Novel cationic-chalcone phthalocyanines for photodynamic therapy eradication of S. aureus and E. coli bacterial biofilms and MCF-7 breast cancer

- Description: New tetrasubstituted zinc (II) and indium (III) phthalocyanines bearing dimethylamino chalcone group (complexes 3 and 4) as well as their quaternized analogs (3a and 4a) have been assessed for their photodynamic therapy (PDT) of cancer as well as photodynamic antimicrobial chemotherapy activities against biofilms and planktonic cultures of pathogenic bacteria of Staphylococcus aureus and Escherichia coli. Compared to the non-quaternized phthalocyanines 3 and 4, the cationic phthalocyanines 3a and 4a exhibit a higher photodynamic inactivation against the planktonic cells with log reduction values above 9 at a concentration of 1.25 µM. This was attributed to the positive charge which enhances cellular uptake. More interestingly, 3a and 4a show a higher photodynamic inactivation (less than 3% of S. aureus survived) on their biofilm counterparts thanks to their stronger affinity to these cells. 3a and 4a Pcs also exhibited interesting PDT activity against MCF-7 cancer cells giving IC50 values of 17.9 and 7.4 μM, respectively following 15 min irradiation. The obtained results in this work show that the positively charged phthalocyanines 3a and 4a are potential antibacterial photosensitizers that show some selectivity toward the Gram-positive and Gram-negative bacteria as well as MCF-7 breasts cancer cells.

Public programming comprises a sequence of strategies for publicizing and marketing archives to the public, which are crucial in archival operations as they promote better use of archives and records. South African National Archives and Records. Services depends on provisions of the National Archives of South Africa Act of 1996 in its strive to promote access and use of archives as sources of information. . unknown and therefore not used to their maximal utility value, likely due to low-key and ineffective archival public programming initiatives. Consequently, the primary purpose of this research study was to evaluate the extent of public programming strategies used by the Eastern Cape Provincial Archives and Records Services (EAPARS). The study used a mixed-method, embracing qualitative and quantitative research approaches. As an integrated record-keeping framework, Record Continuum was used as a theoretical framework to guide the study. Sixteen copies of questionnaire were distributed among staff and users ECPARS. The research uncovered that ECPARS does not have specific regulations and policies on public programming, but relies on the more general provisions of the Provincial Archives and Records Services Act of 1996 as a guiding instrument in executing its public programming operations. The study further revealed various factors, such as the limited budget and inadequate staff, as key hindering factors in the smooth execution of archival public programming plans by ECPARS. The study recommends a review and update by ECPARS of how it can specifically effectively implement the provisions of the 1996 Act to the modern ways of taking public archives to the people. The research also recommends that ECPARS engage additional staff or outsourced services to strengthen its the public programming of its archival resources and services workforce for Eastern Cape Provincial Archives and Records. The study concludes that improved financial resources, use of ICTs, and coherent policy framework, and public awareness creation and education can facilitate better to acquire, widen access to use of its archives. - Description: Wounds are usually accompanied by complications such as excessive bleeding and bacteria invasion. The design of wound dressings that rapidly stop excessive bleeding and inhibit bacterial invasion is crucial to promoting accelerated wound healing. To meet the abovementioned requirements in wound dressings, topical gels were prepared from sodium alginate SA and carboxymethylcellulose CMC. The wound dressings were loaded with an antifibrinolytic agent, tranexamic acid TA, essential oils, and a variety of metal-based nanoparticles, and carbon-based biomaterials. The scanning electron microscopy SEM and X-ray diffraction XRD confirmed the successful formation of the nanoparticles. The prepared formulations exhibited in vitro drug release kinetics that best fitted with the Korsmeyer-Peppas model. These gels exhibited good spreadability and viscosity, showing a shear-thinning behaviour with pH between 6.7 and 7.3, signifying suitability for skin application and ease of application. The prepared topical gels exhibited significant antibacterial effects against gram-negative and gram-positive strains of bacteria. SA EO-based formulations showed high antibacterial activity across all bacterial strains, followed by SA-based formulations compared to CMC-based formulations, which exhibited moderate antibacterial activity. Moreover, the prepared gels showed good cytocompatibility, promoted cell proliferation, and exhibited 80 percent wound closure on day 3 compared to the untreated group, which showed a 38 percent wound reduction in vitro. Excellent blood clotting properties were observed with CMC-based gels compared to other formulations. However, all the prepared formulations exhibited outstanding blood clotting ability compared to the control, showing that they can promote rapid blood coagulation. However, these scaffolds are commonly encapsulated with therapeutic agents to enhance their biological activities, including antibacterial efficacy. In this research, two types of polymer-based scaffolds were formulated and evaluated as effective formulations for the treatment of chronic wounds: sponges and topical gels. Sponges were formulated from cross-linking of gelatin and PEG. Ag nanoparticles and metronidazole were incorporated into the sponges to improve their antibacterial activity. Topical gels were loaded with essential oils and Ag nanoparticles and prepared from CMC and poloxamer. The prepared sponges and topical gels were evaluated using various analysis and characterization techniques. SEM/EDX, FTIR, and TGA were employed to characterize gelatin/PEG hybrid sponges followed by porosity, in vitro biodegradability, cytotoxicity, and antibacterial studies. FTIR, SEM/EDX, and TGA confirmed their physicochemical properties and successful fabrication of sponges loaded with metronidazole and Ag nanoparticles. The sponges were biodegradable, indicating their capability to induce skin regeneration. The drug release studies showed a rapid release of metronidazole 28.32-71.97 percent from the sponges over the first hour, followed by a sustained drug release. The Ag nanoparticles were released in a sustained manner, suggesting that these sponges can rapidly destroy bacteria and inhibit persisting bacterial infections as well as protect the lesion bed from further bacteria infections. The in vitro antibacterial studies of sponges displayed superior antibacterial activity against most of the Gram-negative and Gram-positive bacteria strains commonly found in chronic wound infections with a MIC value of 15.625 μg/mL. In vitro cytotoxicity experiments revealed excellent biocompatibility with a percent cell viability of more than 70 percent. The in vitro wound scratch healing assay exhibited that the sponges encapsulated with only metronidazole promoted high cell migration than the dual drug-loaded sponges and untreated cells, suggesting its potential to quicken the wound healing process. CMC/Poloxamer topical gels were also characterized by FTIR, followed by pH, viscosity, spreadability, cytotoxicity, and antibacterial studies. The recognition materials were characterized using Fourier transform infrared, cyclic voltammetry (CV), square wave voltammetry (SWV), scanning electron microscopy, energy-dispersive X-ray spectroscopy and X-ray powder diffraction techniques. The electrochemical measurements were performed by employing both CV and SWV methods. We did the optimization of critical parameters affecting the sensor performances through the experimental design and verification. The developed sensor showed a linear range from 10 pM to 100 nM with the calculated quantification and detection limits of 1.86 and 6.19 pM, respectively. The incorporation of AgNP with high electrical conductivity into the MIP matrix enhanced the sensor's performance. Furthermore, the sensor was applied to determine E2 in real water samples without any sample preconcentration steps to achieve the percent recovery of 91.87% to 98.36% and acceptable reusability and storage stability performances.

