- Description: With the emergence of the new field of resource economics, one now has the ability to value and to include natural resources in decision making. This thesis aims to explore the valuation of natural resources by reviewing the economic values, techniques, methods and ecological aspects of restoration. Assegaaibos mountain catchment in the Western Cape Province has been used as a case-study example. A cost-benefit analysis of the restoration of the mountain catchment, in terms of the direct benefits of water and tourism, has been performed. The costs of restoration were observed to see whether they outweighed the benefits (income) derived (water and tourism). The results show that the water and tourism benefits did outweigh the costs of a basic restoration scenario. However, the basic restoration scenario did not fulfil the ecological requirements of the project. The results also illustrated that in the moderate restoration scenario, costs only outweighed the benefits when a three percent discount rate was applied. With the optimistic restoration scenario, costs outweighed benefits only when an eight percent discount rate was used. In the comprehensive restoration scenario, costs were shown to outweigh by far the water and tourism benefits over a thirty-year time frame. However, it should be noted that the deterioration of the environment (accelerated erosion, reinvasion, reduced water quality) was not factored into the costs of failure to rehabilitate.

Climate change is understood to have the most severe impact on the most vulnerable communities and sectors of communities by reducing the availability and accessibility of basic resources such as water, food and energy, impacting severely on human health and wellbeing, and further reducing the capacity of ecosystems to support livelihoods. This will add a layer of significant new complications to the ability of poor communities to maintain or improve standards of living, and further challenge non profit organisations that support such communitiesAnticipatory adaptation to climate change can reduce some of the impacts of climate change, and also address some key development stresses. This thesis aims to provide a framework that is relevant for supporting adaptation to climate change within the context of resource poor communities in a developing country. Its outcome is a strategy that can be used to support climate change adaptation in resource poor communities. A definition for adaptation is proposed as an interpretation of adaptation that is relevant in this context. The intended end goal of adaptation as defined in this research is a better form of development that : supports a harmonious and respectful relationship between humans and their natural resource base; averts further destructive global change or at the very least prevents it from becoming unmanageable; and manages the impacts of past and ongoing destructive change so that there is lowest possible loss of natural, human, or cultural resources. The term transformative adaptation emerged from the research processes to describe adaptation to climate change that concurrently addresses development challenges, is grounded in community development aspirations, integrates programming work across multiple thematic areas and approaches, and addresses the causes of climate change within adaptation interventions. - Description: Generally, peptides are selective and efficacious compounds; as a result, they are the best candidates for drug discovery and development. Most peptide-based drugs have been approved for clinical use. The 2,5-Diketopiperazines (DKPs), also known as cyclic dipeptides, are simple compounds which come from nature. Many DKPs are extracted from animals and plants. For example, cyclo(Proline(Pro)-Leucine(Leu)), cyclo(Proline-Valine(Val)), cyclo(Proline-Phenylalanine(Phe)) etc. In this study cyclo(cis-Proline(Pro)-4F-Proline(Pro)) and cyclo(L- Phenylalanine(Phe)-4I- Valine (Val)) were synthesized from their linear esters using a method modified by Milne et al. (1992). This method yielded good results. However, cyclo(L-Phe-4I-Val) degraded due to physicochemical stability attributes caused by hygroscopicity and photosensitivity. Quantitative analysis and evaluation of physicochemical properties of each dipeptide was achieved by using scanning electron microscopy, thermal analysis (differential scanning calorimetry and thermogravimetric analysis) and X-ray powder diffraction. To determine the purity of the cyclic dipeptide, high performance liquid chromatography and thin layer chromatography was used. The structural elucidation of the cyclic dipeptides was carried by employing infrared spectroscopy, mass spectrometry, nuclear magnetic resonance spectroscopy and, molecular modelling and computational chemistry. The aim of this research was to discover possible biological activity of cyclo(cis-Pro-4-F-Pro) and cyclo(L-Phe-4I-Val) with respect to their antimicrobial, anticancer, anti-diabetic and haematological effects. Cyclo(cis-Pro-4Fluoro-Pro) had no inhibitory effect on the antimicrobial strains tested, and cyclo(L-Phe-4Iodo-Val) could not be tested as the compound decomposed.

