- Description: Much research work has assessed the construction process and discovered that the process is ineffective and besieged with problems. Analysis of these problems has shown that a major part of them are related to the state and workings of the supply chains. Prior research justifies that waste and problems in construction supply chains are extensively present and persistent. This anomaly may be ascribed to the nature of the industry. Horizontal integration that is common place in the construction industry tends to fragment the supply chain, resulting in an unstable production environment occasioned by high unpredictability, much rework, low profits and eventual low level of value creation in the process. Therefore, an increased level of integration of interfaces and processes has been canvassed. The purpose of supply chain management is to achieve the expected increased level of integration of the whole supply chain. Supply chain management is a concept that has flourished in the manufacturing industry through Just in Time production and logistics. Supply chain management represents an autonomous managerial tool, though still largely dominated by logistics. Supply chain management has long been advocated as a means of improving the performance of supply chains in construction. This research study reports on an investigation into the impact of supply chain management on value creation in the South African construction industry. The research discovered that collaborative working is already in the industry and contractors consider supply chain management important for project success. Here construction supply chains were approached from the relationship view point. All issues are encouraged to be viewed and resolved in the supply chain from the relationship perspective.

- Description: The control of modern audio studios is complex. Audio mixing desks have grown to the point where they contain thousands of parameters. The control surfaces of these devices do not reflect the routing and signal processing capabilities that the devices are capable of. Software audio mixing desk editors have been developed that allow for the remote control of these devices, but their graphical user interfaces retain the complexities of the audio mixing desk that they represent. In this thesis, we propose a grid approach to audio mixing. The developed grid audio mixing desk editor represents an audio mixing desk as a series of graphical routing matrices. These routing matrices expose the various signal processing points and signal flows that exist within an audio mixing desk. The routing matrices allow for audio signals to be routed within the device, and allow for the device’s parameters to be adjusted by selecting the appropriate signal processing points. With the use of the programming interfaces that are defined as part of the Studio Connections – Total Recall SDK, the audio mixing desk editor was integrated with compatible DAW applications to provide persistence of audio mixing desk parameter states. Many audio studios currently use digital networks to connect audio devices together. Audio and control signals are patched between devices through the use of software patchbays that run on computers. We propose a double grid-based FireWire patchbay aimed to simplify the patching of signals between audio devices on a FireWire network. The FireWire patchbay was implemented in such a way such that it can host software device editors that are Studio Connections compatible. This has allowed software device editors to be associated with the devices that are represented on the FireWire patchbay, thus allowing for studio wide control from a single application. The double grid-based patchbay was implemented such that it can be hosted by compatible DAW applications. Through this, the double grid-based patchbay application is able to provide the DAW application with the state of the parameters of the devices in a studio, as well as the connections between them. The DAW application may save this state data to its native song files. This state data may be passed back to the double grid-based patchbay when the song file is reloaded at a later stage. - Description: The main goals in electroanalytical sensing are towards improved sensitivity and selectivity, or specificity, of an analyte. There are several approaches to achieving these goals with the main approach being modification of an electrode surface with synthetic or natural catalysts (enzymes), polymers and also utilisation of nanostructured materials. At present, there is a strong movement towards hybrid sensing which couple different properties of two or more surface modification approaches. In this thesis, a range of these surface modifications were explored for analysis and detection of two main analytes: the amino acid, tryptophan (Trp); and, the neurotransmitter, dopamine (DA). Specifically, this thesis aimed to utilise these methods to enhance the sensitivity and selectivity for Trp over an interferent, the indoleamine, melatonin (Mel); and, DA over the vitamin, ascorbic acid (AA). For Trp detection, immobilisation of an enzyme, Tryptophanase (Trpase) resulted in poor selectivity for the analyte. However, enhanced sensitivity and selectivity was achieved through pH manipulation of the electrolyte medium at a Nafion®-modified electrode surface for both Trp and Mel. At pH 3.0, the Mel and Trp anodic peak potentials were sufficiently resolved allowing for an LOD of 1.60 and 1.62 nM,respectively, and permitting the accurate analysis of Trp in a dietary supplement containing Mel. Multi-walled carbon nanotubes (MWCNTs) suspended in Nafion® exhibited further increases in the signal responses of these analytes at pH 3.0 and 7.4 with minimal change in the resolution of the anodic peaks. A lower sensitivity was, therefore, observed at the Nafion® and MWCNT modified electrode compared to the Nafion®-modified electrode at pH 3.0 with LODs of 0.59 and 0.80 nM exhibited for Trp and Mel, respectively. Enhanced selectivity for Trp in the presence of Mel can be achieved with MWCNTs in the presence of metallotetrasulphonated phthalocyanines (MTSPcs) particularly at pH 3.0, owing to cation exchange effects. However, the lack of sensitivity towards Trp, and even Mel, at this CoTSPc and MWCNT modified electrode remains a drawback. For DA, detection at the MWCNT and Nafion® surface resulted in improved sensitivity over that of both the bare electrode (613.0 nM) and the Nafion® modified electrode (1045.1 nM) with a calculated LOD of 133.9 nM at this layer. Furthermore, improvements in the selectivity of DA were achieved at the Nafion® and MWCNT modified electrode as exclusion of AA (150 μM) was achieved. At the MWCNT and CoTSPc surface, AA was excluded up to 130 μM with sensitivity for DA extending as low as 14.3 nM, far greater than observed for Trp and Mel. These concentrations are well within physiological concentration ranges and represent the most significant solution yet in terms of AA exclusion and enhanced sensitivity for DA. An examination of the surface layering by impedance spectroscopy and atomic force microscopy indicates that the success of the hybrid sensor utilising CoTSPc and MWCNTs lay in improved dispersion of MWCNTs and improved electron transfer kinetics, facilitated by the net charge of the materials present. This thesis, thus, showed the utility of a judicious selection of synthetic and biological catalysts, polymers and carbon nanomaterials towards a hybrid approach to the electrochemical sensing of Trp, Mel, DA and AA with focus on sensitivity and selectivity of these analytes.