“Eye on the big prize!”: Iconizing the Democratic Alliance in the Daily Sun

- Description: This article gives a snapshot view of how Mmusi Maimane’s rise to leadership in the Democratic Alliance (DA) in 2015 was reported on in the Daily Sun, South Africa’s biggest-selling national daily newspaper (South African Audience Research Foundation, 2016). Through analysis of a Daily Sun news article exemplifying trends in the positioning of the DA in the tabloid over the first half of 2015, the present study demonstrates how Maimane tried to align the DA around a new iconography (Tann 2010, 2013), centred on the values of “freedom”, “fairness” and “opportunity”. Moreover, the present study also shows how this purported transformation in the DA was treated with scepticism by the news article’s author, who iconizes the DA as incapable of transformation and effective governance. Fine-grained complementary Legitimation Code Theory (LCT) and Systemic Functional Linguistics (SFL) analyses were conducted on this news article. The LCT analysis shows how multiple voices in the news article create conflicting binary constellations, axiologically charged through various linguistic resources, including intertextual references. The analysis, using SFL’s Appraisal system (Martin and White 2005), shows how iconization is accomplished in the news article through evaluative language, coupled with intertextual references, grammatical metaphor andtechnicality to produce syndromes of meaning in the news article. Such iconization works, in this case, to reproduce an attitude of cynicism toward party politics in post-apartheid South Africa. This cynicism foreshadows Maimane’s ultimate lack of success in transforming the discourses of the DA.

The photophysical properties of the prepared complexes were also studied prior to the application. Moreover, a decrease in fluorescence lifetimes was observed upon metalation of complex 1. As anticipated, singlet oxygen quantum yield (ΦΔ)ΦΔ) increased notably upon heavy metal (indium) insertion and upon composite formation. Antimicrobial photodynamic therapy comparative studies were done on quaternized and unquaternized indium porphyrins conjugated to GQDs. Complex 3-GQDs exhibited the highest antibacterial activities compared to other complexes, and this was attributed to the high ΦΔΦΔ which plays an imperative role in photodynamic therapy applications.

- Description: Using artworks emergent from my career as a pracademic and scholar activist, I attempt to share a ‘tactile theory’ of being and doing, that refer mainly to response-abilities (i.e., abilities to respond in accountable ways) in scholar activist educational sociology. I aim to make visible (and tactile) the sometimes-invisible qualities and practices needed for navigating the eroded and dying ecological relations of our generation, as well as warming up and making pliable the heteronormative, capitalist, patriarchal and anthropocentric conventions that are associated with it. In order to warm and sculpt these normative conventions, I argue for the need for ‘suitably strange’ practice. I present six images and associated prose that aim to optimally disrupt these conventions, towards generative rethinking and embodying learning, scholar activism and justice, and from which I explore a tactile theory, an example and related response-ability for each. I end with a reflection of how these suitably strange artefacts can help us develop a new concept of proactive-cognitive justice or ‘justness’.
