Foraging behaviour of the Sooty Albatross in relation to climatic variability and fisheries

They are consequently important for the maintenance of ecosystem structure and function, while also possessing characteristics which are useful for identifying climate-driven changes in oceanography at all trophic levels. This important faunal group is one of the most threatened, with climate change and fisheries bycatch mortality being the primary drivers of population declines. Understanding these threats is important for the management of threatened seabird populations. One impact of climate change and variability in climate occurs through changes in prey availability. As central place foragers, breeding seabirds have trip duration constraints due to the obligation of nest duties. This makes them particularly vulnerable to changes in prey availability within their foraging ranges. The ability to adjust foraging behaviour during environmental conditions which alter prey availability is an important attribute which can contribute to breeding success. Thus, investigating for this foraging behavioural plasticity during different breeding stages provides insights into how a species is likely to be impacted by climate change. The sooty albatross population on Marion Island was studied and displayed a degree of foraging behavioural plasticity during the five-year period of pronounced climatic variability studied. Incubating birds mainly foraged in the subtropical convergence zone (STCZ), the Sub Antarctic Front (SAF) and Subtropical Front (STF) which border it, and the region north of the STF. Brooding birds mainly used the SAF and Polar Frontal Zone (PFZ). Significantly different areas were used between years during both breeding stages. This population showed a lack of consistent habitat preferences which may offer resilience to progressive climatic variability and change. Interactions of sooty albatrosses, as well as white-chinned petrels for comparative purposes, with fisheries were also assessed. The two species were compared to shed light on possible explanations for differences in seabird-fishery interactions between the species. The impact of fisheries-related mortality (bycatch) is dependent on exposure to fishing activity and attraction to fishing boats encountered while foraging. Bycatch mortality has previously been proposed as the main cause of this sooty albatross population’s decline. Exposure and 6 attraction to fishing boats was found to be lower than that of white-chinned petrels which are bycatch prone. However, the current level of attraction exhibited by this sooty albatross population may expose a significant fraction of the population to bycatch risk due to its small population size. Additionally, all fishing boats were encountered north of the STF where non breeding individuals are now known to be distributed. Thus, the impact of fisheries on this population may mainly be mediated by more bycatch prone non-breeding individuals. Overall, the relative impact of fisheries bycatch mortality is suggested to be higher than that of climatic impacts on the sooty albatross population on Marion Island. However, this study did not incorporate dietary and demographic information which would reveal whether there are nutritional differences between years that may impact on breeding success. Towards a community based conservation strategy for wetlands in Uganda

Later, policies and legislation in Uganda favoured agricultural production rather than the conservation of wetlands, a situation that culminated in their degradation. In a bid to counter the effects of wetlands degradation, Wetland Management Agencies were created. These agencies excluded the input of members of the local communities who reside in the wetlands environs. Local communities were not consulted on matters concerning management and conservation strategies, a situation that may have escalated the problem of wetlands degradation. Local knowledge is needed for the effective management and conservation of the wetlands and it is essential that local communities are included in decision making. This research study is more of an initiative for providing information that may be essential for the formulation of a community based conservation strategy for wetlands in Uganda. The study aimed to identify and characterize activities carried out by members of the local communities in wetlands found in central and western Uganda that are responsible for their progressive degradation; to assess the performance of existing Wetlands Management Institutions towards protection and conservation of wetlands in Uganda; to ascertain whether the acts and policies that have been promulgated for the protection of wetlands in Uganda have been adhered to by members of the local communities and to evaluate three community based conservation case studies with two vastly different outcomes in order to come up with the community-based conservation principles Ugandan communities may need to adopt to achieve sustainable utilisation and conservation of wetland resources. To achieve the objectives stated above, the researcher used a multi-disciplinary methodology that required several areas of expertise and sources of information. Hence, to accomplish the study, primary sources of data that included quantitative and qualitative data were collected from the study participants by means of a questionnaire survey, key informant interviews and direct observations. Secondary sources of data centred on published Ugandan acts and policies; textbooks; case study articles and journal articles. The study revealed a host of activities in which members of the local community are involved that have led to the degradation of the wetlands in Uganda. These include but are not limited to crop cultivation, dairy farming and unsustainable wetland resource harvesting. A number of other reasons for the degradation of the wetlands include unclear ownership of the wetlands, unclear wetland boundaries and a lack of awareness of the indirect benefits wetlands provide for members of the local communities. The study disclosed that wetlands management institutions have not been effective in the protection and conservation of wetlands in Uganda, mainly due to political interference and inadequate funding for the institutions’ conservation activities. The acts and policies that were promulgated for the protection and conservation of wetlands have been selectively implemented in some places and in others completely ignored, culminating in the degradation of the wetlands. The study revealed that effective community based conservation of wetlands in Uganda includes: initiation of the planning, implementation, monitoring and evaluation of all wetlands activities being borne by members of the local community; government introducing alternative income-generating activities aimed to reduce over-reliance on wetland resources and education of the local communities regarding the community-based projects’ objectives in order to garner their support. Recommendations emanating from this study are that the Land Act should be amended to specify a particular land tenure system, preferably freehold, which may promote the conservation of wetlands in Uganda; fertilizers (organic) should be introduced in the study area to fertilize the uplands that have reportedly become infertile in order to reduce the pressure the local community members are exerting on the wetlands for agriculture; wetlands management institutions should be empowered financially and political interference in activities aimed at conservation of wetlands in Uganda must cease. There should be uniform implementation of the rules and regulations stipulated in the acts and policies for all members of the public concerning wetlands protection. Lastly, essential community based conservation principles, as stated above, need to be applied carefully and adhered to if effective implementation of community-based conservation of wetlands is to be achieved in Uganda.