Towards an industrial ecology for the Nelson Mandela Bay: Metropolitan area

- Description: The aim of this study is to formulate a model of industrial ecology for wider application in the Nelson Mandel Bay (NMB) Metropole. To achieve this aim, the following objectives will be pursued: • A theoretical investigation of the concept of industrial ecology – in an attempt to establish its meaning, as well as the implications thereof. • A theoretical investigation of the operational implications of industrial ecology, including existing methodologies, such as Material Flow Analysis (MFA) and Life Cycle Assessment (LCA) that can be applied, in order to develop a system of industrial ecology between different companies. • The formulation of a methodology for application to the case study. • Presenting and discussing the research results produced by the application of the industrial ecology related methodology to the case study. • To present a synthesis of the research results of this study. This objective includes a critical evaluation of the research methodology and results, as well as some recommendations for the facilitation of industrial ecology. The methodology of the research involved conducting semi structured interviews, conducting a Material Flow Analysis and Life Cycle Analysis using SIMAPRO software. There interview with Goodyear management took place (see Appendix A), while the management at Sovereign Foods agreed to an interview however they didn’t honour the agreement. During the interview at Goodyear understanding of the operational process was achieved by conducting the interviews. The three areas that were discussed during the interview process were: 1. How the boiler works; 2. Current Air-Emission results; and 3. The process of steam generation. An MFA and an LCA on the production process of comparing the boiler functionality was conducted, so that there is an adequate understanding of the environmental performance of the two different boilers. The results of the MFA are presented as follows, namely: the goal and scope definition and the inventory results. Coal boiler versus fluidised boiler data where modelled. The modelling is done on comparing the flow of chemicals during steam production process, and which chemicals have a significant environmental impact. The LCA results looked at the basis of the symbiotic approach by identifying sustainable choices of design, production and consumption. The findings of this research have been obtained from the investigation of industrial ecology framework planning at Goodyear SA and Sovereign Foods. The findings have revealed that there is a synergy that exists between the two case-study areas. However, this is based on Goodyear changing the technology of the boiler they use (a boiler creates steam for the tyre-building process). The company will be required to use a fluidised boiler, instead of the current coal boiler. The use of the fluidised boiler will ensure that all waste streams at Goodyear will be reused and recycled, thereby creating the framework for industrial ecology. The results that are firstly drawn out of SIMAPRO is the Life Cycle Inventory. In this research, there is a flow of 1437 chemical materials and substances in the production of steam using both coal boiler and fluidized boiler. Due to the number of substance flow elements, the research has presented only the significant substances that influence the environmental performance of steam production using the two different feeder materials, which are coal and chicken matter (see Figure 4.4). In the coal boiler process, sulfur comprises the largest amount of chemical substance. The other top five chemical components that contribute to the steam production process are: nickel, chromium, arsenic and iodosulfuron-methyl-sodium. The scale of figure 4.4 is reduced to show the various number of chemical flows found in the steam production process through SIMAPRO software. The Life Cycle Assessment (LCA) results were presented looking at namely the following characterization, normalization, weighting, single score and damage assessment (see Figure 4.7 – 4.10 p87 – p90). The figures are presented as taken directly from the reports drawn from the SIMAPRO software. Figure 4.7 gives the characterisation results up to 100%, where 0% has a minimal environmental effect; whereas 100% has the most significant environmental effect. Oxidation of coal is compared versus the oxidation of chicken matter, the coal boiler process has higher environmental impacts. Figure 4.8 shows the normalisation results of the coal boiler versus the fluidised boiler for the functional unit steam production. The environmental degradation under normalisation between the two boilers are measured, based on three parameters, namely: human health, ecosystems and resources. In all three parameters that are measured under normalisation, the production of steam using the coal boiler has the highest impacts. The weighting results in Figure 4.9 show a mirror image of the normalisation results. Finally, a gap analysis of the theoretical requirements for industrial ecology is displayed versus what currently exists in the case study area. The gap analysis will assist in the development of recommendations. These will be presented in the next chapter. A gap analysis of findings is discussed leading into the synthesis. The value of this research is that the baseline data of planning for an industrial ecology create a platform that can be used for implementing an industrial ecology between Goodyear SA and Sovereign Foods. This will, in time, have wider applications for the entire NMB metropole.

