Spatial and temporal analysis of the critical zone in the Western rift valley corridor: towards earth stewardship science in East Africa

- Description: Over the coming decades, Africa is expected to experience disturbing effects due to climate change and increased land cover change due to human influences presenting a significant concern for the future well-being of human and biological systems, the latter being the foundation of ecosystem services supplied to humanity. Therefore, unprecedented transdisciplinary cooperation, coordination, and integration amongst researchers, government, and civil society are necessary to increase the resiliency of these systems. This study aims to provide an outline of the Africa Alive Corridors (AAC) as an essential model for the encouragement of sustainable development through Earth Stewardship science. These aims are accompanied by the quantification and forward modelling for land cover change of the Critical Zone over 10 Great Lake Basins across one of the AAC, the Western Rift Valley Corridor (WRVC), in East Africa between the years 2018 and 2060. This approach provides the foundation for implementing improved regional governance, better encouragement of sustainable development beyond the 2030 United Nations Sustainable Development Goals, and education programs, such as the AAC, that promote socio-ecological resilience through Earth Stewardship Science. The AAC archives a portion of East Africa as the WRVC, a corridor along the western branch of the East African Rift System that highlights twenty heritage nodes, primarily great lakes, mountain ranges, national parks, and biological hotspots. These heritage entities are associated with ca. 12-million-years of evolution and transformation of the East African topography and related African Great Lake (AGL) systems. The thesis defines the study area by delineating AGL basins intersected by the WRVC. Across these basins, land cover change analysis provides a platform for an integrated assessment of the projected health of the corridor region. Existing land cover datasets provide the initial conditions of the study area for 2008 and 2013. Land cover between 2008 and 2013 is cross-tabulated using the Land Cover Module in the Terrset software, followed by the iii delineation of sub-models and driver variable identification. The Multi-Layer Perceptron algorithm provides the transition potentials between tree cover, urban area, cropland, wetland, and open area classes. Change quantification and prediction using Markov Chain analysis are then established for 2018, 2030, and 2060. The model successfully simulated future land cover change and concluded that: (1) proximity to existing human activity, proximity to existing tree cover, and population are the primary drivers of change; (2) the dominant land cover of the ten lake basins for 2018 was cropland at ca. 48%, followed by tree cover at ca. 33%; (3) total anthropogenic change over the coming four decades equates to over ca. 52 000 km2 (5 200 000 ha), and particularly (4) an urban area is expected to increase by >130%. This assessment ultimately provides a platform for regional governance development at the basin scale and Earth Stewardship science in East Africa. These changes require transdisciplinary action from researchers to civil society. These complexes were synthesized and studied since many questions regarding the interaction of sulfur donors and platinum still exists. These relate to thermodynamic and kinetic factors and their influence on anticancer action. In this dissertation the properties of novel platinum(II) complexes of a bidentate ligand having an aromatic nitrogen-donor atom in combination with a thioethereal sulfur atom capable of forming a five membered ring with platinum(II) were studied. The general structure of the (NS) -ligands used were N-alkyl-2-methylthioalkyl imidazole. Alkyl groups used were methyl, ethyl and propyl. Although amine complexes of platinum have been extensively studied there are some new aspects of these that are worthwhile investigating. In this dissertation amines having planar attachments which will be at an angle with the coordination plane viz. benzylamine and amines having cyclic aliphatic groups namely cyclopropyl and cyclohexyl were investigated. Some of the (NS) - and amineplatinum(II) complexes were oxidised to their mononitroplatinum(IV) analogues . The motivation for the synthesis of these complexes was the greater kinetic stability of platinum(IV) and recent research has shown that a specific type of platinum(IV) compound shows suitable properties as an anticancer agent. These complexes were characterised by a variety of spectral means (IR, NMR, mass spectroscopy) as well as elemental analysis, solubility determinations, thermal analysis (TGA), ionization studies and finally their anticancer behaviour towards three different cell lines(Hela, MCF 7, Ht29) and in this process they were compared to the behaviour of cisplatin as a reference. A few have shown promising anticancer behaviour.

Ecological assessment and biogeography of coastal vegetation and flora in southern Mozambique

