials. The problems encountered during vaccine development to curtail GBS infection are compounded by the multiplicity of S. agalactiae capsular types which vary in different geographic locations. Medicinal plants are a cheap and convenient option since they are widely used in communities but the phytochemical components of the plants have to be identified and subjected to in vitro testing to evaluate their therapeutic efficacy as antimicrobial agents. - Description: Streptococcus agalactiae (S. agalactiae) also known as group B Streptococcus (GBS) is one of the leading causes of bacterial morbidity and mortality among neonates worldwide. It is the cause of invasive Early Onset Disease (EOD), which occurs in the first 7 days of life and characterised by sepsis, pneumonia and meningitis and Late Onset Disease (LOD) occurring between 7 and 89 days of life. Late onset disease is characterised by meningitis and long term neurological sequelae such as cerebral palsy, hearing impairment and cognitive challenges. S. agalactiae does not only infect neonates, it also infects the elderly, immunocompromised individuals and pregnant and non-pregnant women, causing invasive disease. In the world, 10-40 percent of healthy women are rectally or vaginally colonised with GBS and they face the risk of passing it to their babies during the process of childbirth. Molecular studies on cholera and non-cholera causing vibrio pathogens isolated from some water resources and aquatic animals in Eastern Cape Province, South Africa

- Description: The global socioeconomic and disease burdens of cholera; a disease caused by Vibrio cholerae have been ongoing since it was first reported in India in 1563. Unfortunately, emerging non-cholera causing Vibrio pathogens have been contributing to the malady of Vibrio genus globally in the recent years. The history of cholera outbreaks in South Africa have been well documented but information regarding other non-cholera Vibrio pathogens, is deficient in the literature. This study evaluated the statuses of aquatic animals including crustaceans, molluscs, and fish as important reservoirs of six key pathogenic Vibrio spp. in some important water resources and fish markets in the Eastern Cape, South Africa, between December 2016 and November 2017. Physiochemical parameters that are vital for water quality assessment were determined for all the water sampling points using standard methods. The parameters include pH, electrical conductivity (EC), total dissolved solids (TDS), salinity, temperature, total dissolved solids, turbidity and biological oxygen demand (BOD5). Vibrio spp. densities in water and aquatic animal samples were determined using membrane filtration and Most Probable NumberPolymerase chain reaction (MPN-PCR) methods respectively. Presumptive Vibrio spp. were recovered from the samples by direct plating of water samples and aliquots of aquatic animal homogenates on thiosulfate-citrate-bile salts-sucrose (TCBS) agar. Where necessary, samples were enriched using alkaline peptone water (APW) for optimum isolation of the targeted Vibrio spp. Presumptive isolates were identified and delineated into the six targeted pathogenic Vibrio spp. using PCR method. The antibiogram fingerprint of selected Vibrio spp. against a panel 18 panels of antibacterial agents were also carried out. Multiple antibiotic resistance phenotypes (MARPs) per set of isolates from water sampling sites and aquatic animal types were determined. Finally, multiple antibiotic resistance indices (MARI) for isolates recovered from the samples were determined. All experiments were done in replicate and data compared using one-way analysis of variance and one-tail t-test as appropriate. Correlation and regression analysis were also carried out as appropriate. Uncontrolled agricultural, industrial and domestic wastewater influxes, as well as run-off intrusions into the water resources investigated, were observed throughout the sampling season. The physicochemical qualities of the water resources varied with seasons. While pH observed for all the water sampling sites complied with South Africa and international standards, most sampling sites did not comply with set standards for one or more of the remaining physicochemical parameters. The parameters that frequently exhibited significant correlation and regression with Vibrio spp. densities were TDS, TSS, turbidity and temperature. The densities of Vibrio spp. in the samples studied varied with season and the highest density per sample types were observed in the summer. The prevalence of Vibrio spp. in freshwater, brackish water and aquatic animal samples were 79 percent, 78 percent and 86 percent respectively. The respective prevalence of V. cholerae, V. mimicus, V. fluvialis, V.alginolyticus, V. vulnificus and V. parahaemolyticus in freshwater samples (18 percent, 3 percent, ~1 percent, ~1 percent, <1 percent, < percent), brackish water(12 percent, 3 percent, 28 percent,~1 percent, ~1 percent, 9 percent) and aquatic animals (9 percent, 1 percent, 17 percent, 1 percent, 1 percent, 17 percent) were observed. All isolates from all the sample types showed very high antibiotic resistance against ampicillin and polymyxin B. Furthermore, MARPs were observed among isolates from the three sample types. Multiple antibiotic resistance indices (MARI) above 0.2 was observed at one site on Kowie River, three on Kubusi River and one on Buffalo River. The findings of this study suggest that most South Africa water-bodies especially freshwater resources are not well protected. This need to be addressed in a country like South Africa where availability of the freshwater resource for human activities is a critical issue. Base on the prevalence of pathogenic Vibrio spp. especially MARPs and sites that exhibited above 0.2 threshold for MARI, environmental base Vibrio species monitoring programs are recommended.