- Description: Acquired Immunodeficiency Syndrome (AIDS) is a human disease caused by the human immunodeficiency virus type 1 (HIV-1) and it is one of the biggest social, economic and health challenges in the world. The Joint United Nations Programme on HIV/AIDS (UNAIDS) and World Health Organization (WHO) estimated that between 33.4 to 46.0 million people around the world were living with HIV/AIDS in December 2005 and the highest estimates are in the Sub-Saharan Africa (around 25 million). In more developed countries a combined antiretroviral therapy called highly active antiretroviral therapy (HAART) is used, which results in reduced progression to AIDS in most patients. Despite the beneficial effects of HAART, significant side effects are experienced by treated patients. In addition, most infected people live in countries where the treatment is very expensive or, in many cases, not available at all. These people therefore rely on medicinal plants for health care. In this study, aqueous extracts from Sutherlandia frutescens and Lobostemon trigonus were screened for potential anti-HIV activities in a series of in vitro enzymatic assays, including reverse transcriptase, HIV-1 protease and glycohydrolases. Two extracts of Sutherlandia leaves (SFL-1 and SFL-2) were prepared that inhibited HIV reverse transcriptase and a Lobostemon leaf extract (LTL) was shown to also inhibit this enzyme. All extracts were assayed at 1.25mg/ml. Tannin content was determined for all active extracts using a tannic acid assay. SFL-1 and SFL-2 were found to contain about 6 percent and 7 percent tannins, respectively, and LTL contained 31% tannins by weight. Tannins were removed using polyamide columns and three fractions were collected for each. The extracts were also fractionated with Sephadex G-25, Amberlite IR 120 and Dowex 1-X8 as size exclusion, cation exchange and anion exchange, respectively. Extracts were also fractionated by preparative thin layer chromatography where two compounds were separated from S. frutescens extract with high activity against reverse transcriptase while showing insignificant inhibition towards other enzymes tested. SFL-BFW-10 and SFL-WEF-7 inhibited reverse transcriptase by almost 100 percent and the IC50 values calculated for these compounds were 0.34 and 0.23mg/ml, respectively. - Description: Acquired Immunodeficiency Syndrome (AIDS) is a human disease caused by the human immunodeficiency virus type 1 (HIV-1) and it is one of the biggest social, economic and health challenges in the world. The Joint United Nations Programme on HIV/AIDS (UNAIDS) and World Health Organization (WHO) estimated that between 33.4 to 46.0 million people around the world were living with HIV/AIDS in December 2005 and the highest estimates are in the Sub-Saharan Africa (around 25 million). In more developed countries a combined antiretroviral therapy called highly active antiretroviral therapy (HAART) is used, which results in reduced progression to AIDS in most patients. Despite the beneficial effects of HAART, significant side effects are experienced by treated patients. In addition, most infected people live in countries where the treatment is very expensive or, in many cases, not available at all. These people therefore rely on medicinal plants for health care. The solubility of nitrogen in the ZnO films, as well as its activation upon annealing, was also investigated. Hydrogen is known as a major source for passivation of the acceptors in ZnO:N. Therefore, it is crucial to dissociate the complex(es) formed by nitrogen and hydrogen and diffuse out the hydrogen in order to prevent the reformation of such complexes. High temperatures (≥ 600 C) are required for these purposes. In order to effectively remove the hydrogen impurities from the sample, it is important to know the optical fingerprints of hydrogen and its thermal stability. Therefore, the effects of annealing and hydrogen plasma treatment on bulk ZnO (hydrothermally grown) were first studied. The use of bulk material for this purpose was motivated by the well-resolved photoluminescence (PL) lines observed for bulk ZnO, which allow the identification of the different lines related to hydrogen after plasma treatment. Annealing at 850 C was effective for the removal of most of the hydrogen related transitions in the near-band-edge emission. Also, additional transitions at ~3.364 eV and ~3.361 eV were observed after hydrogen plasma treatment, which were ascribed to hydrogen-Zn vacancy complexes. In this work, a comparative study of the annealing ambient and temperature on ZnO films grown on GaAs substrate, using diethyl zinc (DEZn) and tertiary butanol (TBOH), showed that arsenic diffuses in the ZnO films and gives a shallow level in the band gap, which is involved in an acceptor-bound exciton line at 3.35 eV. This shallow level is visible when annealing is performed in oxygen, but not when annealing is performed in nitrogen, and indeed only for annealing temperatures around 550 C. However, annealing in either ambient also causes zinc to diffuse from the ZnO films into the GaAs substrate, rendering the electrical properties deduced from Hall measurements ambiguous. For ZnO:N, NO was used as both oxygen and nitrogen sources. Monitoring the concentration of nitrogen, carbon and hydrogen in the ZnO films, the formation of different complexes from these impurities were deduced. Furthermore, an investigation of the effect of annealing on the concentrations of impurities showed that their out- diffusion was strongly dependent on the crystalline quality of the ZnO films. For porous ZnO films, obtained at low growth temperatures (≤310 C), the out-diffusion of impurities was efficient, whereas for films grown at higher temperatures, which have improved crystalline quality, the out-diffusion was practically nonexistent. The out-diffusion of unwanted impurities may activate the nitrogen dopant in the ZnO films, as was confirmed by the PL measurements on the different samples grown at different temperatures. PL transitions at ~3.24 eV and ~3.17 eV were related to substitutional NO. These transitions were more dominant in the spectra of samples grown at low temperatures. An additional transition at ~3.1 eV was assigned to a donor-acceptor pair transition involving VZn, instead of NO, as previously reported.

- Description: The low affinity IgE receptor, CD23, is expressed by a wide variety of cells and cleaved from its original 45 kDa size to several smaller soluble CD23 proteins. Soluble CD23 function depends on the form of the protein and its interaction with various ligands. CD23 is believed to play an important role in regulating allergic responses and in inflammation, amongst others. β2 integrins are important in a variety of cell-adhesion reactions during immune-inflammatory mechanisms and the binding of their natural ligands generates outside-in cellular signalling, leading to cell activation. Although the binding of CD23 to β2 integrins contributes to this signalling in monocytes, the interaction site for CD23 is unknown. This study focused on the interaction of three soluble CD23 proteins with the β2 integrins CD11b/CD18 and CD11c/CD18. Differentiated HL60, THP1 and U937 monocytic cells were used to demonstrate the binding of three recombinant CD23 constructs (corresponding to 16, 25 and 33 kDa human soluble CD23) to upregulated CD11b/CD18 and CD11c/CD18. This binding was partially blocked by an antibody specific for the CD11b/CD18 αI domain, demonstrating that αI domains are involved in binding to CD23. Recombinant αI domain proteins of CD11b and CD11c were demonstrated to bind CD23 using ELISA and in surface plasmon resonance spectroscopy. The dissociation constants for CD23-CD11b/CD18 and CD23-CD11c/CD18 are comparable to other integrin ligands. This study has shown that CD23 interacts directly with the αI domains of β2 integrins and that the interaction surface likely spans the lectin domain as well as either the stalk and/or C-terminal tail of CD23. This study also looked at the effect that soluble CD23 proteins had on monocyte biology. It appears that iv sCD23 proteins have little effect on the phagocytic or chemotactic ability of monocytes, while an increase in oxidative burst was shown with the 16 kDa and 25 kDa CD23 proteins. Signalling pathways for the production of reactive oxygen species were investigated and it appears that the CD23 proteins signal mainly through the phosphoinositide-3 kinase pathway, although the mitogen activated protein kinase and Src kinase pathways may also play a role. These data suggest that sCD23 proteins induce outside-in signalling of β2 integrins and are able to change the activation state of CD11b/CD11c by stimulating oxidative burst. This needs to be further investigated by determining how the three sCD23 proteins are binding the CD11 proteins and investigating further leukocyte function and inflammatory responses by the cells.