In vitro effects of endogenous and exogenous cannabinoids on insulin resistance and secretion

- Description: Type 2 diabetes mellitus results from a combination of insulin resistance and impaired insulin secretion. The aim of this study is to investigate the effect of endogenous and exogenous cannabinoids on insulin resistant cell lines, viz skeletal muscle (C2C12) and fat (3T3-L1), and to investigate the effects of these cannabinoids on insulin secretion in pancreatic β-cells (INS 1). Insulin resistance was induced in the cells using 20 ng/mL TNF-α (3T3-L1) and 100 nM insulin (C2C12). Insulin resistant cells were exposed to cannabinoids for 48 hours after which glucose uptake, RT-PCR and Western blot analysis was performed. Additionally, adipokine assays were performed on the 3T3-L1 cells. The insulin resistant 3T3-L1 and C2C12 cells had reduced glucose uptake, decreased IRS-1 and Glut-4 expression indicative of an insulin resistant state. The extract and THC significantly enhanced glucose uptake, IRS-1 and Glut-4 in 3T3-L1 and C2C12 cells. The extract and THC thus have the potential to be an insulin sensitizing agent. Interleukin-6 was significantly decreased by THC. INS 1 cells, cultured under normoglycemic conditions, were exposed to cannabinoids for 48 hours after which glucose-stimulated insulin secretion, radioimmunoassay, oxygen consumption, RT-PCR and Western blot analysis was performed. Insulin stimulatory index was not significantly affected after cannabinoid exposure, except by THC. The cannabinoids decreased insulin content, in a concentration dependent manner, but the inhibition mechanism remains elusive. The cannabinoid Treated cells showed insulin gene expression levels similar to the control, while only THC proved effective in significantly stimulating Glut-2 gene expression. Oxygen consumption studies showed levels lower than the control cells. Most of the cannabinoids inhibited insulin secretion under normoglycemia except THC, while the cannabinoids exhibited the potential to improve insulin resistant adipocyte and myocytes response to glucose and gene regulation.

In contrast to the documented literature, this reaction was observed to occur in two consecutive reaction steps. Geometrical and computational analysis of kinetic data revealed that the reaction proceeds by the following reaction model: Os(VIII) + RCH2OH Os(VI) + RCHO Os(VIII) + Os(VI) Os2(VII) k1 k+2 k-2 The conditional rate constants and molar extinction coefficients were calculated using custom written software. A hydride transfer mechanism, coupled with the synchronous removal of the hydroxyl proton of the alcohol, was postulated. The complexation between osmium(VIII) and osmium(VI) was investigated. Mole ratio titrations and mole fraction plots show that at pH 14.3 a 1:1 complexation occurs between osmium(VIII) and osmium(VI). The equilibrium constants and molar extinction coefficients calculated by these methods were found to be consistent with the parameters obtained from the reduction of osmium tetroxide by primary alcohols at pH 14.3. The formation of a mixed oxidation state dimeric osmium complex (denoted Os2(VII)) has been proposed. Analytic pricing of American put options