Modelling the hydrological responses to changes in land use and cover in the Malaba River Catchment, Eastern Uganda

- Description: Hydrological responses vary from one catchment to another, depending on the nature of land use and cover changes. Modelling the hydrological responses to changes in land use and cover at different catchment spatial scales was the major focus of this study. This study assessed the hydrological responses attributed to changes in land use and extreme weather events resulting into increased sediment loading/concentration, rainfall-runoff generation/volume, streamflow fluctuation and modification of the river channel in the Malaba River Catchment, Eastern Uganda. The hydrological responses were assessed using hydrological models (IHACRES, SCS CN, and SHETRAN) to examine the effect of land use on soil physio-chemical properties susceptibility to rainfall-runoff generation and volume, frequency and severity of extreme weather events, changes in streamflow variations, sediment loading/concentration and river channel morphology. The preliminary study results showed that the frequency of extreme weather events reduced from 4-10 to 1-3 years over the catchment. The performance of the IHACRES model with a Nash-Sutcliffe Efficiency (NSE) of 0.89 showed that streamflow comparatively corresponded with the results obtained the drought indices in predicting the recorded events of severe drought (2005) and flood (1997). Changes in land use and cover types showed that the highest change in the gain of land was experienced from the agricultural land use (36.7 percent), and tropical forest (regeneration) (2.2 percent). The biggest losses in land were experienced in the wetlands (24.6 percent) and bushland and thickets (15.3 percent) land cover types. The SHETRAN model calibrated period had a NSE of 0.78 and 0.81 in the validation period showed satisfactory fits between the measured and simulated streamflow. The agricultural land use (crop growing) had a higher influence on the rainfall-runoff generation and increase in the streamflow than the tropical forest, and bushland cover types in the simulated period. Similarly, the curve number model estimated a comparatively higher surface rainfall-runoff volume generated from the agricultural land use (crop growing) (71,740 m3) than in the bushlands and thickets (42,872 m3) from a rainstorm followed by the tropical forest cover type. This was also reflected in the lower rates of saturated hydraulic conductivity from the agricultural land use (crop growing). The study also showed that human-induced sediment loading due to gold mining activities contributed a much higher impact on the concentration of suspended sediments and streamflow than sediments from rainfall-runoff from the sampled streams. The main contributor of human-induced sediments to the Malaba River were Nankuke River (130.6kg/annum), followed by Omanyi River (70.6kg/annum), and Nabewo River (66.8kg/annum). Human-induced sediment loading had a profound impact on the streamflow variations both in the dry and wet seasons from the sampled tributaries. Lastly, in regard to the effect of land use and cover types on the river channel morphology, tree plantation (cohesion=12, angle of internal friction=27) and bushland and thickets (cohesion=14, angle of internal friction=22) cover types had the most stable river banks compared to the wetland and agricultural land use and cover types that exhibited higher levels of sediment concentration.

Deep learning applied to the semantic segmentation of tyre stockpiles

- Description: The global push for manufacturing which is environmentally sustainable has disrupted standard methods of waste tyre disposal. This push is further intensified by the health and safety risks discarded tyres pose to the surrounding population. Waste tyre recycling initiatives in South Africa are on the increase; however, there is still a growing number of undocumented tyre stockpiles developing throughout the country. The plans put in place to eradicate these tyre stockpiles have been met with collection, transport and storage logistical issues caused by the remoteness and distant locales. Eastwood (2016) aimed at optimising the logistics associated with collection, by estimating the number of visible tyres from images of tyre stockpiles. This research was limited by the need for manual segmentation of each tyre stockpile located within each image. This research proposes the use of semantic segmentation to automatically segment images of tyre stockpiles. An initial review of neural network, convolutional network and semantic segmentation literature resulted in the selection of Dilated Net as the semantic segmentation architecture for this research. Dilated Net builds upon the VGG-16 classification architecture to perform semantic segmentation. This resulted in classification experiments which were evaluated using precision, recall and f1-score. The results indicated that regardless of tyre stockpile image dimension, fairly accurate levels of classification accuracy can be attained. This was followed by semantic segmentation experiments which made use of intersection over union (IoU) and pixel accuracy to evaluate the effectiveness of Dilated Net on images of tyre stockpiles. The results indicated that accurate tyre stockpile segmentation regions can be obtained and that the trained model generalises well to unseen images.