Moringa oleifera leaf powder as a functional antioxidant additive in pork droewors

- Description: The study investigated the effect of Moringa oleifera leaf powder (MLP) on physico-chemical characteristics, antioxidant activity, antioxidant compound content and lipid oxidation in pork droëwors. Firstly, the physico-chemical properties (proximate composition, salt content, water activity (aw), pH) of commercially produced droëwors from different types of meat (beef, game, ostrich) were determined. In the second experiment beef and pork droëwors with similar fat content were produced. Physico-chemical properties and lipid oxidation (thiobarbituric reactive substances (TBARS)) during processing and 26 days of storage at 25 °C and 50percent relative humidity (RH) were measured. In the third experiment, antioxidant compounds (Total Phenolic Compounds (TPC), α-tocopherol, β-carotene) in MLP were quantified. Thereafter, 4 treatments of pork droëwors were produced, with MLP included at 0, 0.5, 1 and 2 g/ 100 g. Physico-chemical properties and TBARS were measured at intervals during drying (0, 1.5, 5.75, 27.25, 72 h) and after 7 days of storage under ambient conditions. Antioxidant activity (ferric reducing antioxidant power (FRAP)), TBARS, α- and γ-tocopherol, and β-carotene contents were measured. In the fourth experiment, three batches of droëwors were produced (C: no antioxidant, M0.75: 0.75 g/ 100 g MLP, VE: 15 mg/ kg α-tocopherol oil) and stored at 25 °C and 50percent relative humidity for 112 days. Drying kinetics and α-tocopherol contents of pork droëwors after drying were measured and the physico-chemical properties and TBARS were followed during storage. Results showed no differences (P > 0.05) in the physico-chemical characteristics of beef, game meat and ostrich droëwors; containing on average 25.8 ± 1.25 g/100 g moisture, 42.0 ± 0.10 g/100 g protein, 32.0 ± 1.68 g/100 g fat, 6.2 ± 0.13 g/100 g ash and 4.2 ± 0.10 g/100 g salt; with aw and pH of 0.79 ± 0.015 and 5.3 ± 0.05, respectively. During processing and storage, TBARS were higher (P < 0.05) in pork droëwors (maximum 3.83 mg MDA/kg DM) than in beef (maximum 0.99 mg MDA/kg DM). Moringa oleifera leaf powder contained high levels of TPC (7.5 ± 0.2 mg gallic acid eq/g) and substantial levels of α-tocopherol (76.7 ± 1.9 mg/100 g) and β-carotene (23.2 ± 2.8 mg/100 g). The FRAP, α-tocopherol and β-carotene content of pork droëwors increased (P < 0.05) proportionally with increasing levels of MLP inclusion. Lipid oxidation occurred more rapidly (P < 0.05) when MLP was not added and was similar (P > 0.05) for all MLP treatments. There was no significant effect of the inclusion of 0.75 g/100 g MLP on the drying curves and physico-chemical characteristics of the droëwors. The α-tocopherol content was higher (P < 0.05) and TBARS during storage were lower (P > 0.05) with MLP addition. The results of the current study give an overview of the composition of commercial droëwors and showed higher susceptibility to lipid oxidation in pork droëwors. Moringa oleifera leaf powder exhibited antioxidant activity in pork droëwors, inhibited lipid oxidation and increased the content of α-tocopherol in the product. It can be concluded that MLP could be used as a functional antioxidant additive in pork droëwors.