- Description: American options are the most commonly traded financial derivatives in the market. Pricing these options fairly, so as to avoid arbitrage, is of paramount importance. Closed form solutions for American put options cannot be utilised in practice and so numerical techniques are employed. This thesis looks at the work done by other researchers to find an analytic solution to the American put option pricing problem and suggests a practical method, that uses Monte Carlo simulation, to approximate the American put option price. The theory behind option pricing is first discussed using a discrete model. Once the concepts of arbitrage-free pricing and hedging have been dealt with, this model is extended to a continuous-time setting. Martingale theory is introduced to put the option pricing theory in a more formal framework. The construction of a hedging portfolio is discussed in detail and it is shown how financial derivatives are priced according to a unique riskneutral probability measure. Black-Scholes model is discussed and utilised to find closed form solutions to European style options. American options are discussed in detail and it is shown that under certain conditions, American style options can be solved according to closed form solutions. Various numerical techniques are presented to approximate the true American put option price. Chief among these methods is the Richardson extrapolation on a sequence of Bermudan options method that was developed by Geske and Johnson. This model is extended to a Repeated-Richardson extrapolation technique. Finally, a Monte Carlo simulation is used to approximate Bermudan put options. These values are then extrapolated to approximate the price of an American put option. The use of extrapolation techniques was hampered by the presence of non-uniform convergence of the Bermudan put option sequence. - Description: American options are the most commonly traded financial derivatives in the market. Pricing these options fairly, so as to avoid arbitrage, is of paramount importance. Closed form solutions for American put options cannot be utilised in practice and so numerical techniques are employed. This thesis looks at the work done by other researchers to find an analytic solution to the American put option pricing problem and suggests a practical method, that uses Monte Carlo simulation, to approximate the American put option price. The theory behind option pricing is first discussed using a discrete model. Once the concepts of arbitrage-free pricing and hedging have been dealt with, this model is extended to a continuous-time setting. Martingale theory is introduced to put the option pricing theory in a more formal framework. The construction of a hedging portfolio is discussed in detail and it is shown how financial derivatives are priced according to a unique riskneutral probability measure. Black-Scholes model is discussed and utilised to find closed form solutions to European style options. American options are discussed in detail and it is shown that under certain conditions, American style options can be solved according to closed form solutions. Various numerical techniques are presented to approximate the true American put option price. Chief among these methods is the Richardson extrapolation on a sequence of Bermudan options method that was developed by Geske and Johnson. This model is extended to a Repeated-Richardson extrapolation technique. Finally, a Monte Carlo simulation is used to approximate Bermudan put options. These values are then extrapolated to approximate the price of an American put option. The use of extrapolation techniques was hampered by the presence of non-uniform convergence of the Bermudan put option sequence. When convergence was uniform, the approximations were accurate up to a few cents difference.

In addition, these feeds have been found to be unsuitable for abalone cultured at elevated water temperatures (> 20°C). The aim of the study was to develop size and temperature specific diets for H midoe through optimisation of dietary protein, energy and lipid levels. Abalone were cultured under farm-like conditions in three partially recirculating temperature controlled systems at either 18, 22 or 24°C and fed formulated diets containing graded levels of protein (18,22 and 26 %) and energy (11.6, 13.5 and 16.2 MJ.kg·I ). Abalone were stocked into baskets at 5 % of available of surface area (n=36) and each diet (n=9) was fed to four baskets of abalone at each of the three temperature regimes for ten weeks. Abalone growth was temperature dependent, with growth declining from 4.33 g.month-I for abalone cultured at 18°C to 0.77 g.month-I at 24°C. Dietary protein could be reduced from 26 to 18 % provided dietary energy levels were maintained at 13.5 MJ.kg- l • A dietary energy level of 11.6 MJ.kg-1 was insufficient to meet the energetic requirements of H midae regardless of the protein content of the diet. The effects of water temperature and body size on the protein requirements of H midae were investigated by culturing abalone at temperatures within the optimal range for abalone farming (i.e. 14, 16 and 18°C). Three size classes of abalone (15, 50 and 80 mm) were fed formulated feed containing graded levels of dietary protein (20, 26, 32, 38 and 44 %) under controlled laboratory conditions for 12 weeks, and, in a separate experiment, under commercial farm conditions for 24 weeks. It was not possible to convincingly define the optimal protein levels for abalone of different sizes in this experiment because growth rates fell below average commercial growth rates obtained on farms. Growth was temperature dependent in the laboratory trial, with the rate of weight gain of the 15 mm (ANOV A: p=0.002) and 50 mm abalone (ANOV A: p=0.02) increasing significantly with an increase in temperature from 14 to 18°C. In the farm trial, dietary protein content did not affect the growth rate of the 10-15 or 80 mm abalone (ANOVA: p>0.05), however, the 50 mm abalone displayed significantly higher weight gain on the 32 % (4.72±0.20 g.month-I ) and 38 % (5.01±0.34 g.month-I ) protein diets compared to those fed the 20 % protein diet (3.75±0.13 g.month-I ) (ANOVA: p=O.OI). Although definition of optimal dietary protein levels were not possible, the effects of dietary protein content and water temperature on the growth of H midae were independent signifying that the protein requirements of abalone are temperature independent. In addition, there was no evidence to indicate that abalone of the different sizes tested here had different dietary protein requirements. The size specific dietary lipid and protein requirements of H midae were investigated by feeding two size classes of abalone (30 and 60 mm initial shell length) diets containing graded levels of dietary lipid (4, 7, 10, 13 and 16 %) and protein (34 - 39 %) for 12 weeks. The 30 and 60 mm abalone were stocked at 7 (n=200) and 9 % (n=36) of the available basket surface area respectively and each diet was fed to four baskets of abalone of each size class. The protein requirements of H. midae are influenced by the amount of available dietary energy and thus it is possible that the ability of abalone to utilise lipids as a source of energy differs in the presence of varying levels of dietary protein. High levels of dietary lipid negatively affected the growth, condition factor and soft tissue glycogen content of both size classes of abalone. This negative effect was greater in the 30 mm size class compared to the 60 mm abalone. The corresponding increase in feed consumption and feed conversion ratio in response to increasing levels of dietary protein also provides evidence that abalone are unable to utilise dietary lipids as an energy source and high levels of dietary lipid probably inhibit the uptake of carbohydrates and protein. High dietary lipid levels did however appear to promote gonad maturation. It was possible to reduce dietary protein from 34 to 20 % without negatively affecting growth through the maintenance of dietary energy levels and thus it is recommended that future experiments on the energy content of formulated feeds should focus on the improved use of carbohydrates. Reductions in the protein portion of formulated feeds for H. midae are possible provided the diet contains sufficient levels of energy supplied from carbohydrates. As the ability of abalone to utilise dietary lipid is limited, lipids are unlikely to play a significant role as an energy source in abalone feeds. Further investigations should focus on the utilisation of various carbohydrate sources in abalone feeds.