Using an intelligent self-service solution to support telecommunication service provisioning

- Description: The increasing competition between telecommunication service providers (TSPs) and the large amounts of human resource costs incurred to provision telecommunication services to small, medium and micro enterprises (SMMEs), highlight the need for effective self-service provisioning methods to reduce costs. Existing service provisioning methods do not effectively support SMMEs in the service provisioning process. The current methods used are manual and Web-based. The manual service provisioning method is labour and time-intensive. TSPs are being forced to cut human resource costs as the profit margins for provisioning services are narrow due to competition. The current web-based systems do not allow SMMEs to effectively purchase services as they provide long textual lists to consult and limited information about services. The objective of this research was to develop an intelligent web-based self-service system to support SMMEs in telecommunication service provisioning, using a Design Science Research methodology. A constraint-based, knowledge-based recommender system was selected as the most appropriate intelligent system to support telecommunication service provisioning. A prototype called SelPro was developed as a solution to address the shortcomings of the current ineffective service provisioning methods available for SMMEs. SelPro incorporates an interactive wizard-style user interface with dynamic recommendations to support novice users as well as an expert interface which provides less user support. An expert review and a user study were conducted to determine if SelPro was usable, useful and effective. SelPro was perceived as being usable for SMMEs wishing to purchase telecommunication services. Expert participants were satisfied with the usability of SelPro and perceived it as being useful, easy to use and easy to learn. SelPro was considered effective because of the high task success ratings provided by the participants using the two interfaces. The research determined that intelligent web-based self-service provisioning can be used to effectively support SMMEs in telecommunication service provisioning. General design recommendations for similar systems were proposed and future work will involve using intelligent systems to support large enterprises in telecommunication service provisioning.

- Description: The increasing competition between telecommunication service providers (TSPs) and the large amounts of human resource costs incurred to provision telecommunication services to small, medium and micro enterprises (SMMEs), highlight the need for effective self-service provisioning methods to reduce costs. Existing service provisioning methods do not effectively support SMMEs in the service provisioning process. The current methods used are manual and Web-based. The manual service provisioning method is labour and time-intensive. TSPs are being forced to cut human resource costs as the profit margins for provisioning services are narrow due to competition. The current web-based systems do not allow SMMEs to effectively purchase services as they provide long textual lists to consult and limited information about services. The objective of this research was to develop an intelligent web-based self-service system to support SMMEs in telecommunication service provisioning, using a Design Science Research methodology. A constraint-based, knowledge-based recommender system was selected as the most appropriate intelligent system to support telecommunication service provisioning. A prototype called SelPro was developed as a solution to address the shortcomings of the current ineffective service provisioning methods available for SMMEs. SelPro incorporates an interactive wizard-style user interface with dynamic recommendations to support novice users as well as an expert interface which provides less user support. An expert review and a user study were conducted to determine if SelPro was usable, useful and effective. SelPro was perceived as being usable for SMMEs wishing to purchase telecommunication services. Expert participants were satisfied with the usability of SelPro and perceived it as being useful, easy to use and easy to learn. SelPro was considered effective because of the high task success ratings provided by the participants using the two interfaces. The research determined that intelligent web-based self-service provisioning can be used to effectively support SMMEs in telecommunication service provisioning. General design recommendations for similar systems were proposed and future work will involve using intelligent systems to support large enterprises in telecommunication service provisioning.