- Description: This study considered aspects of the vegetation of the coastal zone of southern Mozambique, an area with a rich biodiversity. The vegetation and flora were assessed in their current state including the associated anthropogenic pressures. The environmental conditions that determined the distribution of the vegetation were evaluated. A hierarchical classification, description and ecological interpretation of the vegetation and flora of the coastal zone in southern Mozambique are presented. Analysis was based on 242 sample quadrats that were distributed in a stratified manner throughout the study area. The vegetation could be grouped into six distinct types of vegetation: Dune Forest, Coastal Forest, Coastal Grassland, Coastal Miombo, Coastal Savanna and Coastal Woodland. Species diversity was high for the all the vegetation types, particularly in the woody elements. A total of 673 species in 410 genera was recorded. They represented 104 families with Fabaceae and Rubiaceae being the most common. Of these, 6.6% were endemic or near-endemic to the coastal zone of southern Mozambique with Coastal Forest being the habitat with the greatest endemism. All soils from different vegetation types had poor agricultural potential. Soil properties were the strongest defining environmental feature separating the vegetation types: e.g. pH was high in Dune Forest compared to the others. Results from Maxent modelling suggest that the distribution of endemic species is influenced by a combination of climatic and non-climatic variables. Soil type, temperature annual range and precipitation of the driest month were the most important predictor variables. Overlaying the potential distributions of the seven selected species indicated two areas of abundance of endemic species – these should be given attention for conservation. Endemic species are not well protected in southern Mozambique – their sampled and potential habitats are largely outside protected areas. Hence, additional reserves should be created to improve their protection. Most endemic and near-endemics species were found in the south, from Ponta de Ouro (Matutuine, south of Maputo Province) to Manhica district (north of the Maputo Province) forming part of the Maputaland Centre of Endemism. A second concentration of endemism was found in the Inhambane Province, specifically the Inhassoro and Vilanculos districts. This is proposed to be an Important Plants Area (IPA). Because most endemics and near-endemics are found in the Coastal Forest, their main threat is harvesting for charcoal production, although none of the endemic species are specifically targeted for charcoal production. In Inhambane they are also threatened by the tourism industry, agriculture and settlements. The impact of the habitat destruction on endemic species is expected to cause severe declines in the near future. The tourism industry and harvesting of trees for charcoal production and over-frequent fires are the main drivers of vegetation loss in this region. Shifting agriculture, harvesting for firewood and construction materials, cattle grazing (at a minor scale) are also impacts, but these were only observed in a small area of Maputo Province. Although the study was done in coastal zone of southern Mozambique, effective management of whole coastal zone of the country will be required to maintain a functioning and diverse ecosystem. Priorities for management are to ensure that forests are protected, in particular, Dune Forest. Actions are required to minimise degradation of coastal vegetation. Further research on Coastal Forest restoration should be planned, as field observations in this study confirm that forest in the coastal zone of southern Mozambique has resilience.

Rehabilitation of the Orange River Mouth Salt Marsh : seed, wind and sediment characteristics

- Description: The Orange River is an important source of freshwater and like many other wetlands in semi-arid regions, supports various social (Spurgeon, 1998), economic (Spurgeon, 1998; Bornman et al., 2005) and ecological functions. The saltmarsh at the Orange River Mouth has become degraded over time following numerous anthropogenic impacts. As a result the Transboundary RAMSAR site was placed on the Montreux record emphasising the importance for rehabilitation. The potential of the marsh for natural rehabilitation was assessed through three physical factors which were considered to have the most influence on the saltmarsh i.e. 1) sediment 2) water 3) wind. Three sampling areas were chosen to investigate the sediment characteristics of the ORM saltmarsh and the suitability for seed germination and adult survival. Site A was representative of the general marsh area, Site B was thought to have favourable sediment conditions for saltmarsh growth because of the large numbers of seedlings and Site C was prone to inundation by wind blown sediment. The sites were sampled in 2005 (dry conditions) and in 2006 after high rainfall and river flooding. Electrical conductivity (EC) of the sediment throughout the marsh was hypersaline in many instances above the tolerance range for S. pillansii (> 80 mS.cm-1) The freshwater event in 2006 lowered salinity significantly in two of the three sites. Differences in sediment characteristics were also compared for three habitats i.e. driftlines, open sites and under vegetation. Driftlines (C. coronopifolia = 872 seedlings m-2; S. pillansii = 1296 seedlings m-2) and the microhabitat associated with adult plants (C. coronopifolia = 803 seedlings m-2; S. pillansii = 721 seedlings m-2) created favourable conditions for seedling growth, however open unvegetated (C. coronopifolia = 56 seedlings m-2; S. pillansii = 49 seedlings m-2) areas had significantly lower seedling density. Due to the marsh currently being in a desertified state this study aimed to establish whether the remaining vegetation could produce enough seed to revegetate the marsh. Laboratory studies indicated that seeds of both species germinated best in freshwater (0 psu). The germination of S. pillansii seeds was 40 percent at 0 psu compared to 5 percent at 35 psu. After storage under hypersaline conditions (35 psu) C. coronopifolia showed 100 percent seed germination when returned to freshwater whereas storage at 70 psu decreased the viability of S. pillansii seeds. The plants are producing adequate seed that will allow for regrowth and rehabilitation if sediment and groundwater characteristics are suitable for seed germination, seedling growth and adult survival. However the increase in bare areas at the Orange River mouth as a result of salt marsh dieback has increased the available sediment source. The wind blown sediment has covered large areas of the remaining adult salt marsh vegetation, particularly in the northern corner at Site C, causing further die-back.