- Description: Vegetation stress as a form of degradation is a widespread problem in many catchments in the Eastern Cape province. The Keiskamma is one of the catchments where considerable parts of the thicket biome are stressed. This necessitates an assessment of the status of the thicket biome by way of detecting vegetation stress in the area. The underpinnings of vegetation stress are investigated in this study. As a basic method to evaluate the thicket condition, remotely sensed data were acquired. High resolution ASTER imagery for the Keiskamma area at two different dates (2001 and 2005) was used to compute SVI and NDVI as indicators of vegetation stress conditions. A Digital Elevation Model (DEM) was used to derive slope angle and aspect. By way of digitizing from ortho-photo maps, various land-use types were mapped using Arc View GIS. The relationship between land use, terrain, soil erosion and vegetation stress was established. Field based techniques comprising stomatal conductance measurements were used and compared to remotely sensed data. The SVI and NDVI resultant images expressed similarities in areas depicting vegetation stress conditions at both epochs. A strong linear regression between NDVI and stomatal conductance measurements (mmol/m²) serve to confirm that the NDVI is a reliable indicator of vegetation stress condition. Slope angle and aspect were found to have a significant influence on vegetation stress conditions. Similarly variations in soil moisture and soil surface condition have strong implications for vegetation stress. Amongst other land-use types, abandoned lands were found to have the lowest NDVI values implying an association with the worst vegetation stress scenarios. It was concluded that an element of persistent stress conditions exists amongst the thicket vegetation of the Keiskamma catchment. This was mainly due to land use activities in the area.

- Description: Two and a half billion people mostly in sub-Saharan Africa and southern Asia remain without improved sanitation facilities despite the Millennium Development Goal 7's target to halve this number by 2015. While it might be tempting for developing countries such as South Africa to implement the cheapest and most rapidly constructed sanitation services simply to meet the desired target, this could have significant negative implications on human health and the environment over the long-term. As a result, there is a need to ensure that the most appropriate sustainable sanitation technologies are selected during the planning stage. The purpose of this research was therefore to document the development and pilot application of a flexible context-specific decision-support tool for sustainable sanitation technology selection within Buffalo City Municipality (BCM) in the Eastern Cape Province of South Africa. The first step in the development process was to ascertain the current status of sanitation within the municipality, with a specific focus on the main challenges related to the provision, maintenance and performance of these technologies. Thereafter, a participatory approach was employed involving BCM stakeholders to develop a series of sustainability criteria and indicators that took into consideration economic, social, environmental and technical concerns as well as legal requirements. The development process resulted in a list of 38 BCM sustainable sanitation selection criteria that were applied in a pilot study involving rural, urban and peri-urban communities within BCM. Certain criteria related to topographical features and the availability of piped water and land were considered useful for 'coarse screening' while others were applied during 'fine screening'. In order to enhance the context specificity of criteria, each was weighted, through consultation with key BCM stakeholders. This research confirmed that the sanitation situation in BCM was poor and preliminary evidence indicated that sanitation systems were negatively impacting on the quality of water resources within BCM justifying the need for a sustainable sanitation decision support tool. Based on the pilot application of the BCM Sustainability Selection Criteria List, the urine diversion technology was considered the most sustainable technology option in each study site. There were however, certain social criteria that received relatively low sustainability scores and these would need to be addressed prior to the approval of this technology for implementation. The approach adopted in this thesis was considered highly context-specific yet flexible and appropriate for adoption not only by BCM but other municipalities on a range of spatial scales.

The response of the red mangrove rhizophora mucronata lam, to changes in salinity, inundation and light : predictions for future climate change