These components are often volatile in nature and dissipate in a matter of hours. When considering the different types of slow release systems, an overwhelming variety of these systems is available. Each one of the systems is unique in a way, and is designed to perform a particular function, whether it facilitates the controlled release of an active into the body via the skin surface (transdermal delivery) or whether it reduces the rate of loss of an active from the skin surface to the surrounding environment. For the purpose of this study, a previously existing fixative formulation which is believed to reduce the rate of loss of an active component to the environment, through film formation on the skin surface, was investigated. Alternative ingredients or components were incorporated together with the original fixative formulation ingredients into an experimental design which investigates the effect of each group of the components present. 18 formulations with various concentrations of the components within the groups and specified upper and lower limits for each component were formulated. The fixative properties of the formulations were analysed through the incorporation of a fixed amount of a simple fragrance molecule, 4- methoxybenzaldehyde, into each formulation and evaporation studies were conducted in an environmental room at 28±1° C over a period of 5 hours followed by gas chromatography analysis and finally data analyses using statistical methods. The most efficient fixative formulation was established using regression analysis. The fragrance compound in this formulation was found to evaporate at a rate of 0.47 g/L per hour. The least efficient fixative formulation lead to the loss of 0.78 g/L of the fragrance component per hour. From the calculated fragrance concentrations, the rate constant for each individual fixative formulation could be calculated and response surface 8 modelling by backward regression was used in order to determine how each component contributes to the rate of loss of the fragrance compound. Since the sum of the original ingredient and its alternative was constant, each of the original ingredients was coupled directly to its alternative and no conclusion could be made about the contribution of individual components. By increasing the concentration of Hydroxypropylcellulose (HPC) 100K and its alternative HPC 140K, while keeping the effects of the other components constant, a decrease in the rate of fragrance loss was observed. The same conclusion could be made when increasing the concentrations of PEG-12 Dimethicone and its alternative cetyl dimethicone (decreases the evaporation rate). An interaction took place between HPC 100K and PEG-12 dimethicone and their alternatives. The negative effect was, however, not as strong as the combined positive effect on the rate of fragrance loss of the individual components HPC and PEG-12 dimethicone. Evidence suggested that the removal of the components polyvinylpyrrolidone and its alternative, polyurethane-32 (Baycusan® C1003), would improve the effectiveness of the fixative formulation in terms of its slow release properties. A confirmation experiment established that the exclusion of these components from the fixative formulation does improve the “slow release” properties thereof. A larger, more intricate design is required to investigate the effect of each one of the individual components and where the sum of the components (original and its alternative) is not constant.

The presence of a lignotuber and its slow growth seems to hinder the cultivation of this species and wild harvesting is likely to continue. Species distribution modelling indicated that C. intermedia has a climate envelope that spans from the Eastern Cape to the Western Cape along the Cape Fold mountains. This regional distribution is threatened under future climate change scenarios with a range loss and shift identified for climate conditions predicted for 2050. More specifically, its niche was identified as south-facing slopes with sandy to loamy sand soils and water holding capacity that is higher than the surrounding areas. Some keystone Fynbos species were found to share this niche: Leucadendron salignum, Leucospermum cuneiforme, Protea neriifolia, Protea repens, Elegia filacea and Rhodocoma fruticosa. Cyclopia intermedia grows throughout the summer with flowering buds developing in autumn. These enlarge during winter with the flowers opening in early spring. Pods develop in November and seed set occurs during December. Harvesting plants increases their fecundity with cut plants producing more than twice the number of pods and seeds compared to their non-harvested counterparts. This is because resprouting plants produce more flowering stems than uncut plants. Controlled harvesting or localised short-cycle burning could be considered as a management option to improve fecundity of declining Cyclopia intermedia populations in protected areas. C. intermedia harvesting could occur every third year and stem colour was identified as a measure of harvest maturity. Where at least 30 percent of the population has orange (RHS colour 163A) stems the population is suitable for harvesting.

Response of black wildebeest, a specialised grazer, to grass availability

- Description: The black wildebeest Connochaetes gnou, a southern African endemic largely confined to the Grassland biome, was used as a model species to assess the response of a specialised grazer to grass availability, as an indicator of habitat quality. The occurrence of animals in low-quality habitats may have detrimental effects on population performance and persistence. Robust techniques are therefore required to assess the response of animals to varying habitat quality, to ensure sustainability of populations and species. Black wildebeest habitat use was assessed in relation to grass availability and visibility, and faecal analysis was used to estimate variation in diet composition and quality, parasite loads, and faecal pellet dimensions, across a grass gradient. Grass consumption, diet quality, and faecal pellet size and weight of black wildebeest increased with increasing grass availability. While black wildebeest consumed up to 55% browse at grass-poor sites, they consistently preferred grasses, potentially resulting in an increased intake of low-quality grasses at grass-poor sites. This, combined with the increased intake of indigestible fibres in browse, may explain the observed trends in diet quality, which in turn may explain variation in faecal pellet size and weight (indicators of body size and weight). Furthermore, black wildebeest population trends within the study area suggest that populations in grass-poor habitats, outside their natural distribution range, may exhibit reduced population performance. These findings suggest that grass availability may be a useful indicator of habitat quality for specialised grazers. The study also provided an opportunity to explore the utility of various diet assessment techniques for estimating herbivore diet composition and quality. Caution should be applied when interpreting and comparing estimates of diet composition and quality that were estimated using different techniques, because different techniques may yield different diet estimates. The study also suggests that black wildebeest may switch to C3 grasses in the winter (as indicated by isotope analysis).