Muscle nanostructure, fat colour related fatty acids and Check-All-That-Apply visual profiling of South African A2 beef longissimus

- Description: The main objective of this study was to determine the effect of muscle nanostructure on tenderness, fat colour related fatty acids and Check-All-That-Apply visual profiling of South African A2 beef longissimus. Fifty two A-class steers (n=52) of five breeds (Bonsmara (n=19), Beef master (n=7), Hereford (n=9) and Simbra (n=17)) typically processed in different South African feedlots were studied. The animals were humanely slaughtered at an abattoir following the commercial standard procedures. Fat colour measurements (CIE b*) were taken at the P8 site on hot carcasses at the abattoir. Carcass mass (warm and cold) was recorded at the slaughter line before chilling (warm) and after chilling (cold) while pH0 and temperature were measured at 45 minutes post-mortem on the Longissimus thoracis et lumborum (LTL) muscle. Thereafter, a 20 g subsection of the LTL muscle on the left side of each carcass was sampled for muscle nanostructure analysis. At 24 hours post-slaughter, ultimate pH (pH24) was measured, further subsections of the LTL muscle (20 g) were harvested for further muscle nanostructure analysis and 2.5 kg of the LTL muscle was also harvested for analysis of tenderness, fatty acids and sensory evaluation. The muscle nanostructure (myofibril diameter (MYD), myofibril spacing (MYS), muscle fibre diameter (MFD), muscle fibre spacing (MFS) and sarcomere length (SL)) was analysed using the scanning electron microscope (SEM) while tenderness was measured using Warner Bratzler Shear Force after 45mins, 24hrs, 3 and 7 days aging period at -20 ºC refrigerator temperature. Some proximate fractions (total extractable intramuscular fat (Fat %), Fat Free Dry matter (FFDM %) and Moisture content) and fatty acid profile were measured using extracted fat and fatty acid indices (desaturase index, atherogenicity index) were calculated. Lastly a Check-All-That-Apply visual profiling method rated by an 80-member consumer panel was done to assess consumer meat preferences and perceptions of Bonsmara, Hereford and Simbra A2 fresh steaks. At 45mins post-slaughter breed affected (P<0.05) MYD and MYS, while at 24hrs breed affected MFD and MFS only. Early post-mortem pH (pH0) had no effect on the muscle nanostructure and tenderness while ultimate pH (pH24) affected MYD and MYS only. Muscle temperature at 45mins affected (P<0.05) the MYD, MFD and MFS, while 24hrs muscle temperature affected (P<0.05) MYD only. Tenderness was not related to the muscle nanostructure early post-mortem and there was no uniformity on the changes in muscle nanostructure and tenderness from early post-mortem to 24hrs against the multifaceted carcass mass, muscle pH and temperature effects. During ageing, breed affected the MYD and MYS at 45mins, while at 24hrs in addition to the myofibril structure breed also had an effect on muscle fibre bundle characteristics (MFD and MFS). The changes in MYD, MYS and MFS became constant on day 3 while variations in the MFD still progressed. There was no uniformity on the SL pattern during ageing and a very weak linear relationship between tenderness, SL, MFD and MFS was observed. Notably there was also no uniformity on how the muscle texture features; including the surface structure, fibre separation, and the mechanism of muscle contraction and relaxation; changed throughout the ageing period across and within the breeds. Moreover, breed had a significant effect on pentadecyclic acid, total saturated fatty acids (SFA) and docosahexanoic acid. Differences (P<0.05) in SFA, monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), n-6, n-3, fatty acid ratios, and desaturase index were observed among the white, creamy-white and creamy fat colour. The distribution of fatty acid classes was SFA>MUFA>PUFA in descending order of concentrations of mean values (44.19:40.06:15.75, respectively), while the n-6: n-3 ratio was 12:1. In sensory evaluation, consumers indicated how they would prefer to pay premium for meat with guaranteed muscle and fat colour, low fat content, high marbling and tenderness. Consumers observed breed differences (P<0.05) in muscle fibre separation and muscle colour. The overall liking of each attribute was more on moderate bright cherry red lean colour, moderate yellow fat, very abundant marbling and slight separation of muscle fibres. Differences on discrete liking