Ecology of key cerithioidean gastropods in the mangroves of the iSimangaliso Wetland Park, KwaZulu-Natal South Africa

- Description: Gastropods are one of the most diverse species groups in mangrove habitats, however, many of their specific roles in relation to ecological patterns and processes are currently largely unknown. The overall aim of this research project was to provide basic ecological information for key gastropod species from subtropical mangroves within a protected area. South African mangroves cover relatively small areas and are restricted to estuaries, these habitats therefore present unique opportunities and challenges to the species that occur in them. Three gastropod species, Terebralia palustris, Cerithidea decollata, and Melanoides tuberculata, all occur at their natural southernmost range limit within South Africa and were selected based on their prominence and occurrence in mangrove habitats of the iSimangaliso Wetland Park, a UNESCO World Heritage Site. Trophic linkages and resource partitioning, resource utilization rates, and ecological resilience were investigated respectively using: 1) a stable isotope (δ15N and δ13C) approach; 2) an experimental approach to quantify feeding dynamics (ingestion rate, consumption/digestion efficiency and grazing impact); and 3) a mixed-effects modelling approach to relate population responses to environmental variables. The diet of T. palustris was seasonally variable and a number of sources were incorporated by different sized snails, but their grazing impact on microphytobenthos was not significant. The results also indicated an ontogenetic shift in the dietary niche for T. palustris through robust partitioning of resources between different size classes. The diets of C. decollata and M. tuberculata were dominated by different primary resources as a function of where they occurred in the mangroves. Melanoides tuberculata consumed a wide variety of primary resources, a typical trait of an opportunistic generalist species. The ingestion rate of M. tuberculata was not dependent on the availability of microphytobenthos, and was highest when conditions were oligotrophic. The resilience of C. decollata was related to the tree-climbing behaviour of this species and its occurrence was best explained by sediment conductivity. These responses were considered in conjunction to what has previously been reported on the resilience of the mangrove trees. The results of this research project have provided new basic ecological information for all three gastropod species in this data-deficient subtropical region. This information can potentially be used in comparative studies for these species in other regions or in broader scale ecological studies. Cerithidea decollata has in contrast expanded its distributional range in this region. Melanoides tuberculata is a globally invasive species, and as South African populations are within its native range, ecological information from this region is valuable as it can be used to investigate the potential ecological effects following introduction into new habitats beyond the native range. Biological drivers have a significant impact on mangrove ecosystem functioning, particularly in relation to recycling and the retention of organic carbon generated through primary productivity. Understanding the ecological linkages that maintain ecological functioning and stability is therefore an important step towards conserving and sustainably managing threatened ecosystems such as mangrove forests.

- Description: Gastropods are one of the most diverse species groups in mangrove habitats, however, many of their specific roles in relation to ecological patterns and processes are currently largely unknown. The overall aim of this research project was to provide basic ecological information for key gastropod species from subtropical mangroves within a protected area. South African mangroves cover relatively small areas and are restricted to estuaries, these habitats therefore present unique opportunities and challenges to the species that occur in them. Three gastropod species, Terebralia palustris, Cerithidea decollata, and Melanoides tuberculata, all occur at their natural southernmost range limit within South Africa and were selected based on their prominence and occurrence in mangrove habitats of the iSimangaliso Wetland Park, a UNESCO World Heritage Site. Trophic linkages and resource partitioning, resource utilization rates, and ecological resilience were investigated respectively using: 1) a stable isotope (δ15N and δ13C) approach; 2) an experimental approach to quantify feeding dynamics (ingestion rate, consumption/digestion efficiency and grazing impact); and 3) a mixed-effects modelling approach to relate population responses to environmental variables. The diet of T. palustris was seasonally variable and a number of sources were incorporated by different sized snails, but their grazing impact on microphytobenthos was not significant. The results also indicated an ontogenetic shift in the dietary niche for T. palustris through robust partitioning of resources between different size classes. The diets of C. decollata and M. tuberculata were dominated by different primary resources as a function of where they occurred in the mangroves. Despite the use of the SPE method that facilitated the concentration of BMAA from large quantities of water, no free dissolved BMAA was detected in raw or processed fresh water. The fate of exogenous BMAA was therefore investigated firstly by evaluating the efficacy of standard water treatment processes employed in South Africa and secondly by investigating the possibility of BMAA bioaccumulation and biomagnification in aquatic food chains. Standard water treatment processes proved highly efficient at removing free dissolved BMAA, explaining the absence of BMAA in treated tap water. However, the cause of the BMAA absence in raw potable water remained unknown. Uptake of BMAA by model aquatic organisms was investigated in controlled experiments. BMAA uptake was documented in both Ceratophyllum demersum and Daphnia magna, however, BMAA-protein association and biomagnification were not observed in D. magna. BMAA had an inhibitory effect on the oxidative stress enzyme acitivties of both organisms tested (as well as human S9 extracts), resulting in accumulation of detrimental reactive oxygen species (ROS) in the cells. Exposure of crop plants to BMAA in controlled experiments resulted in BMAA uptake, protein association, and subsequent inhibition of the antioxidative enzyme activities. However, BMAA was detected in neither free nor protein-associated form in natural crop plants irrigated with known BMAA-containing bloom water. Post-mortem liver samples of Clarias gariepinus (Catfish) and Crocodylus niloticus (Crocodile), from a natural fresh water ecosystem that experienced frequent cyanobacterial blooms, contained both free and protein-associated BMAA. Higher BMAA concentrations were found in crocodile liver samples compared to fish liver samples, strongly suggesting biomagnification from one trophic level to the next. The influence of genetic relatedness on sociality and demography of female African elephants