White-box and black-box modernization are distinct from one another. Grey-box modernization is a hybrid of the white-box and black-box approaches. Modernization can be utilised to create data services for a Service Oriented Architecture. Since it is unclear which modernization approach is more suitable for the development of data services, a comprehensive evaluation framework is proposed to evaluate which of the white- or black-box approaches is more suitable. The comprehensive framework consists of three evaluation components. Firstly, developer effort to modernize existing code is measured by acknowledged software metrics. Secondly, the quality of the data services is measured against identified Quality of Service criteria for data services in particular. Thirdly, the effectiveness of the modernized data services is measured through usability evaluations. By inspection of the combination of application of each of the evaluation components, a recommended approach is identified for the modernization of data services. The comprehensive framework was successfully employed to compare the white-box and black-box modernization approaches applied to a case study. Results indicated that had only a single evaluation component been used, inconclusive results of the more suitable approach may have been obtained. The findings of this research contribute a comprehensive evaluation framework which can be applied to compare modernization approaches and measure modernization success.

- Description: In this work, the selectivity behaviour of two host compounds, namely N,N’-bis(5-phenyl-5- ibenzo[a,d]cycloheptenyl)ethylenediamine (H1) and N,N’-bis(5-phenyl-10,11-dihydro-5- ibenzo[a,d]cycloheptenyl)ethylenediamine (H2), was assessed in mixtures of difficult-toseparate compounds, including the xylenes and ethylbenzene, pyridine and methylpyridines, and anisole and methylanisoles. These host compounds were synthesized using Grignard addition reactions with phenylmagnesium bromide on dibenzo[a,d]cyclohepten-5-one and 10,11- dihydrodibenzo[a,d]cyclohepten-5-one, respectively. The resultant alcohols were treated with perchloric acid to form the corresponding perchlorate salts, which were then reacted with ethylenediamine to afford the required H1 and H2 host compounds. Yields were 95 ad 52% for this final step in each case. An investigation of the conformations, both from experiment (using single crystal X-ray diffractometry (SCXRD) on the apohost compound) and computational calculations was undertaken. Unfortunately, H1 always crystallized out as a powder unless this host compound formed a complex with a guest species, and so could not be subjected to SCXRD analysis as apohost alone. However, a single crystal structure for apohost H2 was successfully obtained and compared with computational data from a previous investigation. It was observed that the conformation of apohost H2 compared favourably with that of the lowest energy conformer from that work. In single solvent recrystallization experiments with o-, m-, p-xylene (o-Xy, m-Xy, p-Xy) and ethylbenzene (EB), both H1 and H2 formed complexes with only one of the four guest species, namely p-Xy and o-Xy, respectively. The host:guest (H:G) ratios were 1:1 for both of these complexes. In the equimolar guest/guest competition experiments, inclusion complexes only formed when p-Xy or o-Xy, respectively, were present in the mixtures; if p-Xy (for experiments with H1) or o-Xy (H2) was absent from these mixtures, only apohost was recovered from the glass vessels. Three selectivity profiles were constructed for each of these host compounds, in which each one was recrystallized from binary mixed guests in various proportions. These demonstrated an overwhelming preference of H1 for p-Xy and H2 for o-Xy, which concurred with the results from the equimolar experiments. Thermal analysis using H1∙p-Xy revealed that this complex was unstable at ambient conditions, while SCXRD explained this observation: this complex may be defined as a true clathrate since no host‧‧‧guest interactions were identified at all, and the guest was retained in the complex by means of steric factors alone. Furthermore, the guest molecules resided in wide open channels that may have facilitated the guest release process. H2∙o-Xy, on the other hand, was stable at room temperature, and the guest release event only commenced at 69.7 °C (Ton). This too was explained by SCXRD analyses in that interactions between host and guest species were observed in this case. Additionally, the guest molecules were housed in discrete cages, which made their escape more challenging compared with p-Xy in H1∙p-Xy. H2∙o-Xy, on the other hand, was stable at room temperature, and the guest release event only commenced at 69.7 °C (Ton). This too was explained by SCXRD analyses in that interactions between host and guest species were observed in this case. Additionally, the guest molecules were housed in discrete cages, which made their escape more challenging compared with p-Xy in H1∙p-Xy. The polymers showed to have higher absorption and emission intensities than their respective starting monomers. Furthermore, the polymers displayed a reversible response towards Hg2+ in the presence of EDTA. This polymer proved to be a sensitive and reversible chemosensor for Hg2+ ions. - Description: Although the marine environment has generally been considered to be less in need of attention from conservationists, politicians and managers than the terrestrial environment, there has been an increasing appreciation that there is little left in the marine environment that can be considered unaltered by humans. Conservation action is therefore urgently needed to protect marine populations, species and environments from anthropogenic pressures. Since overharvesting seems to be the main cause of the collapse in coastal ecosystems, areas need to be protected so that stocks can recover. However, the proclamation of such marine protected areas (MPAs) need to be well planned to ensure that species are well represented and that their biological requirements are met in the long term. While terrestrial conservation planners have started to incorporate biological, social and economic information into so-called systematic conservation plans, marine planners have been more hesitant to use these approaches due to the greater openness of the ocean (with the associated greater dispersal of marine organisms and propogules, lower levels of endemism, and lack of ownership) and the paucity of available information. The current study aimed to investigate the applicability of systematic approaches to the process of selecting sites for a network of potential replenishment zones for seabream species that could supplement protection already afforded by existing no-fishing reserves. Since many of these seabream species are popular targets in both the recreational and commercial linefishery sectors, they have been overexploited and are in need of additional protection. Information on these species and the diversity of their associated reefs were obtained through consultations with scientists, managers and resource-users (namely SCUBA divers, spearfishers as well as recreational and commercial fishers). These data were obtained during workshops where Participatory Rural Appraisal (PRA) scoring and mapping methods were applied. Although ordinal-scale, rather than interval-or ratio-scale data were obtained, these data could be used in an interactive selection process. Because these methods require relatively cheap materials and few personnel, they could be used to collect information where limited budgets are available. Quantitative targets for the protection of individual species were set on the basis of their status in the Marine Living Resource Act, since this reflected their different requirements for protection. A preliminary investigation indicated that the current no-fishing reserves effectively achieved most of these goals for seabream species. The selection of sites for the replenishment network was based on irreplaceability analyses, while decisions between sites with lower irreplaceability values were based on explicit design rules. A total of 26 sites were required to achieve targets for all species. These replenishment zones could potentially be established, managed and monitored by voluntary community groups. This study therefore indicated that systematic conservation planning can successfully be applied to the marine environment, while the involvement of stakeholders is crucial to ensure that conservation action is supported.