This study attempted to express and purify a recombinant human soluble FceRII to test its effect on a B-lymphoblastoid (RPMI 8866) and a monocytic (U937) cell line. The protein was expressed in Escherichia coli inclusion bodies, before being refolded and purified in a single gel chromatography step. This pure protein was then tested for biological activity by testing its IgE binding func tion. Once proven functional, it was used to test its effect on the cell lines at three concentrations for its apoptotic rescue properties and its cytokine effects. The recombinant protein did not seem to have any significant effect on the apoptotic rescue of either cell line. While the recombinant sFceRII appeared to have a slight effect on the stimulation of IL-1ß and TNFa in the RPMI 8866 cells, there was no apparent effect on the production of NF?B. In U937 cells, the protein did not seem to have any effect on the stimulation of IL-1ß, TNFa or NF?B. However, the cytokine effects of the recombinant protein were tested on isolated PBMCs from a healthy individual and a hyper-IgE syndrome patient. The main objective of the study was to develop a FAST referencing system which can be used for benchmarking by the local apparel industry and, as a basis for encouraging and persuading the industry to adopt this system of fabric quality measurement and assurance and thereby improve their product quality and international competitiveness. Fabric Objective Measurement (FOM) represents a new generation of instrumentally measured parameters which provide a more complete picture of fabric quality, tailorability and clothing performance. The two main FOM systems, FAST and Kawabata, are discussed under FOM in terms of their applications, control charts and their worldwide utilisation. A literature review has been done on the global clothing sector as well as South African clothing industry. The research involved a questionnaire survey of, and interviews with major clothing and retail companies in South Africa with a specific focus on the gap in the South African clothing industry in terms of FOM and other quality related issues. The data and information so captured are presented graphically, statistically analyzed and interpreted, to arrive at the main conclusions and recommendations. Trubok, Newcastle, the only company in South Africa utilizing FOM, was visited in order to obtain hands on experience with the FAST system as operated in a mill. Two different fabrics were tested and the control charts obtained were interpreted. According to the analysis of the questionnaires and interviews, various conclusions could be drawn. When benchmarking a product, quality emerged as the first criterion, 100 percent retailers and manufacturers agreed to this. Most respondents stated that their fabric and garment testing is mostly done in-house while other respondents stated that their fabric and garment testing is mostly done by their respective suppliers. The most commonly used outside laboratories are SGS and ITS. Merchandising and quality complement each other and with proper quality assessment the merchandising workflow becomes smooth, easy and timely delivery of products. All of the respondents (100 percent) supported this fact. Retailers and manufacturers agreed that quality and merchandising are related to each other and hence helping those in achieving product benchmarking (statistically significant at 95 percent confidence level). Retailers and manufacturers conduct fabric and garment tests on a regular/routine basis and mostly use knitted and woven fabrics in garment making. In addition to the above, the worldwide manufacturers and suppliers of the FAST and Kawabata systems were approached to obtain data and information about the number of such systems sold worldwide and their fields of application. This information was considered important in promoting FOM in South Africa. Only one manufacturer is presently using FAST for quality control purposes. Of the manufacturers and retailers covered, most of them were either unfamiliar or totally unaware of FOM and its application. This indicates that there is considerable scope for introducing this highly advanced technology into the textile and clothing manufacturing and retail pipeline in South Africa. Most of the manufacturers and retailers (50 percent) intend to introduce certain new tests in future. The tests that they are planning to introduce in future may include FAST, which is fairly simple, reliable and productive, as well as enhancing the quality of the garment. If used, FOM can improve the quality and competitiveness on the international level which is currently lacking in the South African clothing sector.