Poaching and/or overhunting are known to have direct effects on the demography of elephant populations. The current understanding of the indirect effects of poaching on the sociality and demography of elephant populations is relatively poor, both at the group and an individual level. In this thesis the importance of genetic relatedness (as influenced by poaching) on sociality and demography of the female elephants was studied, using a combination of genetic, observation of behaviour and two decades of demography data collected from the northern subpopulation of TNP. I investigated and characterized the relatedness categories within elephant groups. Using a conceptual model for group size analysis in aggregation economies, I hypothesized that elephant populations subject to social disruptions due to poaching would exhibit characteristics of the free entry model, whereas more stable, closed populations would better fit the group-controlled model. I present a rare quantitative analysis of genetic relatedness and group size patterns among groups of adult female elephants in two wild populations: one in Tarangire National Park (TNP), Tanzania, and another in Addo Elephant National Park (AENP), South Africa. I demonstrate that the group size in African elephant populations is governed by genetic relatedness, and that poaching/overhunting has a significant influence upon the apparent group formation and size in elephants. I then focused on the effect of relatedness on agonistic interactions between adult females. I hypothesized that individual-based aggressive interactions among adult female African elephants would vary according to degree of kinship, with closely related dyads showing less aggression towards each other in resource-limited environments, thereby leading to indirect fitness benefits for individuals. As predicted, females did not show agonistic interactions to their close kin most often, and the frequency and intensity of aggressive interactions was inversely related to the degree of relatedness of the interactants. The effect of group relatedness and structure on reproductive success of individual female African elephant in TNP was also investigated. Adult female reproductive success was significantly influenced by within- group relatedness and structure. Higher reproductive success (with higher frequency of calf production and survival and more female calves produced) were more evident in the closely related groups than groups with low relatedness, suggesting that females from genetically disrupted groups are less likely to be reproductive than those in closely related groups. The possibility of negative effects of poaching on the subsequent generation of poached adult females and the alternative of a positive demographic response through reduced density was assessed by analysing the demographic patterns of the first generation (F1) females of prime-aged adult female African elephants in TNP using within- group relatedness and size. I also compared vital rate (age of first birth and interbirth interval) responses of first generation (F1) cows from Tarangire (poached) elephants with other females from poached (Northern Luangwa National Park, Zambia) and unpoached (Amboseli National Park, Kenya and Addo Elephant National Park, South Africa) populations. Group relatedness had no significant effect on sex ratio of the F1 cows‟ calves. A Speciation study of the chloro-hydroxo complexes of Pt(II)

- Description: In this study a method was developed to identify and quantify platinum(II) complexes of the type [PtCl4-n(OH)n]2- and [PtCl4-n(H2O)n]2-n. Separation of the various species was achieved with the aid of a hyphenated reversed phase HPLC-ICP-MS technique coupled with an ion-pairing reagent, HMHDCl2. The adsorption of HMHD2+ onto a C-18 column was investigated by generating a series of breakthrough curves. It was found that the selectivity for high charge density anions originates from its low surface coverage relative to TBA+, which on the other hand could not separate Pt(II) complexes. The peaks in the chromatographic traces were assigned by following the stepwise ligand substitution of [PtCl4]2- in hydroxide medium with UV/Vis spectrophotometry and HPLC-ICP-MS simultaneously. A computer program was written by the author to analyse chromatographic data by deconvoluting the chromatogram into its individual components and calculating the mole fraction of each component. The validity of the consecutive pseudo-first order model was validated by constructing 3D Mauser diagrams with the raw spectrophotometric data (A1 vs A2 vs A3). Additional software was used to simulate the raw spectrophotometric data and processed chromatographic data. The pseudo-first order rate constants obtained in both cases were in agreement with each other. Hence, peaks were assigned to [PtCl4]2-, [PtCl3(OH)]2-, [PtCl2(OH)2]2-, [PtCl3(H2O)]-. The molar extinction coefficient spectra of [PtCl3(OH)]2- and [PtCl2(OH)2]2- were obtained by simulating the spectrophotometric data at wavelengths from 280 to 450 nm. The reaction of [PtCl4]2- with sodium hydroxide was investigated with UV/Vis spectrophotometry at 25 °C. A rate constant consisting of a first and second order term was obtained. The first order term agreed with what has been reported in the literature for aquation of [PtCl4]2- at 25 degrees C. The influence of temperature was established by conducting the experiment at different temperatures. It was found that the reaction proceeds essentially via aquation at elevated temperatures.