- Description: The evaluation of the potential to use Ultra Performance Liquid Chromatography (UPLC) for the simultaneous quantification of all the actives in a multi-active tablet is described in this work. Part of the evaluation was to ensure that the necessary regulatory requirements were adhered to by ascertaining that an analytical method is suitable for a specific purpose through analytical method validation for the specific multi-active tablet. The UPLC method was also tested for the analysis of similar products, namely tablet formulations that contain similar active ingredients in the same proportions but with an additional active ingredient. A method for the simultaneous determination of paracetamol, caffeine and codeine phosphate was developed using UPLC technology. The UPLC developed method was more efficient than the existing in-house HPLC method. The UPLC method was then validated in accordance to ICH and USP guidelines. The application of this UPLC method for the analysis of similar products containing paracetamol, caffeine, codeine phosphate and one extra active ingredient was very challenging. The low concentration of the additional component, differences in sample matrix and differences in formulations added to the challenges. The direct application for the analysis of products Y and Z was not successful; however the method could be used as a platform for further research. A cost comparison between the UPLC and HPLC methods showed the UPLC method to be more cost effective. Thus, while maintenance costs are higher for the UPLC instrument, column costs are comparable to HPLC columns, but solvent and waste disposal charges decrease considerably due to lower solvent use. The reduction in instrument time dramatically improves the cost effectiveness of UPLC over HPLC due to a concurrent reduction in analyst time requirement. The results of this study show that the analytical costs associated with the analysis of multi-active drugs using HPLC procedures can be reduced substantially by the CONFIDENTIAL INTELLECTUAL PROPERTY OF ASPEN PHARMACARE implementation of UPLC technology. The hypothesis that the enhanced chromatographic power of UPLC can be leveraged to provide faster analysis times hence increased product throughput rates, and lower operating costs for the analysis of multi-active drugs was accepted. These advantages were achieved whilst meeting all regulatory requirements for analytical methods as required by regulatory bodies.

The influence of soil properties on the growth and distribution of Portulacaria Afra in subtropical thicket, South Africa

- Description: Subtropical Thicket is the dominant vegetation biome in the Eastern Cape, and extends through to parts of the Western Cape. It is dominated by Portulacaria afra (spekboom), a woody succulent plant recognised for its importance as an ecosystem engineer and its carbon sequestration potential. Due to excessive grazing from domestic stock, spekboom has been completely removed from some areas. The Subtropical Thicket Restoration Programme (STRP) initiated a large scale restoration programme of planting spekboom cuttings in these degraded areas. Their efforts have been met with varying levels of success and improvement of the programme relies on continuous monitoring and scientific evaluation. I investigated the influence of selected soil properties on spekboom growth, mortality and landscape distribution, at both restoration sites and natural intact areas, and through experiments. Site or location was the most important factor influencing spekboom success at restoration plots, whereby sites in the eastern end of spekboom distribution perform better. Moving westwards slope orientation emerged as an important factor, whereby north facing slopes are preferred by spekboom. Although high levels of soil salinity (NaCl) restricted spekboom growth and affected its health, it could tolerate the levels it was exposed too. Soil pH, above 7, and phosphorous concentration, above 70 mg.kg-1, were the only limiting factors to spekboom survival found in the restoration sites. This preference for acidic soils was mirrored in intact Thicket. However in general, soil is not a major factor influencing spekboom growth and distribution, and spekboom is tolerant of a wide range of soil conditions. Spekbooms constraint is most likely a function of climate, which varies greatly across the biome. This study answered some vital questions regarding the possible influence of soil in spekboom growth and distribution. It disapproved the theory that a catena effect may be responsible for the lack of spekboom growing in bottomland areas. The study also indicates and supports the versatility of spekboom as a plant for restoring degraded lands across a range of different geologies and soil types. To maximise spekboom survival rates, restoration efforts should be focussed towards the eastern end of its distribution and to avoid planting in soils with pH levels higher than 7.