A Speciation study of the chloro-hydroxo complexes of Pt(II)

- Description: In this study a method was developed to identify and quantify platinum(II) complexes of the type [PtCl4-n(OH)n]2- and [PtCl4-n(H2O)n]2-n. Separation of the various species was achieved with the aid of a hyphenated reversed phase HPLC-ICP-MS technique coupled with an ion-pairing reagent, HMHDCl2. The adsorption of HMHD2+ onto a C-18 column was investigated by generating a series of breakthrough curves. It was found that the selectivity for high charge density anions originates from its low surface coverage relative to TBA+, which on the other hand could not separate Pt(II) complexes. The peaks in the chromatographic traces were assigned by following the stepwise ligand substitution of [PtCl4]2- in hydroxide medium with UV/Vis spectrophotometry and HPLC-ICP-MS simultaneously. A computer program was written by the author to analyse chromatographic data by deconvoluting the chromatogram into its individual components and calculating the mole fraction of each component. The validity of the consecutive pseudo-first order model was validated by constructing 3D Mauser diagrams with the raw spectrophotometric data (A1 vs A2 vs A3). Additional software was used to simulate the raw spectrophotometric data and processed chromatographic data. The pseudo-first order rate constants obtained in both cases were in agreement with each other. Hence, peaks were assigned to [PtCl4]2-, [PtCl3(OH)]2-, [PtCl2(OH)2]2-, [PtCl3(H2O)]-. The molar extinction coefficient spectra of [PtCl3(OH)]2- and [PtCl2(OH)2]2- were obtained by simulating the spectrophotometric data at wavelengths from 280 to 450 nm. The reaction of [PtCl4]2- with sodium hydroxide was investigated with UV/Vis spectrophotometry at 25 °C. A rate constant consisting of a first and second order term was obtained. The first order term agreed with what has been reported in the literature for aquation of [PtCl4]2- at 25 degrees C. The influence of temperature was established by conducting the experiment at different temperatures. It was found that the reaction proceeds essentially via aquation at elevated temperatures.

Endozoochory in the subtropical thicket: comparing effects of species with different digestive systems on seed fate

- Description: Endozoochory is an important form of seed dispersal and as such plays a fundamental role in plant population and community ecology. This is especially true in the Subtropical Thicket biome where many plant species are adapted for endozoochory. Endozoochorous seed dispersal is carried out by a wide range of thicket fauna, but changes in the faunal assemblage associated with transformed thicket threatens to disrupt the process of endozoochorous seed dispersal and in turn the regeneration dynamics of thicket plants. Outside of conservation areas, indigenous seed dispersers are often replaced by domestic species, such as goats, which are hypothesized to differ in their seed dispersal efficiencies. The dispersal efficiencies of important thicket seed dispersers that occur in transformed and untransformed thicket were therefore investigated. The influence of gut morphologyphysiology on seed gut passage time, seed mortality and germination in animal species representing different digestive models was measured, as well as the role of black-backed jackal in thicket plant seed dispersal. Black-backed jackal were found to disperse the seeds of few thicket plant species and are therefore not considered important thicket seed dispersers under the climatic conditions prevailing during the study. The average gut passage time of seeds was found to differ significantly between birds and mammals, but not between elephants (hindgut fermentor), goats (ruminant) and pigs (omnivore, foregut fermentor) as was expected considering the differences in their digestive morphology. A significantly shorter gut passage of large seeds compared with small seeds was observed in goats, due to the regurgitation of larger seeds. Seed regurgitation in ruminants may prove to be an important factor contributing to their quality as seed dispersers in transformed thicket. The mortality of seeds was significantly higher in goats than in elephants, probably due to the efficient digestion associated with ruminants. The effect of gut passage on seed germination was not consistent and varied between animal species and across different thicket plant species. It was concluded that seed dispersers that occur in transformed and untransformed thicket differ in their seed dispersal efficiency due, in part, to differences in the nature of their digestive tracts. Apoptotic cell death allows a cell to “commit suicide” in genetically- controlled or programmed mechanism(s). The microenvironment of the tumour is important since a nurturing malignant environment is required for tumour maintenance, progression and ultimately the development of metastasis. Due to the correlation of the tumour microenvironment to aggressive tumour progression, emphasis should be placed on the constituents of the tumour’s microenvironment. In recent years, the understanding of intracellular pathways in cancer cells has increased rapidly, contributing to the development of drugs with more specific targets such as growth factors, signalling molecules, cell adhesion proteins, proteases, cell-cycle proteins, modulators of apoptosis and molecules that promote angiogenesis and metastasis. The main aim of this study was thus to identify a few potential or active compounds from a library of synthetically-derived compounds as possible alternative breast cancer treatment candidates.