An intelligent multimodal interface for in-car communication systems

- Description: In-car communication systems (ICCS) are becoming more frequently used by drivers. ICCS are used in order to minimise the driving distraction due to using a mobile phone while driving. Several usability studies of ICCS utilising speech user interfaces (SUIs) have identified usability issues that can affect the workload, performance, satisfaction and user experience of the driver. This is due to current speech technologies which can be a source of errors that may frustrate the driver and negatively affect the user experience. The aim of this research was to design a new multimodal interface that will manage the interaction between an ICCS and the driver. Unlike the current ICCS, it should make more voice input available, so as to support tasks (e.g. sending text messages; browsing the phone book, etc), which still require a cognitive workload from the driver. An adaptive multimodal interface was proposed in order to address current ICCS issues. The multimodal interface used both speech and manual input; however only the speech channel is used as output. This was done in order to minimise the visual distraction that graphical user interfaces or haptics devices can cause with current ICCS. The adaptive interface was designed to minimise the cognitive distraction of the driver. The adaptive interface ensures that whenever the distraction level of the driver is high, any information communication is postponed. After the design and the implementation of the first version of the prototype interface, called MIMI, a usability evaluation was conducted in order to identify any possible usability issues. Although voice dialling was found to be problematic, the results were encouraging in terms of performance, workload and user satisfaction. The suggestions received from the participants to improve the system usability were incorporated in the next implementation of MIMI. - Description: In-car communication systems (ICCS) are becoming more frequently used by drivers. ICCS are used in order to minimise the driving distraction due to using a mobile phone while driving. Several usability studies of ICCS utilising speech user interfaces (SUIs) have identified usability issues that can affect the workload, performance, satisfaction and user experience of the driver. This is due to current speech technologies which can be a source of errors that may frustrate the driver and negatively affect the user experience. The aim of this research was to design a new multimodal interface that will manage the interaction between an ICCS and the driver. Unlike the current ICCS, it should make more voice input available, so as to support tasks (e.g. sending text messages; browsing the phone book, etc), which still require a cognitive workload from the driver. An adaptive multimodal interface was proposed in order to address current ICCS issues. The multimodal interface used both speech and manual input; however only the speech channel is used as output. This was done in order to minimise the visual distraction that graphical user interfaces or haptics devices can cause with current ICCS. The adaptive interface was designed to minimise the cognitive distraction of the driver. The adaptive interface ensures that whenever the distraction level of the driver is high, any information communication is postponed. After the design and the implementation of the first version of the prototype interface, called MIMI, a usability evaluation was conducted in order to identify any possible usability issues. Although voice dialling was found to be problematic, the results were encouraging in terms of performance, workload and user satisfaction. The suggestions received from the participants to improve the system usability were incorporated in the next implementation of MIMI. The adaptive module was then implemented to reduce driver distraction based on the driver‟s current context. The proposed architecture showed encouraging results in terms of usability and safety. This indicate that the market and effectively the prices of the grain commodities in such countries, are controlled by several local and international economic, political and social factors that are rapidly changing. As a result, the prices of some grain commodities are volatile and trading in such commodities are prone to price-related risks. There are different trading strategies for minimising price-related risks and maximising profits. But empirical research suggests that making the right decision for effective grain commodities trading has been a difficult task for stakeholders due to high volatility of grain commodities prices. Studies have shown that this is more challenging among grain commodities farmers because of their lack of skills and the time to sift through and make sense of the datasets on the plethora of factors that influence the grain commodities market. This thesis focused on providing an answer for the main research problem that grain farmers in South Africa do not take full advantage of all the available strategies for trading their grain commodities because of the complexities associated with monitoring the large datasets that influence the grain commodities market. The main objective set by this study is to design a framework that can be followed to collect, integrate and analyse datasets that influence trading decisions of grain farmers in South Africa about grain commodities. This study takes advantage of the developments in Big Data and Data Science to achieve the set objective using the Design Science Research (DSR) methodology. The prediction of future prices of grain commodities for the different trading strategies was identified as an important factor for making better decisions when trading grain commodities and the key factors that influence the prices were identified. This was followed by a critical review of the literature to determine how the concepts of Big Data and Data Science can be leveraged for an effective grain commodities trading decision support. This resulted in a proposed framework for grain commodities trading. The proposed framework suggested an investigation of the factors that influence the prices of grain commodities as the basis for acquiring the relevant datasets. The proposed framework suggested the adoption of the Big Data approach in acquiring, preparing and integrating relevant datasets from several sources. Furthermore, it was suggested that algorithmic models for predicting grain commodities prices can be developed on top of the data layer of the proposed framework to provide real-time decision support. The proposed framework suggests the need for a carefully designed visualisation of the result and the collected data that promotes user experience. Lastly, the proposed framework included a technology consideration component to support the Big Data and Data Science approach of the framework. To demonstrate that the proposed framework addressed the main problem of this research, datasets from several sources on trading white maize in South Africa and the factors that influence market were streamed, integrated and analysed. Backpropagation Neural Network algorithm was used for modelling the prices of white maize for spot and futures trading strategies were predicted. There are other modelling techniques such as the Box-Jenkins statistical time series analysis methodology. But, Neural Networks was identified as more suitable for time series data with complex patterns and relationships. A demonstration system was setup to provide effective decision support by using near real-time data to provide a dynamic predictive analytics for the spot and December futures contract prices of white maize in South Africa. Comparative analysis of predictions made using the model from the proposed framework to actual data indicated a significant degree of accuracy. A further evaluation was carried out by asking experienced traders to make predictions for the spot and December futures contract prices of white maize. The result of the exercise indicated that the predictions from the developed model were much closer to the actual prices. This indicated that the proposed framework is technically capable and generally useful. It also shows that the proposed framework can be used to provide decision support about trading grain commodities to stakeholders with lesser skills, experience and resources. The practical contribution of this thesis is that relevant datasets from several sources can be streamed into an integrated data source in real-time, which can be used as input for a real-time learning algorithmic model for predicting grain commodities prices. This will make it possible for a predictive analytics that responds to market volatility thereby providing an effective decision support for grain commodities trading. Another practical contribution of this thesis is a proposed framework that can be followed for developing a Decision Support System for trading in grain commodities. This thesis made theoretical contributions by building on the information processing theory and the decision making theory. The theoretical contribution of this thesis consists of the identification of Big Data approach, tools and techniques for eradicating uncertainty and equivocality in grain commodities trading decision making process.