- Description: Mangrove forests are subjected to many environmental factors which influence species distribution, zonation patterns as well as succession. Important driving factors in these forests are salinity, water level fluctuations and available light. This study investigated the response of red mangrove (Rhizophora mucronata Lam.) seedlings to these factors in controlled laboratory experiments. Increase in salinity and prolonged inundation within estuaries are predicted impacts resulting from sea level rise due to climate change. The study investigated the effect of five salinity treatments (0, 8, 18, 35 and 45 ppt) with a semi-diurnal tidal cycle on seedling growth. In a separate experiment the effect of different inundation treatments: no inundation, 3, 6, 9 hour tidal cycles and continuous inundation (24 h) were investigated. Both morphological and physiological responses of R. mucronata seedlings were measured. There was a decrease in growth (plant height, biomass and leaf production) with increasing salinity. Seedlings in the seawater, hypersaline and no inundation treatments showed symptoms of stress, having increased leaf necrosis ("burn marks"). The highest growth occurred in the low salinity (8 ppt) treatment, but the highest photosynthetic performance and stomatal conductance occurred in the freshwater treatment (0 ppt). The typical response of stem elongation with increasing inundation was observed in the 24 hr inundation treatment. In the light and salinity combination study there were ten different treatments of five different light treatments (unshaded, 20 percent, 50 percent, 80 percent and 90 percent shade) combined with two salinity concentrations (18 and 35 ppt). In this study the seedling growth: plant height, biomass, leaf surface area and leaf production were higher in the moderate salinity (18 ppt) treatments compared to the seawater (35 ppt) treatments. Biomass in the 35 ppt experiment decreased with increasing shade as well as in the unshaded treatments. Photosynthetic performance and stomatal conductance were lower for the unshaded treatment in both 18 and 35 ppt salinity compared to all other treatments with the same salinity. This suggests that R. mucronata more shade than sun tolerant, but overall it can be concluded that the species has a broad tolerance range. The results may be relevant in mangrove rehabilitation and predicting responses to climate change. This is important as mangrove ecosystems may adapt to changing sea levels and in order to restore areas it will be necessary to choose the mangrove species which will grow best. The results may also help to increase the protection of existing mangrove habitats.

The study investigated the effect of five salinity treatments (0, 8, 18, 35 and 45 ppt) with a semi-diurnal tidal cycle on seedling growth. In a separate experiment the effect of different inundation treatments: no inundation, 3, 6, 9 hour tidal cycles and continuous inundation (24 h) were investigated. Both morphological and physiological responses of R. mucronata seedlings were measured. There was a decrease in growth (plant height, biomass and leaf production) with increasing salinity. Seedlings in the seawater, hypersaline and no inundation treatments showed symptoms of stress, having increased leaf necrosis ("burn marks"). The highest growth occurred in the low salinity (8 ppt) treatment, but the highest photosynthetic performance and stomatal conductance occurred in the freshwater treatment (0 ppt). The typical response of stem elongation with increasing inundation was observed in the 24 hr inundation treatment. In the light and salinity combination study there were ten different treatments of five different light treatments (unshaded, 20 percent, 50 percent, 80 percent and 90 percent shade) combined with two salinity concentrations (18 and 35 ppt). In this study the seedling growth: plant height, biomass, leaf surface area and leaf production were higher in the moderate salinity (18 ppt) treatments compared to the seawater (35 ppt) treatments. Biomass in the 35 ppt experiment decreased with increasing shade as well as in the unshaded treatments. Photosynthetic performance and stomatal conductance were lower for the unshaded treatment in both 18 and 35 ppt salinity compared to all other treatments with the same salinity. This suggests that R. mucronata more shade than sun tolerant, but overall it can be concluded that the species has a broad tolerance range. The results may be relevant in mangrove rehabilitation and predicting responses to climate change. This is important as mangrove ecosystems may adapt to changing sea levels and in order to restore areas it will be necessary to choose the mangrove species which will grow best. The results may also help to increase the protection of existing mangrove habitats.

Nutrient concentrations were sampled at different positions, in the groundwater, at the water’s edge, in the reed bed and in the main estuary channel at five sites in the East Kleinemonde Estuary. The NH4+ and SRP concentrations showed a definite trend as concentrations decreased from the groundwater or water’s edge into the main estuary channel. Groundwater introduced nutrients to the estuary which were then taken up by the fringing reeds (Phragmites australis (Cavinelles) Trinius ex Steudel). The water column nutrients were either below detectable limits or in very low concentrations. Low TOxN concentrations were consistently found at the different sites and probably resulted from P. australis assimilating N-TOxN for growth more efficiently than N- NH4+. The roots, rhizomes, stems and leaves of the reeds were measured for δ15N as an indicator of nutrient enrichment as nitrogen stable isotope analysis of plant tissue is an effective method for assessing and monitoring septic tank and other anthropogenic inputs. All plant parts for the three sampling sessions at Site 1 (mouth region) in the East Kleinemonde Estuary had significantly higher δ15N signatures (~20 percent) than the leaves at Site 5 (upper reaches). These differences were related to the surrounding land-use of the East Kleinemonde catchment, where the lower part of the estuary has moderate to low residential development and the upper reaches are utilized mainly for livestock farming. The high δ15N signatures at Site 1 were attributed to septic tank wastewater and stormwater run-off entering the estuary. Nutrient enrichment also influenced the morphology of the reeds. Reed biomass, height and stem density was significantly higher at Site 1 compared to Site 5. Measurements of the water column may provide an inaccurate assessment of water quality whereas macrophytes are outstanding potential indicators of nutrient enrichment as they are widely distributed, abundant and long-lived. The findings from this study indicate that δ15N concentrations in reeds can be used to indicate nutrient loading.