- Description: Secondary metabolites derived from plants, especially those used by traditional healers, are at the forefront of new drug development in combating diseases such as cancer and diabetes. Garlic is employed in indigenous medicine all over the world for the treatment of a variety of diseases. Dietary garlic has been recognized for its beneficial health effects. In particular, garlic consumption has been correlated with (i) reduction of risk factors for cardiovascular diseases and cancer, (ii) stimulation of immune function, (iii) enhanced detoxification of foreign compounds, (iv) hepatoprotection, (v) antimicrobial effects, (vi) antioxidant effects, and most importantly (vii) its hypoglycemic and anticoagulant properties. Due to these beneficial properties, garlic and its closely related genera which includes Tulbaghia violacea, may be useful as coadjuvant therapy in the treatment of type 2 diabetes and some of its physiological complications. The aim of this study was to determine if T. violacea has antidiabetic and anticoagulant properties. This was performed in vitro using both aqueous and organic extracts of the roots, leaves and bulbs. An organic extract was able to improve glucose-stimulated insulin secretion (GSIS) in INS-1 pancreatic β-cells and glucose uptake in Chang liver cells. The BO extract had no effect on the glucose uptake in 3T3-L1 an adipose cell line and reduced glucose utilisation in C2C12, a skeletal muscle cell line. Some of the properties displayed by T. violacea in this study are consistent with those found in similar studies with garlic extracts. It was observed that the BO extract increased the membrane potential and Glut-2 expression in INS-1 cells cultured at hyperglycemic levels, however, at normoglycemic levels a reduction was observed. The oxygen consumption increased at both glycemic levels due to treatment with the BO extract. Platelets were exposed to the extracts to determine their effects upon platelet aggregation, adhesion and protein secretion. Since the BO extract displayed the highest potential at inhibiting platelet aggregation and adhesion. A rat model was used in ex vivo studies to determine if the extract exhibited the same effect in a physiological model. It was noted that the BO extract exhibited a higher degree of inhibition on platelet aggregation and adhesion than the positive control, aspirin. The BO extract reduced clotting times in the prothrombin time (PT) test, but prolonged the clotting time in the actived partial thromboplastin time (APTT) assay in the ex vivo model; however, it had no affect on these clotting assays in the in vitro model using human blood. The BO extract increased the D-dimer and Fibrinogen-C levels in the in vitro model, but had no effect on the D-dimer concentrations and lowered the Fibrinogen-C levels in the ex vivo model. The active compounds in the extract remain to be elucidated.

An organic extract was able to improve glucose-stimulated insulin secretion (GSIS) in INS-1 pancreatic β-cells and glucose uptake in Chang liver cells. The BO extract had no effect on the glucose uptake in 3T3-L1 an adipose cell line and reduced glucose utilisation in C2C12, a skeletal muscle cell line. Some of the properties displayed by T. violacea in this study are consistent with those found in similar studies with garlic extracts. It was observed that the BO extract increased the membrane potential and Glut-2 expression in INS-1 cells cultured at hyperglycemic levels, however, at normoglycemic levels a reduction was observed. The oxygen consumption increased at both glycemic levels due to treatment with the BO extract. Platelets were exposed to the extracts to determine their effects upon platelet aggregation, adhesion and protein secretion. Since the BO extract displayed the highest potential at inhibiting platelet aggregation and adhesion. A rat model was used in ex vivo studies to determine if the extract exhibited the same effect in a physiological model. It was noted that the BO extract exhibited a higher degree of inhibition on platelet aggregation and adhesion than the positive control, aspirin. The BO extract reduced clotting times in the prothrombin time (PT) test, but prolonged the clotting time in the actived partial thromboplastin time (APTT) assay in the ex vivo model; however, it had no affect on these clotting assays in the in vitro model using human blood. The BO extract increased the D-dimer and Fibrinogen-C levels in the in vitro model, but had no effect on the D-dimer concentrations and lowered the Fibrinogen-C levels in the ex vivo model. The active compounds in the extract remain to be elucidated.

These regions can be described as the parent material, advancing side, retreating side and the weld zone. The tribological properties of the different friction stir weld regions were analysed in air using different loads, frequencies, experimental duration and surrounding environments. When the applied load was increased some of the weld regions showed an increase in the specific wear rate. For example, the weld zone region showed a decrease when compared to the parent material. In this tribology study the effect of the counter material was evaluated. E52100, Si3N4, SS 316 and alumina counter materials were used in the test procedure which was conducted in air on the weld regions. The parent material was found to have the highest average specific wear rates with the four counter materials when compared to the weld zone samples. The regions affected by the friction stir weld process all showed, on average, lower specific wear rates than the parent material. For the various tested samples, the coefficient of friction and material compatibility that was found to be the most stable, was the alumina ball. Owing to this as well as its chemical stability, alumina was determined to be the best material for further tribocorrosion studies. The friction stir welded samples, that were cut from a cross section of a friction stir weld plate, were analyzed in 3.5% NaCl and dilute Harrison’s solution respectively. The effect of wear on the electrochemical properties was studied under open circuit voltage (OCV) and fixed 0.2 V and 0.4 V applied potential conditions respectively. The effect of wear on the corrosion properties was studied using potentiodynamic polarization. From analyzing the friction stir weld regions in both a wear and wear-free environment, the synergistic properties, namely the effect of wear on corrosion and the effect of corrosion on wear, were able to be calculated. The first electrochemical study was performed under OCV conditions. In the presence of applied wear, the open circuit potential of the friction stir weld samples, in both 3.5% NaCl and dilute Harrison’s solution, was found to decrease from the initial stabilized value. Of interest was the rate at which the OCV recovered. It was found to follow a second order repassivation growth model which was explained in terms of an initial film growth stage, followed by a film thickening stage that results in a new OCV. The second synergized electrochemical-wear study was under fixed 0.2 V and 0.4 V applied potential conditions. In both solutions, 3.5% NaCl and dilute Harrison’s solution, a large increase in the current was observed during the applied wear experiment. The samples submerged in 3.5% NaCl were found to have higher average currents during the wear experiment than those in dilute Harrison’s solution. The rapid decrease in the current, after the applied wear ceased, was found to also follow the second order repassivation model. The electrochemical-wear synergism was done by using potentiodynamic polarization tests. The friction stir weld regions as well as the laser samples were analysed in a wear as well as a wear-free environment in order to determine the effect that wear had on the corrosion properties. The results showed that the weld zone region had a lower corrosion rate than the parent material. This implied that the main weld zone was shown to have the least effect of wear on corrosion and would therefore show the lowest likelihood of failure due to corrosion when compared to the parent material. The wear synergistic factor was found to be between 1 and 1.6 for the friction stir weld regions. This implied that the wear rates of the friction stir welded samples were slightly affected by the presence of a corrosive environment and that the wear properties of the laser welded samples were enhanced by the presence of a corrosive environment. Similar studies performed on laser welded samples showed unusually different results due to the small nature of the laser weld region, with inconclusive results when compared to the friction stir welded samples.