- Description: Eland Tragelaphus oryx are the largest and slowest African antelope, falling within the preferred weight range of the prey of lions Panthera leo. Little is known, however, about the antipredatory strategies employed by eland. Rapid population declines in several small reserves following the reintroduction of lions suggest that the antipredatory strategies of eland may be compromised in spatially limited areas. This emphasises the need for research aimed at investigating how eland respond to lions, particularly in the spatial context, given that lions are increasingly being reintroduced into small reserves in South Africa. This study aimed to investigate lion-eland interactions at multiple scales and determine where on the landscape eland were vulnerable to predation. Adult female eland (n = 3 per site) in breeding herds were collared with GPS satellite collars in two study sites; the Nyathi section of Addo Elephant National Park (Nyathi) and Mountain Zebra National Park (MZNP) in the Eastern Cape, South Africa. A reintroduced population of lions, comprising two males and one female, was present in each study site, with individuals fitted with GPS satellite collars. At a broad spatiotemporal scale, eland did not avoid lions and there was considerable overlap between eland and lion home ranges in both study sites. At a finer spatiotemporal scale however, eland and lion space use was asynchronous in time, except in the case of the female lion in Nyathi where there was considerable overlap at the finest temporal scale investigated (i.e. monthly). Herd-associated eland in Nyathi were killed most frequently in areas associated with high long-term use by the female lion, showing that failure to avoid the female lion at a fine scale increased predation risk for eland. Secondly, eland in Nyathi were killed most frequently in habitat types of intermediate structural density (i.e. mixed habitats), which limited the viewshed of eland but presumably offered increased forage. This habitat type was selected for by eland, despite it being associated with a higher level of predation risk. This suggested that eland in Nyathi traded off safety for forage. In MZNP, predation of herd-associated eland was low which reflected the limited spatial overlap between eland and lion at a fine scale in this reserve. In Nyathi, but not in MZNP, eland responded to lions within 1.5 km by increasing their movement rate, with this response particularly strong at night. The findings of this study show that eland responses to lions are scale-dependent and site-specific. Furthermore, eland responses to lions were shown to be lion gender-specific. This study has increased our understanding of the antipredatory behaviours of eland and demonstrated the importance of investigating prey responses to predation risk at multiple scales.

- Description: The investigation has been focussed on the synthesis and characterisation of camphor-derived chiral auxiliaries that incorporate two camphor skeletons and an evaluation of their stereodirecting potential in ester α-benzylation reactions. Two regioisomeric camphorquinone-derived monoketals were synthesised and identified by 1D- and 2D-NMR, and X-ray crystallography. The stereo-directing potential of the alcohols that resulted from reduction of these ketones as chiral auxiliaries in the alkylation of carboxylate ester derivatives has been studied. The diastereoselectivities shown by NMR spectroscopy range from 14- 30 % d.e. for (1R,2 S, 3R) -2 ,2-[ (1R, 2 S, 3R) -bornane-2,3-dioxy] - bornan-3-ol and 68-74 % d.e. for (1R, 2S ,3R) -3 ,3-[ (1R, 2S ,3R) - bornane-2, 3 -dioxy]bornan-2-ol with selectivities that correlate with the size of the alkyl group in the ester moiety. Trapping of the enolates generated from (1R, 2S ,3R)-2, 2 -[(1R,2 S, 3R) -bornane- 2,3-dioxy]bornan-3-yl propanoate afforded both E- and Z-silyl ketene acetal derivatives in the ratio of 64:36 confirming the formation of both possible enolate structures during enolization. Chiral auxiliaries containing a hemiaminal ether blocking group as well as two chiral alcohols containing monothio-ketal blocking groups have also been synthesised. α-Benzylation of their corresponding propanoate esters afforded the alkylated product with disappointingly low diastereos electivities. Asymmetric reduction of α-keto esters attached to (1R, 2 S, 3R) - 2,2- [ (1R,2 S, 3R) -bornane-2, 3 -dioxy]bornan-3-ol and (1R, 2S ,3R) - 3,3- [ (1R,2 S, 3R) -bornane-2, 3 -dioxy]bornan-2-ol with metal hydrides proceeded with selectivities of up to 30 % d.e. Modelling of the keto ester derivatives at DFT levels provided useful insights into possible conformations adopted by the two α-keto esters and hence the preferred face of attack by metal hydride during reduction.