of each attribute among consumers had an effect on the overall liking of each steak with Hereford steaks being more preferred by consumers. It was concluded that meat from beef animals within the same feeding regime has a different fatty acid composition owing to different breed synthetic pathways and feed ingredients and that meat tenderness is not affected by the muscle nanostructure early post-mortem. However, during ageing, meat tenderness is directly linked to breed related myofibril structure changes in particular the myofibril diameter, spacing between myofibrils and their interaction; while the muscle texture features, fibre diameter, spacing between muscle fibres and sarcomere length explain the non-uniformity of beef tenderness. Also the organoleptic and health related quality of meat assumed more importance among consumers in sensory evaluation. Thus meat classification systems should include more credence aspects or indicators that support credence quality of meat

- Description: The main objective of this study was to determine the effect of muscle nanostructure on tenderness, fat colour related fatty acids and Check-All-That-Apply visual profiling of South African A2 beef longissimus. Fifty two A-class steers (n=52) of five breeds (Bonsmara (n=19), Beef master (n=7), Hereford (n=9) and Simbra (n=17)) typically processed in different South African feedlots were studied. The animals were humanely slaughtered at an abattoir following the commercial standard procedures. Fat colour measurements (CIE b*) were taken at the P8 site on hot carcasses at the abattoir. Carcass mass (warm and cold) was recorded at the slaughter line before chilling (warm) and after chilling (cold) while pH0 and temperature were measured at 45 minutes post-mortem on the Longissimus thoracis et lumborum (LTL) muscle. Thereafter, a 20 g subsection of the LTL muscle on the left side of each carcass was sampled for muscle nanostructure analysis. At 24 hours post-slaughter, ultimate pH (pH24) was measured, further subsections of the LTL muscle (20 g) were harvested for further muscle nanostructure analysis and 2.5 kg of the LTL muscle was also harvested for analysis of tenderness, fatty acids and sensory evaluation. The muscle nanostructure (myofibril diameter (MYD), myofibril spacing (MYS), muscle fibre diameter (MFD), muscle fibre spacing (MFS) and sarcomere length (SL)) was analysed using the scanning electron microscope (SEM) while tenderness was measured using Warner Bratzler Shear Force after 45mins, 24hrs, 3 and 7 days aging period at -20 ºC refrigerator temperature. Some proximate fractions (total extractable intramuscular fat (Fat %), Fat Free Dry matter (FFDM %) and Moisture content) and fatty acid profile were measured using extracted fat and fatty acid indices (desaturase index, atherogenicity index) were calculated. Lastly a Check-All-That-Apply visual profiling method rated by an 80-member consumer panel was done to assess consumer meat preferences and perceptions of Bonsmara, Hereford and Simbra A2 fresh steaks. At 45mins post-slaughter breed affected (P<0.05) MYD and MYS, while at 24hrs breed affected MFD and MFS only. Early post-mortem pH (pH0) had no effect on the muscle nanostructure and tenderness while ultimate pH (pH24) affected MYD and MYS only. Muscle temperature at 45mins affected (P<0.05) the MYD, MFD and MFS, while 24hrs muscle temperature affected (P<0.05) MYD only. Tenderness was not related to the muscle nanostructure early post-mortem and there was no uniformity on the changes in muscle nanostructure and tenderness from early post-mortem to 24hrs against the multifaceted carcass mass, muscle pH and temperature effects. During ageing, breed affected the MYD and MYS at 45mins, while at 24hrs in addition to the myofibril structure breed also had an effect on muscle fibre bundle characteristics (MFD and MFS). The changes in MYD, MYS and MFS became constant on day 3 while variations in the MFD still progressed. There was no uniformity on the SL pattern during ageing and a very weak linear relationship between tenderness, SL, MFD and MFS was observed. Notably there was also no uniformity on how the muscle texture features; including the surface structure, fibre separation, and the mechanism of muscle contraction and relaxation; changed throughout the ageing period across and within the breeds. Moreover, breed had a significant effect on pentadecyclic acid, total saturated fatty acids (SFA) and docosahexanoic acid.