Thus there is a deficit of calibrator sources in the southern hemisphere. Further, some of the most interesting astronomical objects eg. the galactic centre and the nearest galaxies (the small and large Magellanic Clouds) lie in the southern hemisphere and these require high resolution studies. With a major expansion of radio astronomy observing capability on its way in the southern hemisphere (with the two SKA (Square Kilometre Array) precursors, meerKAT (Karoo Array Telescope) and ASKAP (Australian SKA Pathfinder), leading to the SKA itself) it is clear that interferometry and VLBI in the southern hemisphere need a dense network of calibration sources at different resolutions and a range of frequencies. This work seeks to help redress this problem by presenting an analysis of 31 southern sources to help fill the gaps in the southern hemisphere calibrator distribution. We have developed a multi-parameter method of classifying these sources as calibrators. It is believed to have been introduced into South Africa, as an ornamental plant, in 1908 to the Cape Province and Natal. Since its introduction, water hyacinth has spread throughout South Africa to the detriment of all aquatic systems that it has been introduced to directly or indirectly. The weed was first positively identified on the Nseleni and Mposa rivers on the Nseleni Nature Reserve which is a protected area near Richards Bay in KwaZulu- Natal in 1982 and formed a 100% cover of the river by 1983. An integrated management plan was implemented in 1995 and resulted in a reduction of the weed from a 100% cover to less than 20% cover in 5 years. The keys to success of the water hyacinth integrated management plan, presented here, were finding the source of the weed, mapping the extent of the water hyacinth infestation, identifying sources of nutrient pollution, appointing a champion to drive the programme, dividing the river into management units, consultation with interested and affected parties, judicious use of herbicides and biological control and a commitment to follow-up. This study further showed that water hyacinth on the Nseleni and Mposa river systems had a negative impact on the biodiversity of the protected area and the control of water hyacinth resulted in the recovery of the benthic invertebrate, amphibian, reptile, fish and avian fauna. The implementation of this integrated management plan was very cost-effective and serves as a model approach to the control of water hyacinth in both South Africa and the rest of the world.

Due to inadequate or lack of proper information, most patients especially those with type 2 diabetes are diagnosed through complications. Untreated or poorly managed diabetes is now the leading cause of eye disease and kidney failure in the world. Diabetes is the largest cause of kidney failure in the developed world, and is the fourth leading cause of global death by disease in the world (IDF, 2007). At the Kenyatta National Hospital in Nairobi, Kenya, it is the leading cause of all non-accident related amputations. It is with such statistics in mind and the grim reality of poor and inadequate health services that this research is based. The wide use of selected medicinal plants for the treatment and management of diabetes warrants the further study of these plants for potential use and commercialization. The data obtained can also be invaluable for use and reference when using these plants for medicinal purposes. The medicinal plant studied in the research is widely used in Kenya by many communities and was chosen based on ethno-pharmacological references using traditional medicinal practitioners as well as patient’s recommendations. Different in vitro and in vivo assays were studied to try and elucidate the mechanisms of action as well as the organs targeted during treatment using this plant.