The diastereoselectivities shown by NMR spectroscopy range from 14- 30 % d.e. for (1R,2 S, 3R) -2 ,2-[ (1R, 2 S, 3R) -bornane-2,3-dioxy] - bornan-3-ol and 68-74 % d.e. for (1R, 2S ,3R) -3 ,3-[ (1R, 2S ,3R) - bornane-2, 3 -dioxy]bornan-2-ol with selectivities that correlate with the size of the alkyl group in the ester moiety. Trapping of the enolates generated from (1R, 2S ,3R)-2, 2 -[(1R,2 S, 3R) -bornane- 2,3-dioxy]bornan-3-yl propanoate afforded both E- and Z-silyl ketene acetal derivatives in the ratio of 64:36 confirming the formation of both possible enolate structures during enolization. Chiral auxiliaries containing a hemiaminal ether blocking group as well as two chiral alcohols containing monothio-ketal blocking groups have also been synthesised. α-Benzylation of their corresponding propanoate esters afforded the alkylated product with disappointingly low diastereos electivities. Asymmetric reduction of α-keto esters attached to (1R, 2 S, 3R) - 2,2- [ (1R,2 S, 3R) -bornane-2, 3 -dioxy]bornan-3-ol and (1R, 2S ,3R) - 3,3- [ (1R,2 S, 3R) -bornane-2, 3 -dioxy]bornan-2-ol with metal hydrides proceeded with selectivities of up to 30 % d.e. Modelling of the keto ester derivatives at DFT levels provided useful insights into possible conformations adopted by the two α-keto esters and hence the preferred face of attack by metal hydride during reduction.

The final study investigated the mechanism of the protective effect of selenium. The results of the first study suggested that selenium could extend survival time. In the second study the selenium supplemented mice showed a reduction in the extent of the increase in liver weight and a decrease in the amount of lipid peroxidation induced compared to the mice that received only toxin. The histology of the selenium supplemented mice also showed a decrease in the severity and amount of morphological changes in the liver. The third study indicated that the protection shown by selenium might be mediated by an increase in the glutathione peroxidase (GPX) activity in selenium supplemented mice. This increase in GPX activity would increase the removal of the lipid hydroperoxides and prevent the damage they would cause in the cell. A further result indicated an increase in glutathione S-transferase in only the toxin control mice when compared to the selenium supplemented and control mice. ii In conclusion selenium offers protection against microcystin but further studies need to be done to provide statistically valid results to clarify the level of protection.

The scientific work described in this report was part of a bigger project aimed at developing innovative processes to manufacture aroma, flavour & fragrance chemicals through beneficiation of industrial waste streams and other raw materials, available locally from the Forestry, Paper & Pulp industries. The evaluation of a process for the preparation of precursor aroma, flavour & fragrance compounds, in particular geraniol and nerol, from locally available raw materials and industrial waste streams, was investigated. Preparation of geraniol and nerol from linalool (ex α-pinene stream) was investigated using acids or organometallic complexes as catalysts for the corresponding isomerisation reaction. The investigation was conducted in an effort to find a less costly process utilising milder conditions than via the conventional cleavage of β-pinene to myrcene under extreme pyrolysis conditions (>650°C). The transformation of linalool to geraniol/nerol using mineral acids was found to be dominated by secondary reactions such as dehydration and cyclisation, resulting in poor product selectivities and yields. On the other hand, organometallic complexes, in particular vanadium-based complexes (e.g. (OV(OBu)3) produced satisfactory results in the preliminary assessment (conversion of 79.8 percent and selectivity of 98.3 percent). A set of statistically designed experiments was carried out on the (VO(BuO)3 + [(Bu)4N+]OH¯) catalyst system where three variables were tested, i.e. substrate concentration, temperature, and catalyst loading. The selected model for conversion was significant with the “Probability > F” being < 0.0001. The most important contributing variable to the model for conversion was temperature i.e. 83.9 percent. Temperature was still the most important variable for the selectivity response at 65.0 percent contribution level. The response surface generated for the selectivity response was flat indicating a robust method within the parameter range selected.