The aim of this research was to investigate using information visualization (IV) techniques to effectively support web service discovery. The node-and-link network IV technique was selected as the most appropriate IV technique to visualize web service collections. A prototype, called SerViz, was developed as a tool for interactive visualization of web service collections incorporating the node-and-link IV technique and an alphabetical list-based technique. SerViz used the Programmable Web web service collection as the sample web service collection. A usability evaluation was conducted to compare these techniques. Ninety percent of participants preferred the network IV technique for visualizing web service collections. The network IV technique was also faster for browsing. Several usability problems were identified with the network IV technique. This motivated a need for implementing an alternative IV technique in SerViz. The node-and-link tree IV technique was selected as it was more structured than the network IV technique. A usability evaluation was conducted to compare the network and tree IV techniques. Participants slightly preferred the tree IV technique as the technique to visualize web service collections. These techniques comprised an Overview using a nested circles layout, a Tag Cloud and a Partition Layout, which used a novel set-based technique. A prototype, called MyPSI, was designed and implemented incorporating these enhanced IV techniques. The requirements and design of the MyPSI prototype were validated using a conceptual walkthrough. The design of the MyPSI prototype was initially implemented for a desktop or laptop device with mouse-based interaction. A sample personal space of information (PSI) was used to evaluate the prototype in a controlled user study. The user study was used to identify any usability problems with the MyPSI prototype. The results were highly positive and the participants agreed that such a tool could be useful in future. No major problems were identified with the prototype. The MyPSI prototype was then implemented on a mobile device, specifically an Android tablet device, using a similar design, but supporting touch-based interaction. Users were allowed to upload their own PSI using Dropbox, which was visualised by the MyPSI prototype. A field study was conducted following the Multi-dimensional In-depth Long-term Case Studies approach specifically designed for IV evaluation. The field study was conducted over a two-week period, evaluating both the desktop and mobile versions of the MyPSI prototype. Both versions received positive results, but the desktop version was slightly preferred over the mobile version, mainly due to familiarity and problems experienced with the mobile implementation. Design recommendations were derived to inform future designs of IV tools to support accessing PI across multiple devices. This research has shown that IV techniques can be enhanced to effectively support accessing PI across multiple devices. The antifungal activity of an aqueous Tulbaghia violacea plant extract against Aspergillus flavus

- Description: Phytochemical analysis of both HEA1 and the crude plant extract showed the presence of phenolics, tannins and saponins. Saponins were the predominant secondary metabolites and were mostly abundant in the plant extract and to a lesser extent in the active compound. Steroidal saponins, tannins and phenolics were also detected in the plant extract, but only the phenolics were detected in the active compound. The results of the phytochemical analysis showed that those compounds that were not present in the active compound could be removed from the crude extract during the TLC purification process. Investigation on the mechanism of action of the crude plant extract on the sterol production by A. flavus showed that the plant extract affected ergosterol biosynthesis by causing an accumulation of oxidosqualene in the ergosterol biosynthetic pathway resulting in a decline in ergosterol production. An oscillatory response in lanosterol production was observed in the presence of the plant extract, which may be an adaptation mechanism of A. flavus to unfavourable conditions and compensation for the loss of enzyme activity which may have occurred as a result of the accumulation of oxidosqualene. The antifungal activity of the plant extract on ergosterol production by A. flavus may also be due to saponins which target the cell membrane and ergosterol production in fungi. The effect of the plant extract on the fungal cell wall of A. flavus also showed that the plant extract caused a decline in β-(1, 3) glucan production by inhibiting β-glucan synthase. The plant extract also affected the chitin synthesis pathway of A. flavus, by causing a decline in chitin production, which was due to the inhibition of chitin synthase. Investigation of chitinase production using 4MU substrates showed that the plant extract caused an accumulation of chitobioses, by activating chitobiosidases and endochitinases. A decline in N-acetylglucosaminidase activity in the presence of the plant extract was observed and this prevented the formation of N-acetylglucosamine. The accumulation of chitobiosidase and endochitinase may be as a result of autolysis that may be triggered by A. flavus as a survival mechanism in the presence of the plant extract and as a compensatory mechanism for the loss of β-glucans and chitin. The antifungal effect of the plant extract on various components of the cell wall of A. flavus, makes T. violacea aqueous plant extract an ideal chemotherapeutic agent against both human and plant pathogens of Aspergillus. The broad spectrum of antifungal activity of T. violacea against A. flavus also eliminates any chances of the fungus developing resistance towards it and would make it a candidate for use as a potential antifungal agent. Further identification and possible chemical synthesis is needed to shed light on the safety and efficacy of the active compound for further development as a chemotherapeutic agent.