- Description: In a context of declining capture fisheries and public pressure for greater access to marine resources, marine aquaculture is receiving increasing interest from the South African government as a means to increase the diversity of economic activities in coastal regions, thereby providing employment and reducing poverty. The east coast rock lobster Panulirus homarus rubellus is currently harvested by subsistence fisherman along the former Transkei coastline of south-east South Africa and presents a possible opportunity for ongrowing wild juvenile lobsters in culture facilities. Lack of compliance coupled with poor enforcement of the minimum size limit (65 mm carapace length) has resulted in the ongoing harvest of undersize size lobsters by subsistence fishers. Generally, fishers either consume these undersize lobsters or sell them to tourists for low prices. In line with international trends in rock lobster aquaculture, interest has subsequently arisen in the possibilities of ongrowing these undersize lobsters as a means of adding value to the P. h rubellus resource for subsistence fishers. The aim of this physiological study was to assess the biological feasibility of harvesting, transporting and culturing wild caught juvenile lobsters, thereby provide empirical data to inform the development of suitable transport and culture protocols. The experimental objectives were to asses the effect of temperature on growth and survival of P. h. rubellus, as well as the effects of a suite of extrinsic and intrinsic factors on ammonia excretion and oxygen consumption. Juvenile lobsters were collected by hand from near-shore reefs (2-15 m depth) off Mdumbi in the former Transkei, Eastern Cape Province and transported by road (7 hours) to the Port Alfred Marine Research Laboratory where they were held in a recirculating culture system. The effect of temperature over a range of 9.7 °C (18.9±0.7 to 28.6±1.5 °C) on the growth and survival of juvenile P. h. rubellus fed a diet of fresh mussel flesh was investigated. Specific growth rate (SGR) was significantly different between temperatures (p = 0.01), with the highest values recorded for the 24 °C and 28 °C treatments. There was no significant difference in moult increment (MI) between temperatures, however, intermoult period (IMP) differed significantly between temperatures (p = 0.0015) with mean IMP lowest at 24 °C, although not significantly different from the means of the 26 °C and 28 °C treatments. IMP was highest at 19 °C and 21 °C. Apparent feed intake was significantly different between treatments (p < 0.0001) and exhibited a strong positive correlation with increasing temperature. Food conversion ratio (FCR) differed significantly between temperatures (p = 0.02) with 24 °C exhibiting the most efficient FCR. The results for growth rate and food conversion efficiency suggested that 24 °C is optimal for the growout of juvenile P. h. rubellus. In the second study, the effect of body weight, emersion, daily rhythm, feeding and ambient ammonia on the total ammonia nitrogen (TAN) excretion rate was investigated. Body weight (n = 16, range of 16.8 – 322 g) was positively correlated to daytime TAN excretion rate (mg h⁻¹). Re-immersion after one hour emersion in a moist environment was characterized by a significant increase in TAN excretion rate for the first hour compared to pre-immersion levels. The amount of TAN excreted during this period was as expected if basal TAN excretion rates were maintained during emersion. Exposure to an ambient TAN concentration of 4.45±0.78 mg l⁻¹ had a significant effect on the TAN excretion rate, with pronounced ammonia uptake occurring for all animals at this concentration. The third study investigated the effects of body weight, diurnal rhythm, feeding and emersion on lobster oxygen consumption. Body weight was positively correlated to both standard and active oxygen consumption (mg O2 h⁻¹) while body weight was negatively correlated to mass-specific standard oxygen uptake (mg O2 g⁻¹ h⁻¹). Diurnal rhythm exhibited a strong effect on the lobsters oxygen consumption, with average night time values 67% greater than those recorded during the day. This was related to activity driven by intrinsic nocturnal foraging behaviour. Feeding resulted in a classic specific dynamic action (SDA) response, with postprandial oxygen consumption increasing to a peak before decreasing gradually to preprandial levels. Emersion resulted in a significant increase in oxygen consumption, with lobsters rapidly recovering to pre-emersion levels after four hours. Results from these studies suggest that the capture, transport and culture of juvenile P. h. rubellus is biologically feasible. Empirical data generated were used to provide recommendations regarding species optimised transport and culture protocols. A purge time of 48 hours before transport is suggested to ensure that ammonia excretion and oxygen consumption are at basal levels. Furthermore, emersed transport for a period of one hour is characterised by rapid recovery upon re-immersion. In order to prevent the accumulation of stressors, it is suggested that consecutive periods of emersion are interjected with recovery periods (five hours) in water to allow the removal of accumulated ammonia and repayment of the oxygen debt incurred. The recorded ammonia rates indicate that a biological filter size of 4.8 m³ is recommended for 1000 kg of fed lobsters in a culture situation, although this can be reduced considerably if lobsters are being held without feeding (0.72 m³). A flow rate of 112 l kg⁻¹ h⁻¹ is required to meet the metabolic requirements of lobsters. Bottlenecks to the viable commercial culture of P. h. rubellus, and the ability of this practice to provide the socio-economic benefits that were envisioned, are discussed.

- Description: In a context of declining capture fisheries and public pressure for greater access to marine resources, marine aquaculture is receiving increasing interest from the South African government as a means to increase the diversity of economic activities in coastal regions, thereby providing employment and reducing poverty. The east coast rock lobster Panulirus homarus rubellus is currently harvested by subsistence fisherman along the former Transkei coastline of south-east South Africa and presents a possible opportunity for ongrowing wild juvenile lobsters in culture facilities. Lack of compliance coupled with poor enforcement of the minimum size limit (65 mm carapace length) has resulted in the ongoing harvest of undersize size lobsters by subsistence fishers. Generally, fishers either consume these undersize lobsters or sell them to tourists for low prices. In line with international trends in rock lobster aquaculture, interest has subsequently arisen in the possibilities of ongrowing these undersize lobsters as a means of adding value to the P. h rubellus resource for subsistence fishers. The aim of this physiological study was to assess the biological feasibility of harvesting, transporting and culturing wild caught juvenile lobsters, thereby provide empirical data to inform the development of suitable transport and culture protocols. The experimental objectives were to asses the effect of temperature on growth and survival of P. h. rubellus, as well as the effects of a suite of extrinsic and intrinsic factors on ammonia excretion and oxygen consumption. Juvenile lobsters were collected by hand from near-shore reefs (2-15 m depth) off Mdumbi in the former Transkei, Eastern Cape Province and transported by road (7 hours) to the Port Alfred Marine Research Laboratory where they were held in a recirculating culture system. The effect of temperature over a range of 9.7 °C (18.9±0.7 to 28.6±1.5 °C) on the growth and survival of juvenile P. h. rubellus fed a diet of fresh mussel flesh was investigated. Specific growth rate (SGR) was significantly different between temperatures (p = 0.01), with the highest values recorded for the 24 °C and 28 °C treatments. There was no significant difference in moult increment (MI) between temperatures, however, intermoult period (IMP) differed significantly between temperatures (p = 0.0015) with mean IMP lowest at 24 °C, although not significantly different from the means of the 26 °C and 28 °C treatments. IMP was highest at 19 °C and 21 °C. Apparent feed intake was significantly different between treatments (p < 0.0001) and exhibited a strong positive correlation with increasing temperature. Food conversion ratio (FCR) differed significantly between temperatures (p = 0.02) with 24 °C exhibiting the most efficient FCR. The results for growth rate and food conversion efficiency suggested that 24 °C is optimal for the growout of juvenile P. h. rubellus. In the second study, the effect of body weight, emersion, daily rhythm, feeding and ambient ammonia on the total ammonia nitrogen (TAN) excretion rate was investigated. Body weight (n = 16, range of 16.8 – 322 g) was positively correlated to daytime TAN excretion rate (mg h⁻¹). Re-immersion after one hour emersion in a moist environment was characterized by a significant increase in TAN excretion rate for the first hour compared to pre-immersion levels. The black rhinoceros population in the Great Fish River Reserve (GFRR) has increased steadily to a current estimate of one hundred animals since the re-introduction of four animals in 1986. In an effort to contribute to the effective conservation and management of this species, dietary composition was studied in the medium Portulcaria thicket vegetation of the GFRR. This study used a molecular approach to determine the diet of the black rhinoceros of the GFRR by sequencing the ribulose bisphosphate carboxylase large subunit (rbcL) gene in plants and dung. Twenty-three plant species were collected from the reserve, and 802 bp of the rbcL gene were sequenced. These plant sequences were used as a reference database for the identification of plant sequences generated from black rhinoceros dung. Initial studies investigated the amplification, cloning and sequencing of DNA extracted from the dung samples which indicated the viability of the molecular approach. Thereafter, dung generated rbcL DNA was analyzed by GS FLX sequencing. Of the plant sequences identified by comparison to the GenBank database, Carissa bispinosa was the most prevalent. The study further characterized the antioxidant activities and phenolic content of plants eaten by the black rhinoceros using four different assays. Phyllanthus verrucosus, Putterlickia pyracantha, Maytenus capitata, Euclea undulata and Ozoroa mucrunata consistently had high antioxidant activities when assayed against 2,2-azinobis (3-ethyl benzothiazolium-6-sulfonic acid) (ABTSʹ⁺), 2,2-diphenyl-1-picrylhydrazyl (DPPHʹ), and ferric reducing antioxidant potentials (FRAP) and phenolic content when evaluated using the Folin-Ciocalteu assay. The majority of plants investigated showed low antioxidant potentials and low phenolic content. The extent to which antioxidants influenced the browse selection by the black rhinoceros remains inconclusive.