- Description: The South African rookeries of the South Western Indian Ocean (SWIO) loggerhead and leatherback regional management units have been continuously monitored for more than 5 decades, representing one of the longest-running sea turtle beach protection and monitoring programmes in the world. Although such conservation has been successful in protecting adults on land when they come to nest, it might not be enough to ensure population recovery if survival is not ensured across all life stages. Sea turtles are particularly vulnerable to direct threats and changing environmental conditions because they are long-lived, slow maturing, and have low survival rates at early life stages, making them conservation-dependent. These characteristics also render them difficult to conserve as a consequence of their wide geographic distributions, spanning multiple jurisdictions and legislative systems with different conservation targets, and a complex life cycle with changes in habitats and diets at different life stages. Sea turtle populations have been severely depleted as a result of historical over-exploitation and current threats including fisheries bycatch, direct harvest, habitat loss, pollution and pathogens, and climate change. In many cases however, population declines have been reversed by a variety of effective conservation measures such as beach protection programs, and marine protected areas, enabling successful population recovery around the world. Despite protection and apparent increase in abundance, indirect pressures can still affect turtle populations. Therefore, it is fundamental to evaluate the effectiveness of conservation measures (using fitness traits such as female and hatchling sizes) and examine underlying trends. The aim of this thesis was thus to evaluate the fitness of loggerhead and leatherback sea turtles nesting in South Africa. Long-term female size trends over time were evaluated and the populations were split between first-time nesters and experienced nesters. As sea turtle reproductive output (clutch size only) is correlated with female size and evidence is showing that in some populations individual female size is decreasing, the effects of maternal body size on reproductive output and hatchling fitness were investigated for both species. Crawling and swimming speeds were used as proxies of hatchling fitness, because these two traits positively influence their survival. An oceanic model was further used in combination with a particle tracking framework to estimate, for the first time, post-hatchling dispersal of loggerhead and leatherback turtles in the SWIO. I investigated whether the trend in female size for loggerhead turtles nesting in South Africa is a result of their foraging strategy to determine if there is a foraging dichotomy (neritic and oceanic). Despite an apparent increasing population, a significant 3.7 and 3.6 cm decrease in the size of both recruit and remingrant female loggerhead turtles, but a stable mean size of leatherbacks for both age groups was observed. I hypothesise that these contradictory effects are attributable to the very distinct foraging ecologies of the two turtle species and speculate that these variations in carapace length may be driven by environmental change. The reproductive strategy of both loggerhead and leatherback turtles nesting in South Africa supports the optimal egg size theory with larger turtles producing larger clutches but not larger eggs. Egg size was thus constant irrespective of maternal length. Hatchlings with longer carapaces and flippers had higher swimming speed, suggesting that larger hatchlings are fitter (potentially higher survival) than smaller ones by spending less time in high predation risk areas (i.e. beach and nearshore). The model revealed that dispersal trajectories of both loggerhead and leatherback sea turtle hatchlings were very similar and simulations indicated that initial active swimming (frenzy) as well as variability in oceanic conditions strongly influenced dispersal of virtual hatchlings. The trends observed in this study emphasize the importance of long-term monitoring, examining not only abundance but also individual size, to understand population dynamics, support recovery planning and prioritize future conservation practices. Furthermore, the findings of this study have important implications for overall population growth if smaller hatchlings of lower fitness are produced as a result of climate change and thus have decreased dispersal abilities and lower likelihood of survival.

The moisture content of the empty shells as well as the capsule contents were analysed at each time-point. The capsule disintegration time was recorded at each time point. Multi-point dissolution testing was performed at each time point to determine the release of the active substance in each case. Based on the achieved results, the best capsule shell supplier was recommended, and other suggestions were made to improve the capsule formulation.

Within the suite of study systems, the first of two case studies focussed on barrier effects of in-stream structures on fish migration. This was undertaken in the Sundays River. In the second case study, predation and competition dynamics of the non-native piscivorous Micropterus salmoides on estuary-dependent fish was investigated in the estuary headwater regions of the Kowie River system. In all four estuaries, young estuary-spawned fish species dominated the ichthyofaunal community followed by marine-spawned species, despite varied freshwater inflow resulting in headwaters varying in salinity from fresh to hypersaline. Fish community structure however, differed largely between estuaries, with both freshwater abstraction and unnatural elevation of freshwater into estuaries, as a result of inter-basin transfers, affecting these communities. In-stream structures were found to effect upstream movement of fish in two ways, dependent on the type of barrier. Partial (size-dependent) and complete (species-dependent) restriction to upstream migration of fish by causeway-type instream structures were observed. Weir-type in-stream structures acted as a complete barrier to most species, regardless of fish size. Predation of estuary- and marine-spawned fish species by large sized M. salmoides was recorded, although these fish did not contribute significantly to their diet during this study. However, the main dietary components found in smaller sized M. salmoides stomachs overlap with those of juvenile estuary- and marinespawned fish species, suggesting feeding competition between the juveniles of indigenous and non-native fish species.

- Description: The utilisation of estuary headwater environments by young estuary- and marine-spawned fish species was investigated together with the effects of riverflow alteration, in-stream barrier effects and non-native ichthyofauna on the nursery function of these habitats.