It is estimated that about 25 percent of prescribed medicines contain plant products or active compounds derived from plants. In South Africa, traditional medicine forms part of the culture and tradition of most communities. Garlic compounds have been shown to have a variety of antimicrobial properties. Amongst these are antifungal, antibacterial, antiviral and anti protozoal activities. Allicin and its breakdown products have been shown to be the main active compounds which possess these properties. Tulbaghia violacea has been used for the treatment of a variety of illnesses including asthma, fever, oesophageal cancer, constipation and hypertension. This study investigated the antifungal nature of T.violacea on the morphology, spore germination and lipid synthesis of Aspergillus flavus and Aspergillus parasiticus. The results of this study showed that the plant extract inhibited A. flavus growth at a minimal inhibitory concentration of 15mg/ml and was fungicidal at 20mg/ml and above. A. parasiticus was not inhibited at 25mg/ml indicating resistance to the inhibitory component of the plant extract. A measure of metabolic activity using the XTT assay showed reduced metabolic activity in the presence of increasing concentrations of the plant extract. Higher extract concentrations resulted in higher percentage inhibition of fungal growth for both fungal species with up to 98 percent inhibition being observed for the highest extract concentrations for both fungi. Germination was also delayed in the presence of 15mg/ml plant extract concentration by up to 60hr for A. flavus and 48hr for A. parasititcus. The TEM results showed increased thickening of the cell wall with higher extract concentrations. The thickening was greater for A. flavus than for A. parasiticus. Cell wall thickening may be the reason for the delay in germination in both species. Lipid production was reduced in the presence of plant extracts when compared to the control. The plant extracts inhibited triglyceride production at 15mg/ml for both A. flavus and A. parasiticus. The results therefore indicate that T. violacea extracts are antifungal and probably affect germination through interactions with the cell wall. - Description: A six-step synthesis of salbutamol from methyl salicylate with an overall yield of 17% has been completed, although the yield was not optimised. In the process, Zn(BH4)2 was found to selectively reduce a ketone carbonyl group in the presence of an ester unit. In contrast, borane was found to reduce both the ketone and ester carbonyl groups. Reduction of phenacyl bromide with borane in the presence of chiral catalysts based on (R)-alaninol and (R,S)-ephidrine resulted a measure of enantioselectivity in the product. However, the configuration of the alcohol obtained in the case of (R)-alaninol was contrary to expectations based both on experimental trends observed elsewhere as well as our own theoretical predictions. The asymmetric reduction of methyl 5-bromoacetyl-2-benzyloxybenzoate was accomplished with both borane and Zn(BH4)2 in the presence of a range of chiral catalysts. Optically active products were obtained in all cases, although the optical rotations were significantly smaller in the case of Zn(BH4)2. Unfortunately, we were not successful in determining the enantiomeric excesses of these reactions. The use of a NMR lanthanide shift reagent resulted in a complex spectrum that was impossible to interpret unambiguously. This presumably arises from the presence of several Lewis base sites in the product at which complexation with the shift reagent can take place. It was also not possible to determine the optical rotation of salbutamol itself owing to the relatively small amount of material obtained. A conformational analysis of salbutamol, where NMR data was correlated with molecular modelling results, was successfully carried out and revealed a strong preference for that conformer family characterised by O–C–C–N and Ar–C–C–N torsion angles of ca. 60º and 180º, respectively. Interestingly, these conformers are found to be stabilised by OH…N rather than NH…O hydrogen bonding. This study has also confirmed the effectiveness of the MMFF94 force field for conformational analysis studies in compounds of this kind. Lastly, a relatively simple method for modelling the BH3/oxazaborolidine reduction of ketones at the PM3 semiempirical MO level of approximation was devised. This approach has provided insights into the mechanism of the reaction and has furthermore enabled us to predict the enantioselectivities likely to result from various catalysts and ketones. In comparing our theoretical and experimental findings, an anomalous result was observed in the case of (R)-alaninol; this will have to be investigated further, particularly at the experimental level. However, we believe that our approach provides a sound basis for aiding the design and screening of new, potentially better catalysts.