The coordination chemistry of all the above compounds to rhodium(I) is also discussed in the prepared complexes [Rh(COD)(NN)]ClO4, where NN = bisferrocenylimines, and [Rh(COD)(NN)]BF4, where NN = bisferrocenylamines and arylbisamines. X-ray crystal structures of the complexes [Rh(COD)(NN)]ClO4 ([3.2] and [3.3]) have been obtained. Complexes of the type [Rh(COD)(NN)]BF4 were characterized with IR and UV-vis spectroscopy, cyclic voltammetry and conductometry. The catalytic activity of the complexes was also investigated: [Rh(COD)(NN)]ClO4 for the polymerization of phenylacetylene and [Rh(COD)(NN)]BF4 for the hydroformylation of styrene.

There is no data available for purified ostrich cathepsin D, and the aim of this study was, therefore, to isolate, purify and characterise cathepsin D from ostrich skeletal muscle and subsequently investigate the possible role that it may have in the tenderisation process of meat. Cathepsin D was successfully isolated and purified from ostrich skeletal muscle using pepstatin A-agarose chromatography. The purified enzyme was composed of two subunits (14 and 29kDa). The amino acid composition as well as the N-terminal amino acid sequence of both subunits were determined. Kinetic parameters (Km and Vm), thermodynamic parameters (Ea, ∆H, ∆S and ∆G) and functional characteristics (effect of pH, temperature and various inhibitors on cathepsin D activity) were determined and are reported in this study. Ostrich muscle cathepsin D showed a pH optimum of 4 and a temperature optimum of 45°C. The activity of cathepsin D was strongly inhibited by pepstatin A and DTT. Purified ostrich cathepsin D displayed kinetic and functional properties similar to previously reported values from various species. Hence, the focus of the problem is on the prediction of the temporal variations in the horizontal geomagnetic field (@Bx/@t and @By/@t). Artificial neural networks are used to predict @Bx/@t and @By/@t measured at Hermanus, South Africa (34.27◦ S, 19.12◦ E) with a 30 minute prediction lead time. As input parameters to the neural networks, insitu solar wind measurements made by the Advanced Composition Explorer (ACE) satellite are used. The results presented here compare well with similar models developed at high-latitude locations (e.g. Sweden, Finland, Canada) where extensive GIC research has been undertaken. It is concluded that it would indeed be feasible to use a neural network model to predict GIC occurrence in the Southern African power grid, provided that GIC measurements, powerline configuration and network parameters are made available.

They are a useful concept in modeling ideas of uncertainty especially those that arise in Economics, Social Sciences, Statistics and other subjects. In this thesis we define and study some basic properties of keychains with reference to Partially Ordered Sets, Lattices, Chains and Finite Fuzzy Sets. We then examine the role of keychains and their lattice diagrams in representing uncertainties that arise in such problems as in preferential voting patterns, outcomes of competitions and in Economics - Preference Relations.