To determine protein structure, homology modelling was conducted and modelling software was evaluated. Leukocyte integrin homologs were obtained from the PDB and models were generated using online servers and MODELLER. Template homologs were fewer in number and of lower quality in comparison to monomeric extracellular proteins. Models were docked using ClusPro, HADDOCK2.2 and AutoDock vina. Models were evaluated using PROSA, Verify-3D and PROSESS. Higher quality models were generated when using MODELLER to separately model monomeric subunits in three defined domain regions (extracellular, transmembrane and cytoplasmic). Template selection concerning these proteins is critical as an intricate relationship exists between model quality, template quality, template quantity, template resolution, target-template identity and template sequence coverage. Docking monomeric subunits was challenging when using ClusPro and the best ligand docking procedures were completed using AutoDock vina. PROSESS provided the most accurate evaluation of protein models, in comparison to PROSA and Verify-3D. These results indicate that although homology modelling is a powerful tool there is much room for improvement. Experimentally obtained templates should be expanded upon within the PDB and energy functions should cater for both monomeric and transmembrane heterodimeric proteins. Leukocyte integrins appear to adopt a closed conformation, which may still facilitate LDV ligand association within the α/β interface. The α3β1 integrin may interact with laminin-5 through the ELV sequence within the G-domain of the α laminin subunit

- Description: The textile industry in South Africa is a significant contributor to the chemical load that public wastewater treatment plants have to process, and the discharge of toxic substances, especially to the aquatic environment. In order to address this issue at factory level, the processes at a textile plant, JMV Textiles were chosen for investigation into the possibilities of minimization of discharges of harmful substances into the environment. The study followed the principles of cleaner production, and the processes that were investigated and revised were: The conventional peroxide bleaching process employed a peroxide stabilizer that did not biodegrade easily. The bleaching process was changed, so that the prevailing conditions in the fabric and dyebath facilitated bleaching without the addition of peroxide stabilizer, and also reduced the alkali and energy requirements. Polyester‐viscose fabric was pretreated by bleaching with hydrogen peroxide, which seemed to be unnecessarily rigorous, because polyester and viscose, unlike natural fibres, are relatively clean and have good whiteness in comparison with natural fibres. The proposed alternate pretreatment was a scour with detergent and alkali. The pretreatment for all polyester‐cotton fabrics was also a hydrogen peroxide bleach. Due to the ability of medium and dark shades to mask the natural tint of cotton fibres, a simpler alternate pretreatment, consisting of an alkaline scour with sequestering agent, was trialled. An unacceptable proportion of the dyeings on polyester was rejected for dye‐stains and dye‐marks. A possible solution exploited the properties of using the finishing auxiliary chemical to alleviate dye‐stains and dye‐marks during the dyeing stage, instead of using the chemical after dyeing. The highly toxic carrier that was used to facilitate level dyeing of polyester fibres that had unacceptable variation in their dyeing properties, was replaced by a less toxic carrier. The formulation for dyeing polyester was simplified to eliminate the auxiliary chemicals that were not essential to the dyeing process. Pale reactive dyed shades on cellulose and polyester‐cellulose were washed off only with hot water instead of detergent. The revised procedures consequential to the study, offered significant environmental benefits by reducing the concentration and volume of effluent produced, substituting a highly toxic carrier with a less toxic one and saving energy and water. Implementation of the suggested changes also offered financial benefits. The management of the factory, however, accepted and implemented some changes, but wanted further investigations for others and a phased approach to the other suggested changes.

- Description: This study considers the dynamical interaction of two predatory carnivores (Lions (Panthera leo) and Spotted Hyaenas (Crocuta crocuta)) and three of their common prey (Buffalo (Syncerus caffer), Warthog (Phacochoerus africanus) and Kudu (Tragelaphus strepsiceros)). The dependence on spatial structure of species’ interaction stimulated the author to formulate reaction-diffusion models to explain the dynamics of predator-prey relationships in ecology. These models were used to predict and explain the effect of threshold populations, predator additional food and prey refuge on the general species’ dynamics. Vital parameters that model additional food to predators, prey refuge and population thresholds were given due attention in the analyses. The stability of a predator-prey model for an ecosystem faced with a prey out-flux which is analogous to and modelled as an Allee effect was investigated. The results highlight the bounds for the conversion efficiency of prey biomass to predator biomass (fertility gain) for which stability of the three species ecosystem model can be attained. Global stability analysis results showed that the prey (warthog) population density should exceed the sum of its carrying capacity and threshold value minus its equilibrium value i.e., W >(Kw + $) −W . This result shows that the warthog’s equilibrium population density is bounded above by population thresholds, i.e., W < (Kw+$). Besides showing the occurrence under parameter space of the so-called paradox of enrichment, early indicators of chaos can also be deduced. In addition, numerical results revealed stable oscillatory behaviour and stable spirals of the species as predator fertility rate, mortality rate and prey threshold were varied. The stabilising effect of prey refuge due to variations in predator fertility and proportion of prey in the refuge was studied. Formulation and analysis of a robust mathematical model for two predators having an overlapping dietary niche were also done. The Beddington-DeAngelis functional and numerical responses which are relevant in addressing the Principle of Competitive Exclusion as species interact were incorporated in the model. The stabilizing effect of additional food in relation to the relative diffusivity D, and wave number k, was investigated. Stability, dissipativity, permanence, persistence and periodicity of the model were studied using the routine and limit cycle perturbation methods. The periodic solutions (b 1 and b 3), which influence the dispersal rate (') of the interacting species, have been shown to be controlled by the wave number. For stability, and in order to overcome predator natural mortality, the nutritional value of predator additional food has been shown to be of high quality that can enhance predator fertility gain. The threshold relationships between various ecosystem parameters and the carrying capacity of the game park for the prey species were also deduced to ensure ecosystem persistence. Besides revealing irregular periodic travelling wave behaviour due to predator interference, numerical results also show oscillatory temporal dynamics resulting from additional food supplements combined with high predation rates.

- Description: Accelerated soil erosion holds strong links to excessive land degradation, socioeconomic problems and accelerated climate change, particularly in developing countries such as South Africa. An analysis of the properties of severely eroded soils is undertaken to determine which characteristic properties influence the erodibility of the soils at several gully and badland sites in three catchments of the Ngqushwa rural municipality, which is tagged as one of the area’s most severely affected by soil erosion and degradation in the Eastern Cape. Physical, biological and chemical properties of the soils were analysed, including aggregate stability, soil texture, organic matter- and carbon content, clay mineralogy, sesquioxide content as Fe2O3 and cation composition. The distribution of a number of these properties was also determined, particularly between erosion sites, along hillslope profiles and with depth. The study area comprises three catchments, namely Mgwalana, Bira and Gqora which share common soil parent materials, land use history, exhibit similar topography and advanced degree of erosion. A total of 63 soil samples were strategically collected from identified erosion zones and site controls devoid of active erosion; where stability was evident by means of vegetation cover and soil surface condition. Surface samples were collected above 30cm depth and subsurface samples at depths greater than 30cm. To determine the fate of carbon additional samples were collected from runon and sink zones at the Mgwalana catchment along a transect from top to bottom slope. Laboratory analyses was performed to determine the soil properties, whereby C content was measured by the dry combustion method, Fe2O3- and cation content by Atomic Absorption (AA) Spectrometer, texture by the separation method described by Schlichting, Blume & Stahr (1995), clay mineralogy by X-Ray Diffraction and organic matter content by conversion of total C. In addition to these properties being used to infer aggregate stability, the wet sieving method was also used for this purpose and for validation of the other soil properties. The results revealed that no one soil property has a greater influence on aggregate stability than the others, but rather that the stability of the soil is dependent on the combinations of these properties in the soil and the interactions that occur between them. Based on this, the results indicated greater stability for runon, sink and control samples, mainly in the topsoil, which were characterised by a relatively higher C- and organic matter content, loam texture and higher concentrations of Ca2+. Furthermore, the less stable eroded samples were characterised by a more clay rich texture particularly in the subsoil, relatively lower C- and organic matter contents and a greater sodicity due to higher concentrations of Na+. The least stable soils belonged to the Gqora catchment, which were found to consist of higher silt content in the topsoil and a higher Fe2O3 content in the subsoil. The clay mineralogy was relatively uniform across all catchments, comprising of primarily illite and secondly kaolinite. The sample with the highest sodicity in terms of ESP contained smectite clay in addition to illite and kaolinite which is assumed to contribute to this increased Na+ concentration. These findings aid in the conclusion that the chemical characteristics of the soils, in association with biological and, to a less extent, physical properties of the soil exacerbate the erosion problem initiated by the extrinsic contributors, such as climate and topography. Investigation into the fate of carbon on eroded lands revealed a topographically driven dynamic whereby the total carbon content was found to be greater at the top and middle slope positions as well as in the sink zone. These hillslope sections were found to have a lower slope gradient and slightly more pronounced concave shape to those sections with lower C values. These topographic variables influence the degree of gully erosion taking place at different hillslope sections, which tends to be greater where slope angle is increased and convexity exists, resulting in the removal of soil C at these positions and its deposition in areas of accumulation, namely the runon and sink zones of low angle concave slope sections. The findings of this research may be used to develop restoration and management strategies with the ultimate goal to reduce the soils vulnerability to erosion by enhancing those soil properties conducive to greater aggregate stability as determined in the present study.
